Description
R1は、C6−10アリール、C5−10ヘテロアリールまたはC5−10複素環を表し;前記アリール、ヘテロアリールおよび複素環は、1〜3個のRa基で置換されていても良く;
Raは、C1−6アルキル、ハロゲン、OH、アリール(C1−6)アルキル、(C1−6)アルコキシ、(C1−6)アルコキシ(C1−6)アルキル、ハロ(C1−6)アルキル、ニトロ、アミノ、モノ−もしくはジ−N−(C1−6)アルキルアミノ、アシルアミノ、アシロキシ、カルボキシ、カルボキシ塩、カルボキシエステル、カルバモイル、モノ−およびジ−N−(C1−6)アルキルカルバモイル、(C1−6)アルコキシカルボニル、アリールオキシカルボニル、ウレイド、グアニジノ、スルホニルアミノ、アミノスルホニル、(C1−6)アルキルチオ、(C1−6)アルキルスルフィニル、(C1−6)アルキルスルホニル、複素環、複素環(C1−6)アルキルを表し;
Rは、C1−8アルキル、C3−10シクロアルキル、C3−10ヘテロシクロアルキル、C5−10ヘテロアリールまたはC5−11複素環を表し;前記ヘテロアリールおよび複素環は1〜3個のRa基で置換されていても良く;前記アルキルは、アリール、複素環、(C1−6)アルキルチオ、シアノ、ヘテロアリール、グアニジノ、((1−アミノエチル)カルボニル)アミノ、((アミノメチル)カルボニル)アミノ、((2−アミノ)プロプ−2−イル)カルボニル)アミノ、アセトアミド、4−(アミノメチル)フェニル、チオ、t−ブチルスルホニル、(C2−6)アルケニルチオ、(C2−6)アルキニルチオ、アミノ、モノ−もしくはジ−(C1−6)アルキルアミノ、アリールチオ、複素環チオ、(C1−6)アルコキシ、アリール(C1−6)アルコキシ、アリール(C1−6)アルキルチオ、シクロアルキル、シクロアルケニル、カルボキシおよびそれのエステル、ヒドロキシおよびハロゲンからなる群から選択される1〜3個の基で置換されていても良い。
N−t−ブトキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−メチルブチルアミド;
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−メチルブチルアミド;
N−t−ブトキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−2−(4′−テトラヒドロピラニル)−アセトアミド;
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−2−(4′−テトラヒドロピラニル)−アセトアミド;
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−(S)−シクロプロピルブチルアミド;
ならびにこれらの製薬上許容される塩、エナンチオマー、ジアステレオマーもしくはin vivo加水分解可能エステルまたはそれらの混合物がある。

N−t−ブトキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−2−(4′−テトラヒドロピラニル)−アセトアミド;
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−2−(4′−テトラヒドロピラニル)−アセトアミド;
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−(S)−シクロプロピルブチルアミド;

表1に示した実施例3〜144の化合物は、固相で製造し、それについて下記で説明する。
下記のアッセイは、カミングスらの報告(Cummings et al., PNAS, May 14, 2002, vol.99, no.10, page 6603-6606)および2003年2月21日出願のPCT出願US03/05552(2002年2月25日出願の米国特許出願60/359707号)(これらは参照によってその全体が本明細書に組み込まれる)に開示されている。それを用いて、致死因子の阻害薬であると考えられる化合物と反応させた後に、致死因子阻害を測定した。
炭疸菌致死因子活性およびそのような活性に対する化合物の効果を測定する方法で、致死因子基質を用いることができる。そのような方法では、炭疸菌致死因子が活性であるインキュベーション培地を用いて炭疸菌致死因子とともに本明細書に記載の致死因子基質をインキュベーションし、被験化合物を存在させることができる。基質の開裂を、炭疸菌致死因子活性または致死因子活性に対する化合物の効果の評価基準として検出することができる。測定は定性的または定量的なものであることができる。本発明の化合物についての致死因子酵素結合アッセイIC50結果は15μM以下の範囲である。具体的には、N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−メチルブチルアミドおよびN−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−2−(4′−テトラヒドロピラニル)−アセトアミドについてのIC50は、それぞれ0.13μMおよび0.06μMである。
Claims (3)
- N−t−ブトキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−メチルブチルアミド;
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−メチルブチルアミド;
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−2−(4′−テトラヒドロピラニル)−アセトアミド;および
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−(S)−シクロプロピルブチルアミド;からなる群
ならびにこれらの製薬上許容される塩、エナンチオマーもしくはジアステレオマーまたはそれらの混合物から選択される化合物。 - N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−3−メチルブチルアミド;または
N−ヒドロキシ−2(R)−[(4−フルオロ−3−メチルフェニルスルホニル)]アミノ−2−(4′−テトラヒドロピラニル)−アセトアミド;
あるいはそれの製薬上許容される塩、エナンチオマーもしくはジアステレオマーまたはそれらの混合物である請求項1に記載の化合物。 - 請求項1に記載の化合物および製薬上許容される担体を含む炭疽菌から放出される致死因子(LF)の活性を阻害するための医薬組成物。
