Description
本発明は、国立衛生研究所によって付与された助成金番号GM102137、HL094463、CA207824及びGM103390の下での米国政府の支援によってなされた。そのため米国政府は本発明に一定の権利を有する。
ヘパリンは50年以上にわたって抗凝固薬として用いられてきた(Mackman, 2008)。現在ヘパリンは3つの形で販売されている:分画されていない(UF)ヘパリン(平均分子量:~14,000Da);低分子量(LMW)ヘパリン(平均分子量:~6,000Da);及び合成ULMWヘパリン五糖ARIXTRA(R)(分子量:1508.3Da)。UFヘパリンは、その比較的短い半減期及び腎障害のある患者に対する安全性のため、手術や腎臓透析で使用されている(Hirsh et al., 2007)。
このような患者を治療する方法において、この合成ヘパリン類似体は、他のヘパリン化合物よりも約50%~約100%速いクリアランス速度を持つことができる。投与の4時間後に、この合成ヘパリン類似体の抗凝固活性は、約10%未満であることができる。この方法において、この患者は高い出血リスクを有することができる。
またここでは、ここに開示する合成ヘパリン化合物を含む医薬組成物が提供される。

"アラルキル基"はアリール-アルキル-基を指し、ここでアリール基及びアルキル基は既述の通りであり、置換アリール基及び置換アルキル基を含む。アラルキル基の例としては、ベンジル基、フェニルエチル基、又はナフチルメチル基がある。

ここで開示される本発明の原理が、組成物及び方法が用語"患者"に含まれることが意図されている哺乳動物を含むすべての無脊椎動物及び脊椎動物に有効であることを示していることを理解すべきであるが、治療され、スクリーニングされ、試験され、又はサンプル採取される患者は、望ましくはヒト患者である。さらに、哺乳動物は、スクリーニングが望ましいあらゆる哺乳動物種、特に農業及び家畜哺乳動物種を含むと理解される。
その開示された方法、化合物及び治療は、温血脊椎動物の検査、スクリーニング及び/又は治療に特に有用である。したがって、本発明は、哺乳類及び鳥類に関する。
本発明の組成物は、いくつかの実施形態において、薬学的に許容される担体を含む組成物を含む。患者へ投与するためのアデノウイルスベクターを調製するために、任意の適切な医薬製剤を使用することができる。
適切な製剤には、例えば、水性及び非水性滅菌注射液が含まれてもよく、これは、抗酸化剤、緩衝液、静菌剤、殺菌性抗生物質、及び製剤を患者レシピエントの体液と等張にする溶質、並びに懸濁剤及び増粘剤を含んでもよい水性及び非水性滅菌懸濁液を含むことができる。この製剤は、単位用量又は複数用量の容器(例えば、密封されたアンプルとバイアル)で提供でき、また使用直前に滅菌液体キャリア(例えば、注射用の水)を追加することだけが必要な冷凍又は凍結乾燥状態で保存できる。いくつかの例示的な成分は、SDS、マンニトール又は他の糖、及びリン酸緩衝生理食塩水(PBS)である。
本発明の製剤は、上記で特に言及した成分に加えて、問題となる製剤の種類を考慮して、当技術分野で慣用されている他の薬剤を含むことができることを理解されたい。例えば、発熱物質を含まない滅菌水溶液及び非水溶液を使用できる。
本発明の組成物の投与は当業者に知られている任意の方法によることができ、それには、静脈内投与、滑液嚢内投与、経皮投与、筋肉内投与、皮下投与、局所投与、直腸投与、膣内投与、腫瘍内投与、経口投与、頬側投与、経鼻投与、非経口投与、吸入、及びガス注入が含まれるがこれらに限定されない。いくつかの実施態様において、本発明の組成物の投与に適した方法には、静脈内投与が含まれるが、これに限定されない。また代替として、組成物を、他の任意の方法で治療を必要とする部位に堆積させてもよい。本発明の組成物の投与の特定の様式は、様々な要因に依存する。
本発明の組成物の有効な投与量が、それを必要とする患者に投与される。"治療有効量"とは、測定可能な反応(例えば、抗凝固)を引き起こすのに十分な組成物の量である。いくつかの実施態様において、治療有効量は、凝固を防止する(即ち、抗凝固)のに十分な量である。いくつかの実施態様において、治療有効量は、抗凝固療法を必要とする患者の健康、幸福度、予後及び/又は生存率を改善するのに十分な量である。
本発明の組成物中の活性成分の実際の投与量レベルは、特定の患者の所望の治療応答を達成するのに有効な量の活性化合物を投与するように変更可能である。選択された用量レベルは、治療組成物の活性、投与経路、他の薬物又は治療との組み合わせ、治療状態の重症度、及び治療されている患者の状態及び以前の病歴に依存してもよい。しかし、所望の治療効果を達成するのに必要なレベルよりも低いレベルで組成物の投与量で開始し、所望の効果が達成されるまで用量を徐々に増加させることは、当業者の裁量の範囲内である。
当業者は、本明細書に提示される本発明の開示をよく調べた後、特定の製剤、組成物に使用される投与方法及び症状の重症度を考慮して、個々の患者に対する用量を調整することができる。投与量のさらなる計算において、患者の身長と体重、症状の重症度と病期、及び追加の有害な身体状態の存在を考慮することができる。このような調整又は変動、並びにこのような調整又は変動を行う時期及び方法の評価は、医学の当業者にとってよく知られたことである。
酵素の発現と精製
SF9細胞(Invitrogen)中で、Sf-900TM III SFM (Life Technologies)を用いて、6-O-スルホトランスフェラーゼ1(6-OST-1)及び6-O-スルホトランスフェラーゼ3(6-OST-3)の発現を行った。2.0×106 cells/mlの濃度の昆虫細胞に、マウス6-OST-1及びヒト6-OST-3を発現する組換えウイルスを感染させ、27℃のシェーカー中で96時間インキュベートした。この培養液を4,000 rpmで10分間遠心分離して、この細胞をペレット化した。次に、1mMのフェニルメタンスルホニルフルオリド(PMSF、95%エタノール中で100mMストック溶液を新たに作成した。)、Triton X-100 及び 0.2%グリセロールを含む上清を、8,000 rpmで30分間遠心分離し、1.5μMメンブレンでろ過した。次に、得られた培地を、等容量の0.05% Triton-100及び 2%グリセロールを含む40 mM3-(N-モルホリノ)プロパンスルホン酸(MOPS)緩衝液(pH 7.0)と混合した。ヘパリントヨパールゲル(東ソーバイオサイエンス)カラムを使用して、2つの緩衝液(緩衝液Aは、20 mM MOPS, pH 7.0, 100 mM NaCl, 2%グリセロール、及び0.1%還元triton X-100 (Sigma)を含み、緩衝液Bは、20 mM MOPS, pH 7.0, 1 M NaCl, 2%グリセロール、及び0.1%還元triton X-100を含む。)で6-OST-1及び3を精製した。この培地を装填した後、4 mL/minの流速において、280nmにおける吸光度がベースラインに達するまで、カラムを緩衝液Aを使用して洗浄した。60分で0-100%Bのグラジエント溶出を適用し、1.5 mL/minの流速で100%Bを用いてカラムを60分間溶出した。酵素の精製は4℃で行った。
化合物1~3の合成は、六糖(GlcNS-GlcA-GlcNS-IdoA2S-GlcNS-GlcA-pNP)("NS2S 6-mer基質"と呼ぶ)から開始した。
化合物1を合成するために、この基質(25mg)を、総容量100 mLの、50 mM MOPS(PH 7.0)、10 mM MnCl2、7 mM MgCl2、及び2 mL 3-OST-3(0.11 mg/mL)を含む緩衝液中で1.8 mM 3'-ホスホアデノシン5'-ホスホ硫酸(PAPS)と共にインキュベートした。この反応混合物を37℃一晩インキュベートした。少量の反応混合物を陰イオン交換HPLC(TSKgel DNA-NPR-カラム(4.6 mM x 7.5 cm、2.5μm、東ソーバイオサイエンスから入手))に注入することにより、反応の完了を監視した。反応が60%未満の場合、更に3-OST-3酵素とPAPSを追加し、反応混合物を37℃でさらに18~24時間維持した。反応が完了した場合、反応混合物をQ-セファロースクロマトグラフィー(GE Healthcare)にかけた。
化合物3を合成するために、化合物2(4 mg)を、総容量100 mLの、18 mM MOPS(pH 7.0)、5 mM MnCl2、5 mM MgCl2、及び6 mL 3-OST-1(4μg/ mL)を含む緩衝液中で1.3 mM PAPSと共にインキュベートした。この反応混合物を37℃一晩インキュベートした。少量の反応混合物を陰イオン交換HPLCに注入することにより、反応の完了を監視し、生成物をQ-セファロースにより精製した。
化合物4を合成するため、この基質30 mgを、総容量90 mLの、33 mg MOPS(pH 7.0)、10 mM MnCl2、5 mM MgCl2、及び4 mL 3-OST-3(0.11 mg/mL)を含む緩衝液中で、2 mM PAPSと共にインキュベートした。この反応混合物を37℃で一晩インキュベートした。少量の反応混合物を陰イオン交換HPLCに注入することにより、反応の完了を監視した。反応が60%未満の場合、更に3-OST-3酵素とPAPSを追加し、反応混合物を37℃でさらに18~24時間維持した。反応が完了したら、反応混合物をQ-セファロースクロマトグラフィー(GE Healthcare)にかけた。
化合物6を合成するために、化合物5(6.5 mg)を、総容量100 mLの、18 mM MOPS(pH 7.0)、5 mM MnCl2、5 mM MgCl2、及び4.5 mL 3-OST-1(4μg/mL)を含む緩衝液中で及び1.3 mM PAPSと共にインキュベートした。この反応混合物を37℃一晩インキュベートした。少量の反応混合物を陰イオン交換HPLCに注入することにより、反応の完了を監視し、生成物をQ-セファロースにより精製した。
3-OST-1及び3-OST-3の基質要件を決定するために、異なるサイズの構造的に均一な複数のオリゴ糖を使用した。これらのオリゴ糖には、N-硫酸化及び2-O-硫酸化6-mer~12-mer(6-mer2S~12-mer2S)及びN-硫酸化、2-O-硫酸化及び6-O-硫酸化6-mer~12-mer (6-mer2S6S~12-mer2S6S)が含まれる。これらのオリゴ糖は化学酵素的方法1により調製され、それらの構造は図5Cに示されている。
オリゴ糖(0.033 mM)を、50 mM MOP(pH 7.0)、10 mM MnCl2、5 mM MgCl2、2.5μL3-OST-1又は3-OST-3、及び[35S] PAPS(1-3 x 105 cpm)と混合した30μMのPAPSを含む100μL反応緩衝液中で37℃で1時間インキュベートした。この反応混合物に、1.4 mM EDTA、50 mM酢酸ナトリウム、及び150 mM NaClを含む900μLの3 M尿素を加えて反応を停止した。次いで、この反応混合物をDEAEカラムを用いて精製した。
酵素反応速度は、N-硫酸化及び2-O-硫酸化のオリゴ糖のために精製3-OST-3 30μL又はN-硫酸化、2-O-硫酸化及び6-O-硫酸化のオリゴ糖のために精製3-OST-3 60μLの混合物をインキュベートすることによって特徴付けた。PAPS(120μM)を、硫酸ドナーとしての1×105cpmの[35S] PAPS及び0~200μMの様々な濃度のオリゴ糖と、37℃で1時間混合した。35S標識オリゴ糖生成物を精製するために、反応液をDEAEカラムに入れた。35S標識オリゴ糖生成物の量を基質濃度に対してプロットし、Sigma Plotソフトウェアを使用してMichalis-Mentenグラフのカーブフィッティングを行い、Km及びVmax値を得た。
硫酸化オリゴ糖の精製を、Q-Sepharoseカラムを用いて行った。移動相Aは25 mMトリス、pH 7.5であり、移動相Bは、25 mMトリス及び1 M NaCl、pH 7.5が含む。溶出勾配は、1 mL/minの流量で合成されたオリゴ糖の硫酸基数に基づく。310 nm及び260 nmにおける吸収をスキャンして記録した。精製後、サンプルを、5mMリン酸ナトリウム(pH 7.5)を含む緩衝液に対して1000 MWCO膜を使用して、2回透析した。
TSKgel DNA-NPR-カラムを用いて、反応の完了度と精製後の合成オリゴ糖の純度を検出した。移動相Aは25 mMトリス、pH 7.5であり、移動相Bは、25 mMトリス及び1 M NaCl、pH 7.5であった。勾配段階は、0.4 mL/minの流量で100分間で0-100%Bであった。310 nm及び260 nmにおける吸収を使用して、溶離液をモニターした。
合成されたオリゴ糖の分子量構造を、ESI-MS(Thermo LCQ-Deca)により決定した。ESI-MS分析は、負イオンモードで、次のパラメーターを使用して行った:Iスプレー電圧3.0 kV、曲線状の脱溶媒線温度120℃、質量範囲300~1000。
3-O-[34S]硫酸化化合物4を合成するために、基質NS2S 8-mer(2 mg)を、38 mM MOPS(pH 7.0)、10 mM MnCl2、5 mM MgCl2、及び6 mL 3-OST-3を含む総容量20 mLの緩衝液中で、3-OST-3酵素(0.11 mg/mL)及び0.1 mM [34S] PAPSと共にインキュベートした。この反応混合物を37℃一晩インキュベートした。生成物の精製を、Q-セファロースカラムを使用して行った。
タンデム質量スペクトル分析を、負イオンモードのThermo LTQ-FT機器上で、次のパラメーターを使用して取得した:Iスプレー電圧3.5 kV、毛管電圧-40 kV、チューブレンズ-50 V、毛細管温度275℃。タンデム質量の場合、選択されたプリカーサーイオンは、次のパラメーターで実行された:IsoWidth(m/z):3.0、標準化衝突エネルギー(%):50.0、Act.Q:0.250、Act.時間:30、最大注入時間(ms):500.000。MS及びMS/MSデータは、Xcalibur 2.2ソフトウェアを使用して記録し処理された。
NMR実験は、Topsin 3.2ソフトウェアを備えたBruker Avance 700 MHz及び850 MHz分光計により298 Kで行なった。複数のサンプル(0.5~3.0 mg)をそれぞれ0.5 ml D2O(99.996%、Sigma-Aldrich)に溶解し、3回凍結乾燥して交換可能なプロトンを除去した。これらのサンプルを0.5 ml D2Oに再溶解し、NMRマイクロチューブ(外径5 mM、Norrell)に移した。化学シフトは、外部の2,2-ジメチル-2-シラペンタン-5-スルホン酸ナトリウム塩(DSS、Sigma. Co.)を基準とする。重水素化EDTA(Sigma. Co.)を加えて、常磁性イオンによる影響を除去した。1D 1H-NMR実験"zg"パルスシーケンスは、64スキャン及び取得時間3.8秒で実行された。1D 13C-NMR実験"zgdc30"パルスシーケンスは、10,000回のスキャン及び取得時間1.0秒で実行された。2D1H-13C HSQC実験"hsqcgpph"パルスシーケンスは、48スキャン、512インクリメント、緩和遅延1.5秒、取得時間120ミリ秒で実行された。2DスペクトルはGARPカーボンデカップリングを用いて記録された。取得開始前に48回のダミースキャンを使用した。f2で合計2048ポイントが収集された。13Cトランスミッターオフセットは90.0 ppmに設定された。
アンチトロンビン(AT)-五糖複合体(PDB ID:3EVJ)の既存の結晶構造を、システムの開始構造として採用した47。アミノ酸残基25-33及び396はこの結晶構造で解析されていなかったため、Modellerへのキメラインターフェイスを用いて生成した48-51。N-グリカンは、フォンダパリヌクスリガンドから遠位(>15Å)にあるため、除去された。この元のリガンドは、さまざまなシミュレーション用に修正され、キメラを用いて手動で調整された。アミノ酸及び炭水化物残基のパラメーターは、ff14SB7及びGLYCAM06(J-1)力場から得た53, 54。システムはNa +イオンで中和され、複合体の周囲12Åの切り捨てられた8面体ボックス内のTIP3P水モデルで溶媒和された。
AMBER14のpmemd.cudaモジュールを使用して、エネルギーの最小化とMDシミュレーションを実行した52。タンパク質骨格のCα原子は、プロセスのすべての段階でデカルト拘束(10 kcal/molÅ2)で制限されていた。2つのシミュレーションでは、1C4又は2S0立体配座を維持するためにIdoA2S残基d(図4A)の内部拘束が必要であり、前述の設定に従って実行された55。残りの24,000サイクルで共役勾配に切り替える前に、最初の1000サイクルで最急降下法を使用してシステムを最小化した。アンチトロンビン(AT)-オリゴ糖複合体の安定した配置の生成を支援するために、さまざまな原子拘束で2つの最小化を実行した。最初、すべての溶質原子は抑制されていた。その後、リガンドの拘束とタンパク質の側鎖が取り除かれた。静電相互作用をパーティクル-メッシュ エワルド アルゴリズムで処理し56、非結合相互作用の8Aカットオフを採用した。SHAKEアルゴリズムを水素含有結合に適用して、2 fsの積分時間段階が可能になった。カップリング時定数1 psのBerendsenサーモスタットを使用して、システムを60 ps以上に渡ってNVT条件下で300 Kに加熱し、NPT条件下で合計50 nsの間平衡化した。同じくNPT条件下で、さらに追加の150 nsの間、平衡化後のデータセットを収集した。
相互作用エネルギーを、MPBSA.py.MPIモジュールを使用して、単一軌道分子力学-一般化Born Solvent Accessible Surface Area(MM-GBSA)法で計算した57。この分析の前に、すべての水分子とイオンを各複合体から除去し、脱溶媒和エネルギーからの寄与をGB暗黙溶媒和モデルによって近似した(igb=2)58。このシミュレーションを5 nsのビン(bin)に分割し、各ビン内に均等に分布した100個のスナップショットのアンサンブルから、平均相互作用エネルギーの寄与を計算した。GraphPad Prism for Windowsバージョン5.04を用いて統計分析を行った。t検定分析による
Xa因子(Enzyme Research Laboratories, South Bend, IN)を、1 mg/mLのウシ血清アルブミン(BSA)を含むリン酸緩衝生理食塩水(PBS)で60 nMに希釈した。1 mg/mL BSAを含むPBS中で0.65μMの濃度のヒトアンチトロンビン(AT)(Cutter Biologicalから入手)を調製した。発色基質S-2765(Diapharma)を水に溶解して、1mg/mLストック溶液を作成した。PBS中で様々な濃度(10~200 nM)の複数のオリゴ糖を作成した。この溶液は、60μLのアンチトロンビン(AT)と15μLのサンプル溶液の混合物であり、これを撹拌し、室温で2分間インキュベートした。次いでこれにXa因子(90μL)を添加し、室温で4分間インキュベートし、その後30μLのS-2765を添加した。この反応混合物の405 nmにおける吸光度を2分間連続して測定した。IC50値の計算は、サンプル濃度と初期反応速度の関数としてプロットした。
すべての研究は、実験動物の管理と使用に関する公衆衛生サービスのガイドラインに従って、ノースカロライナ大学の施設内動物管理使用委員会(IACUC)承認プロトコルの下で実施した。16匹の400-gルイスラットを4つのグループに均等に分けた。各グループに、エンドトキシンを含まないフォンダパリヌクス(0.46μMkg-1)、化合物11(0.43μMkg-1)、化合物5(0.42μMkg-1)、又は生理食塩水を静脈内投与した。次に、化合物の投与直前に1つのサンプルを採取しておき、化合物を投与した後、指定の時点(0.5、1、2、4、8時間)に採血した。化合物の注入直前に1つのサンプルを採取しておき、化合物注入の0.5、1、2、4、8時間後に、対側大腿骨/伏在静脈から0.8 mLの血液を採取し、150 mMクエン酸塩に入れた。これらのサンプルを遠心分離して、約400μLの血漿を得た。これらの血液サンプルをFXa活性分析にかけた。体液量を維持するために生理食塩水を定期的に皮下投与した。
各化合物について作成した検量線を使用して、反応混合物中の化合物の濃度を読み取り、対応する各FXa活性割合(%)を得た。各時点で各グループ中の4匹のラット間の平均血漿濃度と標準偏差を計算した。これらの値を使用して、時間に対する血漿濃度のグラフをプロットした。
GlcNS3S及びGlcNS3S6S残基を持つオリゴ糖の化学酵素的合成
本研究において、六糖(化合物1~3、図1A)及び八糖(化合物4~6、図1A)の合成を達成した。2つの3-O-スルホトランスフェラーゼ(3-OST)アイソフォーム、3-OST-1及び3-OST-3を使用して、GlcNS3S±6S残基を異なる糖配列に導入した。3-OST-1酵素は、硫酸化を導入し、非還元末端のGlcA残基に結合するGlcNS3S6S残基を形成し、-GlcA-GlcNS3S6S-の二糖単位を形成する。一方、3-OST-3酵素は、硫酸化を導入し、非還元末端のIdoA2S残基に結合するGlcNS3S残基を形成し、-IdoA2S-GlcNS3S-の二糖単位を形成する。3-OST-1は、多くの研究でオリゴ糖の合成に使用されてきたが16,17,27,28、3-OST-3を使用して-IdoA2S-GlcNS3S-二糖単位を含むオリゴ糖を合成することは報告されていない。
オリゴ糖の構造及び立体配座解析
化合物1~6の純度測定及び構造分析を実施した。化合物6(図6F)の分析の代表的なデータを図 2A及び2Bに示す。化合物6は、高分解能陰イオン交換HPLCで単一ピークとして溶出され、この化合物が純粋であることを示す(図2A)。化合物6の分子量は、エレクトロスプレーイオン化質量分析(ESI-MS)により2449.43±0.74と決定された。これは分子量の計算値2448.92に非常に近い(図2B)。化合物6の1 H-NMRスペクトルは、8つのアノマープロトンを明確に示し、生成物が八糖であることが確認される。化合物6の13 C-NMR及び完全なNMR帰属をそれぞれ補足図25及び補足表1に示す。化合物4の3-O-スルホ基を特定するために、タンデムMS分析を実施した。この分析において、安定した同位体標識[34S]スルホ基が3-OST-3酵素によって導入され、化合物4の残基dに3-O-スルホ基が存在することを明確に特定できた。化合物4は化合物5と6の中間体であるので、化合物4のタンデムMS分析は、化合物5及び6のIdoA2S-GlcNS3S6S二糖単位の特定にも役立った。
-GlcNS3S6S-IdoA2S-二糖単位の存在は、抗凝固活性に影響を与える。
ヘパラン硫酸(HS)のオリゴ糖は、アンチトロンビン(AT)と相互作用することにより抗凝固活性を発揮する。したがって、抗FXa活性を示すオリゴ糖はATに結合する筈である。GlcNS3S6S残基を取り囲む糖残基の、AT結合及び抗FXa活性に対する効果を調べた。これらのデータは、-GlcNS3S6S-IdoA2S-二糖単位(即ち、GlcNS3S6S残基の還元末端にIdoA2Sが位置する)の存在が、抗凝固活性の決定に果たす役割を明らかにすることができる(表3)。化合物2及び8は、6つの残基、8つのスルホ基、1つのIdoA2S及び2つのGlcA残基を有するが、化合物8のみが抗FXa活性を示した(図3A及び表3)。予想されたとおり、アンチトロンビン(AT)は化合物8に強く結合するが(Kd値7±2 nM)、ATは化合物2には結合しない(表3)。これらの化合物は、構造的に、IdoA2S残基の位置が異なる。化合物8においては、3-O硫酸化グルコサミン(GlcNS3S6S)残基の還元末端にIdoA2S残基が隣接し、化合物8は-GlcNS3S6S-IdoA2S-二糖単位を持つ。一方、化合物2のGlcNS3S6Sはその残基の還元末端にGlcA残基が隣接し、-GlcNS3S6S-GlcA-二糖単位を有する。
アンチトロンビン(AT)結合ドメイン中のGlcA残基はIdoA2S残基で置換可能である
これらの研究により、新しいAT結合糖配列が明らかにされた。現在知られているAT結合配列は、二糖-GlcA-GlcNS3S±6S-単位を含んでいる33。ヘパラン硫酸(HS)中の-GlcA-GlcNS3S6S-二糖単位を-IdoA2S-GlcNS3S6S-二糖単位で置換することは、AT結合親和性を無効にすると認識されていた34,35。8-merの抗FXa活性及びAT結合分析からのデータは、この主張に明らかに異議を唱えた。IdoA2S-GlcNS3S6S-(-GlcA-GlcNS3S6S-ではない)二糖単位を含む化合物5は、強い抗FXa活性を示す(表3)。等温滴定熱量測定(ITC)を使用したAT結合親和性分析も、化合物5がアンチトロンビン(AT)にしっかりと結合することを実証した(図3B及び表3)。化合物6は、この8量体は-GlcA-GlcNS3S6S-二糖単位を含むので、抗FXa活性と高いAT結合親和性を示す(表3)。
7-mer ヘパラン硫酸(HS)化合物の分析
ここで開示された合成方法を使用して、7-merのヘパラン硫酸(HS)化合物又はヘパリン類似体を展開した。図8Bにその構造が示されている7-merを、抗Xa活性について試験し、その抗Xa活性を実証した(図8A)。図8Bは、そこに示すように、7-merの化学構造を6-mer及び8-merと概略的に比較する。限定ではなく例として、7-merを合成するための少なくとも1つの合成経路を図8Cに示す。いくつかの実施態様において、合成に関連するコストの削減を可能にするので、7-merなどの比較的短いオリゴ糖を有することが好ましい場合がある。
結果の議論
本明細書には、化学酵素的アプローチを使用して3-O-硫酸化オリゴ糖ライブラリーを調製する複数のスキームが開示される。本明細書において、-IdoA2S-GlcNS3S-又はIdoA2S-GlcNS3S6S-二糖単位を含むオリゴ糖を合成するためには、3-OST-3修飾が6-O-硫酸化段階に先行しなければならず、一方、-GlcA-GlcNS3S6S-二糖単位を生成するためには、3-OST-1修飾が 6-O-硫酸化後にのみ起きなければならないことが示されている。この発見は、3-OST-1/ 七糖/PAP37及び3-OST-3/四糖/PAP38の三元共結晶構造によってサポートされている。3-OST-3と四糖基質の6-O-スルホ基との間に相互作用はなく、このことは3-OST-3のオリゴ糖基質は6-O-硫酸化を必要としないという結論と整合する。これとは対照的に、3-OST-1と七糖基質の6-O-スルホ基との間に相互作用が観察され、3-OST-1に結合するには6-O-硫酸化が必要であることを示す。3-OST-1と3-OST-3との間のこの明確でユニークな基質要件は、3-OSTの異なるアイソフォームによって修飾された3-O硫酸化ヘパラン硫酸(HS)は異なる経路を経由して生合成される可能性を提起する。

Claims (12)
- 硫酸化によって修飾された、グルコサミン(GlcN)残基に結合したグルクロン酸(G
lcA)及びイズロン酸(IdoA)残基の二糖繰り返し単位を含む6~8個の糖単位か
ら成る合成ヘパリン類似体であって、
(a)3-O-硫酸化オリゴ糖;
(b)少なくとも1つの、3-OST-3酵素により硫酸化された二糖単位;
(c)少なくとも1つのIdoA2S-GlcNS3S又はIdoA2S-GlcNS3
S6S二糖単位;及び
(d)少なくとも1つのGlcN6S二糖単位;を含み、
(e)GlcA-GlcNS3S6S二糖単位を含まない、合成ヘパリン類似体。 - 前記アルキルが、-CH3若しくは-CH2CH3である、又は前記置換アリールが、P
-ニトロフェニル基である、請求項2又は3に記載の合成ヘパリン類似体。 - (1) 硫酸化によって修飾された、グルコサミン(GlcN)残基に結合したグルクロ
ン酸(GlcA)及びイズロン酸(IdoA)残基の二糖繰り返し単位を含む6~8個の
糖単位から成る糖基質であって、少なくとも1つのIdoA2S-GlcNS3S又はI
doA2S-GlcNS3S6S二糖単位;及び少なくとも1つのGlcN6S二糖単位
;を含み、GlcA-GlcNS3S6S二糖単位を含まない、糖基質を提供する段階、
(2) この糖基質を所望の又は所定の長さの糖に伸長する段階、及び
(3) 3-O-スルホトランスフェラーゼ(3-OST)酵素の3-OST-3アイソ
フォームを使用して少なくとも1つの硫酸化反応を実行する段階、
から成り、これにより合成ヘパリン類似体が合成される、ヘパリン物類似体を合成する方
法。 - 前記糖基質がIdoA2S-GlcNS3S±6S二糖単位を含み、前記方法が更に6-
O-スルホトランスフェラーゼ(6-OST)を使用する6-O-硫酸化段階を含み、こ
こで6-O-硫酸化段階の前に、3-OST-3による3-O-硫酸化が起こる、請求項
5に記載の方法。 - 前記糖基質がGlcA-GlcNS3S6S二糖単位を含み、前記方法が更に6-O-ス
ルホトランスフェラーゼ(6-OST)を使用する6-O-硫酸化段階を含み、ここで6
-O-硫酸化段階の後に、3-OST-1による3-O-硫酸化が起こる、請求項5に記
載の方法。 - 前記伸長段階が、グリコシルトランスフェラーゼを使用することを含む、請求項5~7の
いずれか一項に記載の方法。 - 前記グリコシルトランスフェラーゼが、大腸菌K5のN-アセチルグルコサミニルトラン
スフェラーゼ(KFiA)及び/又はパスツレラ・ムルトシダ由来のヘパロサンシンター
ゼ-2(pmHS2)から成る群から選択される、請求項8に記載の方法。 - 前記伸長段階が、グルクロン酸(GlcUA)、N-アセチル化グルコサミン(GlcN
Ac)及びN-トリフルオロアセチルグルコサミン(GlcNTFA)から成る群から選
択される1又はそれ以上の単糖類を使用することを含む、請求項5~9のいずれか一項に
記載の方法。 - 前記合成ヘパリン類似体を合成する方法の収率が、20%~50%である、請求項5~1
0のいずれか一項に記載の方法。 - 請求項1~4のいずれか一項に記載の合成ヘパリン類似体を含む、抗凝固療法を必要とす
る患者を治療するための医薬組成物。
