Claims (8)
- 伸縮可能な細胞培養用基材表面に0〜80℃の温度範囲内で水和力が変化する温度応答性ポリマーが被覆された、該基材表面上で腱細胞を培養して細胞シートを形成させ、ここで、
前記培養の期間中、当該基材を延伸、収縮、或いはそれらを繰り返す、そして
前記温度応答性ポリマー層の性質の変化を利用することで前記基材表面から細胞シートを剥離する、
ことを含む、腱細胞シートの製造方法。 - 前記基材の延伸、収縮、或いはそれらを繰り返すことが、前記細胞が基材表面に付着した直後から行われる、請求項1記載の腱細胞シートの製造方法。
- 前記腱細胞は、成熟腱細胞、腱前駆細胞、腱組織細胞のいずれか、若しくは2種以上の混合物である、請求項1又は2に記載の腱細胞シートの製造方法。
- 前記腱細胞は、線維芽細胞、血管内皮細胞、筋芽細胞、及びそれらの幹細胞からなる群より選ばれる1種以上をさらに含む、請求項3項記載の腱細胞シートの製造方法。
- 前記腱細胞は、ヒト、サル、イヌ、ネコ、ウサギ、ラット、ヌードマウス、マウス、モルモット、ブタ、ヒツジ、チャイニーズハムスター、ウシ、マーモセット、又はアフリカミドリザルに由来する、請求項1〜4のいずれか1項記載の腱細胞シートの製造方法。
- 前記温度応答性ポリマーが、
ポリ−N−置換アクリルアミド誘導体、ポリ−N−置換メタアクリルアミド誘導体、ポリアクリレート誘導体、ポリメタクリレート誘導体、もしくは、
(メタ)アクリルアミド化合物、N−(若しくはN,N−ジ)アルキル置換(メタ)アクリルアミド誘導体、またはビニルエーテル誘導体からなる群より選ばれる2つ以上の共重合体
からなる、請求項1〜5のいずれか1項記載の腱細胞シートの製造方法。 - 前記温度応答性ポリマーが、ポリ−N−イソプロピルアクリルアミドである、請求項1〜6のいずれか1項記載の腱細胞シートの製造方法。
- 前記基材の材質がポリジメチルシロキサンである、請求項1〜7のいずれか1記載の腱細胞シートの製造方法。
