Claims (18)
- 1.植物または植物器官の炭水化物組成を変える方法であって、植物の糖を分解 しうる(第1)目的酵素をコードするDNA配列を含む発現カセットを有するト ランスジェニック植物を、該植物または植物器官がジャガイモである場合には、 該(第1)目的酵素をコードするDNA配列が微生物由来であるという条件付き で、該酸素コードDNA配列が発現し該植物または植物器官の炭水化物組成が変 化する誘導条件下で生育させることを特徴とする方法。
- 2.前記発現カセットが前記トランスジェニック植物または植物器官の発生の所 定の成熟段階で前記目的酵素の発現を指令しうる調節配列を含むことを特徴とす る請求の範囲第1項記載の方法。
- 3.前記発現構築物が、前記目的酵素の組織特異的発現を指令しうることを特徴 とする請求の範囲第1項記載の方法。
- 4.前記(第1)目的酸素をコードするDNA配列に該発現酵素を所定の細胞組 織またはオルネガネラに指向させうるリーダー配列が備えられていることを特徴 とする請求の範囲第1項記載の方法。
- 5.前記目的酵素の作用の結果として前記トランスジェニック植物または植物器 官において6個までのそノサッカライドユニットを含む可溶性サッカライド含量 が増加することを特徴とする請求の範囲第1項記載の方法。
- 6.前記(第1)目的酵素が、アミラーゼ、グルカナーゼ、セルラーゼ、エンド グルカナーゼ、アラビナナーゼ、ガラクタナーゼ、マンナナーゼ、キシラナーゼ 、フコシダーゼ、ラムノシダーゼ、レバナーゼおよびイヌリナーゼからなる群か ら選択される請求の範囲第1項記載の方法。
- 7.前記(第1)目的酵素をコードするDNA配列が微生物由来である請求の範 囲第1項記載の方法。
- 8.前記(第1)目的酵素をコードするDNA配列が、バチルスリチェニホルミ ス(Bacillus licheniformis)由来のα−アミラーゼお よびアスペルギラスニガー(Aspergillus niger)由来のグル コアミラーゼからなる群から選択される請求の範囲第7項記載の方法。
- 9.前記トランスジェニック植物が前記(第1)目的酵素以外に加えて該第1目 的酵素の作用から生じる澱粉分解産物を基質として使用しうる第2目的酵素をコ ードするDNA構築物を含む一つ以上の発現構築物を含むことを特徴とする請求 の範囲第1項記載の方法。
- 10.前記付加的目的酵素が、グルコアミラーゼ、プルラナーゼ、イソアミラー ゼ、シクロマルトデキストリン−D−グルコスランスフェラーゼ、α−(1−4 )−グルカナーゼ、α−(1−4)−グルコシダーゼ、α−(1−6)−グルコ シダーゼ、β−グルコシダーゼ、D−グルコースイソメラーゼおよびイヌリナー ゼからなる群から選択されることを特徴とする請求の範囲第9項記載の方法。
- 11.前記トランスジェニック植物がトマト、ジャガイモ、コーン、カッサバ、 ニンジン、レタス、イチゴおよびタバコからなる群から選択されることを特徴と する請求の範囲第1項乃至第10項のいずれか1項記載の方法。
- 12.植物のポリサッカライドを分解しうる目的酵素をコードするDNA配列が トランスジェニック植物または植物器官の発生の所定の成熟段階における該目的 酵素の発現を指令しうる調節配列に機能的に結合していることを特徴とする発現 構築物。
- 13.植物のポリサッカライドを分解しうる目的酵素をコードするDNA配列が 35SCaMVプロモーターと機能的に結合していることを特徴とする発現構築 物。
- 14.植物のポリサッカライドを分解しうる目的酵素をコードするDNA配列が 該目的酵素の組織特異的発現を指令しうる調節配列に機能的に結合していること を特徴とする発現構築物。
- 15.請求の範囲第12項乃至第14項のいずれか1項記載の発現構築物を含む ベクター。
- 16.請求の範囲第12項乃至第14項のいずれか1項記載の発現カセットを含 むことを特徴とするトランスジェニック植物。
- 17.請求の範囲第15項記載のベクターを含むことを特徴とするバクテリア。
- 18.請求の範囲第1項または第9項のうちのいずれか1項記載の方法により炭 水化物組成が変化していることを特徴とするトランスジェニック植物または植物 器官。
