Abstract

Description
본 발명은 면역글로불린 계통군에 속하지 않는 사람 단백질 또는 이의 일부, 및 면역글로불린 분자의 불변부의 다양한 부분으로 이루어지는, 유전공학적으로 처리된 가용성 융합 단백질에 관한 것이다. 놀랍게도, 2개의 융합 파트너의 작용성 특성이 융합 단백질내에 보유된다.
유럽 공개특허공보 제0 325 262호 및 제0 314 317호에는 사람 T 세포의 CD4 막 단백질의 다양한 도메인(domain) 및 사람 IgG1 부분으로 이루어진 이에 상응하는 융합 단백질이 기술되어 있다. 이들 융합 단백질중 몇몇은 세포-결합된 CD4 분자와 동일한 친화도로 사람 면역결핍 바이러스의 당단백질 gp120에 결합한다. CD4 분자는 면역글로불린 계통군에 속하므로, 결과적으로 면역글로불린 분자의 구조와 매우 유사한 3차 구조를 갖는다. 이러한 유사한 3차 구조는 또한 T-세포 항원 수용체의 α-쇄에 대해서도 적용되는데, 이러한 융합이 또한 다음 문헌에 기술되어 있다[참조: Gascoigne 등, Proc. Natl. Acad. Sci. 본 발명은 면역글로불린 계통군에 속하지 않는 사람 단백질 또는 이의 일부, 및 여러가지 아류의 면역글로불린 (IgG, IgM, IgA, IgE)의 중쇄 또는 경쇄의 불변부의 다양한 부분으로 이루어진, 유전 공학적으로 처리된 가용성 융합 단백질을 제공하는 것을 목적으로 한다. 사람 IgG, 특히 바람직하게는 사람 IgG1의 중쇄의 불변부가 면역글로불린으로서 바람직하며, 여기서 융합은 힌지영역(hinge region)에서 발생한다. 특정 양태에서, Fc 부분은, 또한 혼입되는 절단 서열에 의해 간단한 방법으로 제거될 수 있고 인자 Xa를 사용하여 절단될 수 있다.
또한, 본 발명은 유전공학적으로 상기 융합 단백질을 제조하는 방법, 및 진단과 치료를 위한 이의 용도에 관한 것이다.

융합 단백질은 공지된 원핵 및 진핵 발현 시스템에서 제조될 수 있지만, 바람직하게는 포유동물 세포(예: CHO, COS 및 BHK 세포)에서 제조될 수 있다.
본 발명에 따른 융합 단백질은 그들의 면역글로불린 부분 때문에, 친화 크로마토그라피에 의해 정제하기가 용이하며 생체내에서 개선된 약물동력학적 특성을 갖는다.
많은 경우에서, 융합 단백질에서의 Fc 부분은 치료 및 진단에서 사용하기에 상당히 유리하므로, 예를 들어, 개선된 약물동력학 특성을 초래한다[참조: 유럽 공개특허공보 제0 232 262호]. 한편, 어떤 용도를 위해서는, 융합 단백질을 발현시키고 탐침하고 정제한 후에 기술된 유리한 방법으로 Fc 부분을 제거하는 것이 바람직할 것이다. 이것은 Fc 부분이 치료 및 진단에서 사용하기에는 방해물임이 입증되는 경우, 예를 들어, 융합 단백질이 면역반응에서 항원으로서 사용되는 경우에 해당된다.
상기 목적을 위해 사용하는 것이 가능할 수 있는, 현존의 다양한 프로테아제가 있다. 예를 들어, 면역글로불린 으로부터 F(ab) 단편을 생성시키기 위해 파파인 및 펩신이 사용되지만[참조: Immunology, ed. Roitt, I. 등, Gower Medical Publishing, London(1989)], 이들은 특정하게 특이적 방식으로 절단하지는 않는다. 대조적으로 응혈인자 Xa는 단백질에서 비교적 희귀한 테트라펩티드 서열 Ile-Glu-Gly-Arg을 인지하고 아르기닌 잔기 다음에 단백질의 가수분해적 절단을 수행한다. 언급된 테트라펩티드를 함유하는 서열은 나가이(Nagai) 및 토게르센(Thogersen)에 의해 유전 공학적 방법으로 하이브리드 단백질에 최초로 도입되었다[참조: Nagai,
K. 및 Thogersen, H.C., Nature, vol. 309(1984), 810-812]. 상기 저자들은 이.콜라이에서 발현된 단백질이 실제적으로 인자 Xa 에 의해 특이적으로 절단되는 것을 보여줄 수 있었다. 하지만, 이러한 단백질이 또한 진핵세포, 특히, 동물세포에서 발현되어 정제 후에 인자 Xa에 의해 절단되는 가능성에 대해서는 아직까지 공개된 예가 없다. 그러나, 동물 세포에서의 본 발명에 따른 단백질의 발현이 바람직한데, 이는 상기 타입의 세포 시스템에서만, 예를 들어, 정상적으로, 천연구조를 보유하므로 생물학적 활성을 보유한 융합 파트너로서의 막-결합 수용체의 분비가 예상되기 때문이다. 세포 배양 상층액으로의 분비는 융합 단백질의 후속의 간단한 정제를 촉진시켜준다.

실시예 1: 트롬보플라스틴 융합 단백질
응혈은 인체에서 가장 중요한 과정이다. 응혈 케스케이드(cascade)의 적당하게 예민한 조절이 있는데, 여기에는 다수의 세포성 인자와 혈장 단백질이 함께 작용한다. 전체로서의 상기 단백질(및 이의 보조인자들)이 응혈인자로 불리워진다. 응혈 케스케이드 최종 생성물은 혈소판의 응집을 유발시키는 트롬빈, 및 혈소판 혈전을 안정화시키는 피브린이다. 트롬빈은 피브리노겐으로부터의 피브린의 형성을 촉매하며 스스로는 프로트롬빈의 제한된 단백질 분해에 의해 형성된다. 활성화된 인자 X(인자 Xa)는 상기 단계에 관여하여, 인자 Va 및 칼슘 이온의 존재하에서, 혈소판 막에 결합하여 프로트롬빈을 절단시킨다.
인자 X가 활성화되기 위해서는 2가지 경로, 즉, 외성 경로와 내성 경로가 존재한다. 내성 경로에서는, 일련의 인자들은 단백질분해에 의해 활성화되어 그들 각각이 활성 프로테아제를 형성한다. 외성 경로에서는, 손상된 세포에 의해 트롬보플라스틴 (조직 인자)의 합성이 증가되고, 증가된 트롬보플라스틴이 인자 VIIa 및 칼슘 이온과 함께 인자 X를 활성화시킨다. 트롬보플라스틴의 활성은 상기 반응에만 한정된다고 앞서 가정되었었다. 그러나, 트롬보플라스틴/VIIa 복합체는 또한 인자 IX의 수준에서 내성 경로를 활성화시키는 것을 방해한다. 따라서, 트롬보플라스틴/VIIa 복합체는 응혈의 가장 중요한 생리학적 활성제중의 하나이다.
그러므로, 트롬보플라스틴은 이의 진단 보조제(하기 참조)로서의 용도와는 별도로, 선천성 또는 후천성 응혈 결핍증을 치료하기 위한 치료학적 약제의 성분으로서 또한 사용될 수 있다고 생각할 수 있다. 이러한 예는 인자 VIII, IX 또는 XI의 결핍에 의해 야기되는 만성 혈우병 또는, 예를 들어, 간 또는 신장 질병의 결과로 초래되는 그 밖의 급성 응혈 장해이다. 외과 수술 개입후의 치료학적 약제로서의 용도를 또한 생각할 수 있다.
트롬보플라스틴은 면역글로불린 계통군에 속하지 않는 온전한 막 단백질이다. 트롬보플라스틴 cDNA 서열은 총 4개의 그룹에 의해 공개되었다[참조: Fisher 등, Thromb. Res., vol. 48(1987), 89-99; Morrisey 등, Cell, vol. 50(1987), 129-135; Scarpati 등, biochemistry, vol. 26(1987), 5234-5238; Spicer 등, Proc. Natl. Acad. Sci. USA, vol. 84(1987), 5148-5152]. 트롬보플라스틴 cDNA는 295개 아미노산 잔기의 폴리펩티드를 암호화하고, 이의 32개의 N-말단 아미노산이
시그날 펩티드로서 작용하는 개방 판독 프레임(open reading frame)을 함유한다. 성숙한 트롬보플라스틴은 263개의 아미노산 잔기를 포함하고 3개-도메인 구조, 즉 i) 아미노 말단 세포외 도메인 (219개의 아미노산 잔기); ii) 막횡단 영역(23개의 아미노산 잔기); iii) 세포질 도메인(카복실 말단; 21개의 아미노산 잔기)를 갖는다. 세포외 도메인에는, N-글리코실화를 위한 3개의 포덴셜 부위가 존재한다(Asn-X-Thr). 트롬보 플라스틴은 정상적으로 글리코실화되지만 글리코실화는 단백질의 활성을 위해 필수적인 것으로 보이지는 않는다[참조: Paborsky 등, Biochemistry, vol. 29(1989), 8072-8077].
트롬보플라스틴은 응혈의 진단상 시험에서 혈장 샘플에 대한 보조제로서 요구된다. 시험받는 환자의 응혈 상태는 1-단계 프로트롬빈 혈병 형성 시간 결정에 의해 알 수 있다(예: Quick's test). 진단상 시험에 요구되는 트롬보플라스틴은 현재 사람 조직으로부터 획득하며, 제조방법은 표준화하기 어렵고, 수율은 낮아서 상당한 양의 사람 출발물질(태반)이 공급 되어야만 한다. 한편, 천연의 막-결합 트롬보플라스틴의 유전 공학에 의한 제조는 복잡한 정제 방법 때문에 또한 어려울 것이라 예상된다. 상기 문제점들은 면역글로불린 부분으로의 본 발명에 따른 융합에 의해 회피될 수 있다.
본 발명에 따른 트롬보플라스틴 융합 단백질은 포유동물 세포(예: CHO, BHK, COS 세포)에 의해 배양 배지로 분비되고 단백질 A-세파로즈상에서의 친화 크로마토그래피로 정제되며 1-단계 프로트롬빈 혈병 형성시간 결정에서 놀랍게도 높은 활성을 갖는다.
트롬보플라스틴 cDNA의 클로닝
공개된 서열[참조: Scarpati 등, Biochemistry, vol. 26(1987), 5234-5238]을 트롬보플라스틴 cDNA를 클로닝하는데 사용한다. 2개의 올리고뉴클레오타이드 프로브 분자(도 1 참조)가 이들 공개된 서열로부터 유도된다. 상기 2개의 프로브 분자는 사람 태반으로부터 cDNA 뱅크를 스크리닝하는 데 사용한다[참조: Grundmann 등, Proc. Natl. Acad. Sci. USA, vol. 83(1986), 8024-8028].
다양한 길이의 cDNA 클론을 수득한다. 후속의 과정에서 사용되는 하나의 클론인 2b-Apr5는 스카르파티 (Scarpati)등에 의해 기술된 cDNA와 동일한 아미노산 서열을 암호화한다.
트롬보플라스틴 융합 단백질을 암호화하는 하이브리드 플라즈미드 pTF1Fc의 작제
플라즈미드 pCD4E 감마 1(유럽 공개특허공보 제0 325 262 A2호; ATCC에 수탁번호 제67610호로 기탁되어 있음)을 사람 CD4 수용체 및 사람 IgG1으로 구성된 융합 단백질을 발현시키는데 사용한다. CD4의 세포외 도메인을 암호화하는 DNA 서열은 제한 효소 Hind III 및 BamHI을 사용하여 상기 플라즈미드로부터 제거한다. 이 경우에, 단지 부분적 절단 만을 효소 HindIII를 사용하여 수행하여, pCD4E 감마 1에 함유되어 있는 2개의 HindIII 부위중 단지 1개(위치 2198)만을 절단해야 한다. 결과로 진핵세포 전사조절서열(프로모터)이 개방 HindIII 부위에 이어지는 개방된 벡터가 초래된다. 개방 BamHI 부위는 펜타-펩티드 링커를 위한 암호화 영역의 출

열안정성 DNA 폴리머라제를 사용한 DAN 증폭은 목적하는 서열이 한쪽 말단 또는 양쪽 말단에서 부착되도록 하는 방식으로 소정의 서열을 변형시킬 수 있다. 트롬보플라스틴 cDNA의 5-비해독 영역(A: 5' GATCGATTAAGCTTCGGAACCCGCTCGATCTCGCCGCC 3') 또는 암호화 영역(B: 5' GCATATCTGGATCCCCGTAGAATATTTCTCTGAATTCCCC 3')에 있는 서열과 하이브리드화 할 수 있는 2개의 올리고뉴클레오타이드를 합성한다. 이들중에서, 올리고뉴클레오타이드 A는 암호화쇄의 서열과 부분적으로 동종성이고 올리고뉴클레오타이드 B는 비-암호화쇄와 부분적으로 동종성이다(도 3 참조).
그리하여, 증폭을 통해(암호화쇄를 기준으로 하여) 암호화 서열의 출발점전의 5' 말단에 있는 HindIII 부위 및 막횡단 영역의 첫번째 3개의 아미노산 잔기를 위한 코돈후의 3' 말단에 있는 BamHI 부위를 함유하는 DNA 단편(827bp)을 수득한다. BamHI 절단 부위에 있는 판독 프레임은 pCD4E 감마 1에 있는 BamHI 부위와의 연결이 트롬보플라스틴 cDNA의 개시 코돈에서 부터 IgG1 중쇄의 정지코돈에 걸쳐 연속적인 판독 프레임과의 유전자 융합을 초래하도록 하는 것이다. 목적하는 단편을 수득하고 HindIII 및 BamHI으로 처리한 후, 상기와 같이 HindIII(부분적으로) 및 BamHI으로 절단시킨 벡터 pCD4E 감마 1으로 연결시킨다. 생성된 플라즈미드를 pTF1Fc라고 명명한다(도 4 참조)
포유동물 세포로의 pTF1Fc의 형질감염
플라즈미드 pTF1Fc에 의해 암호화된 융합 단백질은 이후로 pTF1Fc로 명명한다. pTF1Fc는 COS 세포에서 일시적으로 발현시킨다. 이 목적을 위해, COS 세포를 DEAE-덱스트란의 보조하에 pTF1Fc로 형질감염시킨다[참조: 유럽 공개특허공보 제0 325 262호]. 간접 면역형광법 조사는 형질감염된 세포의 비율이 약 25%임을 나타내었다. 형질감염시킨지 24시간 후에, 세포를 혈청없는 배지로 옮긴다. 이 세포 상층액을 3일이 더 지난 후에 수거한다.
세포 배양 상층액으로부터 pTF1Fc 융합 단백질의 정제
일시적으로 형질감염된 COS 세포로부터의 상층액 170ml을 0.8ml의 단백질 A-세파로즈를 함유한 컬럼에서 배치 방법으로 4℃에서 밤새 수집하고, 10배 용적의 세척 완충액(50mM 트리스 완충액(pH 8.6), 150mM NaCl)으로 세척하고 용출 완충액(93:7 100mM 시트르산: 100mM 시트르산 나트륨)을 사용하여 0.5ml의 분획으로 용출시킨다. 처음의 9개 분획을 각 경우마다 2M 트리스 완충액(pH 8.6) 0.1ml을 사용하여 즉시 중화시킨후 혼합하여, 생성된 단백질을 아미콘 미소농축기(Amicon microconcentrator)(Centricon 30)에서 3회 농축/희석 주기를 수행함으로써 TNE 완충액(50mM 트리스 완충액(pH 7.4), 50mM NaCl, 1mM EDTA)으로 옮긴다. 이런 방법으로 수득된 pTF1Fc를 SDS-PAGE 전기영동에 의해 정제한다[참조: U.K. Lammli, Nature 227(1970) 680-685]. 환원제의 부재하에서, pTF1Fc는

프로트롬빈 혈병 형성 시간 결정에서 정제된 TF1Fc의 생물학적 활성
TF1Fc 융합 단백질은 1-단계 프로트롬빈 혈병 형성 시간 결정에서 낮은 농도(50ng/ml 미만)에서 활성이 있다[참조: Vinazzer, H. Gerinnungsphysiologie und Methoden im Blutgerinnungslabor(1979), Fisher Verlag Stuttgart]. 달성된 혈병 형성 기간은 사람 태반으로부터 분리된 트롬보플라스틴을 사용한 경우 수득한 혈병 형성 시간과 비교한다.
실시예 2: 인터루킨-4 수용체 융합 단백질
인터루킨(IL-4)는 T 세포에 의해 합성되며 B-세포 증식을 자극할 수 있기 때문에 초기에는 B-세포 성장 인자로 명명되었었다. 인터루킨(IL-4)는 상기 세포들에게 많은 영향을 끼쳤다. 특히 한가지는 활성화된 B 세포에서 면역글로불린 아류인 IgG1 및 IgE의 분자의 합성을 자극하는 것이다[참조: Coffmann 등, Immunol. Rev., vol. 102(1988) 5]. 또한, IL-4는 T 세포 및 기타 헤모포이에틱 세포(hemopoietic cell)의 증식 및 분화를 조절한다. 그리하여, 알레르기성 반응 및 기타 면역학적 반응의 조절에 기여한다. IL-4는 특정 수용체에 높은 친화도를 가지고 결합된다. 사람 IL-4를 암호화하는 cDNA가 분리되었다[참조: Idzerda 등, J. Exp. Med., vol. 171(1990) 861-873]. IL-4 수용체가 시그날 펩티드로서 작용하는 25개의 N-말단 아미노산을 갖는 총 825개의 아미노산으로 구성되어 있다는 것
이 cDNA 서열로부터 추론된 아미노산 서열의 분석으로부터 명백하다. 성숙한 사람 IL-4 수용체는 800개의 아미노산으로 구성되며 트롬보플라스틴 처럼 3개-도메인 구조, 즉 i) 아미노말단 세포외 도메인(207개의 아미노산); ii) 막횡단 영역(24개의 아미노산); 및 iii) 세포질 도메인(569개의 아미노산)을 갖는다. 세포외 도메인에는, N-글리코실화를 위한 6개의 포텐셜 부위가 있다(Asn-X-Thr/Ser). IL-4 수용체는 사람 IL-6 수용체, 사람 IL-2 수용체의 β-서브 유니트, 마우스 에리트로포이에틴 수용체 및 래트 프로락틴 수용체와 동종성을 갖는다[참조: Idzerda 등, loc. cit.]. 그리하여, 트롬보플라스틴 처럼, 면역글로불린 계통군의 일원이 아니고 헤마토포이에틴 수용체의 신규 계통군으로 언급되는 동종성 단백질과 함께 할당된다. 상기 계통군의 일원은 4개의 시스테인 잔기를 가지며 통상 막횡단 영역 근처에 위치하는 세포외 도메인에 보존 서열(Trp-Ser-X-Trp-Ser)을 갖는다.

포유동물 세포로의 pIL4RFc의 형질감염

실시예 3: 에리트로포이에틴 융합 단백질
성숙한 에리트로포이에틴(EPO)은 166개의 아미노산으로 구성된 당단백질이고 적혈구의 발달에 필수적이다. 이것은 적혈구의 전구체 세포의 성숙 및 말단 분화

BamHI 절단 부위에 있는 판독 프레임은 pCD4E 감마 1에 있는 BamHI 부위와의 연결이 EPO cDNA의 개시 코돈에서 부터 IgG1의 중쇄의 정지코돈에 걸쳐 연속적인 판독 프레임과의 유전자 융합을 초래하도록 하는 것이다. 목적하는 단편을 수득하고, XhoI 및 BamHI으로 처리한 후, 상기와 같이 XhoI/BamHI으로 절단시킨 벡터 pCD4E 감마 1에 연결시킨다. 생성된 플라즈미드를 pEPOFc라고 명명한다(도 8 참조).독성 데이터엔브렐(ENBREL, etanercept)은 사람의 면역글로불린 G1의 Fc 부분과 종양 괴사 인자 수용체 p75의 세포의 리간드 결합부로 이루어진 이량체 융합 단백질로서, 이에 대한 독성 실험 데이터는 하기와 같다:류마티스성 관절염을 가진 환자 1039명에게 엔브렐을 투여한 후, 연구한 결과이다. 통제된 연구에서, 349명의 환자에게는 엔브렐을 투여하였고, 152명의 환자에게는 위약(placebo)을 투여하였다. 부작용으로 인해 치료를 중단한 환자의 비율은 엔브렐과 위약에서 동일했다(4%).1. 주사 부위 반응(injection site response)통제된 연구에서, 엔브렐로 치료를 받은 환자의 37%에서 주사 부위 반응을 나타내었다. 이들 반응 모두가 심각하지는 않았으며(가려움, 통증 또는 부풀어오름), 일반적으로 치료를 중단할 필요는 없었다. 이러한 반응은 대개 치료후 1달 이내에 발생하였으며, 그 이후로는 발생하는 빈도가 줄어들었다. 이러한 주사 부위 반응은 평균 3 내지 5일 동안 지속되었다. 계속 주사를 한 경우, 예전의 주사 부위에서 홍반이 발생한 환자가 7명이 보고되었다.2. 감염성 질환엔브렐 또는 위약을 투여한 환자에게서 상부 호흡기 감염성 질환('감기') 및 부비강염(sinusitis)이 가장 빈번하게 보고되는 감염질환이었다. 위약-통제된 연구에서, 위약을 투여받은 그룹에서의 상부 호흡기 감염성 질환의 발생률은 16%이었고, 엔브렐로 치료받은 그룹에서는 29%이었다. 그리고, 위약 그룹에서의 환자 년 당 발생횟수(event per patient year)가 0.68이고, 엔브렐로 치료한 그룹에서는 0.82이었다.엔브렐을 평가하기 위한 위약-통제된 시험에서, 증상이 심각한 감염성 질환의 발생 빈도의 증가는 관찰되지 않았다(위약 1.3%, 엔브렐 0.9%). 개방된 표지 및 위약-통제된 시험에서, 엔브렐에 노출된 전체 745명의 환자에서 22가지의 중증 감염성 질환이 관찰되었는 데, 여기에는 기관지염, 패혈증, 폐렴, 설사 등이 있었다.3. 악성18개월 동안 엔브렐로 치료한 745명의 류마티스성 관절염 환자에서 각종의 악성이 7차례 보고되었다.4. 자가 항체환자의 혈청을 가지고 여러 시점에서 자가 항체 여부에 대해 시험하였다. 항-핵 항체(ANA)에 대해 평가한 환자중에서 새로운 양성 ANA(1:40)가 발생한 환자의 비율은 위약으로 처리한 환자(5%)보다 엔브렐(11%)로 처리한 환자에서 더 높게 나왔다. 또한, 새로운 양성 항-이중 쇄 DNA 항체를 발생시키는 환자의 비율도 방사성면역검정(위약 처리한 환자 4% : 엔브렐로 처리한 환자 15%) 및 크리티디아 루실래 검정(위약 처리한 환자 0% : 엔브렐로 처리한 환자 3%)에서 더 높게 나왔다. 엔브렐로 처리한 환자중에서 항-카르디올리핀 항체를 발생시킨 환자의 비율은 위약-처리한 환자에 비하면 이와 비슷하게 증가하였다. 어떠한 환자에서도 루퍼스와 유사한 증후군이나 기타의 새로운 자가면역성 질환을 암시하는 증상이 보이지 않았다. 장기간의 엔브렐을 이용한 치료가 자가면역 질환 발생에 미치는 영향에 대해서는 아직 알려져 있지 않다.5. 기타의 부작용엔브렐로 치료받은 환자중에서 3% 이상의 환자에서 높은 빈도를 갖는 것으로 보고된 증상발생 및 환자년 당 증상발생(event per patient year)은 하기와 같다:위약-통제된 임상 시험에서 부작용을 호소하는 류마티스성 관절염 환자의 비율 및 환자년당 발생 증상 *
* 6개월간 메토트랙세이트 치료를 함께 받은 환자에게서 얻은 데이타를 포함** 3개의 통제된 시험중에서 2개의 시험에서 얻은 데이타를 포함통제된 시험에서 류마티스성 관절염을 앓고 있는 환자 중에서, 엔브렐로 치료받은 349명의 환자중 4%가 심각한 부작용을 나타내었으며, 위약으로 처리한 환자에서는 5%로 나타났다. 통제 및 개방-표지된 엔브렐 시험에서 류마티스성 관절염 환자중에서 관찰된 가장 심각한 증상이 악성 및 감염성 질환이었다. 기타 발생하는 부작용으로는 심장마비, 심근 국소 빈혈, 뇌빈혈, 고혈압, 저혈압, 췌장염, 우울증 등이 있었으나, 빈번하게 발생하지는 않는다.

따라서, 본 발명에서는 면역글로불린 계통군에 속하지 않는 사람 단백질 또는 이의 일부, 및 여러가지 아류의 면역글로불린(IgG, IgM, IgA, IgE)의 중쇄 또는 경쇄의 불변부의 다양한 부분으로 구성된, 유전공학적으로 처리된 가용성 융합 단백질을 제공한다. 사람 IgG, 특히 바람직하게는 사람 IgG1의 중쇄의 불변부가 면역글로불린으로서 바람직하며, 여기서 융합은 힌지영역(hinge region)에서 발생한다. 특정 양태에서, Fc 부분은, 또한 혼입되는 절단 서열에 의해 간단한 방법으로 제거될 수 있고 인자 Xa를 사용하여 절단될 수 있다.
또한, 본 발명은 유전공학적으로 상기 융합 단백질을 제조하는 방법, 및 진단과 치료를 위한 이의 용도에 관한 것이다.
Claims (7)
- 종양 괴사 인자 수용체의 세포외 부분 또는 이의 일부와 IgG, IgM, IgA 및 IgE로 이루어진 그룹으로부터 선택되는 면역글로불린 분자의 불변부로 이루어진 가용성 융합 단백질.
- 제1항에 있어서, 면역글로불린 부분이 사람 면역글로불린 G(IgG) 중쇄의 불변부인 융합 단백질.
- 제2항에 있어서, 면역글로불린 부분이 사람 IgG1 중쇄의 불변부 또는 이의 단백질 A-결합 단편인 융합 단백질.
- 제1항에 있어서, 융합이 힌지 영역(hinge region)에서 발생하는 융합 단백질.
- 융합 단백질을 암호화하는 DNA를 포유동물세포 발현 시스템에 도입시키고 발현시킨 후, 생성된 융합 단백질을 면역글로불린 부분을 통한 친화 크로마토그래 피에 의해 정제함을 특징으로 하여, 제1항에 따른 융합 단백질을 제조하는 방법.
- 제1항에 따른 융합 단백질을 포함하는, 고수준의 종양 괴사 인자와 관련된 질환의 진단용 또는 치료용 약제학적 조성물.
- 제7항에 있어서, 고수준의 종양 괴사 인자와 관련된 질환이 류마티스성 관절염인 약제학적 조성물.
