Description
本発明の発酵グアーミールの製造方法は、水分含量が調節されたグアーミールを熱処理する段階を含む。熱処理の温度に係る発酵グアーミールの細菌数及びトリプシンインヒビター(trypsin inhibitor、TI)を測定した。
水分含量及び発酵温度に係る発酵グアーミールの栄養成分を測定した。
グアーミール100gを用意して、グアーミールの水分含量が40~45%(w/w)になるように水分を供給した。続いてグアーミールを80~95℃で30分間熱処理した後、40℃に冷却させた。前記グアーミール重量の10重量%のバチルス・サブチリス1種(Bacillus subtilis ATCC21770)とバチルス・アミロリケファシエンス4種の(Bacillus amyloliquefaciens K2G、ATCC 23842、ATCC 23843、ATCC 23845)培養液を前記冷却されたグアーミールにそれぞれ接種し、最終の発酵水分を45~50%(w/w)にして、よく混ぜた後、37℃~45℃で14時間~16時間恒温恒湿が維持される条件で発酵させた。本発明に係る水分含量の調節段階、熱処理段階、及び発酵段階が行われていないグアーミール原料群は、対照群として用いた。
グアーミール内には1%~3%のグアーガムが存在し、グアーガムの粘度は、消化器官内で消化物の流れを遅くして消化率を阻害すると知られている。したがって、グアーミール内グアーガムの粘度を減少させうる酵素が本出願の発酵グアーミールに添加されてもよい。
グアーミールは4%~6%のオリゴ糖を含有しており、これらのオリゴ糖の中で難消化性オリゴ糖、すなわちガラクトオリゴ糖(galacto-oligosaccharides、以下GOSと呼ぶ)であるラフィノース(raffinose)とスタキオース(stachyose)が2%~3%である。前記難消化性オリゴ糖は、腸内有害微生物の栄養源として用いられたり、下痢を引き起こすこともあるため、グアーミール内その含量を減少させることが必要である。
本出願に従って製造された発酵グアーミール内トリプシンインヒビター(Trypsin inhibitor、以下TI表記)の含量とインビトロ消化率を測定した。
グアーミールは大豆粕のタンパク質含量と同様またはより高いタンパク質を含有しているが、多くの抗栄養因子と高分子量の低消化率タンパク質を含むため、消化吸収率が低くて幼い家畜に用いるのに難しいという欠点がある。低消化率タンパク質は加水分解されたペプチドの形態に変換したり、消化が容易な低分子量のタンパク質に分解して、タンパク質の消化率を改善させうる。
グアーミール(Guar meal、GM)と大豆粕(Soybean meal、SBM)の割合がそれぞれ1:9、5:5、9:1になるように混合した混合物を用意した後、混合物の水分含量が40~45%(w/w)になるように水分を供給した。前記混合物をよく混ぜた後、これを90~95℃で25~30分間熱処理して、熱処理後には冷却させた。前記冷却されたグアーミール/大豆粕混合物に5重量%~20重量%のバチルス・アミロリケファシエンス2種(Bacillus amyloliquefaciens K2G、ATCC 23843)の培養液をそれぞれ接種し、よく混ぜた後、37℃~45℃で14時間~16時間恒温恒湿が維持される条件で発酵させた。発酵されたグアーミール/大豆粕混合物の栄養成分は、下記表7に示した。
発酵グアーミールの効果を評価するために肉鶏の飼養試験を行った。飼養試験を行なうために4日間適応期間を与えて、総21日(3週)の間に大豆粕と大豆粕の3%、5%、7%を発酵グアーミールに代替した飼料を給餌して肉鶏の体重と給餌率を確認した。肉鶏は80羽であって、4処理×4繰り返し×5羽で試験設計した。
本件出願は、以下の構成の発明を提供する。
(構成1)
次の段階を含む発酵グアーミールの製造方法:
(1)グアーミールを前処理する段階;
(2)前記前処理されたグアーミールにバチルス菌株を接種する段階;及び
(3)前記グアーミールに接種された菌を固体培養して発酵グアーミールを収得する段階。
(構成2)
前記段階(1)の前処理する段階が、
(1-1)水をグアーミールに添加して水分含量を調節する段階;
(1-2)前記水分含量が調節されたグアーミールを熱処理する段階;及び
(1-3)前記熱処理されたグアーミールを冷却させる段階を含む、構成1に記載の方法。
(構成3)
前記段階(1-1)の後のグアーミールの水分含量が、40~45%である、構成2に記載の方法。
(構成4)
前記段階(1-2)の熱処理が、90℃~95℃で行われる、構成2に記載の方法。
(構成5)
前記段階(1-3)の冷却が、固体発酵可能な温度まで下げるものである、構成2に記載の方法。
(構成6)
前記段階(3)の固体培養が、30℃~50℃の温度で15~25時間行うものである、構成1に記載の方法。
(構成7)
前記段階(3)の後に、前記発酵グアーミールを乾燥して粉砕する段階(4)をさらに含む、構成1に記載の方法。
(構成8)
前記バチルス菌株が、バチルス・サブチリスまたはバチルス・アミロリケファシエンスである、構成1に記載の方法。
(構成9)
前記発酵グアーミールが、前記(1)~(3)の段階が行われていないグアーミール内トリプシンインヒビター(trypsin inhibitor、TI)の含量を基準に、30~50%水準のトリプシンインヒビターを含むか、または
前記発酵グアーミールが、前記(1)~(3)の段階が行われていないグアーミール内難消化性オリゴ糖の含量を基準に、10~40%水準の難消化性オリゴ糖を含む、構成1に記載の方法。
(構成10)
構成1~9のいずれか一項に記載の方法によって製造された、発酵グアーミール。
(構成11)
バチルス菌株を含む、構成10に記載の発酵グアーミール。
(構成12)
バチルス菌株が、バチルス・サブチリスまたはバチルス・アミロリケファシエンスである、構成11に記載の発酵グアーミール。
(構成13)

(構成14)
構成1~9のいずれか一項に記載の方法によって製造された発酵グアーミールを含む飼料組成物。
(構成15)
ガラクトマンナナーゼ(Galactomannanase)、セルクラスト(Celluclast)及びビスコザイム(Viscozyme)からなる群から選択される少なくともいずれか1つの酵素をさらに含む、構成14に記載の飼料組成物。
(構成16)
前記酵素が、前記発酵グアーミール重量対比0.1%(w/w)~1.0%(w/w)で含まれるものである、構成15に記載の飼料組成物。
Claims (10)
- 発酵グアーミールの製造方法であって、次の段階:
(1)グアーミールを前処理する段階;
(2)前記前処理されたグアーミールにバチルス菌株を接種する段階;及び
(3)前記グアーミールに接種された菌を固体培養して発酵グアーミールを収得する段階を含み、
該バチルス菌株が、バチルス・アミロリケファシエンスであり、
該段階(1)の前処理する段階が、(1-1)水をグアーミールに添加してその水分含量を調節する段階;(1-2)該水分含量が調節されたグアーミールを熱処理する段階;及び(1-3)該熱処理されたグアーミールを冷却させる段階を含む、前記方法。 - 前記段階(1-1)の後のグアーミールの水分含量が、40~45%である、請求項1に記載の方法。
- 前記段階(1-2)の熱処理が、90℃~95℃で行われる、請求項1に記載の方法。
- 前記段階(1-3)の冷却が、固体発酵可能な温度まで下げるものである、請求項1に記載の方法。
- 前記段階(3)の固体培養が、30℃~50℃の温度で15~25時間行うものである、請求項1に記載の方法。
- 前記段階(3)の後に、前記発酵グアーミールを乾燥して粉砕する段階(4)をさらに含む、請求項1に記載の方法。
