37℃、100mM トリス−HCl中、pH=7.5(アッセイ緩衝液)でのAMC(7−アミノ−4−メチルクマリン)の遊離により、DPP−IV酵素とH−Gly−Pro−AMC基体の反応を記録した。AMCの標準曲線は、濃度31.25μMまで線形であり、これが、形成されるAMCの相対的な蛍光ユニット(RFU)を使用した理由である。360nm励起および465発光フィルタを使用して(積分時間30μs、ゲイン25、フラッシュ数50)、テカンスペクトロフルオールプラス(Tecan Spectrofluor Plus)プレートリーダにより、それを検出した。これらの条件の下で、酵素反応は少なくとも30分間線形であり、酵素依存関係は蛋白質2.5μgまで(700RFUまで)線形であった。抽出された蛋白質の1〜0.8μgを使用して、H−Gly−Pro−AMCのKmは50μMであった。500μMより高い基体濃度は、蛍光発光の検出問題を引き起こし(内部フィルタ効果)、これは、サンプルの希釈により解決することができた。活性阻害剤をできるだけ効率的に検出するように、37℃での60分プレインキュベーション時間を使用して、アッセイを設計した。酵素溶液10μl中の蛋白質エキストラクト0.8〜1μgを(アッセイ緩衝液:100mM トリス−HCl、pH=7.5を使用して)、10μl体積のテスト化合物、および55μlのアッセイ緩衝液(対照の場合、65μlのアッセイ緩衝液)を含むウェルに加えることにより、アッセイを実施した。プレインキュベーション期間の後、25μl、1mMのH−Gly−Pro−AMC基体溶液を添加することにより(終濃度250μM)、反応を始めた。最終試験体積は100μlであり、試験溶液はテスト化合物溶液由来のDMSO1%を含んでいた。反応時間は37℃で30分であり、300μl、1MのNa−酢酸緩衝液、pH=4.2を加えることにより反応を停止させた。形成されるAMCの蛍光(RFU)を、テカンスペクトロフルオールプラスプレートリーダで360nm励起および465発光フィルタを使用して(積分時間30μs、ゲイン25、フラッシュ数50)検出した。対照のRFUとブランクのRFUを使用して、阻害%を算出した。

14.7gのtert−ブチル8−アザビシクロ[3.2.1]オクト−3−イル−エキソ−カルバメート(65mmol)(ジャーナルオブメディカルケミストリ、1991、34、656)、8.93gの2−クロロピリミジン(78mmol)、および、12.7mlの1,8−ジアザビシクロ[5.4.0]ウンデカ−7−オン(DBU)(85mmol)を、1−ペンタノール230mlに溶解し、還流下で4時間加熱した。溶剤を蒸発させ、残渣を250mlのクロロホルム中に溶解し、2×300mlの水で洗浄し、Na2SO4上で乾燥し、溶離液としてn−ヘキサン−酢酸エチル−クロロホルム(1:1:1)を使用して、カラムクロマトグラフィにより精製して白色結晶を得て、それをn−ヘキサンで粉末にした。収率:13.25g(67%)。M.p.:113〜115℃。1H−NMR(400MHz、CDCl3):δ 1.34(s、9H)、1.49(t、2H)、1.66〜1.97(m、6H)、3.89(br、1H)、4.61(d、2H)、6.60(t+br、1+1H)、8.34(d、2H)。
5.7g(22.7mmol)のtert−ブチル(2S)−2−(アミノカルボニル)−4,4−ジフルオロ−2−ピロリジンカルボン酸(テトラヘロンレター、1998、39、1169)を、57mlのジクロロメタン中に溶解し、この溶液にトリエチルアミン3.8ml(27.2mmol)を加えた。得られた混合物に、−15℃でピバロイル塩化物3ml(25mmol)を滴加し、混合物をその温度で1時間撹拌し、次いで、25%のアンモニア水溶液7mlを滴加し、混合物を1時間撹拌した。反応混合物を、水、1N NaOH溶液、次いで水で洗浄し、硫酸ナトリウム上で乾燥し、蒸発させた。ジエチルエーテルを添加すると、上記生成物3.94g(69%)が結晶化した。M.p.:136〜138℃。1H−NMR(400MHz、CDCl3):δ 1.48(s、9H);2.3〜2.9(m、3−CH2)、3.69(br、副)+3.86(m、主)(5−CH2)、4.53(br、2−CH)、6.0(br、主)+6.81(br、副)(NH2)。
3.93g(15.7mmol)のtert−ブチル(2S)−2−(アミノカルボニル)−4,4−ジフルオロ−1−ピロリジンカルボキシレートを、塩化水素25%のエタノール溶液75ml中に溶解し、室温で4時間撹拌した。得られた懸濁液にジエチルエーテル150mlを加え、得られた白色結晶物質をろ過分離した。上記生成物2.55g(87%)が得られた。M.p.:232〜233℃。1H−NMR(400MHz、DMSO−d6):δ 2.43〜2.51(m、副)および2.81〜3.05(m、主)(3−CH2)、3.71(t、2H、5−CH2)、4.46(t、1H、2−CH)、7.81(s、1H、)+8.12(s、1H)(NH2)、10.12(br、2H、NH2 +)。
2.54g(13.6mmol)の(2S)−4,4−ジフルオロ−2−ピロリジンカルボキサミドヒドロクロリドを、25mlのジクロロメタン中に懸濁し、この懸濁液にトリエチルアミン4.1ml(29.3mmol)を加えた。得られた混合物に、−10℃以下でジクロロメタン20ml中のクロロアセチル塩化物1.2ml(15mmol)を滴加した。1時間撹拌した後、懸濁液を酢酸エチル450mlに注ぎ、沈殿したトリエチルアミンヒドロクロリドをろ過分離し、ろ液を蒸発させ、クロロホルム−メタノールの4:1混合物の溶離液を使用して、クロマトグラフィにより精製した。上記生成物3.0g(97%)が、無色の油の形態で得られた。

10.4g(46mmol)の(2S)−1−(2−クロロアセチル)−4,4−ジフルオロ−2−ピロリジンカルボキサミドを、230mlのジクロロメタン中に溶解し、それに対してリンオキシ塩化物13ml(140mmol)を加えた。混合物を24時間加熱した(出発原料が残留している場合は、それをさらに還流する)。還流の間に、溶液が薄い黄色になり、粘着性の固体物質が沈殿した。溶液を別のポットに注ぎ、それに対して炭酸カリウム50gを加えた。1時間撹拌した後、固体の塩をろ過し、溶液を蒸発させた。薄い黄色の油が得られ、それをn−ヘキサンで粉末にした。得られた黄色の結晶を集め、ジエチルエーテル70mlを加えた。このようにして不純物が溶解して、純粋な白い固体結晶質生成物が得られた。収率:6.0g(56%)。M.p.:86〜87℃。

6.13gの8−(2−ピリミジニル)−8−アザビシクロ[3.2.1]オクト−3−イル−エキソ−アミン(30mmol)、5.74gの(2S)−1−(2−クロロアセチル)−4,4−ジフルオロ−2−ピロリジンカルボ−ニトリル(27.5mmol)、および、12.5mlのトリエチルアミン(90mmol)を250mlの乾燥アセトニトリル中に溶解し、70℃で3時間、次いで室温で一晩撹拌した。次いで混合物を蒸発して、褐色がかった濃い油が得られ、それを、溶離液としてクロロホルム−メタノール(6:1)を使用して、カラムクロマトグラフィにより精製して、固体生成物を得て、それを純粋なエタノールにより結晶化させた。収率:5.7g(77%)。M.p.:162〜163℃。

63mgの1−(2−ピラジニル)−4−アミノ−ピペリジン(0.32mmol)、62mgの(2S)−1−(2−クロロアセチル)−4,4−ジフルオロ−2−ピロリジンカルボニトリル(0.32mmol)、および、ポリマーに結合した285mgの2−tert−ブチルイミノ−2−ジエチルアミノ−1,3−ジメチル−ペルヒドロ−1,3,2−ジアザホスホリン(PBEMP)(0.73mmol)を、乾燥アセトニトリル20ml中に溶解し、55℃で8時間撹拌した。樹脂をろ過により除去し、ろ液を減圧により濃縮し、溶離液としてクロロホルム−メタノール(9:1)を使用して、カラムクロマトグラフィにより残渣を精製して、油を得て、それをジエチルエ−テル中の塩酸で処理し、標記化合物83mg(60%)が白色結晶として得られた。M.p.:158〜160℃。1H−NMR(400MHz、DMSO−d6):δ 1.54(m、2H)、2.15(m、2H)、2.80〜2.95(m、4H)、4.20〜4.25(m、4H)、4.55(d、2H、)、5.20(t、1H)、7.00(d、1H)、7.87(dd、1H)、8.50(d、1H);9.38(br、2H)。
