Description

【発明の詳細な説明】 本発明は、ａ）Krameria（クラメチア）種の親水性抽
出物、およびｂ）Mesua ferreaの脂肪親和性抽出物を含
む、抗菌性活性を有する新規の薬剤および化粧用調製物
に関する。
先行技術 種々のKrameria種、好ましくはKrameria triandraの
根、樹幹樹皮および葉から塩素化溶媒、脂肪族エーテル
およびケトン、さらに脂肪族および芳香族エステルによ
って抽出し、neolignane（ネオリグナン）（とくに、Eu
pomatenoid（ユーポマテノイド）６および２−（２、４
−ジヒドロキシフェニル）−５−プロペニルベンゾフラ
ン）中で濃縮して得られる抽出物がグラム陽性およびグ
ラム陰性細菌、真菌および嫌気性菌株に対して抗菌性活
性を有することは、文献（Martindale,“The Extraphar
macopeia",28th Ed.（1982）;Cannizaro,Boll.Soc.Ita
l.Biol.Sperim.1,22,1964;V.Hoppe,Drogenkunde Bdl Wa
lter De Gruyter Ed.,1975;および英国特許第2.184.353
A号）から公知である。
これら抽出物は、プロトン性溶媒を用いての抽出によ
ってでも得ることができるが、それらの活性成分のフェ
ノールの性質から本来親水性である。
純粋成分のEupomatenoid 6および２−（２、４−ジヒ
ドロキシフェニル）−５−プロペニルベンゾフランはこ
れら抽出物から、EP 0 464 297 B1に報告されているよ
うに、シリカゲル上のクロマトグラフィーを用いて回収
することができる。
Mesua ferreaの花、とくに蕾から、ヘキサン、塩化メ
チレンなどの非プロトン性溶媒またはより好ましくは二
酸化炭素を用いて超臨界（hypercritical）条件下で、
例えば、圧力110〜260バール、とくに200バールの下
で、温度35〜65℃、好ましくは45℃で、植物性原料を抽
出することによって、置換されたクマリンおよびキサン
トンを含む脂肪親和性抽出物が得られ、これはとくにグ
ラム陽性、グラム陰性および嫌気性細菌菌株に対してKr
ameria triandraから調製される抽出物の活性に匹敵す
る活性を有する。
発明の開示 本発明は、以下を含んでなる抗菌性活性を有する配合
物に関する。
− Krameria種、好ましくはKrameria triandraの親水
性抽出物、および − Mesua ferreaの脂肪親和性抽出物。
1:10〜10:1の範囲、好ましくは1:1の比の該配合物
が、単一抽出物の抗菌性活性の相乗作用をもたらすこと
が見出されたのは予め予見できない驚きであった。
本発明の配合物はまた、食品および化粧品産業におけ
る保存料として用いることができる。したがって、本発
明はまた、保存料として以下を含んでなる食物用または
化粧用調製物に関する。
ａ）Krameria種、好ましくはKrameria triandraの親水
性抽出物、および ｂ）Mesua ferreaの脂肪親和性抽出物。
本発明はまた、食物用および化粧用調製物のための保
存料としての以下の使用に関する。
ａ）Krameria種、好ましくはKrameria triandraの親水
性抽出物、および ｂ）Mesua ferreaの脂肪親和性抽出物。
次表は、Krameria triandraおよびMesua ferreaから
それぞれ調製された二例の抽出物ならびにそれらの1:1
の配合物からなる抽出物のインビトロ抗菌性活性の平均
値を示す。
M.I.C.で表される該活性は、当業者に公知の従来の方
法と一致すると評価されているものである。
本発明は、したがって、局所的投与（ゲル、クリー
ム、ローション、乳剤および固体調製物）ができて、上
記の配合物を含む抗菌性活性を有する薬剤および化粧用
調製物に関する。該調製物は、薬剤技術においてよく知
られた従来法、例えば、“Remington's Pharmaceutical
Handbook",Mack Publishing Co.,N.Y.,USKに記述され
た方法によって適切な賦形剤、とくに抗酸化剤とともに
調製されよう。
次の実施例は、本発明をさらに詳細に説明する。
実施例Ｉ Mesua ferreaの脂肪親和性抽出物の調製 微細にすり砕いたMesua ferreaの蕾1kgを超臨界条件
下でCO2を用いて5Lの抽出器中で抽出する。最初の抽出
は34℃で90バールの圧力下で約25Lの二酸化炭素を用い
て行う。得られる抽出物を捨てて、植物材料を温度を45
℃に、圧力を220バールに上げて第二の抽出に供する。1
5.6gのオレンジ色の極めて濃厚な油が得られる。この油
をそのまま直接に使用するか、または従来の化学的分離
法によるさらなる分画に供する。
実施例II Krameria triandraの抽出物の調製 Krameria triandraの根の樹皮2kgのすり砕いた後で、
それぞれ5Lのアセトンで３回抽出処理する。合わせた抽
出物を小容量に濃縮し、その濃縮物を0.8Lのアセトン：
水（1:1）（v/v）に懸濁する。得られる懸濁物を0.5Lの
塩化メチレンで２回抽出して、塩化メチレン相を無水硫
酸ナトリウム上で乾燥し、乾固するまで濃縮する。約26
％のEupomatenoid6および14％の２−（２、４−ジヒド
ロキシフェニル）−５−プロペニルベンゾフランを含む
赤茶けた固体85gが得られる。
実施例III ヘキサンを用いるMesua ferreaの抽出物の
調製 微細にすり砕いたMesua ferreaの蕾1kgを3Lのヘキサ
ンを用いて４回還流下で抽出する。合わせたヘキサン抽
出物を活性炭（5g）を用いて脱色して、減圧下で油状に
なるまで濃縮する。14.5gの抽出物が得られるが、これ
はさらなる分画なしで使用することができる。

  1. 【請求項１】抗菌活性を有する次の成分を含んでなる薬
    剤および化粧用調製物： ａ）Krameria種、好ましくはKrameria triandraの親水
    性抽出物、および ｂ）Mesua ferreaの脂肪親和性抽出物。
  2. 【請求項２】Krameria抽出物とMesua ferrea抽出物の比
    が1:10〜10:1の範囲である請求項１に記載の薬剤調製
    物。
  3. 【請求項３】該比が1:1である請求項２に記載の薬剤調
    製物。
  4. 【請求項４】抗菌活性を有する医薬物の調製のための、
    次の成分の使用： ａ）Krameria種、好ましくはKrameria triandraの親水
    性抽出物、および ｂ）Mesua ferreaの脂肪親和性抽出物。
  5. 【請求項５】保存料として次の成分を含んでなる食物用
    および化粧用調製物： ａ）Krameria種、好ましくはKrameria triandraの親水
    性抽出物、および ｂ）Mesua ferreaの脂肪親和性抽出物。
  6. 【請求項６】食物用または化粧用調製物のための保存料
    としての、次の成分の使用： ａ）Krameria種、好ましくはKrameria triandraの親水
    性抽出物、および ｂ）Mesua ferreaの脂肪親和性抽出物。
