Abstract
본 발명은 최소 하나의 프롤린 잔기를 포함하는 2-8 아미노산 길이를 갖는 펩타이드가 풍부한 DPP-IV 억제 활성을 갖는 단백질 가수분해물, 이러한 단백질 기수분해물 또는 이의 혼합물에서 유래된 펩타이드 유도체 및 특히 비만, 제2형 당뇨병 및 자가면역 질환의 그룹으로부터 선택되는 DPP-IV 매개 컨디션의 예방 및/또는 치료를 위한 의약, 식품 보조제, 음료 또는 식품 제조용으로 사용되는 이러한 단백질 가수분해물 또는 단리된 펩타이드 또는 혼합물의 용도를 개시한다.

Description
본 발명은 단리된 펩타이드와 같은 특정 펩타이드가 풍부한 단백질 가수분해물, 및 이러한 단백질 가수분해물 또는 단리된 펩타이드의 DPP-IV 매개 컨디션의 예방 및/또는 치료를 위한 의약, 식품 보조제, 음료 또는 식품 제조용 용도에 관한 것이다.
디펩티딜 펩티다아제 IV(DPP-IV)는 N-말단 디펩티다아제 활성을 함유하는 다작용 막전위 글리코단백질이다. 이는 간, 신장, 소장, 침샘, 혈세포 및 혈장과 같은 여러 조직의 대부분의 포유류 세포에 존재한다. 그러나 DPP-IV의 생리학적 역할에 대해 거의 알려진 바 없다.
DPP-IV는 면역, 염증 및 내분비 작용을 포함하는 세포성 프로세스에 연루되어 있다. 시험관내에서 DPP-IV는 예를 들어, 글루카곤성 펩타이드 1(GLP-1)과 같은 다수의 호르몬 및 케모카인을 분할하는 것으로 나타났다.

그러나, GLP-1은 혈장에서 신속히 분해되어 약 1-2분의 매우 짧은 반감기를 갖는다. GLP-1의 분해를 주로 담당하는 효소는 DPP-IV이다. DPP-IV의 억제는 따라서 GLP-1의 순환 반감기를 연장시켜 GLP-1 수준이 치료제로서 작용할 수 있도록 증가된다.
DPP-IV는 또한 T 세포 활성화 및 성장에 관여하는 것으로 알려져 있다. 면역 시스템에서, DPP-IV는 T 세포의 표면상에서 일차적으로 발현된다. 단백질, 특히 우유 단백질은 일반적으로 생물학적으로 활성적인 범위의 펩타이드의 전구체로 알려져 있다. 단백질이 생물학적으로 활성적인 분자의 전구체라는 사실은 어떠한 상기 DPP-IV 매개 컨디션을 보조할 수 있는 식품과 같이 특히 기능성 식품의 개발에 매력적이다. 식품 단백질 가수분해물은 자주 사용되는 식품 성분이며 천연 기원이어서 비만, 제2형 당뇨병 및 면역 질환과 같은 DPP-IV 매개 컨디션을 예방 또는 치료하는 것과 같이 DPP-IV의 억제가 필요한 경우에 기능적 효과를 얻기위해 합성 성분이 요구되지 않는다.
따라서, 상술한 DPP-IV 매개 컨디션의 예방 및 치료를 보조할 수 있도록 DPP-IV 활성을 억제하는 단백질 가수분해물을 제공하는 것이 요구된다. 예를 들어, 우유 단백질 가수분해물과 같은 단백질 가수분해물은 DPP-IV 억제에 사용될 수 있다. 그러나, 이러한 가수분해물내에서 이러한 억제를 담당하는 팩터들은 아직 알려지지 않았다. 가수분해된 단백질 시료는 매우 복잡하며 최대 수백개의 다른 분자들을 함유할 수 있으며, 이는 이러한 시료에서 바이오액티브 화합물을 밝혀내기 어렵게 한다.
본 발명자들은 DPP-IV 활성을 억제하는 단백질 가수분해물로부터 바이오액티브 펩타이드를 확인하였다. 이러한 바이오액티브 펩타이드 또는 이러한 바이오액티브 펩타이드가 풍부한 단백질 가수분해물은 비만, 제2형 당뇨병 및 면역 질환과 같은 DPP-IV 매개 컨디션의 자연적 예방 및 치료에 유용하게 사용될 수 있다. 약 40 단백질 가수분해물은 HPLC에 의한 조질의 추출물의 분리를 가능케 하는 방법인 온라인 액체 크로마토그래피(LC)-생화학 검출(BCD)-매스 스펙트로미터(MS)에 적용되었으며, 이후 바이오액티브 화합물의 존재를 온-라인 생화학 어세이를 통해 검출되었다. 온-라인 분획 및 특성화 방법은 Van Elswijk et al.(J. Chromatography A(2003) 1020:45-58)에 의해 광범위하게 기술되어 있다.
따라서, 제 1 견지로 본 발명은 DPP-IV 억제 활성을 갖는 단백질 가수분해물에 관한 것이며, 상기 가수분해물은 적어도 하나의 프롤린 잔기를 포함하는 2-8 아미노산의 길이를 갖는 펩타이드가 풍부하다. 이러한 펩타이드는 시험관내에서 DPP-IV 활성을 억제할 수 있으며 비만, 제2형 당뇨병 및 면역 질환과 같은 DPP-IV 매개 컨디션의 자연적 예방 및/또는 치료에 사용될 수 있는 것으로 발견되었다.
DPP-IV는 최종 N-말단 잔기로서 리신(K) 및 아르기닌(R)에 대한 선호와 함께 제 2 N-말단 잔기(끝에서 두번째 N-말단 잔기)로서 프롤린(P)을 갖는 기질에 대한 선호를 나타내는 것이 잘 알려져 있다(Davy et al. Plant Physiol.(2000) 122:425-432).
약 40 단백질 가수분해물로부터, 본 발명자들은 DPP-IV 활성을 억제할 수 있는 약 21 펩타이드 서열을 확인하였다. 이들 펩타이드는 3-7 아미노산으로 다양한 길이를 가지며 소수성 특성으로 특히 상기 펩타이드 서열내에 프롤린(P)의 존재로 일반적으로 특성화된다. 프롤린은 또한 C-말단 잔기 또는 끝에서 두번째 C-말단 잔기로 발견된다. 프롤린 잔기는 대부분 루신, 발린 또는 페닐알라닌(F)에 의해 플랭킹되며(flanked by), 또한 보다 적은 정도로 글루타민(Q), 알라닌, 히스티딘(H), 이소루신(I), 글리신(G), 메티오닌(M), 및 티로신(Y)에 의해 플랭킹될 수 있다. 21 펩타이드 서열 외에, 단지 하나의 억제 서열이 확인되었으며, 여기서 프롤린 잔기는 하전된 잔기가 히스티딘인 하전된 잔기에 의해 플랭킹되었다. 마찬가지로, 극성 아미노산 세린, 트레오닌, 아스파라긴 및 글루타민 중, 글루타민만이 DPP-IV를 억제하는 것으로 발견된 펩타이드에서 플랭킹(flanking) 잔기로서 확인되었다. 가장 강력한 DPP-IV 억제 활성은 프롤린 잔기가 제 2 N-말단 위치상에 위치하며, 이 프롤린 잔기가 발린, 이소루신 및/또는 루신의 어느 한 측면으로 플랭킹되는, 가장 바람직하게는 루신의 한쪽 또는 양쪽 모든 측면에 플랭킹되는 펩타이드에 대해서 발견되었다.
본 명세서에 사용된 "단백질 가수분해물"은 최소 정도의 가수분해도로, 즉, 5%, 바람직하게 10%, 보다 바람직하게 20%, 가장 바람직하게 30%의 펩타이드 결합 총량의 가수분해 펩타이드 퍼센트로 하나 이상의 단백질 가수분해로부터 유래된 펩타이드의 혼합물을 칭한다. 바람직한 가수분해도는 5-70%, 보다 바람직하게 10-50%, 가장 바람직하게 20-40%이다. 가수분해물은 2-8 아미노산의 펩타이드를 20-100wt%, 보다 바람직하게 25-75wt%, 가장 바람직하게 30-60wt% 함유한다. 특히 바람직한 가수분해물은 3-7 아미노산의 펩타이드를 20-80wt%, 가장 바람직하게 30-60wt% 함유하는 것이다. 바람직하게, 500 Da 이상의 분자량을 갖는 펩타이드 중량 퍼센트는 75%미만이며, 바람직하게 70%미만이며, 그리고 1000 Da이상의 분자량을 갖는 펩타이드의 중량 퍼센트는 50%미만, 바람직하게 35%미만이다.
상기 단백질은 하나의 단백질 공급원으로부터 유래될 수 있으며 그 이상의 단백질 공급원으로부터 유래될 수 있다. 이러한 단백질 공급원의 예는 미생물(효모, 박테리아, 균류), 식물(예, 콩, 면, 옥수수, 밀), 동물, 및 우유, 혈액, 고기, 달걀 및 젤라틴과 같은 동물 유래 단백질 공급원이다. 따라서, 하나 이상의 단백질은 예를 들어, 미생물의 단백질, 식물 단백질, 고기 조각, 생선, 갑각류 또는 연체동물로부터 유래된 단백질, 우유 단백질 및 달걀 단백질과 같은 동물 단백질일 수 있다.
단백질 가수분해는 당해 기술분야에 알려진 어느 수단에 의해 수행될 수 있다. 이의 예는 화학적 가수분해 또는 효소적 가수분해 방법을 포함한다. 화학적 가수분해 방법의 비제한적 예는 당해 기술분야에 잘 알려져 있으며, 예를 들어 시아노겐 브로마이드를 이용한 가수분해, 예를 들어 염산을 이용한 산 가수분해 또는 하나 이상의 단백질을 포함하는 하나 이상의 단백질 공급원의 발효 수단에 의한 가수분해를 포함한다. 효소적 가수분해 방법의 비제한적 예는 당해 기술분야에 잘 알려져 있으며 정제된 효소 제조물 또는 조 효소 제조물을 이용한 가수분해를 포함한다. 사용되는 효소 제조물은 엔도- 또는 엑소- 펩티다아제, 프로테아제, 또는 이의 혼합물을 포함할 수 있으며, 이의 예는 트립신, 키모트립신 A, B 및 C, 펩신, 레닌, 미생물성 알카리 프로테아제, 파파인, 피신, 브로멜라인, 카텝신 B, 콜라게나아제, 미생물성 중성 프로테아제, 카르복시펩티다아제 A, B 및 C, 카로시나아제, 안세리나아제, 스타필로코커스 아우레우스(Staphylococcus aureus)의 V8 프로테아제 및 당해 기술분야에 잘 알려진 다수의 다른 것들을 포함한다. 또한 이러한 프로테아제의 혼합물이 사용될 수 있다. 또한 예를 들어, Alkalase, Chymotrypsine 800s, Neutrase, flavourzyme(모두 Novo Nordisk(덴마크)로부터 이용가능), Protex 6.0L, Peptidase FP(모두 Genencor(미국)로부터 이용가능), Corolase L10(Rohm, 독일), Pepsin(Merck, 독일), 파파인, 판크레아틴, 프로리더 N 및 Protease N(Amano, 일본)과 같은 상업적으로 이용가능한 효소 제조물 또는 이의 혼합물이 사용될 수 있다. 재조합 DNA 기술 수단에 의해 제조된 효소도 이용될 수 있다.
단백질 가수분해물은 바람직하게 N-말단 끝에서두번째의(penultimate) 프롤린 잔기를 갖는 펩타이드를 형성하는 효소를 이용한 단백질 분해에 의해 획득된다. 이러한 효소는 예를 들어, Cys 엔도프로테아제 EP-A 및 EP-B와 같은 하나 이상의 시스테인 엔도프로테아제일 수 있으며 또한 상술한 바와 같이 다른 어느 적절한 효소일 수 있다.
DPP-IV 억제 활성을 갖는 단백질 가수분해물은 만일 하기에 나타낸 것과 같이 측정되는 DPP-IV 억제 활성이 출발 단백질 가수분해물에 비해 증가되는 경우 "적어도 하나의 프롤린 잔기를 포함하는 2-8 아미노산의 길이를 갖는 펩타이드가 풍부한" 것으로 간주되며, 그리고/또는 이러한 펩타이드의 존재는 예를 들어 Van Elswijk 등(상기 참조)에 기재된 바와 같은 질량 스펙트로미터와 같은 일반적인 단백질 또는 펩타이드 분획 및 분석 기술 수단 뿐만 아니라 HPLC 또는 예를 들어, 시퀀셜 에드만 디그래데이션(sequential Edman degradation) 또는 ELISA와 같은 특정 펩타이드에 대해 표적하는 특정 친화도 어세이와 함께 사용하는 다른 분획 기술과 같은 다른 기술에 의해 입증될 수 있다. 단백질 가수분해물은 바람직하게 우유 단백질 가수분해물, 보다 바람직하게 카세인 단백질 가수분해물은 DPP-IV 억제 활성을 (더) 분리하도록 추출, 침전, 여과, 한외여과, 마이크로여과 또는 통상적인 컬럼 크로마토그래피(바람직하게 이온 교환 또는 친화 크로마토그래피), 또는 상기 기술의 어느 조합의 수단에 의해 분획화될 수 있다. 펩타이드의 혼합물 또는 단일 펩타이드를 포함하는 분획은 출발 단백질 가수분해물에 비해 DPP-IV에 대해 증가된 억제 영향을 갖는 것으로 확인될 수 있다. 이러한 펩타이드의 혼합물 또는 단일 펩타이드가 또한 본 발명에 포함된다. 이러한 펩타이드는 적절한 숙주에서 DNA 암호화의 발현과 같은 재조합 DNA 기술 수단 또는 화학적 합성에 의해 제조될 수 있는 것으로 또한 예상된다.
상기 단백질 가수분해물은 어느 단백질 가수분해물일 수 있으며, 특히 식품 단백질 가수분해물일 수 있으며, 이러한 가수분해물은 식품 등급이며 비교적 쉽게 얻을 수 있는 것으로 간주된다.
본 명세서에 사용된 가수분해물은 실시예 1의 DPP-IV 억제 어세이에서 IC50(즉, DPP-IV 활성의 50%를 억제하는 억제제(특히 단백질 가수분해물)의 농도)가 최대 1000㎍/ml, 바람직하게 800㎍/ml, 보다 바람직하게 600㎍/ml, 보다 바람직하게 400㎍/ml, 그리고 가장 바람직하게 최대 300㎍/ml으로 나타나는 경우 "DPP-IV 억제 활성"을 나타낸다.
본 발명의 바람직한 구현으로, 프롤린이 우유 단백질 가수분해물에 풍부하며 본 발명에서 확인된 여러 펩타이드 서열이 우유 단백질에서 발견되고 쉽게 얻을 수 있으므로 상기 단백질 가수분해물은 우유 단백질 가수분해물이다. 우유 단백질 가수분해물은 예상되는 DPP-IV 억제 활성을 제공하는 것으로 발견되었다. 우유는 어느 포유류로부터 기원할 수 있으며, 특히 소, 양 또는 염소, 바람직하게는 소(보바인 밀크)로부터 기원할 수 있다.
본 발명에 따른 바람직한 구현으로, 상기 단백질 가수분해물은 카세인 가수분해물이다. 본 발명에서 확인된 다수의 펩타이드 서열은 하나 이상의 카세인 서열에서 발견되며, 따라서 카세인으로부터 제조된 가수분해물로부터 쉽게 얻어질 수 있는 것으로 발견되었다. 다른 구현으로, 단백질 가수분해물은 예를 들어 β-락토글로불린으로부터 얻어지는 유장(whey) 단백질 가수분해물이다.
본 발명의 다른 바람직한 구현으로, 상기 펩타이드는 3-7 아미노산의 길이를 갖는다. 가장 강력한 DPP-IV 억제제는 이러한 길이를 갖는 것으로 발견되었다.

제 3 견지로, 본 발명은 본 발명에 따른 단백질 가수분해물 또는 본 발명에 따른 단리된 펩타이드 또는 혼합물의 DPP-IV 매개 컨디션의 예방 및/또는 치료를 위한 의약, 식품 보조제, 음료 또는 식품 제조용 용도에 관한 것이다.
본 명세서에 사용된 용어 "예방"은 증상이 아직 관찰되지 않으나 DPP-IV 매개 컨디션의 발생을 방지하는 것을 칭한다. 이러한 경우에 하나 이상의 단백질 가수분해물이 유해한 DPP-IV 매개 컨디션이 발생되는 것을 예방하는데 사용될 수 있으며, 따라서 이를 요구하는 특정 대상자에서 어느 환자의 건강을 개선 혹은 안정화하는데 사용될 수 있다.

다른 구현에서 본 발명에 따른 상기 단백질 가수분해물 또는 단리된 펩타이드는 혈당의 수준을 낮추기 위한 의약, 식품 보충제, 음료 또는 식품의 제조를 위해 사용된다. 혈당 수준은 가수분해물의 섭취에 의해 감소되어 향상된 글루코스 제어의 결과를 보이는 것으로 나타났으며, 이는 특히 당뇨병에 유익하다.

"Remington's Pharmaceutical Sciences Handbook", Mack Pub. Co., N.Y. U.S.A.,에 설명된 바와 같은 잘 알려진 방법 및 부형제를 이용하여 조제될 수 있는 상기 배합물의 예는 구강 투여를 위한 타블렛, 캡슐, 시럽 등이며, 반면 비경구 투여를 위한 적절한 형태는 수용가능한 액체, 이식제 등 내의 멸균 용액 또는 서스펜션이다.
약량학은 치료될 병리학적 상태의 유형 및 심각성, 환자의 체중 및 성별 등의 일부 요소에 따르며, 숙련자에 의해 쉽게 결정될 수 있을 것이다. 바람직하게는 본 발명의 상기 가수분해물은 활성 펩타이드의 농도에 따라 2mg 내지 10g의 수준으로 투여된다. 가수분해물의 바람직한 투여량 값은 일일 10 mg 내지 5g, 더욱 바람직하게는 50 mg 내지 1g이다. 활성 분획, 특히 2-8 아미노산을 갖는 펩타이드 분획으로 투여의 바람직한 값은 개체당 일일 1mg 내지 1g의 펩타이드 혼합물이며, 이러한 분획에 대한 보다 바람직한 투여량 값은 10 내지 500 mg/일이다.

하나 또는 그 이상의 단백질 가수분해물은 바람직하게는 상술한 효소적 가수분해에 의해 획득되며, 효소적 기수분해가 적절한 정도의 가수분해를 제공하고 편리하게 수행되기 때문이다. 더욱이, 효소적 가수분해에 사용된 상기 효소는 단백질 가수분해물로부터 예를 들어 겔 여과 크로마토그래피와 같은 단순한 컬럼 크로마토그래피에 의해 쉽게 분리되거나 열, 산, 염기, 또는 억제제의 첨가에 의해 불활성화될 수 있다.

활성의 시험관내 측정
DPP-IV 활성은 DPP-IV 기질로 Gly-Pro-p-니트로아닐리드(Sigma G-0513)를 사용하여 385nm에서 흡수의 증가를 측정하여 결정될 수 있다. DPP-IV 활성의 감소는 억제의 척도이다.
13.152mg Gly-Pro-p-니트로아닐리드(기질; Sigma G-0513)를 1ml의 pH 8.0 트리스 버퍼에 용해하고, DPP-IV(sigma D-7052)를 pH 8.0 트리스 버퍼로 1.1 Unit/ml로 희석하였다. 상기 기질을 pH 8.0 트리스 버퍼로 50배 희석한다. 상기 시료는 단백질 가수분해물을 pH 8.0 트리스 버퍼에서 1wt.% 단백질 용액으로 희석하여 제조한다. 상기 시료는 그 후 연속적으로 희석되어 시료 농도의 범위를 획득한다. 50 μl의 다른 연속 희석 시료 및 50 μl의 희석된 기질은 그 후 96 웰 판(microtiter plate)의 웰(well)에 피펫으로 옮겨진다. 후속적으로, 100μl의 희석된 효소가 96 웰 판에 피펫으로 옮겨진다. 그 후, 385nm에서 흡수의 증가가 결정되고 다양한 단백질 가수분해물의 다른 농도에서의 DPP-IV 활성이 결정되며, 그로부터 IC50(즉, DPP-IV 활성의 50%를 억제하는 억제제의 농도(특히 단백질 가수분해물))을 얻을 수 있다. 모든 가수분해물은 5% 이상의 가수분해정도(DH)를 갖는다. 대조군으로서, 0%의 가수분해정도(DH)를 갖는 가수분해되지 않은 두 개의 단백질(DMV International NL로부터의 나트륨 카제인염 및 Davisco Foods, USA로부터의 Bipro)이 실험되었다. 상기 가수분해정도는 o-프탈디알데히드 방법을 이용하여 결정되었으며, 이는 당해 기술분야에 잘 알려져 있다. 따라서, 많은 단백질 가수분해물, 특히 카세인 가수분해물이 290-1000 μg/ml범위의 IC50을 갖지만, 가수분해되지 않은 단백질은 효소를 억제하지 않는 것으로 결정되었다.

가수분해물 1:
1000 그램의 카세인(DMV international, NL)을 7.2 리터의 물에 용해시킨다. pH 및 온도를 8.8 및 55 ℃로 각각 맞추며, 그 후 부피는 8 리터로 증가하였다. 40 그램의 알칼라제(Alkalase)(Novo Nordisk, DK), 4 그램의 프로테아제 N(Amano, Japan) 및 4 그램의 플라보자임(Flavourzyme)(Novo Nordisk)을 상기 혼합물에 첨가하였다. 18 시간의 인큐베이션 후 상기 반응은 90℃에서 10분간의 열 불활성화에 의해 정지되었다. 50℃로 냉각한 후 상기 혼합물을 3kDa 한계의 여과 막으로 통과시키고 투과액을 건조시킨다.
가수분해물 2:
1400 그램의 카세인(DMV international, NL)을 7.2 리터의 물에 용해시킨다. pH 및 온도를 각각 8.3 및 50 ℃로 맞추고, 그 후 부피는 8 리터로 증가하였다. 1.5 그램의 트립신 PTN 6.0(Novo Nordisk, DK)를 200ml의 에탄올과 함께 상기 혼합물에 첨가하였다. 상기 혼합물을 그 후 2시간 동안 지속적인 교반 하에서 인큐베이 션한다. 9 그램의 Corolase LAP F(Rohm, DE)를 100ml의 에탄올과 함께 첨가한다. 21 시간의 인큐베이션을 더한 후 상기 반응은 90℃에서 10분간의 열 불활성화에 의해 정지된다. 상기 혼합물은 후속적으로 50℃로 냉각되고 건조된다.
가수분해물 3:
1000 그램의 유장단백질(Whey protein) 농축액(WPC80; Arla, DK)을 7.2 리터의 물에 용해시킨다. pH 및 온도를 각각 8.3 및 60 ℃로 맞추고, 그 후 부피는 8 리터로 증가하였다. 12 그램의 프로테아제 N(Amano, Japan)을 250ml의 에탄올과 함께 첨가하였다. 상기 혼합물을 그 후 18시간 동안 지속적인 교반 하에서 인큐베이션한다. 9 그램의 Corolase LAP F(Rohm, DE)를 100ml의 에탄올과 함께 첨가한다. 상기 반응은 90℃에서 10분간 열을 가하여 정지된다. 상기 결과 혼합물은 50℃로 냉각되고 3kDa 여과기를 통과하여 건조된다.
가수분해물 2의 분석:
상기 건조된 가수분해물 2를 물에 재용해하고 3kDa 여과 막을 통해 나노여과(nanofiltration)한다. 상기 잔존물 및 여과물은 DPP-IV 억제 효능 및 양의 균형(잔존물 & 여과액 대 총 가수분해물)에 대해 실험되었다. DPP-IV 억제는 실시예 1에서 설명된 바와 같이 수행된 반면 양의 균형에 대해서는 o-프탈디알데히드 방법이 사용되었다. 요약하면, 이 방법은 단백질 및 펩타이드에 존재하는 일차 아민기를 ortho-프탈디알데히드로 수정하는 것에 기초한다. 그 후 상기 시료를 0.1 보랙 스(borax), 1%(w/v) 나트륨 도데실술페이트, pH 9.3에 용해한다. 혼합 후, 상기 ortho-프탈디알데히드 시약을 첨가한 후 10 분간 실온에서 인큐베이션 한다. 흡수성이 3340nm에서 측정되었다. 상기 양의 균형은 잔존물과 투과액의 흡수성을 가수분해물 내의 단백질 총량과 연관시켜 결정된다. 표 1은 출발 가수분해물과 비교할 때 투과액의 DPP-IV 활성이 두 배 증가함을 나타낸다.

실시예 3: 단백질 가수분해물 내 바이오액티브 화합물의 추가 검증
실시예 2에 따라 제조된 다양한 단백질 가수분해물이 DPP-IV 억제 활성에 대해 더욱 조사되었다. 가수분해된 단백질 시료는 Van Elswijk et al.에서 설명된 고-분해능 검사(HRS) 장치에서 DPP-IV 억제 활성에 대해 분석되었다.
DPP-IV 억제는 실시예 1에서 개시된 바와 같은 생물학적 분석의 체제에 기초하여 기질 변환을 통해 검출되었다. DPP-IV 억제 활성 자료에 추가하여, HPLC 방출의 일부가 질량 분석계로 향하게 하여 화학적 정보가 실시간으로 획득되었다. 이러한 방식으로, 두 활성 정보뿐만 아니라 생화학적 활성 화합물의 화학적 특성이 단 일의 크로마토그래프 실행 동안 획득되었다. 적용된 상기 분석 조건은 기질 포화의 적용에 의해 최대 효소 활성을 보증하였다. DPP-IV 억제 활성을 갖는 상기 화합물이 표 2에 확인되었다.

표 2에서, 상대 효능은 본 실험에서 획득된 최고 신호(LPL 및 IPI에 대해)와 대비하여 실시예 1의 DPP-IV 억제 분석에서 화합물에 의해 나타난 신호이다. 가수분해물의 대응하는 펩타이드의 추정 양 및 이들 가수분해물의 IC50에 기초하여, 100% 효능은 약 5μM의 IC50에 상응하는 것으로 추정되며, 이는 6%의 상대 효능은 약 120μM의 IC50에 상응함을 의미한다.

제조 및 활성
가수분해물 4, 5 및 6:
100 그램의 나트륨 카제인염을 교반하면서 1800g의 물에 용해한다. pH는 pH7로 맞춘다. 그 후 물을 2000ml의 최종 부피로 첨가한다. 물에 용해된 표 3에서 확인된(브로멜린, Scintillase CS150L(Genencor), 파파인) 1 그램의 프로테아제가 첨가되고 상기 혼합물은 50℃의 내부 온도로 24시간 동안 인큐베이션 되었다. 상기 효소적 반응은 그 후 95℃에서 5분간 가열하여 정지되고 그 후 상기 혼합물은 동결-건조되었다. 상기 동결-건조된 생성물은 물에 재용해되고 3000rpm에서 원심분리되었다. 상기 상등액(supernatant)은 한계 값 1kDa의 UF 여과를 통해 희석여과(diafiltrate)된다. 상기 출발물질, 상기 건조된 원심분리 펠렛, 상기 건조된 UF 잔존물의 DPP-IV 억제 효과가 결정되었다(표 3, 도 1, 2, 3).

상기 분말은 예비혼합된 반면 상기 Mg 스테아레이트는 혼합의 마지막까지 보류되었다. 상기 타블렛은 직접타정법에 의해 조제되었다(압축 압력 20 kN, 경도: 160N).

함유하는 열-처리된 요구르트 음료의 제조
우유를 물과 혼합한다. 실시예 3에 따른 상기 가수분해물 2, 설탕 및 말티톨 이 첨가되었고 연속적인 교반으로 용해되었으며 후속적으로 저온살균 (Pasteurisation)(90℃, 5분) 되었다. 발효 온도(42℃)로 냉각 후, 상기 접종원이 첨가되었다. 발효는 pH가 4.3에 도달할 때까지 진행되었다. 상기 pH는 락트산을 이용하여 3.8-4.0으로 낮추어졌다. 상기 팩틴이 강한 교반 하에서 첨가되었다. 상기 혼합물은 70℃까지 가열되었고, 120/20 바(bar)에서 균질화되고 향이 첨가되었다. 충전 후 상기 생성물은 저온살균되었다(80℃, 3 분).

모든 건조 성분이 물에 용해되었고 pH가 시트르산(총 +/- 0,14%)으로 먼저 pH 3.8까지 맞춰지고, 그 후 말산(총 +/- 0,66%)으로 pH 3.5까지 맞춰졌다. 상기 용액은 70℃도 예비가열되고 후속적으로 펙틴 예비혼합물(물에 4%)이 첨가되었다. 균질화한 후(150 바(bar)) 상기 생성물은 충전되고 무균적으로 저온살균되거나 저온살균되고 충전되었다.
Claims (26)
- LPL, MPLW, GPFP, PLLQ, KVLP, LPQYL, LPVPQ, VPYPQ, VPLGTQ 및 LPVPQK로부터 선택된 펩타이드가 20-100중량% 함유된 단백질 가수분해물을 활성 성분으로 포함하는, 비만, 제2형 당뇨병 및 면역 질환으로 구성되는 그룹으로부터 선택된 DPP-IV 매개 컨디션의 치료 및 예방을 위한 의약.
- 제 1항에 있어서, 상기 단백질 가수분해물은 우유 단백질 가수분해물인 것을 특징으로 하는 의약.
- 제 2항에 있어서, 상기 단백질 가수분해물은 카세인 가수분해물인 것을 특징으로 하는 의약.
- 제 1항에 있어서, 3-7 아미노산의 길이를 갖는 펩타이드를 20-80wt% 함유하는 것을 특징으로 하는 의약.
- 삭제
- 제 1항에 있어서, 상기 펩타이드는 VPYPQ, VPLGTQ, LPVPQK, KVLP 및 LPL로부터 선택되는 것을 특징으로 하는 의약.

- 제 1항 내지 제 4항 및 제 6항 중 어느 한 항에 있어서, 상기 DPP-IV 매개 컨디션은 면역 질환인 것을 특징으로 하는 의약.
- 제 10항에 있어서, 상기 면역 질환은 류마티즘성 관절염, 다발성 경화증 및 그레이브즈 병으로 구성되는 그룹으로부터 선택되는 자가면역 질환인 것을 특징으로 하는 의약.
- DPP-IV 억제 활성을 가지며, LPL, MPLW, GPFP, PLLQ, KVLP, LPQYL, LPVPQ, VPYPQ, VPLGTQ 및 LPVPQK로부터 선택되는 단리된 펩타이드 또는 이의 혼합물.
- 제 12항에 있어서, 상기 단리된 펩타이드는 VPYPQ, VPLGTQ, LPVPQK, KVLP 및 LPL로부터 선택되는 것을 특징으로 하는 단리된 펩타이드 또는 이의 혼합물.
- 제12항 또는 제13항에 따른 펩타이드를 하나 이상 함유하는 단백질 가수분해물.
- LPL, MPLW, GPFP, PLLQ, KVLP, LPQYL, LPVPQ, VPYPQ, VPLGTQ, 및 LPVPQK로부터 선택된 펩타이드가 20-100중량% 함유된 단백질 가수분해물을 활성 성분으로 포함하며, 비만, 제2형 당뇨병 및 면역 질환으로 구성되는 그룹으로부터 선택된 DPP-IV 매개 컨디션을 개선시키는, DPP-IV 활성 억제를 위한 식품.
- 제 15항에 있어서, 상기 단백질 가수분해물은 우유 단백질 가수분해물인 것을 특징으로 하는 식품.
- 제 16항에 있어서, 상기 단백질 가수분해물은 카세인 가수분해물인 것을 특징으로 하는 식품.
