【発明の詳細な説明】
遺伝子治療のための改良されたAAVベクター 発明の背景
アデノ随伴ウイルス(AAV)は、約4.6kbの1本鎖DNAゲノムを有する
パルボウイルスである。他のウイルスと異なり、AAVは、天然には欠損のある
ウイルスであり、ウイルス産生性感染を樹立するためにはヘルパーウイルス(例
えば、アデノウイルスまたはヘルペスウイルス)との同時感染を必要とする。A
AV感染との関連が見いだされているヒトの疾患はない(ブラックロウ(Blackl
ow)ら、1968)。AAVの宿主の範囲は広い。レトロウイルスと異なり、A
AVは、インビトロおよびインビボで休止細胞および分裂細胞の両方に感染する
ことができ(フロッテ(Flotte)ら、1993;カプリット(Kaplitt)ら、1
994:ポドサコフ(Podsakoff)ら、1994;ラッセル(Russell)ら、10
04)、また異なる種や組織タイプに由来する細胞にインビトロで感染すること
ができる(レブコウスキー(Lebkowski)ら、1988;マクローリン(McLaugh
lin)ら、1988)。ヘルパーウイルスが存在しないところで感染が起きると
、野生型AAVは、アデノウイルスとの重複感染によりレスキュー(rescure)
されるまで、プロウイルスとして細胞のゲノム内に組み込まれる(ハンダ(Hand
a)ら、1977;チェウング(Cheung)ら、1980;ローリン(Laughlin)
ら、1986)。
AAVゲノムは比較的単純であり、短い逆末端繰り返し配列(inverted termin
al repeat)(ITR)が隣接する2つの読みとり枠(ORF)を含有する。IT
Rは、特にウイルス複製、レスキュー、パッケージングおよび組み込みに必要な
シス作用性配列を含有する。ITRの組み込み機能により、AAVゲノムが、感
染後に細胞の染色体に組み込まれることが可能になる。
非構造タンパク質または複製(Rep)タンパク質およびキャプシド(Cap
)タンパク質は、それぞれ5’ORFと3’ORFによりコードされる。rep
遺伝子から4つの関連するタンパク質が発現される。Rep78とRep68は
、
p5プロモーターから転写され、下流のプロモーター(p19)は、Rep52
とRep40の発現を指令する。大きいRepタンパク質(Rep78/68)
は、AAV複製ならびにウイルス遺伝子発現の制御に直接関与する(総説につい
ては、ムジクズカ(Muzyczka)、1992を参照されたい)。cap遺伝子は、
第3のウイルスプロモーター(p40)から転写される。キャプシドは、重複す
る配列の3つのタンパク質から構成され、最も小さいもの(VP−3)が最も豊
富に存在する。逆末端繰り返し配列は、複製、パッケージ、および組み込みのた
めにシスで必要な唯一のAAV配列である(サムルスキー(Samulski)ら、19
89)ため、多くのAAVベクターは、Repタンパク質およびCapタンパク
質をコードするウイルス遺伝子を含有せず、末端繰り返し配列の間に挿入された
外来遺伝子のみを含有する。
AAVが遺伝子治療におけるベクターとして興味があるのは、その生物学のい
くつかの特徴的な性質に由来する。このAAVベクターを使用することにより、
遺伝子治療の多くの目標について理想的な、安定な遺伝的形質転換が達成される
かも知れない。さらに、AAVの組み込み部位は、ヒトの第19染色体上にある
ことは、充分確立されている。この予測可能性により、宿主の遺伝子を活性化も
しくは不活性化するかまたはコード配列を妨害する可能性のある、細胞ゲノムへ
のランダムな挿入(このような現象のために、組み込みがランダムなレトロウイ
ルスのようなベクターの使用が限定される)という危険性を避けることができる
。repタンパク質はAAVの組み込みを仲介するため、AAVベクターの作成
時にこのタンパク質を除去すると、組み込みパターンが変化してしまう。さらに
、AAVはヒトの疾患と関係がなく、ウイルス由来の遺伝子治療ベクターの場合
のような多くの心配をしなくてよい。
AAVを基礎とするベクターのこの魅力的な面にもかかわらず、組換えウイル
スのストックを大量かつ高力価で産生することができないため、遺伝子治療のた
めのその評価における急速な進歩は妨げられている。組換えAAV(rAAV)
ベクターの産生のための従来法は、アデノウイルスに感染した293細胞への、
ベクターを含有するプラスミドとAAVのRepおよびCapタンパク質をコー
ドする第2のヘルパープラスミドとの同時感染である(例えば、レブコウスキー
(Lebkowski)ら、1988;サムルスキー(Samulski)ら、1989、エヌ・
ムジクズカ(Muzyczka,N.)、1992、カプリット(Kaplitt)ら、1994
;エイナーハンド(Einerhand)ら、1995)。この方法は面倒であり、rA
AVの収率が低く、典型的には104〜105感染単位または形質導入単位/mlで
ある。この方法を改良する方法としては、ポリリジンを介してプラスミドDNA
とアデノウイルス粒子の複合体を作成してトランスフェクション効率を上げ(マ
モウナス(Mamounas)ら、1995)、ベクター配列を組換えアデノウイルスの
一部として提供し(スラシャー(Thrasher)ら、1995)、そしてSV40レ
プリコンに結合させてヘルパープラスミドのコピー数を増幅させる方法があった
(チオリニ(Chiorini)ら、1995)。
前記アプローチを使用して高力価の組換えAAVのストックを産生することが
できないため、遺伝子治療ベクターとしてのAAVの開発の進展は制限されてき
た。この制限は今日まで、組換えAAVゲノムの複製とパッケージングにトラン
スで必要なAAVタンパク質の産生不足に由来すると考えられてきた。ベクター
産生のためのトランスに基づく方法は、トランスで必要なタンパク質のレベルを
調節してAAVベクターを産生させることである。これらのタンパク質のレベル
を上げる試みには、AAV rep遺伝子をHIV LTRプロモーター(エフ
・アール・フロッテ(Flotte,F.R.)ら、Gene Therapy 2: 29-37,1995)の制
御下に置いて、タンパク質レベルを上げること、およびrepタンパク質を発現
する細胞株を開発する(キュー・ヤング(Yang,Q.)ら、J.Virol.68: 4847-4
856,1994)ことがあった。
高力価のAAVベクターのストックの産生が制限されることはまた、組換えA
AVベクターの設計にAAVシス要求(cis-required)成分を含有させることが
できないことにも由来する。ベクター産生のためのシスに基づく方法は、AAV
ベクター粒子にパッケージングされる組換えDNAに、シス(縦列)で必要なD
NA配列を与えるものである。このトランスとシス機能は関連している。トラン
スで必要なタンパク質は、ベクターの産生に必要であるが、これは機能活性を有
するためには、組換えAAVゲノム中にシス作用性配列を必要とする。従って、
AAVベクターの高収率の産生には、標準的な遺伝子治療ビヒクルとしてのAA
Vの開発を進行させるために、トランスに基づく改良とシスに基づく改良の共同
作用が必要である。
従って、野生型AAVおよびアデノウイルスヘルパーの汚染のない、高力価の
組換えAAV(rAAV)調製物の産生を可能にする方法および組成物に対する

3B1細胞 − E1とE2Aが欠失した組換えアデノウイルスベクターを補
足するアデノウイルスゲノムの一部を発現するように修飾したヒト胚腎細胞株。
1996年8月28日にアメリカンタイプカルチャーコレクション(ATCC)
(12301 パークローンドライブ(Parklwan Drive)、ロックビル(Rockvi
lle)、メリーランド州)に寄託され、ATCC CRL−12183を割り当
てられた。
Cfu − コロニー形成単位。抗生物質耐性遺伝子を有するレトロウイルス
およびアデノ随伴ウイルスベクターについては、感染後の抗生物質耐性細胞コロ
ニーの数。1つの感染細胞から1つのコロニーが生成すると仮定する。
発現プラスミド − プラスミド中で運搬される遺伝子が細胞内で発現される
ように作成した、細胞内で自律的に増殖することができる染色体外遺伝成分。
シスで − 同じDNA分子から。
トランスで − 異なるDNA分子から。
挿入突然変異誘発 − 宿主細胞ゲノムへの外来遺伝子(例えば、ウイルスD
NA)の挿入による、標的遺伝子への突然変異の導入。細胞への外来DNAの導
入の結果としての細胞性遺伝子の突然変異の出現。
ITR − 逆末端繰り返し配列。ウイルスの両端で逆転した形で繰り返され
るDNA配列。
プロモーター−トランス遺伝子カセット − 遺伝子産物の産生を指令するの
に必要な成分を含有するDNA配列と、遺伝子自身のDNA配列の組合せ。
形質導入 − 生物の細胞への外来DNAの導入(インビボ)。
トランスフェクション − 培養細胞への外来DNAの導入(インビトロ)。
化学的手段を使用する外来DNAの導入による真核細胞の遺伝的修飾。一過性ト
ランスフェクションでは、組み込みされていない外来DNAから発現が起き、ト
ランスフェクション後数日間検出される。
トランス遺伝子 − 別の生物に安定に取り込まれた遺伝子。
休止細胞 − 活発な分裂をしていない細胞。
組換え − 2つまたはそれ以上のDNA分子(この場合はウイルス配列)の
間の物理的相互作用であり、分子の間でDNA配列の交換が起きる。
力価 − 1ml当たりに産生されるウイルス粒子の数。産生されたウイルス粒
子の数の測定系は、問題のウイルスにより大きく異なる。最大数の細胞への、ウ
イルスにより運搬される治療用遺伝子の導入を可能にするため、遺伝子治療の成
功には一般的に高力価が必要である。
ベクター − 生物に遺伝子を提供するために使用される、通常は生物物質(
例えば、ウイルス)であるビヒクル。細胞への外来遺伝子の導入を促進する試薬
。
LacZ遺伝子 − 細胞性遺伝子マーカーとして使用される細菌の遺伝子。
その発現は、基質X−galの存在下での青い発色により検出される。
パッケージング細胞 − AAVからのcapおよびrep遺伝子を含有する
プラスミドでトランスフェクションされた細胞。
本明細書に記載の研究の目的は、rAAVパッケージングに必要な律速成分を
決定することであった。基礎的なレベルでこの過程が理解できれば、rAAV産
生のすべての方法にとって有益であろう。repまたはcap遺伝子(または両
方)の発現を選択的に増加させることにより、Capタンパク質産生がrAAV
産生の1つの律速因子であることが証明される。
A.トランス産物の準備
1.アデノウイルスタンパク質
前述のように、アデノウイルスタンパク質は、ウイルス産生性AAV感染を引
き起こすのに必要である。ヘルパーアデノウイルスの非存在下では、AAVは細
胞性ゲノム中に組み込まれ、細胞がアデノウイルスで感染されるまで潜伏する。
ヘルパー機能に必要なアデノウイルス遺伝子には、特にE1A、E1B、E2
A、E4 ORF6、およびVA RNAがある(エヌ・ムジカ(Muzycka,N.
)、Curr.Top.Micro.Immunol.158: 97-129,1992)。組換えAAVベクター
を作成するための標準的方法は、必要なヘルパータンパク質の充分なレベルを提
供できるようにするために、標的細胞のアデノウイルス感染に依存してきた。A
AVベクター産生中に起きる野生型アデノウイルスの好ましくない産生を避ける
ために、欠失を含むアデノウイルスが、必須のアデノウイルスタンパク質をトラ
ンスで提供する細胞株で使用される。あるいは、温度感受性アデノウイルス複製
突然変異体が、非許容温度で使用される。
本発明の1つの実施態様において、プラスミドの複製を可能にするAAV I
TR配列が結合した必須のアデノウイルスヘルパー遺伝子を含有するヘルパープ
ラスミドが提供される。ヘルパープラスミドは、E1A、E1B、E2A、E4
ORF6、およびVA RNA遺伝子を含有する。これらの各遺伝子はまた、
それ自身のプロモーターを含有し、これにより転写が起きる。ヘルパープラスミ
ドで使用できる別のプロモーターには、CMV、RSV、MMTV、EIA、E
F1a、アクチン、サイトケラチン14、サイトケラチン18、PGKならびに
当業者に公知の他のものがあるが、これらに限定されない。
複製しているプラスミドへの必須のアデノウイルス遺伝子の提供は、宿主細胞
をアデノウイルスで感染させることの代替法である。プラスミド中のAAV I
TR配列は、AAV repタンパク質の制御下の複製開始点として機能して、
プラスミドのコピー数を増加させる。コピー数の増加により、プラスミド上の遺
伝子によりコードされるタンパク質のレベルが増加する。すなわち、本発明のヘ
ルパープラスミドは、AAVベクター産生に必要なアデノウイルスタンパク質を
提供し、アデノウイルス産生の可能性を排除する。さらなる利点は、プラスミド
の複製は遺伝子コピー数を投入レベルより上昇させるため、アデノウイルスタン
パク質のレベルは、投入したプラスミドDNAの量により限定されないことであ
る。
別の実施態様において、複製開始点には、SV40複製開始点、エプスタイン
バー(EBV)複製開始点、ならびに当業者に公知の他の複製開始点があるが、
これらに限定されない。例えば複製開始点が活性化タンパク質(例えば、T抗原
を必要とするSV40複製開始点、EBNAタンパク質を必要とするEBV複製
開始点)を必要とする場合、その活性化タンパク質は、細胞株供給源を作成する
ために安定なトランスフェクションにより提供されるか、または適切な遺伝子を
含有するプラスミドによる一過性トランスフェクションにより提供される。
本発明のヘルパープラスミドを作製するために、標準的組換えDNA技術が使
用できる(例えば、Current Protocols in Molecular Biology,エフ・アウスベ
ル(Ausubel.,F.)ら、ワイリーアンドサンズ(Wiley and Sons)、ニューヨー
ク州 1995を参照)。このような方法には、アデノウイルス遺伝子とITR
配列の境界の適合性のある制限部位、または制限部位を含有するDNAリンカー
配列を使用するもの、ならびに当業者に公知の他の方法がある。アデノウイルス
DNA情報については、アール・ジェイ・ロバーツ(Robert,R.J.,)、Adenov
irus DNA: The Viral Genome and Its Expression、ダブリュー・オーバーフラ
ー(Overfler,W.)編、マチヌス・ニホフ・パブリッシング(Matinus Nihoff P
ublishing)、ボストン、1986)を参照されたい。例えば、pBR322(
ニューイングランドバイオラボズ(New England Biolabs)、ビバリー、マサチ
ューセッツ州)、pRep9(インビトロゲン(Invitrogen)、サンジエゴ、カ
リホルニア州)またはpBS(ストラタジーン(Stratagene)、ラホイヤ、カリ
ホルニア州)のような分子生物学の分野で日常的に使用されるプラスミドが、ア
デノウイルス遺伝子やAAV ITRが挿入されるヘルパープラスミドの基礎と
して使用できる。アデノウイルス遺伝子は、任意の位置でヘルパープラスミド中
に入れられる。本発明のこのような複製しているヘルパープラスミドの具体的な
実施態様を、図1に示す。
ヘルパープラスミドは、AAV repおよびcapタンパク質の供給源と組
合せると、組換えAAV、ならびに組換えAAVゲノムの作成に使用される。当
該分野で公知の技術を用いてプラスミドにより細胞をトランスフェクションする
と、AAV rep遺伝子発現の開始に必要なアデノウイルス遺伝子産物が得ら
れる。
2.AAVタンパク質
組換えAAVベクターのストックを作成するために、標準的アプローチにより
、AAVベクタープラスミドとともに標的細胞を同時トランスフェクションする
ために使用されるプラスミド上でAAV repおよびcap遺伝子が提供され
る。トランスフェクションの結果として産生されるrepおよびcapタンパク
質のレベルは、AAVベクターの産生を最大にすることと関係がある。これは、
repタンパク質がcap遺伝子の転写を活性化し、組換えゲノムのパッケージ
ングに関与するAAV構造タンパク質が産生されるためである。
これらのAAVタンパク質のレベルを上昇させてベクター産生を増強する試み
は、問題がある(アール・エム・コチン(Kotin,R.M.)、Human Gene Therapy
5: 793-81,1994)。問題の1つは、細胞に対する高レベルのrepタンパク質
の毒性のようである。
本発明のこの面において、AAV repおよびcap遺伝子は、AAV I
TR配列を含有する複製しているヘルパープラスミド上で提供される。repタ
ンパク質は複製開始点としてITRを活性化し、プラスミドを複製させ、その結
果コピー数が増加する。この方法の利点は、repタンパク質レベルは、プラス
ミドを用いるトランスフェクションの効率に依存するのみでなく、トランスフェ
クション後のプラスミドの複製の機能に依存することである。AAV repお
よびcap遺伝子を含有する複製しているヘルパープラスミドの例を図2に示す
。本発明のこの面の他の実施態様において、複製開始点には、SV40複製開始
点、エプスタインバー(EBV)複製開始点、ならびに当業者に公知の他の複製
開始点があるが、これらに限定されない。例えば複製開始点が活性化タンパク質
(例えば、T抗原を必要とするSV40複製開始点、EBNAタンパク質を必要
とするEBV複製開始点)を必要とする場合、その活性化タンパク質は、細胞株
供給源を作成するために安定なトランスフェクションにより提供されるか、また
は適切な遺伝子を含有するプラスミドによる一過性トランスフェクションにより
提供される。
さらに別の実施態様において、AAV repおよびcap遺伝子は、複製し
ていないプラスミド上で提供され、これは複製開始点を含有しない。このような
複製していないプラスミドはさらに、ウイルスの産生を最適化するために、細胞
の複製器官が、複製している組換えAAVゲノムに向いていることを確実にする
。さらにある研究は、高レベルrepタンパク質は細胞に毒性があり(ムジカ(
Muzycka)、N.,Curr.Top.Micro.Immunol.158: 97-129,1992)、複製して
いないプラスミド上にrep遺伝子を提供することはこの可能性を低下させるか
も知れないことを示唆している。このような複製していないプラスミドによりコ
ードされるAAVタンパク質のレベルは、これらの遺伝子の発現を指令するため
の特定のプロモーターの使用により調節される。このようなプロモーターには、
AAVプロモーター、ならびに外来性のプロモーター(例えば、CMV、RSV
、MMTV、EIA、EF1a、アクチン、サイトケラチン14、サイトケラチ
ン18、PGK)、ならびに当業者に公知の他のものがあるが、これらに限定さ
れない。複製していないAAVヘルパープラスミドの例を図3に示す。
これらのヘルパープラスミドにより産生されるrepおよびcapタンパク質
のレベルは、目的のタンパク質のレベルに最適の各遺伝子のプロモーターを選択
することにより、個々に制御される。特定の遺伝子(例えば、rep遺伝子)の
具体的な調節は、誘導性プロモーターの使用により達成することもできる。この
ような誘導性プロモーターには、MMTV、メタロチオネイン、ならびに当業者
に公知の他のものがあるが、これらに限定されない。
repおよびcap遺伝子を含有するAAVゲノム配列のパッケージングを防
ぐに、repおよびcap DNA断片を含有するプラスミドが、最適なパッケ
ージングの長さ以上にDNAを伸長させる「詰め物」("stuffer")断片をAA
Vゲノムに導入することにより修飾される。すなわち、ヘルパー断片はAAVウ
イルス粒子中にパッケージングされない。これは安全な特徴であり、最適なパッ
ケージングサイズを超えない組換えAAVベクターゲノムのみがウイルス粒子中
にパッケージングされることを確実にする。詰め物配列を取り込むAAVヘルパ
ー断片は、野生型のゲノムの4.6kbの長さを超え、野生型の105%を超える
長さはパッケージングされない。詰め物断片は、例えばβ−ガラクトシダーゼ遺
伝子のような非ウイルス供給源から得られる。
本発明のヘルパープラスミドを作製するために、標準的組換えDNA技術が使
用され(例えば、Current Protocols in Molecular Biology,エフ・アウスベル
(Ausubel.,F.)ら、ワイリーアンドサンズ(Wiley and Sons)、ニューヨーク
州 1995を参照)、遺伝子とAAV ITR配列(使用される場合)の間の
境界の適合性のある制限部位、または制限部位を含有するDNAリンカー配列の
利用、ならびに当業者に公知の他の方法がある。アデノウイルスDNA配列情報
については、エー・ストリバスタバ(Strivastava,A.)ら、J.Virol.45: 55
5-564,1983 を参照されたい。分子生物学の分野で日常的に使用されるプラスミ
ドが、骨格として使用できる。例えば、AAV遺伝子の挿入については、pBR
322(ニューイングランドバイオラボズ(New England Biolabs)、ビバリー
、マサチューセッツ州)、pRep9(インビトロゲン(Invitrogen)、サンジ
エゴ、カリホルニア州)またはpBS(ストラタジーン(Stratagene)、ラホイ
ヤ、カリホルニア州)、および複製しているプラスミドの場合は、AAV IT
Rがある。
3.ハイブリッドヘルパープラスミド
組換えAAVベクターのストックの作成には、組換えAAVゲノムの転写活性
化、複製およびパッケージングを促進するために、トランスで提供されるAAV
とアデノウイルスタンパク質の両方が必要である。標準的方法は、標的細胞への
AAV遺伝子の、プラスミドをベースにした提供を利用してきた。アデノウイル
スによる標的細胞の感染は、アデノウイルス遺伝子を提供するために使用される
。この多工程プロトコールには、トランスフェクションと感染の共同が必要であ
る。さらに、アデノウイルスによる細胞の感染は、アデノウイルスの産生を可能
にし、これは純粋なAAVベクターのストックを作成しようとする場合は好まし
くない。さらに、ベクターの作成に必要とされない多くのウイルス遺伝子の導入
は、AAV産生に必須のタンパク質のより効率的な産生に向かうはずの転写およ
び複製機構を迂回させる。AAVベクターは感染により導入されたアデノウイル
ス遺伝子を使用して産生されているが、高収率のベクター産生にはまだ問題があ
る。従って、トランスで必要なタンパク質をコードする遺伝子のより効率的な提
供は、AAVベクターの産生を改良するであろう。
本発明は、AAVの高収率のためにトランスで必要なすべての必須のウイルス
タンパク質の単純な提供方法を与える。必須のタンパク質をコードするAAVお
よびアデノウイルス遺伝子を運搬するために、ハイブリッドプラスミドが作製さ
れる。このようなプラスミドの利点は、特にトランス作用性タンパク質をコード
するすべての遺伝子を提供する細胞への単一の侵入、そのようなすべての細胞の
調和した供給、および不要なアデノウイルス遺伝子の排除に由来するアデノウイ
ルス産生を避けることができること、などである。このようなプラスミドを図4
に示す。このプラスミドは、必須のアデノウイルス遺伝子(E1A、E1B、E
2A、E4 ORF6、およびVA RNA)を含有する。このプラスミドはま
た、AAV repおよびcap遺伝子、ならびにプラスミドの複製に必要なA
AV ITR配列を含有する。この遺伝子は、それ自身のプロモーターを使用し
て転写される。またプロモーターには、CMV、RSV、MMTV、E1A、E
F1a、アクチン、サイトケラチン14、サイトケラチン18、PGK、並びに
当業者に公知の他のプロモーターがあるが、これらに限定されない。アデノウイ
ルス遺伝子は、図4に示す順序でプラスミド中に挿入されるか、または任意の数
の異なる位置に、当業者に公知の方法により挿入される。
トランスで必要とされるすべての必須の遺伝子は、クローニングを簡単にする
ために2つのプラスミド上で提供される。すなわち、ハイブリッドヘルパープラ
スミド上のAAVおよびアデノウイルス遺伝子は、クローニングのための任意の
最適な配置で、2つのプラスミドにより運搬される。すなわち、AAVおよびア
デノウイルス遺伝子は、別々のプラスミド上にある必要はない。
本発明のヘルパープラスミドを作製するために、遺伝子とITR配列の間の境
界の適合性のある制限部位、または制限部位を含有するDNAリンカー配列の利
用、ならびに当業者に公知の他の方法などの、標準的組換えDNA技術(例えば
、Current Protocols in Molecular Biology,エフ・アウスベル(Ausubel.,F.
)ら、ワイリーアンドサンズ(Wiley and Sons)、ニューヨーク州 1995を
参照)が使用された。アデノウイルスDNA配列の情報の文献は、前記で引用さ
れている。例えば、pBR322(ニューイングランドバイオラボズ(New Engl
and Biolabs)、ビバリー、マサチューセッツ州)、pRep9(インビトロゲ
ン(Invitrogen)、サンジエゴ、カリホルニア州)またはpBS(ストラタジー
ン(Stratagene)、ラホイヤ、カリホルニア州)のような日常的に使用され
るプラスミドが、アデノウイルスやAAV遺伝子およびAAV ITRの挿入の
ために使用できる。アデノウイルス遺伝子は、任意の位置でヘルパープラスミド
中に入れられる。
4.組換えAAVベクターの産生
トランスで必要な必須のタンパク質を提供するヘルパープラスミドは、組換え
AAVベクターのストックを作成するために使用される。これらのプラスミドは
、任意の数のトランスフェクション方法(特に、リン酸カルシウムトランスフェ
クション、リポフェクチンまたは当業者に公知の他の方法)を使用して標的細胞
に導入される。ヘルパープラスミド対目的の遺伝子を含有するベクタープラスミ
ドの量の比率は、1:1〜1:10の範囲である。この方法では、組換えAAV
ベクターが産生され、このベクタープラスミドは、AAV ITRが隣接する組
換えAAVゲノムを含有する。
組換えAAVベクターは、8個のローラーボトル中で293細胞を使用して産
生される(1X109細胞/ml)。細胞は、ヘルパープラスミドとAAVベクタ
ープラスミドの両方により、ベクター:ヘルパー比1:10でトランスフェクシ
ョンされる。プラスミドは、任意の数のトランスフェクション方法(リン酸カル
シウム、リポフェクチンまたは当業者に公知の他の方法があるが、これらに限定
されない)を使用して標的細胞に導入される(例えば、Current Protocols in M
olecular Biology,エフ・アウスベル(Ausubel.,F.)ら、ワイリーアンドサン
ズ(Wiley and Sons)、ニューヨーク州 1995を参照)。好適な実施態様に
おいて、リポフェクチンがトランスフェクションに使用される。アデノウイルス
感染は、感染多重度(MOI)20で開始される。アデノウイルス株は欠失ウイ
ルスでもよく、この場合、補足性遺伝子が、細胞株または非許容性温度(39℃
)で複製できない温度感受性突然変異体(例えば、ts149)中に組み込まれ
る。トランスフェクション/感染された細胞は、次に適切な温度で2日間インキ
ュベートされる。インキュベート後、細胞を採取し、次にベンゾナーゼ(benzon
ase)(アメリカンインターナショナルケミカル(American International Chem
ical)、ナチック(Natick)、マサチューセッツ州)の存在下で、3回の凍結融
解サイクルを行って溶解する。次に溶解物に1%デオキシコール酸と0.2
5%トリプシンを加え、次に37℃で30分インキュベートする。次に細胞溶解
物(2個のローラーボトル/勾配)を、SW28ロータ中でCsCl段階勾配(
1.5μg/ml〜1.36g/ml)にかけ、26Kで4℃で6時間遠心分離する
。画分を得て、次にNVT65ロータを用いて2回の平衡勾配で精製し、60K
で4℃で20時間遠心分離する。平衡勾配からの画分を屈折計でスクリーニンク
化、プールする。プールした画分を、1%ショ糖含有PBSで4℃で2時間の透
析を3回行う。
5.組換えAAVベクター産生のための測定法
トランスで必要なタンパク質の供給源としてのヘルパープラスミドの効率は、
AAVベクターのストックの収率から測定される。ウイルスの収率を測定するた
めに、AAV感染中心測定法を用いて、感染性子孫の産生について測定する。組
換えAAVベクターは、産生後、前述の精製プロトコールを使用して回収される
。測定法は、感染性AAV子孫が産生プロトコールで産生されるかどうかを示す
。
この測定法は、10%FBS、ペニシリン/ストレプトマイシン、およびグル
タミンを補足したDME培地中の0.1ml/ウェルでウェル当たり1×105細
胞の密度で、293細胞を1日目にプレートに蒔く。24時間後、細胞をMOI
20のアデノウイルスとMOI 2の野生型AAVで感染させる。ウイルスを
同じ培地に懸濁し、各ウェルに0.1mlを加えて感染を開始させる。AAVベク
ターの試料をウェルに加え(25〜100μlの溶解物または希釈物)、プレー
トを適切な温度(野生型については37℃、アデノウイルス温度感受性突然変異
体については39℃)で24〜30時間インキュベートする。感染の翌日、培地
を注意深く細胞から除去する。5mM EDTAを含有する0.2mlの冷PBSを
各ウェルに加え、プレートを氷の上に置く。
次にろ過装置の準備をして、PBSであらかじめ湿らせたニトロセルロースフ
ィルターを所定の位置に置き、ろ過装置の上の部分に5mlのPBSを加える。プ
レート中の細胞を、ピペットで再懸濁する。ろ過装置のPBS緩衝液に0.05
mlの細胞懸濁液を加え、回転させて混合する。装置に吸引をかけて、細胞をフィ
ルター上に沈着させる。フィルターを10分間風乾する。変性溶液、次に中和溶
液を使用して、フィルター上で細胞を直接溶解する。各溶解後5分間紙の上でフ
ィルターを乾燥し、次に10分間風乾する。フィルターを2×SSCで洗浄し、
10分間風乾し、真空オーブン中で2時間焼く。上記のように調製し2×SSC
で湿らせたフィルターを使用して、目的の遺伝子を検出するプローブへのハイブ
リダイゼーションを行う。30〜40mlのクイック−ハイブ(Quick-Hyb)(登
録商標)(ストラタジーン(Stratagene)、ラホイヤ(La Jolla)、カリホルニ
ア州)を使用して、回転水浴中で68℃で2〜4時間インキュベートして、フィ
ルターをプレハイブリダイゼーションすることができる。標識したプローブを、
次にクイックーハイブ(Quick-Hyb)(登録商標)溶液に加え、68℃で一晩イ
ンキュベートする。翌日フィルターを洗浄する(2×SSC、0.5%SDSで
室温で5分間、2×SSC、0.1%SDSで室温で15分間、次に0.1×S
SC、0.5%SDSで65℃で2時間)。フィルターを−80℃で一晩フィル
ムに感光させて、オートラジオグラフィーを作成する。
オートラジオグラフィーでフィルター上の感染中心の数を数える。感染中心の
数に、試料の調製物に使用した希釈率を掛けて、出発物質の力価を決定する。
産生されたAAVベクターがレポーター遺伝子を含有する場合、AAVベクタ
ーの力価を決定するための別の方法が使用できる。例えば、lacZ遺伝子を使
用する場合、遺伝子産物(β−ガラクトシダーゼ)の発現についてX−galを
使用して、感染細胞を染色することができる。力価は、例えばプレートのウェル
中の青く染色された細胞を数えることにより定量される。
B.シス作用性配列を含有するAAVベクタープラスミド作製体
本発明はまた、効率的な複製とパッケージングのために必要なAAVゲノム中
のシス作用性配列を利用するベクタープラスミド設計により、組換えAAVベク
ターの産生を増加させる手段を提供する。本発明はまた、このようなシス作用性
配列を提供するように設計されたベクタープラスミドを提供する。
現在のベクタープラスミドの設計では、組換えゲノムを作成するためにおよび
他のAAV配列を提供しないようにするために、AAV ITR配列の間に目的
の遺伝子を置く。AAV ITR配列は、ベクター産生中の組換えゲノムの複製
とパッケージング、ならびにAAVベクターによる導入後の細胞へのベクターD
NAの組み込みを促進する、シス作用性機能を有する。すなわち、ITR配列は
、
組換えAAVベクターの設計では維持される。しかしAAVベクターの高力価の
産生が困難なことは、ITR自身は、高力価のベクターのストックの産生に必要
なすべてのシス作用性機能を提供するには不充分であることを示す。従って、組
換えAAVゲノムの複製および/またはパッケージングの効率を向上させるため
に、ベクター作製体内にITR以外に他のシス作用性AAV配列が必要である。
AAVゲノム内のシス作用性成分は、AAV repタンパク質との相互作用
を介してゲノムのレスキューと複製を促進すると考えられている。repタンパ
ク質がAAV ITR内の部位ならびにAAV p5およびp19プロモーター
内の部位に結合することは公知である(ディー・エム・マカーティー(McCarty
,D.M.)ら、J.Virol.65: 2936-2945,1991 ;ディー・エム・マカーティー
(McCarty,D.M.)ら、J.Virol.68: 4988-4997,1995)。シス作用性パッケ
ージング成分はまた、最大の粒子産生のために組換えAAVベクターゲノム中で
必要であるようである。
本発明は、ITR配列以外にAAV配列を含有するベクター骨格作製体を使用
してAAVベクター産生を改良するための方法を提供する。AAVベクター骨格
は、rep結合部位または決定的に重要なパッケージング配列を含有するAAV
ゲノム断片を含有してもよい。すべてのシス作用性AAV配列の正確な数と位置
はまだ規定されていないため、すべてのシス作用性配列(まだ規定されていない
ものも含む)を含むために、AAVゲノムのかなりの部分を含有するベクタープ
ラスミドの作製が重要である。これらのベクタープラスミド作製体は組換えAA
Vベクターのストックの産生を改良するが、本発明のさらなる有用性は、改良さ
れたベクター産生により必須のシス作用性配列が機能的に同定されることである
。
このようなシス作用性配列を含有するベクター作製体は、公知の技術を用いて
調製される。(例えば、Current Protocols in Molecular Biology,エフ・アウ
スベル(Ausubel.,F.)ら、ワイリーアンドサンズ(Wiley and Sons)、ニュー
ヨーク州 1995を参照)。AAVゲノム中の既知の制限部位の存在は、組換
えAAVベクター内に挿入するためのサブゲノム断片を得るのに使用される。組
換えゲノムがAAV粒子のパッケージング能力を超えないように、断片の長さが
選択される。必要であれば、「詰め物」DNA配列を作製体に加えて、比較のた
めに標準的なAAVゲノムのサイズを維持する。このような断片は、非ウイルス
性供給源、例えばlacZ、または公知の他の遺伝子から得られ、これらを当業
者は利用することができる。
本発明は、AAV ITRが隣接する目的の遺伝子を含有するベクタープラス
ミドにAAVサブゲノム断片を加える一連のベクタープラスミド作製体を提供す
る。図5を参照されたい。これらの断片のサイズは、1.7〜2.1kbの範囲で
ある。これらの断片はAAVゲノムのコード領域または非コード領域を含有する
ため、パッケージングへの作用は、トランスおよびシスで作用する成分に起因す
るかも知れない。記載した作製体のさらなる修飾物(例えば、より小さいAAV
断片の挿入)は、本発明の範囲内にあり、当業者はこれを容易に作成することが
できる。ゲノム中のコード領域より小さいサブゲノム断片を使用すると、ベクタ
ー産生について観察される作用は、シス作用性が特徴である。例えば、サブゲノ
ム断片を最適のベクター産生のために必要な最小の長さにするには、標準的欠失
解析法が適している。規定されたAAVシス作用性断片(例えば、rep応答性
成分)は、ベクタープラスミド内に特異的にクローン化される。
本発明は、遺伝子治療で使用されるAAVベクターの収率を決定するための、
効率的なレポーター測定法を提供する。こうして、高力価のストックの産生によ
り、最も効率的な作製体の設計が同定される。
AAV配列が加えられる場合、産生効率の改良を測定するために、レポーター
遺伝子とAAV ITR配列を含有するプラスミドが使用される。このプラスミ
ドは、追加の作製体(例えば、プラスミドpTR−CAT、図6に示す)を作成
するために、AAVサブゲノム断片の挿入により修飾することができる。このプ
ラスミドは、CMVプロモーターの制御下のクロラムフェニコールアセチルトラ
ンスフェラーゼ(CAT)遺伝子、ハイブリッドイントロンおよびBGHポリA
部位を含む発現カセットを含有する。発現カセットは、AAV ITR領域をp
UC19にクローニングすることにより得られる高コピー数プラスミドであるp
UC−TR中にクローン化された。(ニューイングランドバイオラボズ(New En
gland Biolabs)、ビバリー、マサチューセッツ州)。pUC−TRに発現カセ
ットを挿入すると、pTR−CATが得られた。リンカーを挿入するかまたは
当業者に公知の他の技術を使用して、CATカセットの5’末端および3’末端
に、制限部位を付加してもよい。AAVサブゲノム断片は、これらの制限部位で
挿入される。このレポータープラスミドは、発現カセットの5’末端または3’
末端に位置する、図5に示すサブゲノム断片を収容できるであろう。最適化する
ためには、両端への断片の挿入を試験して、挿入体の位置による位置的影響を調
べることが必要であろう。断片はまた、ベクタープラスミドの両端に挿入するこ
とができる。規定されたシス作用性成分の1つまたはそれ以上のコピーは、産生
されるベクターの量を増加させることができる。
新規の作製体のパッケージングには、AAV repおよびcap遺伝子(例
えば、例1に記載のpIM45を使用して)、ならびに必須のアデノウイルス遺
伝子のパッケージングが提供されることが必要である。AAVゲノムを含有する
プラスミドは、新規の作製体と同時トランスフェクションされる。本発明のアデ
ノウイルス感染またはアデノウイルスヘルパープラスミド(セクション1を参照
、前述)は、他の必要な遺伝子を提供する。
感染性子孫(前記セクション5を参照)の量を測定するために、適切なプロー
ブを利用する感染中心測定法が使用される。あるいは、AAVベクター中のレポ
ーター遺伝子産物を直接測定できる(例えば、このレポーター遺伝子が存在する
場合は、CAT酵素測定法が使用される、例えばpTR−CAT(Current Prot
ocols in Molecular Biology,エフ・アウスベル(Ausubel.,F.)ら、ワイリー
アンドサンズ(Wiley and Sons)、ニューヨーク州 1995))。
レポーター遺伝子とAAVシス作用性断片を含有する最終のプラスミド作製体
が、AAV粒子のためのパッケージング長さを超えないという条件下で、ベクタ
ープラスミド作製体を測定するのに別のレポーター遺伝子を使用してもよい。他
のレポーター遺伝子産物は、適切な生化学的測定法を使用して、感染性AAV粒
子中に検出される。
本発明は、ヒト第19染色体上のAAV組み込み座を発現することができる非
ヒトトランスジェニック哺乳動物を提供する。非ヒトトランスジェニック哺乳動
物の例には、トランスジェニックウシ、ヒツジ、ヤギ、ブタ、ウサギ、ラットお
よびマウスがある。
本発明のポリペプチドの生理学的および行動的役割を解明する動物モデル系は
、種々の方法を用いて目的のポリペプチドの発現が変更または修飾されているト
ランスジェニック動物を作成することにより、産生される。このような方法の例
には、トランスジェニック動物を産生するために、微量注入法、レトロウイルス
感染または当業者に公知の他の手段により、適切な受精した胚への、目的のポリ
ペプチドをコードする核酸の正常なまたは突然変異体の挿入がある。例えば、カ

目的のポリペプチドの発現または構造の制御を変化させるために、別の方法
(トランスジェニック動物での未変性の遺伝子座との、これらの遺伝子の突然変
異体または正常なものとの相同的組換え)が使用できる(カペッチ(Capecchi)
ら、Science 244: 1288,1989 ;チマー(Zimmer)ら、Nature 338: 150,1989
を参照)。相同的組換え法は当該分野で公知である。相同的組換えは、未変性の
(内因性)遺伝子を組換えまたは突然変異遺伝子で置換して、未変性の(内因性
)タンパク質を発現できないが、例えばヒトAAV組み込み座を発現させる組換
えタンパク質を発現することができる動物を産生することができる。
相同的組換えに比較して、微量注人法は、宿主遺伝子を取り出すことなく、宿
主ゲノムに遺伝子を付加する。微量注入法は、内因性および外因性ポリペプチド
の両方を発現することができるトランスジェニック動物を産生する。トランス遺
伝子の発現を制御する手段を提供するために、誘導性プロモーターを核酸のコー
ド領域に結合することができる。組織特異的制御成分はコード領域に結合して、
トランス遺伝子の組織特異的発現を引き起こすことを可能にする。トランスジェ
ニック動物モデル系は、組み込みおよび長期のトランス遺伝子発現の同定と確認
のためのベクター組成物のインビボスクリーニングに有用である。
C.AAVヘルパープラスミドの作製
repおよび/またはcap遺伝子の発現レベルによりrAAVパッケージン
グが制限されるかどうかを調べるために、一連のヘルパープラスミドを作製した
(図11)。RepおよびCapタンパク質の発現は、内因性AAVプロモータ
ー(p5とp40)を、それぞれRSV LTRとCMV IEプロモーターで
置換することにより増加させた。出発のヘルパープラスミドであるpIM45(
マカーティー(McCarty)ら、1991)は、野生型AAVゲノムを包含するが
末端繰り返しを持たない配列を含有する(ヌクレオチド145〜4493)。p
IMRSVは、pIM45を修飾したもの(RSV LTRがp5を置換してい
る)である。p40はRepコード領域内に位置するため、p40をCMV I
Eプロモーターで置換できるようにするために(p5repΔ−CMVcapの
ように)、rep遺伝子とcap遺伝子を分離した。この方法により、p40と
CMVプロモーターの下流に431塩基対の配列の直接の繰り返しを有するベク
ターが作成された。組換えによる野生型AAVの生成を防ぐために、この作製
体のrep ORF内に存在するp40プロモーターを、部位特異的突然変異誘
発により不活性化した。pIM45のように内因性AAVプロモーターからre
pおよびcap遺伝子を発現するように、しかしp5repΔ−CMVcapに
直接比較できるように、p5repΔ−p40capを作製したが、Repおよ
びCapコード領域は分離した。RSVrepΔ−CMVcapとRSVrep
Δ−p40capは、それぞれp5repΔ−CMVcapとp5repΔ−p
40capの誘導体であり、p5はRSV LTRで置換されている。
D.AAVヘルパープラスミドからのrepおよびcap遺伝子発現
各AAVヘルパープラスミドから発現されるRepおよびCapタンパク質の
量を、ウェスタンブロット解析により推定した(図12)。Adts149感染
(MOI=20)の存在下で293細胞にトランスフェクションして産生された
4つのRepタンパク質は、293細胞を野生型AAV(wtAAV)とAdt
s149で同時感染した後に検出される対応するタンパク質と一緒に泳動する。
各ヘルパープラスミドについて、Rep78とRep52が、産生される主要な
タンパク質である。スプライスされたメッセージから翻訳されるRep68とR
ep40は、低レベルで観察された。これらはまた、wtAAV感染で主要でな
いタンパク質として検出された。
p5プロモーターをRSV LTRで置換すると、Rep78のレベルの上昇
が観察された。これは、3つのすべてのヘルパー(pIMRSV、RSVrep
Δ−CMVcapおよびRSVrepΔ−p40cap)についてであった。す
べての作製体でRep52はp19転写体から得られたため、Rep52の量に
変化はなかった。
3つのカプシドタンパク質(VP1、VP2、およびVP3)は、すべてのヘ
ルパープラスミドから、wtAAV感染で観察された1:1:10の比で産生さ
れた(図12)。p40をCMV IEプロモーターで置換すると、すべての3
つのカプシドタンパク質の合成が増加した(図12レーン2対レーン4)。しか
し、RSV LTRからのrep遺伝子の発現は、p40からのcap発現に対
してダウンレギュレーション作用を有するようであった。すなわち、repのプ
ロモーター制御性発現としてp5を含有する親プラスミドに対して、pIMRS
Vについてはカプシドタンパク質のレベルが低下した(pIM45;レーン3と
2を比較されたい)。CMV IEプロモーターからのカプシドタンパク質発現
について、同様のしかしあまり顕著ではない作用が観察された(レーン5対レー
ン4)。後者の場合、cap mRNAにもノーザン解析で対応する低下が見ら
れ、Rep78の過剰発現により、CMVプロモーターの転写性ダウンレギュレ
ーションを引き起こすことを示唆していた。AAVpAシグナルがrepおよび
cap ORFを分離しているp5repΔ−p40cap中のように、rep
およびcap遺伝子を、別の転写単位から発現した時、Capタンパク質合成も
またpIM45に対して低下した。
一過性トランスフェクションで産生されるAAVタンパク質の全レベルは、M
OI 10でwtAAV感染で観察されたものと匹敵したことに注目されたい。
Rep78、Rep52およびカプシドタンパク質は、AAVプロモーター(そ
れぞれ、p5、p19およびp40)から発現した時、wtAAV感染中で没さ
れたものと類似のレベルで現れ、異種プロモーターであるRSV LTRとCM
V IEからの発現は、ウイルス感染で観察される以上の量に増加させた。これ
は、トランスフェクション効率は20〜50%の範囲であるのに、MOI 10
での感染はより高い効率で起きることを考慮すると、特に重要である。これはト
ランスフェクションされた細胞当たりの各ウイルス遺伝子産物の濃度は、wtA
AV感染より一過性トランスフェクションにおいて高いことを示唆する。
E.pIM−RSVamの作製およびcap遺伝子ダウンレギュレーションの解 析
RSV LTR−rep遺伝子カセットを含有するヘルパープラスミドでのカ
プシドタンパク質の発現のダウンレギュレーションの機構をさらに解析するため
に、rep ORF内にアンバー突然変異を有するpIMRSVの誘導体を作成
した(pIMRSV−am)。もしダウンレギュレーションがシスの変化(すな
わち、p5をRSV LTRで置換)によるなら、これはアンバー突然変異体中
で保持されるはずである。これに対して、もしこれが完全長Repタンパク質の
合成に依存するなら、ダウンレギュレーション作用は、突然変異体中で緩和され
るはずである。
アデノウイルス(Ad)感染の存在下および非存在下でヘルパーとしてpIM
RSV−amを使用して、293細胞中で一過性トランスフェクションを行なっ
た。核タンパク質を単離し、ウェスタンブロットで解析した(図13)。pIM
45をヘルパーとして使用すると、293細胞中でEIAとEIB遺伝子の発現
のために、Adの非存在下で高レベルのRep78が現れ、従ってAdの添加に
より見かけ上誘導がなくなったことを示した(この作製体によるRep78の誘
導はHeLa細胞で観察される)。Adで感染させると、スプライスされたRe
pタンパク質、Rep68およびRep40が出現し、Rep52のレベルがわ
ずかに上昇する。予測されるように、RSVプロモーターからのRep78の発
現は、Ad感染には応答せず、ヘルパーウイルスの同時感染の存在下および非存
在下で同じ高レベルが現れる。pIMRSV−amでトランスフェクションした
細胞では、少量の完全長Rep78が観察され、突然変異が必ずしも完全でない
ことを示している。細胞をpIMRSV−amとアンバーサプレッサーtRNA
とで同時トランスフェクションすると、pIMRSVで観察されたレベルまでR
ep78の産生が回復する。pIMRSV−amをRep発現性プラスミドであ
るpRSVrepと同時トランスフェクションすると、高レベルのRep78が
トランスで得られる。
カプシドタンパク質発現を平行して解析した(図13)。pIM45でトラン
スフェクションした細胞をAdで感染すると、カプシドタンパク質の合成が有意
に増加した。この増加はpIMRSVでは観察されない(レーン5と6)が、p
IMRSV−am突然変異体では起きる。pIMRSV−amをサプレッサーt
RNAと同時トランスフェクションするか、またはpRSVrepで同時トラン
スフェクションしてトランスでRepタンパク質を供給すると、pIMRSV表
現型が回復する。
同じ実験の試料についてノーザン解析を行って、RNAレベルでこの現象を調
べた(図14)。pIM45では、比較的低レベルのp5転写体(4.2kb)が
観察されたが、p19転写体(3.6kb)の方がより多く存在し、Ad感染で増
加した。p5とp19からのスプライスされた転写体(それぞれ3.9および3
.3kb)は検出されなかった。p5をRSV LTRで置換すると、この作製体
に
より産生されるRep78のより高レベルに寄与する4.2kbの転写体が増加し
た。興味深いことに、アンバー突然変異を導入するとこの転写体の量はさらに増
加した。サプレッサーtRNA(レーン5)の同時トランスフェクションにより
Repの合成を回復するか、またはpRSVrep(レーン6)との同時トラン
スフェクションによりトランスで供給すると、4.2kbの転写体の合成は再度低
下した。これらの結果は、RSV LTRからの転写はRepによりダウンレギ
ュレーションされていることを示唆する。
pIM45では、Ad感染により、完全長(2.6kb)およびスプライスされ
た(2.3kb)p40 mRNAの両方が顕著に増加し、カプシドタンパク質合
成の増加を反映する。p5やp19から得られるものよりp40転写体が多いこ
とは、アデノウイルス感染KB細胞でwtAAVからラボウ(Labow)ら(19
86)により観察されたものと類似している。一般に2つのp40 mRNAの
比率は、Ad感染によりスプライスされた2.3kb転写体の方が多くなっている
。pIM45に対して、pIMRSV作製体ではAd感染によりp19またはp
40転写体のレベルの増加は観察されない。顕著なことは、pIMRSV−am
では、ウェスタン解析(図13、レーン14)で観察されたAd感染によるカプ
シドタンパク質合成の増加は、cap mRNAのレベルの増加により反映され
ないことである。pIMRSV−amで観察されるカプシドmRNAのレベルは
、親のプラスミドであるpIMRSVのレベルと似ている。pIMRSV−am
とサプレッサーtRNA、およびpIMRSV−amとRepを発現するプラス
ミドpRSVrepとの同時トランスフェクションで、同じ低レベルが観察され
る。ノーザン解析は、pIMRSVで観察される低レベルのカプシドタンパク質
合成は、Ad感染に応答してp40からの転写を活性化することができないこと
によるRNAレベルで説明できるかも知れないことを示唆する。さらに、これら
の結果は、pIMRSV−am突然変異体によりAd感染した後のカプシドタン
パク質産生の増加は翻訳後の作用であり、突然変異がカプシド合成に対するRe
p介在性の翻訳阻害作用を緩和したことを、示唆する。
F.rAAVパッケージング:RSVrepとCMVcapヘルパープラスミド の比較
ヘルパープラスミドのシリーズを、rAAVを産生する能力について比較した
。pTRlacZベクタープラスミドとともにそれぞれをAd感染293細胞に
トランスフェクションし、粗溶解物中のrAAVの収率(表1)を、力価測定法
により測定した。cap遺伝子をCMVプロモーター(p5repΔ−CMVc
ap)の制御下に置いてカプシドタンパク質発現を増加させると、rAAV収率
が約9倍増加した。これに対して、rep遺伝子発現を増強するためにp5をR
SV LTRで置換すると、rAAV収率が低下した。p40を含有する作製体
(pIMRSVまたはRSVrepΔ−p40cap)にRSV LTRを加え
ると、rAAVの力価が10〜20倍低下し、一方RSVrepΔ−CMVca
pヘルパーは、p5を含有する同等の作製体よりrAAVのパッケージング効率
が5倍低かった。これらの結果は、ウェスタン解析で観察されたカプシドタンパ
ク質発現の差と相関する。前述のように、p40−cap作製体とのRep78
の過剰産生の結果として、カプシドタンパク質産生の大幅な低下が観察され、C
MVプロモーターからのカプシドタンパク質発現でより小さな作用が観察された
。異なるヘルパー作製体を比較するこれらの実験の結果は、rAAVの産生にお
いてCap(しかし、Repタンパク質合成ではない)が律速因子であることを
示唆する。表は、プラスミド上で必要なアデノウイルス初期遺伝子(E1Aおよ
びE1B、E4、E2A、VA)を提供することにより、アデノウイルスの非存
在下で組換えAAV(rAAV)を提供することの実現可能性を示す実験の結果
を提供する。
表1は、アデノウイルスの存在下での従来の産生法(293標準条件)からの
rAAVの収率の、アデノウイルスの非存在下で得られたものとの比較である。
「アデノウイルスの非存在下」の場合では、いくつかの異なる細胞株を使用した
。各細胞株は、アデノウイルスE4遺伝子も含有するように作成した293細胞
株(これはアデノウイルスE1遺伝子を含有する)の誘導体である。VK2−2
0細胞株は、フランク・グラハム(Frank Graham)から得、他のORF6株(2
C4と3B1)は実験室で作成した。VK2−20株とORF6株は、rAAV
を産生するために、すでにE1A、E1BおよびE4遺伝子を含有するため、E
2AとVA遺伝子は、E2Aプラスミドでトランスフェクションして供給する必
要
がある。E2VA5’→3’およびE2VA3’→5’は、このプラスミドの2
つのクローンである(挿入体は反対の方向にある)。(細胞はまた、rAAVを
産生できるように、rAAVベクタープラスミドとヘルパープラスミドでトラン
スフェクションされる)。この実験の結論は、アデノウイルスの非存在下でrA
AVを産生することは可能であり、この方法は、従来法と(より多くではなくて
も)同程度に多い収率がある、というものである。
表2は、ヘルパーDNAとしてpIM45とp5repΔ−CMVcapを使
用する大規模rAAV産生の結果である。修飾ヘルパーを使用すると、rAAV
IU/細胞の収率はほとんど10倍増加することが顕著である。この結果はまた
、精製された物質の高力価(IU/mlと粒子/mlの両方で)に反映されている。表
には、アデノウイルス(Adts149)の存在下および非存在下で測定したIU
/mlを示す。他の研究者(フェラーリ(Ferrari)ら、1996、フィッシャー
(Fisher)ら、1996)が報告しているように、rAAVの力価は、Ad感染
の存在下では約1000倍高い。これらの調製物の粒子:IU比は、20〜12
0(IU+Ad)または4〜7×104(IU−Ad)である。前者の値は、す
でに報告されている範囲内である(サムルスキー(Samulski)ら、1989)。
精製法により、持続しているが可変レベルのAdts149汚染(<103 IU/
ml〜107 IU/ml)を与えるが、ストックはwtAAVの汚染がない(下記参照
)。
H.wtAAVの複製と汚染レベルの解析
ベクターの複製を測定し、wtAAV汚染のレベルを評価するために、前述の
ような小規模の一過性トランスフェクションからの試料について、ハート(Hirt
)解析を行なった。すべてのヘルパーDNAは、TRlacZベクターの複製を
支持した(図15、パネルA)が、RSV LTR−repカセットを含有する
ヘルパープラスミドを用いる各トランスフェクションでは、ベクターの複製レベ
ルは低下しているようであった(臭化エチジウムで染色したゲルは、等量のDN
Aが各レーンにあることを示した)。これは、RSV−repを含有するヘルパ
ーで得られたウイルス収率の低下を説明する助けになるかも知れない。アンバー
突然変異体であるpIMRSV−amをヘルパーとして用いると低レベルの複製
が観察され、図13のパネルAに示すように、少量の完全長Repタンパク質が
この突然変異体により合成されることを確認していた。同じブロットをwtAA
Vゲノムからの断片とプローブ結合させると、wtAAVウイルスDNAがウイ
ル
ス複製している証拠は観察されなかった(図15、パネルC)。しかし、あるレ
ーンで高分子量DNAへのハイブリダイゼーションがあった。これは、wtAA
Vプローブとある程度の相同性を有する細胞性配列との交差ハイブリダイゼーシ
ョンであるかも知れない。しかし、陰性対照(にれ)レーンにシグナルは現れな
かったため、別の説明は、このシグナルは293細胞ゲノムへのヘルパーDNA
の組み込みの証拠であるというものである。興味深いことに、この高分子量バン
ドは、RSV LTR−repではなくp5−repカセットを有するヘルパー
でトランスフェクションした細胞でのみ現れ、repの過剰発現は組み込みを阻
害するか、またはp5内の配列(すなわち、Rep結合部位またはRRS)が組
み込みにはシスで必要であるかも知れないことを示唆している。この組み込み仮
説を支持するのは、突然変異体pIMRSV−amでトランスフェクションした
細胞ではシグナルは現れないという観察であり、この現象がRep合成に依存す
ることを示唆している。これらのトランスフェクションから平行して採取した溶
解物を使用して、Adts149の存在下で293細胞の第2のプレートを感染
させ、Hirt試料を調製した。もし実際に少量の混入wtAAVが存在するな
ら、ウイルスは再感染で増幅されているであろう。サザン解析とwtAAVプロ
ーブを用いるハイブリダイゼーション(図15、パネルD)は、wtAAVウイ
ルスDNAが複製している証拠を示さなかった。二重測定のブロットをlacZ
断片(図15、パネルB)とプローブ結合させても、複製しているベクターDN
Aは観察されなかった。これらの条件下でwtAAV(すなわち、rep遺伝子
発現)の存在はベクター複製を可能にしたであろうから、この後者の結果は、w
tAAVが欠如していることのさらなる証拠である。
rAAVパッケージングに必要なRepおよびCapタンパク質の発現を別々
に増強するために、標準的ヘルパープラスミド中のAAVプロモーターを強い異
種プロモーターで置換した。これらの実験は、cap遺伝子の発現が増加すると
rAAVの収率が約10倍改善されることを示し、カプシドタンパク質のレベル
がrAAVの産生の1つの律速因子であることを示唆した。これに対して、re
pの過剰発現はrAAVの収率を増加させなかったため、rep遺伝子の発現は
おそらく律速因子ではない。しかし、Repタンパク質合成の増加はいつもカプ
シドタンパク質産生の低下と一緒であったため、この点に関して結論づけをする
ことはできない。しかしプラスミドpRSVrepΔ−CMVcapの場合は、
capタンパク質の産生は、p5repΔ−CMVcapで観察されたものに比
較してほんのわずかに低下(せいぜい2倍、しかしそのレベルはpIM45で得
られたものよりまだ高かった)しており、一方Rep78発現は有意に増加して
いた(約5倍)。これらの条件下で、p5repΔ−CMVcapに対してrA
AVの収率の増加はなく、実際パッケージング効率は、わずかに低下していた。
これらの結論は、p5がrep発現を制御した作製体(pAAV/Ad)に比較
して、HIV LTRからrepを発現する作製体(pHIVrep/p40c
ap)を使用するとrAAVの収率が10倍増加したという従来の研究(フロッ
テ(Flotte)ら、1995)に基づく結論と矛盾する。
RepおよびCapタンパク質合成の全体のレベルは、DNAを有する細胞の
数および各細胞内に存在するDNA分子の数の両方により制限されるため、標準
的プロトコールによるAAVベクター産生を制限する別の関連する因子は、トラ
ンスフェクション効率である。トランスフェクション効率を上昇させるため、プ
ラスミドDNAを、ポリリジンで修飾した複製コンピタントなアデノウイルスと
複合体を形成させると、rAAVパッケージングが、標準的リン酸カルシウム法
(マモウナス(Mamounas)ら、1995)に対して40〜240倍上昇した。標
準的rAAV産生法に対する多くの修飾は、チオリニ(Chiorini)ら(1995
)により行われ、293細胞をリン酸カルシウムでトランスフェクションする代
わりに、COS細胞を、90%までの報告されたトランスフェクション効率で電
気穿孔した。これらの研究で使用されたヘルパープラスミドはまた、SV40レ
プリコンを含有し、おそらく各トランスフェクション細胞内のrepおよびca
p遺伝子のコピー数を増加させた。この方法で、103 rAAV粒子/細胞を超
えるパッケージング効率が達成された。あるいは、非効率的なトランスフェクシ
ョン工程を避けるために、パッケージング細胞株が作製されている。ベクターD
NAを安定な細胞株に導入した時、rAAVの収率が同時トランスフェクション
に対して5倍上昇し、104粒子/細胞が得られたことが報告されている(フロ
ッテ(Flotte)ら、1995)。クラーク(Clark)ら(1995)は、アデノ
ウ
イルスの感染のみでrAAVの産生を可能にする、ベクターとAAV rep/
cap遺伝子を含有する細胞株を作製した。この系は30IU/細胞(約400
粒子/細胞)を与え、これは本明細書に記載の改良されたヘルパープラスミドで
達成されたものに匹敵する値である。他の研究者の経験を考えると、これらの実
験で使用されたパッケージングプロトコールは、トランスフェクションした細胞
内のヘルパープラスミドを複製するか、または新しいヘルパー作製体を使用して
パッケージング細胞株を作成することにより、さらに改良できる可能性がある。
p5をRSV LTRで置換して得られたカプシドタンパク質発現に対して観
察された作用は、AAV遺伝子セグメントに関して他の研究者の研究を確認した
。複製での機能以外に、AAVRepタンパク質(主にRep78/68;キョ
スチオ(Kyostio)ら、1994;ホラー(Horer)ら、1995)は、転写レギ
ュレーターとして作用することが知られている。アデノウイルス感染がないと、
Repタンパク質はAAVプロモーターからの転写を抑制し(トラッチン(Trat
schin)ら、1986、トレンペとカーター(Trempe and Carter)、1988、
ビートン(Beaton)ら、1989、キョスチオ(Kyostio)ら、1994)、一
方逆に、アデノウイルス感染に応答してこれらのプロモーターからの転写を活性
化する(ラボウ(Labow)ら、1986、トラッチン(Tratschin)ら、1986
)。マカーティ(McCarty)ら(1991)は、アデノウイルスの存在下でのp
19とp40プロモーターのRep介在活性化は、p5とp19の両方の上流に
存在する配列にシスで依存することを証明した。この知見と一致するのは、p5
を欠失させRSV LTRで置換(pIMRSVプラスミドのように)するとア
デノウイルス感染でp40転写の誘導がなかったことである。同様にこの作製体
ではp19 RNAのレベルの増加もなかった。誘導が観察されないのは、これ
はrep遺伝子の発現とは無関係に起きたため、シスで必要な配列が除去された
ためであった。Repタンパク質合成をアンバー突然変異で妨害した時またはR
epタンパク質をトランスで供給した時、p40の転写活性化は回復しなかった
(図4)。pIM45に対して、pIMRSVはp5の上流に84塩基対(塩基
対191〜275)のみ欠如し、この欠失は、マカーティ(McCarty)ら(19
91)により報告されたもの(塩基対191〜320)より限定されて
おり、従ってRep活性化に必要な推定の制御領域の位置をさらに規定している
。塩基対191と275の間の領域は、主要後期転写因子(USF;チャン(Ch
ang)ら、1989)、YY1(シ(Shi)ら、1991)およびRep(マカー
ティ(McCarty)ら、1994;キョスチオ(Kyostio)ら、1995)ならびに
p5 TATAボックスの結合部位を含有することが知られている。
pIMRSV−am突然変異体のp40プロモーターからの転写は、アデノウ
イルス感染に応答してRepにより活性化されなかったが、カプシドタンパク質
合成は増加することが観察された。この作用は、Repの翻訳阻害活性に起因す
るかも知れない。アデノウイルス感染の非存在下の293細胞では、トレンペと
カーター(Trempe and Carter)(1988)は、rep遺伝子含有ベクターに
比較してRepの非存在下では、p40 mRNAのレベルが低下し、CATタ
ンパク質発現が増加することを観察した。pIMRSVamでトランスフェクシ
ョンした細胞では、アデノウイルス感染によりカプシドタンパク質の合成は有意
に上昇している。しかしこの増加は、p40 mRNAの定常状態に変化無しで
起き、これは翻訳作用であることを示している。これと比較して、pIM45で
トランスフェクションした細胞ではカプシドタンパク質産生も増加するが、この
場合2.6kbおよび2.3kbのp40 mRNAのレベルも同時に増加する。p
IMRSVamによるカプシドタンパク質の合成の見かけの誘導は、Repタン
パク質が発現される場合は起きないため、rep遺伝子の突然変異のトランス作
用である。Rep78はpIMRSVにより産生される主要なRepタンパク質
であるため、これはおそらく阻害作用の主要なメディエーターであるが、Rep
68の役割は排除できない。これらは結果は、アデノウイルス感染はp40 m
RNAの翻訳の効率を有意に上昇させることができる(ウェスト(West)ら、1
987;ジャニク(Janik)ら、1989)が、この作用はRepタンパク質に
より打ち消されることを示唆する。アデノウイルスの存在下での翻訳阻害が、R
epの正常な機能として起きるのか、またはpIMRSV作製体中のrep遺伝
子の過剰発現のアーティファクトであるのかは不明である。あるいは、阻害は、
p40 mRNAのレベルが低い時にのみ起き、p40からの転写がアデノウイ
ルス感染により普通に増加する時打ち消されるのかも知れない。p40からの転
写の誘導は、この場合p5の上流の配列の除去により妨害された。
これらの実験は、Repタンパク質が異種プロモーターからの発現のレプレッ
サーとして作用できることのさらなる証拠を提供する。Repタンパク質は、い
くつかの異種遺伝子の発現をダウンレギュレーションすることが知られている(
ラボウ(Labow)ら、1987;アントニ(Antoni)ら、1991;リットナー
(Rittner)ら、1992;オエルゼ(Oelze)ら、1994;ホラー(Horer)
ら、1995)。前述の実験では、CMV IEプロモーターからのcap遺伝
子の発現ならびにRSV LTRからのrep遺伝子の発現の両方とも、Rep
によりダウンレギュレーションされた。Repは、CMV IEプロモーターか
らのcat遺伝子の発現のレベルを低下させることがすでに証明されており(ヘ
イルブロン(Heilbronn)ら、1990)、本明細書で得られた結果と同様に、
この作用は小さかった(約2倍)。CMV IEとRSV LTRプロモーター
の両方について、阻害はRNAレベルで起きたが、RNAの定常状態はノーザン
解析により測定されたため、この作用は転写またはmRNA安定性のレベルであ
るかも知れない。RNAレベルでのダウンレギュレーションもまた、HIV L
TRとHPV18 URRプロモーターの場合に証明されており、主にRep7
8/68が原因であるとされている(アントニ(Antoni)ら、1991;ホラー
(Horer)ら、1995)。
以下の例は、本発明を例示するものであり、その範囲を限定するものではない
。例1:AAVヘルパー遺伝子を含有する複製していないヘルパープラスミドを使 用する組換えAAV産生
複製していないプラスミド上のAAVrepおよびcap遺伝子を提供し、遺
伝子発現のために異種プロモーターを使用するヘルパープラスミドが、repお
よびcap遺伝子がそれ自身のプロモーターから発現される対照から得られるも
のより高く、AAVベクターの力価を増加させるかどうかを調べるために、実験
を行なった。試験したヘルパープラスミドを図7に示す。
pIM45は、未変性のAAVプロモーターの制御下のAAV repおよび
cap遺伝子を含有し、3’末端にAAVポリA部位を含有する(ディー・エム
・マカーティ(McCarty,D.M.)ら、J.Virol.65: 2936-2945,1991)。
AAVゲノム中のヌクレオチド2287〜4049を削除するために、PCR
を用いてpIM45からcap領域を削除して、pRep*30を作成すること
により、pRSVrep−p40capとpRSVrep−CMVcapを作製
した。NheIとNotI末端を含有する、pRep*30から単離したPCR
断片のヌクレオチド275〜4464を、pRep9(インビトロゲン(Invitr
ogen)、サンジエゴ、カリホルニア州)のNheIとNotI部位の間にクロー
ン化して、pRSVrepを作成した。SmaIとSfiIで消化したpIM4
5中に、pRSVrepからのXbaI(代用)−SfiI断片をクローン化し
て、pRSVrep−p40capを作成した。pRSVrepからのこの同じ
XbaI(代用)−SfiI断片を、SmaIとSfiIで消化したpIM−C
MVcap(後述)にクローン化して、pRSVrep−CMVcapを作成し
た。
p40プロモーターを不活性化するためにAAVゲノムに1823位(T:C
)、1826位(A:C)、および1832位(G:A)で3点突然変異を導入
して、pIM−CMVcapを作製した。AAVゲノムからのヌクレオチド18
50〜4460とBamHI末端を含有するPCR断片をpCMVBのBamH
I部位に挿入して、pCMVcapBを作成した(クローンテク(Clonteck)、
パロアルト、カリホルニア州)。CMVプロモーターを含有するSphI断片を
pCMVcapBから単離し、pIM45内の930位のSphI部位に挿入し
て、pIM−CMVcapを作成した。
ヘルパー/ベクターの比10:1で293細胞にヘルパープラスミドをトラン
スフェクションした(16.5μg/総DNA)。使用したAAVベクタープラ
スミドはpTR−lacZであり、これは、フロリダ大学のニコラス・ムジクズ
カ(Nicholas Muzyczka)博士により開発された。このプラスミドを図8に示す
。AAVベクターの単離と精製は、セクション4(前述)に記載のように行なっ
た。
293細胞をヘルパーアデノウイルスとAAVで同時感染させて、AAVの収
率を測定した。これにより感染時間が減少し、従って測定の感度が上昇した。力
価測定のために、293細胞を96ウェルプレートに、DMEM/10%FBS
(ペニシリン/ストレプトマイシンおよびグルタミンを含有)中で5×105細
胞/ml(100μl/ウェル)で蒔き、37℃で1日間増殖させた。次にMOI
20を使用してAdts149ウイルスと、ウイルス準備ストックの1:10
0、1:200および1:400などの希釈率のAAVを用いて細胞を同時感染
させた。力価を確認するために、異なる希釈率を使用した。
39℃で2日間感染を進行させた後、培地を吸引し、細胞を3.7%ホルムア
ルデヒドで5分間インキュベートし、リン酸緩衝化生理食塩水(PBS)で洗浄
した。次に細胞をX−Gal(5−ブロモ−4−クロロ−3−インドリル−β−
D−ガラクトピラノシド)を用いて37℃で1〜24時間染色し、細胞中の青色
の着色の存在についてスクリーニングして、AAVベクター中に含有されるla
cZ遺伝子の発現を検出した。終点希釈の決定に基づく力価測定解析プログラム
(Titer Analysis program)を使用する換算を用いて、力価(IU/ml)を測定し
た。
図7に示す種々のヘルパープラスミド(および特異的クローン)を使用して、
repおよびcapタンパク質発現のレベルを測定するために、ウェスタンブロ
ット解析を行なった。図9は、repタンパク質発現のウェスタンブロット解析
を示し、図10はcapタンパク質発現のウェスタンブロット解析を示す。ウェ
スタンブロット解析において標準的方法を使用した(Current Protocols in Mol
ecular Biology,エフ・アウスベル(Ausubel.,F.)ら編、ワイリーアンドサン
ズ(Wiley and Sons)、ニューヨーク州 1995)。
力価データを表3に示す。ベクターのストックの力価をIU/mlで示す。実験は
、ウェスタンブロットで証明される特にcap発現のレベルの増加により、記載
の力価により証明されるように、AAVベクターであるpTRlacZの産生が
増加することを証明する。
例2:細胞株、ウイルスおよびプラスミドDNA
10%胎児牛血清(FBS:アービンサイエンティフィック(Irvine Scienti
fic)、サンタアナ、カリホルニア州)、20mMグルタミン、100単位/ml
ペニシリンおよび100μg/ml ストレプトマイシン(ギブコビーアールエル
(Gibco-BRL)、ゲーサーズバーグ(Gaithersburg)、メリーランド州)を補足
したダルベッコー改変イーグル培地−高グルコース(DME:アービンサイエン
ティフィック(Irvine Scientific))中で、293細胞細胞株であるアデノウ
イルス5−形質転換ヒト胚腎細胞株(グラハム(Graham)ら、1977)
を、37℃で5%CO2中で増殖させた。これらの実験でヘルパーウイルスとし
て使用したアデノウイルス5型突然変異体であるts149(Ad5ts149
;エンシンガーとギンスバーグ(Ensinger and Ginsberg)、1972)は、ア
デノウイルス初期領域2(スティルマン(Stillman)ら、1982)によりコー
ドされるDNAポリメラーゼ中の温度感受性突然変異のために、非許容性温度(
39℃)でウイルスDNAを複製する能力が低下している。Ad5ts149を
許容性温度(33℃)で293細胞中で増殖させ、CsCl濃度勾配遠心分離に
より精製した。
組換えAAVベクターであるpTRlacZならびにヘルパープラスミドpI
M45(マカーティ(McCarty)ら、1991)をコードするプラスミドDNA
は、エヌ・ムジクズカ(N.Muzyczka)(フロリダ大学)により供与された。p
TRlacZは、AAVの末端繰り返し配列の間に挿入された、CMV IEプ
ロモーターの転写制御下にある大腸菌(E.coli)LacZ遺伝子(細胞質性)
から構成される。アンバーサプレッサーtRNAであるpSVtsSu+(アン
バー)をコードするプラスミド(カポネ(Capone)ら、1985)は、ユー・エ
ル・ラジバンダリー(U.L.RajBhandary)(MIT)から得た。repコード領
域内にアンバー突然変異を含有するAAVゲノムクローンであるpNTC3(チ
ェジャノブスキーとカーター(Chejanovsky and Carter)、1989)は、アー
ル・オーエンス(Rl.Owens)(NIH)により供与された。
A.プラスミド作製
出発プラスミドとしてpIM45を使用して、内因性AAVプロモーター(p
5とp40)を、それぞれラウス肉腫ウイルスの長い末端反復配列(RSV L
TR)とCMV IEプロモーターで置換した。すべての操作は、標準的クロー
ニング法に従って行なった(サムブルーク(Sambrook)ら、1989)。すべて
の制限酵素およびDNA修飾性酵素は、ニューイングランドバイオラボズ(New
England Biolabs)、ビバリー(Beverly)、マサチューセッツ州)から得て、製
造業者の説明書に従って使用した。プラスミドDNAは、キアゲン(Quiagen)
(チャッツワース(Chattsworth)、カリホルニア州)から得られるキットを用
いて精製した。
まず塩基対1852と4440の間のAAVゲノム配列から構成されるDNA
断片(カプシドタンパク質をコードし、AAV mRNAポリアデニル化部位を
含む)をVentポリメラーゼ(ニューイングランドバイオラボズ(New Englan
d Biolabs)、ビバリー(Beverly)、マサチューセッツ州)を使用してPCR(
サイキ(Saiki)ら、1988)により増幅して、CMV IE−capカセッ
トを作製した。この断片をpCMVβ(クローンテク(Clonteck)、パロアルト
、カリホルニア州)のBamHI部位の間に挿入して、プラスミドpCMVca
pを作成した。
Repをコードする最小の配列を得るために、BamHI部位(塩基対104
5)とApaI(塩基対2283)の間のrep遺伝子配列をPCRで増幅し、
BamHIとApaIで消化したpIM45プラスミド内に挿入した。その結果
は、塩基対2283(Rep停止コドンのすぐ下流)と塩基対4049のApa
I部位の間の欠失である。このプラスミド(pIMrepΔ)を使用して、RS
V LTRからRep78/68 mRNAが発現される作製体を得る。塩基対
276(Rep78/68mRNA開始コドンのすぐ上流)から塩基対4459
に伸長する2.4kbのrep遺伝子断片を、pIMrepΔからPCRで増幅し
、pRep9発現ベクター(インビトロゲン(Invitrogen)、サンジエゴ、カリ
ホルニア州)のNheI部位とNotI部位の間に挿入してpRSVrepを作
成した。
AAVイントロンのこの領域中でRepおよびCapタンパク質コード配列は
重複するため、pIMrepΔとpCMVcapの間(塩基対1852と228
3の間)に431塩基対が共通して存在する。pIMrepΔにCMV IE−
cap断片を挿入してp5repΔ−CMVcapを作成する前に、pIMre
pΔ内のp40配列を突然変異させてプロモーターを不活性化させた。これは、
共通の配列の間の組換えの結果として、野生型AAVが作成されるのを防ぐため
に行なった。重複伸長PCR(ヒグチ(Higuchi)ら、1988)により突然変
異誘発を行なった。隣接するプライマー−1(5'-GGATTACCTCGGAGAAGCAGTGGATCC
-3’;AAVゲノムの塩基対1024〜1050)と突然変異誘発性プライマー
−1(5'-GTTTGGGTTCACTGATGTCTGCGTCACTG-3’;AAV塩基対1821〜184
1;突然変異したヌクレオチドに下線をしてある)を使用して、第1のPCRの
鋳型としてpIMrepΔを使用した。この結果は、p40 TATAボックス
の領域中への3つの塩基対突然変異の導入である:TATAAGTGAGからCATCAGTGAA)
。GからAへの変化は、BanII部位を除去して、制限解析によるスクリーニン
グを可能にする。隣接するプライマー−2(5'-GTGTGGAATCTTTGCCCAGATGGGCCCGG
TTT-GAGCTTC-3';AAV塩基対2260〜2283、4049〜4066)と突
然変異誘発性プライマー−2(5'-CAGTGACGCAGACATCAGTGAACCCAAACG-3';AAV
塩基対1821〜1841)を使用して、第2のPCRの鋳型としてpIMre
pΔを使用した。上記PCR産物をゲル精製後、最初の2つの生成物をアニーリ
ングし、隣接プライマーのみを用いて最終増幅工程を行って第3のPCRを行い
、こうして1285塩基対のDNA断片を作成した。この断片をBamHIとA
paIで消化し、pIMrepΔ骨格の対応する部位にクローン化した。得られ
たプラスミドはpIMrepΔ/p40Δである。CMV IEプロモーターと
cap遺伝子カセットを含有するpCMVcapからのSphI断片を、pIM
repΔ/p40ΔのユニークなSphI部位に挿入して、ヘルパープラスミド
p5repΔ−CMVcapを作製した。同様にp5repΔ−p40capを
作製するために、AAV塩基対1715から4461に伸長するSphI末端を
有するPCR断片をpIM45から作成し、pIMrepΔ/p40ΔのSph
I部位にクローン化した。
プラスミドpIM45、p5repΔ−CMVcapおよびp5repΔ−p
40cap中のp5プロモーター領域を、まずXbaIでpRSVrepを切断
して、RSV LTRプロモーターで置換した。DNAポリメラーゼI−クレノ
ウ断片でXbaI部位を平滑末端にし、DNAをSfiIで制限切断して、RS
Vプロモーターとrep遺伝子の5’末端を含有する断片を放出させた。次にこ
の断片を、親プラスミドのSmaI部位とSfiI部位の間にクローン化した。
pIMRSVへアンバー突然変異を導入するために、突然変異(AAVゲノム
の塩基対1033で)を含有するSfiI−BamHI断片を、プラスミドpN
TC3(チェジャノフスキー(Chejanovsky and Carter)、1989)から単離
し、pIMRSVの対応する部位にクローン化した。例3:一過性トランスフェクションおよびrAAV複製とパッケージングの解析
小規模実験のために、トランスフェクションの48時間前に6cmのシャーレに
1×106細胞の密度で293細胞を接種した。トランスフェクションの前に3
7℃で1時間、感染多重度(MOI)20でDME−10%FBS中のAd5t
s149で細胞を感染させた。トランスフェクション操作は、リン酸カルシウム
プロフェクションキット(ProFection Kit)(プロメガ(Promega)、マジソン
、ウィスコンシン州)を使用して、製造業者の説明書に従って行なった。一般に
、rAAVパッケージングのために、各シャーレに1.5μgのベクターDNA
(すなわち、pTRlacZ)と15μgのヘルパーDNAの混合物を入れた。
37℃で5時間インキュベート後、培地を新鮮なDME−10%FBSで交換し
て、感染/トランスフェクションを停止させた。次にシャーレを39℃(Ad5
ts149の非許容性温度)に移した。
rAAVパッケージングのために、トランスフェクションの48時間後に臨床
的遠心分離機で低速遠心分離により細胞を採取した。各シャーレからのペレット
を100μlのリン酸緩衝化生理食塩水(PBS)に再懸濁し、凍結−融解を4
回行ってrAAVを放出させた。溶解物を56℃で30分インキュベートiして
、アデノウイルスを熱不活性化した。溶解物を2回目の低速遠心分離を行い、細
胞破片をペレットにし、上清を集めた。終点希釈法により293細胞(+/−A
d5ts149による同時感染;MOI=20)で、rAAVの力価を測定した
。細胞をX−Gal(5−ブロモ−4−クロロ−3−インドリル−β−D−ガラ
クト−ピラノシド)を用いて20〜24時間染色後、カーバー(Karber)法に基
づくコンピュータープログラム(リン(Lynn)、1992)を使用して力価を計
算した。
トランスフェクションの48時間後に、ランスフェクションされた細胞中のベ
クターDNAの複製を、ハート(Hirt)分画法(ハート(Hirt)、1967)に
従って、染色体外DNAを単離して測定した。DNAをDpnIで制限切断(導
入DNAを消化するため)してから、アガロースゲル電気泳動とサザン解析を行
なった。使用したlacZと野生型AAVプローブは両方とも50量体オリゴヌ
クレオチドであり、それぞれ5'-ACTGCTGCCAGGCGCTGATGTGCCCGGCTTCTGACCATGCGGT
CGCGTTC-3'と5'-TCGGAGGAAGCAAGGTGCGCGTGGACCAGAAATGCAAGTCCTCGGCCCAG-3'(A
AVヌクレオチド1501〜1550)であった。これらを、標準的方法(サム
ブルーク(Sambrook)ら、1989)に従ってT4ポリヌクレオチドキナーゼを
使用して[γ−32P]で標識した。フィルターをハイブリダイズさせ、ノーザン
ブロット解析のために後述するように洗浄したが、プレハイブリダイゼーション
、ハイブリダイゼーションおよび最終洗浄工程は60℃で行なった。A.タンパク質抽出と免疫ブロッティング
種々のヘルパープラスミドからのRepおよびCapタンパク質発現の解析の
ために、まず293細胞を前述のようにトランスフェクションした。メンデルソ
ン(Mendelson)ら(1986)の記載した方法に従って、トランスフェクショ
ンの48時間後に核画分を調製した。試料の容量をDNA含量(260nmの光学
密度で)に対して標準化し、15〜20μlの試料緩衝液(500mMトリス−塩
酸(pH6.8)、10%ドデシル硫酸ナトリウム(SDS)、20mM EDT
A、10%β−メルカプトエタノール、10%グリセロール、および0.2%ブ
ロモフェノールブルー)と混合し、5分間沸騰させてから使用した。
10%ポリアクリルアミド/0.1%SDSゲルで電気泳動後、タンパク質を
ゲルから、ハイボンド(Hybond)ポリビニルピロリドンジフルオリド(アマシャ
ム(Amersham)、アーリントンハイツ(Arlington Heights)、イリノイ州)膜
に移した。染色の前に、フィルターを、TBST(10mMトリス−塩酸(pH8
.0)、150mM NaCl、および0.05%ツイーン−20)に溶解した5
%ミルク粉末中で室温で1時間ブロッキングした。RepおよびCapウェスタ
ンに使用した一次抗体は、いずれもマウスモノクローナル抗体(アメリカンリサ
ーチプロダクツ(American Research Products)、ベルモント、マサチューセッ
ツ州)である:それぞれ、抗AAV Repタンパク質、303.9(TBST
中で1:10希釈率で使用した)、およびAAVの抗VP1、VP−2、および
VP−3、B1(TBST中で1:5の希釈率で使用した)。これらを、室温で
2時間フィルター上で激しく攪拌してインキュベートした。TBSTで洗浄(3
×15分)後、フィルターを、2次抗体であるヤギ抗マウスIgG(Fab特異
的)ペルオキシダーゼ結合体(シグマ(Sigma)、セントルイス、ミズーリ州)
で室
温で1時間インキュベートした。次にフィルターを前述のように洗浄し、ECL
キット(アマシャム(Amersham))を用いて発色させた。B.RNAの単離とノーザン解析
RNAzol B(テルテスト社(Tel-Test,Inc.)、フリエンドスウッド(
Friendswood)、テキサス州)を用いて製造業者の説明書に従って、トランスフ
ェクションした293細胞から総RNAを単離した。電気泳動の前に、10μg
の各RNAを変性カクテル(50%DMSO、10%ホルムアルデヒド、20mM
MOPS(モルホリンプロパンスルホン酸)、pH7.0、10mM酢酸ナトリ
ウム、1mM EDTA)とローディング(loading)色素(5%グリセロール、
0.1mM EDTA、0.04%ブロモフェノールブルー、0.04%キシレン
シアノール)と一緒にして、65℃で15分間加熱した。電気泳動は、1%アガ
ロース/0.65%ホルムアルデヒドゲルで、MOPS泳動緩衝液(20mM M
OPS、pH7.0、10mM酢酸ナトリウム、1mM EDTA)中で行なった。
毛細管作用により、10×SSC(1.5M NaCl、0.15Mクエン酸ナ
トリウム、サムブルーク(Sambrook)ら、1989)中で、ジーンスクリーン(
GeneScreen)ナイロン膜(エヌイーエヌ−デュポン(NEN-dupont)、ボストン、
マサチューセッツ州)に一晩移した。
フィルターを65℃で4〜5時間プレハイブリダイズし、次にハイブリダイゼ
ーション緩衝液(5×SSC、0.5%SDS、5×デンハルツ溶液(サムブル
ーク(Sambrook)ら、1989)、100μg/ml変性サケ精子DNA)中で6
5℃で一晩プローブとハイブリダイズさせた。プローブは、ランダムプライマー
標識キット(ストラタジーン(Stratagene)、ラホイヤ(La Jolla)、カリホル
ニア州)を使用して[α−32P]dATP(比活性、3,000Ci/mmol;エヌ
イーエヌ−デュポン(NEN-dupont)、ボストン、マサチューセッツ州)で標識し
たpIM45の1.6kbのHincII断片(AAV塩基対2397〜3987)
である。フィルターを2×SSC、0.5%SDS中で室温で5分間、2×SS
C、0.1%SDS中で室温で15分間、次に0.1×SSC、0.5%SDS
中で65℃で2時間洗浄して、フィルムに感光させた。C.大規模トランスフェクションとrAAV精製
rAAVの大規模増殖のための293細胞のトランスフェクションの前に、細
胞がトランスフェクションの日に60〜80%のコンフルエンスに達するように
、細胞をローラーボトルに接種した(最終密度は約1×108細胞/ボトル)。
トランスフェクションは、ボトル当たり脂質#53:DOPE(リー(Lee)ら
、1996)、22μgのベクターDNAおよび218μgのヘルパーDNAを
使用して、OptiMem培地(ギブコビーアールエルライフテクノロジーズ(
GibcoBRL Life Technologies)、ゲーサーズバーグ(Gaithersburg)、メリーラ
ンド州)中で行なった。細胞を、トランスフェクション時にMOI 20でAd
5ts149を感染させ、39℃で48時間インキュベートしてから、採取した
。
採取の時、静かに振盪してボトルから細胞をはがした。細胞を、ソーバル(So
rvall)RC−3Bスイング型バケットロータで遠心分離(2500rpm、4℃、
15分間)し、凍結させた。rAAVの精製のために、細胞を、2mM MgCl2
、0.7mM CaCl2、10%グリセロール、および0.1%ツイーンを含有
するPBSに再懸濁した。
ベンゾナーゼ(Benzonase)(登録商標)(ニコメッドファーマ社(Nycomed Pha
rma A/S)、コペンハーゲン、デンマーク)(10μl/1×108細胞)を加え
、懸濁液を室温で1時間振盪してインキュベートした。トリプシン(ギブコビー
アールエルライフテクノロジーズ(GibcoBRL Life Technologies))を最終濃度
0.25%になるように加え、懸濁液を室温で1時間振盪してインキュベートし
た。細胞破片を遠心分離(3000rpm、15分、4℃、ソーバル(Sorvall)R
C−3B)して、溶解物を0.45μMフィルターでろ過した。
次に溶解物を、CsClの段階勾配(4時間、26,000rpm、4℃、SW
28ロータ)で遠心分離し、ここで、上層と下層はそれぞれ、1.37g/mlと
1.5g/ml CsClであった。上層を集め(CsCl界面とAd5ts14
9バンドの間)、1.41g/ml CsClに調整し、1.41g/mlのCsC
l平衡勾配で遠心分離した(16〜20時間、4℃、35,000rpm、NVT
.65ロータ)。画分を集め、屈折計で測定し、1.36〜1.41の密度を有
する画分をプールし、PBS/1%ショ糖に対して4℃で6時間透析した。
ショ糖を最終濃度が5%になるように加え、精製したウイルスを分注して−80
℃で保存した。D.精製したrAAVストックの性状解析
精製したrAAVのストックを、前述の終点希釈法により、Ad5ts149
(MOI=20)の存在下および非存在下で力価を測定した。混入しているAd
5ts149の力価は同様の方法で測定したが、染色は、抗アデノウイルス(ヘ
キソン)/FITC結合体(ケミコン(Chemicon)、テメキュラ(Temecula)、
カリホルニア州)を使用してヘキソンについて行なった。混入している野生型A
AVのレベルは、既に記載されている(エイナーハンド(Einerhand)ら、19
95)感染中心測定法を使用して測定した。
AAV粒子力価は、サムルスキー(Samulski)ら、1989の修飾した方法を
使用して定量した。まず精製したrAAV試料を、0.1%SDS中のプロテイ
ナーゼKで処理した。適切な希釈物ならびに標準曲線DNA(TRlacZにつ
いては、pTRlacZ DNAを標準物質として使用した)を、変性溶液(0
.5M NaOH、1.5M NaCl)で室温で10分間処理し、スロットブ
ロット装置(シュレイチャーアンドシュエル(Schleicher and Schuell)、キー
ン(Keene)、ニューハンプシャー州)を使用して、1mlの容量をジーンスクリ
ーンプラス(GeneScreen Plus)(アマシャム(Amersham))膜に適用した。適
用後、スロットを300μlの0.5M酢酸アンモニウム(pH5.2)で洗浄
した。フィルターを乾燥し、前述のようにハイブリダイズした。プローブ(pT
RlacZのPvuII断片)を、プライムイットフルオア(Prime-It Fluor)標
識キット(ストラタジーン(Stratagene)、ラホイヤ(La Jolla)、カリホルニ
ア州)を使用して標識した。前述のように一連の洗浄(ただし、最後の洗浄は6
5℃で10分間行なった)後、フィルターをイルミネーター(Illuminator)検
出キット(ストラタジーン(Stratagene)、ラホイヤ(La Jolla)、カリホルニ
ア州)で発色させた。試料のシグナルを標準曲線と比較して、粒子濃度を推定し
た。例4:細胞株の作成
293−MT−DBP:プラスミド作製。親プラスミドpREP−7(インビ
トロゲン(Invitrogen)、サンジエゴ、カリホルニア州)は、プラスミド染色体
外維持のためにEBV複製開始点とEBNA−遺伝子を、そしてDNA選択のた
めにヒグロマイシン耐性遺伝子を含有する。pREP/MT/DBPを作製する
ために、pREP−7のRSVプロモーターをメタロチオネイン(MT)プロモ
ーター(これは、重金属で誘導される)で置換した。DNA結合タンパク質(D
BP)をコードするE2A遺伝子を、MTプロモーターの下流にクローン化した
。
293/MT/DBP細胞株の作成。リン酸カルシウムトランスフェクション
により、293細胞をpREP/MT/DBPクローン0.5でトランスフェク
ションした。トランスフェクションした細胞をヒグロマイシンで6週間選択した
。選択した細胞をクローンで拡大し、誘導後のDBPの発現(免疫蛍光法)とE
l−/E2Aベクターを補足する能力に基づき判定した。
3B1および2C4細胞株:親プラスミドは、アデノウイルスE4 6と6/
7読みとり枠(ORF)の発現カセットを含有する。発現を進めるのに使用した
プロモーターは、低レベルの基礎活性を有する突然変異ヒトメタロチオネインプ
ロモーターである(マカロフ(Makarov)ら、Nuc.Acids Res.22(8): 1504-150
5(1994))。
3B1と2C4細胞株の両方とも、アデノウイルス2型のE1領域で形質転換
したヒト胚腎細胞である293細胞から得られた。3B1と2C4細胞株の両方
とも、E1とE4が欠如した組換えアデノウイルスベクターを補足する能力を有
する。この細胞株は、アデノウイルス2型のE4読みとり枠6と6/7(アデノ
ウイルスヌクレオチド34082〜32913)の発現を進めるための、突然変
異ヒトメトロチオネインプロモーター、SV40スプライス、およびポリアデニ
ル化シグナルを含有する。E4機能の補足のために、ORF6と6/7の発現は
、100μM Zn2+、2μM Cd2+を添加して誘導できる。簡単に説明する
と、293細胞を親プラスミドでトランスフェクションした。各クローンがG4
18で選択できるように、細胞をpSV2Neoで同時トランスフェクションし
た。例5:AAVS1組み込み座を有するトランスジェニックマウス
非相同的組換えによるアデノ随伴ウイルスDNAの組み込みのためのヒト第1
9染色体上の好適な部位の性状解析。700〜800のCD−1マウスに精製し
たDNA断片(AAVS1の0.7kb EcoRI−SacI断片:コチン
(Kotin)ら、EMBO J.11: 5071-5078)を注入した。500〜600個の卵は生
存し続け、分割した。注入した卵を19匹のマウスに移植すると、148匹のマ
ウスの子が産まれた。
染色体DNAをマウスの尾から単離し、サザン解析によりスクリーニングした
。6匹の陽性マウスが見つかった(#66、73、85、93、123、147
)(表4)。
陽性のF0をwtCD−1と交配させ、72匹のF1子供を産ませた。F1子
孫から染色体DNAを単離し、PCR(DMSOの存在下で陽性試料から250
塩基対の断片を産生した)(表5)を使用してスクリーニングした。スクリーニ
ングの結果に基づき、全部で43匹のマウスを飼育した(表6)。
A.F1トランスジェニックの一次培養物の樹立
簡単に説明すると、尾の試料を切断し、前の試料のようにトリプシン処理した
。遊離している細胞と塊の細胞を、6ウェルプレートのDMEM中の10%子ウ
シ血清に入れた(1ウェル/尾)。4日後に細胞を解析すると、いくつかの付着
した細胞は繊維芽細胞様の形態を有した。この時点で細胞の塊を除き、新鮮な培
地と交換した。一次F1培養物からの細胞をAAV(10 MOI)で感染させ
る(AAV力価 9×10E9/ml=9×10E6/μL)(1×106細胞/
プレート)。感染の48時間後細胞を採取し、ハート(Hirt)解析を行なった。
開示された実施態様について本発明を説明したが、本発明の精神を逸脱するこ
となく種々の変更が可能である。従って本発明は、以下の請求の範囲によりのみ

Claims (1)
- 【特許請求の範囲】 1.目的のポリペプチド又はタンパク質をコードするDNA配列を細胞に提供 するための組成物であって、 AAV repタンパク質またはAAV repタンパク質をコードする核酸 配列;および目的のタンパク質もしくはポリペプチドをコードするDNA配列ま たは遺伝子転写体を含む遺伝的作製体;および第1のAAV ITRもしくはそ の一部、および第2のAAV ITRもしくはその一部(ここで、第1および第 2のAAV ITRは、目的のポリペプチドをコードするDNA配列の端に存在 し、プロモーターは、ポリペプチドをコードするDNA配列を制御する)、 からなる上記組成物。 2.AAVの第1および第2のITRと、細胞特異的発現を可能にするプロモ ーターを含む少なくとも1つのカセットとからなる、部位特異的組み込みと細胞 特異的発現のための発現ベクターであって、プロモーターは異種遺伝子に結合し 、カセットは逆末端繰り返し配列の間に存在することを特徴とする、上記ベクタ ー。 3.ITR配列は、他のプロモーターの非存在下で核酸の発現を促進する、A AVのITR配列と核酸からなるAAVベクター。 4.薬剤学的に許容される担体中の請求の範囲第2項記載のベクター。 5.請求の範囲第2項記載のベクターで細胞を感染させる、目的のポリペプチ ドを細胞に提供するための方法。 6.パッケージング細胞株293−MT−DBP(ATCC CRL1218 1)。 7.パッケージング細胞株2C4(ATCC CRL12182)。 8.パッケージング細胞株3B1(ATCC CRL12183)。 9.ヒト第19染色体のAAV組み込み座をコードするDNAを発現する、非 ヒトトランスジェニック哺乳動物。
