Claims (14)
- 1.式 ▲数式、化学式、表等があります▼ (式中、MはRu、OsおよびReから成る群から選択され、u・t=vであり 、uは1または2であり、tはIまたは2であり、Ll、L2およびL3は同一 でもまたは異なっていてもよく、それぞれ▲数式、化学式、表等があります▼, ▲数式、化学式、表等があります▼または▲数式、化学式、表等があります▼ であり、但し、R1、R2、R3、R4、R5およびR6は同一でもまたは異な っていてもよく、それぞれH、1〜5個の炭素原子を有するアルキル基、▲数式 、化学式、表等があります▼または▲数式、化学式、表等があります▼;であり 、R1、R2、R3、R4、R5およびR6、の少なくとま一つは▲数式、化学 式、表等があります▼または▲数式、化学式、表等があります▼; であり、nは1〜20の範囲の整数であり、XはO、S、SO2、COOおよび CONHから成る群から選択され、aおよびbは−N(CH−(CH2)2)2 、−NH−CH−(CH3)2、O−(CH2)2−CN、O−CH3またはモ ルホリノであり、但しaおよびbは同一ではなく、ZはMと会合する対イオンで あり、リガンドL1、L2およびL3は化合物が電気化学ルミネッセンスが可能 であるように選択される)を有する化合物。
- 2.L1、L2およびL3が総て置換されたまたは未置換のビピリジルであり、 nは4〜7である、請求項1に記載の化合物。
- 3.式 ▲数式、化学式、表等があります▼ 〔式中、MはRu、OsおよびReから成る群から選択され、L1、L2および L3は同一でもまたは異なっていてもよく、それぞれ▲数式、化学式、表等があ ります▼,▲数式、化学式、表等があります▼または▲数式、化学式、表等があ ります▼であり、但し、R1、R2、R3、R4、R5およびR6は同一でもま たは異なっていてもよく、それぞれH、1〜5個の炭素原子を有するアルキル基 であり、但し、R1〜R6の一つは、式 ▲数式、化学式、表等があります▼ のリンカーであり、nは1〜20の範囲の整数であり、XはO、S、SO2、C OOおよびCONHから成る群から選択され、yは1〜20の整数であり、zは 0または1であり、Bは成体分子またはその合成類似体であり、ホスホジエステ ル基は前記の生体分子に結合している)を有し、電気化学ルミネッセンスを生じ ることができる化合物。
- 4.式 ▲数式、化学式、表等があります▼ (式中、Mはルテニウム、オスミウムまたはレニウムであり、R1、R2、R3 、R4およびR5は同一でもまたは異なっていてもよく、それぞれH、1〜4個 の炭素原子を有するアルキル基であり、nは1〜20の整数であり、XはO、S 、SO2、COOおよびCONHから成る群から選択され、yは1〜20の整数 であり、zは0または1であり、mは1〜1000の整数であり、NABは修飾 されていてもまたは修飾されていなくともよい核酸塩基である)を有し、電気化 学的ルミネッセンスが可能な錯体。
- 5.前記の核酸がポリメラーゼ増幅反応のプライマーである、請求項4に記載の 錯体。
- 6.前記の核酸がハイブリダイゼーションプローブである、請求項4に記載の錯 体。
- 7.試料中に存在する目的とする核酸の分析物質を検出する方法であって、前記 の試料を式 ▲数式、化学式、表等があります▼ (式中、Mはルテニウム、オスミウムまたはレニウムであり、R1、R2、R3 、R4およびR5は同一でもまたは異なっていてもよく、それぞれH、1〜4個 の炭素原子を有するアルキル基であり、nは1〜20の整数であり、XはO、S 、SO2、COOおよびCONHから成る群から選択され、yは1〜20の整数 であり、zは0または1であり、mは1〜1000の整数であり、NABは修飾 されていてもまたは修飾されていなくともよい核酸塩基である)を有するプロー ブと、このプローブが目的とする前記の分析物質に選択的に結合して、目的とす る分析物質−プローブ錯体を形成するような条件下で接触させ、この錯体の電気 化学的ルミネッセンスを測定する段階を含んでなる方法。
- 8.前記の目的とする分析物質が核酸配列であり、前記のプローブがこの目的と する分析物質に相補的な核酸プローブである、請求項7に記載の方法。
- 9.ポリメラーゼ連鎖反応または他のプライマーに対する反応の増幅生成物にお いて目的とする核酸を検出するように上記反応を行う方法であって、(a)前記 のポリメラーゼ連鎖反応または他のプライマーに対する反応において、式 ▲数式、化学式、表等があります▼ (式中、Mはルテニウム、オスミウムまたはレニウムであり、R1、R2、R3 、R4およびR5は同一でもまたは異なっていてもよく、それぞれH、1〜4個 の炭素原子を有するアルキル基であり、nは1〜20の整数であり、XはO、S 、SO2、COOおよびCONHから成る群から選択され、yは1〜20の整数 であり、zは0または1であり、mは1〜1000の整数であり、NABは修飾 されていてもまたは修飾されていなくともよい核酸塩基である)を有するプライ マーを取り込み、 (b)ポリメラーゼ連鎖反応または他のプライマーに対する反応を行い、(c) 目的とする増幅した核酸の電気化学的ルミネッセンスを測定することから成る、 方法。
- 10.ポリメラーゼ連鎖反応または他のプライマーに対する反応の生成物中に目 的とする核酸を検出する方法であって、(a)前記の反応中の少なくとも1個の 核酸を、式式 ▲数式、化学式、表等があります▼ (式中、Mはルテニウム、オスミウムまたはレニウムであり、R1、R2、R3 、R4およびR5は同一でもまたは異なっていてもよく、それぞれH、1〜4個 の炭素原子を有するアルキル基であり、nは1〜20の整数であり、XはO、S 、SO2、COOおよびCONHから成る群から選択され、yは1〜20の整数 であり、zは0または1であり、mは1〜1000の整数であり、NABは修飾 されていてもまたは修飾されていなくともよい核酸塩基である)を有する化合物 で標識し、 (b)ポリメラーゼ連鎖反応または他のプライマーに対する反応を行い、前記の 核酸を増幅生成物中に取り込み、 (c)目的とする前記の核酸の電気化学的ルミネッセンスを測定する段階から成 る方法。
- 11.化合物の相対的ECL容量の、そのビピリジル類似体の相対的ECL容量 に対する比が0.10以上である、請求項1に記載の化合物。
- 12.化合物の相対的ECL容量の、そのビピリジル類似体の相対的ECL容量 に対する比が0.10以上である、請求項3に記載の化合物。
- 13.目的とする分析物質−プローブ錯体の相対的ECL容量の、そのビピリジ ル類似体に対する相対的ECL容量の比が0.10以上である、請求項7に記載 の方法。
- 14.錯体の相対的ECL容量の、そのビピリジル類似体の相対的ECL容量に 対する比が0.25以上である、請求項4に記載の錯体。
