Abstract
백금 등의 전이금속의 미분말을 포함하는 초산화음이온 또는 일산화질소의 분해제. 바람직하게는 전기 분해제는 1,000mL당 1mM 이하의 비율로 전이금속 콜로이드를 포함하는 수성의 분해제로 제공되며, 생체 내의 과잉 초산화음이온 또는 일산화 질소를 효율적으로 제거할 수 있다.

Description
본 발명은 활성산소의 일종인 초산화음이온의 분해제에 관한 것이다. 본 발명의 초산화음이온의 분해제는 환원수나 의약품으로 이용할 수 있다. 또한, 본 발명은 일산화질소의 분해제에 관한 것이다.
생체 내, 특히 미토콘드리아, 마이크로솜, 백혈구 등에서, O2 -(초산화음이온), H2O2(과산화수소), HOㆍ(히드록시라디칼) 및 여기분자종(勵起分子種)인 1O2(일중항산소) 등의 높은 반응성을 보이는 활성산소종(라디칼)이 많이 발생하고 있으며, 생태방어, 생화학 반응 등을 포함한 생체제어에 관여하고 있다고 알려져 있다. 또한, 일산화질소(NO)는 단수명의 불안정 라디칼 종이며, 이 물질도 활성산소의 일종으로 생체 내에서 중요한 기능을 가지고 있음이 명백하다고 알려져 있다(참조: 현대화학, 1994년 4월호 특집).
정상적인 세포에서 이들 활성산소종의 생성량은 주반응인 산화환원반응등량 의 1% 정도이며, 분해효소 등에 의하여 순차적으로 대사되고 있다. 사람이 호흡으로 체내에 공급한 산소의 95% 이상은 통상의 대사과정을 거쳐 물로 되지만, 나머지 수 %는 미토콘드리아나 마이크로솜의 전자전달계에서 활성산소로 되고, 이들은 대개의 경우 초산화물 불균등화효소(superoxide dismutase: SOD), 카탈라아제, 글루타티온 퍼록시다아제 등의 항산화효소에 의해 제거되고 있다.
그러나, 이들의 항산화효소로 모든 활성산소를 제거할 수 없으며, 활성산소의 일부는 단백질, 지질, 핵산 등을 산화시킨다. 이들의 산화된 물질은 다른 생체방어기구에 의하여 일부 복원되지만, 천천히 비가역적으로 산화손상된 물질이 생성되며, 그 결과, 질병이나 노화와 관련이 되고 있다.
또한, 나이를 먹음에 따라, 초산화물 불균등화효소 등의 항산화효소의 발현량이 감소한다는 것이 잘 알려져 있다. 노화에 의한 활성산소종 대사능력의 저하나 병에 의한 활성산소종의 과잉생성에 따라 이들의 산화물 대사가 충분하지 못하여 축적되어 버리면 비특이적으로 지질 등의 세포성분을 노화시키고, 그 장해로부터 세포사가 유도될 수도 있다. 이것이 노화나 알츠하이머 등과 같은 많은 병의 한 원인이 되고 있다.
활성산소가 관여하는 질병으로는, 암, 당뇨병, 아토피성 피부염, 알츠하이머, 망막색소 변성증 등을 들 수 있지만, 사람에 발생하는 병의 90%는 어떤 형태로든 과잉상태의 활성산소가 관여하고 있다고 알려져 있다. 특히, 생체 내의 미토콘드리아에서는 산소의 90% 이상이 대사되며, 세포 내에서 가장 많이 활성산소가 생성된다. 미토콘드리아 내에서 생성되는 활성산소와 항산화계의 균형이 유전병이나 연령으로 인하여 유지되지 않으면, 미토콘드리아로부터 처리되지 않은 활성산소가 새어 나와 세포에 손상을 주어, 노화나 세포자멸(apoptosis)에 의한 세포사가 일어나게 된다.
활성산소를 제거시키는 수단으로는 물을 전기분해하여 산화환원전위를 최대 -200mV 내지 -250mV로 만든 환원수가 개발되어 있고, 또한, 전기분해에 의해 pH를 9 내지 11로 만든 알칼리 이온수가 개발되어 있다(참조: 谷腰欣司저, 「오늘부터 박식시리즈 철저히 쉬운 물의 책」, 초판, 닛칸공업신문사, 2001년 11월, p.100-124). 또한, 고활성 금속미립자, 예를 들어, 백금 클로이드는 활성산소의 일종인 H2O2(과산화수소)를 분해시킨다고 알려져 있다(참조: 특개평 제 10-68008호 공보, [0040]단락). 그러나, 백금 콜로이드가 생체 내에서 초산화물이나 일산화질소의 제거작용을 가지는 것에 관해서 보고된 문헌은 없다.
또한, 고활성 금속미립자의 제조방법에 대해서는 옛날부터 여러종류가 알려져 있다(참조: 특공소 제 57-43125호 공보, 특공소 제 59-120249호 공보 및 특개평 제 9-225317호 공보 등).
발명의 개시
본 발명자들은 생체 내에서 생성되는 활성산소 중 O2 -(초산화음이온) 및 일산화질소를 효율적으로 소실시키고, 생체 내에서 이들의 활성산소의 과잉상태를 해소하기 위한 수단을 제공하기 위하여 예의연구를 수행하였다. 본 발명자들은 전이금속 미분말, 특히 귀금속인 백금 미분말에 착안하여, 이들의 미분말이 세포 내에 침입가능하고, 미토콘드리아의 내부에도 침입가능한 것과 이들의 미분말이 미토콘드리아의 내부에서 초산화음이온 및 일산화질소를 소실하는 능력을 가짐을 확인하였다. 본 발명은 상기의 지견을 기초로하여 완성되었다.
즉, 본 발명에 의하여, 전이금속의 미분말을 포함하는 초산화음이온 분해제가 제공된다. 이 발명의 바람직한 실시태양에 따르면, 전이금속이 귀금속의 미분말인 상기의 초산화물 분해제가 제공된다. 이 분해제는 생체 내에서 초산화음이온을 분해할 수 있다.
다른 관점으로는, 본 발명에 의하여, 전이금속의 미분말을 포함하는 일산화질소 분해제가 제공된다. 이 발명의 바람직한 실시태양에 따르면, 전이금속이 귀금속의 미분말인 상기의 일산화질소 분해제가 제공된다.
이들 발명의 바람직한 실시태양에 따르면, 상기 미분말이 백금 미분말 또는 백금합금의 미분말인 상기 분해제; 전이금속 콜로이드를 포함하는 수성(水性)의 상기 분해제; 1000mL 중에 1mM 이하의 비율로 전이금속 콜로이드를 포함하는 수성의 상기 분해제가 제공된다.
또 다른 관점으로는, 사람을 포함하는 포유류 동물의 생체 내에서 초산화음이온 또는 일산화질소를 소거하는 방법이며, 전이금속의 미분말을 생체에 투여하는 단계를 포함하는 방법이 제공된다. 이 발명의 바람직한 실시태양에 따르면, 전이금속 콜로이드를 포함하는 물을 투여할 수 있다.
발명을 실시하기 위한 최량의 형태
본 발명의 분해제에 있어서 전이금속의 종류는 특별히 한정되지 않으며, 구체적인 금속의 예로는 금, 니켈, 백금, 로듐, 팔라듐, 이리듐, 루테늄, 오스뮴 등의 금속 또는 이들의 합금을 들 수 있다. 전이금속으로는 귀금속인 것이 바람직하다. 귀금속의 종류는 특별히 한정되지 않으며, 금, 루테늄, 로듐, 팔라듐, 오스뮴, 이리듐 또는 백금 중 어느 하나를 이용하여도 좋지만, 바람직한 귀금속은 루테늄, 로듐, 팔라듐 또는 백금이다. 특히 바람직한 귀금속은 백금이다. 귀금속의 미립자는 2종 이상의 귀금속을 포함하여도 좋다. 또한, 적어도 1종의 귀금속을 포함하는 합금의 미립자 또는 1종 또는 2종 이상의 귀금속의 미립자와 귀금속 이외의 1종 또는 2종 이상의 금속의 미립자를 포함하는 혼합물을 사용할 수도 있다. 예를 들어, 금 및 백금으로 된 합금 등을 사용하여도 무방하다. 이들 중 바람직한 것으로는 백금 또는 백금을 포함하는 합금이며, 특히 바람직한 것은 백금이다.
귀금속의 미립자로서는 비표면적(比表面的)이 크고, 표면 반응성이 우수한 콜로이드 상태를 형성할 수 있는 미립자가 바람직하다. 미립자의 입경은 특별히 한정되지 않지만, 50nm 이하의 평균입경을 가지는 미립자를 사용할 수 있으며, 바람직하게는 평균입경이 20nm 이하, 보다 바람직하게는 평균입경이 10nm 이하, 보다 더 바람직하게는 평균입경이 1 내지 6nm 정도의 미립자를 사용할 수 있다. 특히 미토콘드리아 내부로의 침입을 위해서는 평균입경이 1 내지 6nm 정도인 것이 바람직하다. 또한, 아주 작은 미립자를 사용할 수도 있으며, 생체 내로의 침투를 높이기 위해서 바람직하다. 수성 매체로서는 물 이외에, 생체에 대한 독성이 낮고 물과 임의의 비율로 섞이는 유기용매, 예를 들어, 에탄올, 에틸렌글리콜 등을 이용할 수 있다. 바람직하게는 수성 매체로 물을 이용할 수 있다.
귀금속 미립자의 제조방법은 다수 알려져 있으며(참조: 특공소 제 57-43125호 공보, 특공소 제 59-120249호 공보 및 특개평 제 9-225317호 공보, 특개평 제 10-176207호 공보, 특개 제 2001-79382호 공보, 특개 제 2001-122723호 공보 등), 당업자는 이들의 방법을 참조함에 따라 미립자를 용이하게 제조할 수 있다. 예를 들어, 귀금속 미립자의 제조방법으로, 침전법 또는 금속염 환원반응법이라고 불리는 화학적 방법이나 연소법이라고 불리는 물리적 방법 등을 이용할 수 있다. 본 발명의 분해제로서는 어느 하나의 방법으로 제조된 미립자를 이용하여도 무방하지만, 제조의 용이성과 품질면으로 볼때, 금속염 환원반응법으로 제조된 미립자를 이용하는 것이 바람직하다.
금속염 환원반응법으로서는 예를 들어, 수용성 또는 유기용매 가용성의 귀금속염 또는 귀금속 착체의 수용액 또는 유기용매용액을 제조하고, 이 용액에 수용성 고분자를 가한 후, 용액의 pH를 9 내지 11로 조절하고, 불활성 조건하에서 가열환류함으로써 환원하여 금속미립자를 얻을 수 있다. 귀금속의 수용성 또는 유기용매 가용성의 염의 종류는 특별히 한정되지 않지만, 예를 들어, 초산염, 염화물, 황산염, 질산염, 술폰산염 또는 인산염 등을 이용할 수 있으며, 이들의 착체를 이용하여도 무방하다.
금속염 환원반응법에 이용하는 수용성 고분자의 종류는 특별히 한정되지 않지만, 예를 들어, 폴리비닐프롤리돈, 폴리비닐알콜, 폴리아크릴산, 시클로덱스트린, 아미노펙틴 또는 메틸셀룰로스 등을 이용할 수 있으며, 이들을 2종 이상 조합하여 사용하여도 무방하다. 바람직하게는 폴리비닐피롤리돈을 사용할 수 있으며, 보다 바람직하게는 폴리(1-비닐-2-피롤리돈)을 사용할 수 있다. 또한, 수용성 고분자의 대체 또는 수용성 고분자와 함께 각종의 계면활성제, 예를 들어 음이온성, 비이온성 또는 지용성 등의 계면활성제를 사용하는 것도 가능하다. 알콜을 이용하여 환원하는 경우에는 에틸알콜, n-프로필알콜, n-부틸알콜, n-아밀알콜 또는 에틸렌글리콜 등이 이용된다. 그러나, 귀금속 미립자의 제조방법은 상기에 설명한 방법에 한정되지 않는다.
이상의 방법으로 제조된 금속 미분말은 통상, 사용한 용매를 매체로하는 콜로이드 상태로 수득되므로, 이를 그대로 본 발명의 초산화음이온 분해제 또는 일산화질소 분해제로 이용할 수 있다. 사용한 유기용매를 제거하는 경우에는 가열에 의해 유기용제를 제거하고, 본 발명의 분해제를 금속 미분말로 제조할 수도 있다. 가열건조에 의해 수득된 금속 미분말은 초산화음이온 분해제 또는 일산화질소 분해제로의 특성을 소실하지 않는다.
본 발명의 분해제는 환원수의 형태로 제조할 수 있다. 환원수라 함은 생체 내에서 산화물질을 환원하는 작용을 가지는 물을 의미한다. 본 발명에 따라, 첨가한 분해제의 양에 대하여 초산화음이온 및/또는 일산화질소를 분해하는 환원수를 제조할 수 있지만, 예를 들어, 물 1,000mL당 0.033mM 정도의 분해제를 첨가한 환원수에서도 충분한 환원효과, 즉 초산화음이온 및/또는 일산화질소의 분해효과를 얻을 수 있다. 예를 들어, 본 발명의 환원수는 상기 분해제를 1mM 이하의 비율로 포함하는 것이 바람직하다. 상기 비율로 포함하는 환원수를 사람을 포함하는 포유류 동물에 투여함에 따라, 생체 내에서의 과잉의 초산화음이온 및/또는 일산화질소의 대부분은 소실된다.
본 발명의 분해제의 바람직한 실시태양으로는 나노미터(nm) 단위의 입경을 가지는 금속 미분말을 포함하고 있으며, 전기 금속 미분말은 생체 내에 투여된 후에 세포 내로 침투되고, 또한, 미토콘드리아 내에 침투하여 미토콘드리아 내에서 생성되는 초산화음이온 또는 일산화질소를 소실시킨다. 따라서, 본 발명의 분해제는 활성산소에 기인한다고 여겨지는 전기 질병, 특히 근위축성측색경화증(FALS) 등의 예방 또는 치료에 유효하다고 기대된다. 또한, 환원수의 형태로 제공되는 본 발명의 분해제는 건강식품으로서의 음료수 또는 스포츠 드링크로 이용할 수 있으며, 그 자체를 의약 또는 화장료로서 사용할 수 있는 것 외에, 건강식품의 제조나 의약 또는 화장료 등의 제조에 이용할 수도 있다.
또한, 본 발명의 일산화질소 분해제를 예를 들어, 담배 등의 필터에 배합함으로써 담배의 연기에 포함된 일산화질소를 효율적으로 분해할 수 있다. 예를 들어, 담배의 필터 내에 활성탄 등과 함께 넣거나 또는 활성탄에 대체하여 본 발명의 분해제의 미립자를 고체상태로 배합하여 일산화질소를 분해할 수 있으며, 또한, 수성 콜로이드 상태의 본 발명의 분해제를 물 파이프 내에 충진해 두어, 담배의 연기를 전기 물 파이프 내로 유도함으로써, 담배의 연기에 포함된 일산화질소를 효율적으로 제거할 수 있다.
도 1은 본 발명의 일산화질소 분해제의 작용을 나타낸 그래프이다.
이하, 실시예에 의하여 본 발명을 보다 구체적으로 설명하지만, 본 발명의 범위가 하기의 실시예에 한정되는 것은 아니다.
실시예 1
아릴 냉각관과 삼방 콕크를 접속한 100mL 2구 환저 플라스크에 와코순약 주식회사 시약인 폴리(1-비닐-2-피롤리돈)를 0.1467g 넣고, 증류수 23mL로 용해시켜, 10분간 교반칩으로 교반한 후, 염화백금산 결정(H2PtCl6ㆍ6H2O: 와코순약주식회사 시약)을 증류수에 용해하여 농도를 1.66 x 10-2M로 조정한 염화백금산 수용액을 2mL 가하고, 다시금 30분간 교반하였다. 반응계 내를 질소치환하여 특급에탄올 25ml 가하여 질소분위기 하에서 온도 100℃로 2시간 환류하였다. 반응액의 UV를 측정하여, 백금이온피크가 소실되고, 금속고체 특유의 산란에 의한 피크의 포화를 확인함으로써 환원반응이 완료한 것을 확인하였다. 용매를 증발 농축기로 제거한 후, 다시금 12시간에 걸쳐 동결건조하여 백금미분말(본 발명의 분해제)을 수득하였다.
수득된 분해제를 미리 pH 7.8로 조정한 농도 0.1M의 인산나트륨 완충액에 용해하여 0.66mM, 0.495mM, 0.330mM, 0.165mM, 0.083mM, 0.033mM의 농도로 콜로이드 상태의 분해제를 포함하는 분산액을 수득하였다. 현미경 하에서 관찰한 바, 전기 백금미립자의 입경은 6nm 이하였다.
실시예 2
히포크산틴/크산틴옥시다제의 조합 및 페나디엔메트설페이트/NADH(환원형 니코틴아미드아데닌디뉴클레오티드)의 조합에서 각각 발생하는 O2 -(초산화음이온)를 이용하여, 수득된 분해제의 초산화음이온 분해능을 아래와 같이 측정하였다.
(A) 히포크산틴/크산틴옥시다제 계
시료용기에 농도 8.8M의 DMPO(5,5-디메틸-1-피롤린-N-옥사이드: 라보텍사 제품 스핀트랩제) 20㎕, 농도 1mM의 히포크산틴(시그마사 제품) 50㎕, MilliQ(정제수: 밀리포아사 제품) 50㎕, 농도가 다른 상기의 각 시료 5종의 50㎕를 각각 순차 가하여 혼합한 후, 농도 0.04U/mL의 크산틴 옥시다제(로슈사 제품) 50㎕를 가하고, 45초 후에 ESR 측정장치(일본전자주식회사 제품 JES-FA200)를 사용하여, ESR 스펙트럼을 측정하였다. 표준물질(망간)과의 대비로 O2 -(초산화음이온)양을 측정하였으며, 그 결과를 하기 표 1에 나타내었다. 표 1에서, 괄호안의 수치는 농도 0일 때의 값을 100으로 한 상대수치이다.

상기 시료 중 4종(농도 0.033mM, 0.083mM, 0.165mM 및 0.330mM)에 대하여, 시료용기에 농도 8.8M의 DMPO 20㎕, NADH(후나코시사 제품), 페나딘메트설페이트(와코순약사 제품), 상기 시료 50㎕를 순차 가하여 혼합한 후, 1분간에 상기와 동일하게 하여, ESR 스펙트럼을 측정한 결과를 하기 표 2에 나타내었다. 표 2에서, 괄호 내의 수치는 농도 0일 때의 값을 100으로 한 상대값이다.

비교예 1
폴리(1-비닐-2-피롤리돈) 또는 백금착체인 시스플라틴(PtCl2(NH3)2)을 사용하여, 폴리(1-비닐-2-피롤리돈) 또는 백금농도를 실시예와 동일하게 하여, O2 -(초산화음이온) 양을 측정하였지만, 공시험(백금농도=0)과의 사이에 차이가 확인되지 않았다.

분석용 키트로서 NO2/NO3 Assay Kit-C II(주식회사 도진화학연구소)를 사용하여, 백금 콜로이드에 의한 NO의 소거능을 검토하였다. 전기 키트는 NO가 가수분해하여 발생된 NO2를 측정한다. 마이크로 플레이트 리더(BIORAD사 제품 Model 550)를 사용하여 검출파장을 570nm로 설정하고, 각 시료에 대하여 3회 측정하였다. 마이크로 플레이트로는 96웰 마이크로 플레이트를 사용하였다. 또한, NO 도너로서는 NOC7(주식회사 도진화학연구소)을 이용하였다. 분석은 기본적으로 키트에 부속되 어 있는 시험서에 약간 수정을 가하여 실시하였다. 마이크로 플레이트의 각 웰에 시료 8㎕를 가하고, 완충용액 84㎕와 NOC7 8㎕를 혼화하여, 30분간 방치하였다. 시약 A 50㎕를 혼화하여 5분간 방치하고, 또한, 시약 B 50㎕를 혼화하여 10분간 방치한 후, 마이크로 플레이트 리더로 검출파장 570nm로 검출하였다. NO로부터 발생하는 NO2와 백금 나노 콜로이드가 포함되면 조금 흡광도가 상승하지만, NO2의 분해 등은 일어나지 않음이 밝혀졌다. 각 시료에 대하여, 각각 NO 소거능을 분석하였고, 그 결과를 도 1에 도시하였다. 이 결과로부터 백금 나노 콜로이드에는 NO 소거능이 있음이 밝혀졌다.
본 발명의 초산화음이온 분해제 및 일산화질소 분해제는 생체에 투여함으로써, 생체 내의 과잉의 초산화음이온 및/또는 일산화질소를 분해할 수 있다.
Claims (5)
- 전이금속의 미분말을 포함하는 초산화음이온 또는 일산화질소의 분해제.
- 제 1항에 있어서,전이금속은 귀금속(noble metal)인 것을 특징으로 하는분해제.
- 제 1항에 있어서,백금 미분말 또는 백금합금의 미분말을 포함하는 것을 특징으로 하는분해제.
- 제 1항 내지 제 3항 중 어느 한 항에 있어서,전이금속 콜로이드를 포함하는 수성인 것을 특징으로 하는분해제.
- 제 4항에 있어서,1,000mL당 1mM 이하의 비율로 전이금속을 포함하는 것을 특징으로 하 는분해제.
