Claims (13)
- 페스티바이러스의 자가 단백질 분해효소 Npro의 자가 단백질 분해 기능이 있는 첫 번째 폴리펩티드, 및 이 첫 번째 폴리펩티드의 자가 단백질 분해성 활성에 의해 융합 단백질로부터 절단될 수 있도록 첫 번째 폴리펩티드 C-말단에서 첫 번째 폴리펩티드와 연결된 이종 폴리펩티드인 두번째 폴리펩티드를 포함하는 융합 단백질을 코딩하는 핵산 분자.
- 제1항에 있어서, 상기 페스티바이러스는 돼지 콜레라 바이러스 (CSFV), 국경병 바이러스 (BDV) 및 소 바이러스성 설사 바이러스 (BVDV)의 군으로부터 선택된 것인 핵산 분자.
- 제2항에 있어서, 상기 페스티바이러스는 CSFV인 핵산 분자.
- 제3항에 있어서, 첫 번째 폴리펩티드가 하기 아미노산 서열 또는 자가 단백질 분해 활성을 가진 그의 유도체의 아미노산 서열을 포함하는 것인 핵산 분자.(1)-MELNHFELLYKTSKQKPVGVEEPVYDTAGRPLFGNPSEVHPQSTLKLPHDRGRGDIRTTLRDLPRKGDCRSGNHLGPVSGIYIKPGPVYYQDYTGPVYHRAPLEFFDEAQFCEVTKRIGRVTGSDGKLYHIYVCVDGCILLKLAKRGTPRTLKWIRNFTNCPLWVTSC-(168)
- 제3항에 있어서, 첫 번째 폴리펩티드는 CSFV의 자가 단백질 분해효소 Npro 또는 자가 단백질 분해 활성을 지닌 그의 유도체의 Glu22 내지 Cys168 아미노산 서열을 포함하고 추가로 N-말단에 Met를 가지며, 이종 폴리펩티드는 CSFV의 자가 단백질 분해효소 Npro의 Cys168 아미노산에 직접적으로 연결되어 있는 것인 핵산 분자.
- 제3항에 있어서, 첫 번째 폴리펩티드는 CSFV의 자가 단백질 분해효소 Npro 또는 자가 단백질 분해 활성을 지닌 그의 유도체의 Pro17 내지 Cys168 아미노산 서열을 포함하고 추가로 N-말단에 Met를 가지며, 이종 폴리펩티드는 CSFV의 자가 단백질 분해효소 Npro의 Cys168 아미노산에 직접적으로 연결되어 있는 것인 핵산 분자.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 핵산 분자가 DNA 분자인 핵산 분자.
- 제1항 내지 제6항 중 어느 한 항에 따른 핵산 분자 및 하나 이상의 발현 조절 서열을 포함하며, 미리 정해진 박테리아 숙주 세포에 적합한 발현 벡터.
- 제8항에 있어서, 상기 박테리아 숙주 세포가 이. 콜라이 (E. coli) 세포인 발현 벡터.
- 제8항에 있어서, 상기 발현 벡터가 플라스미드인 발현 벡터.
- 제8항에 따른 벡터를 포함하는 박테리아 숙주 세포.
- 제11항에 있어서, 상기 숙주 세포가 이. 콜라이 세포인 박테리아 숙주 세포.
- (i) 제11항 또는 제12항에 따른 박테리아 숙주 세포를, 융합 단백질이 발현되며, 숙주 세포 내에서 첫 번째 폴리펩티드의 자가 단백질 분해 활성에 의해 상기 융합 단백질로부터 이종 폴리펩티드의 자가 단백질 분해성 절단이 일어나는 조건 하에서 배양하는 단계, 및(ii) 절단된 이종 폴리펩티드를 단리하는 단계를 포함하는 목적 이종 폴리펩티드의 생산 방법.
