Description
Claims (15)
- DNA断片混合物貯蔵溶液(stock solution)を製造する第1ステップ;
キトサン貯蔵溶液を製造する第2ステップ;
前記DNA断片混合物貯蔵溶液及び前記キトサン貯蔵溶液を混合して撹拌した後、滅菌してDNA断片混合物及びキトサンを含む充填液を製造する第3ステップ;
前記充填液を無細胞性真皮移植片に充填する第4ステップ;を含む
ことを特徴とする生着率を増加させた移植用真皮層の製造方法。 - 前記製造方法は、
DNA断片混合物を緩衝溶液に入れて50℃乃至70℃で撹拌しながら1時間乃至3時間の間溶解させてDNA断片混合物貯蔵溶液を製造する第1ステップ;
キトサンを酸性緩衝溶液に溶解させてキトサン貯蔵溶液を製造する第2ステップ;
前記DNA断片混合物貯蔵溶液及び前記キトサン貯蔵溶液をDNA断片混合物及びキトサンの重量割合が20:1−10,000:1になるように混合して65℃乃至75℃で1時間乃至2時間の間撹拌した後、滅菌してDNA断片混合物及びキトサンを含む充填液を製造する第3ステップ;
前記充填液を真空薬物充填装置を用いて無細胞性真皮移植片に充填する第4ステップ;を含む
請求項1に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記DNA断片混合物は、魚類の精巣または精液から分離したものである
請求項1または2に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記魚類は、サケ類である
請求項3に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記DNA断片混合物は、分子量が50kDa乃至10,000kDaである
請求項1または2に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記DNA断片混合物は充填液の総重量を基準に0.01重量%乃至3重量%である
請求項1または2に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記キトサンは、分子量が3kDa乃至1,000kDaである
請求項1または2に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記キトサンは、充填液の総重量を基準に1x10−6重量%乃至0.15重量%である
請求項1または2に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記第4ステップの無細胞性真皮移植片は真皮から細胞及び不純物が除去され、ウイルス不活化処理後、凍結乾燥処理して生物学的に不活性化されたコラーゲン使用組織補充材である
請求項1または2に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記真皮は、自家、同種、または異種から分離した真皮である
請求項9に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記第4ステップの充填は圧力を用いて無細胞性真皮移植片の内部に粘性がある充填液を流入させてコラーゲンの間の空いた空間を詰めるようにする
請求項1または2に記載の生着率を増加させた移植用真皮層の製造方法。 - 前記第4ステップの真空薬物充填装置は、充填液を一方向に連結し、陽圧または陰圧を発生させて連結された充填液が無細胞真皮移植片の内部のコラーゲンの間の空間を詰めて充填されるようにする
請求項2に記載の生着率を増加させた移植用真皮層の製造方法。 - 無細胞性真皮移植片に、DNA断片混合物及びキトサンを含む充填液が充填された
ことを特徴とする生着率を増加させた移植用真皮層。
- 請求項13の移植用真皮層を含む皮膚損傷治療剤。
- 前記皮膚損傷は、擦過傷(abrasion)、切開(incision)、裂傷(laceration)、皮膚裂離(skin avulsion)、皮膚打撲(skin bruise)、穿刺負傷(puncture wound)、及び手術による皮膚切除群より選択される1種以上が原因となる
請求項14に記載の皮膚損傷治療剤。
