Description
1つの態様において、本発明は、アンドロゲン受容体拮抗薬と組み合わせて良性前立腺肥大(BPH)、前立腺癌、とりわけCRPCを含む前立腺新生物の治療に用いるインスリン様成長因子(IGF)受容体拮抗薬に関する。
他の実施形態において、本発明は、治療上有効量のIGF受容体拮抗薬をそれを必要とする患者に投与すること、及び治療上有効量のアンドロゲン受容体拮抗薬を同じ患者に同じ日に、又はIGF受容体拮抗薬の投与の1、2、3、4、5、6若しくは7日前若しくは後にさらに投与することを含む、良性前立腺肥大(BPH)、前立腺癌、とりわけCRPCを含む前立腺新生物の治療の方法に関する。
本発明は、前立腺新生物の治療に関する。
「前立腺新生物」により、本発明の態様は、前立腺新生物が、良性及び悪性腫瘍を含む前立腺癌、とりわけ去勢抵抗性前立腺癌であり、並びに良性前立腺肥大である場合も含む。
1つの態様において、本発明は、前立腺癌の治療に用いるインスリン様成長因子(IGF)受容体拮抗薬に関する。他の実施形態において、前立腺癌は、ホルモン感受性前立腺癌である。他の実施形態において、前立腺癌は、複合アンドロゲン遮断後の前立腺癌である。他の実施形態において、前立腺癌は、抗血管新生療法により治療された前立腺癌である。他の実施形態において、前立腺癌は、化学療法薬により治療された又は治療される。他の実施形態において、前立腺癌は、放射線療法により治療された又は治療される前立腺癌である。他の実施形態において、前立腺癌は、骨喪失療法、例えば、デノスマブ及びホルモン除去により治療された又は治療される前立腺癌である。

「前立腺新生物」により、本発明のこの態様は、前立腺新生物が、良性及び悪性腫瘍を含む前立腺癌、とりわけ去勢抵抗性前立腺癌であり、並びに良性前立腺肥大である場合も含む。
投与するIGF又はアンドロゲン受容体拮抗薬の「治療上有効量」は、前立腺新生物、特に去勢抵抗性前立腺癌又は良性前立腺肥大を予防、改善又は治療するために必要な最小量である。

化合物
IGF mAb_1は、配列番号39の重鎖及び配列番号40の軽鎖を有するIGFリガンドに対する抗体である。その製造は、国際公開第2010/066868号に開示されている。
IGF mAb_2は、配列番号29の重鎖及び配列番号30の軽鎖を有するIGFリガンドに対する抗体である。その製造は、国際公開第2010/066868号に開示されている。
DU−145(ATCC、HTB−81)、BM−1604(DSMZ、ACC298)、PC−3(ATCC、CRL−1435)、22Rv1(ATCC、CRL−2505)、LNCaP(ATCC、CRL−1740)及びDUCaP細胞(Prof KJ.Pienta、Hallym University、College of Medicine、Seoul、Koreaの実験室で得られた;Lee YG et al., In Vivo 2001;15(2):157-62)は、10%熱不活性化ウシ胎児血清(FCS;JRH、#12103)及び2mM L−グルタミン(GIBCO、#25030)を添加したRPMI1640増殖培地(GIBCO、#31870)中で培養し、NCI−H660(ATCC、CRL−5813)は、5%FCS、4mM L−グルタミン、5μg/mlインスリン、0.01mg/mLトランスフェリン、30nM亜セレン酸ナトリウム、10nMベータエストラジオール及び10nMヒドロコルチゾンを添加したRPMI中で成長させた。C4−2及びC4−2b(両方がMD Anderson Cancer Centerから使用が許可された;Thalmann GN et al., Cancer Res. 1994;54:2577-2581)並びにVCaP(ATCC、CRL−2876)は、10%熱不活性化FCS、2mM L−グルタミン及びR1881(Sigma、#R0908;0.1nMを有するVCaP及び1nMを有するC4−2/C4−2b)を添加したDMEM(Lonza、#12−604F)中で培養した。MDA−PCa−2b(ATCC、CRL−2422)は、20%熱不活性化FCS、25ng/mlコレラ毒素、0.005mMエタノールアミン、100pg/mlヒドロコルチゾン及び45nM亜セレン酸を添加したF−12K(GIBCO、#21127)中で成長させた。Bob細胞(ECACC、#10021102)は、事前に適格性確認済みのヒト組換え表皮成長因子1−53、ウシ下垂体抽出物及びグルタミン、2ng/ml白血病抑制因子、2ng/ml幹細胞因子、100ng/mlコレラ毒素並びに1ng/ml顆粒球マクロファージコロニー刺激因子を添加したケラチノサイト−SFM(Invitrogen、#37010−022)中で培養した。Shmac4(ECACC、#10112302)、Shmac5(ECACC、#10112303)及びP4E6細胞(ECACC、#10112301)は、Stemline Keratinocyte Growth Supplement(Sigma、#S9945)、2mM L−グルタミン及び2%FCSを含むStemline Keratinocyte Medium II(Sigma、#S0196)中で成長させた。細胞は、加湿雰囲気中5%CO2中37℃で75cm2組織培養フラスコ(Nunc、#178905)中に保持した。
以下の方法を用いて、前立腺癌細胞系の成長に対するIGFリガンド中和mAbs及びアンドロゲンシグナル伝達阻害薬の抑制効果を測定した。アッセイは、10%血清を含む細胞増殖培地中で実施した。
細胞1×106個及び4×106個をそれぞれ6ウェルプレート及び10cmディッシュで10%熱不活性化FCSを含む培地中で平板培養し、一夜インキュベートした後、細胞を1μMのMDV−3100及び100nMのIGF mAb_1又は抗体とARシグナル伝達阻害薬との組合せで処理した。24時間後に細胞をプレート上で溶解し、総タンパク質を単離し、Bradfordアッセイによりタンパク質濃度を測定した。細胞溶解物をスナップ凍結し、−80℃で保存した。
VCaP細胞4×105個を1μMのIGF mAb_1及び10μMのMDV−3100並びに両剤の組合せで処理し、6ウェルプレートで37℃で24時間、48時間及び72時間インキュベートした。その後、上清をFACS管に移し、付着細胞をトリプシンにより分離し、それぞれのFACS管中に収集した。遠心分離した後、培地を捨て、細胞ペレットを氷冷70%エタノールで4℃最低限2時間固定した。エタノールを完全に除去した後、固定細胞を低張緩衝溶液(0.1%クエン酸ナトリウム、0.1%(容積/容積)トリトンX−100、100μg/ml DNアーゼ不含有RNアーゼA)中ヨウ化プロピジウム(10μg/ml;Sigma;P4864−10mL)で染色し、暗所で室温で30分間インキュベートした。細胞をBecton Dickinson FACS Canto IIフローサイトメーターを用いて解析し、データをFACS Divaソフトウエアを用いて評価した。
VCaP細胞を1μMのIGF mAb_1及び10μMのMDV−3100並びに両剤の組合せで処理し、平底96ウェルプレートでR1881の非存在下で3連として1ウェル当たり細胞5×104個の密度で96時間インキュベートした。インキュベーションの最後の24時間、3H−チミジン(0.4μCi/ウェル;PerkinElmer、NET355001MC)を加えた。その後、プレートを凍結し、−20℃で24時間インキュベートした。採取のために、プレートを解凍し、40μLのトリプシンを各ウェルに加えて、細胞断片を分離した。懸濁液をフィルタープレートに移した。次いでプレートを蒸留水で3回洗浄し、60℃で3時間乾燥した。1ウェル当たり25μLのMicroscintを加え、液体シンチレーションカウンター(1450 Microbeta Wallac Trilux、Perkin−Elmer)を用いてチミジンの取込み(CPM;1分当たりの計数)を測定することにより増殖速度を測定した。
細胞3×105個/ウェルのVCaP細胞を1ウェル当たり2mL細胞培養培地中に播種した。播種の24時間後に細胞培養培地を除去し、R1881を含まないDMEM+10%FCSで置換した。培地の交換の24時間後に前処理ウェルを採取し、Beckman Coulter(商標)Vi−CELL XR2.03を用いて計数し、10μMのMDV−3100を残りの細胞に加えた。24時間ごとに4回、各時点について3ウェルにおけるVCaP細胞数を測定した。これらの3連から平均値を計算した。世代時間を決定するために、以下の式を用いる。
種々の濃度のMDV−3100及びIGF mAb_1並びに両剤の組合せによる処理によるカスパーゼ−3/7媒介性アポトーシスを受ける細胞の生存細胞画像を取得するために、CellPlayer(商標)96−Well Kinetic Caspase−3/7 Reagent(Essen BioScience;#4440)を用いた。VCaPs細胞50000個/100μl/ウェルを播種し、翌日、R1881の非存在下で増殖培地中で各濃度の両剤により処理した。Caspase−3/7試薬を増殖培地の1ウェル当たり100μl中5μMの最終濃度に希釈し、培地に加えた。プレートをIncuCyte(商標)2011A中のマイクロプレートトレイ内に入れ、位相コントラスト及び蛍光チャンネルを用いて1ウェル当たり3画像を4時間ごとに7日間にわたり取得した。
前立腺癌細胞増殖に対するIGF及びARシグナル伝達遮断の抑制効果
AR及びIGF−1/2阻害の組合せの抗増殖効果を試験するために、2次元細胞増殖アッセイにおいて、10種の前立腺癌細胞系(Bob、C4−2、C−4−2B、DUCaP、MDA PCa 2b、P4E6、PC−3、Shmac4、Shmac5、VCaP)を、AR拮抗薬MDV−3100及びIGFリガンドに対する全長ヒトモノクローナル抗体(IGF mAb_1及びIGF mAb_2)により、単剤として及び組合せで、処理した(表1)。供試細胞系のうちの3つ(VCaP及びDUCaP−両細胞系は同じ前立腺癌患者の異なる転移の部位に由来していた、並びにMDA PCa 2b)は、AR及びIGFシグナル伝達阻害単独の両方に対する単剤での抗増殖反応、並びに組み合わせた場合の効果の増大を示した(図1)。
前立腺癌細胞増殖に対するIGFシグナル伝達及びアンドロゲン合成遮断の抑制効果
アンドロゲン及びIGFシグナル伝達阻害の組合せの可能性を試験する第2のアプローチとして、8種の前立腺癌細胞系(22Rv1、BM1604、DU−145、DUCaP、LNCaP、MDA PCa 2b、PC−3、VCaP)を、CYP17A及びひいてはアンドロゲンの新規合成を選択的に阻害する、アビラテロン酢酸塩により、単独で及びIGFリガンド中和モノクローナル抗体(IGF mAb_1及びIGF mAb_2)との組合せで処理した。これらのアッセイの結果によってもVCaP、MDA PCa 2b及びDUCaP細胞が単剤及び組合せ処理の両方に対して反応性である唯一の細胞系であることが確認された。しかし、アビラテロン酢酸塩による処理は、アビラテロン酢酸塩が抗増殖作用を示す腫瘍細胞による自己分泌アンドロゲン産生を意味する。これは、アビラテロン酢酸塩処理に対して感受性の細胞の数を制限する可能性がある。VCaPの2次元及び3次元増殖アッセイ並びにMDA PCa 2b及びDUCaP細胞の2次元アッセイの結果を図2に示す。これらのデータは、IGFリガンドを中和する抗体との組合せにより細胞増殖に対するアビラテロン酢酸塩の単剤効果を増大させることができることを示唆するものである。
アンドロゲン受容体及びIGF−1Rの存在並びにPTEN及びwt PIK3CAの発現がアンドロゲン及びIGFシグナル伝達阻害薬の組合せに対して感受性の前立腺癌細胞を特徴づける
図3にAR及びIGFシグナル伝達阻害に対して感受性である、VCaP、MDA PCa 2b及びDUCaP細胞系におけるシグナル伝達タンパク質発現を非感受性細胞系PC−3と比較して示す。細胞をMDV−3100及びIGF mAb_1により、単剤として及び組合せで、24時間処理し、タンパク質溶解物を、IGF−1R、AR、PTEN及びAKTのタンパク質発現について、並びにAKT−Ser473のリン酸化について、無処理対照と比較した。反応性の細胞系は、wt AR、IGF−1R及びPTENを発現した。これらの特性は、PC−3又は単剤処理若しくは両剤の組合せのいずれか1つに対して抗増殖反応を示さなかった他の供試細胞系には存在しなかった(表1)。
これらの結果は、アンドロゲン受容体、IGF−1R並びにPTEN(及びwt PIK3CA)の発現の存在下では、アンドロゲン及びIGFシグナル伝達阻害薬の組合せがin vitroでの前立腺癌細胞増殖の阻害における有効性の増大をもたらすことを示すものである。
MDV−3100及びIGF mAb_1の組合せ処理後の長期AKTリン酸化阻害
AKTリン酸化の阻害に関する単剤としての、及び組合せ処理での、MDV−3100及びIGFリガンドmAb(IGF mAb_1)の効果を処理の4時間から120時間までウエスタンブロットにより解析した。両剤の組合せは、抗体処理単独よりもAKTリン酸化の完全且つ長期持続的阻害をもたらした(図4)。
IGF mAb_1及びMDV−3100による組合せ処理はVCaP細胞におけるアポトーシス誘導に対する相乗効果をもたらす
図1に示すデータの裏付けとして、図5に示すトリチウム化チミジン取込みアッセイから得られた結果から、MDV−3100及びIGF mAb_1の両方の単独が細胞増殖に対する抑制効果(約50%)を有するが、両剤の組合せがはるかにより有効であったことが示された。IGF mAb_1単独によるVCaP細胞の処理は、位相コントラスト顕微鏡法(図6A)、カスパーゼ3活性(図7)、FACSベースの細胞周期解析(図8)及びPARP切断(図9)により評価したとき、アポトーシスのさほど大きくない増加をもたらした。これに対して、MDV−3100単独による処理後に認められた細胞数の減少(図6A)は、長い細胞倍加時間(図6B)に起因していた。MDV−3100は、カスパーゼ3活性(図7)、サブG1アポトーシス細胞集団(図8)又はPARP切断(図9)を誘導しなかった。しかし、IGF mAb_1とMDV−3100を組み合わせたとき、サブG1アポトーシス細胞集団の増加(図8)及びPARPの切断(図9)に加えて、カスパーゼ3活性に対する相乗効果が認められた(図7)。
エンザルタミドと組み合わせるIGF mAb_1の提案される試験
はじめに
ここに提案した試験では、CRPC患者における、単独投与したエンザルタミドと比較して、エンザルタミドと組み合わせたIGF mAb_1の安全性及び抗腫瘍活性を検討する。
IGF mAb_1は、各治療サイクルの開始時の1時間静脈内注入による28日サイクルの治療において週1回投与するものとする。エンザルタミドは、各治療サイクル中の連続経口投与により毎日投与するものとする。
IGF mAb_1は、IgG1イソ型の完全にヒトモノクローナル抗体(HumAb)である。Abは、IGF−1及びIGF−2に高い親和力で結合し、両タンパク質により誘発される増殖及び生存促進シグナル伝達を強力に中和する。
エンザルタミドは、アンドロゲン受容体シグナル伝達経路における種々の段階において作用するアンドロゲン受容体拮抗薬である。化学名は、4−{3−[4−シアノ−3−(トリフルオロメチル)フェニル]−5,5−ジメチル−4−オキソ−2−スルファニリデンイミダゾリジン−1−イル}−2−フルオロ−N−メチルゼンズアミドである。分子量は、464.44であり、分子式は、C21H16F4N4O2Sである。エンザルタミドは、転移性去勢抵抗性前立腺癌(CRPC)を有する患者の治療を適応とする。
投与
IGF mAb_1は、各治療サイクルの開始時の1時間静脈内注入による28日サイクルの治療において週1回投与するものとする。エンザルタミドは、各治療サイクル中の連続経口投与により毎日投与するものとする。
合計最大約140例の患者を試験に募集することができる。併用療法の安全性を保証し、第II部推奨用量を決定するために約15〜18例の患者を試験の第I部忍容性及び安全性相に参加させるものとする。試験の第II部において、120例の患者を2つの試験アームの1つに無作為化し、60例の患者が各アームに無作為化されるものとする(アームB=60、アームB=60)。
試験の第I部は、3箇所以上の施設で実施するものとする。試験の第II部は、10箇所以上の施設で実施するものとする。
試験に組み入れられたすべての患者(すなわち、インフォームドコンセントが行われた)の記録は、それらの患者が治験薬により治療されたか否かにかかわらず、治験施設におけるISFで維持管理するものとする。
この試験に組み入れられるべき患者は、診断され、組織学的又は細胞学的に確認されたCRPCを有し、単独のドセタキソール治療の後に受け、進行していなければならない。患者は、あらゆる状況において、アビラテロン又はカバジタキセル治療の前に受け、失敗してもよく、又はしなくてもよい。
1.患者は、前立腺の組織学的又は細胞学的に確認された腺癌を有する。
2.18歳以上の男性患者
3.転移性前立腺癌(病期M1又はD2)の放射線学的証拠を有する患者。試験治療の開始の28日以内の放射性核種骨スキャン、CTスキャン又はMRIにより評価可能な遠隔転移。
4.ドセタキセルの投与を受けている間又はドセタキセルベースの化学療法の完了の120日以内に疾患の進行(生化学的、臨床的又は放射線学的)を有し、治験責任医師の見解では追加のドセタキセルベースの療法から著しい利益を得ることがありそうもない、又はこの薬剤による療法に不耐性であった患者。
5.患者は、以下の少なくとも1つと定義される進行性疾患の証拠を有さなければならない。
a.進行性測定可能疾患:従来の固形腫瘍基準RECIST1.1を用いる。
b.骨スキャン進行:骨スキャンで少なくとも2つの新たな病変。
c.漸増PSA:少なくとも1週間隔で測定した参照値(PSA#1)と比較して少なくとも2回の連続的漸増PSA値。第3のPSA(PSA#3)は、PSA#2より大きい必要があり、もしそうでなければ、第4のPSA(PSA#4)がPSA#2より大きい必要がある。
7.<50ng/mLの血清テストステロンを有する事前の外科的又は内科的去勢を有する患者。去勢の方法が黄体化ホルモン放出レベルホルモン(LHRH)作動薬である場合、患者は、プロトコール治療中LHRH作動薬の使用を自発的に継続しなければならない。
8.Eastern Cooperative Oncology Group全身状態(ECOG PS)0、1又は2。
9.患者が十分な血液学的機能(絶対好中球数[ANC]≧1500/uL、ヘモグロビン≧9g/dL及び血小板≧100000/uL)を有する。
10.患者が十分な肝機能(ビリルビンが正常の上限(ULN)の≦1.5倍、アスパラギン酸トランスアミナーゼ[AST]及びアラニントランスアミナーゼ[ALT]がULNの≦3倍又は肝臓転移が存在する場合には≦5倍)を有する。
12.ディップスティック又はルーチン尿検査(UA)で≦1+の尿蛋白。尿ディップスティック又はルーチン分析で≧2+が示される場合、試験への参加を許容するために24時間尿を収集し、24時間に<1000mgの蛋白を示さなければならない。
13.十分な凝固機能(国際標準化比[INR]が≦1.5且つ部分トロンボプラスチン時間[PTT]がULNを≦5秒上回る[経口抗凝固療法を受けていない限り])。最大投与量抗凝固療法を受けている患者は、他のすべての基準を満たし、安定量の経口抗凝固剤又は低分子量ヘパリン(許容されていない、ワルファリンを除く)の投与を受けているならば適格である。
14.空腹時血漿グルコースが<8.9mmol/L(<160mg/dL)又はHbA1cが<8.0%。
1.転移性疾患に対する2回を超える事前のタキサンベースの細胞障害性化学療法を受けた患者。ドセタキソールの治療中断の後に第2又は第3のドセタキセルベースの療法を受け、その後の疾患の進行を有する患者は、適格である。
2.あらゆる状況において事前のエンザルタミド治療を受けた患者は、適格でない。
3.試験治療の開始前の4週間以内にアビラテロン又はカバジタキセル治療を受けた患者。
4.進行前立腺癌に対するミトキサントロンによる事前の療法を受けた患者(ミトキサントロンによる事前のアジュバント療法は、許容)。
5.試験投薬の開始の4週間以内に以下のいずれかによる治療を受けた患者:化学療法、免疫療法、生物学的療法、分子標的、ホルモン療法、放射線療法(鎮痛の目的のため又は試験治療の開始前の2週間以内に完了させることができる骨折のリスクのある溶解性病変に対する局所放射線療法の場合を除く)。
6.試験治療の開始前の4週間以内又はこの試験と同時のあらゆる治験薬の使用。
7.試験の開始の4週間以内に、強いCYP2C8阻害薬;強い又は中等度のCYP3A4又はCYP2C8誘導薬;狭い治療指数を有するCYP3A4、CYP2C9及びCYP2C19基質による治療を受けた患者。
8.症候性うっ血性心不全の既往歴を有する又は試験前の心エコー若しくはマルチゲート(MUGA)スキャンを受け、LLNを≧10%下回る左室駆出分画(LVEF)を有する患者。
9.QTcF延長が>450ミリ秒又はQT延長が治験責任医師により臨床的に問題となるとみなされた(例えば、先天性長QT症候群)。QTcFは、スクリーニング時に受けた3回のECGsの平均値として計算する。
11.既知又は疑われる軟膜転移を有する患者。
12.制御されていない又は制御不十分の高血圧。
13.制御不十分の糖尿病を有する患者。糖尿病の既往歴を有する患者は、血糖が正常範囲内にあり(空腹<160mg/dL又はULNを下回る)、この状態に対する安定食事又は治療規制を受けているならば、参加することが許容される。
14.既知のヒト免疫不全ウイルス感染又は後天性免疫不全症候群関連疾患。
15.てんかん、発作又は治験責任医師により判断された発作の素因を有する患者。
16.治験責任医師により判断されたプロトコールを順守することが不可能な患者。
17.治験責任医師により判断された活動中アルコール又は活動中薬物乱用。
18.ヒトモノクローナル抗体に対するアレルギーの既往歴。
19.IGF及び/又はIGFR経路を標的とする薬剤による事前の療法。
20.性的に活動的であり、試験中又は実薬療法の終了後少なくとも3ヵ月間医学的に許容される避妊の方法(例えば、インプラント、注射剤、複合経口避妊薬、関係する女性のある種の子宮内器具又は精管切除パートナー、関係する男性用のコンドームなど)を使用する意志がない患者。臨床試験薬の投与中及び臨床試験薬の最終投与後6ヵ月間まで精液を提供しないことに同意する意志がない男性。
21.任意選択の腫瘍生検を受けることになっている患者については、遺伝性出血障害の既往歴、又は治験責任医師により判断された、過去6ヵ月における臨床的に問題となる大出血事象。

剤形: 液体製剤
供給元: Boehringer Ingelheim Pharma GmbH&Co.KG
単位強度(unit strength): 20mlバイアル入りで供給される10mg/mlのIGF mAb_1。適切な用量のIGF mAb_1を生理塩化ナトリウム溶液(0.9%)で希釈する。
使用期間: 疾患の進行又は過度の毒性の出現まで28日サイクルの治療の各週の開始時(1、8、15及び22日目)に1時間。注入時間は、注入反応又は有害事象の場合には1時間以上に延長することができる。
投与経路: 静脈内
開始用量: 1時間i.v.注入により750mg(最大1000mg)の総投与量
追加情報: 用量は、第I部忍容性/安全性及び用量設定相中に調節する

Claims (13)
- アンドロゲン受容体拮抗薬と組み合わせて前立腺新生物の治療に用いるための、インスリン様成長因子(IGF)受容体拮抗薬を含む医薬組成物であって、
前記アンドロゲン受容体拮抗薬がエンザルタミド及びアビラテロンアセテートから選択され、並びに
前記IGF受容体拮抗薬が、
配列番号31(HCDR1)、配列番号32(HCDR2)及び配列番号33(HCDR3)の重鎖相補性決定領域並びに配列番号34(LCDR1)、配列番号35(LCDR2)及び配列番号36(LCDR3)の軽鎖決定領域を有する抗体mAb1、又は
配列番号21(HCDR1)、配列番号22(HCDR2)及び配列番号23(HCDR3)の重鎖相補性決定領域並びに配列番号24(LCDR1)、配列番号25(LCDR2)及び配列番号26(LCDR3)の軽鎖決定領域を有する抗体mAb2、
である、上記医薬組成物。 - 前記抗体mAb1が、配列番号37の重鎖可変領域及び配列番号38の軽鎖可変領域を有し、又は、前記抗体mAb2が、配列番号27の重鎖可変領域及び配列番号28の軽鎖可変領域を有する、請求項1記載の医薬組成物。
- 前立腺新生物が良性前立腺肥大(BPH)又は前立腺癌である、請求項1又は2に記載の医薬組成物。
- 前立腺癌が去勢抵抗性前立腺癌である、請求項3に記載の医薬組成物。
- 抗体mAb1が配列番号39の重鎖及び配列番号40の軽鎖を有する、請求項1〜4のいずれかに記載の医薬組成物。
- IGF受容体拮抗薬と組み合わせて前立腺新生物の治療に用いるための、アンドロゲン受容体拮抗薬を含む医薬組成物であって、
前記アンドロゲン受容体拮抗薬がエンザルタミド及びアビラテロンアセテートから選択され、並びに
前記IGF受容体拮抗薬が、
配列番号31(HCDR1)、配列番号32(HCDR2)及び配列番号33(HCDR3)の重鎖相補性決定領域並びに配列番号34(LCDR1)、配列番号35(LCDR2)及び配列番号36(LCDR3)の軽鎖決定領域を有する抗体mAb1、又は
配列番号21(HCDR1)、配列番号22(HCDR2)及び配列番号23(HCDR3)の重鎖相補性決定領域並びに配列番号24(LCDR1)、配列番号25(LCDR2)及び配列番号26(LCDR3)の軽鎖決定領域を有する抗体mAb2、
である、上記医薬組成物。 - 前記抗体mAb1が、配列番号37の重鎖可変領域及び配列番号38の軽鎖可変領域を有し、又は、前記抗体mAb2が、配列番号27の重鎖可変領域及び配列番号28の軽鎖可変領域を有する、請求項6記載の医薬組成物。
- 前記抗体mAb1が、配列番号39の重鎖及び配列番号40の軽鎖を有する、請求項6又は7記載の医薬組成物。
- IGF受容体拮抗薬を含む前立腺新生物の治療用組成物であって、前記組成物を用いた治療が、治療上有効量のIGF受容体拮抗薬を必要とする患者に投与すること、及び治療上有効量のアンドロゲン受容体拮抗薬を同じ患者にIGF受容体拮抗薬の投与の前又は後の7日以内にさらに投与することを含み、
前記アンドロゲン受容体拮抗薬がエンザルタミド及びアビラテロンアセテートから選択され、並びに
前記IGF受容体拮抗薬が、
配列番号31(HCDR1)、配列番号32(HCDR2)及び配列番号33(HCDR3)の重鎖相補性決定領域並びに配列番号34(LCDR1)、配列番号35(LCDR2)及び配列番号36(LCDR3)の軽鎖決定領域を有する抗体mAb1、又は
配列番号21(HCDR1)、配列番号22(HCDR2)及び配列番号23(HCDR3)の重鎖相補性決定領域並びに配列番号24(LCDR1)、配列番号25(LCDR2)及び配列番号26(LCDR3)の軽鎖決定領域を有する抗体mAb2、
である、上記治療用組成物。 - 前記抗体mAb1が、配列番号37の重鎖可変領域及び配列番号38の軽鎖可変領域を有し、又は、前記抗体mAb2が、配列番号27の重鎖可変領域及び配列番号28の軽鎖可変領域を有する、請求項9記載の治療用組成物。
- 前記抗体mAb1が、配列番号39の重鎖及び配列番号40の軽鎖を有する、請求項9又は10記載の治療用組成物。
- 前立腺新生物の治療用の医薬の製造のためのIGF受容体拮抗薬の使用であって、
前記IGF受容体拮抗薬がアンドロゲン受容体拮抗薬と組み合わせて用いられるものであり、
前記アンドロゲン受容体拮抗薬がエンザルタミド及びアビラテロンアセテートから選択され、並びに
前記IGF受容体拮抗薬が、
配列番号31(HCDR1)、配列番号32(HCDR2)及び配列番号33(HCDR3)の重鎖相補性決定領域並びに配列番号34(LCDR1)、配列番号35(LCDR2)及び配列番号36(LCDR3)の軽鎖決定領域を有する抗体mAb1、又は

前記アンドロゲン受容体拮抗薬がエンザルタミド及びアビラテロンアセテートから選択され、並びに
前記IGF受容体拮抗薬が、
