本明細書において「オリゴヌクレオチド」という用語は概ね、2つ以上の共有結合したヌクレオシドを含む分子として当業者に遍く理解されているものとして定義されている。そのような共有結合したヌクレオシドはまた、核酸分子又はオリゴマーでありうる。オリゴヌクレオチドは通例、実験室で(固相化学合成により)作製され、続いて精製される。オリゴヌクレオチドの配列に言及する場合、共有結合したヌクレオチドもしくはヌクレオシドの核酸塩基部分もしくはその修飾物の配列又は順序について説明する。本発明のオリゴヌクレオチドは人造、且つ化学的に合成されるものであり、精製又は単離されるのが一般的である。本発明のオリゴヌクレオチドは、1以上の修飾ヌクレオシド又はヌクレオチドを含む場合がある。
本明細書において、「アンチセンスオリゴヌクレオチド」という用語は、標的核酸、特に標的核酸上の連続配列にハイブリダイズすることによって標的遺伝子の発現を調節できるオリゴヌクレオチドとして定義される。アンチセンスオリゴヌクレオチドは、本質的に二本鎖ではなく、したがってsiRNAではない。本発明のアンチセンスオリゴヌクレオチドは一本鎖であるが、好ましい。

本発明によれば、標的核酸は、哺乳動物PD−L1をコードする核酸であり、例えば、遺伝子、RNA、mRNA、及びプレmRNA、成熟mRNA又はcDNA配列でありうる。したがって、標的は、PD−L1標的核酸と呼ばれる場合がある。本発明のオリゴヌクレオチドは、例えば、哺乳動物PD−L1の標的エキソン領域であってもよいし、又は例えば、PD−L1プレmRNAの標的イントロン領域であってもよい(表1を参照のこと)。
本明細書において「標的配列」という用語は、本発明のオリゴヌクレオチドに相補的な核酸塩基配列を含む、標的核酸中に存在するヌクレオチドの配列を指す。幾つかの実施形態において、標的配列は、本発明のオリゴヌクレオチドの連続ヌクレオチド配列に対して相補的な、標的核酸上の領域からなる。幾つかの実施形態において、標的配列は単一のオリゴヌクレオチドの相補的配列よりも長く、例えば、本発明の幾つかのオリゴヌクレオチドによって標的化できる標的核酸の好ましい領域を表しうる。

多数の単一ヌクレオチド多型がPD−L1遺伝子において公知であり、例えば、下の表に開示されている(ヒトプレmRNA開始/参照配列は、配列番号2である)。
本明細書において「発現の調節」という用語は、オリゴヌクレオチドの投与前のPD−L1の量と比較して、PD−L1の量を変化させるオリゴヌクレオチドの能力の包括的用語として理解すべきである。あるいは、発現の調節は、対照実験を参照して定量できる。対照は、非標的オリゴヌクレオチド(モック)で処理した生理食塩水組成物、個体もしくは標的細胞で処置した個体又は標的細胞であることが概ね理解されるが、また、ケア基準で処置される個体である場合もある。
高親和性修飾ヌクレオシドは、オリゴヌクレオチド内に取り込まれた場合に、その相補的標的に対するオリゴヌクレオチドの、例えば融解温度(Tm)で測定された親和性を増強するように修飾されたヌクレオチドである。本発明の高親和性修飾ヌクレオシドは、修飾ヌクレオシド当たり好ましくは+0.5〜+12℃、より好ましくは+1.5〜+10℃、最も好ましくは+3〜+8℃の間の融解温度の上昇をもたらす。当該技術分野において多数の高親和性修飾ヌクレオシドが公知であり、例えば、多数の2’置換ヌクレオシド、ならびにロックド核酸(LNA)が包含される(例えば、Freier & Altmann;Nucl.Acid Res., 1997, 25, 4429-4443及びUhlmann;Curr.Opinion in Drug Development, 2000, 3(2), 293-213を参照のこと)。
本発明のオリゴマーは、修飾された糖部分、すなわちDNA及びRNAに見出されるリボース糖部分と比較して、糖部分の修飾を有する1つ以上のヌクレオシドを含む場合がある。
LNAヌクレオシドは、C2’とC4’との間にリンカー基(ビラジカル又はブリッジと呼ばれる)を含む、修飾ヌクレオシドである。これらのヌクレオシドは、文献中では、架橋核酸又は二環式核酸(BNA)とも呼ばれる。

アンチセンスオリゴヌクレオチドのRNアーゼH活性は、相補的RNA分子と二本鎖を成す場合にRNアーゼHを補充する能力を指す。国際公開第01/23613号パンフレットは、RNアーゼHを補充する能力を決定するために使用できるRNアーゼH活性を測定するためのインビトロの方法を提供するものである。オリゴヌクレオチドは典型的に、RNアーゼHを補充する能力を有すると考えられ、相補的な標的核酸配列が提供されているとき、初期速度をpmol/l/minで測定した場合に、定量された初期速度の少なくとも5%、例えば少なくとも10%又は20%超の初期速度を有する(試験対象の修飾オリゴヌクレオチドと同じ塩基配列を有するオリゴヌクレオチドで、但し、オリゴヌクレオチド中の全てのモノマー間のホスホロチオエート結合を有するDNAモノマーのみを含有するものを使用し、且つ本明細書において参照により援用されている国際公開第01/23613号パンフレットの実施例91〜95に記載の方法論を使用した場合)。
本明細書において「ギャップマー」という用語は、1つ以上の親和性を増強する修飾ヌクレオシド(側面又はウイング)を含む領域によって5’及び3’に隣接するRNアーゼH補充オリゴヌクレオチド(ギャップ)の領域を含むアンチセンスオリゴヌクレオチドを指す。様々なギャップマーデザインが、本明細書に記載されており、RNアーゼHを補充する能力によって特徴付けられる。ヘッダー及びテールマーは、側面のうちの1つが欠けている、すなわちオリゴヌクレオチドの末端のうちの1つのみが親和性を増強する修飾ヌクレオシドを含む、RNアーゼHを補充する能力を有するオリゴヌクレオチドである。頭部の場合、3’側面は欠けている(すなわち、5’側面は親和性修飾ヌクレオシドを含む)、テールマーは5’側面が欠けている(すなわち、3’側面は親和性修飾ヌクレオシドを含む)。
LNAギャップマーという用語は、親和性増強修飾ヌクレオシドのうちの少なくとも1つがLNAヌクレオシドである、ギャップマー オリゴヌクレオチドを言う。
混合ウイングギャプマー又は混合側面ギャップマーという用語は、側面領域のうちの少なくとも1つが、少なくとも1つのLNAヌクレオシドと、少なくとも1つの非LNA修飾ヌクレオシド(例えば、2’−O−アルキル−RNA、2’−O−メチル−RNA、2’−アルコキシ−RNA、2’−O−メトキシエチル−RNA、2’−アミノ−DNA、2’−フルオロ−RNA及び2’−F−ANAヌクレオシドのような、少なくとも1つの2’−置換修飾ヌクレオシド)と、を含んでなるLNAギャプマーを指す。幾つかの実施形態において、混合ウイングギャップマーは、一方の側面にLNAヌクレオシド(例えば、5’又は3’)のみを含み、他方の側面(それぞれ3’又は5’)に2’置換修飾ヌクレオシド及び任意でLNAヌクレオシドを含む。
「ギャップブレイカー・オリゴヌクレオチド」という用語は、ギャップ領域が非RNアーゼH補充ヌクレオシド(ギャップブレイカー・ヌクレオシド、E)で破壊された結果、ギャップ領域内に含まれる連続DNAヌクレオシド数が5未満となった場合でもRNアーゼHの補充を維持する能力を有するギャップマーに関して使用される。非RNアーゼH補充ヌクレオシドは、例えば、ヌクレオシドのリボース糖環のC2’とC4’との間の架橋がβ−D−オキシLNA又はScETヌクレオシドのようなβ立体配座にある、LNAのような3’末端立体構造のヌクレオシドである。ギャップブレイカー・オリゴヌクレオチドがRNアーゼHを補充する能力は、典型的に、配列特異的、又は化合物特異的でさえある(Rukov et al.2015 Nucl.Acids Res.Vol. 43 pp. 8476-8487を参照)。本文献には、「ギャップブレイカー」オリゴヌクレオチドが開示されている。このギャップブレイカー・オリゴヌクレオチドは、幾つかの例において標的RNAのより特異的な開裂を提供するRNアーゼHを補充するものである。

本明細書において、コンジュゲートという用語は、非ヌクレオチド部分(結合部分又は領域C又は第3の領域)に共有結合しているオリオゴヌクレオチドを指し、オリゴヌクレオチドコンジュゲートとも呼ばれる。
連鎖(linkage)又はリンカーは、1つの化学基又は関心のあるセグメントを1つ以上の共有結合を介して別の化学基又は関心対象のセグメントに連結する2つの原子間の結合である。結合部分は、オリゴヌクレオチドに直接又は連結部分(例えば、リンカーもしくはテザー)を介して結合させることができる。リンカーは、第3の領域、例えば結合部分(領域C)を、第1の領域、例えば標的核酸に相補的なオリゴヌクレオチド又は連続ヌクレオチド配列(領域A)に共有結合するように機能する。
本明細書において「処置」という用語は、既存の疾患(例えば、本明細書で言及する疾患もしくは障害)の治療、又は疾患の防止、すなわち予防を指す。したがって、幾つかの実施形態において本明細書中で言及される治療は予防的でありうることが認識されるであろう。
免疫応答は、先天性及び適応性の免疫応答に分類される。自然免疫系は、即時的ではあるが非特異的な応答を提供するものである。適応免疫応答は、自然免疫応答によって活性化され、特定の病原体に対して高度に特異的である。抗原提示細胞の表面上に病原体由来抗原が提示されると、適応免疫応答の免疫細胞(すなわち、Tリンパ球及びBリンパ球)が、抗原特異的受容体を介して活性化され、病原性特異的免疫応答、及び免疫記憶の発生につながる。HBV及びHCVのような慢性ウイルス感染症は、ウイルス特異的T細胞の非応答性を特徴とするT細胞枯渇と関連している。T細胞の疲弊は充分に研究され、総説については、例えばYi et al 2010 Immunology129, 474-481を参照のこと。慢性ウイルス感染はまた、先天性免疫細胞であるNK細胞の機能低下に関連する。慢性感染のクリアランスのためには、ウイルス免疫応答の強化が重要である。T細胞及びNK細胞媒介性の病原体に対する免疫応答の回復は、増殖、サイトカイン分泌及び細胞溶解性機能の測定によって評価できる(Dolina et al.2013 Molecular Therapy-Nucleic Acids, 2 e72、及び本明細書中の実施例6)。
本発明は、PD−L1の発現を調節できるオリゴヌクレオチドに関する。調節は、PD−L1をコードする標的核酸又はPD−L1の調節に関与する標的核酸にハイブリダイズによって達成できる。標的核酸は、(例えば、配列番号:1、配列番号:2及び/又は配列番号:3からなる群から選択される配列)哺乳類PD−L1配列でありうる。標的核酸は、PD−L1の発現又は調節を支持する哺乳類の細胞から発現されたプレmRNA、mRNA又は任意のRNA配列であってもよい。
オリゴヌクレオチドデザインとは、オリゴヌクレオチド配列中のヌクレオシド糖修飾のパターンを指す。本発明のオリゴヌクレオチドは、糖修飾ヌクレオシドを含み、また、DNA又はRNAヌクレオシドを含む場合がある。幾つかの実施形態において、オリゴヌクレオチドは、糖修飾ヌクレオシド及びDNAヌクレオシドを含む。本発明のオリゴヌクレオチドに修飾ヌクレオシドを取り込むことによって、標的核酸に対するオリゴヌクレオチドの親和性を増強できる。その場合、修飾ヌクレオシドは、親和性増強修飾ヌクレオチドと呼ばれる場合があり、修飾ヌクレオシドは単位と呼ばれる場合がある。

生体開裂性リンカー(領域B)
コンジュゲートの使用は、多くの場合、薬物動態特性又は薬力学的動的特性の増強に関連する。しかし、結合部分が存在する場合は、例えば、ハイブリダイゼーション又はヌクレアーゼ補充(例えば、RNアーゼH)を防止する立体障害を介して、意図された標的オリゴヌクレオチドの活性が妨げられる可能性がある。オリゴヌクレオチド(領域A又は第1領域)と結合部分(領域C又は第3領域)との間に生理学的に易変的な結合(生体開裂性リンカー)を使用することにより結合部分の存在による特性の改善が可能となる一方、いったん標的組織において結合基がオリゴヌクレオチドの有効な活性を妨げないことが保証される。
リンカーは、少なくとも2つの官能性基(一方はオリゴヌクレオチドに結合し、他方は結合部分に結合する)を有する場合がある。例示的なリンカー官能基は、オリゴヌクレオチド又は結合部分上の求核性基と反応するために求電子性である場合もあれば、あるいは求電子性基と反応するための求核性である場合もある。幾つかの実施形態において、リンカー官能基としては、ホスホロチオエート、ホスフェート、ホスファイト、不飽和(例えば、二重又は三重結合)などが挙げられるが、これらに限定されない。幾つかの例示的なリンカー(領域Y)としては、8−アミノ−3,6−ジオキサオクタン酸(ADO)、スクシンイミジル4−(N−マレイミドメチル)シクロヘキサン−1−カルボキシレート(SMCC)、6−アミノヘキサン酸(AHEX又はAHA)、6−アミノヘキシルオキシ、4−アミノ酪酸、4−アミノシクロヘキシルカルボン酸、スクシンイミジル4−(N−マレイミドメチル)シクロヘキサン−1−カルボキシ−(6−アミド−カプロン酸)(LCSMCC)、スクシンイミジルm−マレイミド−ベンゾイルエート(MBS)、スクシンイミジルN−e−マレイミド−カプロイル酸(EMCS)、スクシンイミジル6−(ベータ−マレイミド−プロピオンアミド)ヘキサノエート(SMPH)、スクシンイミジルN−(α−マレイミドアセテート)(AMAS)、スクシンイミジル4−(p−マレイミドフェニル)ブチレート(SMPB)、β−アラニン(β−ALA)、フェニルグリシン(PHG)、4−アミノシクロヘキサン酸(ACHC)、β−(シクロプロピル)アラニン(β−CYPR)、アミノドデカン酸(ADC)、アリレンジオール、ポリエチレングリコール、アミノ酸等が挙げられる。幾つかの実施形態において、リンカー(領域Y)は、アミノアルキル、例えばC2−C36アミノアルキル基、例えばC6−C12アミノアルキル基を含む。好ましい実施形態において、リンカー(領域Y)は、C6アミノアルキル基である。アミノアルキル基は、標準オリゴヌクレオチド合成の一部として、例えば保護されたアミノアルキルホスホルアミダイト等を使用して、オリゴヌクレオチド(領域A又は領域A−B)に添加できる。アミノアルキルとオリゴヌクレオチドとの間の連結基は、例えば、ホスホロチオエート又はホスホジエステル、又は本明細書中で言及される他のヌクレオシド連結基の1つでありうる。アミノアルキル基は、オリゴヌクレオチドの5’又は3’末端に共有結合している。市販のアミノアルキルリンカーは、例えば、オリゴヌクレオチドの3’末端における連結のための、及びオリゴヌクレオチドの5’末端における連結のための3’−アミノ修飾試薬である5’−アミノ修飾酵素C6が利用可能である。これらの試薬は、Glen Research Corporation(Sterling, Va.)から入手可能である。これらの化合物又は類似のものは、フルオレセインをオリゴヌクレオチドの5’末端に連結することを目的に、Krieg, et al, Antisense Research and Development 1991, 1, 161によって利用された。多種多様な更なるリンカー基が当該技術分野において知られており、オリゴヌクレオチドへの結合部分の結合に有用でありうる。有用なリンカー基の多くに関する総説は、例えば、Antisense Research and Applications, S. T. Crooke and B. Lebleu, Eds., CRC Press, Boca Raton, Fla., 1993, p. 303-350に見出される。アクリジンのような他の化合物は、ポリメチレン結合を介してオリゴヌクレオチドの3’末端リン酸基に結合している(Asseline, et al., Proc.Natl.Acad.Sci.USA 1984, 81, 3297)。上記の基は全ていずれも、単一のリンカー(領域Y)として、又は1つ以上の更なるリンカー(領域Y−Y’又は領域Y−BもしくはB−Y)と組み合わせて使用できる。
更なる態様において、本発明は本発明のオリゴヌクレオチドを製造する方法を提供するものであり、この製法は、ヌクレオチド単位を反応させ、それにより、そのオリゴヌクレオチド内に含有されている共有結合の連続ヌクレオチド単位を形成することを含む。この方法は、ホスホラミダイト化学を使用するものであることが、好ましい(例えば、Caruthers et al, 1987, Methods in Enzymology vol. 154, pages 287-313を参照)。更なる実施形態において、本方法は、連続ヌクレオチド配列を結合部分(リガンド)と反応させることを、更に含む。更なる態様では、本発明のオリゴヌクレオチドもしくは結合オリゴヌクレオチドを、医薬的に許容される希釈剤、溶媒、担体、塩及び/又は補助剤と混合することを含む、本発明の組成物の製造方法が提供されている。
更なる態様において、本発明は、前述のオリゴヌクレオチド及び/又はオリゴヌクレオチドコンジュゲート、ならびに医薬的に許容される希釈剤、溶媒、担体、塩及び/又は補助剤のいずれかを含む、医薬組成物を提供するものである。医薬的に許容される希釈剤としては、リン酸緩衝化生理食塩水(PBS)が挙げられる。医薬的に許容される塩としては、限定されるものではないが、ナトリウム塩及びカリウム塩が挙げられる。幾つかの実施形態において、医薬的に許容される希釈剤は、滅菌リン酸緩衝生理食塩水である。幾つかの実施形態において、オリゴヌクレオチドは、医薬的に許容される希釈剤中に50〜300μMの濃度で使用される。
本発明のオリゴヌクレオチド又はオリゴヌクレオチドコンジュゲートは、例えば、診断薬、治療薬及び予防用の研究試薬として利用できる。
本発明のオリゴヌクレオチド又は医薬組成物は、局所的に(皮膚、吸入、眼もしくは耳)又は経腸(経口もしくは胃腸管など)又は非経口(静脈内、皮下、筋肉内、大脳内、脳室内もしくはクモ膜下腔内)投与できる。
幾つかの実施形態において、本発明のオリゴヌクレオチド、オリゴヌクレオチドコンジュゲート又は医薬組成物は、他の治療剤との併用治療の用途に使用される。治療剤は、例えば、上記の疾患又は障害に対する標準ケアとすることができる。

モチーフ配列及びオリゴヌクレオチド化合物
パスツールモデル:
パスツール研究所(Institut Pasteur)で、HLA−A2.1−/HLA−DR1−トランスジェニックH−2クラスI−/クラスII−ノックアウトマウス(本明細書中ではHLA−A2/DR1マウスと呼ばれる)を作成し、飼育した。これらのマウスは、マウスMHC応答に何ら干渉することのない、ヒト免疫機能試験用のインビボ実験モデルである(Pajot et al 2004 Eur J Immunol.34(11): 3060-9)。
このモデルでは、HBVゲノム(AAV/HBV)を保有する組み換えアデノ随伴ウイルス(AAV)で感染したマウスは、30週間を超えて安定なウイルス血症及び抗原血症を維持する(Dan Yang, et al. 2014 Cellular & Molecular Immunology 11, 71-78)。
プラスミドDNAは、エンドトキシン不含で、Plasmid-Factory(Germany)で製造された。pCMV−S2.Sは、HBs抗原(遺伝子型D)のプレS2ドメイン及びSドメインをコードし、その発現はサイトメガロウイルス即時初期遺伝子プロモーターによって制御される(Michel et al 1995 Proc Natl Acad Sci U S A 92: 5307-5311)。pCMV−HBcは、肝炎コア(HBc)抗原を保有するHBVキャプシドをコードする(Dion et al 2013 J Virol 87: 5554-5563)。
これは、Genetech社で内部的に産生されたマウス抗マウスPD−L1 IgG1抗体クローン6E11であり、Atezolizumabをブロックし、同様のインビトロでのブロッキング活性を有する代理抗体であり、ロッシュ(Roche)で内部的に産生されるアテゾリズマブである。抗体は、12.5μg/gの用量で腹腔内(i.p.)注射によって投与された。
オリゴヌクレオチド合成は、当該技術分野において概ね公知である。以下は適用可能なプロトコールである。本発明のオリゴヌクレオチドは、使用される器具、支持体及び濃度の点で僅かに変化する方法によって製造されたものであってもよい。
5’−O−DMT保護アミダイトのアセトニトリル溶液(0.1M)、及びDCI(4,5−ジシアノイミダゾール)のアセトニトリル溶液(0.25M)を活性化剤として使用して、β−シアノエチル−ホスホラミダイト(DNA−A(Bz)、DNA−G(ibu)、DNA−C(Bz)、DNA−T、LNA−5−メチル−C(Bz)、LNA−A(Bz)、LNA−G(dmf)、又はLNA−T)のカップリングを行った。最終サイクル用に、所望の修飾を有するホスホラミダイトを使用できる。結合基を結合させるためのC6リンカー、又は結合基である。ホスホロチオエート結合の導入のためのチオール化は、水素化キサンタン(アセトニトリル/ピリジン9:1中0.01M)を使用して実施する。フォスフォジエステル結合は、THF/ピリジン/水(7:2:1)中の0.02Mヨウ素を使用して導入できる。残りの試薬は、オリゴヌクレオチドの合成に典型的に使用される試薬である。
粗化合物を、Phenomenex Jupiter C18の10μ150×10mmカラムでの分取RP−HPLCによって精製する。0.1M酢酸アンモニウムpH8及びアセトニトリルを、5mL/分の流速で緩衝液として使用する。収集した画分を凍結乾燥して、典型的には白色固体として精製化合物を得る。

オリゴヌクレオチド及びRNA標的(リン酸結合、PO)二本鎖を、500mlのRNアーゼ不含水中で3mMに希釈し、500mlの2×Tm緩衝液(200mMのNaCl、0.2mMのEDTA、20mMのリン酸ナトリウム、pH7.0)と混合する。溶液を95℃で3分間加熱し、次いで室温で30分間アニールさせる。PE Templabソフトウェア(Perkin Elmer)を用いたPeltier温度プログラマーPTP6を備えたLambda 40 UV/VIS分光光度計で、二本鎖融解温度(Tm)を測定する。温度を20℃から95℃まで上昇させ、次いで25℃まで下げ、260nmで吸収を記録する。一次導関数、ならびに融解及びアニーリングの両方の極大値を使用して、二本鎖Tmを評価する。
試験される生体開裂性リンカー(例えば、DNAホスホジエステルリンカー(POリンカー))を有するFAM標識オリゴヌクレオチドは、関連する組織(例えば、肝臓又は腎臓)及び血清のホモジネートを使用してインビトロで開裂される。
S1ヌクレアーゼ感受性リンカー(例えば、DNAリン酸ジエステルリンカー(POリンカー))を用いたFAM標識オリゴヌクレオチドを、S1ヌクレアーゼ抽出物又は血清中でインビトロ開裂させる。
Tupin et al 2006 Methods Enzymol 417: 185-201に記載されている方法に若干の修正を加えて、AAV/HBVマウス由来の肝細胞を調製した。安楽死させた後のマウスの肝臓に、G25ニードル付きのシリンジを使用して、門脈経由で滅菌PBS10mlを灌流させた。臓器が蒼白のときは、その臓器を、ハンクス平衡塩類溶液(HBSS)(GIBCO(登録商標)HBSS、24020)+5%の胎児牛胎仔血清(FCS)で採取した。採取された肝臓を100μm細胞ストレーナー(BD Falcon、352360)に通してゆっくりと加圧し、30mlのHBSS+5%FCS中に細胞を懸濁させた。細胞懸濁液を5分間50gで遠心分離させた後、上清を4℃にて10分間289gで遠心分離させた。遠心分離後、上清を捨て、ペレットを35%等張パーコール溶液(GE Healthcare Percoll#17-0891-01、RPMI 1640(GIBCO, 31870)に希釈)中で室温にて15ml単位で再懸濁させ、15mlチューブに移した。細胞を更に室温にて25分間1360gで遠心分離させた。上清を吸引により捨て、単核細胞を含有するペレットを、HBSS+5%FCSで2回洗浄した。
細胞をU底96ウェルプレート中に播種し、PBS FACS(1%ウシ血清アルブミンと0.01%アジ化ナトリウムとを含有するPBS)で洗浄した。ラット抗マウスCD16/CD32抗体と生存マーカーLDフィクサブルイエロー(Thermofisher, L34959)とを含有する5μLのPBS FACSで、4℃の暗所で細胞を10分間インキュベートした。次いで、NK P46 BV421(ラットMab抗マウスNK P46、Biolegend、137612)及びF4/80(ラットMab抗マウスF4/80 FITC、BD Biolegend, 123108)に対するモノクローナル抗体(Mab)を含有する25μLのPBS FACSで4℃の暗所にて細胞を20分間染色し、また、2つの補足的面マーカー:PDL1(ラットMab抗マウスPD1 PE、BD Biosciences, 551892)及びPDL1(ラットMab抗マウスPDL1 BV711、Biolegend, 124319)を添加した。
脾細胞及び肝臓単核細胞の両方でICSアッセイを実施した。細胞をU底96ウェルプレートに播種した。細胞を入れたプレートを、陰性対照としての完全培地単独で、又は表9に記載のペプチドとの併用のいずれかで、37℃にて2μg/mlの濃度で一晩インキュベートした。インキュベーションの1時間後に、2μg/mLのブレフェルジンA(Sigma, B6542)を添加した。
主として16〜20merのギャプマーを使用して、ヒトPD−L1転写産物全体に対して遺伝子ウォーク(gene walk)を実施した。有効性試験は、ヒト白血病単球細胞系統THP1及びヒト非ホジキンリンパ腫細胞系統(KARPAS−299)における、インビトロ実験で行った。
THP1及びKarpas−299細胞系統は、当初ECACC(European Collection of Authenticated Cell Cultures)から購入し、加湿インキュベーター内で、供給者が推奨するように37℃にて5%CO2で維持した。
THP−1細胞(RPMI-GLutamax中3.104、10%FBS、1%Pen−Strep(Thermo Fisher Scientific)をオリゴヌクレオチド(4〜5μl)に添加し、これを96ウェル丸底プレートに入れ、最終容量100μl/ウェルで6日間培養した。オリゴヌクレオチドを、単一濃度(20μM)にて、及び25μM〜0.004μM(1:3希釈の水溶液)の用量範囲濃度にて、スクリーニングした。MagNA Pure 96 Cellular RNA Large Volume Kit(MagNA Pure 96 System(Roche Diagnostics))をメーカーの指示に従って使用して、総mRNAを抽出した。遺伝子発現解析用に、内因性対照として使用されるヒトPDL1及びACTBを標的化する予備設計済のTaqmanプライマー(Thermo Fisher Scientific)を用い、QuantStudioマシン(Applied Biosystems)で、TaqMan RNA-to-ct1-Stepキット(Thermo Fisher Scientific)を使用してRT−qPCRを行った。相対的なPD−L1のmRNA発現レベルは、2ΔΔC(T)(2(-Delta Delta C(T)))法及び対照試料(非処置細胞)と比較した割合(%)としての阻害パーセンテージを使用して計算された。
表10から選択されたオリゴヌクレオチドを、KARPAS-299細胞にて、半対数(half-log)連続希釈PBS溶液(50μM、15.8μM、5.0μM、1.58μM、0.5μM、0.158μM、μM、0.05μM、0.0158μMのオリゴヌクレオチド)を用いて試験し、実施例1に記載のインビトロ有効性アッセイにおいて評価した。IC50及び最大の阻害(残渣%PD−L1発現量)オリゴヌクレオチドについて評価した。
有効性及び効能試験は、表6のオリゴヌクレオチドを使用して、MCP−11細胞における用量応答研究におけるインビトロ実験で行った。同じオリゴヌクレオチド、ならびにGalNAc結合型バージョン(表8のCMP ID番号755_2〜765_2)が、ポリ(I:C)誘発C57BL/6J雌マウスにおけるPD−L1のmRNA及びタンパク質の発現を低減させる能力を、インビボで試験した。
10%ウマ血清、2mMのL−グルタミン、0.025mg/mlゲンタマイシン及び1mMのピルビン酸ナトリウムを補給したDMEM(Sigmaカタログ番号D0819)中に懸濁させたMCP−11細胞(当初ATCCから購入)を、96ウェル丸底プレート中のオリゴヌクレオチド(10μl)に8000細胞/ウェルの密度で加えて、5%CO2を含む37℃の加湿インキュベーター内で200μl/ウェルの最終容量で3日間培養した。オリゴヌクレオチドを用量範囲濃度(50μM、15.8μM、5.0μM、1.58μM、0.5μM、0.158μM、0.05μM及び0.0158μM)でスクリーニングした。
マウスPD−L1に対する5mg/kgの非結合オリゴヌクレオチド、又はマウスPD−L1に対する2.8mg/kgのGalNAc結合型オリゴヌクレオチドを、C57BL/6J雌マウス(20〜23g;1群当たり5匹のマウス)に皮下注射した。3日後、10mg/kgポリ(I:C)(LWM、Invivogen)を、マウスに静脈内注射した。ポリ(I:C)注射後5時間目にマウスを屠殺し、肝臓試料をRNA抽出用のRNAlater(Thermo Fisher Scientific)に入れるか、又はタンパク質抽出に備えてドライアイスで凍結させた。
ポリ(I:C)誘導マウスから単離された肝細胞及び非実質細胞における、裸型オリゴヌクレオチド及びGalNAc結合型オリゴヌクレオチドの分布、及びPD−L1mRNAの低減を調べた。
本研究では、AAV/HBVマウスを裸型又はGalNAc結合型PD−L1アンチセンスオリゴヌクレオチドで処置し、肝臓におけるPD−L1のmRNA発現量及びHBV遺伝子発現を評価した。

本研究では、パスツール(Pasteur)から入手されたAAV/HBVマウスを、PD−L1を標的化する抗体又はアンチセンスオリゴヌクレオチドで処置した。アンチセンスオリゴヌクレオチドは、裸型又はGalNAc結合型のいずれかであった。処置中に、抗原提示細胞による効率的なT細胞プライミングを確実にするために、HBs抗原及びHBc抗原に抗するDNAワクチンで動物を免疫した(「材料と方法」の節を参照のこと)。この処置が肝臓内及び脾臓内の細胞母集団ならびにこれらの母集団におけるPD−L1発現に対してどのように影響するか、またHBV特異的T細胞応答を同定できるかどうかを評価した。
雌のHLA−A2/DR1マウスを以下のプロトコールに従って処置した。本研究は、下の表16及び表17に示すような投与レジメンにおいて若干異なる2つの別個のサブ研究で実施された。
サイトメトリーを使用して肝臓単核細胞(「材料と方法」を参照)の表面標識によって、肝臓における細胞集団を分析した。
屠殺の時点での、脾臓及び肝臓由来のマクロファージ、B細胞及びT細胞におけるPD−L1タンパク質の発現を評価した。表面標識抗体混合物中にPD−L1抗体を存在させることによって(「材料と方法」を参照)、サイトメトリーを用い、PD−L1発現細胞の定量が可能となった。
IFNγ及びTNFα産生を検出する細胞内サイトカイン染色アッセイ(「材料と方法」の節を参照)を使用して、炎症性サイトカインを産生するNK細胞、CD4+T細胞、及びCD8+T細胞を検出した。
本研究では、上海から入手されたAAV/HBVマウス(「材料と方法」の節を参照)を、GalNAc結合型PD−L1アンチセンスオリゴヌクレオチドCMP ID番号759_2で処置した。
「材料と方法」の節で上海モデルに記載されているように、本研究では、HBVゲノムを保有する組み換えアデノ随伴ウイルス(AAV)(AAV/HBV)を感染させた雄のC57BL/6マウスを使用した。5mg/kg又は媒体(生理食塩水)のアンチセンスオリゴヌクレオチドCMP ID番号:759_2の両方を、8週間にわたって週1回マウス(1群当たりマウス6匹)に注射した。この注射は、皮下注射(s.c.)として投与された。処置中は毎週、処置後6週間に血液試料を採取した。以下に記載するように、HBV DNA、HBs抗原及びHBe抗原レベルを血清試験で測定した。最初の10週間分の結果を、表21及び図13に示す。出願の時点で依然として調査が進行中であったので、残りの4週間分のデータは得られていない。
HBs抗原・化学発光イムノアッセイ(CLIA)及びHBe抗原CLIAキット(それぞれAutobio diagnostics Co. Ltd., Zhengzhou, China、カタログ番号CL0310−2及びCL0312−2)を使用して、製造業者のプロトコールに従い、感染AAV−HBVマウス血清中の血清HBs抗原及びHBe抗原レベルを測定した。概略説明すると、50μlの血清をそれぞれの抗体被覆マイクロタイタープレートに移し、50μlの酵素結合試薬を加えた。プレートを室温にて振盪機上で60分間インキュベートした後、自動洗浄機を使用して全てのウェルを洗浄緩衝液で6回洗浄した。25μlの基質A、次いで25μlの基質Bを、各ウェルに加えた。プレートを室温で10分間インキュベートした後、Envisionルミネセンスリーダーを使用して発光を測定した。1ngのHBs抗原=1.14IUとした場合、HBs抗原は単位IU/mlで与えられる。HBe抗原は、単位NCU/ml血清で与えられる。
最初に、マウス血清をリン酸緩衝生理食塩水(PBS)で10倍(1:10)に希釈した。MagNA Pure 96(Roche)ロボットを使用して、DNAを抽出した。希釈された血清50μlを処置カートリッジ中で200μlのMagNA Pure 96外部溶解緩衝液(Roche、カタログ番号06374913001)と混合し、10分間インキュベートした。次いで、「MagNA Pure 96 DNA and Viral Nucleic Acid Small Volume Kit」(Roche、カタログ番号06543588001)及び「Viral NA Plasma SV external lysis 2.0」プロトコールを使用して、DNAを抽出した。DNA溶出容量は50μlであった。

Taqmanプローブ(P3_core):56−FAM/AGC TCC AAA /ZEN/TTC TTT ATA AGG GTC GAT GTC CAT G/3IABkFQ(配列番号:786)。
初代ヒト肝細胞におけるPD−L1転写産物を減少させるGalNAc結合型PD−L1アンチセンスオリゴヌクレオチド化合物の能力を、ゲノミクスを用いて調べた。
凍結保存されたヒト肝細胞を、10%ウシ胎児血清、ペニシリン(100U/ml)、ストレプトマイシン(0.1mg/ml)及びL−グルタミン(0.292mg/ml)を補給したWME中で5×106細胞/mlに希釈し、2×105細胞/ウェルの密度でコラーゲン被覆24ウェルプレート(Becton Dickinson AG、Allschwil、Switzerland)中に播種した。細胞を4時間前培養し、細胞培養プレートに付着させてから、最終濃度100μMのオリゴヌクレオチドでの処置を開始した。使用されたオリゴヌクレオチドは、表21及び表8に示すとおりであり、ビヒクルはPBSとされた。播種培地を、ペニシリン(100U/ml)、ストレプトマイシン(0.1mg/ml)及びL−グルタミン(0.292mg/mlを補給した315μlの無血清WMEで置き換えて、1mMのPBS中のオリゴヌクレオチドストック溶液35μlを細胞培養物に添加し、これを細胞上に24時間又は66時間放置した。
Stranded mRNAケミストリーを使用し、2×51bp対末端リード及び30M/試料(Q squared EA)の配列決定ストラテジーを用いて、転写物発現プロファイリングを行った。ウェル中にQiagen RLT緩衝液350μlを添加して、細胞を溶解させ、無作為化スキームで受託した。
GSNAPショートリードアライメントプログラムを使用して、長さ2×51bpのIlluminaペアエンド・シーケンシング・リード(paired-end sequencing reads)を、ヒト参照ゲノムhg19上にマッピングした。SAMTOOLSプログラムを使用して、SAMフォーマットアライメントを、ソートアライメントBAMフォーマットファイルに変換した。hg19に対応するGTFファイルで指定されたNCBI RefSeqからのエキソンアノテーションに基づいて、PD−L1の遺伝子読み取りカウントを推定した。各試料の異なるライブラリサイズを考慮した正規化工程が、DESeq2Rパッケージを使用して適用された。
HBVに感染したASGPR−HepaRG細胞における2つの裸型PD−L1アンチセンスオリゴヌクレオチド及び同等のGalNAc結合型PD−L1アンチセンスオリゴヌクレオチドの効力を比較した。
HepaRG細胞(Biopredic International, Saint-Gregoire, France)を、ウィリアムE培地(10%HepaRG増殖サプリメント(Biopredic)を添加したもの)で培養した。レンチウイルス法を使用して、この細胞系統から、ヒトASGPR1及びASGPR2を安定に過剰発現するHepaRG細胞系統を生成した。CMVプロモーター及びピューロマイシン耐性遺伝子の制御下でヒトASGPR1及び2をコードするSirion biotech(CLV−CMV-ASGPR1-T2a_ASGPR2-IRES-Puro)が必要に応じて産生したレンチウイルスを、増殖用HepaRG細胞に、感染多重度(MOI)を300として形質導入した。形質導入細胞を1μg/mlピューロマイシンで11日間選択し、次いで、確実に導入遺伝子が安定して発現するように、同濃度の抗生物質中で維持した。ASGPR1/2過剰発現を、RT−qPCR(ASGPR1:非形質導入対8560倍、ASGPR2:非形質導入対2389倍)によるmRNAレベル及びフローサイトメトリー分析によるタンパク質レベルの両方で確認した。

末梢血単核細胞(PBMC)のエクスビボでのHBV抗原刺激後に、裸型PD−L1アンチセンス化合物が、慢性的に感染したHBV(CHB)患者のT細胞機能を増強させうるかどうかを調べた。
Claims (24)
- a.PD−L1標的核酸に対して少なくとも90%の相補性を有する10〜30ヌクレオチド長さの連続ヌクレオチド配列を含む、オリゴヌクレオチド(領域A)と;
b.(a)のオリゴヌクレオチドに共有結合された少なくとも1つの、アシアロ糖タンパク質受容体標的化結合部分(領域C)と;
を含んでなる、アンチセンスオリゴヌクレオチドコンジュゲート。 - 前記連続ヌクレオチド配列が、配列番号:1、配列番号:2及び/又は配列番号:3からなる群から選択される標的核酸に対して相補的である、請求項1に記載のオリゴヌクレオチドコンジュゲート。
- 前記連続ヌクレオチド配列が標的核酸のサブ配列に対して相補的であり、前記サブ配列が、配列番号:1上の371−3068位、5467−12107位及び15317−19511位からなる群から選択される、請求項1又は2に記載のオリゴヌクレオチドコンジュゲート。
- 前記連続ヌクレオチド配列が標的核酸のサブ配列に対して相補的であり、前記サブ配列がA221、A360、A180、A160及びA269からなる群から選択される、請求項1から3に記載のオリゴヌクレオチドコンジュゲート。
- 前記オリゴヌクレオチドが、配列番号:466、640、342、287及び566から選択される配列を含む、請求項1から4に記載のオリゴヌクレオチドコンジュゲート。
- 前記連続ヌクレオチド配列が1以上の修飾ヌクレオシド(例えば、1以上の2’糖修飾ヌクレオシドを含む、請求項1から5に記載のオリゴヌクレオチドコンジュゲート。
- 前記1以上の2’糖修飾ヌクレオシドが独立に、2’−O−アルキル−RNA、2’−O−メチル−RNA、2’−アルコキシ−RNA、2’−O−メトキシエチル−RNA、2’−アミノ−DNA、2’−フルオロ−DNA、アラビノ核酸酸(ANA)、2’−フルオロ−ANA、及びLNAヌクレオシドからなる群から選択される、請求項6に記載のオリゴヌクレオチドコンジュゲート。
- 前記全ての修飾ヌクレオシドがLNAヌクレオシドである、請求項6又は7のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- 前記連続ヌクレオチド配列が、少なくとも1つの修飾ヌクレオシド間結合(例えば、少なくとも1つのホスホロチオエートヌクレオシド間結合)を含む、請求項1から8のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- 前記オリゴヌクレオチドがギャップマーである、請求項1から9のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- 前記ギャップマーが式5’−D’−F−G−F’−3’又は5’−F−G−F’−D”−3’を有し、領域F及びF’が独立に、1〜7修飾ヌクレオシドを含み、GがRNアーゼHを補充する能力を有する6〜16ヌクレオシド間の領域であり、及び領域D’又はD”は任意であり、且つ0〜5ホスホジエステルの連結ヌクレオシドを含む、請求項10に記載のオリゴヌクレオチドコンジュゲート。
- 前記アシアロ糖タンパク質受容体標的化結合部分が、ガラクトース、ガラクトースアミン、N−ホルミル−ガラクトサミン、N−アセチルガラクトサミン、N−プロピオニル−ガラクトサミン、N−n−ブタノイル−ガラクトサミン及びN−イソブタノイルガラクトサミンからなる群から選択される少なくとも1つの炭水化物部分を含む、請求項1から11のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- 前記アシアロ糖タンパク質受容体標的化結合部分が1価、2価、3価又は4価である、請求項1から12のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- 前記アシアロ糖タンパク質受容体標的化結合部分が、3価N−アセチルガラクトサミン(GalNAc)部分である、請求項1から13のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- 前記アシアロ糖タンパク質受容体標的化結合部分が、図3の前記3価GalNAc部分である、請求項1から14のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- 前記オリゴヌクレオチドコンジュゲートがCMP ID番号:766_2、767_2、768_2、769_2及び770_2から選択される、請求項1から15のいずれか一項に記載のオリゴヌクレオチドコンジュゲート。
- オリゴヌクレオチド又は請求項1から11のいずれか一項に記載の連続ヌクレオチド配列を含む、アンチセンスオリゴヌクレオチド。
- 請求項1から16に記載のオリゴヌクレオチドコンジュゲート、又は請求項17に記載のオリゴヌクレオチド、ならびに医薬的に許容される希釈剤、溶媒、担体、塩及び/又は補助剤を含む、医薬組成物。
- PD−L1を発現する標的細胞におけるPD−L1の発現を調節するためのインビボ又はインビトロ方法であって、前記方法が、有効量の請求項1から16に記載のオリゴヌクレオチドコンジュゲート、又は請求項17に記載のオリゴヌクレオチドを前記細胞に投与することを含む、方法。
- ウイルスもしくは寄生虫に対する免疫応答の回復に使用するための、請求項1から16に記載のオリゴヌクレオチドコンジュゲート、又は請求項17に記載のオリゴヌクレオチド、又は請求項18に記載の医薬組成物。
- 前記免疫応答の回復が、対照と比較して、肝臓における1以上のHBV抗原に対して特異的なCD8+T細胞の増加である、請求項20に記載の使用。
- 薬品として使用するための、請求項1から16に記載のオリゴヌクレオチドコンジュゲート、又は請求項17に記載のオリゴヌクレオチド、又は請求項18に記載の医薬組成物。
- HBV、HCV及びHDVなどのウイルス性肝炎感染;マラリア、トキソプラズマ症、リーシュマニア症及びトリパノソーマ症などの寄生虫感染;又は肝臓癌もしくは肝臓の転移の治療又は予防に使用するための、請求項1から16に記載のオリゴヌクレオチドコンジュゲート、又は請求項17に記載のオリゴヌクレオチド、又は請求項18に記載の医薬組成物。
- HBV、HCV及びHDVなどのウイルス性肝炎感染;マラリア、トキソプラズマ症、リーシュマニア症及びトリパノソーマ症などの寄生虫感染;又は肝臓癌もしくは肝臓の転移の治療又は予防用の薬品を調製するための、請求項1から16に記載のオリゴヌクレオチドコンジュゲートの使用、又は請求項17に記載のオリゴヌクレオチド、又は請求項18に記載の医薬組成物。
