Abstract
La presente invención proporciona pares de heterodímeros que pueden comprender un primer heterodímero y un segundo heterodímero donde cada heterodímero comprende una cadena pesada de inmunoglobulina o fragmento de esta y una cadena ligera de inmunoglobulina o fragmento de esta. Al menos uno de los heterodímeros puede comprender una o más modificaciones de aminoácidos en los dominios CH1 y/o CL, una o más modificaciones de aminoácidos en los dominios VH y/o VL o una combinación de estas. El o los aminoácidos pueden ser parte de la interfaz entre la cadena ligera y la cadena pesada y se modifican típicamente para crear emparejamiento preferencial entre cada cadena pesada y una cadena ligera deseada de modo que cuando las dos cadenas pesadas y las dos cadenas ligera del par de heterodímeros se coexpresan en una célula, la cadena pesada del primer heterodímero se empareja preferentemente con una de las cadenas ligera en vez de con la otra. De manera similar, la cadena pesada del segundo heterodímero típicamente se empareja preferentemente con la segunda cadena ligera en vez de con la primera.
