본 발명은 세레노아 레펜스(Serenoa repens)와 비티스 비니페라(Vitis vinifera)의 추출물들을 활성성분으로 함유한, 두피장해 치료 및 예방을 위한 약제 및/또는 화장품 조성물에 관한 것이다.

Description
본 발명은 식물기원성분을 함유하는, 두피장해 예방 및 치료용 약제 및/또는 화장품의 경구조성물에 관한 것이다.
더욱 특별히는, 본 발명은 활성성분으로서 세레노아 레펜스(Serenoa repens)와 비티스 비니페라(Vitis vinifera)의 추출물들을 함유한 두피장해 예방 및 치료용 약제 및/또는 화장품의 경구 조성물에 관한 것이다.
비듬(dandruff), 지루(seborrhea) 및 머리카락 손실(hair loss) 또는 원형탈모증(alopecia)이 두피장해시 가장 흔한 질병에 속한다.
많은 연구에서 남성 원형탈모증(androgenetic alopecia)은 유전적으로 사전 소인이 있는 개개인의 생리학적 진행이며, 비록 매우 높은 빈도, 특히 백인중심으로, 유전성 형태의 난해를 보이려한다. 이와 같은 유전성이 비록 강한 상염색체성 일지라도 포함된 유전자수는 아직 확인된바 없다.
안드로겐(androgens)과 남성 원형탈모증의 발전과의 관계는 규명되어 있다 : 예를들면, 대머리(baldness) 유형은 머리카락 조직재생(hair anagen) 성장단계에서 환원시간(reduced time)과 관련이 있으며, 안드로겐(androgens)은 두피 머리카락 소포(follicles) 중에서 더 짧은 조직재생 성장단계를 야기시키는 것으로 알려져있고, 더욱 얇고(finer) 더욱 가늘게(thinner) 된다. 조직 안드로겐, 테스토스테론 및 더 강력한 디하이드로테스토스테론(DHT, dihydrotestosterone)은 혈액순환을 통해서 피부에 도달할 수 있으며, 또는 스테로이드 분지중 특이한 효소에 의해서 머리카락 소포(hair follicle)와 피지선(sebaceous glands) 중에서 국소적으로 생산 될 수 있다.
남성 원형탈모증으로 영향받는 남성과 여성의 두피로부터 인간 머리카락의 머리카락 소포와 피지선 중에 포함되어있는 5-알파-환원효소, DHT의 형성을 중재하는 많은 효소의 동력상수(Kinetic constants)라 평가되었고; 나아가 DHT와 특이적으로 결합하는 안드로겐 수용체를 지방세포(sebocytes)와 인간 머리카락에서 확인된바있다.
최근, 결합연구는 대머리 소지가 있는 머리카락 소포의 피부유두(dermal papilla)는 정상적 소지가 있는 것보다 더 많은 안드로겐 수용체를 포함한다는 점을 보여주고있다. 홀몬경로로서, 비정상 피지(sebum)분비가 일어나고, 나아가 대머리(baldness)를 더욱 나쁘게 발전시킴은 물론 피지와 지루의 과잉생산을 일으킨다.
5-알파 환원효소(5-alpha-reductase)의 두가지 이성체로는 타입1과 타입2가 알려져있다. 전립선(prostate)에는 타입2 이소엔자임이 포함되며, 반면 피부(skin)와 피부부속기(cutaneou appendages)에는 타입1과 타입2가 포함되어있다. 5-알파-환원효소억제제의 원조격인 피나스테라이드(finasteride)는 전립선비대 치료에 사용되고, 또한 남성 원형탈모증 치료에도 활성을 보인다; 나아가, 대머리 심각성과 전립선 비대심각성과의 사이에 관계를 관찰하였다.
5-알파-환원효소에 추가하여 또한 산화 스트레스(oxidative stress)(오염 -pollution; 기압제등 - atmospheric agent)과 다이어트를 통한 올리고 성분 (oligoelements)와 설페이티드 아미노산(메티오닌, 시스테인 및 시스틴등)의 열악한 섭취는 머리카락에 역으로 영향을 준다.
현재 비듬 및 원형탈모증의 치료용 약제학적제제 또는 화장품제제로 상품화된 대다수가 유용성면에서 이와같은 문제점을 해결하는데 만족스럽지 못하였다.
본 발명의 목적은 이와같은 점에 착안한 것으로, 약제학적 및/또는 화장품 경구제제는 두피장해 특히 원형탈모증 및 비듬치료시 탁월한 효과를 주는 식물기원의 활성성분들의 조합물을 포함하며, 여러가지 성분의 상이한 효능의 조합결과로서, 즉 그 중에서도 특히 항안드로겐성 효능(antiandrogenic), 항라디칼 효능 (antiradicalic), 항경시 효능(antiaging activities)을 발휘한다.
본 발명의 조성물은 두피장해 발전에 기여하는 요소에 작용하며, 특히, 다이어트중에 존재하는 안드로겐, 산화스트레스, 올리고 성분 및 설페이티드 아미노산에 작용한다.
더욱 특별하게는 본 발명은 하기성분을 포함하는 경구약제 및/또는 화장품 조성물에 관한 것으로;
a) 세레노아 레펜스(Serenoa repens)의 추출물,
b) 비티스 비니페라(Vitis vinifera)의 표준 추출물로, 유리형태 및/또는 인지질 복합체를 포함한다.
본 발명의 조성물중 성분은 모두 알려져 있으며 약제 및/또는 화장품 분야에서 사용되고 있다. 그러나 단일성분으로 알려져 있으며 개별적으로 분리되어 사용시 본 발명의 조성으로 얻을 수 있는 효과보다는 매우 저조한 효능을 발휘하고 있을 뿐이며, 많은 성분들이 두피장해의 예방 및 치료시 상승작용(synergistic effect)을 발휘됨이 밝혀졌다.
a) 세레노아 레펜스(Serenoa repens)의 추출물은 항안드로겐 적용때문에 양성 전립선 비대증에 식물성 치료효과가 있다. 이 제품은 EP특허 제250,953호에 개시되어 있는 바와같이, 초임계조건에서 탄산가스(CO2)에 의하여 식물로부터 추출된 지방산(fatty acids)의 특정혼합물(specific mixture)을 포함하며, 전립선 분리세포에 대한 생체외 실험에서 안드로겐 수용체에 강한 영향력을 발휘하였으며, 방사선 표지 3H-메틸트리에놀론으로 치환시켜 나타냈다.
b) GB특허 제1,541,469호에 기재된 포도, 비티스 비니페라(Vitis vinifera)의 추출물은 갈릭산(gallic acid)과 함께 카테친(catechin)과 에피카테친 (epicatechin)의 모노머, 다이머, 트리머, 테트라머, 펜타머, 헥사머 및 헵타머로 유리형태로 포함되거나 또는 갈레이트(gallates)와 같이 에스텔화 된 것을 포함한다.
광범위한 연구로 많은 성질이 규명되었다: a) 자유 라디칼 효능과 관련한 모든 현상과 반대로 작용하므로 더많은 반응성 라디칼을 제거가능 하도록 하는 강하고, 안전한 항산화제 푸로파일; b) 잔틴-옥시다제(xanthine-oxidase)를 억제하고, Cu++와 Fe++를 킬레이트 형성하며, 나아가 조직내로의 자유 라디칼의 효소적 방출을 예방하는 능력; c) 콜라겐나제(collagenase), 히알유로니다제 (hyaluronidase), 에라스타제(elastase) 및 베타-글루쿨로니다제(beta-glucuronidase)를 억제하고, UV선, 산화스트레스 및 염증반응발전후 방출되는 단백질 분해효소에 의하여 일으키는 손상에 대하여 혈관 및 연질조직을 보호하는 능력등이다.
위에서 언급한 바와같이, 포도인 비티스 비니페라(Vitis vinifera)의 추출물은 또한 US특허 제4,963,527호에 기재된 바와같이 인지질복합체의 형태로 또한 존재할 수 있다.
본 발명의 조성물은 임의로, 부가적으로 위에서 언급된 성분들 중에 유용하고 보충작용을 갖는 추가성분을 함유할 수 있으며, 예를들면 아연, 철, 세레늄, 마그네슘과 같은 올리고 성분; L-리진, L-푸로린, L-하이드록시푸로린, L-루신, L-이소루신, L-메티오닌, L-시스테인, L-시스틴과 같은 아미노산; 비타민 B 콤푸렉스, 비타민 E 및 비타민 C와 같은 비타민류가 함유될 수 있다.
본 발명의 조성물은 관용의 기술에 따라 경구용량형태(oral dosage forms)로 제형화 될 수 있으며, 예를들면 "Remington : The Science and Practice of Pharmacy", 리핀코트, 윌리암스와 윌킨스편집, 12.(2000)에 기재되어 있는 기술을 응용할 수 있다.
본 발명의 조성물은 정제, 캅셀, 경구제제, 분말, 과립, 로젠즈, 재구성용 분말, 주사액, 또는 현탁액, 및 주입액(liquids for infusions) 또는 좌제의 형태로 제제화 할 수 있다.
경구투여를 위한 정제와 캅셀제는 유니트용량의 형태로 통상 존재할 수 있으며, 결합제, 희석제, 정제화제, 윤활제, 붕해제, 염료, 향료 및 습윤제와 같은 관용의 부형제를 포함할 수 있다. 정제는 이 기술분야에서 잘 알려진 방법에 따라 피복될 수 있다.
본 발명의 태양에 따르면, 조성물은 동시투여를 위하여 2개의 캅셀제 형태로 존재할 수 있으며, 하나는 발명의 추출물들을 포함하고, 다른 하나는 위에서 언급한 올리고 성분(oligoelements)을 포함하는 것이다.
경구액상제제는, 예를들면, 수용성 또는 기름성 용액 또는 현탁제, 유제, 시럽 또는 에릭서 형태로 존재할 수 있으며, 또는 물 또는 사용전 다른 적당한 담체와 재구성용 분말일 수 있다.
이들 액상제제는 현탁화제, 유화제, 비수용성 담체, 보전제, 향료 또는 염료와 같은 관용의 부형제를 포함할 수 있다.
본 발명의 조성물은 다음과 같은 용량범위내에서 매일 섭취하는 성분을 제공할 수 있도록 그런 용량형태로 사용될 수 있다. :
a) 세레노아 레펜스(Serenoa repens) 표준 추출물(40 - 320 mg/일);
b) 유리형태 및/또는 인지질 복합체로의 비티스 비니페라(Vitis vinefera)의 표준 추출물(50 - 300 mg/일과 150 - 900 mg/일, 각각)
올리고 성분(Oligoelements)는 매일 섭취량이 0.1 ~ 100 mg이 제공되도록 이 와같은 량으로 존재할 수 있다.
본 발명의 조성물은 두피장해 치료시 효능을 발휘하고, 모발그램 (trichogram), 비듬(dandruff), 지루(seborrhea) 및 대머리(baldness)에 유익한 효능을 갖으며, 두피장해의 예방시 유효하며, 건강한 머리카락을 보장한다.
약리실험에 대한 결과는 다음에 기재하였다.
비듬(dandruff), 피지(Sebum) 생산과 머리카락 손실에 대한 효과
비듬(두피피부를 벗김)을 갖는 60명의 환자를 4개군으로 나누었다.
첫번째군은 8주동안 매일 실시예 1에 따라 제조된 1캅셀을 주었다. 평가는 치료전 즉시, 치료말기(8주후) 및 치료방해후 4주(재주사)에 수행하였다. 두번째군은 동일실험조건하에서 위약(placebo)을 주었다. 세번째와 네번째군은 각각 비티스 비니페라(Vitis vinifera)와 세레노아 레펜스(Serenoa repens) 추출물 25 mg과 80 mg을 주었다.
결과를 하기 표1에 기재하였는바, 치료 8주후 표피가 벗겨진 세포 (desquamated cells)의 수가(맥 긴리(Mac Ginley)등의 J.Invest. Dermatol. 53, 107,1969에 따라 평가) 85 세포/㎠에서 18 세포/㎠으로 감소되었음이 인정된다. 표피가 벗겨진 세포의 수는 4주(재조사)후에 현저히 감소되었다(21 세포/㎠).

하기 표2에 기재된 결과는 본 발명에 따른 캅셀로 치료후 두피피지의 평균값이 105에서 92 U.S.(U.S. = arbitrary Sebometric Units, 인위 피지측정 유니트)로 현저한 감소가 보였음을 입증하고 있다. 특히 현저한 것은 피지측정 유니트의 값이 치료말기 4주후 현저히 감소되었음을 보인다.(95 U.S.)

머리카락 손실에 대한 효과를 트리코그램(trichogram) 평가에 의해 조사되었 다. 각개인의 더높고 안테로-누칼 전방지역(antero-nuclal frontal areas)으로부터 머리카락(약50개)의 충분한 수를 택하여 구성하였다(보스 케이, 하우즈아트(Bosse K., Hautzart, 18, 35, 1967; Bosse K., Hautzart. 18, 218, 1967). 조직재생(anagen)(성장), 퇴축(catagen)(성숙), 또는 종국(telogen)(휴식)상태에서 머리카락의 백분율을 현미경하에서 각 단일 모간(hair shaft)을 현미경 관찰로 평가되었다.
발육이상 조직재생조건에서, 주로 소형화된 모간을 갖는 상태에서 본 연구를 평가하였다. 종국상태에서 머리카락 백분율이 10~15 %(정상고려)보다 더 높은것이 머리카락 손실의 임상병적조건의 수치로 하였다. 표3에 실험결과를 기재하였는바, 본 발명의 캅셀제로 치료 8주후 조직재생 상태에서 모근(hair bulb)에서 증가를 보였고, 발육이상 조직재생 머리카락 수치가 감소했으며, 결과적으로, 종국상태에서 모근(bulb)의 감소가 관찰되었다.
이 결과는 재조사 4주후에도 계속 볼 수 있었다. 위약군에서는 치료말기 및 재조사 4주후 모두 임상상태가 대립적으로 반대였다.
표 3 - 머리카락 손실에 관한 효과
지루성 피부염(Seborrheic dermatitis)에 관한 효과
두피에 지루성 피부염을 나타내는 40명의 환자를 2군으로 나누었다. 첫번째군은 8주동안 실시예 1에 따라 제조된 캅셀을 주었다. 기계적 지루평가를 치료개시전 즉시, 치료말기(8주후), 및 치료(재조사) 4주후 현탁액을 가하고 수행하였다. 두번째군은 동일한 실험조건하 위약을 주었다. 3번째와 4번째군은 비티스 비니페라 및 세레노아 레펜스 각각을 25 및 80 mg 주었다.
결과를 표4에 기재하였으며, 본 발명의 캅셀치료군에서 피지값이 200 U.S.(U.S.=arbitrary Sebometric Units, 인위 피지측정 유니트)이상인 지루성 피부염을 갖는 환자에서 두피피지 평균값을 현저히 감소시켰다. 이 수치는 재조사 4주후에도 여전히 현저히 낮았다.

[발명의 효과]
본 발명에 따르면, 세레노아 레펜스(Serenoa repens)와 비티스 비니페라 (Vitis vinifera)의 추출물을 활성성분으로 함유한 두피장해의 치료와 예방에 탁월한 효과가 있는 약제학적 조성물 및/또는 화장품 조성물을 얻을 수 있다는 특장점이 있다.
Claims (11)
- 하기 유효성분으로 구성된 두피장해 개선용 화장품 조성물.(하 기)(a) 세레노아 레펜스(Serenoa repens) 추출물,(b) 비티스 비니페라(Vitis vinifera) 추출물의 유리형태 및/또는 인지질 복합체.
- 청구항 1에 있어서, 하기 유효성분 외에 추가로 징크(아연), 커퍼(동), 철, 셀레니움, 마그네슘 중에서 선택된 1종 또는 그 이상의 올리고 성분; L-리진, L-하이드록시푸로린, L-루신, L-이소루신, L-메치오닌, L-시스테인, L-시스틴 중에서 선택된 1종 또는 그 이상의 아미노산; 비타민 B 콤푸렉스, 비타민 E 또는 비타민 C 중에서 선택된 1종 또는 그 이상의 비타민류(Vitamins)로 구성된 화장품 조성물.(하 기)(a) 세레노아 레펜스(Serenoa repens) 추출물,(b) 비티스 비니페라(Vitis vinifera) 추출물의 유리형태 및/또는 인지질 복합체.
