Claims (1)
- 【特許請求の範囲】 1. 式(1)の10−デアセチルバッカチン(baccatine)III および10−デアセチル−14β−ヒドロキシバッカチンIII誘導体。 式中、 R1およびR2は、水素原子であるか、またはR1が、水素原子であり、そして R2が、ヒドロキシルもしくはアセチルオキシ基であるか、またはOR1とR2が 、一緒になって、式: の環状炭酸エステル基を形成し; R3は、αまたはβ配向することができて、水素原子もしくはアルキルシリル 基、好ましくはトリエチルシリル(TES)であり; R4は、水素、または残基 か、または式A: [式中、R1’は、炭素原子1〜5個をもつ直鎖または分枝アルキルもしくはア ルケニル基か、またはアリール残基であり;R2’は、炭素原子1〜5個をもつ 直鎖または分枝アルキルもしくはアルケニル基、またはアリール残基、またはt ert−ブトキシ基である]のイソセリン残基である。 2. 次のものからなる群より選ばれる請求の範囲1記載の化合物: 10−デアセチル−10−デヒドロバッカチンIII; 10−デアセチル−10−デヒドロ−14β−ヒドロキシバッカチンIII; 10−デアセチル−10−デヒドロセファロマンニン(cephaloman nine); 10−デアセチル−10−デヒドロ−14β−ヒドロキシバッカチンIII 1,14−炭酸エステル; 13−[(2R,3S)−3−tert−ブトキシカルボニルアミノ−2−ヒ ドロキシ−3−イソブチル−プロパノイル]10−デアセチル−10−デヒドロ −バッカチンIII; 13−[(2R,3S)−3−tert−ブトキシカルボニルアミノ−2−ヒ ドロキシ−3−イソブチル−プロパノイル]10−デアセチル−10−デヒドロ −14β−ヒドロキシバッカチンIII 1,14−炭酸エステル; 13−[(2R,3S)−3−カプロイルアミノ−2−ヒドロキシ−3−イソ ブチル−プロパノイル]−10−デヒドロ−10−デアセチル−14β−ヒドロ キシバッカチンIII 1,14−炭酸エステル。 3. 請求の範囲1記載の10−デアセチルバッカチンIIIおよび10−デ アセチル−14β−ヒドロキシバッカチンIII誘導体の調製方法であって、式B : [式中、R1、R2、R3およびR4は、先に定義された意味をもつ]のシントン( synton)が、a)R2=HもしくはOHか、またはOR1およびR2が一緒 になって炭酸基であり、そしてR4≠Hの場合には、銅(II)塩による処理によ って位置10において酸化され;そして場合に よっては位置7において脱保護され; b)R2=R4=Hの場合には、銅(II)塩による処理によって位置10において 酸化され、そして場合によっては(R3=Hの場合、7において予めシリル化さ れて)、基R4≠Hを導入させるアシル化剤によって位置13においてエステル 化されることを特徴とする方法。 4. R2=OHおよびR4=Hである請求の範囲3記載の式Bのシントンの調 製方法であって、その7−トリアルキルシリル−10−デアセチルバッカチンI II(または7−エピマー)が、二酸化マンガンを用いる処理によって位置13 における選択的酸化、そして位置14における同時β−ヒドロキシル化にかけら れ、得られる7−トリアルキルシリル−10−デアセチル−13−デヒドロ−1 4β−ヒドロキシバッカチンIII(またはその7−エピマー)が、10および 14においてアセチル化され、そして7−トリアルキルシリル−13−デヒドロ −14β−ヒドロキシ−14−アセチルバッカチンIII(またはその7−エピ マー)が、水素化物による還元にかけられて、対応する13β−ヒドロキシ誘導 体を得ることを特徴とする方法。 5. 請求の範囲3記載の式Bのシントン。 6. 式9b: [式中、R2’は、前記意味をもつ]の中間体。 7. R3=Hである請求の範囲1記載の1種以上の式1の化合物を、有効成 分として含有する医薬組成物。 8. 非経口または経口投与できる、請求の範囲7記載の医薬組成物。 9. 抗腫瘍剤としての請求の範囲1〜2の化合物。 10.心臓病患者における腫瘍の治療に有用な薬物の調製のための、R2’が アルキルもしくはアルケニル基である請求の範囲1のタキサン(taxane) の使用。
