생물학적 활성 플라스미노겐 활성제의 제조방법

제1도는 본 발명에 의한 t-PA 샘플의 은으로 착색한 겔을 나타낸 것임.
제2도는 제1도에 나타낸 것과 동일한 겔의 면역 블로프(immunoblot)를 나타낸 것임.

Claims (18)

  1. 숙주 세포 배양액 중에서 이 배양액의 배지로부터 다음의 인체조직형 플라스미노겐 활성제의 회수 및 활성에 대하여 장해적인 배지 성분 모두를 실질적으로 제거하고, 동시에 숙주 세포의 표현 용량을 보유시키는 것으로 되는 생물학적 활성 인체 조직형 플라스미노겐 활성제의 제조방법.
  2. 제1항에 있어서, 배양시키기 전 장해 성분들을 제거하는 방법.
  3. 제1항에 있어서, 배양도중 배지 교환에 의하여 배지로부터 장해 성분들을 제거하는 방법.
  4. 제3항에 있어서, 배양 도중 교류 여과에 의하여 배지로부터 장해 성분들을 제거하는 방법.
  5. 제4항에 있어서, 평면 여과 막을 통하여 교류 여과를 행하는 방법.
  6. 제4항에 있어서, 가공 공동 섬유의 벽을 통하여 교류여과를 행하는 방법.
  7. 제4항에 있어서, 나선형 상처 막을 통하여 교류 여과를 행하는 방법.
  8. 전항 중 어느 하나의 항에 있어서, 숙주 세포 배양액이 재조합 숙주 세포 배양액인 방법.
  9. 제8항에 있어서, 재조합 숙주 세포 배양액이 재조합 숙주 세포 배양 현탁액인 방법.
  10. 제1항에 있어서, 인체 조직형 플라스미노겐 활성제의 회수 및 활성에 대하여 장해적인 성분을 하나이상 함유하는 성장 및 세포 복제 지지 배지 중에서 숙주 세포 배양액을 성장시키고, 하나 이상의 장해 성분 모두를 교류 여과에 의하여 숙주 세포 배양액으로부터 실질적으로 제거하고, 이 숙주 세포 배양액에 인체조직형 플라스미노겐 활성제의 회수 및 활성에 대한 장해 성분을 실제로 갖지 아니하며 추가로 세포 복제를 지지할 필요없이 인체 조직형 플라스미노겐 활성제의 숙주 세포 배양액에 의한 표현을 허용할 수 있는 추가배지를 공급하여 장해 성분을 제거하는 방법.
  11. 제1항에 있어서, 성장 및 세포 복제 지지 배지 성분이 혈액, 조직 또는 그의 유도체로부터 유도된 것인 방법.
  12. 제1항에 있어서, 숙주 세포 배양액이 인체 조직형 플라스미노겐 활성제를 생성시킬 수 있는 포유류세포로 된 것인 방법.
  13. 제1항에 있어서, 숙주 세포 배양액이 제조합 표면에 의하여 인체 조직형 플라스미노겐 활성제를 생성시킬 수 있는 포유류 세포로 된 것인 방법.
  14. 제13항에 있어서, 포유류 세포가 재조합 트란스펙트 CHO 세포로 되는 방법.
  15. a) 인체 조직형 플라스미노겐 활성제를 생성시킬 수 있는 세포 b) i)인체 조직형 플라스미노겐 활성제의 회수 및 활성에 대한 장해 성분이 실제로 없으며, ii)숙주 세포 배양액 중에서 세포 복제를 더 지지할 필요 없이 인체 조직형 플라스미노겐 활성제의 표현을 허용할 수 있는 배지로 되는 숙주 세포 배양액.
  16. 재조합 트란스펙트 숙주 세포로 되는 제12항에 의한 세포배양액.
  17. 제조합 트란스펙트 포유류 세포로 되는 제12항에 의한 세포배양액.
  18. 제17항에 있어서, 포유류 세포가 재조합 트란스펙트 CHO 세포로된 것인 세포 배양액.
