Description
嗅覚は、ヒトの感覚系のうち、最も複雑でかつ十分な解明が進んでいないものの1つである。嗅覚受容体(OR)の活性化から知覚にかけては、なおもさらなる研究を要する多くのステップが存在する。個々の匂い物質および混合物に対するORコードがどのようにして知覚に変換されるかが解明されれば、この知見を様々な分野において顕著な利点をもたらすべく利用することができる。こうした分野には、匂いのモジュレーター、例えば不快な匂いの知覚を遮断する悪臭中和剤、非生分解性または毒性化合物に代わる新たなフレーバーおよびフレグランス成分、並びに匂いの増強剤が含まれ、これらによって、天然源由来の供給源化合物に対する我々の依存性が制限されると考えられる。「嗅覚コード」の組み合わせの典型は、全ての単一のORは複数の匂い物質によって活性化されることができ、かつ逆にほとんどの匂い物質は複数のORを活性化しうるという所見に集約される。マウスゲノム中には、約1,200種の異なるORが存在する。一方で、ヒトは約400種のORを有する。どちらの場合にも、ORレパートリーは世界中の何千もの匂い物質によって活性化され、そしてこの組み合わせの複雑さによって、我々が知覚しうる嗅覚が広がる。しかし、2014年現在で従来の脱オーファン化法を用いて特定されているのは、わずか95種のマウスOR(約8%)および41種のヒトOR(約10%)に対する匂い物質またはリガンドのみである。さらに、主にインビトロで特定されたヒトORとの生理的関連性に関して試験が行われたリガンドは、ほとんどない。
本明細書においては、匂い物質および/または香り化合物により活性化される嗅覚受容体を特定する方法であって、以下:
a)非ヒト哺乳動物種から、各神経細胞が前記嗅覚受容体を発現するネイティブ嗅神経細胞を含む単離された嗅上皮を単一細胞に解離すること;
b)前記嗅細胞に、前記嗅覚受容体の匂い物質または香り受容体結合活性の測定を可能にする指示色素を負荷すること;
c)前記嗅覚受容体と匂い物質または香り化合物とを順次接触させること;
d)匂い物質または香りにより誘発された神経細胞活性の変化を測定すること;
e)匂い物質または香り化合物によって活性化された1種以上の嗅神経細胞を単離すること;
f)前記単離された嗅覚受容体のmRNAを、単離、または単離および増幅すること;
g)次世代シーケンシングにより前記mRNAのトランスクリプトームの少なくとも一部を配列決定すること;および
h)前記トランスクリプトームの配列を同種および他の脊椎動物種のリファレンスゲノム配列と比較することにより、匂い物質受容体および香り受容体からなる群から選択される嗅覚受容体群を特定すること
を含む前記方法が提供される。

実施例1
インドールおよび/またはスカトールに結合する受容体を特定するためのマウス嗅神経細胞のスクリーニング、
マウス由来のネイティブ嗅神経細胞を含む嗅上皮を単離して単一細胞に解離した後、解離バッファーに移して穏やかに機械的および酵素的に解離した。解離プロトコルを最適化するために、新鮮な解離バッファーを作製するための一般的な手順を、Araneda et al. (2004), A Pharmacological Profile of the Aldehyde Receptor Repertoire in Rat Olfactory Epithelium. Journal of Physiology: 555:743-756の記載に従ったが、次の変更を加えた:DNアーゼIIに代えてDNアーゼIにし、Ca2+の濃度を5mMに高め、かつpHを32℃で7.35に調整した。解離された神経細胞を、その後、蛍光顕微鏡検査により2,500超の細胞のスクリーニングが可能なカバースリップ上に播種した。次いで、これらの細胞にカルシウム感受性色素(Fra−2 AM)を31℃で30分間負荷し、かつスクリーニングの準備ができた顕微鏡上に移した。解離された嗅神経細胞を以前に記載された通りにスクリーニングしたが(Malnic et al., 1999; Areneda et al., 2004)、電動式可動顕微鏡ステージを用いることで、スクリーニング可能な細胞数を増加させた。次いで、解離された嗅神経細胞上に(生理食塩水中の)匂い物質の希釈溶液を灌流することによって、これらの細胞を刺激した。
インドールおよびスカトール感受性細胞からのmRNAの単離および増幅による、インドールおよびスカトール感受性受容体に関する「悪臭受容体」遺伝子のcDNA配列の作成
実施例1の活性化された嗅神経細胞をガラス吸引マイクロピペットを用いて単離した後に、引き続き、応答細胞中のmRNAとして発現された匂い物質受容体遺伝子を特定するためにプールして1つのサンプルとした。悪臭受容体遺伝子配列を、次のようにインドールおよびスカトール感受性細胞から特定した:1種以上の悪臭化合物によって活性化される5種超の嗅神経細胞(各6、10、15および15の細胞)の4つの組を、マイクロマニピュレーターに取り付けられたガラス微小電極を用いて単離した。マイクロピペットはホウケイ酸ガラス(品目#B150−86−10、Sutter Instruments)から設計され、かつ次の条件下で微小電極プラー(P−1000型、Sutter Instruments)を用いて引かれたものである:熱、傾斜温度+20℃;Pull=0、Vel=120;圧力=500。引かれたピペットチップを手ではずし、かつカルシウムイメージングアッセイにおいて決定される際にインドールおよびスカトールにより活性化される嗅神経細胞を単離するためにNewportマイクロマニピュレーター(MW3R型)上に固定した。活性化されたOSNを、マイクロピペットの後端に適用される穏やかな吸引により単離した。
マウスおよびヒトの悪臭受容体の特定
カルシウムイメージングとNGSとの組み合わせによりマウスゲノムにおける約1,200種のORのうち5種のマウスインドールおよびスカトールORのサブセットを特定することによって(実施例1、2)、インシリコ法による付加的なマウスおよび新規のヒトインドールおよびスカトールORの迅速な特定が可能となる。類似する全体的なアミノ酸同一性を有するORは、類似の匂い物質応答プロファイルを示すものと考えられるため[Adipietro et al. (2012)]、候補インドールおよびスカトールORのリストを、カルシウムイメージングにより単離されたインドール/スカトール感受性嗅神経細胞のNGS解析に由来するマウスOR配列を用いた公共のマウスおよびヒトゲノムデータベースの検索により拡張した。NGSの結果を同種のリファレンスゲノム配列と比較することにより、マウス悪臭嗅覚受容体を特定した。推定悪臭受容体は、嗅神経細胞由来のNGSサンプル中でmRNAが極めて高レベルの量であるか、または、複数の独立した生物学的複製において存在するが1種以上の悪臭に応答する嗅神経細胞を含まない対照サンプル中には存在しない。標準的なバイオインフォマティクスツールを使用して、相同配列受容体は類似の機能を保持するという仮定の下で(Adipietro et al. (2012))、推定哺乳動物(非ヒト)の1種以上の悪臭受容体に極めて密接に関連する1種以上のヒト匂い物質受容体を特定した。BLASTPおよび/またはBLASTNアルゴリズムのデフォルトパラメーターを用いた。第1表に、特定されたマウス匂い物質受容体(OR)および予測ヒトオーソログを挙げる。
HEK293T細胞中でのマウスOR Olfr743、Olfr746およびOlfr740のインドールおよびスカトール活性
インドールおよびスカトールの双方の悪臭に対する、候補マウス受容体Olfr740(配列番号1および配列番号2)、Olfr743(配列番号5および配列番号6)およびOlfr746(配列番号37および配列番号38)の活性を、細胞ベースのcAMPアッセイで確認した。図5Aおよび5Bにおいて、タグ付きFLAG−Rho(配列番号18)、Olfr743(配列番号6)およびOlfr740(配列番号2)cDNA配列を、Gαolf(配列番号21)と共に共トランスフェクションし、かつインドール(左)またはスカトール(右)の濃度増加に曝露した。図5Cおよび5Dは、インドールおよびスカトールを用いた場合のOlfr740(配列番号2)の活性およびOlfr743(配列番号6)の活性が類似していることを示す反復試験である。図5Eにおいて、タグ付きFLAG−Rho(配列番号18)、Olfr746(配列番号38)cDNA配列を、Gαolf(配列番号21)と共に共トランスフェクションし、かつインドールまたはスカトールの濃度増加に曝露した。匂い物質誘発活性を、HTRFアプローチ(CisBioキット)を用いて、細胞質ゾル内cAMPレベルの測定により検出した。受容体活性の用量依存性増加を、双方の分子に対して3種全ての受容体に関して示す。Gαolf(配列番号21)単独(受容体なし)を用いたHEK細胞の共トランスフェクションを、非特異的活性についての対照として使用した(対照)。活性は、この試験における最も高いシグナルに正規化されたベースライン補正HTRF比と定義される。結果を図5に示す。3種全ての受容体は、悪臭化合物に対する特異的な用量依存的活性を示し、かつそれ自体で、こうした化合物に対する受容体を特定するための方法の正当性を実証している。マウス受容体Olfr740(配列番号1および配列番号2)およびOlfr743(配列番号5および配列番号6)を、インドールおよびスカトールの双方に応答した単離された嗅神経細胞から特定した。マウス受容体Olfr746(配列番号37および配列番号38)を、Olfr740(配列番号2)およびOlfr743(配列番号6)を検索配列として使用したデータベースマイニングによってインシリコで特定した。これらの受容体を、そのN末端でFLAGタグで修飾し、かつウシロドプシン受容体の最初の20のアミノ酸(配列番号18)を一過的にHEK293T細胞中で発現させ、かつ別々にインドールおよびスカトールで刺激することによって、それらが真のインドール/スカトール受容体であることを確認した。ヒトGαサブユニットGαolf(配列番号21)の共発現によってGs伝達経路が活性化され、これによって適したリガンドへの結合時に内部cAMPが増加する。この結果は、Olfr740(配列番号1および配列番号2)、Olfr743(配列番号5および配列番号6)およびOlfr746(配列番号37および配列番号38)がインドール−スカトール受容体であることを裏付けるものである。
HEK293T細胞中でのヒトOR52N2、OR11G2、OR5AC2、OR4C15、OR8S1、OR11H6およびOR11H4、並びにマウスOlfr665およびOlfr740のインドールおよびスカトール活性
候補ヒトインドールおよびスカトール悪臭受容体OR52N2(配列番号11および配列番号12)、OR11G2(配列番号13および配列番号14)、OR5AC2(配列番号75および配列番号76)、OR4C15(配列番号79および配列番号80)、OR8S1(配列番号83および配列番号84)、OR11H6(配列番号15および配列番号16)およびOR11H4(配列番号49および配列番号50)、並びに候補マウスインドールおよびスカトール悪臭受容体Olfr665(配列番号9および配列番号10)およびOlfr740(配列番号1および配列番号2)の活性を、細胞ベースのカルシウムフラックスアッセイにおいて確認した(図6)。細胞で安定的に共発現するRhoタグ付きヒトOR52N2(図6A、B)またはOR11G2(図6C)およびGαolf(配列番号25)を、スカトールまたはインドールの濃度増加に曝露した。図6Bは、インドールおよびスカトールを用いたOR52N2の類似の活性を示す反復試験である。匂い物質誘発OR52N2またはOR11G2活性を、匂い物質曝露後の最大のCalcium5色素(Molecular Devices)蛍光変化を測定することによって検出した。図6Aにおいては、活性は、この試験における最も高い相対的蛍光単位(RFU)に正規化されたベースライン補正RFU比と定義される。図6B〜Jにおいては、受容体活性の測定に相対的蛍光単位を用いる。受容体活性の用量依存的増加を記録し、かつ対応する用量反応曲線を双方の化合物について示す。受容体を欠く細胞株、例えばhOR52N2を、高濃度での非特異的活性についての対照として用いた(「インドール対照」および「スカトール対照」)。ヒト受容体OR52N2(配列番号11および配列番号12)を、インドールおよびスカトールの双方に応答する嗅神経細胞から単離されたインドール/スカトール受容体であるマウスOlfr665(配列番号10)に対するその配列類似性ゆえに特定した。ヒト受容体OR11G2(配列番号13および配列番号14)を、インドールおよびスカトールの双方に応答する嗅神経細胞から単離されたインドール/スカトール受容体であるマウスOlfr740(配列番号2)に対するその配列類似性ゆえに特定した。これらの受容体をRho配列で修飾し、かつHEK293T細胞中で安定的に発現させた。さらなる例として、ヒト受容体OR5AC2、OR4C15、OR8S1、OR11H6、OR11H4、並びにマウス受容体Olfr665およびOlfr740に関するインドールおよびスカトールの用量反応曲線を示す(図6D−J)。ヒト受容体OR5AC2(配列番号75および配列番号76)を、インドールおよびスカトールの双方に応答する嗅神経細胞から単離された受容体であるマウスOlfr207(配列番号78)に対するその配列類似性ゆえに特定した。ヒト受容体OR4C15(配列番号79および配列番号80)を、インドールおよびスカトールの双方に応答する嗅神経細胞から単離された受容体であるマウスOlfr1211(配列番号82)に対するその配列類似性ゆえに特定した。ヒト受容体OR8S1(配列番号83および配列番号84)を、インドールおよびスカトールの双方に応答する嗅神経細胞から単離された受容体であるマウスOlfr257(配列番号86)に対するその配列類似性ゆえに特定した。ヒト受容体OR11H6(配列番号15および配列番号16)を、インドールおよびスカトールの双方に応答する嗅神経細胞から単離された受容体であるマウスOlfr745(配列番号8)に対するその配列類似性ゆえに特定した。ヒト受容体OR11H4(配列番号49および配列番号50)を、インドールおよびスカトールの双方に応答する嗅神経細胞から単離された受容体であるOlfr746(配列番号38)に対するその配列類似性ゆえに特定した。マウス受容体Olfr665(配列番号9および配列番号10)を、インドールおよびスカトールの双方に応答する単離された嗅神経細胞からのNGSデータから直接特定した。マウス受容体Olfr740(配列番号1および配列番号2)を、インドールおよびスカトールの双方に応答する単離された嗅神経細胞からのNGSデータから直接特定した。ヒトGαサブユニットGα15の共発現によってGq伝達経路が活性化され、これによって適したリガンドへの結合時に内部Ca2+が増加する。この結果は、本明細書に開示された方法が、悪臭化合物に対する哺乳動物の匂い物質受容体の迅速かつ確実な特定に有用であることの実証に寄与する。
HEK293T細胞におけるインドール受容体Olfr743の活性の阻害
本明細書に概説した手順を使用して特定した後に、得られた悪臭受容体細胞株を使用して、その活性と可能性として考えられるヒトの知覚とを調節する化合物の化学ライブラリーをスクリーニングすることができる。この試験を、実施例4と同一の条件下に行った。Rhoタグ付き(配列番号18)Olfr743(配列番号6)と共にトランスフェクトされた細胞を、300μMの2−アセトナフトンの存在下または不在下に、インドールの濃度増加に曝露した。2−アセトナフトンは、インドールに対するOlfr743(配列番号6)の活性を阻害する。結果を第4表および図7に示す。アンタゴニスト化合物の存在下では用量反応曲線の右方向へのシフトが認められ、その結果、インドールに対してEC50が4倍増加する。活性は、この試験における最も高いシグナルに正規化されたベースライン補正HTRF比と定義される。この結果は、2−アセトナフトンがインドール受容体に結合して、この受容体の活性を阻害することを示している。
Claims (4)
- インドールおよびスカトールからなる群から選択される化合物によって活性化される匂い物質または香り受容体からなる群から選択される嗅覚受容体の活性を遮断、阻害、調節および/または増強する化合物を同定するための方法であって、以下:
a)前記嗅覚受容体の少なくとも一つまたはそのキメラと化合物とを、インビトロで接触させること、ここで、前記嗅覚受容体は、配列番号2のアミノ酸配列と少なくとも90%の配列同一性を有するアミノ酸配列を含むポリペプチドであるものとする;
b)前記化合物が前記嗅覚受容体の活性を遮断、阻害、調節または増強する程度を測定すること;および
c)前記嗅覚受容体の応答を遮断、阻害、調節または増強する化合物を同定すること;
を含む、前記方法。 - 前記ポリペプチドが、前記アミノ酸配列の一つと少なくとも95%または98%の配列同一性を有するアミノ酸を含む、請求項1に記載の方法。
- 前記ポリペプチドを発現するために組み換えにより改変された細胞を使用することを含む、請求項1または2に記載の方法。
- 前記細胞が、HEK293、CHO、アフリカツメガエル卵母細胞、COS、酵母、細菌および嗅プラコード由来細胞からなる群から選択される、請求項3に記載の方法。
