Claims (1)
- 【特許請求の範囲】 1)哺乳動物細胞における組換え遺伝子の発現のための相同的組換え発現ベクタ ーであって、該ベクターが、組換え遺伝子の発現のためのプロモーター、選択マ ーカーをコードする転写ユニット、および相同的組換えターゲッティングのため のマウスの免疫グロブリンγ2A座位特異的DNA配列を含有する、ベクター。 2)マウス細胞における組換え遺伝子の発現のための請求項1に記載の相同的組 換え発現ベクター。 3)NS/O細胞における組換え遺伝子の発現のための請求項2に記載の相同的組換 え発現ベクター。 4)前記選択マーカーが、gpt、dhfr、抗生物質耐性、またはグルタミン合成酵 素転写ユニットからなる群から選択される、請求項1〜3のいずれかに記載の相 同的組換え発現ベクター。 5)前記選択マーカーが、グルタミン合成酵素転写ユニットである、請求項4に 記載の相同的組換え発現ベクター。 6)前記プロモーターが、CMV-IEプロモーターである、請求項1〜5のいずれか に記載の相同的組換え発現ベクター。 7)前記組換え遺伝子が免疫グロブリン遺伝子である、請求項1〜6のいずれか に記載の相同的組換え発現ベクター。 8)哺乳動物細胞における組換え免疫グロブリン遺伝子の発現のための相同的組 換え発現ベクターであって、該ベクターが、第1の免疫グロブリン遺伝子の発現 のための第1のプロモーター、第2の免疫グロブリン遺伝子の発現のための第2 のプロモーター、選択マーカーをコードする転写ユニット、および相同的組換え ターゲッティングのためのマウス免疫グロブリンγ2A座位特異的DNA配列を含有 する、ベクター。 9)前記第1のプロモーターおよび第2のプロモーターがそれぞれCMV-IEプロモ ーターである、請求項8に記載の相同的組換え発現ベクター。 10)前記選択マーカーがグルタミン合成酵素転写ユニットである、請求項8ま たは9に記載の相同的組換え発現ベクター。 11)マウス細胞における組換え免疫グロブリン遺伝子の発現のための請求項8 〜10に記載の相同的組換え発現べクター。 12)マウス細胞における組換え免疫グロブリン遺伝子の発現のための請求項8 〜10に記載の相同的組換え発現ベクター。 13)NS/O細胞における組換え免疫グロブリン遺伝子の発現のための請求項11 に記載の相同的組換え発現ベクター。 14)ベクターpIgG2A。 15)哺乳動物宿主細胞における組換え遺伝子の発現方法であって、以下の工程 : (a)免疫グロブリンγ2A座位特異的相同的組換えベクターを前記宿主細胞に転 移する工程であって、該ベクターが組換え遺伝子をコードするDNAを含有し、そ して該ベクターが宿主細胞染色体γ2A座位と相同的に組換わる、工程;および (b)選択および組換え遺伝子発現に適切な条件下で該宿主細胞を培養する工程 、を包含する、方法。 16)前記宿主細胞選択が、gpt、dhfr、抗生物質耐性、またはグルタミン欠乏 からなる群から選択される、請求項15に記載の方法。 17)前記ベクターが、前記宿主細胞染色体γ2A座位と、前記宿主細胞γ2A座位 中の任意の部位で相同的に組換わる、請求項16に記載の方法。
