Abstract
本申請案係關於一種易於萃取細胞中之油的新穎裂殖壺菌屬物種品系及一種使用其生產含ω3之油的方法,且本發明之新穎破囊壺菌系列微藻在生物質中具有高脂肪含量且其中具有高含量之不飽和脂肪酸,諸如二十二碳六烯酸及二十碳五烯酸,且極易於自生物質萃取藉由自身或藉由培養及醱酵所產生之生物質及包括不飽和脂肪酸之脂肪組分。因此,該微藻、生物質乾燥產物及自其產生之生物油可有效地用作用於飼料之組成物或用於食品之組成物或其類似物。

本發明係關於一種易於萃取細胞中之油的新穎裂殖壺菌屬物種(Schizochytrium sp.)品系及一種使用其生產含ω3之油的方法。
破囊壺菌(Thraustochytrid)在自然界之各種環境中存活及分佈。其藉由附著至生物體而以共生關係生存,漂浮於海洋環境或淡水或半鹹水環境中,且分佈於各種沈積層中存活。此等破囊壺菌屬於海洋生態食物鏈之最低層,且亦歸類為作為浮游植物之有機異養原生生物微藻(microalgae)。自然環境中之破囊壺菌在功能上負責自然循環元素(諸如硫、氮、磷、鉀及其類似物)之循環及純化。此外,其含有高濃度之多不飽和脂肪酸(polyunsaturated fatty acid,PUFA)
以充當海洋生態系統之來源,該多不飽和脂肪酸包括歸類為ω-3之二十二碳六烯酸(docosahexaenoic acid,DHA)及二十碳五烯酸(eicosapentaenoic acid,EPA)。
包括人類之大部分高級生物體本身無法合成包括二十二碳六烯酸及二十碳五烯酸之多不飽和脂肪酸,因此必須將其作為必需養分食用。多不飽和脂肪酸當中的二十二碳六烯酸及二十碳五烯酸為腦、眼組織及神經系統所必需的脂肪酸,且特定言之,已知其在嬰兒之神經系統(諸如視力及運動神經能力)發育及預防心血管疾病方面起重要作用,且其為腦結構脂質中最豐富的組分。
迄今為止,多不飽和脂肪酸之主要來源為自鯵魚類之油中萃取的魚油,該鯵魚類諸如鯖魚、秋刀魚、鮪魚、鯵魚、沙丁魚、鯡魚及其類似物,且此極適用作用於海水魚之魚類養殖飼料,諸如開口餌料(initial feed)。儘管已在工業上研發自魚油萃取及攝入多不飽和脂肪酸,但亦存在缺點。魚油之品質視魚類物種、季節及捕魚地點而變化,且魚油係經由捕魚產生的,因此其難以連續供應。另外,由於魚油中所包含之重金屬及有機化學物質之污染問題、在加工過程期間雙鍵氧化之問題以及魚油所特有之魚腥味及其類似問題,在製造過程及生產中存在侷限性。
為解決此等問題,近來,正進行關於藉由微生物培養生產包括二十二碳六烯酸及二十碳五烯酸之多不飽和脂肪酸之方法的研究。特定言之,除能夠天然地新合成脂肪酸之外,微藻亦可提供優於魚油之若干優勢。其可經由以工業規模進行培養來穩定地供應,且其使得能夠產生具有相對恆定生化組成之生物質。不同於魚油,由微藻產生之脂質不會具有任何令人不適的氣味。此外,其具有與魚油相比更簡單的脂肪酸組成,此有助於分離主要脂肪酸之步驟。
基於此等優勢,近來,關於多不飽和脂肪酸之生產的研究及工業化正極其快速地推進,該多不飽和脂肪酸包含ω-3不飽和脂肪酸(ω不飽和脂肪酸),諸如二十二碳六烯酸(DHA)、二十碳五烯酸(EPA)、二十碳四烯酸
(arachidonic acid,ARA)、二十二碳五烯酸(docosapentaenoic acid,DPA)及α-次亞麻油酸及其類似物,且該等研究及工業化主要係由破囊壺菌屬物種(Thraustochytrium sp.)及裂殖壺菌屬物種微生物生產多不飽和脂肪酸,該等微生物為一種海洋微藻。舉例而言,揭示了一種用於使用裂殖壺菌屬物種ATCC20888及裂殖壺菌屬物種PTA10208(其為裂殖壺菌屬物種微生物)生產ω-3多不飽和脂肪酸之方法(美國專利第5,130,242號)及另外,一種用於使用破囊壺菌屬物種ATCC10212(破囊壺菌屬物種PTA10212)(其為破囊壺菌系列破囊壺菌屬物種微生物)生產二十二碳六烯酸及二十碳五烯酸之方法。
本發明之一個具體實例提供一種新穎裂殖壺菌屬物種微藻。在一個特定具體實例中,新穎裂殖壺菌屬物種微藻可為裂殖壺菌屬物種CD01-1821微藻(寄存編號KCTC14660BP)。
本發明之另一具體實例提供衍生自裂殖壺菌屬物種微藻之生物質或生物油。
本發明之其他具體實例提供一種飼料組成物,其包含裂殖壺菌屬物種微藻衍生之生物質、生物油或其組合。

本發明之一個具體實例提供一種新穎裂殖壺菌屬物種微藻。
本說明書中所用之術語「破囊壺菌(Thraustochytrid)」意謂破囊壺菌目(order Thraustochytriales)微藻。另外,本說明書中所用之術語「裂殖壺菌屬物種(Schizochytrium sp.)」為屬於破囊壺菌目之破囊壺菌科(family Thraustochytriaceae)的屬名稱中之一者,且可與術語「裂殖壺菌屬物種(裂殖壺菌屬)(Schizochytrium sp.(genus Schizochytrium))」互換使用。此外,術語「微藻(microalgae)」意謂無法用裸眼看到且僅可經由顯微鏡在具有葉綠素之光合植物中看到且在水中以自由浮動方式生存的生物體,且亦稱為浮游植物。存在各種類型之微藻,且由於光合成係不可能的,因此甚至包括僅經由異養生長之品系。
作為本發明中之一個具體實例,在自海岸區域收集環境樣品以獲得破囊壺菌系列之純的經分離群落之後,作為總脂質及脂肪酸之分析的結果,獲得兩種品系亦即具有極佳胞內DHA含量之CD01-1821及CD01-1822。作為培養評估之結果,證實CD01-1821品系更適用於按比例擴大方法(scale-up process),因為該品系在相同醱酵條件下具有更高的總生物質產量及總油含量。
因此,在本發明中,新穎裂殖壺菌屬物種微藻可為裂殖壺菌屬物種CD01-1821微藻(寄存編號KCTC14660BP)。
另外,裂殖壺菌屬物種品系可具有SEQ ID NO:1之18s rRNA核苷酸序列,但不限於此。舉例而言,裂殖壺菌屬物種微藻可具有18S rRNA,其由展示與SEQ ID NO:1之核苷酸序列具有80%或更高、85%或更高、90%或更高、95%或更高、98%或更高或99%或更高之序列一致性的核苷酸序列組成,但不限於此。
本說明書中所用之術語「二十二碳六烯酸(docosahexaenoic acid,DHA)」為具有C22H32O2之化學式的多不飽和脂肪酸中之一者,且對應於ω-3脂肪酸以及α-次亞麻油酸(α-次亞麻油酸(α-linolenic acid):ALA)及二十碳五烯酸(eicosapentaenoic acid,EPA),且常見名稱為二十二碳六烯酸(cervonic acid),且其亦可表達為呈縮寫形式之22:6 n-3。
本說明書中所用之術語「二十碳五烯酸(eicosapentaenoic acid,EPA)」為具有C20H30O2之化學式的多不飽和脂肪酸中之一者,且對應於ω-3脂肪酸以及ALA及DHA,且其亦可表達為呈縮寫形式之20:5 n-3。
按脂肪酸之總重量計,裂殖壺菌屬物種微藻可生產及/或包含35重量%至60重量%之DHA。舉例而言,以脂肪酸之總重量計,裂殖壺菌屬物種微藻可生產40重量%至65重量%、45重量%至65重量%、50重量%至65重量%、40重量%至60重量%、45重量%至60重量%、50重量%至60重量%或55重量%至60重量%之DHA。
按脂肪酸之總重量計,裂殖壺菌屬物種微藻可生產及/或包含0.1重量%至2重量%之EPA。舉例而言,以脂肪酸之總重量計,裂殖壺菌屬物種微藻可生產0.2重量%至2重量%、0.2重量%至1.5重量%、0.2重量%至1重量%、0.3重量%至2重量%、0.3重量%至1.5重量%、0.3重量%至1重量%、0.5重量%至2重量%、0.5重量%至1.5重量%、0.5重量%至1重量%、0.5重量%至0.9重量%或0.6重量%至0.8重量%之EPA。
本發明之另一態樣提供衍生自裂殖壺菌屬物種微藻之生物質或生物油,其包含裂殖壺菌屬物種CD01-1821微藻、微藻之培養物、培養物之乾燥產物或乾燥產物之溶胞產物。

本說明書中所使用之術語「生物質(biomass)」意謂生質能之能量來源,亦即活機體,諸如植物、動物、微生物及其類似者,且亦意謂生態學上單位時間及空間中存在之特定活機體之重量或能量的量。另外,生物質包含細胞所分泌之化合物,但不限於此,且可含有細胞及/或細胞中之內含物以及細胞外材料。在本申請案中,生物質可為裂殖壺菌屬物種微藻本身、其培養物、其乾燥產物、其溶胞產物或藉由培養或醱酵微藻產生之產物,或可為生物質之濃縮物或乾燥產物,但不限於此。
裂殖壺菌屬物種微藻之「培養物」係指藉由培養微藻產生之產物,且具體言之,其可為包含微藻之培養溶液或自培養溶液移除微藻之培養物濾液,但不限於此。裂殖壺菌屬物種微藻(microalgal)培養物之「乾燥產物」自微藻培養物移除水分,且例如其可呈微藻之乾燥微生物細胞形式,但不限於此。此外,乾燥產物之「溶胞產物(lysate)」統稱為對自微藻培養物移除水分之乾燥產物進行裂解的結果,且例如其可為乾燥微生物細胞粉末,但不限於此。可根據所屬技術領域中已知之培養方法,藉由將微藻接種至微藻培養基來製備裂殖壺菌屬物種微藻之培養物,且培養物之乾燥產物及其溶胞產物亦可根據所屬技術領域中已知之用於加工或乾燥微藻或培養溶液之方法來製備。
衍生自裂殖壺菌屬物種微藻之生物質可包含以生物質之總重量計40重量%至85重量%、45重量%至80重量%、50重量%至75重量%、50重量%至70重量%、54重量%至66重量%之粗脂肪。
衍生自裂殖壺菌屬物種微藻之生物質可包含按生物質之總重量
計5至25重量%、5至20重量%、10至20重量%或15至20重量%之粗蛋白質。
衍生自裂殖壺菌屬物種微藻之生物質可包含以脂肪酸之總重量計35重量%或更多或35重量%至60重量%之DHA,且可包含以脂肪酸之總重量計0.1重量%或更多或0.1重量%至2重量%之EPA,且可包含以脂肪酸之總重量計30重量%至40重量%或更多之棕櫚酸。
裂殖壺菌屬物種微藻具有高油含量高油含量,且特定言之高ω-3含量,以使得可縮短複合糖中之培養時間。因此,其對於按比例擴大方法係有利的。
「油含量(oil content)」可與「粗脂肪含量(crude fat content)」互換使用。
生物質可藉由根據一個態樣之衍生自裂殖壺菌屬物種微藻之生物質的製備方法製備。
本發明之其他態樣提供一種組成物,其包含裂殖壺菌屬物種CD01-1821微藻、微藻之培養物、培養物之乾燥產物或乾燥產物之溶胞產物。

本申請案之組成物可進一步包含:穀粒,諸如經研磨或壓碎之小麥、燕麥、大麥、玉米及稻米;植物蛋白質飼料,例如具有大豆及向日葵作為主要組分之飼料;動物蛋白質飼料,例如血粉、肉粉、骨粉及魚粉;糖及乳製品,例如由各種粉末狀奶粉及乳清粉末組成之乾燥組分,且除此之外,可進一步包含營養補充劑、消化及吸收增強劑、生長促進劑及其類似物。
本申請案之組成物可單獨投予或與可食用載劑當中之其他飼料添加劑組合投予。另外,組成物可直接混合作為追肥(top dressing)或飼餵或易於以分開的口服調配物形式投予以飼餵至動物中。當組成物分開投予以飼餵時,其可作為中間釋放或持續釋放調配物,以同樣所屬技術領域中已知之醫藥學上可接受之可食用載劑的組合形式來製備。此可食用載劑可為固體或液體,例如玉米澱粉、乳糖、蔗糖、大豆片、花生油、橄欖油、芝麻油以及丙二醇。當使用固體載劑時,組成物可為呈錠劑、膠囊、散劑、糖衣錠或口含錠或不可分散形式之追肥。當使用液體載劑時,組成物可為明膠軟膠囊或糖漿或懸浮液、乳液或溶液之調配物。

本說明書中所用之術語「培養物(culture)」意謂使微藻在經適當控制之環境條件下生長。本申請案之培養方法可根據所屬技術領域中已知之合適的培養基及培養條件進行。此培養方法可易於由所屬技術領域中具有通常知識者根據所選擇之微藻來調整及使用。
特定言之,本申請案之裂殖壺菌屬物種微藻之培養可在異養條件(但不限於此)下進行。
本說明書中所用之術語「異養(heterotrophic)」為視獲自體外能量來源或養分來源的有機物而定的營養方法,且為與自養對應的術語,且其可與術語『暗培養(dark culture)』互換使用。
培養裂殖壺菌屬物種微藻不特定限於此,但可藉由已知分批培養方法、連續培養方法、分批進料培養方法以及其類似方法進行。本申請案之微藻的培養中所用的任何培養基及其他培養條件可在無特定限制之情況下使用,只要其為用於培養常見微藻之培養基即可。特定言之,本申請案之微藻可藉由在好氧條件下在含有適合碳源、氮源、磷源、無機化合物、胺基酸及/或維生素及其類似物之常見培養基中調節溫度、pH及其類似者來培養。

用於培養裂殖壺菌屬物種微藻之培養基中所包含的碳源可為選自由以下者組成之群中之任何一或多者:葡萄糖、果糖、麥芽糖、半乳糖、甘露糖、蔗糖、阿拉伯糖、木糖及甘油,但用於培養微藻之任何碳源不限於此。
用於培養裂殖壺菌屬物種微藻之培養基中所包含的氮源可為:(i)選自由酵母萃取物、牛肉萃取物、蛋白腖(peptone)及胰腖(tryptone)組成之群的任何一或多種有機氮源,或ii)選自由乙酸銨、硝酸銨、氯化銨、硫酸銨、硝酸鈉、尿素及麩胺酸單鈉(Monosodium glutamate,MSG)組成之群的任何一或多種無機氮源,但任何用於培養微藻之氮源不限於此。

本發明之新穎破囊壺菌系列微藻在生物質中具有高脂肪含量且其中具有高含量之不飽和脂肪酸,諸如二十二碳六烯酸及二十碳五烯酸,且極易於自生物質萃取藉由自身或藉由培養及醱酵所產生之生物質及包括不飽和脂肪酸之脂肪組分。因此,該微藻、生物質乾燥產物及自其產生之生物油可有效地用作用於飼料之組成物或用於食品之組成物或其類似物。

在下文中,將藉由實施例更詳細地描述本發明。然而,此等實施
例意欲說明性地描述一或多個特定具體實例,且本發明之範圍不受此等實施例限制。
實施例1.破囊壺菌系列微藻之分離
為分離破囊壺菌系列微藻,自沿著韓國西海岸區域、Seocheon、Gunsan、Buan及Yeonggwang-gun區域之總共40個區域收集呈海水、葉子及沈積物形式之環境樣品。以產生及觀測到有機沈積物之特定區域為中心進行取樣,且將所收集之環境樣品在7天內輸送至實驗室環境,且進行用於移除除破囊壺菌系列微藻以外之其他污染源(諸如細菌微生物及真菌及原生生物)的操作。經由連續顯微鏡檢查,聚焦於顯示出特徵形態且在生命週期內形成可觀測到之游走孢子或構成在發育階段期間所產生之外質網狀結構的樣品,來分離出破囊壺菌系列微藻細胞(圖1)。作為分離製程中可使用之分離及培養培養基,利用經改良之YEP培養基(酵母萃取物0.1g/L、蛋白腖0.5g/L、MgSO4.7H2O 2g/L、海鹽50g/L、H3BO3 5.0mg/L、MnCl2 3.0mg/L、CuSO4 0.2mg/L、NaMo4.2H2O 0.05mg/L、CoSO4 0.05mg/L、ZnSO4.7H2O 0.7mg/L、瓊脂15g/L)。可獲得經由若干次分離及繼代培養過程移除污染源之純的經分離群落,且將經分離群落再次在包括抗生素調配混合溶液(硫酸鏈黴素0-50mg/L、安比西林(Ampicillin)0-30mg/L、青黴素G 0-30mg/L、硫酸康黴素(Kanamycin sulfate)0-30mg/L)之固體培養基中進行污染源控制及移除過程,且藉此獲得純的可分離群落。
實施例2.經分離微藻之培養評估及優異品系選擇
針對實施例1中之純的經分離群落進行培養評估,且經由此選擇優異品系。
特定言之,使用經改良之GGYEP培養基(葡萄糖5g/L、甘油5g/L、酵母萃取物0.1g/L、蛋白腖0.5g/L、MgSO4.7H2O 2g/L、海鹽50g/L、H3BO3 5.0mg/L、MnCl2 3.0mg/L、CuSO4 0.2mg/L、NaMo4.2H2O 0.05mg/L、CoSO4 0.05
mg/L、ZnSO4.7H2O 0.7mg/L)在250mL燒瓶中在10-35℃、100-200rpm之條件下培養實施例1中之純的經分離群落約2天。基於已進行的培養的結果,選擇29種可在30℃或更高的溫度下生長,且具有極佳生長速率且可確保微生物細胞量之微藻。對於所選擇之微藻品系,在經改良GYEP培養基中以500mL燒瓶規模培養2天,該培養基包括30g/L葡萄糖作為碳源,且培養條件為30℃、150rpm。在確認所有輸入碳源在培養環境中被消耗2天後,回收全部培養溶液且在60℃之乾燥烘箱中乾燥隔夜以獲得生物質。
為了分析經培養微藻微生物細胞之脂質及多不飽和脂肪酸含量,使用如下方法,且微藻衍生之利用經乾燥微生物細胞之含脂肪酸油係藉由以下方法來量測。方法為,將8.3M鹽酸溶液(HCl)添加至5g乾燥微生物細胞,且接著在80℃下水解微藻微生物細胞之細胞壁,且接著添加乙基醚30mL及石油醚20mL且混合30秒,且接著重複離心3次或更多次。回收經分離之溶劑層,且轉移至預稱重之圓底燒瓶中,且接著經由氮氣吹掃移除溶劑,且在乾燥器中冷卻為恆重。藉由在乾燥之後藉由自燒瓶之重量減去空燒瓶之重量來量測經乾燥油之重量,計算總油含量。藉由用甲醇0.5N NaOH及14%三氟硼烷甲醇(BF3)預處理且藉由氣相層析法量測展示油中所包含之二十二碳六烯酸(DHA)之含量。

下表1之「生物質」意指培養溶液中微生物細胞之濃度,且其可與下表2之細胞乾重(dry cell weight,DCW)互換使用。
(在該表中,TFA意謂總脂肪酸,且可與「粗脂肪含量」或「粗脂肪量」或「總脂質」互換使用。)
因此,如表1及圖2中所示,展示2種CD01-1821及CD01-1822品系中的胞內DHA含量極高。
作為脂肪酸分析之結果,在5L規模培養基中針對具有極佳胞內DHA含量之2種CD01-1821、CD01-1822品系進行培養評估。對於種菌培養,將其在500mL燒瓶中在30℃、150rpm之條件下使用滅菌MJW02培養基(葡萄糖30g/L、MgSO4.7H2O 3.0g/L、Na2SO4 15g/L、NaCl 0.8g/L、酵母萃取物1.0g/L、MSG.1H2O 1.0g/L、NaNO3 1.0g/L、KH2PO4 0.8g/L、K2HPO4 1.5g/L、CaCl2 0.5g/L、維生素混合溶液10ml/L)培養約24小時。將種菌培養之燒瓶等分且接種於
5L培養基中。供應與總培養溶液相比28%之葡萄糖碳源且進行培養約72小時,且在滅菌MJW02培養基中在30℃、500rpm、1.5vvm、pH 5-8之培養環境條件下進行培養。
因此,如表2中所示,證實CD01-1821品系更適用於按比例擴大方法,因為該品系在相同醱酵條件下具有高於CD01-1822品系之總生物質產量及粗脂肪量。因此,選擇CD01-1821品系且將其用於品系序列鑑別及額外品系研發。使用光學顯微鏡觀測所選擇之CD01-1821品系之類型且展示於圖3中。
實施例3.複合碳源條件下CD01-1821品系之培養特徵的確認
在基於異養微生物之醱酵中,葡萄糖組分主要用作碳源之原料。此時,葡萄糖為呈90%或更高之精製形式的單醣,且在工業規模醱酵期間產生的成本高於其他碳源原料組分。為了利用廉價碳源原料且經由其獲得價格競爭力,重要的是發現一種可用於藉由微生物醱酵且可通常自廉價碳源組分而非純化葡萄糖培養的品系。
因此,針對實施例2中所選擇之CD01-1821品系及CJM01(KR 10-2100650 B1)品系在具有葡萄糖、果糖或蔗糖作為主要組分之粗糖中進行醱酵培養評估以確認培養特徵。在30L培養基中進行培養,且基於經改良之MJW02培養基,分別用包含以下者之粗糖溶胞產物進行實驗:葡萄糖450g/L;葡萄糖225g/L及果糖225g/L之混合物;葡萄糖225g/L、果糖220g/L及硫酸鹽1.51g/L。培養條件設定為與30℃、500rpm、1.5vvm、pH 5-8之條件相同,且分別供應總培養溶液之35%的碳源。
因此,如表3中所示,CD01-1821品系展示在醱酵中在果糖混合物或粗糖溶胞產物培養基條件而非葡萄糖單組分下在等同或更高含量下之總生物質產量及粗脂肪量。另一方面,當培養基中添加除葡萄糖組分以外之糖組分時,CJM01品系展示雙重碳源消耗及細胞生長模式之雙期生長形式,且展示總培養時間延長之現象。經由對應的實驗結果,可確認CD01-1821品系在複合碳源條件下具有按比例擴大醱酵之可能性。
實施例4.新穎裂殖壺菌屬物種品系CD01-1821之鑑別
對於實施例1及實施例2中之經分離及選擇之微藻品系CD01-1821之生物分子鑑別,分析18S rRNA基因序列。
特定言之,在自純的經分離微藻CD01-1821之群落萃取及分離gDNA之後,使用引子18s-Fwd、LABY-ARev進行PCR擴增反應以用於表4中所述之18s rRNA區域之基因擴增。
對於PCR反應,使用含有taq聚合酶之反應溶液,在95℃下進行變性5分鐘;且接著在95℃下變性30秒、在50℃下黏合30秒且在72℃下聚合2分鐘,重複35次;且接著在72℃下進行聚合反應5分鐘。經由PCR方法擴增之反應溶液在1%瓊脂糖凝膠中進行電泳,且藉此,確認約1000bp大小之DNA片段經擴增,且進行核苷酸序列定序分析。作為分析之結果,經確認,經由NCBI BLAST搜尋,
所獲得之對應序列展示與屬於破囊壺菌科(family)系列微藻之蛞蝓形裂殖壺菌(Schizochytrium limacinum)品系OUC109之18S rRNA基因序列的95.11%同源性,且展示與裂殖壺菌屬物種之裂殖壺菌屬物種品系LY-2012之18S rRNA基因序列的95.0%同源性。經由此,確認經分離之微藻CD01-1821為新穎裂殖壺菌屬物種品系,且命名為裂殖壺菌屬物種CD01-1821品系,且於2021年8月23日寄存於韓國生物科學與生物技術研究所之韓國菌種保藏中心(KCTC)且指定為寄存編號KCTC14660BP。
實施例5. CD01-1821品系培養溶液樣品之粗蛋白質含量之分析
為分析裂殖壺菌屬物種CD01-1821品系培養溶液樣品中之粗蛋白質含量,使用如下方法。
特定言之,精確量測相當於約20至30mg之乾燥微生物細胞,亦即各醱酵溶液乾燥產物(試樣)且置於分解管中,且添加兩種分解促進劑。當硫酸(H2SO4)與硫酸鉀(K2SO4)之比率為1.4至2.0:1.0時,分解促進劑被有效地分解。接著,將濃硫酸(H2SO4)12至15mL添加至分解管,且當藉由在420℃下在分解裝置中分解45至60分鐘,分解溶液之顏色呈透明淺綠色(使用銅促進劑)或透明黃色(使用硒催化劑)時,將其冷卻至室溫。冷卻後,將80mL蒸餾水添加至經分解測試溶液中。在將與混合指示物混合之25mL收集溶液添加至錐形瓶(Erlenmeyer flask)中之後,將其置於蒸餾設備上,且抬升錐形瓶支架以使得蒸餾溶液在蒸餾期間進入收集溶液。將50mL氫氧化鈉溶液(NaOH)(量對應於在分解期間所使用之硫酸的4倍)添加至分解管,且將其在蒸餾設備中蒸餾3至4分鐘。經確認,蒸餾設備的錐形瓶中的收集溶液變成綠色同時收集蒸餾溶液中所包含的氨(NH3)。使用鹽酸溶液(通常0.1N或0.2N)滴定蒸餾溶液直至終點達到淡粉色,且記錄用於滴定之酸的量。在自動裝置之情況下,蒸餾、滴定及計算均自動進行。使用實驗結果,藉由以下計算公式2推導出氮%。藉由將先前推導出之
氮%乘以6.25之平均氮係數來表達蛋白質定量。

*硼酸溶液:藉由添加H3BO3 100g(或400g)、0.1%溴甲酚綠色溶液100mL及0.1%甲基紅溶液100mL製備之恆定體積為10L之1%(或4%)硼酸溶液
對於胺基酸含量分析,自CD01-1821及CD01-2147品系之培養溶液收集約1g之醱酵的溶液乾燥產物(試樣)樣品。在利用6N濃度之HCl溶液對胞內蛋白質進行酸水解之後,將其稀釋且用蒸餾水過濾,且進行液相層析。
因此,如表5中所示,經確認,CD01-1821乾燥微生物細胞中之粗
蛋白質含量為17%。另外,在乾燥微生物細胞中之胺基酸含量中,麩胺酸最高,且其次為酪胺酸、丙胺酸、苯丙胺酸、甘胺酸、精胺酸、絲胺酸、纈胺酸、離胺酸、天冬胺酸、甲硫胺酸、異白胺酸及白胺酸。經由此,確認CD01-1821乾燥微生物細胞中之胺基酸係由麩胺酸、酪胺酸、丙胺酸、苯丙胺酸、甘胺酸、精胺酸、絲胺酸、纈胺酸、離胺酸、天冬胺酸、甲硫胺酸、異白胺酸及白胺酸構成。

- 一種新穎裂殖壺菌屬物種(Schizochytrium sp.)CD01-1821微藻(寄存編號KCTC14660BP)。
- 如請求項1之裂殖壺菌屬物種微藻,其中該裂殖壺菌屬物種CD01-1821微藻具有生產ω-3不飽和脂肪酸之能力。
- 如請求項2之裂殖壺菌屬物種微藻,其中該ω-3不飽和脂肪酸為二十二碳六烯酸(docosahexaenoic acid,DHA)及二十碳五烯酸(eicosapentaenoic acid,EPA)。
- 如請求項3之裂殖壺菌屬物種微藻,其中該微藻產生以該脂肪酸之總重量計35重量%至60重量%的二十二碳六烯酸。
- 如請求項3之裂殖壺菌屬物種微藻,其中該微藻產生以該脂肪酸之總重量計0.1重量%至2重量%之二十碳五烯酸。
- 一種衍生自裂殖壺菌屬物種微藻之生物質,其包含如請求項1之裂殖壺菌屬物種微藻、該微藻之培養物、該培養物之乾燥產物或該乾燥產物之溶胞產物。
- 一種組成物,其包含如請求項6之衍生自裂殖壺菌屬物種微藻之生物質、該生物質之濃縮物或乾燥產物或該生物質之萃取物。
