Abstract
Description

Kriksら、Nature.2011年11月6日;480巻(7378号):547~51頁には、SMAD二重阻害、SHH活性化、およびWnt活性化を含むKSR培地において細胞を培養することによって、中脳DA細胞へのhESCの分化についてのプロトコルが記載されている(以下に記載されているように「bump」を含まない)。しかしながら、KSR培地は規定されておらず、この分化プロトコルを使用して生成された細胞は、移植後にin vivoで不十分な役割しか果たさない。本実施例に従って分化させた細胞は、NB/N2培地においてSMAD二重阻害(SB431542およびLDN193189による)、SHH活性化およびWnt活性化を含む培養プロトコルを利用し、この培養プロトコルでは、Wntの濃度は、培養のD4またはD5において、0.7μMのベースライン濃度から5~10μMの間まで増加させる(すなわち、Wnt「bump」)。
本実施例は、実施例1に記載されているGMP V2A分化プロトコルの改変バージョンを提供する。

7.3.20.1 プレーンNeurobasalにおける細胞の既知の希釈液を作製する(成長因子を含まない)。

7.6.1 順次、各T75フラスコを一度に一つずつ操作する。
7.6.2 既存の培地を吸引し、2mMのL-glut、250nMのLDN193189および10.8μMのSB431542、500ng/mLのSHHおよび0.7μMのCHIR99021を含有するNB/N2/B27培地40mLを穏やかに添加する。
7.7.1 順次、各T75フラスコを一度に一つずつ操作する。
7.7.2 既存の培地を吸引し、2mMのL-glut、250nMのLDN193189および10.8μMのSB431542、500ng/mLのSHHおよび7.5μMのCHIRを含有するNB/N2/B27培地40mLを穏やかに添加する。
7.9.1 順次、各T75フラスコを一度に一つずつ操作する。
7.9.2 既存の培地を吸引し、2mMのL-glut、250nMのLDN193189および10.8μMのSB431542、500ng/mLのSHHおよび7.5μMのCHIR99021を含有するNB/N2/B27培地40mLを穏やかに添加する。
7.10.1 順次、各T75フラスコを一度に一つずつ操作する。
7.10.2 既存の培地を吸引し、2mMのL-glutおよび7.5μMのCHIR99021を含有するNB/N2/B27培地40mLを穏やかに添加する。
7.12.1 7.5μMのCHIRを供給する:
7.12.1.1 順次、各T75フラスコを一度に一つずつ操作する。
7.12.1.2 既存の培地を吸引し、2mMのL-glutおよび7.5μMのCHIR99021を含有するNB/N2/B27培地40mLを穏やかに添加する。
7.12.2 POコーティング
7.12.2.1 各T75フラスコに、バッチ記録番号を番号を増番させて標識した。各タスクを順次実施する。
7.12.2.2 48×T75を、フラスコごとに、DPBS中の15μg/mLのポリ-L-オルニチン15mLでコーティングする。
注記:必要量は720mLであるが、過剰に800mLを作製する。
7.12.2.3 37℃で5%のCO2と一緒に終夜インキュベートする。
7.13.1 3μMのCHIRをプレートに供給する。
7.13.1.1 順次、各T75フラスコを一度に一つずつ操作する。
7.13.1.2 既存の培地を吸引し、2mMのL-glut、20ng/mLのBDNF、200nMのAA、20ng/mLのGDNF、500nMのcAMP、1ng/mLのTGFβ3および3μMのCHIR99021を含有するNB/B27培地40mLを穏やかに添加する。
7.13.2 F/Lコーティング
7.13.2.1 フラスコを4つのバッチで処理することができる。
7.13.2.2 吸引し、次いで、15mLのDPBSを添加する。
7.13.2.3 穏やかに揺すって洗浄し、次いで、合計で3×DPBS洗浄のために、もう2回繰り返す。
7.13.2.4 吸引し、次いで、DPBS中2μg/mLのフィブロネクチン/ラミニン15mLを添加する。
注記:必要量は720mLであるが、過剰に800mLを作製する。
7.13.2.5 37℃で5%のCO2と一緒に終夜インキュベートする。
注記:7.14は、生成の組ごとに2人の独立したオペレーターにより同時に実施することができる。2つの組は、同時にフラスコを処理し、処理時間を低減させることができる。生成物は、均質性を確保するために播種前にプールされなければならない。
注記:収量が必要量より多い場合、48個のフラスコのみを継代することができる。収量が合計1×109個の細胞未満である場合、バッチは中止される場合がある。
注記:このステップでは2つの容器を処理して、同じチューブ内に含めることができる。
注記:さらに容器を処理する必要がある場合は、細胞を4℃で保存することができる。
7.14.23.1 プレーンNeurobasal培地における細胞の既知の希釈液を作製する。
7.14.23.2 20uLの希釈細胞を20uLのAOPIと混合する。
7.14.23.3 20uLの細胞/AOPI混合物をCellometer(登録商標)スライドにロードする。
7.14.23.4 スライドをCellometer(登録商標)に挿入する。
7.14.23.5 プログラムファイル「mDA Neurons AOPI」を選択する。
7.14.23.6 以下のフォーマット「DA01 Lot#MMDDYY d11」を使用してファイル名を与える。
7.14.23.7 希釈値を入力する。
7.14.23.8 F1チャネルを使用して、機械の右側のつまみを使用して細胞に焦点を当てる。
7.14.23.9 焦点があったら、「Count」を押す。
注記:可能であれば、試料を、記録保管のために生きた試料を凍結保存するために提出すべきでもある。生きた試料は、Stem-Cellbanker(登録商標)およびQC実験室における凍結保存プロトコル「USER1」を使用して、単一の細胞として凍結することができる。
7.14.29.1 15億個の細胞を達成するために必要な体積を計算する。
7.14.29.2 計算した体積を2mMのL-glut、20ng/mLのBDNF、200nMのAA、20ng/mLのGDNF、500nMのcAMP、1ng/mLのTGFβ3および3μMのCHIR99021を含有するNB/B27培地の各1L瓶から取り除く。
7.14.29.3 15億個の細胞を含有する計算した体積を各1L瓶に添加する。
注記:細胞収量が3×109個未満であるが1×109個超である場合、細胞を1mL当たり1.5×106個の細胞の最終濃度にするために必要なmL数を計算し、可能な限り多くのフラスコに播種する。
注記:ピペッティングの前に細胞を注意深く懸濁させることを確認する。
注記:均一なコーティングを確保するために、顕微鏡でフラスコを抜き打ち検査すること。
7.15.1 制御点:フラスコのコンフルエンスを抜き打ち検査し、BRに記録する。培養物は100%コンフルエントであるべきである。指定されたフラスコのコンフルエンスに留意すること。
注記:継代の間に最大数のフラスコが達成されない場合には、成熟培地を次の日に使用してもよい。
7.15.2 順次、各T75フラスコを一度に一つずつ操作する。
7.15.3 既存の培地を吸引し、各T75フラスコに対し、2mMのL-glut、20ng/mLのBDNF、200nMのAA、20ng/mLのGDNF、500nMのcAMP、1ng/mLのTGFβ3および10μMのDAPTを含有するNB/B27培地を穏やかに添加する。
7.16.1 順次、各T75フラスコを一度に一つずつ操作する。
7.16.2 既存の培地を吸引し、各T75フラスコに対し、2mMのL-glut、20ng/mLのBDNF、200nMのAA、20ng/mLのGDNF、500nMのcAMP、1ng/mLのTGFβ3および10μMのDAPTを含有するNB/B27培地を穏やかに添加する。
7.17.1 順次、各T75フラスコを一度に一つずつ操作する。
7.17.2 既存の培地を吸引し、各T75フラスコに対し、2mMのL-glut、20ng/mLのBDNF、200nMのAA、20ng/mLのGDNF、500nMのcAMP、1ng/mLのTGFβ3および10μMのDAPTを含有するNB/B27培地を穏やかに添加する。
7.18.1 順次、各T75フラスコを一度に一つずつ操作する。
7.18.2 既存の培地を吸引し、各T75フラスコに対し、2mMのL-glut、20ng/mLのBDNF、200nMのAA、20ng/mLのGDNF、500nMのcAMP、1ng/mLのTGFβ3および10μMのDAPTを含有するNB/B27培地を穏やかに添加する。
注記:7.14は、生成の組ごとに2人の独立したオペレーターにより同時に実施することができる。2つの組は、同時にフラスコを処理し、処理時間を低減させることができる。生成物は、均質性を確保するために播種前にプールされなければならない。

図12に示すように、mDA前駆体の損傷ラットへの移植の4カ月後に、移植片を受けたラットは、偽処置ラットと比較して少ない、1分当たりの回転数を示した。移植の5カ月後には、移植した移植片の免疫細胞化学により、移植片が、mDA形態の典型的なTH染色を示し、TH陽性ニューロンは、GIRK2発現についても陽性であることが示された(図13)。
