Abstract

Se proporciona un kit para detectar múltiples ácidos nucleicos con especificidad de objetivo, que sean capaces de amplificar y detectar simultáneamente múltiples genes por medio de un recipiente de reacción que contenga un tipo de solución de reacción y un tipo de marcadores. La solución puede contener un primer ácido nucleico con especificidad de objetivo (10) y un segundo ácido nucleico con especificidad de objetivo (20) de los cuales cada uno se disocia a la temperatura de desnaturalización T0. La solución contiene adicionalmente: ADN polimerasa (30); un cebador del primer objetivo (13) a temperatura de alineamiento T1 que se une con las primeras cadenas simples derivadas del primer ácido nucleico con especificidad de objetivo; un cebador del segundo objetivo (23) a temperatura de alineamiento T1 con segundas cadenas simples derivadas del segundo ácido nucleico con especificidad de objetivo; una sonda del primer objetivo (15) a temperatura de alineamiento T1 que se une con las primeras cadenas simples derivadas del primer ácido nucleico con especificidad de objetivo; y una sonda del segundo objetivo (25) a una temperatura de detección del segundo objetivo T3 que es menor que la temperatura de alineamiento T1 y la temperatura de elongación T2 uniéndose con las segundas cadenas simples derivadas del segundo ácido nucleico con especificidad de objetivo. A condición de que: se cumpla que T0 sea mayor que T2; T2 no sea menor que T1; y T1 sea mayor que T3. Figura que se seleccionará: Figura 36.
