Abstract
Receptores de antígenos quiméricos (CAR). Los CAR tienen la capacidad de redirigir la especificidad y reactividad de la célula inmune hacia un blanco seleccionado haciendo uso de las propiedades de dominio de unión a ligando. En particular, la presente invención se relaciona con un receptor quimérico de antígeno en donde la unión a ligando extracelular es un scFv derivado de un anticuerpo monoclonal contra CD19, preferentemente 4G7. La presente invención también se relaciona con polinucleótidos, vectores que codifican para los CAR y células aisladas que expresan el CAR en su superficie. La presente invención también se relaciona con métodos para diseñar células inmunes que expresan 4G7-CAR en su superficie que confiere un estado "activado" prolongado en la célula transducida. La presente invención es particularmente útil para el tratamiento de linfomas de células B y leucemia.
