Abstract
Description
本発明は、部分的に、米国国立衛生研究所(NIH)と米国国立神経疾患・脳卒中研究所(NINDS)のNIH/NINDS助成金第NS052671号及びNIH/NINDS助成金第P50 NS047085号の下の政府支援を以て行われた。従って、米国政府は本発明にある特定の権利を有する。
本発明は幹細胞生物学の分野、とりわけ万能性幹細胞又は多能性幹細胞の系譜特異的分化に関連し、それらの幹細胞には非胚性ヒト人工万能性幹細胞(hiPSC)に加えてヒト胚性幹細胞(hESC)、体性幹細胞、疾患を有する患者に由来する幹細胞、又は系譜特異的分化が可能である他のあらゆる細胞が含まれ得るが、これらに限定されない。新規の培養条件を用いて底板中脳始原細胞への、その後さらに、大集団の中脳運命FOXA2+LMX1A+TH+ドーパミン(DA)ニューロンへのhESC及び/又はhiPSCの系譜特異的分化を制御する方法が具体的に記載される。本発明の方法を用いて作製された中脳運命FOXA2+LMX1A+TH+ドーパミン(DA)ニューロンには、インビトロ創薬アッセイにおける使用、神経生物学研究における使用、及び患者におけるドーパミンニューロンの喪失による疾患又は障害を回復させる治療薬としての使用を含むが、これらに限定されない様々な使用法がさらに企図される。さらに、疾患、特にパーキンソン病のモデル化に使用するため、ヒト万能性幹細胞から中脳運命FOXA2+LMX1A+TH+ドーパミン(DA)ニューロンを分化させるための組成物と方法が提供される。加えて、真正DAニューロンはCD142などのマーカー及びA9型神経細胞に富む。
本明細書において使用される場合、「疾患のモデル化」という用語は、障害の結果としてヒトにおいて観察される特定の兆候又は症状を模倣する、実験生物又はインビトロ細胞培養物を用いる方法を指す。1つの実施形態では、神経障害、例えばパーキンソン病(PD)を引き起こす遺伝的変異を有する動物モデルから得られた万能性幹細胞を培養し、PDに関連するニューロンの新しい特徴を特定するために神経細胞に分化させることができる。1つの実施形態では、神経障害、例えばパーキンソン病(PD)を引き起こす遺伝的変異を有する個人から得られたヒト万能性幹細胞を培養し、その個人に観察されたのと同様の欠損を保持する神経細胞に分化させることができる。

次の実施例は本発明の開発中に使用された神経系譜細胞を提供する例となる方法を説明する。
本発明者らは、DA様ニューロンを含有する細胞集団を生じることになるいくつかの他の制御分化方法をこれまでに使用した。これらのDA様ニューロンを移植試験において使用し、治療用途にこれらの細胞をさらに使用することに対する懸念が生じた。例えば、MS5神経誘導を含む、Perrier et al., 2004及び Fasano et al., 2010に記載される方法がロゼット細胞形成を引き起こし、そして、それらの方法が第11日の前駆細胞(例えば、図2、16及び17を参照のこと)を作製するために用いられ、DA様ニューロンを得るためにさらに用いられた。これらのニューロンは、生じた第11日の細胞集団中の低パーセンテージの前駆細胞より生じた。これらのニューロンを使用した移植試験は、テラトーマの発生を引き起こす増殖制御の喪失と共に不適切な神経種の移植後発生が観察されたことに加えて、低い移植後生存率とDA様ニューロンの表現型の喪失を示した。図16及び17を参照のこと。
次の実施例は、小分子候補化合物をスクリーニングするための節Iの例となる細胞の使用、及び、それらの化合物の使用が二重SMAD阻害剤との最初の接触から第11日までに高パーセンテージの中脳底板ニューロンを含有する細胞集団の制御分化を引き起こすかの判定を説明する。このスクリーンの結果は、SHH活性化分子FGF8及びWntの活性化と共に分化第11日までにhESCからFOXA2+/LMX1A+陽性中脳底板細胞の効率的な誘導を引き起こすことを最初に示した。本発明者らは、第11日において所望のFOXA2+/LMX1A+陽性細胞集団を誘導するために必要である分子、及び最適接触時間を明らかにする例となる実験の結果を本明細書において示す。
パーキンソン病(PD)は2番目に最も一般的な神経変性障害であり、世界中で410万〜460万人の患者を冒していると推定され、人数は2030年までに2倍を超えると予想されている。パーキンソン病はアルツハイマー病の後に2番目に最も一般的な神経変性障害であり、米国では約100万人の患者を冒しており、毎年60,000人の新しい患者がパーキンソン病と診断される。その疾患は著しい罹患率と死亡率をもたらすので大きい社会経済的影響を有し、そして、医療費と所得の損失を含むPDの直接的及び間接的損失の合計は米国だけで毎年約250億ドルと推定される。現在のところ加齢に関連し、進行性であり、そして、無力化する障害であるパーキンソン病(PD)の治療法は存在しない。PDは黒質における中脳DAニューロンの選択的喪失を病理学的な特徴とする。従って、PDの基礎的な特徴は、最終的には無力化する運動機能不全を引き起こす中脳ドーパミン(DA)ニューロンの進行性であり、重篤で不可逆的な喪失である。薬理学的療法、運動系療法、遺伝子療法及び外科的療法がPDに向けて開発されてきたが、それらのアプローチの中で適切なDAニューロン機能を回復させることができるものは未だ無い。患者における運動症状の長期制御は多くの場合最適以下で留まり、そして、進行性非ドーパミン応答性運動症状及び非運動症状の重要性を認識している一方で、長期ドーパミン応答性症状制御の基礎的問題は重大な治療必要性のある領域のままである。中枢と末梢の両方神経系を冒す広範囲に及ぶ病態がPDにおいて認識されており、PDの基本的特徴(部分的には運動緩慢、強直、及び身震い)は基本的にDA神経細胞の喪失に関連し、そして、ドーパミン応答性である。従って、PDには、その疾患の大半の運動症状の原因である中脳DAニューロンのどちらかと言えば選択的な喪失のため、神経細胞置換を用いる治療が企図される。健康なヒトの脳は約100万のDAニューロンを有する。従って、1つの実施形態では、CNSにおける大半の他の障害と比べて比較的に少数の生存細胞を必要とするDAニューロン置換が企図される。
本発明者らは、CHIR99021曝露のタイミングがFOXA2/LMX1A中脳底板前駆細胞の誘導を決定することを本発明の開発中に発見した。従って、本発明者らは、LSB/S/F8処理(すなわち、LSB、S、すなわちSHH、及びFGF8(F8)での細胞の処理)のみ又は様々な時点で開始するCHIRと組み合わせた、すなわち、第0日〜第11日、第1日〜第11日、第3日〜第11日、第5日〜第11日、第7日〜第11日の処理の後の分化第11日においてFOXA2/LMX1Aの免疫細胞化学分析について試験し、CHIR処理が無い細胞の2つ組の培養物と比較した。その後、FOXA2+細胞、LMX1A+細胞及び二重標識細胞のパーセンテージの定量を、免疫細胞化学分析において記載されるようにCHIR曝露の差次的開始後の分化第11日に行った。
さらなる分化のためにニューロン成熟を促進する培地(BAGCT、実施例Iを参照のこと)中に前駆細胞FOXA2+/LMX1A+細胞を維持した。比較のために2つの他の技術を用いてDAニューロン前駆細胞を作製した。これまでの方法と本発明の方法から生じる分化細胞の集団の間で次の種類の比較を行った:A)分化第50日におけるLMX1A、FOXA2及びNURR1と組み合わせたTHについての免疫細胞化学分析、B)ロゼット由来培養物と底板由来(LSB/S/F8/CHIR)培養物を比較する総細胞の中のTH+細胞、FOXA2+細胞、LMX1+細胞、及びNURR1+細胞の定量。(C)底板DAニューロン培養物及びロゼット由来DAニューロン培養物における第50日でのセロトニン+(5‐HT)神経細胞サブタイプとGABA+神経細胞サブタイプ(非DAニューロン混入細胞)のパーセンテージの定量。そして、(D)ドーパミンと代謝物を測定するためのHPLC分析:底板由来培養物とロゼット由来培養物の間のDAレベル、DOPACレベル及びHVAレベルの比較。
次の実施例は治療的細胞置換に使用するための本発明の例となる方法の使用について説明する。その分野における1つの主要な課題は、神経過形成又は非中脳ニューロンへの不適切な分化のリスクが無い状態で機能的にインビボにおいて生着するhPSC由来中脳DAニューロンを作製する、又はテラトーマを発生させる能力である。胎児組織移植試験に基づき、NURR1の発現によって示される細胞周期離脱の時が移植に適切な期間であり得ると本発明者らは考えた(およそ分化第25日、図2)。非傷害成体マウスの第25日細胞を使用する最初の試験によって、移植後6週間でhPSC由来FOXA2+/TH+ニューロンの堅固な生存が示された(図12)。パーキンソン病の宿主における、神経過形成を引き起こすことがない、FOXA2+/TH+細胞の長期生存が試験された。この目的のため、まれな腫瘍原性細胞の曝露に特定の感受性を有する、異種移植片の生存を効率的に支援する株(Quintana, et al. Efficient tumour formation by single human melanoma cells. Nature 456:593〜598 (2008)、参照により本明細書中に援用)であるNOD‐SCID IL2Rgcヌルマウスにおいて6‐ヒドロキシ‐ドーパミン(6‐OHDA)傷害(Tabar, et al. Nature Med. 14:379〜381 (2008)、参照により本明細書中に援用)を行った。高い増殖能を有する細胞の混入の可能性を示すために事前精製することなく底板由来DAニューロン培養物とロゼット由来DAニューロン培養物の両方を移植した(150×103細胞/動物)。移植から4.5か月の後に底板由来DAニューロン移植片がFOXA2共発現性TH+細胞から構成される輪郭のはっきりした移植中核とヒト特異的マーカーhNCAMを示した(図4a〜c)。機能分析によってアンフェタミン誘導性回転行動の完全な救出が示された。対照的に、ロゼット由来ニューロン移植片はTH+ニューロンをほとんど示さず、回転行動の有意な減少をもたらさず(図4d)、そして、大規模な神経過形成を示した(20mm3超の移植片体積;図13)。本明細書において報告される、移植に使用されたロゼット由来神経細胞の広範囲の過形成は本発明者らのグループ(Kim, et al. miR−371−3 Expression Predicts Neural Differentiation Propensity in Human Pluripotent Stem Cells. Cell Stem Cell 8:695〜706 (2011)、参照により本明細書中に援用)及び他のグループ(Hargus, et al. Proceedings of the National Academy of Sciences of the United States of America 107:15921〜15926 (2010)、参照により本明細書中に援用)のロゼット由来DA移植片を用いるこれまでの研究に匹敵した。その過形成はより長い生存期間(4.5か月対6週間)、移植前のFACS精製の欠如、及びNOD‐SCID IL2Rgcヌル宿主の選択に起因する可能性があった。増殖性のKi67+細胞の数は底板由来移植片では最小であった(総細胞の1%未満)が、ロゼット由来移植片は増殖性神経前駆細胞からなるポケットを保持した。神経過形成は移植片内の初期前方神経外胚葉性細胞によって引き起こされると考えられている(Elkabetz, et al. Genes Dev. 22:152〜165 (2008); Aubry, et al. Proc. Natl. Acad. Sci. U. SA 105:16707〜16712 (2008)、参照により本明細書中に援用)。この仮説は底板由来移植片ではなくロゼット由来移植片での前脳マーカーFOXG1の発現によって裏付けられた。底板由来移植片とロゼット由来移植片の両方に小パーセンテージのアストログリア細胞が存在したが、大半のGFAP+細胞は宿主起源を示すヒトマーカーについて陰性であった(図13)。
次の実施例は、本明細書に記載される方法による分化により生じる中脳DAニューロンの機能的能力を決定するための本発明の例となる方法の使用について説明する。黒質緻密部(SNpc)DAニューロンは脳における他の大半のニューロンからそれらを区別する電気生理学的表現型をインビボで示す。具体的には、それらのニューロンは低速で(1〜3Hz)突発的に電位上昇を示す。また、この低速の電位上昇は低速の閾値下振動電位を伴う。インビトロで2〜3週間の後、生後初期マウスから培養されたSNpc DAニューロンがこれらの同じ生理的特徴を示す。本発明のmDAニューロンが同等の電気生理学的表現型を示したか判定するため、本発明のmDAニューロンの電気的応答シグネチャについてそれらを試験した。培養第80日〜第100日の本発明の中脳DAニューロンを単一細胞レコーディングにより試験した。hESCから分化したこれらのmDAニューロンが、特定の自律的内部標準添加行動及び振動性膜電位変化を示すことによって一貫して(4/4試験)この特徴的な生理的表現型を示した(図3g〜i)。この挙動は自律的ペースメーカー活性として知られ、そして、中脳DAニューロン及び特にパーキンソン病に最も関係があるサブタイプの中脳ドーパミンニューロン(黒質型中脳DAニューロン)の特異的な特性である。
本明細書における記載は、本明細書に記載される組成物と方法による分化により生じるmDA神経細胞の大規模生産のための例となる方法と使用を示す。規模拡大可能な作成法(すなわち、比較的少数の細胞を含有する培養物からのmDA神経細胞の作製に成功すると考えられている方法)が第25日において移植可能なmDAニューロンが可能である細胞集団を産出することが示された。とりわけ、PINK iPSC細胞について表9を参照のこと)。
(複数の)問題を克服することによって、例えば、混入万能性幹細胞を含むが、これらに限定されない混入細胞集団を除去することによって中脳DAニューロン前駆細胞について細胞集団を濃縮することを目標に、本発明の開発前及び開発中にいくつかの方法を開発及び試験した。初代の胚性マウスニューロン集団、胚性ラットニューロン集団及びマウスESC由来集団を使用して最初の試験を実施した。DAニューロン濃縮に使用するための神経集団の増加という目標を有することに加えて、そのマウスESC試験は以前の方法で問題であったテラトーマの形成を防止する方法の開発を含んだ。これらの戦略には神経マーカー(NCAM)を発現する細胞の陽性選択と共に万能性細胞で発現される細胞表面マーカー(例えば、SSEA1)についての陰性選択が含まれた。マウス細胞を使用して、DAニューロン移植パラダイムにおける神経細胞の濃縮の特定(例えば、SOX1、コリン(Corin)/Lmx1a、Ngn2、TH、Pitx又はDATの発現を有する細胞の特定)において使用されるいくつかの遺伝的レポーター戦略が提唱された。機能試験がNgn2レポーターマウス由来の初代細胞(Thomposon et al., Exp Neurol. 198(1):183〜98 (2006))及びコリンについても選別されたLmx1Aレポーターマウス由来の初代細胞(Jonsson, Exp Neurol. 219(1):341〜54 (2009))において実施された。マウスESC由来集団についてはSOX1(Barraud et al., Eur J Neurosci. 2005 22(7):1555〜69)、TH(Kelly et al., Minerva Endocrinol. 1991 16(4):203〜6)、Pitx3(Hedlund et al., Stem Cells. 2008 26(6):1526〜36)及びDAT(Zhou et al., Stem Cells. 2009 27(12):2952〜61)マウスESCレポーター株を使用して試験が実施された。加えて、本発明者らは本発明の開発中にDAニューロン発生の連続するステージを代表する3種の精製されたマウスESC由来集団、すなわち、中脳前駆細胞(Hes5::GFP)、初期分裂終了後細胞(Nurr1::GFP)、及び成熟DAニューロン(Pitx3::YFP)のインビボ成績を直接比較する包括的な移植試験を実施した。それらの試験はNurr1発現性DA発生を移植に特に適切なものと特定し、そして、精製されたDAニューロンはインビボで効率的な生着が可能であることを示した。さらに、これらの結果は、PDのマウスモデル、ラットモデル及びアカゲザルモデルへのhESC‐DAニューロンの移植に使用するための分化第25日(Nurr1発現の開始日)における細胞の選択のために使用された。
本発明の開発中に収集された細胞表面マーカー発現データは中脳DAニューロンで発現されるいくつかの新規細胞表面マーカーの特定を示した。具体的には、細胞、例えばDAニューロン、本発明の成熟DAニューロン及びA9細胞に成熟するだろう特定の細胞をさらに特定するためのマーカーが見出された。2つの主要な戦略がそのような表面マーカーを特定するために用いられた。第一に遺伝的レポーター株における無作為遺伝子発現スクリーン(図27a)がCD142、及び中脳DAニューロンで選択的に発現し、A9型DAニューロンを特異的に標識するように見えるDCSM1という名前のマーカー(図27b)を含むいくつかの候補マーカーを示した。第2の戦略は、96ウェル形式の242種の市販の抗体を試験するhESC由来DAニューロンにおけるCD細胞表面マーカースクリーンの使用であった(図27c、d)。そのような例となるスクリーンの結果(図27e)によって、CD63、CD99、及びDCSM1に加えてNurr1+DAニューロン期を選択的に標識したマーカー(図27f)であるCD142を含む、中脳DAニューロンにおいて濃縮される少なくとも5個の検証済みのマーカーが特定された。
移植片の組み込みとDA線維伸長の程度はPD患者における胎児移植試験を含む移植方法における課題である。移植片組織及び細胞が出会う1つの問題は患者を治療するときのこれらの移植片からの線維伸長の制限である。この問題は、患者の回復が広範囲の線条体神経再支配を必要とするのでヒトにおいて特に重大である。以前の方法では、組織移植後の充分な神経再支配の達成には線条体のいたる所への細胞供託物の複数回の注入が必要とされた。注入毎に他の外科的リスクと共に線条体の損傷と炎症が引き起こされ得る。そのようなリスクには、患者において卒中又はけいれんを引き起こす可能性があるだろう細胞注入の間の血管の損傷が含まれる。PSAは、(ポリシアル酸化)神経細胞接着分子(NCAM)、NCAM(CD56)、及び同種のものなど、他の細胞表面分子の(ポリシアル酸トランスフェラーゼ(PST)酵素の作用による)翻訳後修飾として特定された天然細胞表面シアル酸ホモ重合体(すなわち、α2,8‐結合シアル酸)である。PSAは、細胞移動と軸索伸長を含む、細胞間相互作用の変化を必要とするいくつかの細胞行動の可塑性の調節に機能するようであった。PSAは、胚においてよく発現されるが、構造的及び生理的可塑性を維持するCNSの局所領域(例えば、海馬、視交叉上核、SVZ)を例外として成体の組織では下方制御された。従って、いくつかの実施形態では、移植された細胞の線維伸長の促進にポリシアル酸(PSA)を使用することが企図された。他の細胞種におけるPSAの使用の例は、全体の参照により本明細書中に援用される国際公開第2006/042105号に記載されている。いくつかの実施形態では、本発明者らは本明細書に記載されるPSAと組み合わせた真正DAニューロンの使用を企図する。
被傷害動物群は野生型細胞又はPSTを発現する細胞又はPST酵素で前処理された細胞からなる3つの用量のうちの1つを受容した。それらの動物群は表11で考察されるパラダイム(アンフェタミン回転、円筒試験、ステッピング試験)に従う行動試験のために処理された。結果生じるニューロンがマウスPST遺伝子について本明細書に記載される結果を示すように、選択された用量(例えば、200,000個、100,000個、50,000個の細胞)をマウスで使用することに成功した。
本発明の細胞作製方法における使用のため、患者への健康上のリスクをさらに減少させるために考えられた実施形態が下に記載される。
好ましい実施形態では、ヒトにおける移植方法、言い換えると、PDの治療のための細胞療法のために細胞を作製し、使用するための方法にヒトESCを使用することが考えられる。特に、本発明の方法においてヒトESCはヒトiPSCの使用よりも多数の利点を有し、一例ではPD細胞療法として使用される移植可能中脳DAニューロンの供給に使用される。特に、DAニューロン誘導のための細胞供給源としての人工万能性幹細胞(iPSC)の使用は、各患者にとって遺伝的に適合した細胞供給源を供給することなど、いくつかの利点を有する。しかしながら、多数の近年の研究により、iPSCの安全性と完全な遺伝的適合性に関する不確実性が生み出された。再プログラム化細胞は、臨床的利用に望ましくない危険な可能性がある遺伝的異常及びエピジェネティックな異常を有することが示されている。さらに、マウスiPSCにおける研究は、iPSC由来細胞が、それらの細胞のヒト移植における使用を後押しする主要な言い分である、完全に免疫適合性であるわけではないことを示した。さらに、移植にiPSCを使用するFDAが認可した方法は存在しない。そして、それぞれ個々の患者のためにGMPに準拠し、QC管理された細胞バンクを作製することを考えることは実際的ではないし、桁違いの費用がかかるだろう。比較すると、hESCと比べたhiPSCの遺伝的安定性が集中的に研究された。hESCは培養状態で時間が経つにつれ変異を獲得することが観察されたが、そのような変異のタイミングと率はhiPSCとは異なるように見え、hESCは概してiPSCよりも遺伝的に安定していると考えられた。例えば、Hussein, et al., Nature 471, 58〜62 (2011); Mayshar, et al. Cell Stem Cell 7, 521〜531 (2010); Lister, et al. Nature 471, 68−73 (2011); Laurent, et al. Cell Stem Cell 8, 106〜118 (2011)を参照のこと。加えて、細胞療法製品及び遺伝子療法製品についてFDAによって要求される厳密な安全性試験を満足させるいくつかのhESC株について標準操作手順が考え出された。FDAはhESC−ベースの細胞療法を臨床使用に進めるように米国の2つのグループに認可した。例えば、ゲロン(Geron)社はhESC由来乏突起膠細胞前駆細胞(GRNOPC1)を用いる第I相試験に入っていた。アドバンスド・セル・テクノロジー(ACT)社はシュタルガルト黄斑ジストロフィー(治験番号NCT01345006)及び乾燥進行性加齢黄斑変性(治験番号NCT01344993)を治療するためにhESC由来網膜色素上皮細胞を使用する、現在行われている2つの第I相/II相試験を有する。

方法の概要:ヒトESC(H9、H1)及びiPSC株(2C6及びSeV6)を改変二重SMAD阻害(Chambers, et al. Nat. Biotechnol. 27:275〜280 (2009)、参照により本明細書中に援用)ベース底板誘導(Fasano, et al., Cell Stem Cell 6:336〜347 (2010)、参照により本明細書中に援用)プロトコルの対象とした。中脳底板のため、及びDAニューロンの新規集団の産出のためにSHH C25II、パルモルファミン、FGF8及びCHIR99021への曝露を最適化した(図1dを参照のこと)。底板誘導後、AA、BDNF、GDNF、TGFβ3及びdbcAMPなどのDAニューロン生存及び成熟因子(Perrier, et al. Proc Natl Acad Sci USA 101:12543〜8 (2004)、参照により本明細書中に援用)の存在下でNeurobasal/B27系の分化培地でさらなる成熟(第11〜25日、又は25日より長く培養状態であり、最大で少なくとも100日培養状態である)を実施した(詳細について方法全体を参照のこと)。結果生じるDAニューロン集団を免疫細胞化学、qRT‐PCR、包括的遺伝子発現プロファイリング、ドーパミンの検出のためのHPLC分析及びインビトロ電気生理学的レコーディングによる詳細な表現型特徴解析の対象とした。半身パーキンソン症げっ歯類動物(動物の脳の一方の側に6OHDA毒素を注入したマウス又はラット)でインビボ試験を実施した。6‐ヒドロキシドーパミン傷害を前に記載されたようにその毒素の定位的注入により受けた成体NOD‐SCID IL2Rgcマウス(ジャクソン・ラブズ社)及び成体スプレイグ・ダウリー・ラット(タコニック・ファームス社)、並びにMPTPの片側のみの頸動脈注射により処置された2匹の成体アカゲザルにおいてそれらの試験を実施した。DAニューロンはそれらの動物の線条体中に定位的に注入され(マウスでは150×103細胞、ラットでは250×103細胞)、そして、サルでは総計7.5×106細胞(6路に供給;脳の各側に3路)。アンフェタミン介在性回転分析並びに焦点性無動症及び四肢使用についての試験(「ステッピング試験」及び「円筒試験」)を含む行動アッセイを移植後に月単位の間隔で実施した。ラットとマウスを移植後の18〜20週間で、及びそれらの霊長類動物を移植後の1か月で殺処理した。それらの移植片の特徴解析を細胞数と移植片体積の立体解析学的分析により、並びに表現型の包括的特徴解析を免疫組織化学により実施した。未分化ヒトES細胞の培養.hESC株H9(WA‐09、XX、2009年10月の時点より27〜55継代)、H1(WA‐01、XY、2010年6月の時点より30〜40継代)並びにiPS細胞株2C6(Kim, et al. Cell Stem Cell 8:695〜706(2011)、参照により本明細書中に援用)(XY、20〜30継代)及びSeV6(XY、20〜30継代;非挿入性4因子センダイ・ベクター・システム(Ban, et al. Proc. Natl. Acad. Sci. U.S.A(2011)108(34):14234〜14239:10.1073/pnas.1103509108、参照により本明細書中に援用)を使用するMRC‐5胚性線維芽細胞由来)を、ヒトES細胞に基づいてcm2当たり0.5×103個からcm2当たり100×103個までの範囲に推定される播種濃度で、至適20%ノックアウト血清代替物(KSR、インビトロジェン社、カリフォルニア州、カールスバッド)含有ヒトES細胞培地(前に記載された(Kim, et al. Cell Stem Cell 8:695〜706 (2011)、参照により本明細書中に援用)において細胞をクラスター化する傾向があるマウス胚性線維芽細胞(MEF、グローバル・ステム社、メリーランド州、ロックビル)上に維持した。ノックアウト血清代替物の使用は0%から40%までの範囲にあり得る。
Claims (20)
- LDN‐193189及びCHIR99021と接触する細胞集団であって、その40%超がフォークヘッドボックスタンパク質A2(FOXA2)について陽性である前記細胞集団であり、LDN‐193189、SB431542、ソニックヘッジホッグ(SHH)シグナル伝達活性化剤及びCHIR99021と以前に接触している前記細胞集団であり、前記ソニックヘッジホッグ(SHH)シグナル伝達活性化剤が、ソニックヘッジホッグ(SHH)C25II及びパルモルファミンからなる群より選択される、前記細胞集団を含む組成物。
- 前記細胞集団の10%超が、フォークヘッドボックスタンパク質A2(FOXA2)/LIMホメオボックス転写因子1α(LMX1A)について二重陽性である細胞、及びフォークヘッドボックスタンパク質A2(FOXA2)/オルトデンティクル(orthodenticle)ホメオボックス2(OTX2)について二重陽性である細胞からなる群より選択される、請求項1に記載の組成物。
- 前記細胞集団の少なくとも20%がNurr1+、CD142、DCSM1、CD63及びCD99からなる群より選択されるマーカーについて陽性である、請求項1に記載の組成物。
- 前記細胞集団が、げっ歯類細胞、霊長類細胞、ヒト細胞及びパーキンソン病(PD)の神経学的症状を有する患者に由来するヒト細胞からなる群より選択される、請求項1に記載の組成物。
- 細胞の底板中脳始原細胞の集団への制御分化を誘導するための方法であって、
(a):(i)細胞集団であって、非胚性幹細胞、胚性幹細胞、人工非胚性万能性細胞及び遺伝子操作万能性細胞からなる群より選択される前記細胞集団;及び
(ii)第1シグナル伝達阻害剤、第2シグナル伝達阻害剤、ソニックヘッジホッグ(SHH)シグナル伝達の活性化剤及び第3シグナル伝達阻害剤であって、SB431542である前記第1阻害剤、LDN‐193189である前記第2阻害剤、ソニックヘッジホッグ(SHH)C25II及びパルモルファミンからなる群より選択される前記ソニックヘッジホッグ(SHH)シグナル伝達活性化剤、及びCHIR99021である前記第3阻害剤;を提供すること、
(b)前記第1阻害剤及び前記第2阻害剤と前記細胞集団を接触させることであって、前記第1阻害剤及び前記第2阻害剤との前記接触がインビトロでの細胞播種から48時間以内にあるように底板中脳始原細胞の前記分化集団を生じさせることができる条件下で前記第1阻害剤及び前記第2阻害剤と接触させること、
(c)前記第1阻害剤及び前記第2阻害剤と前記細胞集団を接触させた後に底板中脳始原細胞の集団を分化させるための条件下で前記細胞を前記ソニックヘッジホッグ(SHH)シグナル伝達活性化剤とさらに接触させること;及び
(d)前記細胞集団を前記ソニックヘッジホッグ(SHH)シグナル伝達活性化剤と接触させた後に前記細胞集団を底板中脳始原細胞の集団に分化させるために前記細胞を前記第3阻害剤とさらに接触させること
を備える方法。 - 前記細胞の前記ソニックヘッジホッグ(SHH)シグナル伝達活性化剤との前記接触が前記細胞集団の前記第1阻害剤及び前記第2阻害剤との接触後少なくとも24時間であり、最大で36時間までにあるように、前記細胞の前記ソニックヘッジホッグ(SHH)シグナル伝達活性化剤との前記接触が底板中脳始原細胞の前記分化集団を生じさせることができる条件下にある、請求項5に記載の方法。
- 前記細胞の前記第3阻害剤との前記接触が前記細胞集団の前記ソニックヘッジホッグ(SHH)シグナル伝達活性化剤との接触後少なくとも24時間であり、最大で36時間までにあるように、前記細胞の前記第3阻害剤との前記接触が底板中脳始原細胞の前記分化集団を生じさせることができる条件下にある、請求項5に記載の方法。
- 前記底板中脳始原細胞集団が40%を超えるフォークヘッドボックスタンパク質A2(FOXA2)+LIMホメオボックス転写因子1α(LMX1A)+細胞を含む、請求項5に記載の方法。
- 神経細胞成熟培地であって、底板中脳始原細胞の底板中脳ドーパミン(DA)ニューロンへの分化のためにN2培地、脳由来神経栄養因子(BDNF)、アスコルビン酸(AA)、グリア細胞株由来神経栄養因子、ジブチリルcAMP及びトランスフォーミング増殖因子タイプβ3を含む前記培地と底板中脳始原細胞の前記集団を接触させるステップe)をさらに備える、請求項5に記載の方法。

- 治療処置のためのインビボ移植方法であって、
(a):(i)底板中脳ドーパミン(DA)ニューロンの集団であって、前記集団の40%超がチロシンヒドロキシラーゼ(TH)を発現する前記集団;及び
(ii)少なくとも1つの神経学的症状であって、身震い、運動緩慢(過度に遅い動作)、屈曲姿勢、姿勢動揺及び強直からなる群より選択される前記神経学的症状を示す対象;を提供すること、及び
(b)ドーパミン(DA)神経機能を提供するためにインビボ移植を可能にする条件下で前記対象に前記底板中脳ドーパミン(DA)ニューロンを移植すること、
を備える方法。 - 前記対象が前記神経学的症状のうちの少なくとも1つの減少を示す、請求項16に記載の方法。
- 底板中脳ドーパミン(DA)ニューロンの前記集団が請求項11に従って処理された細胞の集団に由来する、請求項16に記載の方法。
