Description
本明細書に記載した発明は、ある新規なα−Gal A調製物およびその製造方法、ならびにそれらの調製物を用いてファブリー病またはファブリー病の異型変種を患った患者を治療する方法に関する。考えられる特定の代表的な実施の形態を要約し、以下に詳細に説明している。
ファブリー病は、α−Gal Aの活性不全によって引き起こされる遺伝子疾患である。「α−Gal A欠乏症」とは、患者の体内におけるこの酵素の量的または活性に関する任意の欠乏状態を意味し、血管壁内の組織球における中性糖脂質(例えば、グロボトリアシルセラミドなど)の異常蓄積を起こし、太腿、臀部および生殖器における角化血管腫、発汗減少症、四肢の感覚異常、角膜の輪生、ならびにスポーク様後部水晶嚢下白内障を伴う。この物質が沈着することにより、痛み、重篤な腎および心血管疾患、ならびに卒中を引き起こす。糖脂質が蓄積することにより、ファブリー病を患っている男性において一般的に観察されるような重篤な症状が誘発される場合がある。別の場合には、糖脂質が蓄積することにより、欠損遺伝子をヘテロ接合体として有する女性において時折観察されるような比較的緩和な症状が誘発される場合がある。罹病患者は寿命が非常に縮まり、腎臓、心臓または脳血管の合併症により、ほぼ40才で死に至る。本疾患に対する特異的な治療は存在しない。リソソーム貯留性疾患として分類されるファブリー病は全世界において15,000人以上の患者が存在する。
ファブリー病などのα−GalA欠乏症が疑われる患者は、培養された遺伝子改変細胞(好ましくはヒト細胞)から得た精製ヒトα−GalAを用いて治療することができる。
本発明の方法に従い、細胞(「α−GalA産生細胞」)を増殖させた培地を回収し、または細胞を溶解して内容物を放出させ、次に、レクチンアフィニティークロマトグラフィーを用いないタンパク精製技術に付すことにより、培養細胞からα−GalAタンパク質を単離した。好ましい精製過程については、以下の実施例2に概説している。
本発明は、肝臓およびマクロファージ以外の特定の組織において治療用酵素の取り込みを増加させるための糖タンパク質変形プログラムを提供する。本発明に従う方法を用いることによってグリコシル化ヒトα−GalA調製物が得られ、このとき、オリゴ糖の35〜85%が電荷を帯びており、好ましくは、少なくともオリゴ糖の50%が電荷を帯びている。
α−GalAをホスホリル化することにより、α−GalAの循環半減期および細胞に侵入するα−GalAのレベルを変更することができる。ホスホリル化はα−GalAを発現する細胞内で行うことが好ましい。特に企図していることは、α−GalA産生細胞内にホスホリルトランスフェラーゼをコードしているDNA配列を最初に導入することにより、または、内在性ホスホリルトランスフェラーゼ遺伝子の発現を制御するような制御遺伝子を相同的組換えによって導入することにより、高度にホスホリル化されたグリコシル化α−GalAを得ることである。次に、α−GalAおよびホスホリルトランスフェラーゼを発現するような培養条件下においてα−GalA産生細胞を培養する。その後、ポリヌクレオチドを有しない細胞において産生されたα−GalAと比較して、より高度にホスホリル化されているα−GalA調製物を単離する。そのようなホスホリルトランスフェラーゼ類は、当該分野において既知である。例えば、米国特許第5,804,413号、および5,789,247号を参照のこと。これらを参考として本明細書中に取り入れておく。
末端にガラクトース残基を有し、十分にシアリル化されていないグリカンについて、シアリル化の割合を高めることは、シアリルトランスフェラーゼ遺伝子を有する哺乳類細胞、好ましくはヒト細胞をトランスフェクトすることによって達成することができる。
本発明に従えば、α−GalAをポリエチレングリコールとコンプレックス形成させることにより、ヒトグリコシル化α−GalA調製物の循環半減期を延ばすことができる。ポリ(エチレングリコール)(PEG)は、水に可溶性の高分子(ポリマー)であり、タンパク質に共有結合している場合には、タンパク質の潜在的用途を広げるような方向に特性を変化させる。ポリエチレングリコール修飾(「PEG化」)は十分に確立された技術であり、タンパク質およびペプチド薬剤に関する多くの問題を解決する、または改善する可能性を有するものである。

本発明に従う組成物(すなわち、多様なα−GalAグリコフォームを含む組成物)は、α−GalA調製物に適した任意の経路で投与することができる。精製したα−GalA調製物は、α−GalAタンパク質を十分に産生できない、もしくは欠損型のα−GalAタンパク質を産生する対象、またはα−GalA治療によって利益を享受する対象に対して投与することができる。本発明に従う治療用調製物は、任意の適切な方法により、直接的に(例えば、局所的に、注射、組織部分への内移植または局所投与として)、または全身的に(例えば、経口または非経口的に)投与することができる。
さらに本発明は、アルブミンなどの非α−GalAタンパク質、宿主細胞によって産生された非α−GalAタンパク質または動物組織または体液から単離されたタンパク質を実質的に含有していないα−GalA調製物の新規な製剤を提供する。
さらに本発明は、ファブリー病の患者、ファブリー病の異型変種の患者、または、α−GalAのレベルが低下している、もしくはα−GalAの突然変異型を有する状態にある患者にα−GalA調製物を投与するための方法を提供する。投与量としては、好ましくは0.05〜5.0mg/kg、より好ましくは0.1〜0.3mg/kgであり、1週間または2週間ごとに投与する。好ましい実施態様においては、約0.2mg/kgを2週間ごとに投与する。患者の生涯にわたって規則的に反復投与することが必要である。皮下注射を行うことにより、より長時間にわたって薬物を全身的に循環させることができる。皮下投与量は0.01〜10.0mg/kgであり、好ましくは0.1〜5.0mg/kgであり、2週間または1週間ごとに投与する。筋肉内注射によって投与されるα−GalA調製物の投与量は、皮下注射の場合と同等または異なっており、好ましい実施態様においては、筋肉内投与の投与量はより少量であり、投与頻度も低い。α−GalA調製物は静脈内に投与することもでき、例えば、ボーラス静注、緩速静注または連続的静注などの方法がある。連続的IV輸液(例えば、2〜6時間など)により、血中の特異的濃度を維持することができる。
2つの発現プラスミド、pXAG-16およびpXAG-28を構築した。これらのプラスミドは、α−Gal A酵素の398個のアミノ酸(α−Gal Aシグナルペプチドを含まない)をコードしているヒトα−Gal A cDNA;hGH遺伝子の第1のイントロンにより中断された、ヒト成長ホルモン(hGH)シグナルペプチドゲノムDNA配列;およびポリアデニル化のシグナルを含有する、hGH遺伝子の3’非翻訳配列(UTS)を含有する。プラスミドpXAG-16は、ヒトサイトメガロウイルス即時−早期(immediate-early)(CMV IE)プロモータおよび第1のイントロン(非コードエクソン配列に隣接した)を有し、一方で、pXAG-28は、コラーゲンIα2プロモータおよびエクソン1により駆動し、β−アクチン遺伝子の第1のイントロンを含有するβ−アクチン遺伝子の5’UTSも含有する。
以下のように構築したヒト線維芽細胞cDNAライブラリーからヒトα−GalA cDNAをクローニングした。総RNAからPoly−A+mRNAを単離し、製造業者の使用説明書にしたがってlambda ZapII(登録商標)システム(カリフォルニア州、ラ・ホーヤのストラタジーン社(Stratagene Inc.))用の試薬を用いて、cDNA合成を行った。端的に言えば、「第1鎖」cDNAは、内部XhoI制限エンドヌクレアーゼ部位を含有するオリゴ−dTプライマーの存在下で逆転写により産生した。RNase Hによる処理後に、そのcDNAをDNAポリメラーゼIによりニックトランスレーションして、二本鎖cDNAを産生した。T4 DNAポリメラーゼIによりこのcDNAを平滑末端化し、EcoRIアダプタに連結した。この連結産物をT4 DNAキナーゼにより処理し、Xho Iで切断した。cDNAをセファクリル(Sephacryl(登録商標))−400クロマトグラフィーにより分画した。大型および中型分画をプールし、EcoRIおよびXhoIで切断したlambda ZapIIアームにこれらのcDNAを連結した。次いで、この連結産物をパッケージングし、滴定した。この一次ライブラリーは、1.2×107pfu/mLの力価および925bpの平均挿入サイズを有していた。
5’−CTGGGCTGTAGCTATGATAAAC−3’(オリゴ1;配列番号6)および
5’−TCTAGCTGAAGCAAAACAGTG−3’(オリゴ2;配列番号7)
を用いて、ポリメラーゼ連鎖反応(PCR)によりゲノムDNAから単離した。次いで、このPCR産物を用いて、前記線維芽細胞cDNAライブラリーをスクリーニングし、陽性のクローンを単離し、さらに特性付を行った。製造業者の使用説明書にしたがい、1個の陽性クローン、ファージ3AにLambda ZapIIシステム切除プロトコール(カリフォルニア州、ラ・ホーヤのストラタジーン社)を施した。この方法によりプラスミドpBSAG3Aを得た。この産物は、pBluescriptSK(商標)−プラスミド骨格にα−Gal A cDNA配列を含有している。DNA塩基配列決定により、このプラスミドは、前記cDNA配列の完全5’末端を含まないことが分かった。したがって、ヒトゲノムDNAから増幅したPCRフラグメントを用いてこの5’末端を再構築した。このことを行うために、以下のオリゴヌクレオチド:
5’−ATTGGTCCGCCCCTGAGGT−3’(オリゴ3;配列番号8)および
5’−TGATGCAGGAATCTGGCTCT−3’(オリゴ4;配列番号9)
を用いて268bpのゲノムDNAフラグメント(図2、配列番号2)を増幅した。この断片を「TA」クローニングプラスミド(カリフォルニア州、サンディエゴのインビトロゲン社(Invitrogen Corp.))中にサブクローニングして、プラスミドpTAAGEIを産生した。α−Gal A cDNA配列の大部分を含有するプラスミドpBSAG3A、およびα−Gal A cDNAの5’末端を含有するpTAAGEIの各々をSacIIおよびNcoIで切断した。増幅されたDNAフラグメント内の関連するSacIIおよびNcoIの位置は図2に示している。pTAAGEI由来の0.2kbのSacII−NcoIフラグメントを単離し、同等に切断したpBSAG3Aに接合した。このプラスミドpAGALは、前記α−Gal Aシグナルペプチドをコードしている配列を含む完全α−Gal A cDNA配列を含有する。このcDNAを完全に塩基配列決定し(図3に示したように、α−Gal Aシグナルペプチドを含む;配列番号3)、ヒトα−Gal A cDNAについて発表された配列と同一であることが分かった(Genbank配列HUMGALA)。
α−GalAを発現するpXAG-28の構築に使用するため、以下のようにしてヒトコラーゲンIα2プロモーターを単離した。ヒトコラーゲンIα2プロモーターの一部を含むヒトゲノムDNAの408bpのPCRフラグメントは、次のオリゴヌクレオチド:5’−TTTTGGATCCGTGTCCCATAGTGTTTCCAA−3’(オリゴ72;配列番号16)および5’−TTTTGGATCCGCAGTCGTGGCCAGTACC−3’(オリゴ73;配列番号17)を用いて単離した。
繊維芽細胞内でα−GalAを発現させるため、二次繊維芽細胞を培養し、既報の方法に従ってトランスフェクトした。セルデン(Selden)、WO 93/09222。
α−GalA活性は、イアノー(Ioannou)らによって記載(J. Cell Biol., 119: 1137-1150(1992))されたプロトコールを変形し、水溶性基質である4−メチルウンベリフェリル−α−D−ガラクトピラノシド(4−MUF-gal;リサーチ・プロダクツ(Research Products)社)を用いて測定した。基質を基質緩衝液(0.1Mのクエン酸−リン酸緩衝液、pH4.6)に溶解し、濃度を1.69mg/ml(5mM)にした。一般的には、75mlの基質溶液に10mlの培養上清を加えた。試験管にふたをし、37℃の水槽内で60分間インキュベートした。インキュベート時間の終わりに2mlのグリシンーカルボネート緩衝液(130mMのグリシン、83mMの炭酸ナトリウム、pH10.6)を用いて反応を停止した。固定励起波長が365nmであり、460nmの固定発光波長を検出するモデルTK0100フルオロメーター(ホーファー・サイエンティフィック・インストゥルメンツ(Hoefer Scientific Instruments)社)を用いて各サンプルの相対蛍光を測定した。サンプルの読みとり値は、1mMのメチルウンベリフェロン(シグマ(Sigma)社)のストック溶液から調製した標準サンプルと比較し、加水分解された基質の量を計算した。α−GalAの活性はユニットで表し、α−GalA活性の1ユニットは、37℃において1時間に加水分解された1nmolの基質量と等価である。細胞発現データは、24時間に106個の細胞によって分泌されたα−GalA活性のユニットとして表した。このアッセイを用いることにより、細胞溶解物中、および以下に記載するようなα−GalAの精製過程の様々な段階から得られたサンプル中のα−GalA活性量を測定することもできる。
遺伝子活性化されたα−GalA(GA-GAL)の産生は、実質的に米国特許第5,733,761号に記載されているGA技術を用い、ヒトα−GalAコード配列の上流に制御および構造DNA配列を挿入することによって行った。ここで、前記特許を参考として本明細書中に取り入れておく。遺伝子活性化配列の正確な挿入は、トランスフェクトしたDNAフラグメント上に存在するDNAとヒト細胞内のα−GalA遺伝子座の上流に存在するゲノムDNA配列との間において相同的組換えを行うことによって達成することができる。遺伝子活性化配列は、それ自身がシグナルペプチド解裂部位までのα−GalAコード配列を有するが、該解裂部位は含まれていない。活性化されたα−GalA遺伝子座を含む細胞を単離し、薬剤選択を行い、GA-GAL産生が増加している細胞を単離した。
胞系内の内因性α−GalA遺伝子座の一部を活性化するように設計された配列、およびヒトα−GalA以外のシグナルペプチドをコードしている配列を有する。標的構築体は、バクテリアのneo遺伝子およびマウスのdhfr遺伝子用の発現カセットをも有する。このことにより、安定的に組み込まれた標的フラグメントの選択(neo遺伝子を介して)、およびそれに続いてメトトレキセート(MTX)による段階的選択を用いたdhfr遺伝子の選択を行うことができる。
以下に記載するのは、α−GalAの産生、精製および試験に関する好ましい方法である。精製過程においては、精製過程を通してα−GalAの可溶性、活性、天然由来の型を維持していた。タンパク質は、極端なpH、有機溶媒または界面活性剤にさらされることなく、精製過程中にタンパク質分解を起こすことなく、さらに、凝集体を形成しなかった。精製過程は、α−GalAグリコフォームの分布状態を変化させないように計画した。
実施例2.1は、安定的にトランスフェクトされて酵素を産生している培養ヒト細胞株のコンディションドメディウムからほぼ均質な状態でα−GalAが精製され得ることを示している。α−GalAは、5段階の連続したクロマトグラフィーを用い、α−GalA含有培地から単離した。5つの段階には、酵素の別異の物理学的特性を利用して混在物質からα−GalAを分離するような多様な分離原理を適用した。分離法としては、ブチルセファロース(butyl Sepharose)を用いた疎水性相互作用クロマトグラフィー、ハイドロキシアパタイトを用いたイオン性相互作用、Qセファロース(Q Sepharose)を用いた陰イオン交換クロマトグラフィー、およびスーパーデックス200(Superdex200)を用いた分子ふるいクロマトグラフィーが挙げられる。精製過程の最終段階を実施することに加えて、分子ふるいクロマトグラフィーは、精製されたタンパク質を製剤相溶性緩衝液に交換する有効な手段としても作用する。
冷コンディションドメディウム(1.34l)は、遠心分離およびグラスファイバープレフィルターを用いて0.45μmの酢酸セルロースフィルターを通したろ過により清澄化した。撹拌しながら、1NのHClを滴下することによって、ろ過した冷培地のpHを5.6に調整し、さらに、3.9Mの超純粋硫酸アンモニウムの保存溶液(室温)を滴下し、硫酸アンモニウムの最終濃度を0.66Mにした。培地は4℃でさらに5分間撹拌し、上述したようにろ過し、ブチルセファロース4ファストフロー(butyl Sepharose4 Fast Flow)カラム(カラム容量81ml、2.5×16.5cm、ファルマシア(Pharmacia)社(スウェーデン、ウプサラ)にかけたが、このとき、カラムは0.66Mの硫酸アンモニウムを含む10mMのMES-Tris(pH5.6)(緩衝液A)で平衡化してから使用した。クロマトグラフィーは、4℃においてグラディ−フラクシステム(Gradi-Frac(登録商標) System)(ファルマシア(Pharmacia)社、スウェーデン、ウプサラ)を用いて行い、総タンパク質量および塩濃度を測定するためのUV(280nm)および伝導性モニターを並列に配備した。流速10ml/分でサンプルを添加した後、10カラム容量の緩衝液Aを用いてカラムを洗浄した。緩衝液A(硫酸アンモニウムを含む)から10mMのMES-Tris(pH5.6)(硫酸アンモニウムを含まない)まで直線的に濃度を勾配させたところ、14カラム容量目にブチルセファロース(butyl Sepharose)カラムからα−GalAが溶出した。4−MUF−galアッセイを用い、α−GalA活性に関してフラクションのアッセイを行い、明らかな酵素活性を含むフラクションを集めた。図8および精製のまとめ(表5)に示すように、この段階によって混在タンパク質の約99%が除去された(カラム前のサンプル=総タンパク質量8.14g、カラム後のサンプル=総タンパク質量0.0638g)。
ブチルセファロース(butyl Sepharose)カラムのピークフラクションは、4℃、10mMのMES-Tris(pH5.6)(4l)中で透析した(途中で一度交換した)。透析物の伝導度は、必要に応じてH2OまたはNaClを添加することにより、4℃において1.0mMHOに調整した。その後、サンプルをヘパリンセファロース6ファスト・フロー(Heparin Sepharose6 Fast Flow)(ファルマシア(Pharmacia)社、スウェーデン、ウプサラ)カラム(カラム容量29ml、2.5×6cm)にかけたが、このとき、カラムは9mMのNaClを含む10mMのMES-Tris(pH5.6)(緩衝液B)で平衡化してから使用した。この操作は4℃において流速10ml/分で行った。並列に配備したUV(280nm)および伝導性モニターにより、総タンパク質量および塩濃度を測定した。サンプルの添加後、10カラム容量の緩衝液Bでカラムを洗浄し、次に、3カラム容量で8%の緩衝液C(250mMのNaClを含む10mMのMES-Tris(pH5.6))/92%の緩衝液Bまで直線的に濃度を勾配させ、さらに、8%の緩衝液Cを10カラム容量用いてカラムを洗浄した。続いて、1.5カラム容量で29%の緩衝液Cまで直線的に濃度を勾配させ、続いて、10カラム容量で35%の緩衝液Cまで濃度を勾配させることによってα−GalAを溶出させた。フラクションのα−GalA活性をアッセイし、適切な活性を有するフラクションを集めた。
ヘパリンカラムから得られた活性を有するフラクションを集めたものをろ過し、セラミックハイドロキシアパタイトHC(40μm、アメリカン・インターナショナル・ケミカル(American International Chemical)社、マサチューセッツ州ナティック)カラム(カラム容量12ml、1.5×6.8cm)に直接かけたが、このとき、カラムは1mMのリン酸ナトリウム(pH6.0)(緩衝液D)で平衡化してから使用した。クロマトグラフィーは、室温において、UV(280nm)および伝導性モニターを並列に配備したハイブリッドグラディ−フラク/FPLCシステム(Gradi-Frac/FPLC(登録商標) System)(ファルマシア(Pharmacia)社、スウェーデン、ウプサラ)を用いて行った。サンプルの添加後(5ml/分)、10カラム容量の緩衝液Dを用いてカラムを洗浄した。7カラム容量で42%の緩衝液E(250mMのリン酸緩衝液(pH6.0))/58%の緩衝液Dまで直線的に濃度を勾配させ、続いて、10カラム容量で52%の緩衝液Eまで濃度を勾配させることによってα−GalAを溶出させた。フラクションのα−GalA活性をアッセイし、適切な活性を有するフラクションを集めた。
ハイドロキシアパタイトカラムから得られた活性を有するフラクションを集めて水で約1.5倍に希釈し、室温における最終伝導度を3.4〜3.6mMHOに調整した。ろ過後、サンプルをQセファロースHP(Q SepharoseHP)(ファルマシア(Pharmacia)社、スウェーデン、ウプサラ)カラム(カラム容量5.1ml、1.5×2.9cm)にかけたが、このとき、カラムは10%の緩衝液G(25mMのリン酸ナトリウム(pH6.0)、250mMのNaCl)/90%の緩衝液F(25Mのリン酸ナトリウム(pH6.0))で平衡化してから使用した。クロマトグラフィーは、室温において、ハイブリッドグラディ−フラク/FPLCシステム(Gradi-Frac/FPLC System)(ファルマシア(Pharmacia)社、スウェーデン、ウプサラ)を用いて行い、並列に配備したモニターで総タンパク質量および塩濃度をモニターした。サンプルは流速5ml/分で流し、続いて次のような操作を行った。(1)5カラム容量の10%の緩衝液Gを用いて洗浄、(2)7カラム容量の12%の緩衝液Gを用いて洗浄、(3)3カラム容量で50%の緩衝液Gまで直線的に濃度を勾配、(4)10カラム容量で53%の緩衝液Gまで濃度を勾配、(5)3カラム容量で100%の緩衝液Gまで濃度を勾配、さらに、(6)10カラム容量の100%の緩衝液Gでカラムを洗浄。α−GalAは、段階(3)および(4)の間に主に溶出した。適切な活性を有するフラクションを集めた(「Qプール」と名付けた)。
Qプールは、セントリプレップ−10(centriprep(登録商標)-10)遠心性濃縮器ユニット(アミコン(Amicon)社、マサチューセッツ州ビヴァリー)を用いて約5倍に濃縮し、スーパーデックス200(Superdex200)(ファルマシア(Pharmacia)社、スウェーデン、ウプサラ)のカラム(カラム容量189ml、1.6×94cm)にかけた。カラムは、150mMのNaClを含む25mMのリン酸ナトリウム(pH6.0)を用いて平衡化し、溶出させた。クロマトグラフィーは、室温において、UV(280nm)モニターを並行に配備したFPLCシステム(ファルマシア(Pharmacia)社、スウェーデン、ウプサラ)上で行い、タンパク質の溶出を追跡した。カラムに添加したサンプルの容量は2ml以下、流速は0.5ml/分、フラクションの容量は2mlとした。複数のカラムを用いて分離を行い、フラクションのα−GalA活性をアッセイし、適切な活性を有するフラクションを集めた。
精製過程を実施することにより、高度に精製されたα−GalAが得られた。精製の大部分は最初の2段階で行われ、最後の3つの段階では、残留している微量混在物質を除去することにより、材料に仕上げを施した。最後の段階であるスーパーデックス200(Superdex200)を用いた分子ふるいカラムクロマトグラフィーもα−GalAを製剤相溶性緩衝液に交換する働きをした。
精製ヒトα−GalAの構造的および機能的特性について精査した。還元条件下で4〜15%のSDS−ポリアクリルアミドゲルを用いて行った電気泳動およびその後の銀染色により、得られた生成物は純度が高いことが示された。
本発明に従って産生されたα−GalAのグリコシル化パターンについても評価を行った。適切なグリコシル化は、イン・ビボ(in vivo)におけるα−GalAの活性を最適化するために重要であり、グリコシル化が行われない系において発現されたα−GalAは不活性または不安定である。ハンゾポロウス(Hantzopolous)ら、Gene 57: 159(1987)。グリコシル化はα−GalAが所望する標的細胞内に移行する際にも重要であり、イン・ビボ(in vivo)における酵素の循環半減期にも影響を与える。α−GalAの各サブユニット上には、アスパラギン結合炭化水素鎖の付加が可能な部位が4個あり、このうちの3個のみが埋まっている。デスニック(Desnick)ら、「遺伝病の代謝的および分子的基礎(The Metabolic and Molecular Bases of Inherited Disease)」(マグロウ・ヒル(McGraw Hill)社、ニューヨーク(1995年))、pp. 2741-2780。
上述したように、α−GalAのグリコフォームの分画は、本明細書に記載した精製過程の様々な段階において行われる。本実施例においては、分子の大きさと電荷によってα−GalAを分画した。これらの、または上述したようなその他のクロマトグラフィー技術を組み合わせることによってα−GalAを分画することもできる。
安定的にトランスフェクトされた細胞によって産生されたα−GalAをα−GalA欠乏症に対する有効な治療剤にするためには、影響を受けるべき細胞によって酵素が取り込まれなければならない。α−GalAは、生体のpHレベル(例えば、血中または腸内液中など)においては、活性が最小限に抑えられている。α−GalAは、リソソームの酸性環境内に取り込まれた場合にのみ、蓄積した脂質基質を最大限に代謝する。この内部移行は、α−GalAがM6Pレセプターに結合することによって媒介され、ここで、M6Pレセプターは、細胞表面に発現しており、エンドサイトーシス経路を介して酵素をリソソームに輸送する。M6Pは普遍的に発現され、ほとんどの体細胞はある程度の量のM6Pを発現している。マンノースレセプターは、糖タンパク質上に露出しているマンノース残基に特異的であり、それほど普遍的に存在しているわけではない。一般的には、マンノースレセプターは、マクロファージおよびマクロファージ様細胞上にのみ見出され、このような種類の細胞内にα−GalAが侵入するための追加の手段を提供する。
緩衝溶液および製剤の調製
α−GalAの精製バルクはα−GalA希釈剤を用いて最終濃度に希釈した。製剤化される精製バルクの容量の基づき、α−GalAの濃度(mg/ml)、ならびに最終製剤中におけるα−GalAの所望濃度、必要なα−GalA希釈剤の容量を決定した。α−GalA希釈剤は、適切な量のWFI、塩化ナトリウムおよび一塩基性リン酸ナトリウムを混合し、ならびに水酸化ナトリウム溶液を用いてpHを6.0に調整することにより、使用の24時間以内に調製した。α−GalA希釈剤の組成は表8に示す。
グリコシル化がα−GalAの生体分布に及ぼす効果を調査することを目的として、α−GalAの精製調製物を順次脱グリコシル化し、各型をマウスに注射した。注射4時間後にマウスの臓器を採取し、組織の免疫組織化学を行い、タンパク質の生体分布における変化がわかるように可視化した。
遺伝子治療を行うためには、α−GalAを産生する自己細胞を移植したものが、標的細胞内のα−GalA欠乏状態を「補正する」ように適切に変形された型の酵素を産生しなければならない。ファブリー細胞にトランスフェクトされたヒト繊維芽細胞によるα−GalA産生の効果を評価するため、トランスウェルズ(Transwells(登録商標))(コスター(Costar)社、マサチューセッツ州ケンブリッジ)内において、ファブリー病患者由来の繊維芽細胞(NIGMSヒト遺伝子突然変異細胞貯蔵所(NIGMS Human Genetic Mutant Cell Repository)をα−GalA産生細胞株(BRS-11)と共培養した。ファブリー細胞は、12ウェルの組織培養皿で培養し、そのうちのいくつかには、細胞が増殖可能な表面を有するインサート(inserts)(トランスウェルズ(Transwells)、孔径0.4mm)を入れた。インサートの増殖マトリックスは有孔性であり、巨大分子がマトリックスの上側から下側へ通過することができる。一組のインサートは、最低レベルのα−GalAを分泌する正常ヒト包皮(HF)繊維芽細胞を含んでいたが、別の組は、安定的にトランスフェクトされたヒト繊維芽細胞系BRS-11を含んでおり、これは、大量のα−GalAを分泌した。α−GalA産生細胞と共培養したウェルにおいては、ファブリー細胞が存在している培地中にα−GalAが侵入することができ、ファブリー細胞によって取り込まれると考えられる。
1.SDS-PAGEまたは逆相HPLCにより測定した場合、少なくとも99.5%の均質性まで精製されたヒトα−Gal A調製物を含む組成物であって、該調製物は、多様なα−Gal Aグリコフォームを含み、少なくとも3.0×106ユニット/mgタンパク質の比活性を有し、かつ、実質的にレクチンを含まないことを特徴とする組成物。
(a)平衡緩衝液中、酸性のpHにおいて、陽イオン交換樹脂に該α−GalAグリコフォームを結合させ、
(b)該平衡緩衝液で該樹脂を洗浄することにより、未結合材料を溶出させ;さらに、
(c)10〜100mMの塩溶液、pH4〜5の緩衝溶液、およびそれらの組み合わせよりなる群から選択される溶出液を用いて該α−GalAグリコフォームを溶出させる、工程を含み、
該α−GalAは少なくとも99.5%の均質性まで精製されており、かつ、該調製物はレクチンを実質的に含まないことを特徴とする方法。
(a)GlcNAcトランスフェラーゼIII(GnT-III)の発現をコードしているポリヌクレオチドをα−GalA産生細胞内に導入する、または相同的組換えにより、内在性のGnT-III遺伝子の発現を制御する制御配列を導入し、
(b)α−GalAおよびGnT-IIIを発現するような培養条件下において該α−GalA産生細胞を培養し;さらに、
(c)α−GalAを単離する、工程を含み、
該α−GalA調製物は、前記ポリヌクレオチドを有していない細胞から産生されたα−GalAと比較して、オリゴ糖の電荷が高められていることを特徴とする方法。
(a)シアリルトランスフェラーゼの発現をコードしているポリヌクレオチドをα−GalA産生細胞に導入し、または、相同的組換えにより、内在性シアリルトランスフェラーゼの発現を制御する制御配列を導入し、
(b)α−GalAおよびシアリルトランスフェラーゼを発現するような条件下において該α−GalA産生細胞を培養し;さらに、
(c)前記α−GalAを単離する工程を含み、
該α−GalA調製物は、前記ヌクレオチドを導入していない細胞において産生されたα−GalAと比較して、オリゴ糖電荷が高められていることを特徴とする方法。
(a)ホスホリルトランスフェラーゼの発現をコードしているポリヌクレオチドをα−GalA産生細胞に導入し、または、内在性ホスホリルトランスフェラーゼの発現を制御する制御配列を相同的組換えによって導入し、
(b)α−GalAおよびホスホリルトランスフェラーゼを発現するような条件下において該α−GalA産生細胞を培養し;さらに、
(c)α−GalAを単離する工程を含み、
該α−GalA調製物は、前記ポリヌクレオチドを導入していない細胞において産生されたα−GalAと比較して、ホスホリル化の割合が高められていることを特徴とする方法。
(a)α−GalAをノイラミニダーゼ(シアリダーゼ)と接触させることによってシアル酸残基を除去し、末端ガラクトース部位を露出させ;さらに、
(b)工程(a)の脱シアリル化α−GalAをβ−ガラクトシダーゼと接触させることによって末端ガラクトース残基を除去する、工程を含み
工程(b)の生成物である脱シアリル化脱ガラクトシル化α−GalAは、酵素との接触を行っていないα−GalAと比較して、オリゴ糖鎖上の末端のシアル酸数、またはガラクトース残基数が減少していることを特徴とする方法。
Claims (4)
- 多様なα−GalAグリコフォームを含む精製α−GalA調製物を作成する方法であって、
(a)20mMのリン酸ナトリウムを含む平衡緩衝液中、酸性pHにおいて、陰イオン交換樹脂にα−GalAグリコフォームを結合させる工程、
(b)前記平衡緩衝液で前記樹脂を洗浄することにより、非結合材料を溶出させる工程、および、
(c)30〜130mMの塩溶液、pH6の緩衝溶液、およびそれらの組合せからなる群より選択される溶出液を用いて前記α−GalAグリコフォームを溶出させる工程、
を含み、前記α−GalA調製物は、少なくとも99.5%の均質性まで精製され、かつ該調製物はレクチンを実質的に含まず、さらに
a)前記α−Gal A調製物のオリゴ糖の少なくとも35%が電荷を帯びているか、
b)前記グリコフォームが、2〜4個のシアル酸残基を有する少なくとも20%のコンプレックス型グリカンを含むか、
c)Z数によって測定した前記α−Gal A調製物のオリゴ糖電荷が150以上であるか、
d)前記グリコフォームが、平均して少なくとも25〜50%においてホスホリル化されているか、または
e)前記α−Gal A調製物が、バイセクティングGlcNAc残基を有するN−グリカンを含む、方法。 - クロマト集束クロマトグラフィー、金属キレートアフィニティークロマトグラフィーおよびイムノアフィニティークロマトグラフィーからなる群より選択される精製工程をさらに含む、請求項1記載の方法。
- 前記工程(a)において、前記α−GalAグリコフォームをpH6.0で陰イオン交換樹脂に結合させる、請求項1または2記載の方法。
- 前記工程(c)において、130mMの塩溶液を含む溶出液を用いて前記α−GalAグリコフォームを溶出させる、請求項1〜3いずれか1項記載の方法。
