Claims (1)
- 【特許請求の範囲】 1.i)約8.0〜11.0のレンジ内のpHにおける約0.01−0.25モルのバッファー 中のタンパク質の第1水性混合物、及びii)以下の式: G−LM−PEG−LM−G により表される架橋剤の第2水性混合物、から本質的に成る接着剤組成物。上記 式中、−PEG−は、以下の式: −O−(CH2−CH2−O−)a− (式中、aは、20〜300の整数である。)により表されるジラジカル断片であり ;−LM−は、式−C(O)−のカーボネートのジラジカル、式−(CH2)bC(O) −(式中、bは、1〜5の整数である。)のモノエステル・ジラジカル、式−C (O)−(CH2)c−C(O)−(式中、cは、2〜10の整数であり、そしてそのジ ラジカルの脂肪族部分はは飽和であっても不飽和であってもよい。)のジエステ ル・ラジカル、式−C(O)−O−(CH2)d−O−C(O)−(式中、dは、2〜 10の整数である。)のジカーボネート・ジラジカル、又は以下の式−R−C(O )−,−R−C(O)−(CH2)c−C(O)−、若しくは、−R−C(O)−O− (CH2)d−O−C(O)−(式中、cは、2〜10の整数であり、dは、2〜10の整 数であり、そしてRは、ラクチド、グリコリド、トリメチレン・カーボネート、 カプロラクトン及びp−ジオキサノンから成る群から選ばれた1〜10のモノマー 断片をもつポリマー又はコポリマーである。)により表されるオリゴマー・ジラ ジカルから成る群から選ばれたジラジカル断片であり;そして−Gは、スクシン イミジル、マレイミジル、フタルイミジル、イミダゾールイル、ニトロフェニル 又はトレジルから成る群から選ばれた脱離基である。 2.請求項1に記載の接着剤組成物であって、その第1混合物中のタンパク質 が、約20〜60wt/vol%の血清アルブミンで存在するもの。 3.請求項1に記載の接着剤組成物であって、その第2混合物中の架橋剤の濃 度が、その架橋剤の重量平均分子量が約1,000〜5,000のレンジ内にあるとき、約 50〜300mg/mlであり、そしてその第2混合物中の架橋剤の濃度が、その架橋剤 の重量平均分子量が約5,000〜15,000のレンジ内にあるとき、約300〜800mg/ml であることを特徴とする組成物。 4.請求項1に記載の接着剤組成物であって、その第1混合物と第2混合物の 組合せ物が、初めに液体であり、そして次に、約1分未満でその組織表面上で硬 化して、その組織に結合し、そして約4〜60日以内に吸収される柔軟で実質的な マトリックスが得られることを特徴とする組成物。 5.i)約0.01−0.25モルのバッファー濃度及び約8.0〜11.0のレンジ内のpH におけるバッファー中のタンパク質の第1水性混合物、とii)以下の式: G−LM−PEG−LM−G により表される架橋剤の第2水性混合物との混合物を形成することから成る組織 接着剤の製造方法。上記式中、−PEG−は、以下の式: −O−(CH2−CH2−O−)a− (式中、aは、20〜300の整数である。)により表されるジラジカル断片であり ;−LM−は、式−C(O)−のカーボネートのジラジカル、式−(CH2)bC(O) −(式中、bは、1〜5の整数である。)のモノエステル・ジラジカル、式−C (O)−(CH2)c−C(O)−(式中、cは、2〜10の整数であり、そしてそのジ ラジカル の脂肪族部分は飽和であっても不飽和であってもよい。)のジエステル・ジラジ カル、式−C(O)−O−(CH2)d−O−C(O)−(式中、dは、2〜10の整数 である。)のジカーボネート・ジラジカル、又は以下の式−R−C(O)−,− R−C(O)−(CH2)c−C(O)−、若しくは、−R−C(O)−O−(CH2)d− O−C(O)−(式中、cは、2〜10の整数であり、dは、2〜10の整数であり 、そしてRは、ラクチド、グリコリド、トリメチレン・カーボネート、カプロラ クトン及びp−ジオキサノンから成る群から選ばれた1〜10のモノマー断片をも つポリマー又はコポリマーである。)により表されるオリゴマー・ジラジカルか ら成る群から選ばれたジラジカル断片であり;そして−Gは、スクシンイミジル 、マレイミジル、フタルイミジル、イミダゾールイル、ニトロフェニル又はトレ ジルから成る群から選ばれた脱離基である。 6.請求項1に記載の接着剤混合物を組織に適用し、そしてそれに接着させる 段階を含む、インビボにおける組織接着方法。 7.請求項1に記載の接着剤混合物を肺組織中の空気洩れ部位に適用し、そし てそれを硬化させる段階を含む、肺組織中の空気洩れをシールするインビボにお ける方法。 8.請求項1に記載の接着剤混合物を外科手術部位の周囲の組織に適用し、そ してそれを硬化させる段階を含む術後の接着を防止するためのインビボにおける 方法。 9.請求項1に記載の接着剤混合物を組織に適用し、そして接着させて、血液 又は他の液体の洩れを防ぎ又はコントロールする段階を含む、組織をシールする ためのインビボにおける方法。
