Claims (1)
- 【特許請求の範囲】 1.焼結温度で安定剤の存在下に、ハイドロキシアパタイトを不溶性の安定化 リン酸三カルシウムに変換することにより作製される安定化リン酸カルシウム相 を含む、その上の骨細胞活性を一貫して支持することが可能な形態を提供する生 物的に活性のある焼結組成物。 2.前記安定化リン酸三カルシウムが、主にαリン酸三カルシウムである請求 項1に記載の組成物。 3.前記組成物が、粉末、フィルム、厚いコーティング、または立体塊材料で ある請求項2に記載の組成物。 4.前記フィルムが厚さ約0.1μmから10μmである請求項3に記載の組成物。 5.焼結前のハイドロキシアパタイトが、ケイ素、アルミニウム、ジルコニウ ム、バリウム、チタン、およびそれらの混合物から成るグループから選択される 成分を含む基質上に提供される請求項3に記載の組成物。 6.前記安定剤が、焼結中に前記基質から遊離されるか、あるいは焼結前にハ イドロキシアパタイトに溶液として添加される請求項5に記載の組成物。 7.前記安定剤が、ケイ素、ゲルマニウム、クロム、バナジウム、ニオブ、チ タン、ホウ素、アルミニウム、ジルコニウム、およびそれらの混合物から成るグ ループから選択される請求項3、5または6に記載の組成物。 8.前記組成物は石英基質上にコーティングされ、焼結時にケイ素が石英から 、形成されつつあるリン酸カルシウム相の中に遊離され、αリン酸三カルシウム を安定化する請求項2に記載の組成物。 9.前記ケイ素が焼結前にハイドロキシアパタイトに溶液として添加される請 求項3に記載の組成物。 10.前記ケイ素がオルトケイ酸四プロピルである請求項1または請求項9に 記載の組成物。 11.前記リン酸カルシウム相のハイドロキシアパタイト対αリン酸三カルシ ウムの比が50:50から20:80である請求項1に記載の組成物。 12.前記組成物がpH約6.4から7,3の生理学的溶液に不溶である請求項1に記 載の組成物。 13.ハイドロキシアパタイトを焼結により主にαリン酸三カルシウムに変換 するステップと、リン酸相内部で形成されたαリン酸三カルシウムを安定化させ 、不溶化する安定剤を提供するステップとを含んでなる、その上の骨細胞活性を 支持するのに適した形態を有するリン酸カルシウム相の人工焼結組成物を安定化 するため方法。 14.前記組成物が、粉末、フィルム、コーティング、または立体固体である 請求項13に記載の方法。 15.前記ハイドロキシアパタイトが、ケイ素、アルミニウム、ジルコニウム 、チタン、ホウ素、ゲルマニウム、クロム、バナジウム、ニオブ、およびそれら の混合物から成るグループから選択される成分を含む基質上に提供される請求項 14に記載の方法。 16.前記安定剤が、前記基質から焼結中に作製されるハイドロキシアパタイ ト相の中に遊離される請求項15に記載の方法。 17.前記安定剤が、ケイ素、アルミニウム、ジルコニウム、チタン、ホウ素 、ゲルマニウム、クロム、バナジウム、ニオブ、およびそれらの混合物から成る グループから選択される請求項14または16に記載の方法。 18.前記ハイドロキシアパタイトは石英基質上に塗布され、焼結時にケイ素 が石英から、形成されつつあるリン酸カルシウム相の中に遊離され、αリン酸三 カルシウムを安定化する請求項17に記載の方法。 19.前記ケイ素が焼結前にハイドロキシアパタイトに溶液として添加される 請求項13に記載の方法。 20.前記ケイ素がオルトケイ酸四プロピルである請求項13または19に記 載の方法。 21.前記リン酸カルシウム相のハイドロキシアパタイト対αリン酸三カルシ ウムの比が50:50から20:80である請求項13に記載の方法。 22.ハイドロキシアパタイトの焼結が、約900℃から1100℃の温度で行われ る請求項13に記載の方法。 23.構造中に相互に結合された微細孔を有する相互に緩く結合された丸い顆 粒の球状表面形態を有する焼結安定化リン酸カルシウム相を含んでなる、骨細胞 活性を支持するための焼結された微細孔を有する人工多結晶構造。 24.前記構造が図10の前記球状表面形態を有する請求項23に記載の多結 晶構造。 25.前記球状顆粒が、0.5〜1μmの範囲の側方向への大きさを有する請求項 24に記載の多結晶構造。 26.a)基質を支持する構造と、 b)リン酸カルシウム相を不溶化し、安定化させる安定剤の存在下に、焼結温度 でハイドロキシアパタイトをリン酸三カルシウムに変換することにより作製され た安定化リン酸カルシウム相の層と、 c)前記安定化されたリン酸カルシウム相の層上に培養された骨芽細胞により沈 着された境界層と、 d)この様に培養された骨芽細胞により分泌された鉱物化コラーゲンに富んだ基 質と を含んでなる移植可能な石灰化骨基質。 27.前記基質が骨芽細胞を含む骨細胞を含まない請求項26に記載の移植可 能な石灰化骨基質。 28.前記基質が、骨芽細胞を含む患者の骨細胞を含む請求項26に記載の移 植可能な石灰化骨基質。 29.前記基質が、破骨細胞により吸収され得る請求項26に記載の移植可能 な石灰化骨基質。 30.請求項1または2の組成物を用いて作製されるセラミック塊の微細孔構 造物。 31.前記構造が内部に大きな孔を有する請求項30に記載のセラミック塊の 微細孔構造物。 32.請求項1または2の焼結組成物によりコーティングされた移植可能な装 置。 33.請求項1の組成物を本質的に含む移植可能な装置。 34.安定化され、不溶化されたαリン酸三カルシウム複合体を含む基質上に 安定化リン酸カルシウム相の人工焼結フィルムに、生理学的培地中に懸濁された 骨芽細胞懸濁液を組成物に塗布するステップを含んでなる機能性骨細胞の培養方 法。 35.安定化され不溶化されたαリン酸三カルシウムを含むリン酸カルシウム 相の焼結フィルムを有する基質と、 前記基質に付着させたマルチウェル骨細胞培養装置と を含んでなる骨細胞の活性を監視し、定量するキット。 36.相互に結合された微細孔を有する相互に緩く結合された球状顆粒で形成 される球状表面形態を有する人工安定化組成物を提供するステップと、 生理学的培地中に懸濁された骨芽細胞懸濁液を組成物に塗布するステップと、 培養した骨芽細胞から分泌された鉱物化コラーゲンに富む骨基質をインキュベ ーションするステップと、 単離されたコラーゲンに富む骨基質を患者に移植するステップと を含んでなる鉱物化コラーゲンに富む基質のex vivoでの作製方法。
