Abstract

본 발명은 중심물질인 지방산이 사람의 혈청 알부민으로 감싸여진 직경이 약 12㎛ 미만인 미립자 및 이것을 제조하는 방법에 관한 것이다. 이들은 분산된 마이크로버블에서 얻어진 것과 동일하거나 또는 이보다 큰 산란세기를 갖지만 저장 및 생체내에 사용할때 분산된 마이크로버블을 기본으로하는 콘트라스트제보다 훨씬 안정한 초음파 화상형성용 콘트라스트제로서 유용하다.

Claims (27)

  1. 하나 이상의 지방산으로 된 중심물질을 사람의 혈청 알부민으로 감싼 형태로 이루어진, 평균 직경이 12㎛이하인 입자들로 구성된 진단용 화성형성제.
  2. 제1항에 있어서, 입자의 평균 직경이 0.1-12㎛인 화상형성제.
  3. 제2항에 있어서, 평균 직경이 0.1-10㎛인 화성형성제.
  4. 제3항에 있어서, 평균 직경이 0.1-8㎛인 화성형성제.
  5. 제1항에 있어서, 지방산의 탄소원자수가 6-18인 화성형성제.
  6. 제5항에 있어서, 지방산이 포화된 화성형성제.
  7. 제5항에 있어서, 지방산이 직쇄인 화상형성제.
  8. 제2항에 있어서, 지방산이 불포화된 화상형성제.
  9. 제5항에 있어서, 지방산이 분지쇄를 갖고있는 화상형성제.
  10. 제1항에 있어서, 지방산과 섞일수있는 보조약을 더 포함하는 화상형성제.
  11. 제10항에 있어서, 보조약이 생리학적으로 적당한 오일 및 계면활성제들에서부터 선택된 화상형성제.
  12. 제11항에 있어서, 보조약이 콜레스테롤인 화상형성제.
  13. 제10항에 있어서, 보조약의 양이 지방산의 전체 중량의 0-50wt%로 존재하는 화상형성제.
  14. 제1항에 있어서, 사람의 혈청 알부민 대 지방산의 중량비가 10:1-1:1인 화상형성제.
  15. 제14항에 있어서. 상기 비율이 4:1인 화상형성제.
  16. 사람의 혈청 알부민으로 피복된 지방산 미립자의 분산물을 만들고 이렇게 만들어진 분산물을 가열하여 사람의 혈청 알부민을 응고시키는 것으로 구성된, 하나 이상의 지방산 중심물질이 사람의 혈청 알부민으로 감싸여진 평균 직경이 12㎛이하인 입자들로 구성된 진단용 화상형성제를 제조하는 방법.
  17. 제16항에 있어서, 분산물을 90℃ 보다 높은 온도까지 가열하는 방법.
  18. 제17항에 있어서, 분산물을 최소한 45분간 가열하는 방법.
  19. 제18항에 있어서, 분산물을 최소한 1시간동안 가열하는 방법.
  20. 제16항에 있어서, 분산물을 60-600RPM의 속도로 교반하는 방법.
  21. 제16항에 있어서, (a) 지방산을 용매에 용해시켜 지방산 용액을 만들고, (b) 지방산 용액을 사람의 혈청알부민 용액과 혼합하여 사람의 혈청 알부민으로 피복된 지방산 미립자의 분산물을 만든뒤, (c) 만들어진 분산물을 빠르게 교반하면서 90℃보다 높은 온도까지 가열하여 사람의 혈청 알부민을 응고시키는; 단계들로 구성된 방법.
  22. 제16항에 있어서, (a) 지방산 염의 용액을 산성화시켜 지방산의 유제를 만들고, (b) 사람의 혈청 알부민을 부가한 다음, (c) 이를 가열하여 사람의 혈청 알부민을 응고시키는; 단계들로 구성된 방법.
  23. 제22항에 있어서, 단계(b)로 넘어가기전에 지방산 용액내으로 가스를 뿜어올리는 방법.
  24. 제23항에 있어서, 가스가 산소로 구성된 방법.
  25. 제24항에 있어서, 최소한 8시간동안 가스를 뿜어올리는 방법.
  26. 제25항에 있어서, 최소한 24시간동안 가스를 뿜어올리는 방법.
  27. 제26항에 있어서, 6일동안 가스를 뿜어올리는 방법.
