で示される、ペリンドプリルtert−ブチルアミン塩の新しいβ結晶形、その製造方法及びこれを含む医薬組成物に関する。

ペリンドプリル及びその薬学的に許容しうる塩、そして特にそのtert−ブチルアミン塩は、有用な薬理学的性質を有する。その主な性質は、アンギオテンシンI変換酵素(即ち、キニナーゼII)を阻害することであり、そしてこれは、一方では、オクタペプチドのアンギオテンシンII(血管収縮物質)へのデカペプチドのアンギオテンシンIの変換を妨げ、他方では、不活性ペプチドへのブラジキニン(血管拡張物質)の分解を妨げる。これら2つの作用は、心血管疾患、特に動脈高血圧及び心不全におけるペリンドプリルの有用な作用に寄与する。

ペリンドプリル、その製造法及び処置におけるその使用は、ヨーロッパ特許明細書EP 0,049,658に記載されている。

この化合物の薬剤としての価値から見て、これを非常に高い純度で入手することが重要であった。また、工業規模に容易に変換できる製造法によって、特に迅速な濾過及び乾燥が可能な形で、これを合成できることが重要であった。最後に、この結晶形は、完全に再現性があり、容易に処方され、そして温度、光、湿度又は酸素レベルについて特殊な要求なく長期にわたる貯蔵が可能であるよう充分に安定である必要があった。

特許明細書EP 0,308,341は、ペリンドプリルの工業的合成法を記載している。しかしこの文書は、再現性あるようにこれらの性状を示す形でペリンドプリルを入手するための条件を特定していない。

本出願人らは今や、ペリンドプリルの特定の塩である、tert−ブチルアミン塩が、特に処方に有用な性状を示す、充分に明確で、完全に再現性ある結晶形で入手できることを見い出した。

本発明はまた、式(I)の化合物のβ結晶形の製造方法であって、
−第1の実施態様に従って、ジクロロメタン中のペリンドプリルtert−ブチルアミン塩の溶液を加熱還流し、次いで急速に0℃に冷却し、得られる固体を濾過により回収すること、又は
−第2の実施態様に従って、酢酸エチル中のペリンドプリルtert−ブチルアミン塩の溶液を加熱還流し、次いで急速に5℃に冷却し、得られる固体を濾過により回収すること、のいずれか
を特徴とする、製造方法に関する。

・本発明の結晶化プロセスでは、任意の製造法により得られる式(I)の化合物を使用することができる。有利には、特許明細書EP 0,308,341に記載された製造方法により得られる式(I)の化合物が使用される。

・本発明に従ったプロセスの第1の実施態様において、ジクロロメタン中の式(I)の化合物の濃度は、好ましくは100〜200g/lである。

・本発明に従ったプロセスの第2の実施態様において、酢酸エチル中の式(I)の化合物の濃度は、好ましくは70〜90g/lである。

本発明はまた、1つ以上の適切で不活性な非毒性賦形剤と一緒に、活性成分として式(I)の化合物のβ結晶形を含む医薬組成物に関する。本発明の医薬組成物としては、特に経口、非経口(静脈内又は皮下)又は鼻内投与に適したもの、錠剤又は糖衣錠、舌下錠、ゼラチンカプセル剤、トローチ剤、坐剤、クリーム剤、軟膏剤、皮膚用ゲル剤、注射用製剤、飲用懸濁剤などに言及することができる。

有用な用量は、障害の性質及び重篤度、投与経路並びに患者の年齢及び体重により変化させることができる。これは、1日当たり1〜500mgで変化させることができ、これを1〜数回で投与する。

特許明細書EP 0,308,341に記載される方法によって得られたペリンドプリルtert−ブチルアミン塩135gを、ジクロロメタン1100mlに溶解して還流加熱した。次に溶液を0℃に冷却し、得られた固体を濾過により回収した。
