Abstract
HLA 제I형 유전자의 제2 및 제3엑손의 특이적 핵산서열 증폭용 프라이머 및 증폭된 DNA중에 함유된 다형상 서열 확인용 탐침이, 다양한 공급원으로부터의 잡종 또는 동종 샘플을 타이핑하고 혈청학적방법으로는 구별되지 않는 대립형질 변이체를 검출하는데 사용된다. 이러한 HLA 제I형 DNA타이핑 시스템을 수행하기가 간단하고 신속하며, 신속하게 검출시그날을 생성하며, 조직 타이핑에 사용할 수 있는 포워드(forward) 또는 리버스(reverse)도트-블럿 포맷에 이용할 수 잇으므로 개체의 신원을 파악하고 병에 걸리기 쉬운 개체를 확인할 수 있다.

제1도는 2개의 프라이머 쌍 및 고정화된 탐침을 갖는 다수의 막 스트립으로 제I형 HLA A, B 및 대립형질을 확인하는 본 발명의 하나의 양태를 나타낸다.
Claims (17)
- (a) 제I형 HLA 대립형질 제2 또는 제3엑손 서열을 함유하는 샘풀중의 핵산을 증폭시키고, (b) 단계(a)에서 증폭시킨 핵산을, 탐침이 단지 정확히 상보적인 서열에만 하이브리드화하는 조거하에서, 올리고뉴클레오타이드 탐침의 패널에 하이브리드화시킨 후, (c) 단계(b)에서의 탐침 하이브리드화의 양상으로부터, 샘플중의 핵산이 기인하는 제I형 HLA 대립형질을 결정함을 특징으로 하여, 샘플중의 핵산의 HLA 제I형 DNA타입을 결정하는 방법.
- 제1항에 있어서, 제I형 HLA 대립형질이 A, B 및 C 대립형질로 이루어진 구릅 중에서 선택되는 방법.
- 제2항에 있어서, 제I형 HLA 대립형질이 A 및 B 대립형질인 방법.
- 제1항 내지 3항중 어느 한 항에 있어서, 단계(a)의 증폭을 한쌍의 올리고뉴클레오타이드 프라이머를 사용한 폴리머라제 쇄 반응으로 수행하는 방법.
- 제4항에 있어서, 제2엑손 서열을 증폭시키는 방법.
- 제5항에 있어서, 프라이머가 DB308(서열확인번호 1) 및 DB309(서열확인번호 2)인 방법.
- 제4항에 있어서, 제3엑손 서열을 증폭시키는 방법.
- 제7항에 있어서, 프라이머가 DB337(서열확인번호 4) 및 DB311(서열확인번호 3)인 방법.
- 제4항에 있어서, 제2 및 제3엑손 서열 모두를 증폭시키는 방법.
- 제9항에 있어서, 프라이머가 DB308(서열확인번호 1), DB309(서열확인번호 2, DB337(서열확인번호 4), 및 DB311(서열확인번호 3)인 방법.
- 제1항 내지 10항중 어느 한 항에 있어서, 올리고 뉴클레오타이드 탐침의 패널이 고체지지체상에 고정되고, 각각의 탐침이 고체 지지체상의 분리된 별개의 위치에 위치하는 방법.
- 올리고뉴클레오타이드 탐침의 패널을 포함하는, 제1항 내지 11항중 어느 한 항에 따른 방법을 수행하기 위한 키트.
- 제12항에 있어서, 제2 및/또는 제3엑손 서열을 증폭시키기 위한 한 쌍의 올리고뉴클레오타이드 프라이머를 추가로 포함하는 키트.
- 제12항 내지 13항에 있어서, 탐침의 패널이 하기로 이루어진 그룹중에서 선택된 2개 이상의 탐침을 포함하는 키트, DB312(서열확인번호 5), DB313(서열확인번호 6), DB314(서열확인번호 7), DB315(서열확인번호 8), DB316(서열확인번호 9), DB317(서열확인번호 10), DB318(서열확인번호 11), DB319(서열확인번호 12), DB320(서열확인번호 13), DB321(서열확인번호 14), DB322(서열확인번호 15), DB323(서열확인번호 16), DB324(서열확인번호 17), DB325(서열확인번호 18), DB326(서열확인번호 19), DB327(서열확인번호 20), DB328(서열확인번호 21), DB329(서열확인번호 22), DB330(서열확인번호 23), DB331(서열확인번호 24), DB332(서열확인번호 25), DB333(서열확인번호 26), DB334(서열확인번호 27), DB335(서열확인번호 28), DB336(서열확인번호 29), RAP10(서열확인번호 30), RAP11(서열확인번호 31), RAP12(서열확인번호 32), RAP13(서열확인번호 33), RAP14(서열확인번호 34), RAP15(서열확인번호 35), RAP16(서열확인번호 36), RAP17(서열확인번호 37), RAP18(서열확인번호 38), RAP19(서열확인번호 39).
