Claims (9)
- (a)포르스콜린(6-위치에 OH를 갖는 일반식(Ⅰ)의 화합물)을 염화수소의 존재하에 아세톤과 반응시켜서 1,9-이소프로필덴 오도체를 생성시키고, (b)이 유도체를 알칸올 중에서 알칼리로 처리하여 7-데아세틸 유도체를 수득하며, (c)이 유도체를 유기 염기의 존재하에 β-할로프로피오닐할라이드로 처리하여 7β-아크릴로일옥시-7-데아세틸 프로스콜린의 1,9-이소프로필리덴 유도체를 형성시키고, (d)이 유도체를 알칼리로 처리하여 6β-아크릴로일옥시-7-데아세틸 포르스콜린의 1,9-0-이소프로필리덴 유도체를 수득하며, (e)이를 7β-아세록시 유도체로 변형시키고, (f)이를 일반실(여기서, R1및 R2는 하기 정의하는 바와 같다)의 아민과 반응시켜서 6-위치에 (3-치환된 아미노)프로피오닐옥시 유도체를 형성시키며, (g)1,9-위치에서 수득된 화합물을 탈보호시켜서 일반식(Ⅰ)의 화합물을 수득하고, (h)이를, 경우에 따라,약제학적으로 허용되는 염으로 전환시킴을 특징으로 하여(여기서, 단계(e)와 (f)의 순서 및 또한 단계 (f)와 (g)의 순서는 교체될 수 있다), 일반식(Ⅰ)의 6β-(3-치환된 아미노)프로피오닐옥시포르스콜린 유도체 및 이의 약제학적으로 허용되는 염을 제조하는 방법.상기식에서, R1및 R2는 각각 수소, 알킬, 아릴, 아르알킬 또는 디알킬아미노알킬을 나타내거나; R1및 R2가 이들이 결합되어 있는 질소와 함께 추가의 헤테로 원자(예 : N, O 및 S)를 함유할 수 있고 하나 이상의 위치에서 알킬, 알콕시, 하이드록실, 할로겐 또는 아릴에 의해 임의로 치환될 수 있는 헤테로사이클을 형성한다.
- 제1항에 있어서, 단계(c)에서 β-할로프로피오닐 할라이드가 β-클로로프로피오닐 클로라이드인 방법.
- 제1항 또는 제2항에 있어서, 단계(f)에서 아민이 디메틸아민인 방법.
- 제1항 내지 제3항중 어느 한항에 있어서, 단계 (a)/(b) 및 단계(e)/(f)를 윈-포트(one-pot)반응으로 수행하는 방법.
- 7β-아크릴로일옥시-6β-하이드록시-1,9-0-이소프로필리덴-8,13-에폭시-labd-14-엔-11-온.
- 6β-아크릴로일옥시-7β-하이드록시-1,9-0-이소프로필리덴-8,13-에폭시-labd-14-엔-11-온.
- 7β-아세톡시-6β-아크릴로일옥시-1,9-0-이소프로필리덴-8,13-에폭시-labd-14-엔-11-온.
- 7β-아세톡시-6β-〔3-(디메틸아미노프로피오닐)옥시〕-1,9-0-이소프로필리덴-8,13-에폭시-labd-14-엔-11-온.
