(5)プロピオン酸菌が、プロピオニバクテリウム・フロイデンライヒ ET−3株である、(1)〜(3)のいずれかに記載の組成物。
(6)食品組成物または医薬組成物である、(1)〜(5)のいずれかに記載の組成物。
(7)抗肥満用である、(1)〜(6)のいずれかに記載の組成物。

プロピオニバクテリウム・フロイデンライヒET−3株は平成13年8月9日付で、独立行政法人産業技術総合研究所特許生物寄託センター(日本国茨城県つくば市東1丁目1番地1中央第6)に寄託され、その後、国際寄託に移管され受託番号FERM BP−8115が付与されている。なお、Budapest Notification No. 282 (http://www.wipo.int/treaties/en/notifications/budapest/treaty_budapest_282.html)に記載されるとおり、独立行政法人製品評価技術基盤機構(IPOD、NITE)が独立行政法人 産業技術総合研究所 特許生物寄託センター(IPOD、AIST)から特許微生物寄託業務を承継したため、プロピオニバクテリウム・フロイデンライヒET−3株は、現在は、独立行政法人製品評価技術基盤機構(IPOD、NITE)(千葉県木更津市かずさ鎌足2−5−8 120号室)に受託番号:FERM BP−8115のもとで寄託されている。
(1)試験方法
雄性で8週齢のC57BL/6Jマウスを5群に分け、通常食(AIN−93M)(オリエンタル酵母工業株式会社製)あるいは高脂肪食(HFD−60)(オリエンタル酵母工業株式会社製)を与えた。高脂肪食を与えた群には、フラクトオリゴ糖(以下、「FOS」とも記載する)、プロピオン酸菌(Propionibacterium freudenreichii ET−3株、受託番号:FERM BP−8115、以下、「P菌」とも記載する)、あるいはFOSおよびプロピオン酸菌(以下、「FOS+P菌」とも記載する)の両方を投与して8週間飼育した。なお、高脂肪食を与えるが、FOSおよびプロピオン酸菌を投与しない群をコントロール群とした。FOSにはメイオリゴ(商標)P(株式会社 明治フードマテリア製)を使用し、高脂肪食中に5質量%含有させて投与した。プロピオン酸菌は、GAMブイヨン(日水製薬株式会社製)で賦活培養した菌液を生理食塩水で洗浄後、5×109cfu/mLに菌数を調整した。この菌液を、1日1回0.2mLの用量で、試験開始日から終了日まで毎日強制経口投与した。なお、プロピオン酸菌を投与していない群(通常食群、コントロール群およびFOS投与群)には、生理食塩水を1日1回0.2mLの用量で、試験開始日から終了日まで毎日強制経口投与した。試験期間中の体重、解剖時の精巣周囲脂肪重量および試験終了日に採取した糞便中のプロピオン酸濃度を測定し、体重増加量を試験開始時と終了時の体重から算出して評価に用いた。

注入量:10μ
通常食群と高脂肪食を投与した4群との比較では、高脂肪食を投与した4群で明らかな体重増加が確認された(図1参照)。高脂肪食を投与した4つの群では、プロピオン酸菌投与群で6週目以降の体重がコントロール群より低値を示し、FOSとプロピオン酸菌の両方を投与した群においてこれらの群の中で最も低値を示した。また、体重増加量も、FOS投与群やプロピオン酸菌投与群でも低値を示しているものの、FOSおよびプロピオン酸菌を投与した群(FOS+P菌)で最もコントロール群との差が大きく、FOSとプロピオン酸菌とを同時に摂取することで、体重の増加が抑制されることが分かった(図2参照)。
