Description
GAGは、高度に伸長したコンフォメーションを取り、空間充填性で、圧縮力に耐性であるマトリックスを形成する傾向がある。4つの主要なグループのGAGが、それらの糖残基、それら残基間の連結のタイプ、並びに硫酸基の数及び位置により区別されている。それらには以下のものが含まれる。(1)ヒアルロナン、(2)コンドロイチン硫酸及びデルマタン硫酸、(3)ヘパラン硫酸及びヘパリン、及び(4)ケラタン硫酸。
ヒアルロナンは、皮膚、軟骨、及び眼等の、全ての組織にて様々な量で見出され、成体動物では、全てではないとしても、ほとんどが流体である。ヒアルロナンは、特に初期胚に豊富に存在する。関節軟骨では、HAは、軟骨の機能にとって重要な大型凝集体を形成することができる。更に、細胞運動性及び免疫細胞接着は、細胞表面受容体RHAMM(ヒアルロナン媒介性運動性受容体)及びCD44により媒介される。
哺乳類型ヒアルロニダーゼは、加水分解活性及びトランスグリコシダーゼ活性を両方とも有しており、HA及びコンドロイチンを分解することができる。結合組織では、HAに結合している水和水は、組織間に空間を生み出し、したがって、細胞運動及び増殖に資する環境が生み出される。HAは、迅速な発育、再生、修復、胚形成、発生学的発生、創傷治癒、血管新生、及び腫瘍形成を含む、細胞運動に関連する生物学的現象に重要な役割を果たす。
CD44は、特異的にHAと結合するが、あるコンドロイチン硫酸含有プロテオグリカンも認識される場合がある。CD44は、HAへの接着及び遊走、HA分解、及び腫瘍転移を含む、種々の細胞機能及び生理機能に役割を果たす。また、CD44は、細胞外マトリックス結合、細胞遊走、リンパ球活性化、リンパ球ホーミング、及び気管支平滑筋細胞の増殖に役割を果たすことが示されている(非特許文献1)。CD44受容体は、細胞外ドメインのその可変領域で、複雑なパターンの選択的スプライシングを示す。
CD44は、HAの特に重要な白血球受容体であると考えられており、したがって、喘息の病因に関与している可能性がある。加えて、HAのレベルは、対照マウスでは実験的喘息中に増加するが、抗体処置マウスでは著しく低減され、CD44が、HA代謝に(具体的には、高分子量HAの炎症促進性低分子量形態への分解)に関与していることが支持される。これは、HA由来オリゴ糖がトール様受容体と結合及び活性化することができるため、特に重要であり得る。明らかに、上記結果の中で最も印象的な側面は、抗CD44処置の有益効果の大きさが著しいことである。
CD44−HA相互作用は、T細胞の肺内への遊走を媒介するという概念の通り、Gal−9は、BW5147マウスT細胞のHAへのCD44依存性接着を阻止した。Gal−9は、細胞外マトリックスのCD44依存性白血球認識を調節することにより、気道のアレルギー性炎症及びAHRを阻害すると結論付けられた(非特許文献3)。
シェーグレン症候群(SS)は、全身性エリトマトーデス(SLE)及び関節リウマチ(RA)等の関連自己免疫障害よりも一般的であるが、SSの科学的及び医学的研究は、著しく遅れている。これは、SS遺伝学の分野において特に当てはまり、現在までの努力は、候補遺伝子手法に極度に依存している(非特許文献6)。強直性脊椎炎(AS)は、中軸骨格の偏好を示す一般的な炎症性リウマチ疾患であり、人口の0.2%に影響を及ぼす。現行の診断基準は、臨床的変化及び放射線学的変化の複合に依存し、平均診断時間は5〜10年である(非特許文献7)。結果は、活性化CD44を坦持するTリンパ球の循環は、慢性炎症条件下で上昇し、これらは、自己免疫又は慢性炎症性疾患の活性の信頼できるマーカーを供給することができる活性化循環細胞の病因的に重要な亜集団を代表することができることを示唆する(非特許文献8)。
クローン病(CD)及び潰瘍性大腸炎(UC)を含む炎症性腸疾患(IBD)は、臨床的特徴及び免疫学的特徴が乾癬と共通している。ゲノムワイド関連研究により、共通の感受性遺伝子が見出された。しかしながら、乾癬、乾癬性関節炎、及びIBDのリスク間の関連性を評価する疫学的データは希である。この研究は、米国女性における、乾癬、乾癬性関節炎、並びに付随的なCD及びUC間の関連性を評価することを目的としていた。乾癬性関節炎を伴う乾癬は、付随的なCDのリスク増加と関連している(非特許文献9)。
皮膚筋炎(DM)は、筋肉及び皮膚の炎症を特徴とする多発性筋炎(PM)と関連する結合組織疾患である。DMは、最も頻繁に皮膚及び筋肉に影響を及ぼすが、関節、食道、肺、及びあまり一般的ではないが心臓にも影響を及ぼす場合のある全身性障害である。血管炎は、炎症により血管を破壊する一群の障害である。動脈及び血管は両方とも影響を受ける。血管炎の病態生理は、十分に理解されていないが、続いて起こる炎症応答は、一般的に十分に説明されている(非特許文献10)。ベーチェット病(BD)は、あらゆるサイズの動脈及び血管の両方に関与する唯一の全身性血管炎である。リューマチ診療所では、ベーチェット病が頻繁に見受けられる。ベーチェット病は、いくらか重症な病的状態、及びある場合には致命的転帰さえも示す(非特許文献11)。
特許文献2には、癌及び血管新生依存性疾患を阻害するための、CD44を含むHA結合タンパク質及びペプチドの使用が開示されている。この刊行物では、B16マウス黒色腫及びルイス肺癌の肺転移を阻害するために、メタスタチン、軟骨リンクタンパク質の38kDa断片、並びにこの断片に由来するHA結合ペプチドが使用される。HA結合ペプチドの場合、ニワトリCAMでのB16黒色腫の増殖、及びHAでの内皮細胞遊走が阻害されている。両刊行物では、HA結合ペプチドの使用は、ヒアルロン酸に結合するそれらの能力と直接的に関連する。
これを受けて、本発明者は、更に本発明を発明し、その表面細胞受容体CD44とのHAの相互作用によると考えられる、ヒアルロン酸の組織付着のこの独特の特徴を、このグリコサミノグリカンが他の化合物とコンジュゲートされた場合に維持することができるかどうかを検証する。したがって、更に、本発明者は、HAを標的送達媒体として使用して、薬物をCD44豊富な部位に導くことができるかどうかを検証するために、薬物をHAにコンジュゲートさせる。前述したように、CD44が、炎症、感染症、又は癌が存在する状況中に過剰発現される場合、関連薬物は、リガンドHAが受容体CD44に結合するため、標的部位に容易に到着し、標的部位で相対的に高い濃度を保持することができる。
炎症部位又はCD44豊富な部位に対するHAの付着効果に伴って、コンジュゲート薬物は、標的部分に特に凝集し、その部位での薬物の濃度が比較的より高くなるため療法効力が増強され、したがって、使用される薬物の量が減少され、安全性プロフィールがより良好になるはずである。薬物又は色素のHAとのコンジュゲーションが成功したことを確認し、更にHA結合効果を確認するために、本発明の発明者は、色素をHAにコンジュゲートさせ(HA−色素)、細胞系及びマウスを別々に処置することを含む実験を実施した。
図2A及び図2Bは、細胞系HCT15(CD44がより少ない結腸直腸腺癌)における様々な作用時間での実験を示し、図2C及び図2Dは、細胞系HT29(CD44が豊富な結腸直腸腺癌)における様々な作用時間での実験を示す。HT29(図2C及び図2D)の結果は、HA−色素のコンジュゲーションが成功し、HT29のCD44豊富な領域に結合し(図2C)、更にHT29細胞内に進入したことを示す(図2D)。これは、本発明の発想が適切及び有効であることを意味し、薬物又は色素をHAにコンジュゲートさせることができ、HAは、CD44に結合するその能力を保持することも意味する。
HT29の結合面積の比率は、74.15%であるが、HCT15は、25.85%である。結果は、色素をHAとコンジュゲートさせた場合、色素の濃度は、HAがCD44豊富な部位に結合するため、増加したことを示すことができる。
好ましい実施形態では、感染症には、下気道感染症、HIV/エイズ、下痢性疾患、結核、マラリア、麻疹、百日咳、破傷風、髄膜炎、梅毒、B型肝炎、敗血症、及び熱帯病が含まれる。炎症性異常は、非常に様々なヒト疾患の根底にある障害の大きなグループである。好ましい実施形態では、炎症性疾患は、尋常性ざ瘡、アレルギー性疾患、喘息、自己免疫疾患、セリアック病、慢性前立腺炎、糸球体腎炎、過敏症、炎症性腸疾患、骨盤内炎症性疾患、再潅流傷害、関節リウマチ、サルコイドーシス、移植拒絶反応、血管炎、及び間質性膀胱炎が含まれる。
大多数の抗アレルギー薬は、フェキソフェナジン、セチリジン、マレイン酸クロルフェニラミン、プソイドエフェドリン、及びブデソニド等の、抗ヒスタミン、抗炎症剤、うっ血除去薬、及びステロイドに分割することができる。大多数の抗感染症薬は、皮膚又は粘膜に外部的に使用され、血流に進入せず、肝臓で新陳代謝されない局所的抗感染症手段、及び経口又は注射投与で内臓感染症を治療するために使用される全身性抗感染症手段に分割することができる。大多数の抗炎症薬は、非ステロイド抗炎症剤、又はNSAID、COX−2アンタゴニスト、及びステロイドに分割することができる。
実施例1:結腸組織でのHAの付着(IVIS画像システム−バージョン3) 手順:
1. 0.25gの高分子量ヒアルロン酸ナトリウム粉末(HHA;Mv:2MDa;Freda社)及び0.25gの低分子量ヒアルロン酸ナトリウム粉末(LHA;Mv:0.35MDa;Freda社)を、それぞれ50mLのPBS緩衝液(リン酸緩衝生理食塩水)に添加して、0.5%の溶液を形成し、その後、粉末が完全に溶解するまで6時間撹拌した。0.25gの中分子量ヒアルロン酸ナトリウム粉末(MHA;Mv:1MDa;Freda社)を、50mLのPBS緩衝液に添加し、その後、粉末が完全に溶解し、下記ステップで使用する準備ができるまで、6時間撹拌した。
(1)蛍光性HA(HA−f)を、0.39gのMES遊離酸(2−(N−モルホリノ)エタンスルホン酸、Calbiochem社)により調製し、100mLのdd水に溶解した。
(2)溶液A:65mgのフルオレセインアミン粉末(アイソマーI、Fluka社)を、9mLの95%EtOH溶液に溶解し、その後、光が禁止された条件下で10分間撹拌した。
(3)溶液B:359mgのEDC粉末(N−(3−ジメチルアミノプロピル)−N−エチルカルボジイミド塩酸塩、Sigma社)を、9mLのMES緩衝液に溶解し、その後10分間撹拌した。
(4)溶液C:216mgのNHS粉末(N−ヒドロキシスクシンイミド)、Sigma社)を、9mLのMES緩衝液に溶解し、その後、10分間撹拌した。
(5)3mLの溶液Aを、50mLの0.5%HA溶液にゆっくりと落下させ、その後、光が禁止された条件下で10分間撹拌した。
(6)3mLの溶液B及び5mLの溶液Cを、ステップ(5)の溶液に別々に落下させ、その後、光が禁止された条件下で10分間撹拌した。
(7)0.02MのMES緩衝液を、容積が100mLになるまで、ステップ(6)の溶液にゆっくりと添加し、その後、光が禁止された条件下で室温にて24時間撹拌した。
(8)透析溶液としての5Lのdd水中の透析チューブ(MW:12000〜14000)に、反応後の産物を注ぎ、その後、光が禁止された条件下で4℃にて5日間、12時間毎に透析溶液を換え、透析溶液が蛍光を示さなくなるまで撹拌した。
(9)透析後の液体を、50ccのプラスチック遠心分離管に配分し、その後、−20℃の冷蔵庫で一晩保存し、次に、光が禁止された条件下にて凍結乾燥機で乾燥した。
(10)乾燥したHA−f粉末を、−20℃の冷蔵庫で保存した。
(11)50mgのHA−f粉末を、10mLのPBS緩衝液にゆっくりと添加し、その後、粉末が完全に溶解するまで、6時間撹拌した。
HA−色素コンジュゲーションの以下の全プロセスは、暗所で維持されなければならない。
HA−ADHの合成
1. HA(0.34MDa、50mg)を水に溶解して、4mg/mLの濃度を得た。
2. 5倍過剰のADH(114.8mg)をこの溶液に添加した。
3. 反応混合物のpHを、0.1NのHClを添加することにより4.75に調整した。
4. 次に、1当量のEDC(25.1mg)を固体形態で添加した。反応混合物のpHを、0.1NのHClを添加することにより4.75に調整した。
5. 15分の反応後、0.1NのNaOHを添加することにより反応を停止させ、反応混合物のpHを7.0に調整した。
6. その後、反応混合物を、前処理した透析チューブ(Mwカットオフ3500)に移し、100mMのNaClに対して、次に25%EtOH/水の4サイクル、最後に水に対して、徹底的に透析した。その後、この溶液を、0.2μmセルロースアセテート膜でろ過し、瞬間凍結し、凍結乾燥した。
7. ADHの置換度を、1HNMRで測定した。
1. 88mgのHA−ADH(DS=36%)を、35mLの水に溶解した。
2. 9.5mgのFITCを、10mLのDMSOに溶解した。
3. HA−ADH溶液及びFITC溶液を混合する。
4. 室温で48時間撹拌した後、MWCO12000〜14000の透析バッグを使用して、溶液を、0.3MのNaCl及び純水で交互に3日間透析した。
5. その後、この溶液を2日間凍結乾燥した。
6. 最後に、置換度を、UVスペクトラムで決定した。
(1)1×105個のHT29細胞及びHCT15細胞(ヒト結腸癌、CD44陽性細胞)を、3.5cmディッシュ内の顕微鏡スライドに接種した。
(2)示されている色素濃度、1μMのHA−色素(HA:0.34MDa)を、示されている時間、細胞にそれぞれ添加した。
(3)インキュベーション後、細胞をPBSで洗浄し、その後3.7%ホルムアルデヒドで固定した。
(4)HA−色素と細胞との相互作用の観察を、共焦点顕微鏡で実施した。
手順:
1. 100mgのHA(10K〜700KDa)を25mLのDD水に溶解した。
2. 0.8当量の水酸化テトラブチルアンモニウム(TBA−OH)を、HA溶液に添加し、16時間撹拌した。
3. この溶液を乾燥し、HA−TBA白色固形物を得た。
4. 40mgのHA−TBAを、1mLのDD水に溶解し、その後、この溶液に30mgのEDC及び18mgのNHS粉末を添加し、室温で5分間撹拌した。
5. 4mgのセレコキシブを、2mLのジメチルスルホキシド(DMSO)溶液に溶解した。
6. この混合物(HA−TBA、EDC、NHS、及びセレコキシブ)を、室温で72時間撹拌した。
7. 透析器バッグ(MWCO:1200〜1400)を使用することにより、混合物を、割合が2対1であるDMSO及びDD水に対して1日間透析した。溶液は3回交換した。
8. その後、透析器バッグ(MWCO:1200〜1400)を使用することにより、混合物を、0.3MのNaClに対して2日間透析した。溶液は、1日2回交換した。
9. HA−セレコキシブ溶液を凍結乾燥機で脱水することにより、HA−セレコキシブ粉末を得た。
図3には、HA−セレコキシブコンジュゲート体の構造が示されている。
手順:
1. RAW264.7細胞(1×106細胞/ウェル、1000μL)を、24ウエル培養プレートで、24時間37℃にて5%CO2の存在下で培養した。
2. 10%FBSを含有するDMEMに培地を変更し、細胞を、種々の濃度の化合物(100nMセレコキシブ、HA−セレコキシブ(100nMセレコキシブと等しい)、及び4.9μg/mLのHA)で4時間処理し、その後1μg/mLのリポポリサッカリド(LPS)で24時間処理した。
3. 細胞培地を収集し、PGE2の測定に使用した。
4. Nunc−Immuno 96ウエルプレートを、ヤギポリクローナル抗マウスIgG二次抗体でコーティングした。
5. 細胞培地の分割量を、一次PGE2モノクローナル抗体及びトレーサPGE2アセチルコリンエステラーゼを有する免疫プレートに、暗所で室温にて一晩添加した。
6. 翌日、ウエルを吸引し、100μLの洗浄緩衝液(Cayman Chemical社、アンアーバー、ミシガン州、米国)で5回洗浄した。
7. 200μLのエルマン試薬を各ウエルに添加し、直射日光を避けて室温にて60〜90分インキュベートした。
8. 穏やかな回転/振とうを使用して、発色に必要な時間を減少させた。
9. 吸光度を412nmで測定した。
10. 各試料のPGE2濃度を、PGE2標準曲線から算出した。
11. PGE2量パーセントは、LPS処理された対照群の1つで除算し、次にこの値に100を掛けた、各試料のPGE2レベルの関数としてであった。
12. PGE2の実際の量は、LPS処理した対照群の場合、1370ng/mLであると推定した。
手順:
HA−ADHの合成:
1. HA(50mg)を水に溶解して、4mg/mLの濃度を得た。
2. 5倍過剰のADH(114.8mg)をこの溶液に添加した。
3. 反応混合物のpHを、0.1NのHClを添加することにより4.75に調整した。

1. EDC及びNHSを使用して、ブデソニドスクシナートのカルボキシル基をHA−ADHのアミン基と化学コンジュゲーションすることにより、HA−ADH−ブデソニドスクシナートを形成した。
2. 7.4mgのブデソニドスクシナートを、1mLのDMSOに溶解し、14.5μmolのEDC及び14.5μmolのNHSを含有する1mLの純水と5分間反応させた。
3. 5分後、混合溶液を、20mgのHA−ADHを含有する8mLの水:DMSO=1:1(v/v)に滴加した。
4. 混合溶液を、室温(約30℃)で24時間撹拌した。
5. 24時間後、MWCO12000〜14000の透析バッグを使用して、この溶液を、0.3MのNaCl及び純水で交互に3日間透析した。
6. その後、この溶液を2日間凍結乾燥し、4℃で保管した。
7.最後に、置換度を、1H−NMRで決定した。
結果:図5には、HA−ADH−ブデソニドコンジュゲート体の構造が示されている。
手順:
1. Raw264.7細胞を、フェノールレッド(Gibco−BRL社、ウィーン、オーストリア)を含まない培養培地に懸濁し、106細胞/mLに調整し、10μMのブデソニド、378μg/mLのHA、及びHA−ADH−ブデソニド(10μMブデソニドと等しい)で処理した。
2. 薬物で4時間処理した後、1μg/mLのLPSを、24時間培地に添加した。
3. 培地で処理した細胞は、陰性対照群としての役割を果たしたに過ぎず、陽性対照群としてLPS(1μg/mL)で処理した。
4. その後、培養上清を除去し、誘導型NOS活性を測定するために、グリース反応により酸化窒素を蓄積させた。
5. 100μLのグリース試薬(2.5%のリン酸中の1%スルファニルアミド、0.1%のナフチルエチレンジアミン二塩酸塩)を、100μLの培養上清に添加し、発色は、ELISAマイクロプレートリーダ(SpectraMax(登録商標)M2eマルチモードプレートリーダ、Molecular Devices社、米国)を用いて550nmで評価した。
6. フェノールレッドを含まない培養培地に溶解した亜硝酸ナトリウムの系列希釈を用いて標準曲線を生成した。
結果:図6に示されているように、LPSのみの群と比較すると、HA+LPS、bude.+LPS、及びHA−bude+LPS(「HA−bude」は、「HA−ADH−ブデソニド」を表わす)、群HA+LPS及びHA−bude+LPSは、治療効果を示している。炎症インデックスは、ここでは亜硝酸塩である。
手順:
1. EDC及びNHSを使用して、フェキソフェナジンのカルボキシル基をHA−ADHのアミン基と化学コンジュゲートすることにより、HA−ADH−フェキソフェナジンを合成した。
2. 7.8mgのフェキソフェナジンを、1mLのDMSOに溶解し、73.3μmolのEDC及び73.9μmolのNHSを含有する1mLの純水と5分間反応させた。
3. 5分後、混合溶液を、20mgのHA−ADHを含有する8mLの水:DMSO=1:1(v/v)に滴加した。
4. 混合溶液を、室温(約30℃)で24時間撹拌した。
5. 24時間後、MWCO12000〜14000の透析バッグを使用して、この溶液を、0.3MのNaCl及び純水で交互に3日間透析した。
6. その後、この溶液を2日間凍結乾燥し、4℃で保管した。
7. 最後に、置換度を、1H−NMRで決定した。
結果:図7には、HA−ADH−フェキソフェナジンコンジュゲート体の構造が示されている。
手順:
1. RAW264.7細胞(1×106細胞/ウェル、1000μL)を、37℃にて24時間、5%CO2の存在下で24ウエル培養プレートにてインキュベートした。

4. Nunc−Immuno 96ウエルプレートを、ヤギポリクローナル抗マウスIgG二次抗体でコーティングした。
5. 細胞培地の分割量を、一次PGE2モノクローナル抗体及びトレーサPGE2アセチルコリンエステラーゼを有する免疫プレートに、暗所で室温にて一晩添加した。
6. 翌日、ウエルを吸引し、100μLの洗浄緩衝液で5回洗浄した。
7. 200μLのエルマン試薬を各ウエルに添加し、直射日光を避けて室温にて60〜90分インキュベートした。
8. 穏やかな回転/振とうを使用して、発色に必要な時間を減少させた。
9. 吸光度を412nmで測定した。
10. 各試料のPGE2濃度を、PGE2標準曲線から算出した。
11. PGE2量パーセントは、LPS処理された対照群の1つで除算し、次にこの値に100を掛けた、各試料のPGE2レベルの関数としてであった。
12. PGE2の実際の量は、LPS処理した対照群の場合、1370ng/mLであると推定した。
結果:図8に示されているように、薬物のみの群(フェキソフェナジン)は、治療効果を示した。しかしながら、HAをフェキソフェナジンとコンジュゲートすると(HA−fexo、「HA−fexo」は、「HA−ADH−フェキソフェナジン」を表わす)、薬物のみよりも良好な治療効果を示す。
手順:
1. プレドニゾロン21−ヘミエステルの合成:
2. ピリジン(8mL)中のプレドニゾロン(1g;2.77mmol)及び無水コハク酸(1.125g;12.55mmol)の溶液を、室温で撹拌した。
3. 24時間後、反応混合物を、氷(25g)、水(25mL)、及び濃HCl(10mL)の混合物に注いだ。
4. 分離した結晶をろ過で収集し、水で洗浄し、乾燥し、トルエンから再結晶させ、再び一晩乾燥した。
5. 調製したヘミエステルを、1H−NMR分光法で特徴付けた。
1. 溶液1:73.3μmol(14mg)のEDC及び73.9μmol(8.5mg)のNHSを、1mLの水に添加した。
2. 溶液2:14.5μmol(6.4mg)のプレドニゾロンヘミエステルを、1mLのDMSOに溶解した。
3. 溶液3:20mg(ADH:14.5μmol)のHA−ADH(D.S=31%)を、8mLの共溶媒(水:DMSO=1:1)に溶解した。
4. 溶液1&2を混合し、5分間撹拌して、プレドニゾロンヘミエステルのカルボニル基を活性化する。
5. 5分後、混合物を、撹拌混合下で溶液3に滴加した(1mL/分)。
6. 混合溶液を、室温(約30℃)で24時間撹拌した。
7. 24時間後、MWCO12000〜14000の透析バッグを使用して、この溶液を、0.3MのNaCl/純水で交互に3日間透析した。
8. その後、この溶液を2日間凍結乾燥した。
9. 最後に、HA−ADH−Predの置換度を、1H−NMRで決定した。
結果:図9には、HA−ADH−プレドニゾロンコンジュゲート体の構造が示されている。
1. RAW264.7細胞(1×106細胞/ウェル、1000μL)を、24時間37℃にて5%CO2の存在下で、24ウエル培養プレートにてインキュベートした。
2. 10%FBSを含有するDMEMに培地を変更し、細胞を、種々の濃度の化合物(86.7μMプレドニゾロン、HA−ADH−プレドニゾロン(86.7μMプレドニゾロンと等しい)、及び70μg/mLのHA)で4時間処理し、その後1μg/mLのLPSで24時間処理した。
3. 細胞培地を収集し、PGE2の測定に使用した。
4. Nunc−Immuno 96ウエルプレートを、ヤギポリクローナル抗マウスIgG二次抗体でコーティングした。
5. 細胞培地の分割量を、一次PGE2モノクローナル抗体及びトレーサPGE2アセチルコリンエステラーゼを有する免疫プレートに、暗所で室温にて一晩添加した。
6. 翌日、ウエルを吸引し、100μLの洗浄緩衝液で5回洗浄した。

手順:
1. 30匹の8週齢の雄Sprague−Dawelyラット(BioLASCO Taiwan CO.,Ltd.)を準備した。ラットを、無作為に4つの群に分割した(賦形剤群及びプレドニゾロン群は、n=9、HA−ADH−プレドニゾロン群及びHA群は、n=6)。Institute of Taiwan Animal Technologyのげっ歯動物設備の1つのケージに、ラットを2匹ずつ入れた。
2. 10mg/mLの加熱死ミコバクテリウムを含有する0.1mLのCFA(完全フロイントアジュバント、10mg/mL、Chondrex Inc.社)を右後肢の足蹠に皮下注射する。
3. 針は、足蹠の皮膚下に足首に向かって挿入されるべきである。これにより、流入膝窩リンパ節に対するアジュバントの送達が最大化される。
4. 重症及び急性炎が、注射の30数分以内に観察され、3〜4日以内にピークに達し、20〜25日間続くことが多い。
5. アジュバント誘導性関節炎(AIA)の誘導後の14日目に、プレドニゾロン又はHA−プレドニゾロン(両方とも10mg/kg)、又は対照としての生理食塩水を静脈内注射することにより、ラットを処置した。
6. 処置後の20日目にラットを犠牲にし、その後、組織を単離した。
関節炎測定:
足掌の厚さ及び足首周囲:足掌の厚さは、Mitsutoyo社製デジマチックカリパス等のカリパスを使用して、足掌直径を測定することにより決定した。足首周囲は、2つの垂直な直径、側方−横方向の直径(a)及び前後方向の直径(b)をデジタルカリパスで測定し、以下の式を使用することにより決定した。
また、図12に示されているように、HAのみでさえ、この種類のインデックス(足首周囲の変化)の点で、RA症状を強化した。しかしながら、薬物をHAとコンジュゲートさせると(HA−ADH−Pred群)、その治療効果は、薬物のみよりも更に良好であった。これは統計的に有意である。
Claims (12)
- グリコサミノグリカン及び活性化合物に由来するコンジュゲート体からなる化合物であって、前記活性化合物が、官能基により、アジピン酸ジヒドラジド、ポリペプチド、ペプチド、脂質、及びアミノ酸からなる群から選択される1つのリンカーを介して前記グリコサミノグリカン、その誘導体、又はその塩のカルボキシル酸基に間接的にコンジュゲートされて、共有結合によるコンジュゲーションを形成し、前記活性化合物が、フェキソフェナジン、及びステロイドのうちの何れかからなり、前記ステロイドが、ブデソニドスクシナート及びプレドニゾロンスクシナートからなる群から選択される化合物。
- 前記グリコサミノグリカンがヒアルロン酸である、請求項1に記載の化合物。
- 前記ヒアルロン酸が、5kDaから2000kDaまでの範囲に含まれる平均分子量を有する、請求項2に記載の化合物。
- 急性又は慢性炎症の治療に使用するための請求項1に記載の化合物。
- 前記炎症が、感染症、傷害、自己免疫疾患、又はアレルギーにより誘導又は引き起こされる、請求項4に記載の化合物。
- 前記自己免疫疾患が関節リウマチである、請求項5に記載の化合物。
- 炎症を治療するための方法であって、請求項1に記載のグリコサミノグリカン及び活性化合物に由来するコンジュゲート体からなる治療上有効量の化合物を、その必要性のある対象体(ヒトを除く)に投与するステップを含む方法。
- 前記炎症が、感染症、傷害、自己免疫疾患、又はアレルギーにより誘導又は引き起こされる、請求項7に記載の方法。
- 少なくとも1つの賦形剤及び/又は希釈剤と組み合わせて、請求項1に記載のグリコサミノグリカン及び活性化合物に由来する少なくとも1つのコンジュゲート体を含む医薬組成物。
- 前記組成物が炎症を治療するためもののである、請求項9に記載の医薬組成物。
- 前記炎症が、感染症、傷害、自己免疫疾患、又はアレルギーにより誘導又は引き起こされる、請求項10に記載の医薬組成物。
- 前記組成物が、経口投与用又は注射可能な投与用に設計されている、請求項9に記載の医薬組成物。
