Description
3nmの酸化鉄ナノ粒子の合成
オレイン酸鉄錯体(iron oleate)1.8g(2mmol)とオレイン酸(oleic acid)0.57g(2mmol)、オレイルアルコール(oleyl alcohol)1.61g(6mmol)をジフェニルエーテル10gと混合した後、丸底フラスコに入れて1時間程度80℃で排気させることにより気体を除去する。その後、アルゴンを流して不活性環境を形成した後250℃まで10℃/minで昇温して反応させると、反応が進むにつれて反応物の色が黒色に変わることが確認された。250℃まで昇温してから30分間反応させると3nmのナノ粒子が製造され(図1、図2)、30分間反応させた後、迅速に冷却してから過量のアセトンで洗浄する。洗浄した後に得られた沈殿物をクロロホルム(chloroform)又はヘキサン(hexane)の有機溶媒に分散させる。
2.3nmの酸化鉄ナノ粒子の合成
2.3nmのナノ粒子は、オレイン酸(oleic acid)無しに前記オレイン酸鉄錯体(iron oleate)0.9g(1mmol)とオレイルアルコール(oleyl alcohol)3.22g(12mmol)をジフェニルエーテル10gと混合した後、実施例1と同一の条件下で熱分解して合成する。
1.8nmの酸化鉄ナノ粒子の合成
1.8nmのナノ粒子は、オレイン酸(oleic acid)無しに前記オレイン酸鉄錯体(iron oleate)0.9g(1mmol)とオレイルアルコール(oleyl alcohol)3.22g(12mmol)をジフェニルエーテル10gと混合した後、20℃/minの昇温速度で200℃まで昇温し、200℃で30分間反応させて合成する。それ以外は実施例1と同様にした。
1−オクタデセンを使用した3.3nmのナノ粒子の合成
オレイン酸鉄錯体1.8g、オレイン酸0.57g、及びオレイルアルコール1.6gを1−オクタデセン10gと混合した後、10℃/minの昇温速度で250℃まで昇温し、250℃で30分間成長させて合成する。それ以外は実施例1と同様にした。
オレイルアミン(oleyl amine)を使用した3.5nmのナノ粒子の合成
オレイン酸鉄錯体1.8g、オレイン酸0.57g、及びオレイルアミン1.6gをジフェニルエーテル10gと混合した後、10℃/minの昇温速度で250℃まで昇温し、250℃で30分間成長させて合成する。それ以外は実施例1と同様にした。
1.6nmの酸化鉄ナノ粒子の製造
公知の合成法(Nature mater、3(2004)、891)を参照して従来合成法では見つけることができなかった粒子の成長速度を制限する方法を用いることにより酸化鉄ナノ粒子を合成し、これを遠心分離して小さい酸化鉄ナノ粒子を得た。オレイン酸鉄錯体(iron oleate)の熱分解によりナノ粒子を合成する際の一般的な合成温度は320℃であり、この温度で成長が急激に進められが、300℃では十分なエネルギーが供給されないため成長速度が非常に低下する。従って、320℃にて反応を進める際に得られなかった成長段階中の極小のナノ粒子を300℃にて反応を進めながら得ることができる。
2.4nmの酸化鉄ナノ粒子の製造
実施例6と同様なナノ粒子の成長速度を制限する方法を用いて、2.4nmの酸化鉄ナノ粒子を合成した。
3.5nmの酸化鉄ナノ粒子の製造
多数の核を製造する方法を用いて公知の合成法(Nature mater、3(2004)、891)を参照し、温度を変化させることにより小さい粒径のナノ粒子の合成を試みた。オレイン酸鉄錯体(Fe−oleate)を熱分解する際に核生成温度は約270℃である。これを用いて核生成温度である270℃で長く残存することにより多量の核生成を図った。これにより、多量の核を形成した後、粒子一つ当りに付着される鉄原子の個数が減少されて大きさが小くなる。
2.3nmの酸化鉄ナノ粒子の合成
オレイン酸0.64gと1,2ヘキサデカンジオール0.59gがジフェニルエーテル15.81gに混合された溶液を70℃程度で1時間程度排気させて不純物を精製する。アルゴンを流して不活性雰囲気を形成した後、アルゴンガスを止める。鉄ペンタカルボニル(iron pentacarbonyl)0.3mlを注入し、3.3℃/minの速度で250℃まで昇温した後、250℃で30分間反応させると、2.3nmのナノ粒子が製造される。それ以外は実施例1と同様にした。
昇温速度による粒径及び分布
オレイン酸鉄錯体1.8g、オレイン酸0.57g、及びオレイルアルコール1.6gをジフェニルエーテル10gと混合した後、20℃/minの昇温速度で250℃まで昇温し、250℃で30分間成長させて2.7nmの均一なナノ粒子を合成する。それ以外は実施例1と同様にした。
製造された酸化鉄ナノ粒子の物理化学的特性
ナノ粒子の磁性を振動試料磁力計(Vibrating Sample Magnetometer;VSM)を用いて測定した。図12は実施例で合成された2.3、3、12nmのナノ粒子の磁化度−磁場(magnetization−magnetic field;M−H)の曲線を示す。図12の(a)は実施例1の方法で合成された3nmのナノ粒子の5K、300KにおけるM−H曲線であり、(b)は大きさによるナノ粒子の300KにおけるM−H曲線の変化を示し、(c)は実施例2の方法で合成された2.3nmのナノ粒子のゼロ磁場冷却(Zero field cooling)と磁場冷却(Field cooling)M−T曲線であり、(d)は実施例1の方法で合成された3nmのナノ粒子のM−T曲線であり、(e)は比較例3の方法で合成された12nmのナノ粒子のM−T曲線であり、(f)は実施例6の方法で合成された1.6nmのナノ粒子の5K、300KにおけるM−H曲線であり、(g)は実施例6の方法で合成された1.6nmのナノ粒子のM−T曲線であり、(h)は実施例9で合成された2.3nmのナノ粒子の5K、300KにおけるM−H曲線である。
Phospolipid PEG(リン脂質−PEG)を用いた酸化鉄ナノ粒子の親水化
実施例1で製造された3nmの酸化鉄ナノ粒子10mgを10mlのクロロホルムに分散させた後、Phospholipid−PEG{1,2−distearyl−sn−glycero−3−phosphoethanolamine−N[methoxy(polyethylene glycol−2000)]}10mgを追加して攪拌してからクロロホルムを徐々に蒸発させた後、水を添加して分散させた。DLS(Dynamic Light Scattering)分析結果、流体力学的径15nmの粒子を得た。
PEG−Phosphate(PO−PEG)を用いた酸化鉄ナノ粒子の親水化
POCl30.15gとpolyethylene glycol methyl ether(Mn:2000)6gを7mlのTetrahydrofuran(THF)溶液に入れた後、4時間攪拌した。THFを除去してPEG−Phosphate(PO−PEG)を得た。実施例1で製造された3nmの酸化鉄ナノ粒子10mgと100mgのPEG−Phosphate(PO−PEG)をエタノールに混合して密封し、70℃で4時間攪拌して配位子を置換する。N−Hexaneで三回洗浄してエタノールを蒸発した後、水を添加して水に分散させて流体力学的径11.7nmの粒子を得た。
親水化させた酸化鉄ナノ粒子のMR in vitro弛緩特性
実施例13、17、比較例6の方法で親水化させた酸化鉄ナノ粒子のMR造影能力を測定するために、1.6nm(実施例6)、2.4nm(実施例7)、3nm(実施例1)、4nm(実施例20)、7nm(比較例5)の酸化鉄ナノ粒子を0.5、0.25、0.13、0.063、0.031、0.016、0.0078、0.0039mg/mlの濃度でファントム(phantom)を準備した。1.5T MR映像の場合、MRスキャナ(GE Health care、signa excite)に頭部コイル(head coil)を用いて得た。T1値はIR−FSEシーケンス(sequence)を用いて得られ、使用されたパラメータは次の通りである(TR/TE/TI=4000ms/8.4ms/50〜4000ms)。T2値はCPMGシーケンス(sequence)を用いて得られ、使用されたパラメータは次のとおりである(TR/TE=5000ms/16〜200ms)。

様々な濃度のナノ粒子(0、25、100μg Fe/mL)で培養されたMCF−7細胞のin vitro T1秤量されたMR映像を1.5T MRスキャナで得た。PO−PEGでキャッピングされた3nmのナノ粒子25及び100μg Fe/mLで標識された細胞でT1信号に相当する増加が観察された反面、標識されていない細胞は明るくなかった(図15の(a))。ナノ構造化した物質はエンドソーム(endosome)で一般的にクラスタ(cluster)になるが、3nmのナノ粒子は低い容積異方性により脱イオン数及び細胞環境においてもT1造影効果を提供する。反面、細胞phantom T1秤量されたMR映像で12nmの粒子で標識された細胞は、はるかに少ない信号増加を示し、さらに、前記細胞は高濃度で暗くなった(図15(b))。12nmの酸化鉄ナノ粒子で培養された細胞の弱化したT1信号は、大きい磁性ナノ粒子の凝集体の強い磁性モメントから誘導されたsusceptibility効果によるものと考えられる。
表面をPO−PEGで処理した3nmの酸化鉄ナノ粒子のin vivo MR映像
実施例13によって製造(投入量:2.5mg Fe/kg)された、PO−PEGでキャッピングされた3nmの酸化鉄ナノ粒子の注入前後に、3T MRIスキャナ上でリストコイル(wrist coil)を使用して、ラット(rat)の動的時間分解MR血管撮影及び3d−FLASH映像を得た。前の造影映像は後の造影映像から抽出され、生成された映像はOsriX(バージョン3.8.1;32ビット;ジュネーブのOsiriX社)の最大強度の投映(MIP)プロトコルを使用して再現した。動的時間分解MR血管撮影は、1.24秒の補間臨時解像度及び次のパラメータで得た:フリップ角=20、ETL=1、TR=3.1ms、TE=1.13ms、視野角(FOV)=75×140mm2、マトリックス=256×106、スライス厚/ギャップ=2.5mm/0mm、及びNEX=1.3d−FLASHの映像パラメータは次の通りである。フリップ角=25、ETL=1、TR=25ms、TE=5.1ms、視野角(FOV)=110×65mm2、マトリックス=256×169、スライス厚/ギャップ=1.0mm/0mm、及びNEX=2。
リン酸モノサッカライドを用いた酸化鉄ナノ粒子の親水化改質及びMR in vivo映像
表面上でオレイン酸でキャッピングされた酸化鉄ナノ粒子を実施例1の方法で製造した。3nmの酸化鉄ナノ粒子100mgをTHF(テトラヒドロフラン)8ml中に分散した後、水2ml中でグルコース6−リン酸ナトリウム塩200mgの水溶液と混合した。混合した溶液を攪拌させ、60℃で4時間反応させた。冷却した後、混合溶液のTHFの上部の相が分離され、表面上でグルコース6−ホスフェートでキャッピングされた酸化鉄ナノ粒子の下部の相に水を加えて酸化鉄ナノ粒子の安定したコロイドを製造した。動的光散乱方法(Malvern Zetasizer Nano ZS)を使用して測定された、表面上でグルコース6−ホスフェートを有するナノ粒子の流体力学的径は3.8nmであった。グルコース6−ホスフェートでキャッピングされた3nmのナノ粒子を使用するMR映像を実施例16によって実行した。
クエン酸を使用する酸化鉄ナノ粒子の親水化の改質
表面上でオレイン酸でキャッピングされた酸化鉄ナノ粒子が実施例1の方法を使用して製造された。3nmの酸化鉄ナノ粒子100mgをTHF(テトラヒドロフラン)8ml中に分散させた後、水2ml中のクエン酸ナトリウム400mgの水溶液と混合した。混合した溶液を攪拌させ、60℃で4時間反応させた。冷却した後、混合溶液のTHFの上部の相が分離され、表面上でクエン酸でキャッピングされた酸化鉄ナノ粒子の下部の相に水を加えて酸化鉄ナノ粒子の安定したコロイドを製造した。動的光散乱方法(Malvern Zetasizer Nano ZS)を使用して測定された、表面上でクエン酸を有するナノ粒子の流体力学的径は10nmであった。
ベタイン(betaine)を使用する酸化鉄ナノ粒子の親水化の改質
表面上でオレイン酸でキャッピングされた酸化鉄ナノ粒子を実施例1の方法で製造した。3nmの酸化鉄ナノ粒子150mgをn−ヘキサン25ml中に分散させ、エタノール25ml中にベタイン(2−(トリメチルアザニウミル(trimethyl azaniumyl))アセテートハイドロクロライド)600mgと混合した。混合した溶液を攪拌させ、50℃で8時間反応させた。冷却した後、溶媒を3000rpmで10分間遠心分離して配位子交換されたナノ粒子を分離した。配位子交換されたナノ粒子ベタインに水を加えて酸化鉄ナノ粒子の安定したコロイドを製造した。動的光散乱方法(Malvern Zetasizer Nano ZS)を使用して測定された、表面上でベタインを有するナノ粒子の流体力学的径は7nmであった。
4nmの酸化鉄ナノ粒子の合成
実施例6の方法を参照し、粒子成長率を調節して4nmの酸化鉄ナノ粒子を合成した。より具体的に、4nmのナノ粒子を次のように合成した。オレイン酸鉄錯体(iron oleate)1.8g及びオレイン酸0.57gを1−オクタデカン10gと混合した後、10℃/minで318℃まで昇温し、30分間318℃にて反応させてから反応生成物を室温まで迅速に冷却した。その後、室温で冷却された生成物にエタノールを加えて沈殿させた後、4nmの酸化鉄を生成した。
親水化改質された酸化鉄ナノ粒子のMTT分析(assay)実験
湿潤大気37℃及び5%CO2濃度下にて、ヒト乳癌細胞株、即ち、MCF−7細胞を、10%胎児血清(FBS)及び1%ペニシリン/ストレプトマイシン(それぞれ100U/ml及び100μg/ml、Gibco社製)を含むダルベッコ(Dulbecco)の改質されたイーグル培地(DMEM、Welgene)上で成長させた。細胞内の吸収を観察するために、MCF−7細胞を8−ウェルチャンバースライド(8−well chamber slide)上で接種した後、培養された細胞を3nm及び12nmの酸化鉄ナノ粒子それぞれに混合して、PO−PEGで表面改質された。24時間後、細胞をPBSで洗浄してから4%パラホルムアルデヒドを使用して固定させた。共焦点(confocal)のレーザスキャニング顕微鏡(LSM510、Carl Zeiss、Germany)を使用して蛍光像を得た。
MALDI−TOF質量分光計
酸化鉄ナノ粒子10mg/ml及びマトリックスとして使用された9−ニトロアントラセン10mg/mlをクロロホルム中に溶解した。ナノ粒子及び9−ニトロアントラセンを1:100の相対比でブレンドした後、混合物の液滴(droplet)をLDI物質に加えた後、大気中に蒸発させた。物質はMALDI−TOF分光計(Voyager−DETM STR Biospectrometry Workstation、Applied Biosystems Inc.)に位置させ、レーザ照射させて線形モデル及びカチオン検出モデルそれぞれで500〜300,000Daの範囲でナノ粒子の質量を測定した。図19は、MALDI−TOFを使用してナノ粒子の分子量の結果を示す。図19の(a)は1.6nmの酸化鉄ナノ粒子のTEM映像であり、(b)はMALDI−TOFを介して得た1.6nmの酸化鉄の分析結果を示し、ここで、ナノ粒子は9,000Daの分子量を有し、(c)は2.4nmの酸化鉄ナノ粒子のTEM映像であり、(d)はMALDI−TOFを介して得た2.4nmの酸化鉄の分析結果を示し、ここで、ナノ粒子は65,000Daの分子量を有し、(e)は熱重量分析装置(thermogravimetric analysis;TGA)を介して1.6nmの酸化鉄ナノ粒子のコア(core)部分の質量分析結果を示し、ここでコア質量は35.8%であり、これは1.6nmの粒子のそれぞれが35.8%のコア分画を有しコアの分子量が3,330Daであることを意味する。
グルコース6−ホスフェート−エタノールアミンを使用する酸化鉄ナノ粒子の親水化の改質
(実施例23−1)
グルコース6−ホスフェート−エタノールアミンの合成
グルコース6−ホスフェートナトリウム塩1g、EDC(1−エチル−3−[3−ジメチルアミノプロピル]カルボジイミドハイドロクロライド)0.68g、及びNHS(N−ヒドロキシスクシンイミド)0.4gをMES(2−(N−モルホリノ)エタンスルホン酸)緩衝溶液10mlと混合した。混合した溶液を攪拌させて30℃で30分間反応させた。反応させた後、エタノールアミン(2−アミノエタノール)0.22mlを溶液と混合した後、30℃で12時間反応させた。MES緩衝溶液を除去した後、グルコース6−ホスフェート−エタノールアミンを得た。
グルコース6−ホスフェート−エタノールアミンを使用する酸化鉄ナノ粒子の親水化の改質
表面上でオレイン酸でキャッピングされた酸化鉄ナノ粒子を実施例1の方法を使用して製造した。3nmの酸化鉄ナノ粒子100mgをTHF(テトラヒドロフラン)10ml中に分散した後、水2mlの中でグルコース6−ホスフェート−エタノールアミン300mgの水溶液と混合した。混合した溶液を攪拌させて60℃で4時間反応させた。冷却した後、混合溶液のTHFの上部の相が分離され、表面上でグルコース6−ホスフェート−エタノールアミンでキャッピングされた酸化鉄ナノ粒子の下部の相に水を加えて酸化鉄ナノ粒子の安定したコロイドを製造した。表面上でグルコース6−ホスフェート−エタノールアミンでキャッピングされた3nmのナノ粒子を使用する流体力学で酸化鉄ナノ粒子の安定したコロイドを製造した。動的光散乱方法(Malvern Zetasizer Nano ZS)を使用して測定された、表面上でグルコース6−ホスフェート−エタノールアミンを有するナノ粒子の流体力学的径は8nmであった。
グルコース6−ホスフェート−PEGを使用する酸化鉄ナノ粒子の親水化の改質
(実施例24−1)
グルコース6−ホスフェート−PEGの合成
グルコース6−ホスフェートナトリウム塩1g、EDC(1−エチル−3−[3−ジメチルアミノプロピル]カルボジイミドハイドロクロライド)0.68g、及びNHS(N−ヒドロキシスクシンイミド)0.4gをMES(2−(N−モルホリノ)エタンスルホン酸)緩衝溶液10mlと混合した。混合した溶液を攪拌させて30℃で30分間反応させた。反応させた後、ジエチルアミン0.23mlを溶液と混合した後30℃で12時間反応させた。反応後、前記溶液を、mPEG−COOH(メトキシ−ポリエチレングリコール−カルボキシル、Mn:5000)17.75g、EDC1.361g、及びNHS0.817gをMES緩衝溶液40mlと混合して製造されたEDC活性化されたmPEG−COOH溶液と混合した。混合した溶液を攪拌させ、30℃で24時間反応させた。透析過程により副産物を除去した後、DIWを除去してグルコース6−ホスフェート−PEGを得た。
グルコース6−ホスフェート−PEGを使用する酸化鉄ナノ粒子の親水化の改質
表面上でオレイン酸でキャッピングされた酸化鉄ナノ粒子を実施例1の方法を使用して製造した。3nmの酸化鉄ナノ粒子100mgをTHF(テトラヒドロフラン)10ml中に分散させた後、水2ml中でグルコース6−ホスフェート−PEG1gの水溶液と混合した。混合した溶液を攪拌させて60℃で4時間反応させた。冷却した後、混合溶液のTHFの上部の相が分離され、表面上でグルコース6−ホスフェート−PEGでキャッピングされた酸化鉄ナノ粒子の下部の相に水を加えて酸化鉄ナノ粒子の安定したコロイドを製造した。
表面上でグルコース6−ホスフェート−PEGでキャッピングされた3nmのナノ粒子を使用する流体力学により酸化鉄ナノ粒子の安定したコロイドを製造した。動的光散乱方法(Malvern Zetasizer Nano ZS)を使用して測定された、表面上でグルコース6−ホスフェート−PEGを有するナノ粒子の流体力学的径は14nmであった。
昇温速度による粒径及び分布
オレイン酸鉄錯体1.8g、オレイン酸0.57g、及びオレイルアルコール1.6gをジフェニルエーテル10gと混合した後、3.3℃/minの昇温速度で250℃まで昇温し、250℃で30分間成長させると、ほぼ6nmのナノ粒子が混合された不均一なナノ粒子を合成する。それ以外は実施例1と同様にした。
昇温速度による粒径及び分布
オレイン酸鉄錯体1.8g、オレイン酸0.57g、及びオレイルアルコール1.6gをジフェニルエーテル10gと混合した後、5℃/minの昇温速度で250℃まで昇温し、250℃で30分間成長させると、ほぼ6nmのナノ粒子が混合された不均一なナノ粒子を合成する。それ以外は実施例1と同様にした。
12nmのナノ粒子の合成
12nmのナノ粒子は、「J.Park et al.Nat.Mater 2004、4、891」に開示された方法と同様に合成する。オレイン酸鉄錯体1.8gとオレイン酸0.28gを1−オクタデセン(octadecene)10gと混合した後、3.3℃/minの昇温速度で318℃まで昇温し、318℃で30分間成長させて合成した。図20は合成されたナノ粒子を観察したTEM写真である。
ベヘン酸(Behenic acid)を用いた酸化鉄ナノ粒子の合成
バルキー(bulky)な界面活性剤を使用し、立体障害(steric hindrance)によりナノ粒子が大きく成長することを妨害して小さい粒径のナノ粒子を製造した。オレイン酸(oleic acid)より立体障害が大きいベヘン酸(behenic acid)を合成時に添加してナノ粒子が大きく成長することを妨害した。
7nmの酸化鉄ナノ粒子の合成
7nmのナノ粒子は、オレイン酸鉄錯体1.8gとオレイン酸0.57gを1−オクタデセン10gと混合した後、5℃/minの昇温速度で318℃まで昇温し、318℃で30分間成長させて合成した。図22は合成されたナノ粒子を観察したTEM写真である。
4nmの酸化鉄ナノ粒子が凝集されている親水化カプセルの製造
4nmの酸化鉄ナノ粒子(40mg)とPLGA(poly(lactic−co−glycolic acid))40mgをエチルアセテート溶液に分散させてからPluronicRF127(BASF corporation、difunctional block copolymer)溶液4mlと混合した後、攪拌してカプセル化した。TEM観察結果(図8)、多数のナノ粒子が凝集された形態でカプセルに囲まれていることが分かった。DLS(Dynamic Light Scattering、Maker:Malven)で測定した流体力学的径(z−average)は117nmであった(図23)。
1,2−ヘキサデカンジオール(1,2−hexadecanediol)を用いた合成
オレイン酸鉄錯体1.8g、オレイン酸0.57g、及び1,2−ヘキサデカンジオール1.55gをジフェニルエーテル10gと混合した後、10℃/minの昇温速度で250℃まで昇温し、250℃で30分間成長させて合成する。それ以外は実施例1と同様にした。TEM観察結果、若干不均一な6nmのナノ粒子が合成される。オレイルアルコールの代わりに二つのヒドロキシル(hydroxyl)基を有する1,2−ヘキサデカンジオールで合成すると、低い温で熱分解されるが、4nmの小さい大きさは得ることができなかった(図24)。
Claims (16)
- 鉄を中心原子とし、4〜25の炭素原子(C4〜C25)を含むカルボキシレート基(carboxylate)が前記中心原子に配位子として結合されている鉄錯体を、C4〜C25を含む脂肪酸及びC4〜C25を含む脂肪族アルコール又はC4〜C25を含む脂肪族アミンと200℃〜310℃にて反応させて酸化鉄ナノ粒子を製造する段階を含み、酸化鉄の大きさが、前記脂肪酸及び前記脂肪族アルコール又は前記脂肪族アミンのモル比を調節することにより調節され、前記酸化鉄ナノ粒子を製造する段階が、室温から200〜310℃まで10℃/min以上の昇温速度で加温することにより行われる、酸化鉄ナノ粒子の製造方法。
- 製造された酸化鉄ナノ粒子を冷却し、洗浄して得られた沈殿物を有機溶媒に分散させる段階をさらに含む請求項1に記載の酸化鉄ナノ粒子の製造方法。
- 酸化鉄ナノ粒子の粒径は4nm以下であることを特徴とする請求項1又は2に記載の酸化鉄ナノ粒子の製造方法。
- 鉄錯体はC10〜C22を含む脂肪酸基が配位子として結合されたことを特徴とする請求項1〜3のいずれかに記載の酸化鉄ナノ粒子の製造方法。
- 鉄錯体はオレイン酸鉄錯体(iron oleate)であることを特徴とする請求項4に記載の酸化鉄ナノ粒子の製造方法。
- 脂肪酸及び脂肪族アルコール又は脂肪族アミンは、それぞれ10〜22の炭素原子(C10〜C22)を含むことを特徴とする請求項1〜5のいずれかに記載の酸化鉄ナノ粒子の製造方法。
- 脂肪酸及び脂肪族アルコールは、それぞれオレイン酸(oleic acid)及びオレイルアルコール(oleyl alcohol)であり、脂肪族アミンはオレイルアミン(oleyl amine)であることを特徴とする請求項6に記載の酸化鉄ナノ粒子の製造方法。
- 3.5nm以下の平均粒径を有し、20K以上の温度で常磁性又は擬似常磁性を有し、前記平均粒径±1nmの範囲内の均一な粒径を有する、磁気共鳴画像(MRI)T1造影剤に用いるための酸化鉄ナノ粒子。
- 請求項8に記載の酸化鉄ナノ粒子を含む磁気共鳴画像(MRI)T1造影剤。
- 酸化鉄ナノ粒子の表面は、リン脂質ポリエチレングリコール(phospolipid polyethyleneglycol)(phospholipid−PEG)、ポリエチレングリコール−ホスフェート(PO−PEG)、モノサッカライドホスフェート若しくはそれらの誘導体、クエン酸又はベタイン(betaine)で親水性に改質されることを特徴とする請求項9に記載の造影剤。
- 酸化鉄ナノ粒子の表面は、グルコース6−ホスフェート、グルコース6−ホスフェート−エタノールアミン又はグルコース6−ホスフェート−ポリエチレングリコールで改質されることを特徴とする請求項9に記載の造影剤。
- a)20K以上の温度で常磁性あるいは擬似常磁性を有し、
b)粒径は1nm〜4nmであり、
c)表面は親水性物質で囲まれており、
d)粒子同士の凝集が生じないこと
を特徴とする請求項8に記載の酸化鉄ナノ粒子。 - 親水性物質は、リン脂質ポリエチレングリコール(phospolipid polyethyleneglycol)、ポリエチレングリコール−ホスフェート(PO−PEG)、グルコース6−ホスフェート、グルコース6−ホスフェート−エタノールアミン、グルコース6−ホスフェート−PEG、クエン酸又はベタインである請求項12に記載の酸化鉄ナノ粒子。
- 請求項12又は13に記載の酸化鉄ナノ粒子が水に分散されている酸化鉄ナノ粒子コロイド。
- 請求項12又は13に記載の酸化鉄ナノ粒子を含む磁気共鳴画像T1造影剤。
- 請求項14に記載の酸化鉄ナノ粒子コロイドを含む磁気共鳴画像T1造影剤。
