Abstract
Description
本出願は、2010年5月12日に出願された、米国特許仮出願シリアル番号61/3
33,831号の利益を主張するものであり、当該出願を本明細書にその全容において、
かつあらゆる目的について援用するものである。
本発明は、多能性幹細胞からのインスリン産生細胞への分化を促進させるための方法を
提供する。特に、本発明は、膵臓内胚葉系に特徴的なマーカーを発現する細胞集団であっ
て、その集団の50%超の細胞がPDX1及びNKX6.1を共発現する、細胞集団を産
生する方法に関する。
に適したインスリン産生細胞即ちβ細胞の供給源の開発に注目が集まっている。1つの手
法として、例えば、胚性幹細胞のような多能性幹細胞から機能性のβ細胞を生成すること
がある。
3つの胚葉(外胚葉、中胚葉、及び内胚葉)を含む細胞のグループを生じる。例えば、甲
状腺、胸腺、膵臓、腸、及び肝臓等の組織は、内胚葉から中間段階を経て発達する。この
プロセスにおける中間段階は、胚体内胚葉(definitive endoderm)の形成である。胚体
内胚葉細胞は、HNF-3 β、GATA4、MIXL1、CXCR4、及びSOX17
などの多くのマーカーを発現する。
-十二指腸ホメオボックス遺伝子、PDX1を発現する。PDX1が存在しない場合、膵
臓は、腹側芽及び背側芽の形成を越えて発達しない。したがって、PDX1の発現は、膵
臓器官形成において重要な工程に格付けされる。成熟した膵臓は、他の細胞型の中でも、
外分泌組織及び内分泌組織を含む。外分泌組織及び内分泌組織は、膵臓内胚葉の分化によ
って生じる。
例えば、Lumelskyら(Science 292:1389、2001年)は、マ
ウスの胚幹細胞の、膵島と同様のインスリン分泌構造への分化を報告している。Sori
aら(Diabetes 49:157,2000)は、ストレプトゾトシン誘発糖尿病
のマウスにおいて、マウスの胚性幹細胞から誘導されたインスリン分泌細胞が糖血症を正
常化することを報告している。
シチド3-キナーゼ(LY294002)の阻害剤でマウス胚性幹細胞を処理することに
より、β細胞に類似した細胞が生じたことを開示している。

腸形成の終了時に対応する期間中の、胚性幹細胞分化の4日目に培養液に添加すると、P

の特徴を獲得するプロセスである。分化細胞又は分化を誘導された細胞は、細胞系内でよ
り特殊化された(「拘束された」)状況を呈している細胞である。分化プロセスに適用し
た際の用語「拘束された」は、通常の環境下で特定の細胞型又は細胞型の小集合に分化し

多能性幹細胞は、ステージ特異的胚抗原(SSEA)3及び4、並びにTra-1-6
0及びTra-1-81と呼ばれる抗体を使用して検出可能なマーカーのうちの1つ以上

均及び標準偏差を算出した。総PDX1、NKX6.1及びCDX-2の発現細胞を、全
細胞集団中の割合として報告した。
、細胞集団のおよそ92%±4%がPDX1を発現した。しかしながら、PDBu(プロ
テインキナーゼC活性化因子)による処理は、基本培地(処理1)、又はALK5阻害剤
II+ノギン(処理2)のいずれかで処理された細胞集団と比較して、PDX1発現集団
におけるNKX6.1発現細胞の割合に顕著な増加を引き起こした(図2A)。PDBu
で処理された群では、全集団の88%±4.2%がNKX6.1を発現し、処理2を受け
た細胞の62%±8%はNKX6.1を発現し、処理1を受けた細胞の46.7±0.2
%はNKX6.1を発現した。ステージ4のNKX6.1発現細胞のほとんどは、PDX
1も発現した。これらの観察結果は、所与の細胞集団から得たPDX1発現とNKX6.
1発現との画像をオーバーレイすることによって確認された(図1A)。これらのデータ
は、プロテインキナーゼC活性化因子を追加した培地で細胞を処理することによって、膵
臓内胚葉系に特徴的なマーカーを発現する細胞集団においてPDX1及びNKX6.1を
共発現する細胞の割合が増加したことを示している。

Bで処理された細胞集団の90%が、NKX6.1を発現した。これら処理の全てにおけ
る総PDX1発現細胞に有意差は観察されなかった。図2Bを参照。
発現の上方調節を容易にし、かつ膵臓内胚葉系に特徴的なマーカーを発現する細胞集団内
におけるPDX1及びNKX6.1を共発現する細胞の割合を増加させることができるこ
とを示している。
重症複合免疫不全(SCID)-beige(Bg)マウスへの、本発明の細胞の移植

滅菌した餌と水を自由に利用できるような状態で、マウスをmicroisolator
ケージ内に収容した。外科手術を準備するため、マウスをイヤータグで同定し、体重を測
定し、手持ち式glucometer(LifeScan;OneTouch)を用いて
血糖を測定した。

購入した。細胞充填後にデバイスを医療用接着剤silicone type A(Do
w Corning,カタログ#129109)によりシールし、SICD/Bgマウス
(動物番号3及び4)の皮下に移植した。5~0 vicrylを用いて連続縫合するこ
とで筋を縫合し、皮膚を創傷クリップにより閉じた。マウスには手術後に1.0mg.k
g Metacamを皮下投与した。マウスを麻酔から覚めさせ、完全に回復させた。
は様々な間隔で3g/kgグルコースを腹腔内投与し、グルコース注入の60分後に、ヘ
パリンを少量含有している遠心管に後眼窩静脈洞から血液を吸引した。血液を遠心分離し

co Diagnostics,NH)を用いてヒトCペプチドレベルを測定した。
の両方の動物血清中でヒトCペプチドが検出され、時間経過に伴って増加した(図3A及
び図3B)。3ヵ月の終わりに、腎臓皮膜及びTheracyte社製デバイスを移植さ
れた群の両方の動物の100%において、グルコース投与に応答して顕著な量の循環ヒト
Cペプチドを検出することができた。3ヵ月後の腎臓皮膜群におけるグルコース刺激され
たヒトCペプチドの血中濃度は1.7±0.5ng/mLであり(n=4)、Thera
cyte社製デバイス移植群では1±0.5ng/mL(n=2)であった(図3C)。
X6.1共発現細胞集団が、インビボでインスリン分泌細胞に更に分化する能力を有する
ことを示している。インスリン分泌細胞への更なる分化の可能性は、局所環境に依存しな
い。本発明者らは、腎臓皮膜及び皮下部位の中の免疫保護デバイス内の両方において、P
DX1及びNKX6.1共発現細胞がインスリン分泌細胞へと更に分化し得ることを示し
ている。
本発明の細胞集団を形成するための代替方法

現する細胞の割合の増加に与える追加因子の影響を調べた。ステージ4の4日目に培養液
を2回サンプリングし、上の実施例1に記載の通りに画像解析を行った。ISL1及びN
EUROD1の発現も記録した。
X1の発現に陽性であった。PDX1発現細胞のほとんどもまたNKX6.1に共陽性で
あった。表1に示されるように、PKC活性化因子のみの追加は、膵臓内胚葉系に特徴的
なマーカーを発現する細胞集団におけるNKX6.1発現細胞の形成を促進した。ステー
ジ4の4日目までに、全集団の93%はNKX6.1陽性であり、NKX6.1発現細胞
のほとんど全てはPDX1の発現に陽性であった。
5)は、観察されたNKX6.1発現の増加に影響を与えなかった。細胞集団の57.1
%はニューロD1を発現し、細胞集団の52.4%はISL1を発現し、この処理の後に
内分泌前駆体細胞集団の割合が増加したことを示唆している。表1を参照されたい。

明らかにした。図4A~図4D参照。その一方で、NGN3の発現の顕著な増加が、AL
K5阻害剤2及びPDBuを与えられた(処理5)細胞で観察された。図4A~図4D参

の影響もまた調べた。PDBuと組み合わせた50ng/mLの濃度でのFGF10の添
加(処理6)は、細胞集団の90%がNKX6.1を発現する膵臓内胚葉系に特徴的なマ
ーカーを発現する細胞集団を生成したが、NKX6.1発現細胞の多くはCDX2陽性で
もあった。表1参照。PDX1、NKX6.1及びPTF1 αのmRNAレベルは、P
DBu及びノギンで処理された細胞で観察されるレベルよりも増加しなかった。図4A~
図4Dされたい。
P阻害剤の組み合わせを使用して、細胞の90%超の細胞がPDX1及びNKX6.1を
共発現する、膵臓内胚葉系に特徴的なマーカーを発現する細胞集団を生成することができ

を生成した。表2を参照されたい。NKX6.1発現細胞のほとんどはPDX1も発現し
た。
KC活性化因子を加えることにより、ステージ4の4日目までにNKX6.1及びPTF
1 αのmRNAレベルの上昇がもたらされた(図5A~図5D)という点で、PCR分
析はIN Cell analysisも支持した。
