本発明は、ジペプチジルペプチダーゼ-IV阻害活性を有する新規ペプチドに関する。
ジペプチジルペプチダーゼ-IV(di-peptidyl peptidase-IV:以下、「DPP-IV」ともいう)は、N末端ジペプチダーゼ活性を有する多機能性の貫通膜型糖タンパク質である。DPP-IVは、大部分の哺乳類の、肝臓、腎臓、小腸、唾液腺、血液細胞及び血漿等の種々の組織の細胞上に存在する。
DPP-IVは生体内での様々な役割が果たすことが想定されており、その1つとしてインクレチンであるグルカゴン様ペプチド1(Glucagon-like peptide-1:以下、「GLP-1」という)及びグルコース依存性インスリン分泌刺激ポリペプチド(Glucose-dependent insulinotropic polypeptide:以下、「GIP」という)の分解酵素であることが知られている。
DPP-IVは生体内での様々な役割が果たすことが想定されており、その1つとしてインクレチンであるグルカゴン様ペプチド1(Glucagon-like peptide-1:以下、「GLP-1」という)及びグルコース依存性インスリン分泌刺激ポリペプチド(Glucose-dependent insulinotropic polypeptide:以下、「GIP」という)の分解酵素であることが知られている。
GLP-1は、食後に放出されるものであり、インスリンの生合成及び分泌に対するグルコース誘導性の刺激、グルカゴン分泌抑制、遺伝子発現の調整、β細胞に対する栄養性の効果、食物摂取の抑制、及び胃内容排出の緩徐化をはじめとする、多面的な作用を有する。GIPもまた、食後に放出され、血糖濃度依存的に膵臓からのインスリン分泌を促進する作用を有する。
DPP-IVを阻害すると、GLP-1及びGIPの分解が抑制され、血中のこれらインクレチン濃度が上昇する。その結果、インスリン分泌が促進され、血糖値を低下させることが知られている。このインクレチンによるインスリン分泌促進の作動条件は血糖値が高いことであるため、2型糖尿病の中でもインスリン分泌低下による糖尿病において、DPP-IVを阻害することは、従前のインスリン分泌促進剤で生じる低血糖の副作用が生じにくいと考えられる。
DPP-IVを阻害すると、GLP-1及びGIPの分解が抑制され、血中のこれらインクレチン濃度が上昇する。その結果、インスリン分泌が促進され、血糖値を低下させることが知られている。このインクレチンによるインスリン分泌促進の作動条件は血糖値が高いことであるため、2型糖尿病の中でもインスリン分泌低下による糖尿病において、DPP-IVを阻害することは、従前のインスリン分泌促進剤で生じる低血糖の副作用が生じにくいと考えられる。
このように、DPP-IV活性を阻害する作用を有するDPP-IV阻害剤は、新しいタイプの糖尿病治療薬の有効成分として期待されており、すでに複数の医薬が上市されている。また、さらなるDPP-IV阻害剤となり得る成分の探索も、引き続き行われている(特許文献1等)。
また、糖尿病治療においては食餌療法と運動療法が第一選択であり、それでも血糖値コントロールが不十分な場合に糖尿病治療薬を用いるのが通常である。かかる状況においては、DPP-IV阻害活性剤をサプリメントや食品添加物等の形態で食餌により摂取することの需要がある。
そこで、DPP-IV阻害活性を有し、さらに飲食品として適用し得る成分の探索も種々行われている。例えば、乳タンパク質由来のオリゴペプチドのいくつかについてDPP-IV阻害活性が見出されている(特許文献2~5)。特に、カゼイン由来のトリペプチドについてもDPP-IV阻害活性が確認されている(特許文献6~7)。
そこで、DPP-IV阻害活性を有し、さらに飲食品として適用し得る成分の探索も種々行われている。例えば、乳タンパク質由来のオリゴペプチドのいくつかについてDPP-IV阻害活性が見出されている(特許文献2~5)。特に、カゼイン由来のトリペプチドについてもDPP-IV阻害活性が確認されている(特許文献6~7)。
本発明は、優れたDPP-IV阻害活性を有するペプチドを提供することを課題とする。
本発明者らは、上記の課題を解決すべく鋭意研究を行った結果、乳タンパク質に由来する特定のアミノ酸配列を含むオリゴペプチドが、高いDPP-IV阻害活性を有することを見出し、本発明を完成するに至った。
すなわち、本発明の第一の態様は、以下の(a)~(g)に記載のいずれかの配列を含むオリゴペプチドである。

次に、本発明を詳細に説明する。ただし、本発明は以下の実施形態に限定されず、本発明の範囲内で自由に変更することができるものである。

例えば、配列(a)Leu-Pro-Valを含むオリゴペプチドとしては、配列(h)Leu-Pro-Val-Proを含むオリゴペプチドを好ましく挙げられる。
本発明においてオリゴペプチドは、アミノ酸長が3~10残基のものが好ましく、3~7残基のものがより好ましく、3~4残基のものがさらに好ましい。

(a)~(h)に記載のいずれかの配列を含むオリゴペプチドは、その塩の形態であってもよい。塩としては、例えば、カリウム、ナトリウム等のアルカリ金属類;カルシウム、マグネシウム等のアルカリ土類金属類等が挙げられる。
本発明のオリゴペプチドは、例えば、(1)所望の(a)~(h)に記載のいずれかのアミノ酸配列を含むタンパク質やペプチドを加水分解酵素等にて分解し、得られた分解物から分離精製して得る方法、(2)ペプチドの合成法によって本発明のオリゴペプチドを合成した後、得られた粗合成物から所望の本発明のオリゴペプチドを分離精製して得る方法、(3)本発明に係る本発明のオリゴペプチドを生産する植物、動物又は微生物から抽出し、得られた抽出物から分離精製する方法等により得ることができる。
以下に(1)及び(2)の方法について具体的に説明する。

(1)加水分解により得る方法
本発明のオリゴペプチドは、所望の(a)~(h)に記載のいずれかのアミノ酸配列を含むタンパク質やペプチドをタンパク質加水分解酵素や、酸・アルカリ等により加水分解し、得られた加水分解物から本発明のオリゴペプチドを分離精製して得ることができる。原料となるタンパク質やペプチドを含むものとしては、例えば、乳、大豆、卵、小麦、大麦、米、じゃが芋、さつま芋、えんどう豆、トウモロコシ、畜肉、魚肉、魚介などに由来するタンパク質などが挙げられ、これらのうち乳タンパク質であるカゼインが特に好ましい。
本発明のオリゴペプチドは、所望の(a)~(h)に記載のいずれかのアミノ酸配列を含むタンパク質やペプチドをタンパク質加水分解酵素や、酸・アルカリ等により加水分解し、得られた加水分解物から本発明のオリゴペプチドを分離精製して得ることができる。原料となるタンパク質やペプチドを含むものとしては、例えば、乳、大豆、卵、小麦、大麦、米、じゃが芋、さつま芋、えんどう豆、トウモロコシ、畜肉、魚肉、魚介などに由来するタンパク質などが挙げられ、これらのうち乳タンパク質であるカゼインが特に好ましい。
以下にカゼインを原料として用いる場合を例に挙げて、加水分解酵素による処理によって本発明のオリゴペプチドを得る方法を説明する。
カゼインタンパク質は、本発明のオリゴペプチドを一次構造中に含むタンパク質であって、適宜加水分解酵素で消化したときに本発明のオリゴペプチドが生成可能なものである。
まず、酵素で加水分解する前に、原料タンパク質を水又は温湯に分散し、溶解してタンパク質水溶液を調製する。当該タンパク質水溶液の濃度は、特に限定されないが、通常、タンパク質濃度として2質量%以上、さらに好ましくは5~15質量%程度の濃度範囲に設定するのが好適である。
さらに、前記タンパク質水溶液を、ナトリウム型又はカリウム型陽イオン交換樹脂(好適には強酸性陽イオン交換樹脂)を用いたイオン交換法、電気透析法、限界ろ過膜法、ルーズ逆浸透膜法等で脱塩し、適宜pH調整やカルシウム濃度調整を行うのが好適である。脱塩の際には、カラム式やバッチ式の何れを採用してもよい。また、タンパク質水溶液を、脱塩前等に適宜、加熱殺菌をおこなってもよい。
カゼインタンパク質は、本発明のオリゴペプチドを一次構造中に含むタンパク質であって、適宜加水分解酵素で消化したときに本発明のオリゴペプチドが生成可能なものである。
まず、酵素で加水分解する前に、原料タンパク質を水又は温湯に分散し、溶解してタンパク質水溶液を調製する。当該タンパク質水溶液の濃度は、特に限定されないが、通常、タンパク質濃度として2質量%以上、さらに好ましくは5~15質量%程度の濃度範囲に設定するのが好適である。
さらに、前記タンパク質水溶液を、ナトリウム型又はカリウム型陽イオン交換樹脂(好適には強酸性陽イオン交換樹脂)を用いたイオン交換法、電気透析法、限界ろ過膜法、ルーズ逆浸透膜法等で脱塩し、適宜pH調整やカルシウム濃度調整を行うのが好適である。脱塩の際には、カラム式やバッチ式の何れを採用してもよい。また、タンパク質水溶液を、脱塩前等に適宜、加熱殺菌をおこなってもよい。
次いで、前記タンパク質水溶液を、加水分解処理する。当該加水分解処理として、例えば酵素処理、酸処理、アルカリ処理、熱処理等が挙げられ、これらの2種以上の処理を適宜組み合わせてもよい。
酵素処理には、植物由来、動物由来、微生物由来等のタンパク質分解酵素を使用でき、これらから1種又は2種以上組み合わせて使用できる。当該タンパク質分解酵素としては、エンドプロテアーゼが好適である。
前記エンドプロテア-ゼとしては、例えば、セリンプロテアーゼ、メタロプロテアーゼ、システインプロテアーゼ、アスパラギン酸プロテアーゼが挙げられ、これらを1種又は2種以上選択して用いることができる。このうち、セリンプロテアーゼ及び/又はメタロプロテアーゼを用いるのが好適である。
また、プロテアーゼは、アルカリ性プロテアーゼ、中性プロテアーゼ及び酸性プロテアーゼに分類される。このうち中性プロテアーゼを用いるのが好適である。
酵素処理には、植物由来、動物由来、微生物由来等のタンパク質分解酵素を使用でき、これらから1種又は2種以上組み合わせて使用できる。当該タンパク質分解酵素としては、エンドプロテアーゼが好適である。
前記エンドプロテア-ゼとしては、例えば、セリンプロテアーゼ、メタロプロテアーゼ、システインプロテアーゼ、アスパラギン酸プロテアーゼが挙げられ、これらを1種又は2種以上選択して用いることができる。このうち、セリンプロテアーゼ及び/又はメタロプロテアーゼを用いるのが好適である。
また、プロテアーゼは、アルカリ性プロテアーゼ、中性プロテアーゼ及び酸性プロテアーゼに分類される。このうち中性プロテアーゼを用いるのが好適である。
前記タンパク質分解酵素は、市販品を用いることができる。前記タンパク質分解酵素として、例えば、ビオプラーゼ(ナガセケムテックス社製)、プロレザー(天野エンザイム社製)、プロテアーゼS(天野エンザイム社製)、PTN6.0S(ノボザイムズ社製)、サビナーゼ(ノボザイムズ社製)、GODO B.A.P(合同酒精社製)、プロテアーゼN(天野エンザイム社製)、GODO B.N.P(合同酒精社製)、ニュートラーゼ(ノボザイムズ社製)、アルカラーゼ(ノボザイムズ社製)、トリプシン(ノボザイムズ社製)、キモトリプシン(ノボザイムズ社製)、スブチリシン(ノボザイムズ社製)、パパイン(天野エンザイム社製)、ブロメライン(天野エンザイム社製)等が挙げられ、これらから1種又は2種以上の酵素を選択して用いてもよい。
これらのうち、スブチリシン(subtilisin:例えば、ビオプラーゼ)、トリプシン(trypsin:例えばPTN6.0S)、及びバシロリシン(bachillolysin:例えばプロテアーゼN)から選ばれる1種又は2種以上の中性プロテアーゼが好ましく、より好ましくはこれら3種を組み合わせて用いる。
これらのうち、スブチリシン(subtilisin:例えば、ビオプラーゼ)、トリプシン(trypsin:例えばPTN6.0S)、及びバシロリシン(bachillolysin:例えばプロテアーゼN)から選ばれる1種又は2種以上の中性プロテアーゼが好ましく、より好ましくはこれら3種を組み合わせて用いる。
前記タンパク質に対するエンドプロテア-ゼの使用量は、特に限定されず、基質濃度、酵素力価、反応温度及び反応時間等により適宜調整すればよいが、一般的には、タンパク質中のタンパク質1g当り2,000~11,000活性単位の割合で添加することが好ましい。
前記タンパク質分解酵素による加水分解条件を適宜調整することにより、所望のペプチドを得ることができる。

前記タンパク質分解酵素による加水分解前に、前記原料タンパク質溶液のpHを、炭酸カリウム、水酸化ナトリウム等の食品上使用可能な塩類を用いて、使用酵素の至適pHに調整することもできる。前記原料タンパク質溶液のpHは、好ましくは5~10、より好ましくは7~10に調整する。
前記タンパク質分解酵素の反応温度は、使用酵素の最適温度の範囲で行うことが望ましく、好ましくは30~60℃、より好ましくは40~60℃で行う。
前記タンパク質分解酵素の反応保持時間は、酵素反応の分解率をモニターしながら、好ましい分解率に達するまで反応を続ければよく例えば0.5~24時間で行うことが可能であり、好ましくは1~15時間、より好ましくは3~10時間である。特に、原料に前記カゼインタンパク質を用いた場合の分解率は20~30%であることが好ましい。
前記タンパク質分解酵素の反応保持時間は、酵素反応の分解率をモニターしながら、好ましい分解率に達するまで反応を続ければよく例えば0.5~24時間で行うことが可能であり、好ましくは1~15時間、より好ましくは3~10時間である。特に、原料に前記カゼインタンパク質を用いた場合の分解率は20~30%であることが好ましい。
なお、原料タンパク質の分解率の算出方法は、ケルダール法(日本食品工業学会編、「食品分析法」、第102頁、株式会社光琳、昭和59年)により試料の全窒素量を測定し、ホルモール滴定法(満田他編、「食品工学実験書」、上巻、第547ページ、養賢堂、1970年)により試料のホルモール態窒素量を測定し、これらの測定値から分解率を次式により算出する。

前記タンパク質分解酵素による加水分解は、当該酵素を加熱して失活させて終了させればよい。加熱失活処理の加熱温度と保持時間は、使用した酵素の熱安定性を考慮し、十分に失活できる条件を適宜設定することができる。例えば、100℃以上(好適には110~130℃)で失活させる場合には1~3秒間、100℃未満60℃以上で失活させる場合には3~40分間で行うことが好適である。
加熱処理の方式としては、バッチ方式、連続方式のいずれの方式も可能であり、連続方式として、プレート熱交換方式、インフュージョン方式、インジェクション方式等の方式を用いることができる。
なお、前記の加熱失活処理は、加水分解物の殺菌処理として併用することも可能であり、常法による加熱処理方法等を用いることができる。
加水分解終了後、必要に応じて分解液のpHを、好ましくは6~8、より好ましくは7.0±0.5、さらに好ましくは7.0±0.3とするのが好適である。
加熱処理の方式としては、バッチ方式、連続方式のいずれの方式も可能であり、連続方式として、プレート熱交換方式、インフュージョン方式、インジェクション方式等の方式を用いることができる。
なお、前記の加熱失活処理は、加水分解物の殺菌処理として併用することも可能であり、常法による加熱処理方法等を用いることができる。
加水分解終了後、必要に応じて分解液のpHを、好ましくは6~8、より好ましくは7.0±0.5、さらに好ましくは7.0±0.3とするのが好適である。
なお、本発明に係るタンパク質分解物の製造において、カルシウム濃度未調整の溶液を加水分解した場合には、得られた分解液を、前記のような脱塩処理し、カルシウム濃度を調整してもよい。次いで、常法により加熱して酵素を失活させる。反応加熱温度と反応保持時間は使用した酵素の熱安定性を配慮し、十分に失活できる条件を適宜設定することができる。加熱失活後、常法により冷却し、そのまま利用することもでき、必要に応じて濃縮して濃縮液を得ることもでき、更に濃縮液を乾燥し、粉末製品を得ることも可能である。
また、前記タンパク質水溶液を酸処理又はアルカリ処理にて加水分解する際には、タンパク質水溶液のpHを調整して処理すればよい。当該pH調整による処理の場合には、タンパク質水溶液のpHが、好ましくはpH5以下又はpH9以上であり、より好ましくはpH4以下又はpH10以上である。このようにpH処理された水溶液は、室温にて数分以上、好ましくは5分~1時間、放置又は撹拌することによって、酸処理又はアルカリ処理の加水分解物を得ることができる。ここで、「室温」とは、4~40℃程度であるが、10~30℃が好適である。
また、前記タンパク質水溶液を、熱処理にて加水分解してもよい。このタンパク質水溶液は、pH未調整でもよく、またpH調整(具体的には、酸性(pH5以下)、中性(pH6~8)、アルカリ性(pH8以上))してもよい。熱処理は、4~100℃程度で、上記酸アルカリ処理のような条件にて行えばよい。
また、前記タンパク質水溶液を、熱処理にて加水分解してもよい。このタンパク質水溶液は、pH未調整でもよく、またpH調整(具体的には、酸性(pH5以下)、中性(pH6~8)、アルカリ性(pH8以上))してもよい。熱処理は、4~100℃程度で、上記酸アルカリ処理のような条件にて行えばよい。
タンパク質加水分解物中の本発明のオリゴペプチドの含有率は特に限定されないが、その下限値は、本発明の効能をより良好に発揮させる観点から、好ましくは0.001質量%以上であり、0.005質量%以上であり、より好ましくは0.01質量%以上であり、その上限値は、本発明の分解物の製造効率の観点から、好ましくは1質量%以下であり、より好ましくは0.5質量%以下であり、さらに好ましくは0.4質量%以下であり、よりさらに好ましくは0.3質量%以下である。
なお、本発明において、対象のペプチド含有量は、下記の方法にて測定できる。
(a)試料粉末を、1.0mg/mLとなるように、0.2%ギ酸水溶液に希釈溶解し、10分間超音波破砕したのち、0.22μm口径のPVDFフィルター(Millipore社製)でろ過して粉末溶液を調製し、下記測定条件によるLC/MS分析を実施する。一方、測定対象のペプチドの化学合成標準ペプチド(ペプチド研究所社製)の溶解液を濃度別に数点調製し、下記測定条件によるLC/MS分析を実施し、検量線を作成する。
前記粉末溶液の分析におけるピークのうち、標準ペプチドと分子量及びリテンションタイムが一致するものを、標準ペプチドと同一の配列として同定する。標準ペプチドのピーク面積と資料粉末のピーク面積を対比することにより、前記粉末溶液中に対象ペプチドの含有量を求める。

得られたタンパク質加水分解物は、未精製のままの状態で使用しても効能を発揮することが可能である。すなわち、タンパク質加水分解物を、好ましくは乳タンパク質加水分解物を後述の飲食品や医薬組成物の態様として、摂取や投与に供してもよい。
また、さらに、得られたタンパク質加水分解物に対して、適宜公知の分離精製を行ってもよい。例えば、得られたタンパク質加水分解物に対して分子量分画を行い、本発明に係る本発明のオリゴペプチドの分子量に該当する分画を含むタンパク質分解物を得ることができる。目的物質を含む画分は、後述するDPP-IV阻害作用を指標として決定することができる。
分子量分画として、例えば、限外ろ過、ゲルろ過等の方法が採用でき、これにより不要な分子量のペプチドや遊離アミノ酸の除去率を高めることができる。
限外ろ過の場合には、所望の限外ろ過膜を使用すればよく、ゲルろ過の場合には、所望のサイズ排除クロマトグラフィーに用いるゲルろ過剤を使用すればよい。
さらに、脱塩や不純物を除去したり、純度を高めたりするために、公知の分離精製方法例えば、イオン交換クロマトグラフィー、吸着クロマトグラフィー、逆相クロマトグラフィー、分配クロマトグラフィー等の各種クロマトグラフィー、溶媒沈殿、塩析、2種の液相間での分配等の方法を用いてもよい。
また、さらに、得られたタンパク質加水分解物に対して、適宜公知の分離精製を行ってもよい。例えば、得られたタンパク質加水分解物に対して分子量分画を行い、本発明に係る本発明のオリゴペプチドの分子量に該当する分画を含むタンパク質分解物を得ることができる。目的物質を含む画分は、後述するDPP-IV阻害作用を指標として決定することができる。
分子量分画として、例えば、限外ろ過、ゲルろ過等の方法が採用でき、これにより不要な分子量のペプチドや遊離アミノ酸の除去率を高めることができる。
限外ろ過の場合には、所望の限外ろ過膜を使用すればよく、ゲルろ過の場合には、所望のサイズ排除クロマトグラフィーに用いるゲルろ過剤を使用すればよい。
さらに、脱塩や不純物を除去したり、純度を高めたりするために、公知の分離精製方法例えば、イオン交換クロマトグラフィー、吸着クロマトグラフィー、逆相クロマトグラフィー、分配クロマトグラフィー等の各種クロマトグラフィー、溶媒沈殿、塩析、2種の液相間での分配等の方法を用いてもよい。
分離精製したペプチドの分画物は、本発明のオリゴペプチドが含まれているかどうかを確認することを目的として、質量分析法によりペプチドの同定を行うことができる。
このようにして得られた本発明のオリゴペプチドは、ペプチド溶液のまま使用することもでき、また、必要に応じて、該溶液を公知の方法により、濃縮した濃縮液として使用することもできる。また、該濃縮液を公知の方法により乾燥し、粉末にして使用することもできる。

(2)合成により得る方法
本発明のオリゴペプチドは、化学合成又は生合成によっても製造することができる。
ペプチドの化学合成は、オリゴペプチドの合成に通常用いられている液相法または固相法によって行うことができる。合成されたペプチドは必要に応じて脱保護され、未反応試薬や副生物等を除去して、本発明のオリゴペプチドを単離することが可能である。このようなペプチドの合成は、市販のペプチド合成装置を用いて行うことができる。
ペプチドの生合成は、宿主生物にペプチド発現ベクターを導入して生成・分泌させるといった常法により行うことができる。
目的とするペプチドが得られたことは、後述するDPP-IV阻害作用を指標として確認することができる。
本発明のオリゴペプチドは、化学合成又は生合成によっても製造することができる。

目的とするペプチドが得られたことは、後述するDPP-IV阻害作用を指標として確認することができる。
本発明のオリゴペプチドは、DPP-IV活性を阻害する作用を有する。
ここで、DPP-IV阻害活性を有するとは、本発明のオリゴペプチド存在下におけるDPP-IV活性が、非存在下における活性に比べて小さいことをいい、同条件における阻害率が好ましくは10%以上、より好ましくは30%以上、さらに好ましくは50%以上であることをいう。また、酵素の50%阻害濃度(IC50)が、好ましくは1000μM以下、より好ましくは500μM以下、さらに好ましくは100μM以下であることをいう。
なお、DPP-IV阻害活性、阻害率、IC50は、定法により確認することができる。
ここで、DPP-IV阻害活性を有するとは、本発明のオリゴペプチド存在下におけるDPP-IV活性が、非存在下における活性に比べて小さいことをいい、同条件における阻害率が好ましくは10%以上、より好ましくは30%以上、さらに好ましくは50%以上であることをいう。また、酵素の50%阻害濃度(IC50)が、好ましくは1000μM以下、より好ましくは500μM以下、さらに好ましくは100μM以下であることをいう。
なお、DPP-IV阻害活性、阻害率、IC50は、定法により確認することができる。
したがって、本発明のオリゴペプチドは、DPP-IV阻害剤の有効成分として好ましく含有させることができる。ここで、本発明のオリゴペプチドは乳タンパク質加水分解物の態様で剤に含まれていてもよく、言い換えると本発明のオリゴペプチドを一種又は二種以上を含む乳タンパク質加水分解物は、DPP-IV阻害剤の有効成分として好ましく含有させることができる。
DPP-IVが、生体内の生理機能に関与している物質を分解することで、種々の疾患や症状が生じる場合がある。このため、DPP-IVを阻害すると、通常DPP-IVによって分解される生体内の生理機能に関与している物質の分解が抑制されその寿命が延びることを利用して、DPP-IVに起因する疾患や症状の予防、改善又は治療が可能となる。
前記生理機能に関与する物質として、例えば、GLP-1やGIPといったインクレチンが挙げられる。GLP-1の分解を抑制することによって、インスリンの生合成及び分泌に対するグルコース誘導性の刺激、グルカゴン分泌抑制、遺伝子発現の調整、β細胞に対する栄養性の効果、食物摂取の抑制、及び胃内容排出の緩徐化などが作用する。また、GIPの分解を抑制することによって、膵臓からのインスリン分泌に対するグルコース誘導性の刺激が作用する。これらによって、上昇した血糖値の正常化、並びに空腹感及び体重の改善・調整に寄与することが可能である。
前記生理機能に関与する物質として、例えば、GLP-1やGIPといったインクレチンが挙げられる。GLP-1の分解を抑制することによって、インスリンの生合成及び分泌に対するグルコース誘導性の刺激、グルカゴン分泌抑制、遺伝子発現の調整、β細胞に対する栄養性の効果、食物摂取の抑制、及び胃内容排出の緩徐化などが作用する。また、GIPの分解を抑制することによって、膵臓からのインスリン分泌に対するグルコース誘導性の刺激が作用する。これらによって、上昇した血糖値の正常化、並びに空腹感及び体重の改善・調整に寄与することが可能である。
また、DPP-IVの作用によって、血管の内皮細胞の機能が低下したり、血管の内皮細胞が障害されることが知られている。この血管の内皮細胞の機能低下や障害によって、血管の緊張増加による血管収縮、動脈硬化、血栓形成等の血管障害が生じ、これらの原因によって臓器の血流障害が生じ、臓器機能不全となり、糖尿病の合併症が誘発されることとなる。
近年、DPP-IV阻害薬を投与することで内皮細胞の機能が改善することが多数報告されている(例えば、Endocrine Journal 2011, 58 (1), 69-73、J Am Coll Cardiol. 2012, 59(3), 265-76、Diabetes Care. 2011, 34(9), 2072-7、Cardiovascular Diabetology 2011, 10(85)等)。かかる内皮細胞機能改善効果は、単に血糖値を下げることによる改善の他に、インクレチンによる血管の保護作用を介していると考えられている。高血糖により内皮細胞の機能が低下し、血管のしなやかさが失われると、血圧が上昇し、上昇した血圧がさらに血管を傷めるという悪循環が、心臓・腎臓・脳といった臓器への悪影響となって現れる。そのため、DPP-IV阻害剤は循環器系の治療においても、重要な役割を果たすものと考えられている。
近年、DPP-IV阻害薬を投与することで内皮細胞の機能が改善することが多数報告されている(例えば、Endocrine Journal 2011, 58 (1), 69-73、J Am Coll Cardiol. 2012, 59(3), 265-76、Diabetes Care. 2011, 34(9), 2072-7、Cardiovascular Diabetology 2011, 10(85)等)。かかる内皮細胞機能改善効果は、単に血糖値を下げることによる改善の他に、インクレチンによる血管の保護作用を介していると考えられている。高血糖により内皮細胞の機能が低下し、血管のしなやかさが失われると、血圧が上昇し、上昇した血圧がさらに血管を傷めるという悪循環が、心臓・腎臓・脳といった臓器への悪影響となって現れる。そのため、DPP-IV阻害剤は循環器系の治療においても、重要な役割を果たすものと考えられている。
以上のことから、本発明のオリゴペプチド及び本発明のオリゴペプチドを一種又は二種以上を含む乳タンパク質加水分解物、並びにこれらを含有する本発明のDPP-IV阻害剤は、DPP-IV阻害活性を有することにより、血糖値上昇抑制作用、高血糖改善作用、血管内皮機能低下抑制作用、血管内皮障害抑制作用、血管障害抑制作用、血管内皮細胞の保護作用、食欲抑制作用等を示す。
なお、本明細書における「血糖値上昇抑制」とは、血糖値低下を含む意味であるが、特に「正常値以上又は必要以上に上昇した血糖値を下げることができること」を意味するものである。血糖値の正常値の判断は、日本糖尿病学会の診断基準(2012年改訂)を参考にすればよい。なお、血糖値上昇は、食後に生じる上昇を指すものであってよい。
さらに、本発明のオリゴペプチド及びそれを含有する本発明のDPP-IV阻害剤は、DPP-IV阻害活性を有することにより、DPP-IVに起因する疾患や症状の予防、改善又は治療が可能と考えられる。よって、本発明のオリゴペプチド及びそれを含有する本発明のDPP-IV阻害剤は、ヒトを含む動物に摂取又は投与して、DPP-IVに起因する疾患や症状等の予防、改善及び/又は治療を図るための方法に使用することができる。
ここで、疾患や症状等の予防とは、疾患や症状等を罹患(発症)していない適用対象において、疾患又は症状等の発生を防止すること、該発生を遅延させること、及び該発生の危険性を低下させることを含む。
なお、本明細書における「血糖値上昇抑制」とは、血糖値低下を含む意味であるが、特に「正常値以上又は必要以上に上昇した血糖値を下げることができること」を意味するものである。血糖値の正常値の判断は、日本糖尿病学会の診断基準(2012年改訂)を参考にすればよい。なお、血糖値上昇は、食後に生じる上昇を指すものであってよい。
さらに、本発明のオリゴペプチド及びそれを含有する本発明のDPP-IV阻害剤は、DPP-IV阻害活性を有することにより、DPP-IVに起因する疾患や症状の予防、改善又は治療が可能と考えられる。よって、本発明のオリゴペプチド及びそれを含有する本発明のDPP-IV阻害剤は、ヒトを含む動物に摂取又は投与して、DPP-IVに起因する疾患や症状等の予防、改善及び/又は治療を図るための方法に使用することができる。
ここで、疾患や症状等の予防とは、疾患や症状等を罹患(発症)していない適用対象において、疾患又は症状等の発生を防止すること、該発生を遅延させること、及び該発生の危険性を低下させることを含む。
本発明のオリゴペプチドを投与する対象(被投与者)及び摂取させる対象(摂取者)は、動物であれば特に限定されないが、通常はヒトである。また、成人、小児、乳児、新生児等のいずれであってもよい。また、性別は特に限定されない。
本発明の別の側面は、DPP-IV阻害剤、又はDPP-IVに起因する疾患や症状の予防、改善及び/若しくは治療のための組成物の製造における、本発明のオリゴペプチドの使用である。

本発明のオリゴペプチドの摂取(投与)量は、本発明に係る本発明のオリゴペプチド1種類ずつの摂取量として、例えば、成人において1μg/日~10mg/日の範囲が好ましく、5μg/日~5mg/日の範囲がさらに好ましい。
なお、摂取(投与)の量や期間にかかわらず、本発明のオリゴペプチドは1日1回又は複数回に分けて投与することができる。

本発明のオリゴペプチドの摂取(投与)経路は、経口又は非経口のいずれでもよいが経口が好ましい。また、非経口摂取(投与)としては、経皮、静注、直腸投与、吸入等が挙げられる。
本発明のDPP-IV阻害剤は、前述の通りDPP-IVに起因する疾患や症状の予防、改善又は治療に好適に用いることができるため、医薬組成物に含有させる態様が好ましい。すなわち、DPP-IVに起因する疾患や症状の予防、改善又は治療のための医薬組成物も本発明の一態様である。
DPP-IVに起因する疾患や症状として、例えば、高血糖症、糖尿病、糖尿病合併症、血管内皮障害、血管障害等が挙げられるがこれらに限定されない。糖尿病としては、好ましくは2型糖尿病が挙げられ、より好ましくはインスリン分泌低下による2型糖尿病が挙げられる。なお、DPP-IVに起因する疾患や症状には、DPP-IVが直接的に関与するものの他、DPP-IVが間接的に関与するものであってよい。
さらに、高血糖症、糖尿病及び高血糖状態によって引き起こされる種々の疾患や症状も、本発明の医薬組成物の対象となり得る。かかる疾患や症状としては、例えば、糖尿病性の細小血管症(例えば、網膜症、腎症、神経障害等)及び大血管合併症(例えば、狭心症・心筋梗塞等の虚血性心疾患、脳梗塞、閉塞性動脈硬化、壊疽等)等が挙げられる。
DPP-IVに起因する疾患や症状として、例えば、高血糖症、糖尿病、糖尿病合併症、血管内皮障害、血管障害等が挙げられるがこれらに限定されない。糖尿病としては、好ましくは2型糖尿病が挙げられ、より好ましくはインスリン分泌低下による2型糖尿病が挙げられる。なお、DPP-IVに起因する疾患や症状には、DPP-IVが直接的に関与するものの他、DPP-IVが間接的に関与するものであってよい。

医薬組成物の投与経路は、経口又は非経口のいずれでもよいが経口が好ましい。また、非経口摂取(投与)としては、経皮、静注、直腸投与、吸入等が挙げられる。

製剤化に際しては、本発明のオリゴペプチドの他に、通常製剤化に用いられている賦形剤、pH調整剤、着色剤、矯味剤等の成分を用いることができる。また、他の薬効成分や、公知の又は将来的に見出されるDPP-IVに起因する疾患や症状を予防、改善及び/又は治療し得る成分等の他の医薬を併用することも可能である。
また、本発明の効果を妨げない限りにおいて、他のペプチドが併存してもかまわない。
加えて、製剤化は剤形に応じて適宜公知の方法により実施できる。製剤化に際しては、適宜、通常製剤化に用いる担体を配合して製剤化してもよい。かかる担体としては、賦形剤、結合剤、崩壊剤、滑沢剤、安定剤、矯味矯臭剤等が挙げられる。

結合剤としては、例えば、上記賦形剤の他、ゼラチン;ポリビニルピロリドン;マクロゴール等が挙げられる。
崩壊剤としては、例えば、上記賦形剤の他、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、架橋ポリビニルピロリドン等の化学修飾されたデンプン又はセルロース誘導体等が挙げられる。
滑沢剤としては、例えば、タルク;ステアリン酸;ステアリン酸カルシウム、ステアリン酸マグネシウム等のステアリン酸金属塩;コロイドシリカ;ピーガム、ゲイロウ等のワックス類;硼酸;グリコール;フマル酸、アジピン酸等のカルボン酸類;安息香酸ナトリウム等のカルボン酸ナトリウム塩;硫酸ナトリウム等の硫酸塩類;ロイシン;ラウリル硫酸ナトリウム、ラウリル硫酸マグネシウム等のラウリル硫酸塩;無水珪酸、珪酸水和物等の珪酸類;デンプン誘導体等が挙げられる。
安定剤としては、例えば、メチルパラベン、プロピルパラベン等のパラオキシ安息香酸エステル類;クロロブタノール、ベンジルアルコール、フェニルエチルアルコール等のアルコール類;塩化ベンザルコニウム;無水酢酸;ソルビン酸等が挙げられる。
矯味矯臭剤としては、例えば、甘味料、酸味料、香料等が挙げられる。
なお、経口投与用の液剤の場合に使用する担体としては、水等の溶剤等が挙げられる。

本発明の医薬組成物を摂取するタイミングは、例えば食前、食後、食間、就寝前など特に限定されないが食前が好ましい。
本発明のオリゴペプチドを経口摂取される組成物とする場合は、飲食品の態様とすることが好ましい。
本発明のオリゴペプチドは、前述のようにDPP-IV阻害活性を有することにより、血糖値上昇抑制作用、高血糖改善作用、血管内皮機能低下抑制作用、血管内皮障害抑制作用、血管障害抑制作用、血管内皮細胞の保護作用、食欲抑制作用等を示すため、これらの作用を企図して用いられる組成物の態様とすることができる。特に好ましい態様としては、食後血糖値上昇抑制用の組成物が挙げられる。
本発明のオリゴペプチドは、特に用途を限定せずに飲食品の態様とすることができる。また、前述した種々の作用に係る用途に供される飲食品とすることもでき、特に好ましくは食後血糖値上昇抑制用の飲食品とすることができる。

本発明のオリゴペプチドが飲食品(飼料を含む)の態様である場合、DPP-IVに起因する疾患や症状の改善に関する用途や、脳機能低下予防、記憶力低下予防等の用途が表示された飲食品として提供・販売されることが可能である。また、本明細書に係る本発明のオリゴペプチドは、これら飲食品等の製造のために使用可能である。
かかる「表示」行為には、需要者に対して前記用途を知らしめるための全ての行為が含まれ、前記用途を想起・類推させうるような表現であれば、表示の目的、表示の内容、表示する対象物・媒体等の如何に拘わらず、全て本発明における「表示」行為に該当する。
また、「表示」は、需要者が上記用途を直接的に認識できるような表現により行われることが好ましい。具体的には、飲食品に係る商品又は商品の包装に前記用途を記載したものを譲渡し、引き渡し、譲渡若しくは引き渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネット等)方法により提供する行為等が挙げられる。
また、「表示」は、需要者が上記用途を直接的に認識できるような表現により行われることが好ましい。具体的には、飲食品に係る商品又は商品の包装に前記用途を記載したものを譲渡し、引き渡し、譲渡若しくは引き渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネット等)方法により提供する行為等が挙げられる。
一方、表示内容としては、行政等によって認可された表示(例えば、行政が定める各種制度に基づいて認可を受け、そのような認可に基づいた態様で行う表示等)であることが好ましい。また、そのような表示内容を、包装、容器、カタログ、パンフレット、POP等の販売現場における宣伝材、その他の書類等へ付することが好ましい。
また、「表示」には、健康食品、機能性食品、経腸栄養食品、特別用途食品、保健機能食品、特定保健用食品、栄養機能食品、機能性表示食品、医薬用部外品等としての表示も挙げられる。この中でも特に、消費者庁によって認可される表示、例えば、特定保健用食品、栄養機能食品、若しくは機能性表示食品に係る制度、又はこれらに類似する制度にて認可される表示等が挙げられる。具体的には、特定保健用食品としての表示、条件付き特定保健用食品としての表示、身体の構造や機能に影響を与える旨の表示、疾病リスク減少表示、科学的根拠に基づいた機能性の表示等を挙げることができ、より具体的には、健康増進法に規定する特別用途表示の許可等に関する内閣府令(平成二十一年八月三十一年内閣府令第五十七号)に定められた特定保健用食品としての表示(特に保健の用途の表示)及びこれに類する表示が典型的な例である。
かかる表示としては、例えば、「食後の血糖値上昇を緩やかにする」、「食後の血糖値上昇を抑える」、「食後の血糖値が気になる方へ」、「高めの血糖値を下げる」、「高血糖の改善のために」、「血管内皮細胞をまもる」、「食欲を抑えたいときに」等と表示することが挙げられる。
以下に実施例を用いて本発明をさらに具体的に説明するが、本発明はこれら実施例に限定されるものではない。
<実施例1>各ペプチドのDPP-IV阻害活性の評価
乳タンパク質のアミノ酸配列に由来する904種のトリペプチドを有機合成し、各ペプチドのDPP-IV阻害活性を評価した。
DPP-IV阻害活性の測定は、ウチダらの方法(Uchida, M. et al., J. Pharmacol. Sci. 117, 63-66, 2011)に準じて行った。すなわち、酵素(DPP-IV)として液体コントロール血清IワコーC&C(富士フィルム和光純薬株式会社製)、基質としてGly-Pro-AMC(Enzo Life Sciences社製)を用いた。384ウェルブラックプレート(nunc社製)の各ウェルに、液体コントロール血清IワコーC&Cを50mM Tris-HClで25倍希釈したものを5μL、100μMペプチド溶液を5μL添加し、37℃で10分間インキュベートした。各ウェルに10μM Gly-Pro-AMCを10μLずつ添加して混合後、37℃で30分間インキュベートした。蛍光強度値の測定にはマイクロプレートリーダー(SH-9000, コロナ電気株式会社製)を用いた(Ex.380nm/Em.460nm)。

904種のトリペプチドについてDPP-IVに対する阻害率ごとに分けた数の割合を図1に示す。阻害率20%未満のペプチドが89%、阻害率20%以上50%未満のペプチドが8%、阻害率50%以上のペプチドが3%であった。すなわち、DPP-IV阻害率の高いトリペプチドは、ごく一部の特定のアミノ酸配列を持つものだけであった。
<実施例2>DPP-IV阻害活性を有するトリペプチドのIC50の評価
実施例1においてDPP-IV阻害率が50%以上であったトリペプチドのDPP-IV阻害活性をIC50値で評価した。
DPP-IV阻害活性評価はカトウらの方法(Kato , T. et al., Biochem. Med. 19, p351, 1978)に準じて行った。すなわち、酵素(DPP-IV)としてRecombinant human DPPIV(Enzo Life Sciences社製)、基質としてGly-Pro-AMC(Enzo Life Sciences社製)を用いた。384ウェルブラックプレート(nunc社製)の各ウェルにDPP-IV溶液(10.4 μU/μL)を5μL、各濃度のペプチド溶液を5μL添加し、37℃で10分間インキュベートした。各ウェルに10μM Gly-Pro-AMCを10μLずつ添加して混合後、37℃で15分間インキュベートした。蛍光強度値の測定にはマイクロプレートリーダー(SH-9000, コロナ電気株式会社製)を用いた(Ex.380nm/Em.460nm)。
阻害率は下記の式で算出した。

IC50算出のため、ペプチド濃度を段階的に希釈し、それぞれの阻害率を求め、その結果を基にペプチド濃度と阻害率の関係式を求めた。この関係式を用いて50%阻害率となるペプチド濃度を算出し、IC50とした。
実施例1においてDPP-IV阻害率が50%以上であったトリペプチドのDPP-IV阻害活性をIC50値で評価した。

IC50算出のため、ペプチド濃度を段階的に希釈し、それぞれの阻害率を求め、その結果を基にペプチド濃度と阻害率の関係式を求めた。この関係式を用いて50%阻害率となるペプチド濃度を算出し、IC50とした。
結果を表1に、評価したトリペプチドのうち、DPP-IV阻害活性を有することが知られていない新規配列からなるトリペプチドのIC50を示す。LPVペプチド、IPTペプチド、PPQペプチド、APFペプチド、PPLペプチド、PPFペプチド、APSペプチドが新規のDPP-IV阻害ペプチドとして得られた。また、実施例1においてDPP-IV阻害率が20%未満であったトリペプチドから選出した4種類のトリペプチド(Ile-Pro-Pro、Val-Pro-Pro、Val-Glu-Pro、Val-Ala-Pro)については、いずれもIC50が1000μMより大きく、DPP-IV阻害活性が低かった。
<実施例3>LPV配列を含むテトラペプチドのIC50評価
実施例2においてIC50値が最も低かったLPVペプチドの配列を含むテトラペプチド(LPVPペプチド)のDPP-IV阻害活性をIC50値で評価した。
DPP-IV阻害活性評価は実施例2と同様の方法で行った。
実施例2においてIC50値が最も低かったLPVペプチドの配列を含むテトラペプチド(LPVPペプチド)のDPP-IV阻害活性をIC50値で評価した。
DPP-IV阻害活性評価は実施例2と同様の方法で行った。
表2に評価したLPVPペプチドのIC50値を示す。LPVPペプチドはLPVペプチドに比べてIC50値が低いことから、高いDPP-IV阻害活性をもつ新規のペプチドである。
<実施例4>カゼイン加水分解物のDPP-IV阻害活性及び食後血糖値上昇抑制作用の評価
(1)本発明のオリゴペプチドを含むカゼイン加水分解物の取得
市販のカゼインナトリウム125 gを875 gの精製水に溶解した。該カゼインナトリウム水溶液を85℃で10分間加熱殺菌し、50℃に温度調整し、水酸化ナトリウムを添加してpHを7.0に調整した。タンパク質分解酵素1.0、2.5、又は4.0重量%を添加して加水分解をし、本発明のオリゴペプチドを含む溶液3種類を得た。その後、酵素を失活させ、常法により濃縮し、乾燥し、カゼイン加水分解物A、B、Cを得た。
(1)本発明のオリゴペプチドを含むカゼイン加水分解物の取得
市販のカゼインナトリウム125 gを875 gの精製水に溶解した。該カゼインナトリウム水溶液を85℃で10分間加熱殺菌し、50℃に温度調整し、水酸化ナトリウムを添加してpHを7.0に調整した。タンパク質分解酵素1.0、2.5、又は4.0重量%を添加して加水分解をし、本発明のオリゴペプチドを含む溶液3種類を得た。その後、酵素を失活させ、常法により濃縮し、乾燥し、カゼイン加水分解物A、B、Cを得た。
(2)カゼイン加水分解物中の本発明のオリゴペプチド含量の測定
以下の手順で、カゼイン加水分解物における本発明のトリペプチド5種(LPVペプチド、IPTペプチド、PPQペプチド、APFペプチド、PPLペプチド)及びテトラペプチド(LPVPペプチド)の各含量を測定した。
(1)で取得したカゼイン加水分解物A、B及びCをそれぞれ、1.0mg/mLとなるように、超純水で溶解し、10分間超音波破砕したのち、0.22μm口径のPVDFフィルター(Millipore社製)でろ過して溶液を調製した。また、各種オリゴペプチドの化学合成標準ペプチド(フナコシ社製)の溶解液を0.01、0.1、及び1.0 μg/mLの濃度で調製し、検量線用の標準溶液を作成した。以上のペプチド調製液と標準溶液を、下記測定条件でLC/MS分析し定量した。なお、前記ペプチド調製液のMS2ピークのうち、標準ペプチドと分子量及びリテンションタイムが一致するものを、標準ペプチドと同一の配列として同定した。定量値は、MS2ピークの面積値を検量線と対比することで算出した。
以下の手順で、カゼイン加水分解物における本発明のトリペプチド5種(LPVペプチド、IPTペプチド、PPQペプチド、APFペプチド、PPLペプチド)及びテトラペプチド(LPVPペプチド)の各含量を測定した。

使用機器
・質量分析計:TSQ Quantum Discovery MAX(サーモフィッシャーサイエンティフィック社製)。
・HPLC構成:システムコントローラーCBM-20A、オートインジェクターSIL-20AC、送液ポンプLC-20AD×2、カラムオーブンCTO-20AC、UV/VIS検出器SPD-20A、オンラインデガッサDGU-20A5 (以上、島津製作所社製)
・カラム:XBridge Peptide BEH C18 Column, 300Å(2.1mm×250mm)(Waters社製)。
測定条件(HPLC)
・移動相A:0.2重量% 蟻酸-水溶液、移動相B:0.2重量% 蟻酸-アセトニトリル溶液。
・タイムプログラム:2%B (0分) - 40%B (12分) - 80%B (15分) - 80%B (17%) - 2%B (17分) - STOP (30分)。
・試料注入量:10 μL、液体流量:200 μL / min、カラム温度:40℃
測定条件(質量分析計)
・イオン化モード: ESI(+)、スプレー電圧:+4.0kV、キャピラリー温度:300℃、シースガス圧:30Arb、補助ガス圧:10Arb
・分析モード:SRM
・質量分析計:TSQ Quantum Discovery MAX(サーモフィッシャーサイエンティフィック社製)。
・HPLC構成:システムコントローラーCBM-20A、オートインジェクターSIL-20AC、送液ポンプLC-20AD×2、カラムオーブンCTO-20AC、UV/VIS検出器SPD-20A、オンラインデガッサDGU-20A5 (以上、島津製作所社製)

・分析モード:SRM
各カゼイン加水分解物中に含まれる本発明のオリゴペプチド量の測定結果を表3に示す。
(3)カゼイン加水分解物のDPP-IV阻害に対するIC50の評価
前述の実施例2と同様の方法で、カゼイン加水分解物A、B及びC並びにカゼインそれぞれのIC50を評価した。
結果を表4に示す。カゼインと比較し、DPP-IV阻害効果が高かった。

(4)カゼイン加水分解物による食後血糖値上昇抑制試験
被験者に、水100mLにカゼイン加水分解物B 1gとデキストリン 5gを溶解した試験溶液、又は水100mLにデキストリン 5gを溶解した対照溶液を摂取させ、摂取5分後に米飯200gを摂取させた。米飯摂取から0、15、30、45、60、90、及び120分経過後にメディセーフファインタッチディスポ(テルモ(株))を用いて指先から採血し、ワンタッチベリオビュー(ジョンソン・エンド・ジョンソン(株))を用いて血糖値を測定した。
米飯摂取前(0分)に対する血糖値変化量について、被験者2名のデータを平均した結果を図2に示す。カゼイン加水分解物を含む試験溶液を摂取した場合には、対照溶液を摂取した場合と比較して、血糖値変化量が低く推移した。

- 以下の(a)~(g)に記載のいずれかの配列を含むオリゴペプチド。
(a)Leu-Pro-Val
(b)Ile-Pro-Thr
(c)Pro-Pro-Gln
(d)Ala-Pro-Phe
(e)Pro-Pro-Leu
(f)Pro-Pro-Phe
(g)Ala-Pro-Ser - (h)Leu-Pro-Val-Proを含む、請求項1に記載のオリゴペプチド。
- 請求項1又は2に記載のオリゴペプチドから選択される一種又は二種以上のオリゴペプチド含む、乳タンパク質加水分解物。
- 請求項1又は2に記載のオリゴペプチドから選択される一種又は二種以上のオリゴペプチドを有効成分として含有する、ジペプチジルペプチダーゼ-IV(DPP-IV)阻害剤。
- 請求項1又は2に記載のオリゴペプチドから選択される一種又は二種以上のオリゴペプチドを含む乳タンパク質加水分解物を含有する、DPP-IV阻害剤。
- 請求項4又は5に記載のDPP-IV阻害剤を含有する医薬組成物。
- 請求項1又は2に記載のオリゴペプチドから選択される一種又は二種以上のオリゴペプチドを有効成分として含有する、食後血糖値上昇抑制用組成物。
- 請求項1又は2に記載のオリゴペプチドから選択される一種又は二種以上のオリゴペプチドを含む乳タンパク質加水分解物を含有する、食後血糖値上昇抑制用組成物。
- 飲食品である、請求項7又は8に記載の組成物。
