<1>細胞培養構造体
まず、本発明のスフェロイドを製造するために用いることができる細胞培養構造体について説明する。
このように、凹凸構造の幅を3μm以下にすることで、実施例5に記載のように、平板効率が同素材からなる平滑の細胞培養基材と比して高い細胞培養構造体が得られる。この理由としては、実施例10に記載のように、凹凸構造の幅が小さくなる程、播種された細胞は多くの仮足を成長させるようになり、細胞培養構造体への接着性が向上するためであると考えられる。ここで仮足とは、細胞質の一時的な突出で、細胞の接着や遊走に際して形成されるものである。仮足は、ガンの浸潤・転移にも関与している。運動時には、細胞膜付近でアクチンの重合による繊維ができ、この繊維がネットワークを形成することで仮足ができる。
上述したように製造された細胞培養構造体は、細胞を培養するための容器であって、容器内に凹凸の幅が3μm以下の凹凸構造が形成されている細胞培養容器210に用いることもできる。
次に、上述した細胞培養構造体上又は細胞培養容器上で培養した本発明のスフェロイドについて説明する。
Claims (3)
- セル間の幅が3μm以下で平面方向の形状が正三角形、正方形、正六角形のいずれかであると共に最小内径が3μm以下のセルを等方的に連続して形成された細胞接着面として機能する凹凸構造であって、ナノインプリント技術によって精密に制御して形成された前記凹凸構造を有するスフェロイドを製造するために用いることができる細胞培養構造体上で細胞を培養して得られたことを特徴とするスフェロイド群。
- セル間の幅が3μm以下で平面方向の形状が正三角形、正方形、正六角形のいずれかであると共に最小内径が3μm以下のセルを等方的に連続して形成された細胞接着面として機能する凹凸構造であって、ナノインプリント技術によって精密に制御して形成された前記凹凸構造を有するスフェロイドを製造するために用いることができる細胞培養構造体上で細胞を培養することを特徴とするスフェロイド群の製造方法。
- 請求項1記載のスフェロイド群の、薬剤のスクリーニングのための使用。
