Claims (10)
- a)적합한 트랜스아미나제 1의 존재하에서 아스파르테이프 및 α-케토글루타르에이트를 반응시켜 옥살아세테이트 및 글루타메이트를 수득한 후, b)적합한 트랜스아미나제 2의 존재하에서 글루타메이트 및 구조식(Ⅱ)의 HMPB를 반응시켜 α-케토글루타메이트 및 L-포스피노트리신을 수득하는 반응단계를 특징으로 하는 커플링된 효소반응(여기서, 아스파르테이트대 HMPB의 몰비는 0.5 내지 1.5대 1, 바람직하게는 0.8 내지 1.2대 1, 특히 동물비이고 글루타메이트 또는 α-케토글루타르에이트는 촉매량으로 첨가한다)으로, 구조식(Ⅱ)의 4-(하이드록시메틸포스피닐)-2-옥소부티르산(HMPB)으로 부터 구조식(Ⅰ)의 L-2-아미노-4-(하이드록시메틸포스피닐)부티르산(L-포스피노트리신)을 제조하는 방법.
- 제1항에 있어서, 글루다메니트 또는 α-케토글루타르에이트를 HMPB에 대해 0.01 내지 1대 1, 바람직하게는 0.01 내지 0.2대 1, 특히 0.05 내지 0.2대 1의 몰비로 첨가하는 방법.
- 제1항 또는 제2항에 있어서, 트랜스아미나제 1이 글루타메이트/옥살아세테이트 트랜스아미나제 활성(GOT활성)을 갖는 방법.
- 제3항에 있어서, GOT를 돼지, 에스케리치아 콜라이 또는 바실러스, 바람직하게는 에스케리치아 콜라이 또는 바실러스로부터 수득하는 방법.
- 제1항 또는 제2항에 있어서, 트랜스아미나제 2가 L-포스피노트리신 트랜스아미나제 활성을 갖는 방법.
- 제5항 있어서, L-포스피노트리신-특이적 트랜스아미나제를 에스케리치아 콜라이로 부터 수득하는 방법.
- 제1항 내지 제6항중 어느 한 항에 있어서, 옥살아세테이트가 다중 하전된 양이온, 바람직하게는 Al3+, Mg2+, Zn2+, Mn2+, Fe2+또는 Fe3+의 존재하에서 피루베이틀 탈카복실화되는 방법.
- 제7항에 있어서, 트랜스아미나제 1 및 트래스아미나제 2가 피루베이트를 알라닌으로 전환시키지 않는 방법.
- 제1항 내지 제8항중 어느 한 항에 있어서, 하나 이상의 트랜지스아미나제가 고정화된 형태로 존재하는 방법.
- 제9항에 있어서, 고정화된 트랜스아미나제가 컬럼 반응기내에 존재하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
