Description
本発明は、米国国立神経疾患・脳卒中研究所によって交付された1R01NS071835の下で、政府の支援を受けてなされた。政府は本発明に一定の権利を有する。
本発明は、タウに対する抗体およびそれらの使用方法に関する。
微小管結合タンパク質タウの凝集は、アルツハイマー病(AD)や前頭側頭型認知症を含むいくつかの神経変性障害と関連している。ADでは、病理学的タウ凝集が、おそらくは既存の神経ネットワークに沿って、脳全体に進行性に拡がる。ADは認知症の最も一般的な原因であり、深刻さを増しつつある公衆衛生上の問題である。これは現在、米国において500万人を苦しめていると推定され、それが2050年までに1300万人に増加すると予想されている。アルツハイマー病は、記憶や認知機能の喪失につながり、最終的には自立できなくなる。これは患者とその家族に大きな人的および経済的犠牲を強いる。ADや、タウの凝集に関連する他の神経変性疾患は、重症度が高く、人口に占める罹患率が増加しつつあることから、より良い処置および検出方法を開発することが急がれている。

アルツハイマー病と全てのタウオパチーとの間の最低限のつながりは、タウの凝集状態である。これらの罹患状態のいずれにおいても、単量体型タウはポリマー状の規則的原線維に転化することが知られている。原線維性タウ凝集体から構成される神経原線維変化(NFT)は、タウオパチーの神経病理学的特徴である。本出願人らは、脳におけるタウ病態の拡がりが「ドナー(donor)」細胞から放出されて第2の「レシピエント(recipient)」細胞に進入し、直接的なタンパク質−タンパク質接触を介してレシピエント細胞におけるタウのさらなるミスフォールディングと凝集を誘導する、タウ凝集体の一形態によって引き起こされうることを発見した。この細胞から細胞へのタウ凝集体の拡がりを容易にする特別な形態のタウ凝集体を「タウシード(tau seed)」と呼び、その活性を本明細書では「シーディング活性」と呼ぶ場合がある。というのも、この形態のタウ凝集体は、それが進入した細胞(すなわち「レシピエント細胞」)におけるタウ凝集の種晶または核になるからである。
ヒトでは、エクソン2、3、および10の選択的スプライシングによって生成するタウのアイソフォームが6種類存在する。これらのアイソフォームは352アミノ酸から441アミノ酸の範囲にある。エクソン2およびエクソン3はそれぞれN末端に29アミノ酸インサートをコードしている(これはNと呼ばれ、したがってタウアイソフォームは2N(両インサート)、1N(エクソン2のみ)、0N(どちらもなし)になりうる)。どのタウアイソフォームでも、微小管結合ドメインが3回繰り返されている。C末端にエクソン10を含めると、エクソン10によってコードされる4つ目の微小管結合ドメインが含まれることになる。したがってタウアイソフォームは、微小管結合ドメインが4回繰り返されている場合(エクソン10が含まれている場合)と、微小管結合ドメインが3回繰り返されている場合(エクソン10が排除されている場合)とがある。本発明の抗タウ抗体は、タウのアイソフォームのいずれかに結合する抗体を含みうる。例示的一実施形態において、本発明の抗タウ抗体は、エクソン10を含むタウのアイソフォームに結合する抗体を包含しうる。
表A.本発明の抗体
一態様において、本発明は、生きている対象に投与される機能的治療組成物において使用するための抗体を提供する。もう一つの態様において、本発明は、生きている対象から得られた生物学的液体の試料中のタウ凝集体を検出するために、イムノアッセイにおいて使用するための抗体を提供する。もう一つの態様において、本発明は、生きている対象から得られた生物学的液体の試料中のタウ凝集体の量を測定する目的で、イムノアッセイにおいて使用するための抗体を提供する。対象から得た生物学的液体の試料中のタウ凝集体の量は、多量または少量のタウ凝集体を有すると対象を分類するために使用することができ、さらに、対象がその生涯にわたってタウ凝集に関連する症状および/または疾患を発症するリスクを予測するために使用することができる。
一態様において、本発明は、対象の脳におけるタウ凝集の拡がりを低減する方法を含む。もう一つの態様において、本発明は、タウシードによって誘導されるタウの細胞内凝集を低減するための方法を含む。それぞれの態様において、本方法は、対象に薬学的有効量の抗タウ抗体を投与することを含む。適切な抗体は上記I項で述べた。好ましい一実施形態では、抗体が、表1の抗体および表2の抗体(そのヒト化抗体、キメラ抗体または免疫学的フラグメントを含む)からなる群より選択される。
一態様において、本発明は、対象から得た生物学的液体の試料中のタウ凝集体を検出するための手段を提供する。もう一つの態様において本発明は、対象から得た生物学的液体の試料中のタウ凝集体の量を測定するための手段を提供する。本方法は、一般に、(i)対象から生物学的液体の試料を得ること、および(ii)タウに特異的に結合する抗体を使って、試料中のタウ凝集体の量を測定することを含む。適切な抗体は上記I項で述べた。適切な対象は上述した。

本明細書にいう「抗体」は、タウを特異的に認識する免疫グロブリン由来分子を指す。本発明の抗体は、完全長抗体(IgM、IgG、IgA、IgE)であるか、抗体フラグメント(Fab、F(ab’)2、scFv)であることができる。抗体はキメラ抗体であるか、ヒト化抗体でありうる。
ニューロンおよびグリアにおける微小管結合タンパク・タウの凝集は、アルツハイマー病(AD)、進行性核上性麻痺、および前頭側頭型認知症を含む20を超える神経変性障害と関連している。ヒト研究から得られた最近の証拠は、タウ病態は脳中にランダムに分布するのではなく、ニューロン接続の既存のネットワークと関係していることを示唆している。ADの原線維性タウ病態は既知の解剖学的接続に沿って進行するが、ネットワークが変性する機序はわかっていない。重要なことに、最近の病理学的研究は、ヒト脳およびマウス脳ではタンパク質凝集体がある細胞から別の細胞へと移動しうることを示唆している。さらにまた、タウ、SOD−1、α−シヌクレインおよびポリグルタミン(polygutamine)などの組換えヒト疾患関連タンパク質の原線維型は、細胞外間隙から容易に取り込まれて、細胞内ミスフォールディングを誘発する。これらの現象は、エキソソームおよびトンネリングナノチューブが経細胞的拡がり(trans−cellular spread)を媒介すると提唱されたプリオン伝播を連想させる。タウ凝集体が細胞から細胞へとタンパク質ミスフォールディングを拡げるのは、直接的な細胞間接触によるのか、それとも細胞外間隙を介して起こるのかは、未解決の問題である。さらにまた、病理学的タウ種が、凝集体が「ドナー」細胞から放出され、第2の「レシピエント」細胞に進入し、他の間接的な機序ではなくて直接的なタンパク質−タンパク質接触によって、さらなるミスフォールディングを誘導するという、凝集の真の経細胞伝播を媒介しうるのかどうかも、まだ決定されていない。ここでは、タウ原線維が細胞外間隙に直接放出されるかどうか、そしてこの機序によって凝集を伝播することができるかどうかを検証する。
標準的な技法を使って2系列の抗タウ抗体、すなわちHJ8系列(組換えヒトタウに対するマウスモノクローナル抗体)およびHJ9系列(組換えマウスタウに対するマウスモノクローナル抗体)を作製した(表1)。結合エピトープは多くの抗体についてマッピングされている(表A)。

マウスタウとヒトタウの両方を認識するタウ抗体を使って、野生型マウスとP301Sヒトタウトランスジェニックマウス(P301S tgマウス、方法の項に詳細)の両方のISF試料から、タウを免疫沈降させた。ISF中の単量体型タウの量は比較的低いので、免疫沈降アッセイにおいてうまく作用する2つの抗タウモノクローナル抗体を使用した。免疫沈降後に、免疫ブロットによってタウを解析した。内在性マウスタウアイソフォームは48〜62kDaに泳動する。野生型脳溶解物では、タウがSDS−PAGE上で4つの独立したバンドに現れた(図14A)。野生型マウスでは最も豊富な種が48kDaに泳動した。P301S tgマウス脳では、4つの内在性マウスタウバンドに加えて、過剰発現したヒト1N4Rタウが、55kDaに泳動する強いバンドとして観察されると共に、タウ分解産物に相当すると思われる39kDaバンドとして観察された。
タウ遺伝子は6つのタンパク質アイソフォームをコードし、複数の変異が優性遺伝性神経変性疾患を引き起こす。スプライシングに依存して、タウタンパク質は、リピートドメイン(RD)と呼ばれる、タンパク質の易凝集性コアを構成する3つまたは4つのリピート領域を有する。タウRDの発現はトランスジェニックマウスにおける病態の原因となり、完全長タウの切断が患者において原線維を含むフラグメントを形成するという証拠がある。このコンストラクトは培養細胞中で確実に原線維を形成するので、完全長タウではなく、このコンストラクトを使用した。タウ凝集を増加させることが知られているさまざまな変異、すなわちΔK280(ΔKと呼ぶ)、P301L、およびV337Mを、4リピートRDタンパク質に工学的に組み入れた。P301L変異体とV337M変異体を合わせて一つのタンパク質(LMと呼ぶ)にすることにより、以前に記載したものと同様に凝集能が著しく増加しているRDの変異体型を作製した。この「非生理的」変異体は、細胞内凝集体の効率のよい形成に依存するミスフォールディングの伝達事象および経細胞伝播のアッセイを容易にし、類似しているがより弱い(less robust)「生理的」ΔK変異体の凝集表現型を補う。βシート形成および原線維化を阻害するが、不定形凝集体の形成は阻止しない2つのプロリン置換、I227PおよびI308P(PPと呼ぶ)も、ΔK変異体に工学的に組み入れた。各形態の変異体タウを、カルボキシル末端で、シアン蛍光タンパク質もしくは黄色蛍光タンパク質(CFPまたはYFP)またはHAタグのいずれかに融合した。コンストラクトを図15Aに図解する。
以前、本出願人らは、ある細胞からのタウ封入体が共培養中のナイーブ細胞に伝達されるであろうことを決定した。しかし、これらの伝達された凝集体がレシピエント細胞中でさらなる凝集を誘導できるかどうかも、凝集の誘導が直接的なタンパク質−タンパク質相互作用に基づくかどうかも証明されていなかった。まず、あるドナー細胞集団に由来するRD(LM)−HA凝集体が、共培養時に、異なるレシピエント集団中のRD(ΔK)−YFPと共に封入体を形成するかどうかを調べた。一つの細胞群に易凝集性RD(LM)−HAをトランスフェクトし、別個の群にはRD(ΔK)−YFPをトランスフェクトした。翌日、それらの細胞集団を一緒に再プレーティングして、48時間、共培養した。固定後に、HA抗体を使ってそれらを免疫染色し、X−34で対比染色した。RD(LM)−HAとRD(ΔK)−YFPが封入体中に共局在している細胞が数多く観察された(図17A)。これらの封入体はX−34についても陽性に染色されることが多く、これはベータシート構造を示している。FRETアッセイを使ってこれらの研究を拡張することにより、RD(LM)−HAを発現する細胞との共培養によって誘導されるRD(ΔK)−CFP/YFPの凝集をモニターした。この例では2つの細胞集団を共培養した。ドナー集団はRD(LM)−HAを発現し、レシピエント集団はRD(ΔK)−CFP/YFPを発現した。βシート耐性型のタウRD(PP)−HAまたはモックトランスフェクト細胞を陰性対照として使用した。48時間後に、細胞単層からのFRETを測定した。RD(PP)−HAまたはモックトランスフェクト細胞との対比で、RD(LM)−HAとの共培養によって誘導されるFRETの著しい増加が観察された(図17B)。RD(LM)−HA細胞をRD(WT)−CFP/YFPレシピエント細胞と共培養すると、FRETシグナルの小さな増加が観察された(非掲載データ)。これらの結果から、一つの易凝集性タウ種がある細胞から別の細胞へと移動して、ベータシートに富む封入体での共局在を誘発することが示唆された。凝集体の放出が細胞死後に起こる可能性はあるが、さまざまなトランスフェクト集団のヨウ化プロピジウム染色では、その証拠は観察されなかった(図17C)。
これらの結果は、共凝集が、直接的なタンパク質接触により、レシピエント細胞中の対応タンパク質と接触するドナー由来のタウRD間の分子間会合を伴って起こったことを強く示唆するものの、その当否を正式に扱うことはできなかった。FRETを使ってこの問題に取り組んだ。まず、RD(LM)−CFPをドナー細胞集団内で発現させ、同時に、RD(LM)−YFPを第2のレシピエント集団内で発現させた。48後に、共焦点顕微鏡法とFPRの両方を使って、細胞単層からのFRETを測定した。共焦点顕微鏡法を使って、YFPのフォトブリーチングの前後で、CFPシグナルを測定した。〜14.2%という平均FRET効率が記録されたことから、封入体は直接接触したRD(LM)−CFPとRD(LM)−YFPを含有することが示された(図18A)。次に、異なる形態の非標識RDを使ってRD−CFPの凝集を誘導することにより、相対的FRETシグナルをFPRで比較した。まず、RD(ΔK)−CFPとRD(LM)−HAをドナー細胞集団内で共発現させ、第2のレシピエント細胞集団内でRD(ΔK)−YFPを発現させた。RD(LM)−HAは、RD(ΔK)−CFPの凝集および移動の強化因子として働き、続いてそれがRD(ΔK)−YFPレシピエント細胞に伝達されるのを促進する。これにより、共培養細胞では、小さいが再現性のあるFRETシグナルの増加がもたらされた。このシグナルは、CFPタグ付きまたはYFPタグ付きのRDコンストラクトが、βシート形成を阻止するPP変異を含有する場合には、消失したことから(図18B)、このペアのどちらのメンバーも、ベータシート構造を形成する能力を有さなければならないことが示された。これらの結果は、先の実験と合わせて考えると、直接接触によるミスフォールディングの伝播が起こること、すなわち凝集体はある細胞を出て、第2の細胞中でネイティブフォールドタンパク質のミスフォールディングを誘発することを示唆している。このデータから、ミスフォールディングの増幅は、逐次的細胞共培養(serial cell co−cultures)でも起こるかもしれないことが暗示された。「ドナー」細胞集団を凝集シードに事前ばく露することにより、レシピエント細胞集団中に検出される最終凝集は増加するだろうと予測された。3つの細胞集団を順次培養することによって、これを調べた。第1集団は、さまざまな形態の非蛍光性RD−HAを発現して、凝集「シード」を形成した。第2群はCFPまたはRD(ΔK)−CFPを発現して、凝集体維持に関して非許容的(CFP)または許容的RD(ΔK)−CFP)である。ミスフォールディングを増幅させるために、これら2つの群を48時間、共培養した。次に、第1群と第2群の複合物の50%を、RD(ΔK)−YFPを発現する第3の細胞群と共に、48時間、共培養した。この第3のレシピエント群は、RD(ΔK)−CFP細胞内凝集および伝播の程度を示す「レポーター」として働いた。RD(ΔK)−CFP集団へのRD(LM)−HAの事前ばく露により、易凝集性タウに事前ばく露しなかった細胞と比較して、最終FRETは2.6倍増加した。予想どおり、第2細胞集団における純粋なCFPを発現する細胞の介在は、タウRD「シード」への事前ばく露の効果を完全に阻止した(図18C)。これらのデータは全体として、逐次的に培養された細胞集団内でのタウ凝集の増幅を示している。
タンパク質凝集体が細胞間を移動する機序はわかっていない。例えばトンネリングナノチューブを介したプリオンタンパク質伝播を推論するものもいたし、エキソソームを示唆するものもいた。タウタンパク質に対する抗体はインビボで病態を低減すると以前に報告されているので、タウ凝集体は細胞外間隙に直接放出されるのかもしれないという仮説を立てた。細胞−細胞接触に基づく経細胞移動が細胞外培地の体積には依存しないはずであるのに対して、タウの経細胞移動は、SOD1について述べられているように、細胞外体積に鋭敏であるかもしれないと予測した。手始めに、さまざまな培地体積の設定下で共培養の効果をまず調べた。細胞培養培地体積を増加させると、凝集体の経細胞移動の効率は低減することが観察された(図19A)。さらに、RD(LM)−HAを発現する細胞からの条件培地の導入は、RD−CFP/YFPを発現する細胞における凝集を誘発するのに十分であった(図19B)。これらの結果は、細胞外間隙を通る細胞間でのタウの移動と合致したが、タンパク質がエンドソームに封入されているかどうかは決定することができなかった。
伝播アッセイにおけるHJ9.3の活性により、原因となるタウ種を明確にする機会が得られた。HJ9.3を使って細胞培地からタウを抽出した。HJ9.3または対照IgGを、さまざまなRDコンストラクト(wt、PP、ΔK、LM)を発現する細胞培地に加えた。抗体は、48時間の培養期間の最初または最後のどちらかに加えた。プロテインG−アガロースビーズを用いる抗体/抗原複合体のアフィニティ精製のために、培地を回収(harvest)した。複合体を洗浄してから、ウェスタンブロットによる解析のために、SDSローディング緩衝液中で煮沸した。HJ9.3は細胞培地からタウRD種を特異的に捕捉したが、IgGには感知しうる効果はなかった(図21A)。培養期間の終了時に添加するのではなく、HJ9.3が培養期間中ずっと存在する場合は、培地中に存在するタウタンパク質が〜10倍増加することが観察された(図21B)。RD(ΔK)−HAおよびRD(LM)−HAトランスフェクト細胞の培地には、RD凝集体と合致する高次分子量種も認められた。RD(PP)−HAタウは、培地中に存在する量が最も少ないタンパク質であり、ウェスタンブロットでは高次種は観察されなかった。先に述べた実験の時間経過(0時間、3時間、6時間、9時間、12時間、24時間および48時間)は、培地中のタウレベルの時間依存的な増加を示した。これは、HJ9.3インキュベーションが条件培地中に存在するタウタンパク質の定常状態レベルを実際に増加させていることを含意する(図21C)。これらのデータは全体として、HJ9.3が、細胞への凝集体の取り込みを妨害することによって、細胞から細胞への伝播を阻止することを示し、細胞の内外へのタウ凝集体の定常状態フラックスと合致している。
完全長組換えヒトタウに対するさらに2つの抗体の活性を伝播アッセイで調べた。RD(LM)−CFP細胞とRD(ΔK)−YFP細胞を、異なるタウエピトープを標的とする異なるモノクローナル抗体(HJ8.5、HJ9.3およびHJ9.4)の存在下および非存在下で48時間、共培養した(図22A)。Aβペプチドに対するHJ3.4抗体を陰性対照として使用した。3つの抗タウ抗体はいずれも、RD−タウの凝集促進変異体の細胞間での経細胞伝播を阻止した(図22B)。陰性対照HJ3.4は経細胞伝播を阻止しなかった。HJ8.5、HJ9.3およびHJ9.4はELISAでもRD−タウ原線維を検出した(図22C)。
鋳型を介したコンフォメーション変化(templated conformational change)と凝集の経細胞伝播とが関与するプリオン様の機序が、タウオパチーおよび他の神経変性疾患の容赦ない進行の説明になりうることは、以前に提唱されている。これは、ドナー細胞からのタンパク質凝集体の放出、レシピエント細胞への進入、およびネイティブフォールドタンパク質との直接的接触によるミスフォールド状態の増幅からなるだろう。しかし、タウオパチーのこのモデルを裏付ける機序的証拠は不完全であり、このようなタウミスフォールディングの経細胞伝播は今まで証明されたことがなかった。実施例3〜7には、分泌されたタウ凝集体による培養細胞におけるタウ凝集の経細胞伝播が記載され、有望な機序が提案されている。まず、トランスフェクト細胞におけるRDタウ原線維の自発的形成が、抽出物のX−34染色およびAFMを使って実証された。次に、2つの別個の細胞に由来するタウが、細胞内封入体中で同所共在することが、共焦点顕微鏡法を使って観察された。これは、易凝集型のタンパク質RD(LM)−HAを発現する細胞と共培養した時の、タウRD(ΔK)−YFPの界面活性剤不溶性の増加と関連した。また、この凝集の増加は、同じ細胞内で共発現したRD(LM)−CFP/YFP間でも、FRETを使って実証された。これは、アクセプターフォトブリーチング(顕微鏡法)とスペクトル法(FPR)によって検出された。次に、別々の細胞集団において発現したRD(LM)−CFPとRD(LM)−YFPの間のFRETを使って、伝播が直接的なタンパク質接触によって起こることを実証した。次に、この方法を拡張して、逐次的培養条件における細胞集団内でのタウタンパク質ミスフォールディングの増幅を実証した。タウ凝集の経細胞伝播は、細胞外間隙中に直接放出された原線維によって媒介される、なぜなら伝達は細胞外体積に鋭敏であり、条件培地は細胞内凝集を増加させることができ、抗タウ抗体(HJ9.3)が細胞間伝播を妨害し、細胞外タウ原線維をトラップしたからである。こうして、本出願人らは、さまざまな技法を使って、鋳型を介したコンフォメーション変化による経細胞凝集体伝播を実証した、また、本出願人らは、これらの現象の説明となる簡単なモデルを提案する(図29)。
抗体
最も長いマウス組換えタウアイソフォームmTau40(432aa)と最も長いヒトタウアイソフォームhTau40(441aa)はEva Mandelkowの研究室で生産され、タウELISAではそれらを標準として使用した。ヒトタウとマウスタウの両方(残基218〜225にあるエピトープ)を認識するマウスモノクローナル抗体Tau−5は、L.Binderの研究室から得た(LoPresti et al., 1995;Porzig et al., 2007)。モノクローナル抗体HJ8.1およびHJ9.3は、タウノックアウトマウス(The Jackson Laboratory)において、それぞれヒトタウおよびマウスタウに対して免疫化することによって生じさせたマウスモノクローナル抗体である。どちらの抗体も、ウェスタンブロット上で、免疫沈降によって、そしてELISAアッセイにおいて、マウスタウとヒトタウを認識する。HJ9.3はタウの微小管結合領域(MTBR)を認識する。同様にヒトタウおよびマウスタウ(残基194〜198にあるエピトープ)を認識するマウスモノクローナル抗体BT−2は、Pierceから入手した。タウに対するウサギポリクローナル抗体(ab64193、リピートドメイン領域中にあるエピトープ)はAbcam(マサチューセッツ州ケンブリッジ)から購入した。ヘマグルチニンHAに対するマウスモノクローナル抗体(HA.11クローン16B12)はCovance(カリフォルニア州エメリービル)から購入した。ウサギポリクローナルGFP抗体(sc−8334)はSanta Cruz Biotechnologyから購入した。
タンパク質発現には微小管結合タンパク質タウの4リピートドメイン(RD)をコードする配列を使用した。野生型に加えて、さまざまなタウ変異体を作製した:ΔK280Δ(K); P301L/V337M(LM);ΔK280/I227P/I308P(PP)。これらの配列を、C末端ヘマグルチニン(HA)タグと共にpcDNA3.1(Invitrogen)にサブクローニングするか、またはpEYFP−N1もしくはpECFP−N1(Clontech)にサブクローニングして、C末端蛍光タンパク質融合物を作製した。
P301SヒトT34アイソフォームタウ(1N4R)を過剰発現するP301S tgマウス(PS19系統)は以前に作出し、特徴づけたもので、B6C3背景を持つ。P301S tgマウスはThe Jackson Laboratoryから入手した。タウノックアウトマウスはThe Jackson Laboratoryから入手した。同年齢および同遺伝的背景の非トランスジェニックマウス同腹仔を野生型マウスとして使用した。どの実験でも、本研究では雄と雌の両方を使用した。
免疫沈降および免疫ブロット解析.P301Sタウトランスジェニックマウスおよび野生型マウスから海馬微小透析試料を1.0 l/分の流量で15時間収集した。製造者の説明書に従ってHJ8.1またはHJ9.3タウ抗体でコーティングしたDynabeads(Invitrogen)により、ISFを免疫沈降した。沈降した画分を還元4-12%ビス−トリスミニゲル(Invitrogen)にローディングし、ニトロセルロース膜に転写した。沈降した抗体の妨害を排除するために、ビオチン化BT−2抗体(Pierce)およびポリHRPコンジュゲートストレプトアビジン(Thermo Scientific)を使用した。HEK293細胞は、10%ウシ胎児血清、100μg/mLペニシリンおよび100μg/mLストレプトマイシンを補足したダルベッコ変法イーグル培地(DMEM)で培養した。培養物は5%CO2の湿潤雰囲気下、37℃で維持した。一過性トランスフェクションには、Lipofectamine/Plus試薬と600ngの適当なDNAコンストラクトとを使用し(Invitrogen、米国カリフォルニア州カールズバッド)、製造者の推奨に従って、Optimem培地にプレーティングした細胞をトランスフェクトし、24時間後または48時間後に、さらなる解析のために回収(harvest)した。
HEK293細胞を12穴プレートに400,000細胞/ウェルでプレーティングした。翌日、細胞に600ngのプラスミドをトランスフェクトした。48時間後に、37℃で3分間、0.05%トリプシンを使って、細胞を回収(harvest)し、7000×gで手短にペレット化し、プロテアーゼ阻害剤を含有するPBS中の1%Triton 100μl中で溶解した。次に、14,000×gで10分間の遠心分離によって可溶性サイトゾルタンパク質を収集した。ペレットをRIPA/SDS緩衝液に再懸濁し、20,000×gで15分間の遠心分離後に、核酸のベンゾナーゼヌクレアーゼ消化を行うことにより、不溶性タンパク質を得た。共培養実験の場合は、RD(LM)−HAおよびRD(ΔK)−YFPがトランスフェクトされた同数の細胞を48時間、共培養してから、回収(harvest)し、ウェスタンブロッティングを行なった。各フラクションから等量のHEK293細胞タンパク質抽出物を、4%−20%ポリアクリルアミドゲル(Biorad);1:2000希釈のタウRDに対する抗体(RD領域中のエピトープを認識するもの)(ab64193、Abcam、マサチューセッツ州ケンブリッジ)および/または1:1000希釈のGFPに対する抗体(sc−8334、Santa Cruz Biotechnology, Inc.)を使って解析した。化学発光に基づくペルオキシダーゼコンジュゲート二次抗体反応を行い、X線フィルムで検出した。定量はImage J解析ソフトウェアを使って行なった。
HEK293細胞を12穴プレートに300,000細胞/ウェルでプレーティングした。翌日、上述のように、600ngのプラスミドを細胞にトランスフェクトした。コトランスフェクト細胞には、150ngのRD−CFPコンストラクトと450ngのRD−YFPコンストラクトの組み合わせを与えた。15時間後に、37℃で3分間、0.05%トリプシンを使って、細胞を回収(harvest)し、細胞の一部を96穴プレートに4つ一組にして再プレーティングするか、顕微鏡法による撮像のためにibidi μ−スライド(ibidi GmbH、ドイツ)に再プレーティングした。次に細胞をさらに48時間培養してから、4%パラホルムアルデヒドで固定し、解析した。
12穴プレートにおいてHEK293細胞にRD(ΔK)−YFPかRD(LM)−HAのどちらか一方をトランスフェクトした。15時間後に、細胞をibidi μ−スライド上に一緒に再プレーティングし、さらに48時間培養した。次に、それらを固定し、顕微鏡法による解析のために、抗HA抗体とX−34で染色した。
12穴プレート中の2つのHEK293細胞集団に、300ngのRD(LM)−HAと300ngのRD(ΔK)−CFPとを一緒にコトランスフェクトするか、またはRDΔ(K)−YFPをトランスフェクトした。15時間後に、等パーセンテージのそれら2つの集団を、96穴プレートフォーマットで48時間、共培養した。次に、4%パラホルムアルデヒドで細胞を固定し、蛍光プレートリーダー(FPR)を使ってFRET解析を行なった。FRET顕微鏡解析の場合は、12穴プレート中の2つのHEK293細胞集団に、600ngのRD(LM)−CFPまたはRD(LM)−YFPをトランスフェクトした。15時間後に、等パーセンテージのそれら2つの集団を、ibidi μ−スライド上で48時間、共培養した。次に、4%パラホルムアルデヒドで細胞を固定し、FRETアクセプターフォトブリーチングを行なった。
12穴プレートにおいてHEK293細胞に600ngのさまざまな形態の非蛍光RD−HAをトランスフェクトし、24時間培養した。第2の細胞群にはCFPまたはRD(ΔK)−CFPをトランスフェクトした。次に、等パーセンテージの第1集団と第2集団を48時間、共培養した。この時点で、この集団の50%を、96穴プレートに、RD(ΔK)−YFPをトランスフェクトした細胞の集団と共にプレーティングし、48時間培養した。次に、FPRを使ったFRET解析のために、4%パラホルムアルデヒドで細胞を固定した。
12穴プレート中でHEK293細胞に600ngのRD(LM)−HAをトランスフェクトするか、または150ngのRD(ΔK)−CFPコンストラクトと450ngのRD(ΔK)−YFPコンストラクトの組み合わせをコトランスフェクトした。15時間後に、37℃で3分間、0.05%トリプシンを使って、細胞を回収(harvest)した。RD(ΔK)−YFP/CFPを発現する細胞とRD(LM)−HAを発現する細胞を同数ずつ、さまざまな量の細胞培養培地中で、48時間、共培養した。次に、4%パラホルムアルデヒドで細胞を固定し、FRET解析を行なった。条件培地実験では、トランスフェクションの15時間後に、トランスフェクション複合体を含有するRD(LM)−HA細胞からの培地を新鮮な培地で置き換えた。37℃で3分間、0.05%トリプシンを使って、RD(ΔK)−YFP/CFPを発現する細胞を回収(harvest)し、96穴プレートに再プレーティングした。24時間後に、RD(LM)−HAをトランスフェクトした細胞から条件培地を収集し、RD(ΔK)−YFP/CFPを発現する細胞に加えた。48時間後に、4%パラホルムアルデヒドで細胞を固定し、FRET解析を行なった。
前述のようにコトランスフェクション実験および共培養実験用にトランスフェクトしたHEK293細胞を、FRETアクセプターフォトブリーチング顕微鏡法のために調製した。画像は全て、C−Apochromat 40× 1.2NAレンズ(Carl Zeiss Advanced Imaging Microscopy、ドイツ07740イエーナ)を使って得た。100×(CFP)。デジタル画像は、Zeiss Axiovert 200MでZeiss LSM510 Meta NLO多光子/共焦点レーザー走査型顕微鏡システムを使って取得した。撮像に使用したチャンネルは次のとおりだった:ドナーCFPは458nmアルゴンレーザーを使って刺激し、480〜520nm帯域フィルタで蛍光を収集した;アクセプターYFPは514nmアルゴンレーザーを使って刺激し、ロングパス560nmフィルタで蛍光を収集した。強度がFRET効率の概算値を反映する画像を作成するために、初期CFP画像の値をフォトブリーチング後に得られる最終CFP画像からピクセルごとに差し引き、この相違に100を掛けて、最終CFP画像強度で割った:100×(CFP最終−CFP初期)/CFP最終。部分アクセプターフォトブリーチングについて適正な調整を行なった。画像算術変換およびグレースケールからカラーへの画像変換は、NIH ImageJ 1.44ソフトウェアを使って行なった。
スペクトルFRET測定値(FRET/ドナー)は、既述の方法に従い、TecanM1000蛍光プレートリーダーを使って得た。ドナーとアクセプターが同じタンパク質に融合されていない場合、スペクトルFRET測定値は、細胞内で発現させるドナータンパク質とアクセプタータンパク質の相対量の注意深い制御に依存する。プレートリーダーでの値は全て、まず、モックトランスフェクト細胞に対して背景差分をとった。各ウェル中のYFPシグナル(Smpl485ex/528em FRET)を使ってRD−YFP発現レベルを見積もり、実験条件下では、RD−CFP/YFPが独立して変動しないとも仮定した。これは、見かけのFRETの変化が単にRD発現レベルの変動によるものである可能性を排除するのに役立つ。ドナー励起シグナル(435nm)によるアクセプター活性化(528nm)の総FRET測定値に対する相対的寄与は、アクセプターの「クロスオーバー活性化」分率X(ここでX=435ex/528emで測定されたRD−YFPシグナルを485ex/528emで測定されたシグナルで割ったもの)を決定することによって補正した。この「クロスオーバー活性化」は、実験中に直面するRD−YFPの発現レベルが異なっても、本質的に一定である。各試料のFRET「実測」値を435ex/528emで記録し、「ドナー」値(CFP)を435ex/485emで記録する。各ウェルの「実際の」FRET/ドナー値は、
FRETactual=(Smpl435ex/528em−X*(Smpl435ex/528em))/Smpl435ex/528em
として反映される。
RIPA不溶性タンパク質をトランスフェクトHEK293細胞から抽出し、マイカチップ(Ted Pella, Inc)上で10分間インキュベートした。次に、試料を100μlのddH2Oで2回すすぎ、室温で放置して乾燥させた。翌日、MFP−3D原子間力顕微鏡(Asylum Research)を使って原子間力顕微鏡観察を行なった。
前述のように共培養実験用にトランスフェクトされたHEK293細胞を、免疫蛍光およびX−34染色用に調製した。4%パラホルムアミド中、室温で15分間、固定した後、細胞を2回、室温(RT)のPBSで5分間洗浄し、PBS中の0.25%Triton X−100中、室温で10分間、透過処理した。PBS中に1%正常ヤギ血清、20mg/ml BSA、0.25%Triton X−100を含有するブロッキング溶液を使って、細胞を、室温で3時間、ブロッキングした。HAに対する一次マウスモノクローナル抗体(Covance、カリフォルニア州エメリービル)をブロッキング溶液に1:2000希釈し、4℃で細胞に終夜、適用した。次に、0.1%Triton X−100を含有するPBSで細胞を5分ずつ3回洗浄し、ブロッキング溶液に1:400希釈した抗マウスAlexa546コンジュゲート二次抗体(Invitrogen)と共にインキュベートした。次に、0.1%Triton X−100を含有するPBSで、5分ずつ3回、細胞を洗浄し、40%エタノール、60%PBS、および20mM NaOHの溶液中に調製した1μM X−34に、室温で10分間ばく露した。次に、40%EtOH、60%PBS中で、2分ずつ3回、細胞を洗浄し、1×PBSで5分ずつ2回すすいだ。共焦点顕微鏡法(405共焦点顕微鏡−Zeiss)を使って画像をキャプチャした。HJ9.3抗体による伝播阻止の機序を特徴づけるために、HEK293細胞にRD(ΔK)−YFPをトランスフェクトするか、モックトランスフェクションを行なった。RD(ΔK)−YFP細胞またはモックトランスフェクト細胞をHJ9.3の存在下で48時間培養してから、細胞を4%PFAで固定し、0.25%Triton X−100で透過処理し、次にヤギ抗マウスAlexa546標識二次抗体にばく露した。共焦点顕微鏡法(共焦点顕微鏡−Zeiss)を使って画像をキャプチャした。
HEK293細胞を96穴プレートに75,000細胞/ウェルでプレーティングした。翌日、細胞に、100ngのさまざまな形態の非蛍光性RD−HAプラスミドを、4つ一組にしてトランスフェクトするか、DNAなしのトランスフェクション複合体にばく露した。翌日、トランスフェクション複合体を含有する培地を取り除き、新鮮な培地で置き換えた。細胞死の陽性対照として、非トランスフェクト細胞を、37℃で30分間、さまざまな濃度のスタウロスポリン(1、2、4、20μM)で処理した。次にスタウロスポリン溶液を取り除き、全ての細胞を、37℃で10分間、5μg/mlのヨウ化プロピジウムにばく露した。次に、ヨウ化プロピジウム溶液をフェノール非含有培地で置き換え、励起波長535nmおよび放射波長617nmのプレートリーダーで蛍光を読み取った。
トランスフェクト細胞集団を、それだけで、またはマウスモノクローナル抗体HJ9.3(2.5ng/μlの抗体に相当する1:1000)もしくはプールマウスIgG抗体の存在下で、3時間、6時間、9時間、12時間、24時間または48時間、共培養した。条件培地を収集し、プロテインG−アガロースビーズ(Pierceの50%スラリービーズ100μl)を培地に加え、回転させながら4℃で終夜インキュベートした。18時間後に、500μlの結合緩衝液(Pierce)を試料に加え、2000×gで3分間遠心分離した。上清を捨て、この洗浄ステップを3回繰り返した。次に、ビーズに結合したタンパク質を、高塩濃度溶出緩衝液(50μl)を使って、室温で5分間インキュベートすることで溶出させた。次に試料を2000×gで3分間遠心分離し、上清を収集した。この溶出ステップを1回繰り返し、溶出液を合計100μlとした。最初にHJ9.3にもIgGにも暴露していない条件培地のもう一つの試料を、HJ9.3(1:1000)またはIgG抗体と共に、回転させながら4℃で終夜インキュベートした後、上述と同じ免疫沈降プロトコールに付した。全ての条件からの試料を、4−20%ポリアクリルアミドゲル(BioRad)で解析し、TBS/Tween中の5%粉乳に1:2000希釈したタウRDに対するウサギポリクローナル抗体(ab64193、Abcam、マサチューセッツ州ケンブリッジ)で検出した。化学発光に基づくペルオキシダーゼコンジュゲート二次抗体反応を行い、X線フィルムで検出した。
HEK293細胞を10cmプレートで〜80%コンフルエントまで平板培養した。次に、細胞に24μgのRD(LM)−YFPコンストラクトをトランスフェクトするか、mCherryレンチウイルスで形質導入した。翌日、37℃で3分間、0.05%トリプシンを使って処理することにより、細胞を回収(harvest)し、ペレット化し、新鮮培地に再懸濁した。2つの細胞集団を、それらだけで、または1:1000希釈もしくは1:10,000希釈のタウ−RDに対するマウスモノクローナル抗体HJ9.3(1:1000は2.5ng/μlの抗体に相当する)の存在下で、48時間、共培養した。その後、細胞を回収(harvest)し、1%FBSおよび1mMのEDTAを含有するハンクス平衡培地に再懸濁した。サイトメトリーの直前に事前混合した細胞を陰性対照として使用した。MoFlo高速セルソーター(Beckman Coulter)を使って細胞をカウントし、二重陽性細胞のパーセンテージを各条件について解析した。各条件につき3つの生物学的複製を用意し、各実験条件で50,000細胞を解析した。
この研究ではP301SヒトT34アイソフォーム(1N4R)を発現するP301Sタウトランスジェニックマウスを使用した。これらのマウスは6ヶ月齢でタウ病態を発生させる。そこで、抗体を6ヶ月齢時に脳室注射によって左側脳室に注入し、これらの注入を12週間続けた。処置後に、免疫組織化学ならびにELISAおよび免疫ブロッティングによる生化学的解析のために、マウス脳を加工した。
タウは、タウオパチーと総称されるいくつかの神経変性疾患において細胞内凝集体を形成する微小管結合タンパク質である。これらには、アルツハイマー病(AD)、進行性核上性麻痺(progressive supranculear palsy)(PSP)、大脳皮質基底核変性症(CBD)、および前頭側頭型認知症(FTD)が含まれる。タウは、微小管に結合しその集合を促進する可溶性の高い天然解鎖(natively unfolded)タンパク質である。タウオパチーでは、適切に染色した場合に変性神経突起および細胞体内に可視化される過剰リン酸化神経原線維変化(NFT)中に、タウが蓄積する。タウ病態の量は、進行性神経機能障害およびシナプシス消失、ならびにヒトおよびトランスジェニックマウスモデルにおける機能低下と相関する。
以前、本発明者らは、タウ凝集体は培養細胞によって取り込まれるが、単量体は取り込まれないこと、そして内部移行したタウ凝集体は、レシピエント細胞において細胞内タウ凝集を誘発することを観察した(Frost et al., 2009;Nat Rev Neurosci 11, 155−159;Kfoury et al., 2012;J Biol Chem 287, 19440−19451)。8つのマウスモノクローナル抗体のHJ8系列(完全長ヒトタウに対して産生させたもの)および5つの抗体のHJ9系列(完全長マウスタウに対して産生させたもの)を、タウ凝集体を含有する脳溶解物の添加によって誘導される細胞タウ凝集を測定するKfoury et al.(2012;J Biol Chem 287, 19440−19451)に既述の適合細胞バイオセンサー系で特徴づけた。どの抗体もタウ単量体に効率よく結合し、神経原線維変化を染色するという事実にもかかわらず、シーディングの阻止に関する抗体の効果は一様でなかった。ここに提示する研究には、シーディングを阻止する能力が異なる3つの抗体を選択した。

P301S脳溶解物中に存在するシーディング活性を評価するために、本発明者らが以前に記載した細胞バイオセンサー系(Kfoury et al., 2012)を適合させた。これは、シアン蛍光タンパク質または黄色蛍光タンパク質のいずれかに融合されたΔK280変異を含有するタウのリピートドメイン(aa243〜375)(RD(ΔK)−CFP/YFP)の発現に基づく。これらの細胞への外因性凝集体の取り込みは、RD(ΔK)−CFP/YFPの細胞内凝集を誘発し、それは、蛍光プレートリーダーで記録される蛍光共鳴エネルギー移動(FRET)によって検出される。バイオセンサー細胞系に添加された12ヶ月齢P301Sマウスからの清澄化脳溶解物は、RD(ΔK)−CFP/YFPレポーターの強い凝集を誘導したことから、タウシーディング活性の存在が示された(図33A)。12ヶ月P301S脳ホモジネートマウスからのシーディング活性は、50nM(単量体換算)の組換え完全長原線維にほぼ匹敵する(非掲載データ)。
マウスコロニーにおいて、P301Sマウスが最初に細胞内タウ病態を発生させるのは5ヶ月齢からである。慢性的脳室内(ICV)投与によって3つの抗体の効力を調べるために、6ヶ月齢で各マウスの左側脳室にカテーテルを外科的に埋め込み、Alzet皮下浸透圧ミニポンプにより、3ヶ月間にわたって抗タウ抗体を持続的に注入した(図35A)。抗Aβ抗体HJ3.4およびリン酸緩衝食塩水(PBS)を陰性対照として使用した。6週間後に、各ポンプを、新鮮な抗体溶液またはPBSを充填したものと置き換えた。脳切離時に、各マウスの左側脳室におけるカテーテルの配置をクレシルバイオレット染色によって検証した(図35B)。カテーテルが正しく設置されていたマウスだけを解析に含めた。インビボで6週間後の抗体の安定性を調べるために(図35A)、動物から取り出した時に、残存するポンプ内容物を収集し、SDS−PAGEおよびクーマシーブルー染色を使って抗体を評価した。軽鎖および重鎖は無傷であって断片化しておらず、ウェスタンブロット上でタウ結合活性を保っていた(非掲載データ)。注入中のCSFおよび血清における抗タウ抗体の濃度を見積もるために、ビオチン化HJ8.5(HJ8.5B)を48時間投与した(約7.2μg/日)(図35A)。遊離HJ8.5Bの濃度はCSF中では7.3μg/ml、血清中では6.2μg/mlであったことから、CNSから末梢への抗体の有意なクリアランスが示された(表6)。ヒトタウに結合したHJ8.5BもCSFと血清の両方に検出されたが、濃度は遊離抗体より低かった(表6)。

3ヶ月間の処置後のP301Sマウスにおけるタウ病態の程度を決定するために、タウ病態に関する多重染色を行なった。脳切片をまず抗ホスホタウ抗体AT8による免疫染色によって評価した(図36)。AT8は、マウスタウとヒトタウの両方のリン酸化残基Ser202およびThr205に結合する(図36)。PBSおよびHJ3.4で処置されたマウスでは、AT8が、複数の脳領域において、特に梨状皮質、嗅内皮質、扁桃体、および海馬において、ニューロン細胞体およびニューロピルを強く染色した(図36Aおよび図36B)。HJ8.5処置はAT8染色を著しく低減し(図36C)、ニューロピルでは特にそうであった。HJ9.3およびHJ9.4もAT8染色を減少させたが、その効果はわずかに低かった(図36Dおよび36E)。梨状皮質(図37A)、嗅内皮質(図37B)、および扁桃体(図37C)におけるAT8染色の定量的解析により、全ての抗タウ抗体処置マウスにおけるホスホ−タウの著しいが一様ではない低減が証明された。HJ8.5抗体は梨状皮質、嗅内皮質、および扁桃体におけるAT8染色を著しく低減した。HJ9.3はHJ8.5と比較して効果がわずかに減少しており、HJ9.4は嗅内皮質でも扁桃体でも有意な効果があったが、梨状皮質では効果がなかった(図37)。海馬は、他の領域と比べてはるかに多様なAT8染色を主に細胞体において呈し、それゆえに処置群には対照群との対比で統計的有意差がなかった(図37D)。雄P301Sマウスは雌よりタウ病態が多いと報告されているので、両性別と処置の効果も評価した(図38)。処置の効果に加えて、雄マウスで解析された全ての脳領域においてAT8染色は有意に多かった(表7)。しかし、性別に関する調整後も、抗体の効果は依然として著しく有意であり、事実上同一であった(表8)。雄および雌における処置群と対照も別々に比較したところ、抗体HJ8.5の効果は依然としてもっとも有意であった(図38Aおよび図38B)。

タウアミロイド沈着について調べるために、チオフラビンS(ThioS)を使って脳切片を染色した(図39)。盲検評価者を使ってThioS染色を半定量的に評価し、全ての対照および抗タウ抗体処置マウスにおいて、1(染色なし)から5(最大染色)までのスコアを与えた。半定量的評価によれば、HJ8.5処置は、PBSおよびHJ3.4と比較して、ThioS染色を有意に低減した(図39Aおよび図39B)。PBS、HJ8.5、およびHJ9.3で処置されたマウス(各群からn=6)を、タウホスホ残基Ser396およびSer404を認識するPHF1モノクローナル抗体でも染色した。AT8染色とPHF1染色は有意に相関したことから(r=0.630、p=0.005)(図40A)、異なるタウエピトープに対する2つの抗ホスホタウ抗体は類似する結果を与えることが示された。
皮質における可溶性タウおよび不溶性タウのレベルを決定するために、RAB(水性緩衝液)、放射性免疫沈降アッセイ(RIPA)(界面活性剤緩衝液)、および70%ギ酸(FA)による逐次的な生化学的抽出を行うことで、最終ペレットを可溶化した。全タウは、ヒトタウとマウスタウの両方を同じKD(0.34pM)で検出する抗タウ抗体HJ8.7によるELISAで定量した。処置抗体がELISAを妨害する可能性は、解析に先立って陽性対照試料にこれらの抗体をスパイクし、妨害がないことを観察することによって排除した(非掲載データ)。図37における病理学的解析で評価した全てのマウスを解析した。RAB可溶性画分(図42A)またはRIPA可溶性画分(図42B)中の全タウレベルは全ての群間で類似していた。界面活性剤不溶性/70%FA可溶性画分は、ELISAおよびウェスタンブロットに先立ち試料を中和することによって解析した。研究された各動物を解析したところ、HJ8.5およびHJ9.3は界面活性剤不溶性タウを対照との対比で>50%減少させることがわかった(図42C)。代表的な試料(各群からn=4)は、HJ8.5およびHJ9.3で処置されたマウスにおける不溶性タウレベルの減少を、ウェスタンブロットで例証している(図40C)。HJ9.4処置群ではPBSまたはHJ3.4との対比で不溶性タウレベルに相違はなかった。平均AT8染色が図37における結果の平均値を反映している各群N=6匹のマウスにおいて、界面活性剤不溶性/70%FA可溶性画分中のヒトタウおよびマウスタウも特異的に評価した。70%FA可溶性画分にはマウスタウよりヒトタウの方が有意に多く存在し、抗体はこの画分におけるヒトタウを有意に低下させたが、マウスタウは有意に低下させなかった(図42Dおよび図42E)。これらと同じ試料において、AT8免疫反応性シグナルのレベルをELISAによって評価した。AT8シグナルは、この画分における全タウについてみられたものと同様に、抗体処置試料の方が低くかった(図42F)。
9ヶ月齢のP301Sタウトランスジェニックマウスの研究において、対照群と抗タウ抗体処置群を、さまざまな行動について比較した。自発運動活性、探索行動、または感覚運動機能の尺度に、群間の相違はなかった(図44)。P301Sマウスにおける認知機能欠損を回復する抗タウ抗体処置の能力を、条件恐怖手法でマウスの成績を評価することによって、査定した。1日目には、マウスの4つの処置群の全てが、トレーニングチャンバにおける最初の2分間に、類似するレベルのベースラインフリージングを呈した。これは、rmANOVAによって確認され、ここでは処置に関わる有意な全体的主効果または交互作用は何も明らかにならなかった(図45A)。加えて、4つの群の全てが音−ショック(T/S)条件刺激−無条件刺激(CS−US)ペアリング中に、類似するレベルのフリージングを示した(図45A)。3回のT/Sペアリングで群間にフリージングレベルの相違が全般的に欠如していることは、有意でない処置の効果と、有意でない遺伝子型×分数交互作用によって実証された。
タウオパチーの病理発生に関するモデルの一つでは、ある細胞中で生産された凝集体が細胞外間隙に漏れ出すか放出されて、隣接する細胞または接続された細胞における凝集を促進すると考えられている。脳溶解物からのタウシーディング活性を特異的に阻止する治療用抗体の選択により、以前になされた能動的または受動的タウワクチン接種の報告と比べて、それ以上ではないとしても、少なくとも同じ程度には強い、強力なインビボ応答が予測されることが観察された。細胞外タウ凝集体の存在に対して高感度な細胞バイオセンサーアッセイで実験を開始した。P301Sトランスジェニックマウスからの脳溶解物は、さらなる細胞内凝集を誘導することができるシーディング活性を含有することがわかった。一群の抗タウ抗体をスクリーニングした後、タウシーディング活性の阻止に関してさまざまな活性を持つ3つを選択した。これらの抗体を、P301Sタウオパチーマウスに、病態が始まった時点(6ヶ月)から開始して、3ヶ月にわたってICV注入した。抗体の注入により、CSFと血清の両方にかなりの濃度の抗体が存在する結果となり、CNSから末梢へと抗体が流出するという以前の報告と合致した。インビトロでもっとも強力な抗体であるHJ8.5による処置は、タウ病態を大きく低減し、ニューロピルからそれを著しく減少させた。この効果は、複数の独立した染色、不溶性タウの生化学的解析、および脳溶解物中に存在する残存タウシーディング活性の解析によって検出された。さらに、この処置は、このモデルにおいて検出される行動欠陥の一つを改善した。全ての抗体が、細胞へのタウ凝集体の取り込みを阻止し、このアッセイでは細胞外凝集体の存在下または非存在下で、どの抗体も細胞内には観察されなかった。これらの抗体の効力は、かつては細胞自律的であると考えられていた神経病態の病理発生における細胞外タウの明確な役割を暗示している。さらにまた、この研究は、細胞内病態の状況下で凝集体フラックスが起こりうることを示唆する本発明者らの先の知見を拡張するものであり、細胞外種を標的とすることによって凝集体のクリアランスを補助することができる治療法の可溶性を高めるものである。最後に、この研究は治療用抗体の設計に関して重要な意味を有し、特にシーディング活性を標的とすることで最も効果的な薬剤が得られるであろうことを示唆している。
抗体.HJ9.3およびHJ9.4マウスモノクローナル抗体は、タウノックアウトマウス(The Jackson laboratory)をマウスタウに対して免疫化することによって産生させ、HJ8.5およびHJ8.7はモノクローナル抗体は、タウノックアウトマウスをヒトタウに対して免疫化することによって産生させた。HJ9.3、HJ9.4およびHJ8.7モノクローナル抗体は、マウスタウとヒトタウをどちらも認識する。しかしHJ8.5モノクローナル抗体はヒトタウだけに結合する(エピトープは残基25〜30にある[NCBIリファレンス配列:NP_005901])。HJ9.3抗体はタウのRD領域(残基306〜320にあるエピトープ)を認識する。HJ9.4抗体はタウのN末端領域(残基7〜13にあるエピトープ)を認識する。対照抗体として、ヒトAβ配列のN末端領域(残基1〜16にあるエピトープ)を認識するHJ3.4マウスモノクローナル抗体を使用した。HJ8.5、9.3、および9.4モノクローナル抗体はIgG2bアイソタイプである。ウサギポリクローナルタウ抗体(ab64193、エピトープはリピートドメイン領域に位置する)はAbcamから購入した。マウスモノクローナルビオチン化BT−2抗体はヒトタウとマウスタウ(残基194〜198にあるエピトープ)を認識する。これはPierceから購入した。ラット抗マウスモノクローナルCD68抗体はAbD SeroTecから購入した。ビオチン化AT8抗体はThermo scientificから購入した。
10%ウシ胎児血清、100μg/mLペニシリンおよび100μg/mLストレプトマイシンを補足したダルベッコ変法イーグル培地(DMEM)中でHEK293細胞を培養した。培養物を、37℃、5%CO2の湿潤雰囲気で維持した。一過性トランスフェクションのために、HEK293細胞を、optimem培地中、250,000細胞/ウェルで、12穴プレートにプレーティングし、Lipofectamine 2000試薬と600ngの適当なDNAコンストラクト(Invitrogen)とを使用し、製造者の推奨に従って、トランスフェクトした。コトランスフェクト細胞には、150ngのRD(ΔK280)−CFPコンストラクトと450ngのRD(ΔK280)−YFPコンストラクトの組み合わせを与えた。15時間後に、37℃で3分間、0.05%トリプシンを使って、細胞を回収(harvest)し、次に96穴プレートに4つ一組にして再プレーティングし、15時間培養した。次に、全ての抗タウモノクローナル抗体(HJ8.5、9.3および9.4)またはHJ3.4抗体(モノクローナル抗Aβ抗体)と共にプレインキュベートしたP301S脳溶解物[プロテアーゼ阻害剤(Roche)およびホスファターゼ阻害剤(Roche)を含む1×TBS中に調製]を、回転させながら4℃で16時間、さまざまな濃度(0.125μg/ml、0.25μg/ml、0.5μg/ml、1μg/mlおよび2μg/ml)で加えた。異なる抗体で3ヶ月間処置したP301Sマウスにおけるシーディング活性を決定するために、全ての処置マウスのRAB可溶性画分も、さまざまな濃度で細胞に加えた。細胞をさらに24時間培養してから4%パラホルムアルデヒドで固定し、FRET解析を行なった。
3ヶ月齢タウノックアウト(KO)マウス、3ヶ月齢野生型(WT)マウス、3ヶ月齢P301Sマウスおよび9ヶ月齢PBS処置P301Sマウスの異なるRAB可溶性画分中に存在するタウ種を分離するために、既述の半変性界面活性剤アガロースゲル電気泳動(SDD−AGE)法にわずかな変更を加えて使用した。試料を、0.2%SDSを含む緩衝液G(20mMトリス、200mMグリシン)中の水平1.5%アガロースゲルで泳動した。試料を、試料緩衝液(60mMトリス−HCl(pH6.8)、0.2%SDS、5%グリセロール、および0.05%ブロモフェノールブルー(bromphenol blue))中、室温で7分間インキュベートした。電気泳動後に、レムリ(Laemmli)緩衝液(緩衝液G/0.1%SDS)中、4℃で、タンパク質をゲルからImmobilon−P PVDFシート(Millipore)に転写した。抗タウ特異的ウサギポリクローナル抗体(Abcam)を1:2000で使って、膜をブロッティングした。GE ECL Plusシステムを使ってブロットを発光させた。
患者の血漿試料中の病理学的タウ凝集体を検出するためにELISAアッセイを開発した。このアッセイで使用される抗体には、マウスモノクローナル抗タウHJ9.3およびHJ9.2が含まれる。ワンステップ抗体ビオチン標識キット(One−step Antibody Biotinylation Kit)を使ってHJ9.3をビオチン化する(HJ9.3−Bio)。このサンドイッチELISAではHJ9.3とHJ9.2を捕捉抗体として等濃度で利用する。96穴ハーフエリアプレート(Costar 3690)を、炭酸水素塩緩衝液(pH9.6)中に調製した20μg/mlのHJ9.2/HJ9.3(50μl/ウェル)でコーティングし、4℃で終夜インキュベートする。4%BSA/PBSを使ったブロッキングステップ後に、血漿試料(試料緩衝液(0.25%BSA/PBS、300nMトリス(PH7.4〜8.0)、1×プロテアーゼ阻害剤)中に1:4希釈)を3つ一組にしてウェルに適用する(50μl/ウェル)。次に、プレートを4℃で終夜インキュベートする。検出のために、0.5%BSA/PBS中に0.3μg/mlで調製したHJ9.3−Bioを、ウェルに、37℃で1.5時間加えた。TMB super slow基質を使った酵素反応による最終的検出には、0.5%BSA/PBS中に1:4,000希釈の二次ストレプトアビジン−ポリHRP40抗体(50μl/ウェルおよび室温で振とう機上、暗所で1.5時間)を使用する。本ELISAは血漿中の希少種の検出を最適化するために設計された。初期の実施形態には、凝集体のトラッピングを最適化するために、ELISAプレートの表面を抗体ペアでコーティングすることが含まれた。ただし、アッセイの感度を増加させるために、より大量の液体試料から、抗体被覆ビーズを使用することも、同様に妥当であるだろう。健常若年参加者から収集した陰性血漿を使って、アッセイのバックグラウンドシグナルを計算した。実験試料におけるタウシードの存在は、陰性血漿からのシグナルに対する誘導倍率として報告される。
一つの集団からもう一つの集団へのタウ凝集体の伝播を測定するために細胞伝播アッセイを設定した。リピートドメイン(RD)から構成されるタウのフラグメントを、CFPタグ付き型の凝集を促進するための2つの疾患関連変異(LM:P301L/V337M)を持つタグ無し型として、または1つの疾患関連変異(ΔK:ΔK280)として使用した。一つの細胞群に、RD(LM)とRD(ΔK280)−CFPをトランスフェクトし、もう一つにはRD(ΔK280)−YFPをトランスフェクトした。96穴フォーマットで4つ一組にして成長させた細胞からのFRETを蛍光プレートリーダーで記録した。FRETシグナルは、RD−CFP凝集体がRD−YFPを含有する細胞に移動するか、その逆の結果として生じる。複数の抗体を、表示したさまざまな希釈度で培地に加えた。抗体の出発濃度は約1mg/mlとした。例えば10−3希釈とは約1μg/mlの最終濃度を示す。24時間後に細胞を固定し、FRET測定値を記録した。個々の抗体に関するデータを図47に提示する。いくつかの抗体は凝集の経細胞伝播を極めて強力に防止した(例えばHJ8.2、HJ9.1)。別の抗体は、それよりは中間的に有効であり(例えばHJ9.3)、いくつかは本質的に有効でなかった(HJ8.7)。各グラフにおいて、最初のバーは、伝播のベースライン効率に相当する抗体が添加されていない培地を表す。
