本発明の融合タンパク質は、例えば、固相ペプチド合成(例えば、Merrifield固相合成)又は組み換え産生によって得ることができる。組み換え産生については、例えば、上記の融合タンパク質(フラグメント)をコードする1つ以上のポリヌクレオチドを単離し、そして、宿主細胞において更にクローニング及び/又は発現させるために1つ以上のベクターに挿入する。このようなポリヌクレオチドは、従来の手順を用いて容易に単離及び配列決定することができる。1つの実施態様では、本発明のポリヌクレオチドのうちの1つ以上を含むベクター、好ましくは、発現ベクターが提供される。当業者に周知の方法を用いて、適切な転写/翻訳制御シグナルと共に、融合タンパク質(フラグメント)のコード配列を含む発現ベクターを構築することができる。これら方法は、インビトロ組み換えDNA技術、合成技術、及びインビボ組み換え/遺伝子組み換えを含む。例えば、Maniatis et al., Molecular Cloning: A Laboratory Manual, Cold Spring Harbor Laboratory, N.Y. 以下は、本発明の方法及び組成物の例である。上に提供した一般的記載を鑑みて、様々な他の実施態様を実施できることが理解される。
標準的な方法を用いて、Sambrook et al., Molecular cloning: A laboratory manual; Cold Spring Harbor Laboratory Press, Cold Spring Harbor, New York, 1989に記載の通りDNAを操作した。分子生物学的試薬は、製造業者の指示書に従って用いた。ヒト免疫グロブリン軽鎖及び重鎖のヌクレオチド配列に関する一般的情報は、Kabat, E.A. et al., (1991) Sequences of Proteins of Immunological Interest, Fifth Ed., NIH Publication No 91-3242に記載されている。
DNA配列は、二本鎖配列決定によって決定した。
所望の遺伝子セグメントは、状況によって、適切なテンプレートを用いるPCRによって作製したか、又は自動遺伝子合成による合成オリゴヌクレオチド及びPCR産物からGeneart AG(Regensburg, Germany)によって合成した。正確な遺伝子配列が入手不可能であった場合、最も近縁なホモログの配列に基づいてオリゴヌクレオチドプライマーを設計し、そして、適切な組織に由来するRNAからRT−PCRによって遺伝子を単離した。唯一の制限エンドヌクレアーゼ切断部位に隣接する遺伝子セグメントを標準的なクローニング/シーケンシングベクターにクローニングした。形質転換された細菌からプラスミドDNAを精製し、そして、紫外分光法によって濃度を求めた。サブクローニングした遺伝子フラグメントのDNA配列を、DNAシーケンシングによって確認した。それぞれの発現ベクターにサブクローニングできるように好適な制限部位を有する遺伝子セグメントを設計した。全てのコンストラクトを、真核細胞において分泌するためのタンパク質を標的とするリーダーペプチドをコードする5’末端DNA配列を有するように設計した。配列番号39〜47は、例示的なリーダーペプチド及びそれらをコードするポリヌクレオチド配列を提示する。
IL−2受容体の結合親和性を調べるために、ヘテロ二量体IL−2受容体を発現させるツールを作製した。「knobs-into-holes」技術(Merchant et al., Nat Biotech. 16, 677-681 (1998))を用いて、IL−2受容体のβサブユニットを、ヘテロ二量体化するように操作したFc分子(Fc(ホール))(配列番号21及び22(ヒト)、配列番号27及び28(マウス)、並びに配列番号33及び34(カニクイザル)を参照)に融合させた。次いで、IL−2受容体のγサブユニットを、Fc(ホール)とヘテロ二量体化するFc(ノブ)バリアント(配列番号23及び24(ヒト)、配列番号29及び30(マウス)、並びに配列番号35及び36(カニクイザル)を参照)に融合させた。次いで、このヘテロ二量体Fc−融合タンパク質を、IL−2/IL−2受容体相互作用を分析するための基質として用いた。IL−2Rαサブユニットは、AcTev切断部位及びAvi Hisタグを有する単量体鎖(配列番号25及び26(ヒト)、配列番号31及び32(マウス)、並びに配列番号37及び38(カニクイザル))として発現させた。それぞれのIL−2Rサブユニットを、IL−2Rβγサブユニットコンストラクトについては血清を含み、そして、αサブユニットコンストラクトについては血清を含まない、HEK EBNA293細胞において一時的に発現させた。IL−2Rβγサブユニットコンストラクトは、プロテインA(GE Healthcare)、次いで、サイズ排除クロマトグラフィー(GE Healthcare、Superdex 200)によって精製した。IL−2Rαサブユニットは、NiNTAカラム(Qiagen)においてHisタグを介して、次いで、サイズ排除クロマトグラフィー(GE Healthcare、Superdex 75)によって精製した。様々な受容体コンストラクトのアミノ酸及び対応するヌクレオチド配列を配列番号21〜38に示す。
遺伝子合成及び/又は古典的な分子生物学的技術によってDNA配列を作製し、そして、MPSVプロモータ及び上流の合成polyA部位の制御下で哺乳類発現ベクターにサブクローニングし、各ベクターは、EBV OriP配列を有していた。リン酸カルシウムトランスフェクションを用いて、指数関数的に成長しているHEK293−EBNA細胞を哺乳類発現ベクターでコトランスフェクトすることによって、以下の実施例で適用する融合タンパク質を産生した。あるいは、懸濁液中で成長しているHEK293細胞を、ポリエチレンイミン(PEI)によってそれぞれの発現ベクターでトランスフェクトした。あるいは、無血清培地で産生するために、安定的にトランスフェクトされたCHO細胞プール又はCHO細胞クローンを用いた。次いで、上清から融合タンパク質を精製した。簡潔に述べると、20mM リン酸ナトリウム、20mM クエン酸ナトリウム(pH7.5)において平衡化されたプロテインA(HiTrap ProtA、GE Healthcare)を用いる1段階アフィニティーによって、融合タンパク質を精製した。上清の投入後、カラムを、まず20mM リン酸ナトリウム、20mM クエン酸ナトリウム(pH7.5)で洗浄し、続いて13.3mM リン酸ナトリウム、20mM クエン酸ナトリウム、500mM 塩化ナトリウム(pH5.45)で洗浄した。融合タンパク質を、20mM クエン酸ナトリウム、100mM 塩化ナトリウム、100mM グリシン(pH3)又は10mM クエン酸ナトリウム(pH3.0)で溶出した。画分を0.5M Na2HPO4(pH8.0)(1:10)で中和し、プールし、そして、最終製剤バッファ:20mM ヒスチジン、140mM NaCl(pH6.0)中で、サイズ排除クロマトグラフィー(HiLoad 16/60 Superdex 200、GE Healthcare、又はHiLoad 26/60 Superdex 200、GE Healthcare)によって精製した。精製タンパク質サンプルのタンパク質濃度は、アミノ酸配列に基づいて計算したモル吸光係数を用いて、280nmにおける光学濃度(OD)を測定することによって求めた。また、分子量は、アミノ酸配列に基づいて求めた。融合タンパク質の純度及び分子量は、還元剤(5mM 1,4−ジチオトレイトール(dithiotreitol))の存在下及び非存在下でSDS−PAGEによって分析し、そして、クマシーブルー(SimpleBlue(商標)SafeStain、Invitrogen)で染色した。NuPAGE(登録商標)Pre-Castゲルシステム(Invitrogen)を、製造業者の指示書に従って用いた(4〜20% Tris−グリシンゲル又は3〜12% Bis−Tris)。あるいは、分子の純度及び分子量は、製造業者の指示書に従ってCaliper LabChip GXIIシステム(Caliper Lifescience)を用いて、還元剤の存在下及び非存在下でCE−SDS分析によって分析した。融合タンパク質サンプルの凝集量は、25℃で、2mM MOPS、150mM NaCl、0.02% NaN3(pH7.3)ランニングバッファ中でSuperdex 200 10/300GL分析サイズ排除カラム(GE Healthcare)を用いて分析した。あるいは、抗体サンプルの凝集量は、25℃で25mM K2HPO4、125mM NaCl、200mM L−アルギニン一塩酸塩、0.02%(w/v) NaN3(pH6.7)ランニングバッファ中でTSKgel G3000 SW XL分析サイズ排除カラム(Tosoh)を用いて分析した。
以下の条件下で、組み換えIL−2Rβγヘテロ二量体を用いて、ヒト、マウス、及びカニクイザルIL−2Rβγヘテロ二量体に対する融合タンパク質の親和性を、Biacore T200(GE Healthcare)において表面プラズモン共鳴(SPR)によって求めた:リガンド:SAチップに固定化されたビオチン化ヒト、マウス、及びカニクイザルIL−2Rβノブγホールヘテロ二量体(固定化レベルは、それぞれ、194、114、及び116RUであった)、検体:DP47GS IgG-IL-2(配列番号13、15、及び19を参照)、DP47GS IgG-(IL-2)2(配列番号17及び19を参照)、DP47GS IgG-IL-2 N88D(配列番号15、19、及び48を参照)、DP47GS IgG-(IL-2 N88D)2(配列番号19及び50を参照)、及びDP47GS IgG-(IL-2 E95A)2(配列番号19及び52を参照)、温度:25℃、バッファ:HBS−EP、検体濃度:100nMから1.2nMに低下(1:3希釈)、流速:30μL/分、会合:120s、解離:2つの最高濃度については600s、そして、より低濃度については120s、再生:60s 3M MgCl2、フィッティング:1:1ラングミュア結合モデル、RI≠0、Rmax=ローカル。親和性は、反応速度定数Kon及びkoffに基づいて求めた。また、以下の条件下で、組み換え単量体IL−2Rαサブユニットを用いて、ヒト、マウス、及びカニクイザルIL−2Rαサブユニットに対する融合タンパク質の親和性を求めた:リガンド:アミンカップリングを介してCM5チップに固定化されたヒト、マウス、及びカニクイザルIL−2Rαサブユニット(固定化レベルは、それぞれ、240、245、及び220RUであった)、検体:DP47GS IgG-IL-2(配列番号13、15、及び19を参照)、DP47GS IgG-(IL-2)2(配列番号17及び19を参照)、DP47GS IgG-IL-2 N88D(配列番号15、19、及び48を参照)、DP47GS IgG-(IL-2 N88D)2(配列番号19及び50を参照)、及びDP47GS IgG-(IL-2 E95A)2(配列番号19及び52を参照)、温度:25℃、バッファ:HBS−EP、検体濃度300nMから0.41nMに低下(1:3希釈)、流速:30μL/分、会合:120s、解離:180s、再生:10mM グリシン(pH1.5)、60秒間。親和性は、定常状態分析によって求めた。
高親和性三量体Il−2受容体は、α(IL−2RA、CD25)、β(IL−2RB、CD122)、及びγ(IL−2RG、CD132)鎖で構成され、そして、約10pMのKDを有する。CD25単独では、IL−2に対して低い親和性(KD約10nM)しか有しない。IL−2RAの非存在下で幾つかの細胞型において発現するIL−2RB/IL−2RG二量体もIL−2に結合するが、中親和性(KD約1nM)である。IL−2受容体を介するシグナル伝達は、IL−2RB鎖及びIL−2RG鎖によって媒介される。結晶構造解析から、IL−2RAは、IL−2RBにもIL−2RGにも接触しないと思われる。三量体受容体の協同性の基礎は、CD25がIL−2RB及びIL−2RGに提示するために細胞表面においてIL−2を捕捉するとき、あるいはIL−2の立体構造がCD25に誘導されて変化するときのエントロピー低減であり、これによって複合体が安定化されると提唱されている。FOXP3+制御性CD4+T細胞では、受容体のβ鎖及びγ鎖に比べて化学量論的に大きく過剰のIL−2RAが存在し、これは、α鎖の二量体、又は更に大きな複合体が、IL−2の結合を支援するという仮説を支持する。また、ある細胞におけるCD25は、高親和性細胞間相互作用で、IL−2を別の細胞におけるIL−2RB/IL−2RG二量体に提示することができるという証拠も存在し、これは、高親和性IL−2受容体を構成する3本の鎖間の独自の関係を強調するものである。
三量体IL−2RのIL−2誘導オリゴマー化に続いて、それぞれ、IL−2RB及びIL−2RGの細胞内ドメインと連結するJAK1及びJAK3細胞質タンパク質チロシンキナーゼが活性化される。これらキナーゼは、次にリン酸化されるSTAT5a及びSTAT5bに対するドッキング部位として作用する特定のIL−2RBチロシン残基をリン酸化する。幾つかのシグナル伝達経路のIL−2誘導活性化により、最終的に、IL−2/IL−2R経路に関連する様々な機能に寄与する標的遺伝子が転写される。様々な細胞型が様々なレベルのIL−2受容体IL−2RA分子及びIL−2RB分子を発現する(図6及び7)ので、高及び中親和性受容体の様々な組み合わせによって媒介されるIL−2に対する統合シグナル伝達応答について理解するために、多色フローサイトメトリーによって個々の細胞内のpSTAT5aレベルを測定した。
新鮮ヒト末梢血単核球(PBMC)を、NK細胞及びCD8+T細胞の生存及び成長に好ましいことが知られている条件で、IL−2以外の更なる刺激なしに、インビトロで培養した。ヒトNK細胞は、検出可能なCD56をほとんど又は全く発現しないもの(CD56−/dimと称する)及び高レベルのCD56を発現する「活性化」サブセット(CD56brightと称する)に細分することができ;血液中のNK細胞の大部分は、CD56−/dimであり、CD56brightとして分類されるのはわずかである。CD56brightである「活性化」NK細胞は、CD56を発現しないNK細胞(すなわち、CD56−/dim)の前駆体であると考えられる。DP47GS IgG-(IL-2)2及びDP47GS IgG-(IL-2N88D)2(それぞれ、0、5、50、及び500ng/mL)と共にインビトロで6日間後、NK CD56−/dim細胞、NK CD56bright細胞、及びCD8+T細胞の成長及び生存における差を検出した。CD56bright細胞(メジアン:101×103対38×103、p<0.005)(図16A)、及び、CD8+T細胞(メジアン:細胞13.6×103個対細胞2.5×103個、p<0.0001)(図16B)でも、最高増加を刺激するDP47GS IgG-(IL-2)2による刺激間に細胞数の著しい差がみられた。
免疫低下宿主として軽度に放射線照射された新生仔NOD.PrkdcscidIL2rgnull(NSG)マウスを用い、そして、ヒト胎児肝臓CD34+幹細胞をIP注射で移植することによって、ヒト化マウスを構築した。8〜10週齢において、各マウス由来の血液を試験してヒト生着が生じたことを確認した。ProleukinとDP47GS IgG-(IL-2)2とを比較したところ、インビボで影響を受けた一次細胞は、NK細胞及びTregであり;DP47GS IgG-(IL-2)2処置後、NK細胞及びTregの両方が実質的に増加し、NK細胞は比例的に大きく応答することが見出された。Proleukinのインビボにおける効果は、同様であったが、群内のNK細胞及びTregに対する効果は、それほど一致しておらず、そして、増加の程度は、DP47GS IgG-(IL-2)2と比べて低かった。また、ヒト化マウスは、IL−2融合タンパク質のインビボにおける効果を識別することができた。野性型DP47GS IgG-(IL-2)2は、Treg(図17A)及びNK細胞(図17B)の両方を増加させ、一方、DP47GS IgG-(IL-2N88D)2は、NKに対して全く効果を有していなかった(図17B)が、野性型IgG-(IL-2)2よりも著しく大きくTregを増加させた(図17A)。
ヒト末梢血で観察された通り、IL−2(Proleukin)で刺激したカニクイザル末梢血中のTregサブセットでは、pSTAT5aが優先的かつ同様に用量依存的に誘導される(データは示さない)。3つのTregサブセットにおいてDP47GS IgG-(IL-2)2及びDP47GS IgG-IL-2のpSTAT5aを誘導する能力を直接比較すると、pSTAT5aの最高レベルの50%に達するのに必要なDP47GS IgG-(IL-2)2は、DP47GS IgG-IL-2よりも2〜8倍少なかった。様々なカニクイザルTregサブセットにおけるDP47GS IgG-IL-2対DP47GS IgG-(IL-2)2 によるpSTAT5aの活性化についてのEC50を表4に要約し、結果は、ヒト末梢血でみられたもの(表3)と著しく類似している。
マウスにおいて機能試験を実施する前に、野性型及びN88Dの一価及び二価IL−2免疫コンジュゲートの薬物動態(PK)特性を、NOD、NOD.scid、及びNOD.scid.Il2rα-/-マウスにおいて評価した(図23)。
様々な分子形態及び立体構造のヒトIL−2のインビボにおいてFoxP3+Tregを刺激する能力を比較するために、ビヒクル、Proleukin(組み換えヒトIL−2)、DP47GS IgG-IL-2、DP47GS IgG-(IL-2)2、又はDP47GS IgG-(IL-2N88D)2をマウスに皮下注射し、そして、既に決定した24時間の最適時間において、細胞表面CD25及び細胞内FOXP3の発現の変化についてTregを評価した(図24)。幼若健常BALB/cマウス(n=3/処置群、n=4/ビヒクルコホート)に、Proleukin(20,000又は100,000IU/マウス)、DP47GS IgG-IL-2(60、300、又は1500IU/マウス)、DP47GS IgG-(IL-2)2(60、300、又は1500IU/マウス)、又はDP47GS IgG-(IL-2N88D)2(60、300、又は1500IU/マウス)を皮下注射した。用量は、0.5% 滅菌濾過マウス血清を含有する滅菌PBS(pH7.2) 100μLで構成されるビヒクル中で送達した。24時間後、マウスを安楽死させ、そして、脾臓を切除した。脾細胞の単一細胞懸濁液をL−15培地 1mL中で作製し、そして、更に加工するまで氷上で保管した。単一細胞懸濁液の濾過アリコート 40μLをFACSチューブに移し、そして、FACSバッファ 2mLで洗浄した(600×g、5分間)。次いで、細胞表面抗原:CD4(クローンRM4−5、蛍光色素(fluorochrome) A700)、CD25(eBio7D4、Af488)、CD44(IM7、e605)、CD62L(MEL−14、PE)、ICOS(C398.4A、PE/Cy7)、CD103(2E7、APC)に対する蛍光色素コンジュゲート抗体と共にサンプルをインキュベートした。染色は、FACSバッファ(PBS(pH7.2)+0.2% BSA) 100μL中4℃で30分間実施した。細胞表面染色後、サンプルをFACSバッファ 4mLで洗浄(600×g、5分間)した後、(eBioscienceの細胞内染色プロトコールに従って)細胞内染色した。簡潔に述べると、サンプルを固定/透過処理バッファ(eBioscience #00-5521) 200μL中に再懸濁させ、そして、4℃で1時間インキュベートした。1×透過処理バッファ(eBioscience #00-8333) 1mLをサンプルに添加した後、FACSバッファ 3mLを添加し、そして、洗浄(600×g、5分間)した。細胞内抗原Ki−67(B56、PerCP Cy5.5)及びFOXP3(FJK−16S、e450)を、1×透過処理バッファ 100μL中4℃で1時間染色した。サンプルをFACSバッファ 4mLで洗浄(600×g、5分間−2回)し、そして、BD Fortessa(商標)分析機においてデータを取得し、そして、FlowJoソフトウェア(FlowJo, LLC)を用いて解析した。Tregをリンパ球ゲート内のシングレットからCD4+FOXP3+と定義し;この集団から、全てのサンプルについてCD25及びFOXP3の平均蛍光強度(MFI)を計算した。
非ヒト霊長類において機能試験を実施する前に、IL−2免疫コンジュゲートの薬物動態(PK)特性を、生物学的ナイーブ健常成体カニクイザルにおいて評価した(図25)。サル(n=2/用量)に、0.5% カニクイザル血清を含有するPBS中滅菌DP47GS IgG-IL-2又はDP47GS IgG-(IL-2)2のショートボーラスを静脈内(i.v.)注射し、そして、30分〜72時間にわたる注射後の様々な時点で失血させた。ヒトIL−2を捕捉するために、マウス抗ヒトIL−2mAb(BD Pharmingen、カタログ#555051、クローン5344.111)を用いて96ウェルプレートをコーティングして、カニクイザル血清サンプル中のヒトIL−2を評価した。次いで、ビオチン化マウス抗ヒトIL−2mAb(BD Pharmingen、カタログ#555040、クローンB33-2)を用いてヒトIL−2を検出した。IL−2結合を可視化し、そして、ユウロピウムコンジュゲートストレプトアビジンを用いて定量した。アッセイにおけるカニクイザル血清を用いた検出レベルは、0.05ng/mL IL−2であった(図中点線)。処置前に採取したカニクイザル血清は、検出可能なIL−2を有していなかった(したがって、<0.05ng/mL)。
インビボにおいてDP47GS IgG-IL-2で処置したカニクイザルは、Tregの絶対数が用量依存的に増加したことに加えて、投与後7日目にベースラインを超えて倍数増加した(それぞれ、図26A及び26B)。3〜6年齢の両性別の正常健常カニクイザルを全ての試験で用い、そして、1回を超えて用いた動物は存在しなかった。麻酔下にある間、様々な用量のDP47GS IgG-IL-2(n=4〜6)又はビヒクル(n=3)を外側足背にSC注射した。DP47GS IgG-IL-2の個々の用量は、体重に基づいており、そして、0.5% 滅菌正常カニクイザル血清を含有する滅菌PBS(pH7.2)のビヒクル中で注射用に製剤化したものであった。処置後の様々な時点で血液サンプルを回収し、そして、Advia Automated Hematology Analyzer、並びに上に詳述した(表4についての実験手順)細胞表面及び細胞内マーカーを用いて血液学的変化(CBC及び差異)について試験した。処置後7日目における全血CD4+CD25+FOXP3+制御性T細胞の変化を、全血1mm3当たりの絶対細胞数(図26A)及びTregの倍数変化(図26B)として図26に示し;全てのデータを平均±SDとして表す。より高用量の25μg/kg及び36μg/kg DP47GS IgG-IL-2では、それぞれ、ほぼ3倍(111〜255%の範囲、n=6)及び4倍(110〜470%の範囲、n=6)の平均Treg増加が観察された。DP47GS IgG-IL-2のSC用量を更に低減すると(12、6、及び2μg/kg)、Treg数の変化及び倍数変化は、対応して減少し続けた。理論に束縛されることは望まないが、12μg/kg DP47GS IgG-IL-2の用量によるTregの〜2倍の増加(67〜133%の範囲、n=6)は、幾つかの自己免疫疾患及び炎症性疾患に対して望ましいTregの増加を表し得る。ヒトにおいてTreg数は広い範囲で存在し(血液1mm3当たり20〜90個のTreg;CD4+T細胞の4〜10%)、そして、個体内のIL−2によって誘導されるTregの増加によって、機能抑制が全体的に増大すると推測することが合理的である。
低用量IL−2処置のインビボにおける細胞特異性は、重要なパラメータである。本発明者らは、カニクイザル又はマウスに投与した後の様々な時点で採取した血液中のエクスビボpSTAT5aレベルを測定することによって、DP47GS IgG-IL-2又はProleukinによって誘導されるインビボ細胞活性化を高感度でモニターできることを見出した。インビトロでモニターすることができる全ての細胞集団のインビボ応答(図8〜10)もエクスビボで調べることができる。
機能的に活性のTregは、免疫抑制性サイトカインCTLA−4、IL−10、及びTGFβを産生することができ、そして、転写因子FOXP3の安定な発現を必要とする。ヒト及びカニクイザルTregにおけるFOXP3の発現は、FOXP3遺伝子座内のTreg特異的脱メチル化領域(TSDR)における10個のCpG DNAメチル化部位の脱メチル化に依存する。同様に、CTLA−4の発現及び産生は、ヒト及びカニクイザルのCTLA−4遺伝子座における7個のCpG DNAメチル化部位の脱メチル化に依る。したがって、本発明者らは、血液中のTreg数を著しく増加させることが既に示されている用量及び回数で投与されたDP47GS IgG-IL-2(n=4、25μg/kg s.c.)及びDP47GS IgG-(IL-2)2(n=4、6μg/kg s.c.)による処置前及び処置後に、様々なCD4+T細胞サブセットにおけるFOXP3及びCTLA−4の脱メチル化状態を評価した。試験では生物学的ナイーブ成体雄カニクイザルのみを用い、そして、その体重は、9.1〜10.9kgであった。処置は、初期ベースライン血液回収の3週間後に投与し、そして、処置後血液を4〜5日後に回収した。ヘパリン化血液 30mL由来のPBMCを、FACSAria(商標)(Becton Dickinson)を用いて、Treg(CD4+CD25+CD127low)、メモリーTエフェクター(CD4+CD45RA−)、ナイーブTエフェクター(CD4+CD45RA+)、及びCD4+CD25−CD127−細胞を含む関連CD4+サブセットに選別した。
DP47GS IgG-(IL-2N88D)2のPK特性を、生物学的ナイーブカニクイザルにおいて評価した(図37)。サル(n=2/用量)に、0.5% カニクイザル血清を含有するPBS中30又は100μg/kg 滅菌DP47GS IgG-(IL-2N88D)2を静脈内(iv)及び皮下(sc)注射し、そして、30分〜14日間にわたる注射後の様々な時点で失血させた。上記の通り、カニクイザル血漿サンプル中のヒトIL−2を評価した。アッセイにおけるカニクイザル血漿を用いた検出レベルは、0.05ng/mL IL−2であった。処置前に採取したカニクイザル血漿は、検出可能なIL−2を有していなかった(したがって、<0.05ng/mL)。
野性型DP47GS IgG-IL-2及びDP47GS IgG-(IL-2)2によってインビボで処置したカニクイザルにおける既に記載した試験では、ヒト全血アッセイにおけるインビトロ活性を反映していたTregの絶対数の2〜4倍の用量依存的増加(それぞれ、図26及び33)、すなわち、6〜9倍のインビトロ差(表2及び3)が示された。ヒト全血におけるインビトロ効力の喪失(表3)から、DP47GS IgG-(IL-2N88D)2について30及び100μg/kgのインビボ用量を選択した。以前の試験と同様に、0.5% 滅菌正常カニクイザル血清を含有する滅菌PBS(pH7.2)のビヒクル中で注射用に製剤化した、体重に基づく個々の用量のDP47GS IgG-(IL-2N88D)2を、iv又はsc経路によって注射した(n=2/用量及び注射経路)。処置前(−4及び−5日目)、処置直前(0日目)、及び処置後の様々な時点(1〜14日目)に血液サンプルを回収し、そして、血液学的変化(CBC及び差異)並びに既に記載した細胞表面及び細胞内マーカーについて試験した。
上記の通り、IL−2処置のインビボ細胞特異性は、重要なパラメータであり、そして、本発明者らは、投与後の様々な時点で採取した血液中のpSTAT5a、細胞表面CD25、及び細胞内Ki−67をエクスビボにおいて測定することによって、インビボにおける細胞サブセットの活性化をモニターできることを実証した。
ヒトにおけるProleukinは、1×106〜4.5×106IU/ヒトの用量で毎日投与したとき、好酸球増多を刺激する。カニクイザルでは、高用量のProleukinによる反復処置後又は226pmol/kgの用量のDP47GS IgG-IL-2による単回処置後に、動物の100%で同様の好酸球増多がみられた(図44)。図44の結果は、好酸球数が正常であるか増加しているかにかかわらず、試験した全ての動物を表す(コロニーベースラインは、試験前の動物:(n=44及びn=30)及び様々なIL−2処置群(n=5〜9)について求めた)。Proleukin及びDP47GS IgG-IL-2の効果とは全く対照的に、170及び570pmol/kg(それぞれ、30及び100μg/kg)のインビボ用量のDP47GS IgG-(IL-2N88D)2による好酸球に対する効果は、実質的に存在していなかった。DP47GS IgG-(IL-2N88D)2によって全身好酸球増多が誘導されなかったことは、これら様々な処置によって誘導されるTreg拡大の程度に鑑みて特筆すべきである(図43において比較)。
予備的な試験において、本発明者らは、4000IUの用量のDP47GS IgG-IL-2が、インビボにおいてマウスFOXP3+制御性T細胞を活性化することを観察し;次いで、この用量を用いて、マウスにおける古典的T依存性免疫応答、すなわち、遅延型過敏(図45)及びIgG抗KLH応答(図46)を抑制する能力を評価した。

- (i)免疫グロブリン分子と、(ii)野性型IL−2分子と比べたとき、中親和性IL−2受容体に対する変異体IL−2分子の親和性を低減するアミノ酸変異を含む2つの変異体インターロイキン−2(IL−2)分子とを含む融合タンパク質。

- 移植拒絶反応又は移植片対宿主病の処置又は予防において使用するための、請求項1〜29若しくは34のいずれか一項記載の融合タンパク質又は請求項35記載の医薬組成物。
- それを必要としている個体における疾患を処置するための医薬を製造するための、請求項1〜29又は34のいずれか一項記載の融合タンパク質の使用。
- 前記疾患が、自己免疫疾患である、請求項40記載の使用。
