Description
D−フルクトースをプシコースに転換させる微生物を分離するために、1%(w/v)プシコースが添加された最小培地(KH2PO4 2.4g/L、K2HPO4 5.6g/L、(NH4)2SO4 2.6g/L 、3mM MnSO4、7H2O 0.1g/L、酵母抽出物1g/L)を用いた。根圏土壌(rhizosphere soil)1gを0.85%(w/v)NaCl、10mLに懸濁し、100μlを取って寒天培地に塗抹した後、30℃で培養した。前記寒天培地で形成されたコロニーのうちの形状や大きさが異なるコロニーを選別した後、最小培地(KH2PO4 2.4g/L、K2HPO4 5.6g/L、(NH4)2SO4 2.6g/L 、3mM MnSO4、7H2O 0.1g/L、酵母抽出物1g/L)に各コロニーを接種し、30℃で24時間振とう培養し、遠心分離して菌体のみを回収した。回収した菌体は、0.85%(w/v)NaClで洗浄した後、菌体濃度20%(w/w)に50%(w/w)D−フルクトース、3mM MnSO4を添加した50mM Tris−Cl緩衝液(pH8.0)を入れて浮遊させ、55℃で2時間菌体と反応させた。前記反応結果物は遠心分離して反応液から菌体を除去し、上澄み液をHPLCを介してプシコースの生産を確認した。前記HPLC分析は、Aminex HPX−87Cカラム(BI0-RAD)が装着されたHPLC(Agi lent、USA)のRefractive Index Detector(Agilent 1260 RID)を利用し、移動相溶媒は水を用いており、温度は80℃、流速は0.6ml/分で行った。HPLC分析により、D−フルクトースからプシコースを最も多く生産した菌株2種(LIS1及びLIS2)を選定した(図1及び2)。
カイスティア・グラニュリLIS1及びカイスティア・デフルビイLIS2と同種の他の菌株及び異種のカイスティア属菌株もプシコースの生産が可能であるかを確認した。
Claims (6)
- カイスティア(kaistia)属微生物を含む、D−プシコース生産用組成物。
- 前記カイスティア属微生物が、カイスティア・グラニュリ(Kaistia granuli)、カイスティア・デフルビイ(Kaistia defluvii)カイスティア・ゲウムホネシス(Kaistia geumhonensis)、カイスティア・アディパタ(Kaistia adipata)、カイスティア・ダルセオネシス(Kaistia dalseonensis)、カイスティア・ヒルジニス(Kaistia hirudinis)、カイスティア・ソライ(Kaistia soli)及びカイスティア・テラエ(Kaistia terrae)からなる群から選択される1種以上のカイスティア属微生物である、請求項1に記載のD−プシコース生産用組成物。
- 前記プシコース生産用組成物が、D−フルクトース(D-fructose)をさらに含む、請求項1または2に記載のD−プシコース生産用組成物。
- カイスティア(kaistia)属微生物をD−フルクトースと接触させる段階を含む、D−プシコースの製造方法。
- 前記接触が、pH5.0〜9.0の条件で、40℃〜90℃の温度条件で、または0.5時間〜48時間行うことである、請求項4に記載のD−プシコースの製造方法。
- 寄託番号KCCM11916Pとして寄託された、カイスティア・グラニュリ(Kaistia granuli)LIS1菌株。
