Abstract
Description
スラウストキトリッドファミリー微細藻類菌株を分離するために、次の実験を行った。
実施例2−1:人為突然変異法による変異株の選択
本出願においては、実施例1で分離したスラウストキトリウム属(Thraustochytrium genus)KC01菌株のガンマ線照射変異によりドコサヘキサエン酸(Docosahexaenoic acid: DHA)の生産がさらに向上した菌株を分離するために、次の実験を行った。
実施例2−1で選択したCJM01とKC01菌株の多価不飽和脂肪酸含有オイル生産能を比較するために、次のように培養した。
また、CJM01菌株の培養特性を確認するために、前記培養液における総アミノ酸含有量を確認した。
実施例2で選択したアミノ酸低減及びDHA含有量が向上したCJM01菌株を用いて、DHA含有量及び生産性をさらに向上させるために、培地条件の最適化を行った。
Claims (9)
- 野生型に比べてドコサヘキサエン酸(DHA)生産量が増加し、アミノ酸生産量が減少したスラウストキトリウム属(Thraustochytrium genus)CJM01微細藻類(寄託番号:KCTC13538BP)。
- 前記スラウストキトリウム属CJM01微細藻類は、脂肪酸総重量に対して40〜60重量%のドコサヘキサエン酸を含む、請求項1に記載の微細藻類。
- 前記スラウストキトリウム属CJM01微細藻類は、ドコサヘキサエン酸生産性が0.5〜0.7(g/l/h)である、請求項1に記載の微細藻類。
- 請求項1に記載のスラウストキトリウム属CJM01微細藻類を培養するステップと、前記微細藻類、その培養物、その乾燥物又はその破砕物からドコサヘキサエン酸含有バイオマスを回収するステップとを含むバイオマス製造方法。
- 前記培養は、従属栄養条件下で行うものである、請求項4に記載のバイオマス製造方法。
- 前記培養は、炭素源及び窒素源を含む培地を用いて行うものである、請求項4に記載のバイオマス製造方法。
- 前記炭素源は、グルコース、フルクトース、マルトース、ガラクトース、マンノース、スクロース、アラビノース、キシロース及びグリセリンからなる群から選択される少なくとも1種である、請求項6に記載のバイオマス製造方法。
- 前記窒素源は、i)酵母抽出物、牛肉抽出物(beef extract)、ペプトン(peptone)及びトリプトン(tryptone)からなる群から選択される有機窒素源、又はii)酢酸アンモニウム、硝酸アンモニウム、塩化アンモニウム、硫酸アンモニウム、硝酸ナトリウム、ウレア及びMSG(Monosodium glutamate)からなる群から選択される無機窒素源である、請求項6に記載のバイオマス製造方法。
- 請求項1に記載のスラウストキトリウム属CJM01微細藻類を培養するステップと、前記微細藻類、その培養物、その乾燥物又はその破砕物からドコサヘキサエン酸含有脂質を回収するステップとを含むバイオオイル製造方法。
