本開示の主題は、ヒト幹細胞から誘導される4つの主要な外胚葉系統、CNS、神経堤、頭蓋プラコード、および非神経外胚葉の細胞、ならびに神経障害の細胞ベースの処置および薬物発見のためのそれらの使用に関する。
ヒトにおいて、初期発生細胞型は単離および研究するのが困難である。多能性幹細胞(PSC)の方向付けられた分化によって、基礎生物学およびトランスレーショナルバイオロジーに適用するための体系的手法において、初期運命決定にアクセスするモデル系が提供される。in vivoでの発生の間に作用することが公知である発生経路のin vitroでのモジュレーションに基づき、自然分化パラダイムおよび方向付けられた分化ストラテジーなどの初期系統へPSCを分化させるためのいくつかのストラテジーが存在する。様々な分化プラットホームにわたって帰結に大きく影響を及ぼす因子として、フィーダー細胞の使用、単層に基づくストラテジー対胚葉体に基づくストラテジー、または複合培地組成が挙げられる。例えば、多くの公開されたプロトコルには、血清または所望の運命を誘導するためのKSRなどの血清置換因子を含有する培地が関与する。これらの試薬の製造におけるバッチ間変動は、分化の再現性に影響を及ぼし、目的の特異的細胞型を生成するために骨の折れるロット試験の遂行を必須とすることが多い(Blauwkampら、2012年)。KSRなどの複雑な試薬に対するこのような広範な品質管理ストラテジーは、任意の単一のプロトコルについては実行可能であるが、これらは、モジュール形式で何十もの、またはおそらく何百もの定義された細胞型を生成することを目的とする、より意欲的なストラテジーの開発を妨げる。

1種または複数のNC、CPまたはNNE系統マーカーを発現するin vitroで分化した細胞(「幹細胞から誘導されたNC、CPまたはNNE前駆体」とも称される)を神経変性障害または下垂体障害を処置するために使用することができる。本開示の主題は、神経変性障害または下垂体障害を処置する方法であって、有効量の本開示の幹細胞から誘導された前駆体を神経変性障害または下垂体障害を患っている被験体に投与するステップを含む方法を提供する。

ヒト多能性細胞(hPSC)は、潜在的に、身体の任意の細胞型を生じることができる。長期的な目的は、in vitroで完全なヒトの系統樹を再現するためのストラテジーの開発である。このような努力は、3つの胚葉のそれぞれへのアクセスをもたらすモジュラー分化プラットホームを確立することに依拠する。ここで、本発明者らは、4つの主要な外胚葉系統のすべて(CNS、神経堤、頭蓋プラコード、非神経外胚葉)を十分に定義された条件下で並行して誘導するためのストラテジーを提示する。遺伝子レポーター株を使用して、本発明者らは、非CNS外胚葉誘導体を誘導する際のBMPシグナル伝達に対する用量および時間に依存する役割を実証する。本発明者らは、初期細胞運命の決定の機構を精査する遺伝子編集ツールを適用し、頭蓋プラコードの運命を増大させる化合物を化学的にスクリーニングする本発明者らのプラットホームの有用性をさらに実証する。オンデマンドでの4つの外胚葉系統への再現可能なアクセスは、in vitroでヒト外胚葉系統の多様性を再現する途中の最初のマイルストーンである。
4つの外胚葉系統を誘導するためのdSMADiベースの分化プロトコルは、化学的に定義したシステムを使用すると、NEの運命への偏りを生じる
4つの主要な外胚葉系統は、神経外胚葉(NE)、神経堤(NC)、頭蓋プラコード(CP)および非神経外胚葉(NNE)を含む。これらの系統のそれぞれは、図1Aにまとめたように伝統的なKSRベースのプロトコルを使用してdSMADi条件を修正することによって生成することができる。興味深いことに、KSR条件下で、パターニング因子を含むかまたは減らす最適な時点は、誘導の48時間後である(Dincerら、2013年;Micaら、2013年)。この時点で、dSMADiによる継続によって前部NEが生じ、CHIR99021によるWntシグナル伝達の活性化により頭蓋NCが生じ、BMP阻害剤であるLDN193189の除去により頭蓋プラコードが生じ、またはLDN193189と組み合わせたSU5402によるFGFシグナル伝達のブロッキングによりNNEの運命が誘発される。定義された転写因子および他の系統特異的マーカーを使用して、初期の外胚葉系統のそれぞれを一意的に特定することができる。NEの生成は、SOX1およびPAX6の発現、ならびにTFAP2Aの非存在によってマークされる。TFAP2Aの発現によって、非神経外胚葉から誘導される細胞型から外胚葉が分離される。TFAP2Aと組み合わせて、SOX10対SIX1の発現によって、それぞれ、NC対プラコードの同一性が特異的にマークされる。NNEに対する特異的転写因子が存在するかどうかは不明のままであるが、SOX10とSIX1の両方の非存在下でのTFAP2Aの発現は、これらの培養条件下でNNEを確実に特定しているようである(図1B)。
ほとんどの外胚葉系統プロトコルが、デフォルトで、PAX6陽性NEを生じた理由を理解するために、本発明者らは、転写因子AP2α(TFAP2A)の誘導について調査した。TFAP2AはNC、CPおよびNNEにおいて高度に発現され、それぞれ、NCおよびCPに対するSOX10およびSIX1などの他の系統に制限されたマーカーの発現に先行する分化の2日以内に上方調節される(Dincerら、2013年)。多くのシグナル伝達分子がレチノイドならびにWNTおよびBMPシグナル伝達の活性化剤などのTFAP2Aの発現を誘導することが報告されている(Luoら、2003年;Xieら、1998年)。興味深いことに、KSR条件下では、これらのシグナル伝達因子はいずれも、堅固なTFAP2A発現にもかかわらず、非CNSの運命の誘導の間に添加されない(Dincerら、2013年)。したがって、本発明者らは、KSRベースの培地はTFAP2Aの誘導に十分な内因性シグナルを誘発することができるが、E6はこれらの因子を欠いていると仮定した。したがって、本発明者らは、関連するシグナル伝達分子を直接添加することによってTFAP2Aの発現を回復させることを試みた。
BMPシグナル伝達は、ニワトリ胚の発生においてNNEおよびプラコードの形成に重要であることが示されている(図2A)(GrovesおよびLaBonne、2014年)。本発明者らは、BMPシグナル伝達を外部から刺激することによってTFAP2Aの発現を誘導し、PAX6+NEを抑制することを考えた。本発明者らは、用量依存的な処置の3日以内に、TFAP2Aの発現が急速に上方調節されることを観察した(図2BおよびC)。高濃度(20ng/ml)で、細胞はTFAP2A陽性となり、SOX10およびSIX1の発現を欠き、NNEが強力なBMPシグナル伝達の活性化により誘発されることを示した(図2D)。FGF経路をさらに阻害することによってCP誘導をさらにブロックし、それによって、NNE誘導の効率を増大させる。NNEを規定の条件を使用してケラチノサイトへと最終分化に供する際に、本発明者らは、未成熟(K14陽性)および成熟上皮細胞(K18陽性)の両方を達成することができた(図2E)。
本発明者らは、BMP4のショートパルス(1ng/ml)と組み合わせたWNTシグナル伝達の活性化によりほぼ均質なSOX10陽性NC集団を生成できることを観察した(図3Aおよび図11)。興味深いことに、このような低濃度のBMP4で、TFAP2Aは弱くしか誘導されず(図2F)、NCが、最初に、TFAP2Aを発現しないがまたは低レベルのTFAP2Aを発現する初期前駆細胞から生じる可能性があることを示唆する。しかし、WNTとBMPの両方を添加することは、TFAP2Aと同じく、非神経外胚葉の運命の別のマーカーであるDLX3も活性化するように相乗的に作用する(図3B)。さらに、NCの分化によって、それぞれ、Isl1およびMash1染色によりマークされる自律神経および感覚神経を生じうる(図3C)。ここで表されたデータは、BMPシグナル伝達の初期の、用量依存的誘導によって、以前に報告されたよりも高い効率および低い可変性を有するNCを含む非CNS外胚葉の運命の形成が可能となることを実証する。
本発明者らは、外胚葉に特異的なレポーター株を生成したため、4つのヒト外胚葉系統すべての転写発現特性を決定しようとした。NNE系統を除いて、本発明者らは、それぞれのレポーター株(すべての株はWA−09hESCから誘導された)を使用してGFP陽性細胞を選別し、これらの精製細胞のRNAシークエンシングを実施した。不偏クラスタリングアルゴリズムにより、NEはhESCと接近してクラスター化する一方、NNEは他のすべての外胚葉系統から最も離れてクラスター化することが示された。興味深いことに、主成分分析に基づき、NCおよびCPは互いに接近してクラスター化し、これらの細胞が類似する転写プロファイルを有するが、それぞれ、SOX10およびSIX1の発現においては異なることが示唆された(図4AおよびB)。次いで、差次的に発現した遺伝子は、共有の、および特有の発現プロファイルを有するものへとグループ化される(図4C)。次いで、このような発現パターンを遺伝子オントロジー分析(Edgarら、2013年)に供した(図4D)。細胞外マトリックス再編成に関連する遺伝子は、非CNSから誘導された細胞型のすべてにおいて非常に豊富に存在する。このことは、分化の間の初期BMPシグナルがECMを介して部分的に作用する可能性があり、またはECMに関連する転写物の豊富な細胞型を少なくとも誘導することを示す。逆に、NEに関連するオントロジーはシナプス伝達および神経系の発達に関与する。NCに特異的な個々のオントロジーは細胞接着およびカルシウム結合を誘導し、一方、CPはシナプス伝達およびイオン膜輸送に対して濃縮される。まとめると、4つの外胚葉系統に対する転写発現プロファイルは全体として異なっており、表された特異的系統のそれぞれに関連する捕捉機能である。
ここで、4つの主要な外胚葉系統を誘導するために提示された新規条件は、堅固かつ効率的である。したがって、本発明者らは、外胚葉系統の特異化の間に重要な役割を果たすものを特定するための摂動研究を実施しようとした。非CNS運命に関与する本発明者らの研究の1つの最良の候補は、dSMADiの間に、初期に高度に発現されることから、TFAP2Aであった。非CNS系統がTFAP2Aの発現に依存するか否かを対象として、本発明者らはCRISPR/Cas9系を使用してTFAP2AノックアウトhESC株を生成した。2種のガイドRNAを使用して、フレームシフトの欠失を誘導し、陽性クローンをシークエンシングして、欠失の程度および性質を決定した(図12Aおよび12B)。TFAP2Aの発現の消失を高BMPの存在下で短い3日の誘導を使用して確認した(図5Aおよび12C)。TFAP2A対野生型hESCのさらなる比較研究により、CPまたはNNE条件下で、野生型では分化の6日目にE−カドヘリンの堅固な上方調節が示されたが、TFAP2Aノックアウト細胞では示されなかった(図5B)。これらのデータにより、TFAP2Aが、NCまたはCPの運命へのEMT様移行から細胞を保護する場合のあるE−カドヘリンの発現を促進することが示唆される。
化学的に規定された系に変換することによって、最も主要な外胚葉系統に対して優れた収率を有するモジュール分化プラットホームが得られる。しかし、CPの運命の誘導は比較的低いままであった(約40%)。CP誘導の効率をさらに増強し、HTSアッセイにおける本発明者らのプラットホームの適性を実証するために、本発明者らは、Six1::H2B−GFPレポーター株に関して、Library of Pharmacologically Active Compounds(LOPAC)を使用して小分子スクリーニングを実施した(図6A)。本発明者らは、対照の分化で観察されたレベルを超えてSix1の発現を増加させた3種の有効な候補を特定した。セロトニン受容体アゴニストであるBRL−5443;パルテノライド、NF−kBおよびSTATが媒介するコンフォメーション転写を阻害する能力を有する植物ホルモン;ならびにフェナントロリン、メタロプロテアーゼ阻害剤。主要なヒットのさらなる検証に関して、フェナントロリンがSix1を発現する細胞のパーセンテージを確実に増強することを確認した(図6C、13A、13B)。
オンデマンドでhPSCから多数の特異的細胞型を誘導する目的は、適切な分化プラットホームの利用可能性に依存する。現在利用可能なプロトコルは、培地の組成および培養技術に多くの矛盾が生じやすい。ここで、本発明者らは、化学的に規定された系を使用して、4つの主要な外胚葉系統のすべてを誘導するストラテジーを提示する。in vivoでの発生の合図となる因子(cue)の適用により、分化の成功がおおいに増強され、本発明者らは、4つのシグナル伝達経路のモジュレーションが、初期の外胚葉系統の選択の十分な多様性を再現するのに十分であることを示す。CNS対非CNSの運命の線引きは、BMPによる用量依存的処置に依拠する。NC、CPおよびNNEなどの任意の特異的系統を促進する具体的なBMP濃度は非常に狭い。BMPは、NCを生成するWNTの活性化、CPを生成するFGFの活性化、およびNNEを生成するFGFの阻害と共同で作用することができるTFAP2Aを上方調節することにより、少なくとも部分的に作用する。本発明者らの結果により、最小限の培地系において、少量の小分子を使用するin vivoでの発生を模倣することによって、外胚葉細胞の運命の決定を再現することができることが実証される。この外胚葉の分化のプラットホームは、非常に堅固であり、発生経路の遺伝学的解剖および小分子スクリーニングに適している。本研究によって、TFAP2Aの損失は、NEまたはNNEの形成に影響を与えないが、NCおよびCPの運命の誘導にはおおいに影響を及ぼすことも実証された。
hPSCの培養および分化
H9 ESC(WA09)および改変レポーター株(40〜70継代)をマウスの胎仔線維芽細胞フィーダーのKnockout Serum Replacement(KSR)(Life Technologies)ベースの培地(DMEM−F12、20%のKSR、L−グルタミン酸および10ng/mlのFGF2)で培養した。KSR hPSCをビトロネクチンでコーティングしたE8培地(Life Technologies)に直接移し、分化が実施される4〜5継代前の間適応させた。KSRベースの分化を以前に記載したように実施し、神経外胚葉および神経堤(Micaら、2013年)、ならびにプラコードおよび非神経外胚葉(Dincerら、2013年)の通りに実施した。
ドナープラスミドを構築し、Infusion Cloning System(Clontech)を使用してpUC19にクローニングした。TALEN配列をTAL Effector Nucleotide Targeterソフトウェア(Cermakら、2011年;Doyleら、2012年)を使用して予測した。
総RNA(Trizol)を少なくとも二連で分化の12日目にGFP選別細胞から(NE、NCおよびプラコード)またはバルクで(NNE)単離した。6千万の読み取りデータが生じ、Tophat v1.2(Trapnellら、2012年)を使用してアライニングした。次いで、HTseq(Andersら、2015年)を使用して読み取りデータをカウントし、差次的に発現した遺伝子をDESeq(AndersおよびHuber、2010年)を使用して計算した。差次的に発現した群を、遺伝子オントロジーによる分類およびLifeMap Gene Analytics(Edgarら、2013年)を使用するシグナル伝達経路濃縮について分析した。得られたRNAシークエンシングデータセットをGEOにアップロードする。
CRISPR/Cas9系を使用してノックアウトhESC株を生成した。手短に述べると、2種のガイドRNAをCRISPR設計ツール(Congら、2013年)を使用して予測し、TOPO−Bluntベクター(Maliら、2013年)(Life Technologies)にクローニングした。Cas9−GFP(5ug)と両方のガイドRNA(それぞれ1ug)をH9 hESCへヌクレオフェクトし、ROCK阻害剤を含むKSR培地中マトリゲルをコーティングしたディッシュに再播種した。24時間後、細胞をGFRについて選別し、ROCK阻害剤を含むKSR培地中のMEFフィーダー層に播種した。次いで、コロニーを単離し、TFAP2Aの標的化領域をPCRによって増幅させ、TOPO TAベクターにクローニングし、フレームシフトの変異を特定するためにシークエンシングした。

ヒト多能性幹細胞(hPSC)は、再生医療において適用するための潜在的に制限されない細胞供給源を表す。ホルモン生成細胞は、細胞療法の適用に特に適している場合があり、下垂体機能障害の処置は有効な治療標的である場合がある。以前の研究により、in vivoでの下垂体の発生に関与する発生の相互作用を模倣する3Dオルガノイド培養を使用するマウスのESCからの下垂体系統の誘導が実証された。
十分に規定した条件下でのhPSCからの頭蓋プラコードの誘導
本発明者らは、hPSCからの腺下垂体を含む頭蓋プラコードの生成について最近報告した(Dincerら、2013年)。しかし、本発明者らの以前のプロトコルは、下垂体系統の細胞を生成するために最適化されておらず、いくつかの不明確な試薬、例えば、ノックアウト血清代替物(KRS)、マトリゲルおよびマウスの胎仔線維芽細胞(iMEF)フィーダーを含有した。これらの構成成分は、所与の分化プロトコルの堅固さに対する可変性のかなりの発生源であり、ヒトへの移行に適する細胞療法の開発を複雑化している。さらに、PSCからの下垂体前葉系統の細胞の生成効率は、マウスとヒトの細胞の両方について公開されたすべての研究において低いものであった。最後に、マウスのESCにおける3D培養の研究によって示唆された誘導プロセスの間に、視床下部細胞との直接的相互作用が必要であるか否か(Sugaら、2011年)、または制限された数の規定のシグナルが下垂体の運命を誘発するのに十分であるか否かを、最終的に評価することが重要であった。
下垂体の複雑な形態発生は、初期の胚段階の間に起こる(マウスで約E10)。腺の前葉および中葉は下垂体プラコードに対応する口側外胚葉から誘導されるが、後下垂体は神経外胚葉から発生する(図16A)。誘導のための組織の相互作用およびFGF、BMPおよびSHHを含む種々の規定のシグナル伝達経路は、in vivoでの嚢の陥入、適切な腺の発生およびホルモンサブタイプの特異化に重要であることが示された(図16A、上のパネル)(検討のため、(Zhuら、2007年)を参照のこと)。ここで、本発明者らは、プラコードの運命へとhPSCの分化を方向付けるこれらの重要なシグナル伝達経路をモジュレートすることが、下垂体アイデンティティを誘発するのに十分であるか否かを評価した(図16A、下のパネル)。本発明者らのデータにより、SHH、FGF8およびFGF10に時間を決めて曝露することにより、PITX1/2、LHX3/4ならびにHESX1およびSIX6を含む下垂体前葉の発生に関連する遺伝子を堅固に誘導することが示される(図16B)。下垂体分化条件下での発現は、PITX1、LHX3、LHX4およびSIX6に対する抗体を使用するタンパク質レベルについても確認された(図16C)。
腺下垂体の主な機能は、ストレス応答(副腎皮質刺激ホルモン[ACTH])骨格成長(成長ホルモン[GH])、代謝(甲状腺刺激ホルモン[TSH])および生殖機能(プロラクチン[PRL]、および卵胞刺激ホルモン[FSH]、黄体形成ホルモン[LH])を含むヒトの身体における重要な事象を制御する6つの異なるホルモンを分泌することである。したがって、本発明者らは、30日間の分化後に、本発明者らの培養におけるホルモンのサブタイプの存在を評価した。本発明者らは、その培養において、ACTH、GH、PRLならびにFSHおよびLH発現細胞を検出することができた(図18A)。細胞培養上清のELISA測定によって、細胞が、ACTH、GHおよびFSHの分泌の基本的速度を示すことが確認された(図18B)。下垂体前葉におけるホルモンの放出は、種々の標的器官によって分泌される種々の因子から、および門脈を介して視床下部細胞によって放出される上流の因子からのいくつかのフィードバック機構によってしっかりと調節される。したがって、下垂体細胞の機能的応答は、これらの種々の調節刺激と密接にリンクする必要がある。分化30日目のhPSCから誘導された下垂体細胞は、CRF、ストレシンまたはウロコルチンによる刺激に応答して、ACTHの放出の誘導を示した。対照的に、グレリンやソマトクリニンなどの不適当な刺激への曝露はACTHの放出を誘発しなかった(図18C)。一方、CRFを除くソマトクリニンへの曝露は、GH放出において堅固な増加を誘発した(図18D)。最後に、本発明者らは、ナファレリンに曝露した際のFSHの誘導を示すことができた(図18E)。
下垂体腺内での種々のホルモン前駆体系統の分化(Tabar、2011年)は、種々のパターニング事象に関与するしっかりと調節された空間的および時間的プロセスである(図16A)。本発明者らのhPSCベースの培養系における前駆体の運命の多様性に対処するために、本発明者らは、Fluidigmプラットホームを使用する単一細胞のqRT−PCR実験を実施した。本発明者らは、多能性幹細胞から成熟ホルモン発現細胞への全下垂体発生にわたる34種の遺伝子についてプロービングした。本発明者らは、T(Brachyury)、MYODおよびSOX17などの有効な中胚葉または外胚葉の夾雑物についてアッセイするためのプライマーも含めた。分化30日目および60日目の主成分分析(PCA)により、ほとんどの細胞が明確な時間に依存する変化を示し、いくつかの細胞だけが、計画より前に移動するか(すなわち、30日目の細胞が60日目のプロファイルを示す)、または遅延する(すなわち、60日目の細胞が30日目の特性を保持する)ことが示された(図19A、B)。スクリープロット(図19B)により、データの変動のほとんどを説明するPCA成分が規定された。階層的クラスタリングによって、いくつかのより小さなサブクラスターと共に散在する2つの主要なクラスターにおいて生じる時間軸に非常に沿う細胞の分離が確認された(図19C)。
下垂体機能低下症は、非常に多様で複雑な疾患である。下垂体機能障害の原因に応じて、影響するホルモンの種類は様々となりうる。例えば、GH不足は、通常、先天性遺伝疾患を有する患者で観察される(van Gelderenおよびvan der Hoog、1981年)が、後に放射線処置を受けた患者でも生じる場合がある(SklarおよびConstine、1995年)。対照的に、下垂体の自己免疫疾患である、リンパ球性下垂体炎は、主にACTHに影響を及ぼす(Rivera、2006年)。したがって、将来のhPSCから誘導された下垂体細胞の広範な適用のために、細胞代替療法が、所与の患者集団の特異的な需要に対してカスタマイズされる必要を有する場合がある。本発明者らの標準的条件により、背側のACTH+細胞が最も得られるため、本発明者らは、さらなるシグナルを使用して、より腹側の細胞型の生成を増強することができるか否かを調べた。
in vivoで生存し、機能するhPSCから誘導された下垂体細胞の能力を評価するために、本発明者らは、30日目の細胞を下垂体切除したラットに移植した。傍咽頭方法を使用する下垂体(図21A)の外科手術による除去の後、ラットにおける下垂体機能低下症を、血中のACTHレベルを測定することによって確認した。下垂体切除に成功したラットを2つの実験群、マトリゲルのみの注射を受ける偽群(n=4)とhESCから誘導された下垂体細胞を含有するマトリゲルを受ける移植群(n=7)に分けた(30日目、BMP2処理を行わない標準条件)。2×106個の細胞を皮下に移植した後、処置群および対照群の両方について、移植後7週間、血流中の下垂体ホルモンレベルをモニタリングした(図21B〜D)。移植の3週間後からスタートして、移植群のホルモンレベルは、1週間の時点で比較して増加し始め、一方偽群のレベルはほとんど変化しないままであった。ACTHレベルは、実験の7週間の期間、移植対対照群で一定の高いレベルのままであった(図21B)。損傷していない動物で観察されたACTHレベルと比較して、移植群は、ACTHレベルの約60%を回復した(データは示さず)。2種の他のホルモン、すなわち、GHおよびLHのレベルの増加は、より可変的で、試験した時点で全く有意ではなかった。しかし、本発明者らは、移植の3週間および7週間後のGHレベルでは有意な増加を検出することができた(図21C)。LHレベルの有意な増加は観察されなかった(図21D)。移植した動物のホルモンレベルを週齢の一致するインタクトなラットのレベルと比較し、ACTHについて約40%、GHについて約28%、およびPRLについて約20%であることが分かった(図35)。移植細胞の機能を評価する最終段階で、本発明者らは、ホルモン分泌によって影響を受けた下流因子の測定を実施した。ACTHの放出に際し、通常のHPA軸応答の一部として副腎により分泌されるグルココルチコイドの増加が存在する(WebsterおよびSternberg、2004年)。ヒトでは、ACTHは、コルチゾールの放出を誘発するが、げっ歯類の主なグルココルチコイドはコルチコステロンである(Wand、2008年)。したがって、本発明者らは、両方の実験群でコルチコステロンのレベルを測定した(図21E)。移植群では、移植の7週間後までに、統計的に有意な差を生じるコルチコステロンの一貫して高いレベルが示された。これらのデータにより、hPSCから誘導された細胞によって放出されたヒトのACTHは、宿主の副腎からのステロイドホルモンの放出を誘発することができることが示される。移植の7週間後に、動物を屠殺し、移植片を組織学的に分析した(図21F〜G)。本発明者らは、移植片に関して6つの下垂体前葉ホルモンのそれぞれを発現する細胞を検出することができた(図21F)。これらのデータにより、in vivoでの細胞の生存が確認され、in vivoでのさらなる分化と細胞の成熟が示唆される。移植片の立体解析学的定量化によって、移植片当たり平均3.08×106±0.42×106(平均±SD;n=3)個のヒトの細胞が示され、免疫組織化学によりSIX1とヒトの核抗原(hNA)の同時発現により決定されるように、大多数(95%超)はプラコードのアイデンティティを有した。移植片におけるKi67陽性増殖細胞の比率は、hNA+集団の9.6%±0.6%(平均±SD;n=3)であった。移植片全体は、多能性関連表面マーカーであるSSEA−4およびTra1−60について陰性に染色された。腫瘍の徴候は、移植の7週間後まで検出されなかった。移植片の立体解析学的定量化により、動物当たり平均18212±2969個のACTH+細胞が示され(光学的細胞分画法)、81±10mm3の移植片体積を有した(Cavalieri estimator;平均±SD;n=3)。
ESCおよび培養条件
ヒト多能性幹細胞H9(WA−09、XX、35〜50継代)、MEL−1(XY、20〜40継代)、HUES−6(XX、24〜40継代)、hiPSC(胎仔の線維芽細胞株MRC5(ATCC CCL−171)から誘導した社内で生成したhiPSC(Chamberら、2009年)、XY、15〜30継代)および改変レポーター細胞株(すべてH9バックグラウンド、40〜75継代)をEssential8培地(E8)(Fisher Scientific)(Chenら、2011年)を使用するVTN−N(Fisher Scientific)で維持し、EDTAを使用して1週間に2回継代した(Chen、2008年)。細胞を1ヵ月に1回、マイコプラズマの夾雑について試験した。
神経外胚葉への分化を以前に記載したもの(Chambers 2009年)に若干の修正を加えて実施した。手短に述べると、細胞をE8+Y−27632(Tocris)中でVTN−Nでコーティングしたディッシュに1cm2当たり250000個の細胞で播種した。24時間後(0日目)、培地を10μMのSB431542(Tocris Biosciences)、500nMのLDN193189(Stem Cell Technologies)および1μMのXAV939(Tocris Biosciences)(5日目まで)を補充したEssential6(E6)(Chen)に交換した。5日目から、XAV939を培地から除去した。培地を11日目まで毎日交換した。
標準的な(本文で別段言及していなければ)下垂体の成熟として、30日目の培地は、さらなる30日のために「E6のみ」に交換した。パターニング実験(本文で示した)として、高濃度のFGF8(100ng/ml、背方化)、高濃度のBMP2(20ng/ml、腹方化)または中間の濃度で両方(FGF8 50ng/ml、BMP2 10ng/ml)のいずれかをE6培地に補充した。
製造業者のプロトコルに従って、Phase−lockチューブ(5Prime)と組み合わせてTRIzol(Fisher Scientific)試薬を使用して、少なくとも3つの独立した実験から全RNAを抽出した。iScript(BioRad)を使用して、1μgの全RNAをcDNAに逆転写した。定量的RT PCRとして、本発明者らは、BioRadのCFX96 Thermal CyclerでQuantiTectプライマーアッセイ(Qiagen)と組み合わせたSSoFast EvaGreen Mix(BioRad)を使用した。すべての反応を製造業者のプロトコルに従って実行した。遺伝子発現をグリセルアルデヒド3−リン酸デヒドロゲナーゼ(GAPDH)および対照の細胞型(図で示されている)に対して正規化した。結果は、ΔΔCt方法(LivakおよびSchmittgen、2001年)を使用して計算した。
単一細胞のPCR分析として、Accutaseを使用して細胞を剥離させた。40μmの細胞ストレーナーを通して濾過した後、BDFACS Aria III機器でDAPI陰性細胞を選別し、実質的に細胞調製物からデブリと死細胞を一掃した。選別された細胞懸濁液を1ml当たり400000個の細胞の濃度まで調整した。製造業者のマニュアルに従ってFluidigm C1システムを使用して単一細胞を捕捉した。各C1チップ毎の捕捉率を標準組織培養明視野顕微鏡を使用して微視的に確認した。捕捉率は以下の通りであった:30日目:91%(87/96)60日目:94%(90/96)60日目 FGF8:93%(89/96)60日目 FGF8/BMP2:89%(85/96)60日目 BMP2:93%(89/96)。細胞を溶解し、RNAを抽出し、製造業者のプロトコルに従って、ウェットラボで試験したFluidigmのDELTAgeneアッセイと組み合わせてC1を使用してcDNAに転写した(図27)。ウェットラボで試験したDELTAgeneアッセイは、Fluidigmから直接購入した。得られたcDNAを1:5に希釈して、製造業者のマニュアル(「Fast Gene Expression Analysis Using EvaGreen on the BioMark or BioMark HD System」)に従って、EvaGreen化学と組み合わせたFluidigm BioMarkシステムを使用して単一細胞のPCR増幅に供した。Fluidigmの96.96 Dynamic Arrayを使用してPCRを実行した。各プライマー対をチップ上に技術的に二連でランした。偽陰性の数を増加させることを対価として偽陽性の細胞(calls)を最小限にするために、技術的プライマー反復間で一貫した増幅結果を生じる単一細胞のみを考慮した。全体の不一致率は低かった(プライマー対当たり3%超)。FluidigmのSINGuLAR Analysis Toolset for R(バージョン3.0.2(2013−09−25)「Frisbee Sailing」)と組み合わせてFluidigmのリアルタイムPCR分析ソフトウェアを使用して発現データを分析した。
細胞を1回PBSで洗浄した後、細胞を4%(v/v)のパラホルムアルデヒドで20分間固定し、PBSで2回洗浄し、PBS中0.1%(v/v)のTriton X−100を使用して透過処理し、細胞をPBS中10%(v/v)のFCSを用いて室温で1〜5時間ブロッキングした。細胞をPBS中2%のFCS(v/v)で希釈した一次抗体と一緒に、4℃で終夜インキュベートした。この研究で使用した一次抗体のリストを図28に提供する。一次抗体のインキュベーション後、細胞をPBSで2回洗浄し、その後、PBSで希釈した適当なAlexaFluorとコンジュゲートした二次抗体と一緒に、室温で1時間インキュベートした(1:1000;Molecular Fisher Scientific)。PBSで2回洗浄した後、核をDAPIを使用して染色した。さらに2回の洗浄ステップの後、HamamatsuのORCA CCDカメラを備えたOlympusのIX71倒立顕微鏡を使用して、細胞の蛍光画像を撮った。
BD Lyoplate(商標)細胞表面マーカーのスクリーニングでは、30日目の細胞を、Accutaseを使用して、1cm2当たり100000個の細胞密度で96ウェルイメージングプレートに再播種した。4時間後、付着期の細胞をバイオイメージングのために使用者のマニュアルに従って染色した。細胞をOperetta High Content Imaging System(Perkin Elmer)で分析した。画像を処理し、Harmonyソフトウェアパッケージ(Perkin Elmer)を使用して分析した。
in vitroでホルモン放出を刺激するために、上述のように、細胞を24ウェルプレートで分化させた。分化の30日目に、細胞をPBSで1回洗浄し、溶媒または刺激物のいずれかを含有する250μlの新鮮な培地を各ウェルに添加した。12時間後に、上清を除去し、2000gで5分間遠心分離して、デブリをペレット化した。上清を新しい反応チューブに移し、急速冷凍して、ELISA測定まで−80℃で保存した。使用した刺激物は以下の通りであった:CRF(Tocris、1μM)、ストレシンI(Tocris、2μM)、グレリン(Tocris、1μM)、ソマトクリニン(Accurate Chemical、1μg/ml)、ナファレリン(Tocris、1μM)およびウロコルチン(Tocris、500nM)。
細胞上清中または動物の血清中のホルモン濃度をELISA測定を使用して分析した。細胞培養物の上清中のホルモン濃度を、製造業者のマニュアルに従って伝統的な単一ホルモンELISAキットを使用して評価した。ACTH(Calbiotech、ラットとヒトのACTHを検出する)、hGH(R&D Systems、ヒトに特異的)、FSH(Calbiotech、FSH(lumELISA、ヒトに特異的)およびコルチコステロン(Abcam)。プレートをEnSpire Multimodeプレートリーダー(PerkinElmer)を使用して読み取った。in vivoでの試料のホルモン濃度を、伝統的なELISA(ACTHに対してのみ、1:2に希釈した血清)またはLuminex technology(Millipore)を使用する種特異的(ヒトまたはラット)Milliplex multiplex ELISAのいずれかを使用して分析した。磁気ビーズをベースとするサンドイッチイムノアッセイを製造業者のマニュアルに従って実施した。二連のウェルで25μlの希釈していない血清試料をLuminex FlexMap 3D(Luminex Corp、Austin、TX)によって分析した。サイトカインの濃度を5−p log分析を使用するLuminexのXponent 4.1およびEMD−MilliporeのMilliplex Analyst v5.1によって決定した。
オスの無胸腺ヌードラット(RNUラット Crl:NIH−Foxn1rnu、Charles River Laboratories)を8週齢で傍咽頭アプローチを使用して下垂体切除した。血漿のACTHを下垂体切除の1週間後に測定し、下垂体機能低下症を確認した。下垂体を切除したラットを2つの群に無作為化した:偽対照(n=4)、ヒトESから誘導した下垂体細胞を皮下移植した群(n=7)。シリンジ(1ml、BD biosciences)とマトリゲル(BD biosciences)を注射前に氷上で冷やし、注射前にマトリゲルがゲルを形成するのを防いだ。ラットの首の毛を剃り、ベタジンおよび70%のエタノールを用いて調製した。0.9mlのマトリゲルを2百万個のヒト下垂体細胞の懸濁液(100μlのessential E6培地中)と混合した。マトリゲルと細胞の混合物をラットの首の皮下組織に注射した。

Claims (35)
- 幹細胞の分化を誘導するためのin vitro方法であって、幹細胞の集団を有効量の形質転換成長因子ベータ(TGFβ)/アクチビン−Nodalシグナル伝達の1種または複数の阻害剤、およびBMPシグナル伝達の1種または複数の活性化剤と少なくとも約2日間接触させるステップを含み、幹細胞の前記集団を、有効量の
(a)Wntシグナル伝達の1種または複数の活性化剤、
(b)FGFシグナル伝達の1種または複数の活性化剤、および
(c)FGFシグナル伝達の1種または複数の阻害剤
の1つまたは複数とさらに接触させて、1種または複数の神経堤、頭蓋プラコード、または非神経外胚葉系統マーカーを発現する分化細胞の集団を生成するin vitro方法。 - 幹細胞の前記集団を形質転換成長因子ベータ(TGFβ)/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤と少なくとも約12日間接触させるステップを含む、請求項1に記載の方法。
- 形質転換成長因子ベータ(TGFβ)/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤とBMPシグナル伝達の1種または複数の活性化剤を、幹細胞の前記集団に同時に接触させる、請求項1に記載の方法。
- 幹細胞の前記集団を、Wntシグナル伝達の1種または複数の活性化剤と接触させ、ここで、前記細胞が、1種または複数の神経堤系統マーカーを発現する、請求項1に記載の方法。
- 形質転換成長因子ベータ(TGFβ)/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤とWntシグナル伝達の1種または複数の活性化剤を、幹細胞の前記集団に同時に接触させる、請求項4に記載の方法。
- 幹細胞の前記集団を、FGFシグナル伝達の1種または複数の活性化剤と接触させ、ここで、前記細胞が、1種または複数の頭蓋プラコード系統マーカーを発現する、請求項1に記載の方法。
- FGFシグナル伝達の前記活性化剤を、前記細胞のTGFβ/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤との最初の接触の約2日後に、幹細胞の前記集団に接触させる、請求項6に記載の方法。
- 前記頭蓋プラコード系統マーカーが、水晶体プラコード系統マーカーである、請求項6に記載の方法。
- 幹細胞の前記集団をWntシグナル伝達の1種または複数の活性化剤と接触させるステップをさらに含み、前記細胞が、1種または複数の三叉神経プラコード系統マーカーを発現する、請求項6に記載の方法。
- Wntシグナル伝達の前記活性化剤を、前記細胞のTGFβ/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤との最初の接触の約2日後に、幹細胞の前記集団に接触させる、請求項9に記載の方法。
- 幹細胞の前記集団を、FGFシグナル伝達の1種または複数の阻害剤と接触させ、ここで、前記細胞が、1種または複数の非神経外胚葉系統マーカーを発現する、請求項1に記載の方法。
- 形質転換成長因子ベータ(TGFβ)/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤とFGFシグナル伝達の1種または複数の阻害剤を、幹細胞の前記集団に同時に接触させる、請求項11に記載の方法。
- 幹細胞の前記集団の、TGFβ/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤との最初の接触から約12日目またはその後に、前記神経堤、頭蓋プラコード、または非神経外胚葉系統マーカーの1種または複数を発現する分化細胞の集団へと分化させる、請求項1に記載の方法。
- TGFβ/アクチビン−Nodalシグナル伝達の前記1種または複数の阻害剤が、SB431542、その誘導体、およびそれらの混合物からなる群より選択される小分子を含む、請求項1に記載の方法。
- Wntシグナル伝達の前記1種または複数の活性化剤が、Wntシグナル伝達の活性化のためにグリコーゲン合成酵素キナーゼ3β(GSK3β)を低下させる、請求項4または9に記載の方法。
- Wntシグナル伝達の前記1種または複数の活性化剤が、CHIR99021、そのWNT3A誘導体、およびそれらの混合物からなる群より選択される小分子である、請求項4または9に記載の方法。
- FGFシグナル伝達の前記1種または複数の活性化剤が、FGF2、その誘導体、およびそれらの混合物を含む、請求項6に記載の方法。
- 前記1種または複数の神経堤系統マーカーが、SOX10からなる群より選択される、請求項1に記載の方法。
- 前記1種または複数の頭蓋プラコード系統マーカーが、SIX1、PAX3、PITX3、クリスタリンアルファA、クリスタリンアルファB、およびこれらの組合せからなる群より選択される、請求項1に記載の方法。
- 前記1種または複数の非神経外胚葉系統マーカーが、TFAP2A、および検出可能なSIX1およびSOX10発現の非存在からなる群より選択される、請求項1に記載の方法。

幹細胞の集団を有効量の

(b)少なくとも約2日間のBMPシグナル伝達の1種または複数の活性化剤、
(c)SHHシグナル伝達の1種または複数の活性化剤、および
(d)FGFシグナル伝達の2種またはそれより多い活性化剤
と接触させるステップを含み、
細胞の前記集団を、(c)および(d)に約20日間またはそれより長い間接触させて、
1種または複数の下垂体プラコード系統マーカーを発現する分化細胞の集団を生成するin vitro方法。 - 1種または複数の神経堤、頭蓋プラコード、または非神経外胚葉系統マーカーを発現するin vitroで分化した細胞の集団であって、請求項1から22のいずれか一項に記載の方法に従って幹細胞の集団から誘導されるin vitroで分化した細胞の集団。
- 請求項23に記載のin vitroで分化した細胞の集団を含む組成物。
- 被験体における神経変性障害または下垂体障害を処置する方法であって、有効量の請求項23に記載のin vitroで分化した細胞を、神経変性障害を患っている被験体へと投与するステップを含む方法。
- 前記下垂体障害が下垂体機能低下障害である、請求項25に記載の方法。
- 神経変性障害または下垂体障害を処置するための医薬の製造における、請求項23に記載のin vitroで分化した細胞集団の使用。
- 前記下垂体障害が下垂体機能低下障害である、請求項27に記載の使用。
- 幹細胞の分化を誘導するためのキットであって、有効量の1種または複数の

を含むキット。 - 幹細胞の分化を誘導するためのキットであって、有効量の
