Abstract
Description
a)大豆粕に水分を添加して熱処理する段階;
b)前記熱処理された大豆粕を冷却した後、バチルス・アミロリケファシエンスK2G菌株を接種する段階; 及び
c)前記大豆粕に接種されたバチルス・アミロリケファシエンスK2G菌株を固体培養して発酵大豆粕を得る段階を含むことを特徴とする。
本発明者らは、タンパク分解酵素の生成能に優れた菌株を分離するために、多様な伝統的な発酵食品(キムチ、味噌類、伝統的な地酒、塩辛など)から約3000種の微生物を分離し、これらのうち同定を通じて確認された約1308種の飼料適合性(短尾飼料協会、プロバイオティクス)バチルス菌を中心にタンパク分解酵素の発現が高く、抗菌活性を有し、生育速度が速い多機能菌株を見出そうとした。
家畜に対して食中毒を起こす代表的な病原菌である大腸菌及びサルモネラ菌の生育や増殖を抑制することができる菌株を分離し、発酵大豆粕の生産に適用してサルモネラ菌がない無毒性製品を開発するために、前記実施例1で分離した14種のタンパク分解酵素の高生産菌株を対象に、病原菌に対する抗菌活性を測定した。
前記実施例2の結果によると、寒天培地上でCJ823はサルモネラ菌に対して優れた抗菌活性を示した。しかし、実際の大豆粕発酵過程の中でもサルモネラ菌に対する増殖抑制力を表すかどうかを確認するために下記実験を行った。
前記実施例1〜3で選別されたCJ823は、タンパク分解酵素を高濃度で生産し、病原菌に対して優れた抗菌活性を示すが、発酵過程中に生産される酵素により原料大豆の糖質、大豆タンパク由来のレバン型のフルクタン(levan form fructan)とポリグルタミン酸(polyglutamate)の重合により粘着性粘液質が生成された。大規模な工業化の過程で、このような粘液質の過量生成は、撹拌を困難にして発酵物内の溶存酸素、温度などを制御するのに問題となり、移送を困難にするなどの問題が発生した。
粘液質生成能の低下したU304変異株の同定のために、新たなNA寒天培地に菌を接種し、37℃で16時間培養した。形成されたコロニーは、0.8%のNaCl滅菌溶液で希釈した後、63種の乾燥培地及び生化学反応物で構成されたBCL ID card(bioMNitek Inc.、Hazewood、USA)に注入し、その結果は15分間隔でVITEK 2 Compact software(bioMVitek)に統合的に保存、分析され、14時間後に同定が完了した。
Claims (11)
- 固体発酵により発酵大豆粕を製造する方法において、寄託番号KCCM11471Pとして寄託されたバチルス・アミロリケファシエンスK2G菌株を大豆粕に接種する段階を含むことを特徴とする発酵大豆粕の製造方法。
- 下記段階を含む請求項1に記載の方法:
a)大豆粕に水分を添加して熱処理する段階;
b)前記熱処理された大豆粕を冷却した後、バチルス・アミロリケファシエンスK2G菌株を接種する段階; 及び
c)前記大豆粕に接種されたバチルス・アミロリケファシエンスK2G菌株を固体培養して発酵大豆粕を得る段階。 - 段階a)において水分が添加された大豆粕が30〜80%(v/w)の水分含量を有する、請求項2に記載の方法。
- 段階a)の熱処理が、大豆粕を70〜130℃で10〜30分間熱処理することである、請求項2に記載の方法。
- 段階b)において大豆粕が30〜50℃に冷却される、請求項2に記載の方法。
- 段階b)においてバチルス・アミロリケファシエンスK2G菌株が接種直後に105〜109 CFU/gの菌数となるように接種される、請求項2に記載の方法。
- 段階c)において固体培養が30〜45℃で12〜48時間行われる、請求項2に記載の方法。
- 段階c)で得られた発酵大豆粕を乾燥及び粉砕する段階をさらに含む、請求項2に記載の方法。
- 寄託番号KCCM11471Pとして寄託されたバチルス・アミロリケファシエンスK2G菌株。
- 請求項1〜8のいずれか一項に記載の方法により製造された発酵大豆粕。
- 請求項10による発酵大豆粕を含む飼料組成物。
