Abstract
Description
図1の切断地図のように、植物体でアフリカ豚熱ウイルス抗原P32タンパク質(感染性サイクルの初期段階でウイルスの内在化に関与すると知られた)を発現させることができるように組換えした植物発現ベクターを作製した。より詳細には、アフリカ豚熱ウイルスのP32タンパク質に対する遺伝子情報を確保し、植物体での発現に最適化した配列でP32タンパク質をコードする遺伝子(配列番号1)を合成した。
(2.1.植物発現アフリカ豚熱ウイルスP32抗原ベクターの一過性発現)
前記実施例1で準備した植物発現ベクターをアグロバクテリウム(Agrobacterium)LBA4404菌株に電気穿孔法を用いて形質転換させた。形質転換されたアグロバクテリウムを5mLのYEP液体培地(酵母抽出物10g、ペプトン10g、NaCl 5g、カナマイシン50mg/L、リファンピシン25mg/L)で28℃の条件で16時間振とう培養した後、1次培養液1mLを50mLの新しいYEP培地に接種して、28℃の条件で6時間振とう培養した。このように培養されたアグロバクテリウムは、遠心分離(7,000rpm、4℃、5分)して収集した後、600nmの波長で吸光度(O.D)値が1.0になるようにインフィルトレーション(Infiltration)バッファー[10mMのMES(pH5.7)、10mMのMgCl2、200μMのアセトシリンゴン]に懸濁させた。アグロバクテリウム懸濁液は、注射針を除去した注射器を用いてNicotiana benthamianaの葉の裏面に注入する方法でアグロインフィルトレーションを行った。
前記実施例2.1で準備した植物の葉からタンパク質を抽出して遠心分離した後に、上清画分(S)にあるタンパク質と沈殿(P)分画分にあるタンパク質、上清画分と沈殿を全部含む画分(Total;T)をそれぞれ分離して、ウェスタンブロッティングで組み換えP32抗原タンパク質の発現を確認した。より詳細には、各画分30μLをSDS試料バッファーと混合した後に加熱した。そして、10%SDS−PAGEゲルに電気泳動してサイズ別に分離したタンパク質バンドを確認し、これをPVDF膜に移動させた後に、5%スキムミルクを用いてブロッキング段階を経た後、ポリヒスチジンと反応する抗体を結合させ、ECL溶液を製造社で提供する方法によって処理して、組み換えP32抗原タンパク質の発現を確認した。
前記実施例2.2で準備したP32抗原タンパク質を用いて抗体血清診断キットの試作品を作製し、スペインASF標準実験室で提供する血清にて反応性及び敏感度テストを進めた。

植物体で発現することを特徴とする、アフリカ豚熱の診断のための抗原生産用組み換えベクター。 - 前記組み換えベクターは、配列番号3のBiP(chaperone binding protein)をコードするポリヌクレオチドをさらに含むことを特徴とする、請求項1に記載のアフリカ豚熱の診断のための抗原生産用組み換えベクター。
- 前記BiPをコードするポリヌクレオチドは、前記P32タンパク質をコードするポリヌクレオチドの5′末端方向に位置することを特徴とする、請求項2に記載のアフリカ豚熱の診断のための抗原生産用組み換えベクター。
- 前記組み換えベクターは、配列番号4のポリヒスチジンをコードするポリヌクレオチドをさらに含むことを特徴とする、請求項1に記載のアフリカ豚熱の診断のための抗原生産用組み換えベクター。
