Abstract
Description
(a)5’および3’トランスポゾン特異的逆位末端配列(ITR)、
(b)該5’ITRと3’ITRの間に位置しかつプロモーターの制御下にあり、導入遺伝子発現プロセシング(TEP)タンパク質またはTEP機能性RNAをコードする少なくとも1つの核酸配列、および
(c)任意選択で、同様に該5’ITRと3’ITRの間に位置しかつ導入遺伝子プロモーターの制御下にある少なくとも1つの導入遺伝子(前記核酸分子は、任意で、ベクターの一部である)。
該方法は、導入遺伝子を含む組換え哺乳類細胞を提供することを含み、
ベクターが該TEPまたはTEP機能性RNAを発現する発現ベクターであり、、前記ベクターによって発現した該TEPまたはTEP機能性RNAが、前記哺乳類細胞における導入遺伝子の発現を、少なくとも10%、少なくとも20%、少なくとも30%、少なくとも40%、少なくとも50%、少なくとも60%または少なくとも70%、任意に増加させることを特徴とする。

前記導入遺伝子を含む上記のベクターのうちの1つを含む組換え哺乳類細胞を提供すること、および、該導入遺伝子を発現させることを含む。ここで、前記MARエレメント(複数可)は、該導入遺伝子の発現を好ましくは少なくとも10%、少なくとも20%、少なくとも30%、少なくとも40%、少なくとも50%、少なくとも60%または少なくとも70%増加させ得る。該ベクターは、単一MAR X_29エレメント、および/または、配列番号3と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有し得る核酸を含んでいてよく、ならびに、4、5、6、7、8、9、10、11、12、13または14週を超える培養後に、該MARエレメントは、該目的導入遺伝子の発現を少なくとも10%、少なくとも20%、少なくとも30%、少なくとも40%、少なくとも50%、少なくとも60%または少なくとも70%増加させ得る。
細胞代謝エンジニアリングで、例えば、発現細胞に固有のプロセスが変更される。例えば、分泌経路の特定のタンパク質が、例えば、過剰発現する。あるいは、組換え経路に影響を与えることによって、組換えイベントが変更される。
タンパク質の分泌は、3つの界全ての生物に共通するプロセスである。この複雑な分泌経路は、特に、細胞質ゾルから細胞の細胞膜を横切るタンパク質の移行を必要とする。タンパク質をその最終目的地に到着させるのに複数のステップおよび様々な因子が必要とされる。哺乳類細胞において、この分泌経路には、シグナル識別粒子(SRP)および分泌複合体(Sec複合体またはトランスロコン)である2つの主な巨大分子アセンブリが関与する。SRPは、9、14、19、54、68および72kDaの質量を有する6つのタンパク質ならびに7S RNAで構成されており、該トランスロコンは、Sec61αβγ、Sec62およびSec63で構成されるドーナツ形粒子である。これらのタンパク質のいくつかのヒト版の受託番号(括弧内)は以下のとおりである:hSRP14(受託番号X73459.1);hSRP9(NM_001130440);hSRP54(NM_003136);hSRPRα(NM_003139);hSRPRβ(NM_021203);hSEC61α1(NM_013336);hSEC61β(L25085.1);hSEC61γ(AK311845.1)。
DNA組換え経路としても知られる組換え経路は、染色体2本鎖切断後のDNA分子の末端接合などのDNA損傷修復、ならびに例えば、減数分裂時の染色体交差またはリンパ球細胞における免疫グロブリン遺伝子の再構成などの染色体DNA分子と非染色体DNA分子の間でのDNA配列の交換もしくは融合をもたらす細胞経路である。3つの主要な組換え経路は、相同組換え経路(HR)、非相同末端結合経路(NHEJ)およびマイクロホモロジー媒介末端結合(MMEJ)経路ならびに代替末端結合(Alt−EJ)経路である。
導入遺伝子は2本鎖切断位置で宿主ゲノムへ組込むために組換え機構を使用する
2本鎖切断(DSB)は、ゲノム損傷の生物学的に最も有害な種類であり、細胞死または多種多様な遺伝子再構成をもたらす可能性がある。正確な修復が、遺伝情報の維持および伝播の成功に必須である。
概念的に、NHEJプロセスの分子機構は、1)一組の酵素が切断されたDNA分子を捕捉し、2)2つのDNA末端をつなぎ合わせて、分子ブリッジを形成し、3)切断された分子を再結合するというように、単純であると考えられている。このような反応を実行するために、哺乳類細胞におけるNHEJ機構には、DNA−PKcs(DNA依存性タンパク質キナーゼの触媒サブユニット)に結合したヘテロ二量体Ku80/Ku70およびその補助因子XRCC4(X線補完チャイニーズハムスター遺伝子4(X−ray−complementing Chinese hamster gene 4))と結合したDNAリガーゼIVの2つのタンパク質複合体、ならびにアルテミス(Artemis)およびXLF(XRCC4様因子、またはケルヌンノス(Cernunnos))などの多くのタンパク質因子が関与する(Delacote et al.,2002)。NHEJは、通常は配列相同性のない2つの切断されたDNA末端を単につなぎ合わせ、小さな挿入および欠失を生じさせるので、たびたびエラープローンDSB修復と見なされる(Moore and Haber,1996;Wilson et al.,1999)。NHEJは、細胞周期全体にわたってDSB修復機構を提供するが、有糸分裂細胞のG0〜G1および初期S期中が特に重要である(Takata et al.,1998;Delacote and Lopez,2008)。NHEJによるDSBの修復は、細菌から哺乳類に及ぶ生物で観察されており、進化の間、保存されていることを示す。
相同性組換え(HR)は非常に正確な修復機構である。相同染色分体は切断された鎖の修復のための鋳型として役割を果たす。HRは、姉妹染色分体が利用可能である場合、細胞周期のSおよびG2期中で起こる。古典的HRは主に3つのステップ:1)切断された末端の5’の切除、2)鎖の侵入および相同性のあるDNA2重鎖との交換、ならびに3)組換え中間体の分解、によって特徴付けられる。異なる経路は、鎖の侵入を実行する能力に依存してDSB修復を完了することができ、合成依存性の鎖のアニーリング(SDSA)経路、古典的2本鎖切断修復(DSBR)(Szostak et al,1983)、切断により誘発される複製(BIR)、および、あるいは、一本鎖アニーリング(SSA)経路を含む。全てのHR機構は、相互接続し、多くの酵素ステップを共有する。
他の組換え経路が失敗するかまたは作用しない場合、DSBは、マイクロホモロジー媒介末端結合(MMEJ)と呼ばれる別のエラープローン修復機構によって修復できる。この経路は、依然として十分に解明される必要性があり、代替末端結合(alt−EJ)とも称される場合があるが、これらの2つのプロセスが同じ機構に基づくか否かは明らかではない。NHEJと区別するこの経路の最も特徴的な特性は、切断されたDNA鎖のアライメント中に、5〜25塩基対のマイクロホモロジーを使用することである(McVey and Lee,2008)。
細胞代謝エンジニアリングのために使用できるこのカテゴリーのタンパク質は、タンパク質分泌経路にも組換え経路にも属さないが、別の方法で発現細胞において固有のプロセスに影響を与える。
非ウイルスベクターの中で、宿主ゲノム内の複数の遺伝子座に高頻度でDNA配列の単一コピーを組込む活性により、トランスポゾンが特に魅力的である。ウイルスベクターと異なり、いくつかのトランスポゾンは、優先的に細胞遺伝子の近くには組込まれないと報告されており、したがって、それらは有害変異を導入しそうにない。さらに、トランスポゾンは容易に生成および操作でき、一般に、逆位反復配列が隣接するカーゴDNAを含むトランスポゾンドナープラスミドおよびトランスポゼース発現ヘルパープラスミドまたはmRNAを含む。いくつかのトランスポゾンシステムは、内因性トランスポゾンコピーに干渉することなく、様々な細胞株においてDNAを動員するように開発された。例えば、ピギーバック(PB)トランスポゾンは、最初にキャベツルーパー蛾から単離され、様々な哺乳類細胞に効率的にカーゴDNAを転移させる。
抗生物質耐性は必ずしも効率的に導入遺伝子発現を反映しないので、強力なGAPDH細胞プロモーター誘導体から発現する緑色蛍光タンパク質(GFP)を指標として用いた。MARエレメントのPBトランスポゾンへの付加が転移効率および導入遺伝子発現に影響し得るか否かを試験するために、かつ構築物中のMARの位置がこれらの効果に影響するか否かを評価するために、一連のトランスポゾンドナー構築物をGFPおよびピューロマイシン耐性(Puro)遺伝子を含有するように設計し、MAR1−68または対照の中性スペーサーDNA配列をプラスミドの異なる位置に挿入した(図1)。挿入していない親のPuro−GFPトランスポゾンプラスミドを転移の対象として用いて、MARの効果またはスペーサー配列付加に比べて増加したトランスポゾンサイズの影響を区別した。
導入遺伝子の転写の増加および/またはより多い導入遺伝子コピーの組込みにより、より高いGFP蛍光レベルが得られ得る。これを、様々な種類のベクターに由来するゲノム組込み導入遺伝子の数を定量することによって評価した。非選別集団からの蛍光細胞の細胞蛍光測定選別後またはピューロマイシン耐性についての選別後のいずれかに、全ゲノムDNAをプールした細胞集団から単離した。細胞β2−ミクログロブリン(B2M)遺伝子に対する導入遺伝子のコピー数を、GFPコード配列の定量的ポリメラーゼ連鎖反応(qPCR)解析によって決定した。抗生物質選択剤の非存在下における、トランスポゼースまたは細胞組換え酵素のいずれかによって組込まれた導入遺伝子の平均数は同等であり、ゲノムあたり約1〜6コピーであり、MARまたは対照配列によって著しい影響は受けなかった(図4A)。しかし、MARがトランスポゾンの端に含まれる場合に、最低コピー数が得られ、この位置においてMARは転移を減少させるという本発明者らの以前の結論を支持する。ピューロマイシンを用いた高発現細胞の選別後に、転移した導入遺伝子の数は同様に2〜7コピーの範囲であった(図4B)。しかし、トランスポゼースの非存在下で組込まれた導入遺伝子のコピー数は、一般に非常に高く、6〜14コピーに及んだ。これは、自然発生的な組込みが通常、単一ゲノム遺伝子座において複数のプラスミドコピーのコンカテマーの組込みをもたらすとう事実(結果は示しめしていない)、ならびに、サイレンシング効果に供した細胞を抗生物質選別によって除去した場合に、より高い導入遺伝子コピー数がより高い発現レベルをもたらすはずであるという事実によって容易に説明できる。抗生物質選別によって優先的に高発現細胞が得られるという以前の結論と合わせて、これは、自然発生的なプラスミド組込みが転移性ベクターよりも様々な数の導入遺伝子コピーをもたらすことも示した。
IgGなどの異種タンパク質の分泌は、CHO細胞などの培養細胞における不適切なポリペプチドプロセシングおよび低いIgG産生によって縮小される。BiPのようなストレス誘導性シャペロンの発現が誘導され、かつこのシャペロンが正しくERに局在して、IgG前駆鎖と相互作用できることが観察された。しかし、それにもかかわらず、インフリキシマブで見られるものなどの特定の可変配列を含有するIgGは正しくプロセシングおよびアセンブルされず、不十分な分泌をもたらす。したがって、これらの細胞におけるUPR応答の活性化は、著しいレベルの免疫グロブリンの修復に依然として効果がない。
IgGのHP(高産生)およびLP(低産生)クローンを、SRPのSRP14成分をコードするベクターおよびネオマイシン耐性プラスミドでコトランスフェクトした。ネオマイシン耐性プール中の個々の細胞を限界希釈法によって分離し、その後、振盪培養皿のバッチ内での成長および免疫グロブリン分泌について試験した。SRP14を発現するLP由来サブクローンは、培養の間中、それらの親対応物よりも著しく多量の抗体を分泌し、HP SRP14−発現サブクローンとして同様の免疫グロブリン抗体価をもたらした(図8Aおよび8B、左のパネル)。
異種SRP14の過剰発現が所定の閾値までIgG分泌を増加させたことを考慮すると、SRP14と直接的または間接的に相互作用する分泌経路の他の構成成分が、SRP14−LPサブクローンにおける制限になり得ると考えるのは妥当であった。したがって、本発明者らは、分泌経路の他の構成成分の過剰発現が、単独でまたはSRP14との組合せのいずれかで、IgG発現も改善し得るか否かを調べた。これらには、SRP14、SRP受容体(SR)のサブユニットならびにトランスロコンのSec61α、βおよびγサブユニットと共にSRP複合体を構成するヒトSRP9およびSRP54タンパク質が含まれる。
組換えに影響を与えるために、重要なDNA組換え遺伝子をサイレンシングできる。ノックダウンの標的は、文献から哺乳類における特定の組換え修復経路に不可欠であると知られる遺伝子である(表D)。これらの遺伝子のほとんどが細胞の生存および発達に必要であるので、これらの永久的なサイレンシングが細胞生存率を低下させた。したがって、RNA干渉(RNAi)を一過的に用いるこれらの標的遺伝子のサイレンシングが好ましい。
3つのsiRNA標的Rad51を、ヘアピン構造を形成できるshRNA配列に変換し、shRNAコード配列を、ピギーバックトランスポゾンベクターに挿入した。該ピギーバックトランスポゾンベクターは、抗生物質選択遺伝子を欠くものの、GAPDHエンハンサーおよびCMVプロモーター融合物の制御下にあり、これに続いてMAR X−29が制御する。懸濁液に適応したCHO−M細胞をトランスポゾンドナープラスミドおよびトランスポゼース発現ベクターで3回トランスフェクトし、その後30個の個々の細胞クローンを、ClonePix装置を用いて無作為に採取した。親細胞ならびにshRNA発現細胞プールおよびクローンプールをGFP発現プラスミド(すなわちPuro−GFP−MAR X_29構築物)と再トランスフェクトし、その後に、GFP発現細胞のポリクローナルプールのピューロマイシン選択を行った。GFP蛍光プロファイルの比較により、親CHO−M細胞と比較して中くらいから強い蛍光を発するより高い割合の細胞(M2細胞集団)または非常に強い蛍光を発する細胞(M3細胞集団)が、shRNAベクターでトランスフェクトした細胞プールから得られたことが示された(図17A)。
プラスミドおよびDNAベクター
PBトランスポゼース発現ベクターpCS2+U5V5PBU3は、アフリカツメガエルのβグロビン遺伝子の5’および3’非翻訳末端領域(UTR)によって囲まれたPBトランスポゼースコード配列を含む。このプラスミドを以下のように構築した:pBSSK/SB10の3’UTR317塩基対断片(S.Ivics博士から快く提供された)をpCS2+U5(Invitrogen/Life Technologies社、ペーズリー、英国)に挿入し、pCS2+U5U3を得た。PBトランスポゼースコード配列(2067塩基対、GenBank受託番号EF587698)は、ATG:biosynthetic社(Merzhausen、ドイツ)によって合成してもらい、2つのUTR間のpCS2+U5U3骨格にクローニングした。PB対照ベクターは、変更していないpCS2+U5プラスミドに相当する(図1、左のパネル)。ATG:biosynthetic社(Merzhausen、ドイツ)によって合成されたPB235塩基対3’および310塩基対5’逆位末端反復配列(ITR)をpBluescript SK−プラスミドに導入することによって、この研究で用いる異なるトランスポゾンベクターを作製した(pBSK ITR3’−ITR5’、図1、右のパネル)。次いで、pRc/RSVプラスミド(Invitrogen/Life Technologies社)のSV40プロモーターの制御下にあるピューロマイシン耐性遺伝子(Puro(登録商標))を2つITRの間に挿入した。この研究で用いるMAR 1−68およびMAR X−29エレメント、ピューロマイシン耐性遺伝子ならびにGFP遺伝子は、前述のとおりである。免疫グロブリン発現ベクターおよびSRP9、SRP14、SRP54、SRPRα、SRPRβ、SEC61A1、SEC61BおよびSEC61Gコード配列は、Le Fournら(Metab.Eng.,Epub 2013 Feb 1)によって説明されたとおりであった。
CHO DG44細胞株を、ヒポキサンチン/チミジン(HT、Gibco社)および10%ウシ胎仔血清(FBS、Gibco社)を補充したDMEM:F12(Gibco社)で培養した。製造業者の取扱説明書に従って、PEI(JetPRIME、Polyplus Transfection社)を用いてトランスフェクションを行った。滴定実験において細胞を、様々な量(0〜1500ngの範囲)のPBトランスポゼースのpDNA源でトランスフェクトし、または細胞を300ngのPBトランスポゼース発現プラスミドと300ngのトランスポゾンドナープラスミドの最適な比でコトランスフェクトした。トランスフェクション後2日目に、実験に応じて細胞をいくつかのペトリ皿に移した。非選択トランスフェクトCHO細胞の分析については、抗生物質選択剤の非存在下で3週間細胞を再播種し、蛍光細胞の百分率およびGFP陽性細胞の蛍光強度を、CyAn ADPフローサイトメーター(Beckman Coulter社)を用いるFACS解析によって決定した。非選択細胞の遺伝子コピー数の分析については、FACSAriaIIを用いて安定GFP陽性CHO細胞を選別した。抗生物質耐性コロニー計数アッセイについては、50,000個のトランスフェクト細胞を100mmプレートに播種し、2週間、5μg/mlのピューロマイシンで選択した。次いで、耐性コロニーを固定し、70% EtOH、0.7%メチレンブルーで10分間染色し、直径が0.5mmを超えるコロニーを計数した。GFP発現研究については、上記のとおりGFP蛍光FACS解析の前に細胞を2週間選択した。
製造業者のプロトコールに従って、DNeasy組織キット(Qiagen社、ヒルデン、ドイツ)を用いて転移アッセイ後のCHO安定細胞プールから全DNAを単離した。ゲノムに組込まれた導入遺伝子のコピー数を、6ngのゲノムDNAを用いて、Eurogentec社のSYBRグリーン−TaqポリメラーゼキットおよびABI Prism 7700 PCR機(Applied Biosystems社)を用いる定量qPCRにより評価した。GFP−フォワード:ACATTATGCCGGACAAAGCCおよびGFP−リバース:TTGTTTGGTAATGATCAGCAAGTTGプライマーを用いてGFP遺伝子を定量し、B2M−フォワード:ACCACTCTGAAGGAGCCCAおよびB2M−リバース:GGAAGCTCTATCTGTGTCAAプライマーを用いて、β−2ミクログロブリン遺伝子を増幅した。B2Mプライマーによって作製した増幅産物については、NCBI BLASTソフトウェアを用いて本発明者らのCHOゲノムアセンブリに対するアライメント後に、CHOハプロイドゲノムあたり1つのヒットが見出された。CHOは2倍体に近い細胞なので、B2Mはゲノムあたり2コピーで存在すると推定された。前述のとおり、B2M参照遺伝子の割合と比較したGFP標的遺伝子コピー数の割合を計算した。
IgG発現細胞を磁気選別するため、トランスフェクトされたCHO−M細胞を、8mMのL−グルタミンおよび1×HT栄養補助剤(共にGibco社から)を補充し、完全培地とも称されるSFM4CHO培地(Thermo Scientific社)中、1mlあたり3×105細胞の細胞密度で播種した。培養4日後に、2×106細胞を洗浄し、PBSに再懸濁し、事前に洗浄した30μlのMyOne T1ストレプトアビジンコーティングダイナビーズ(Invitrogen社)と共に、3μg/mlの最終濃度のビオチン化ヒトIgG(KPL216−1006)と室温で30分間、回転ホイール上でインキュベートした。次いで、細胞およびビーズ混合物を磁石上に置き、標識細胞と非標識細胞を分けた。ビーズをリン酸緩衝食塩水(PBS)溶液で4回洗浄した。最後のPBS洗浄後、これらのビーズおよび細胞を予熱した500μlの完全培地に再懸濁し、24ウェルプレートに移し、37℃、5%CO2でインキュベートした。24時間後、磁気選別したポリクローナル細胞をビーズから分離し、50mLのTPPスピンチューブバイオリアクター(TECHNO PLASTIC PRODUCTS AG社、スイス)の中で細胞が十分な増殖密度に達するまでインキュベーションを続けた。
この研究で用いるクローニングベクターは、Selexis社の哺乳類発現ベクターSLXplasmid_082である。pGL3対照ベクター(Promega社)のルシフェラーゼ配列を、真核生物発現カセットと置換した。該真核生物発現カセットは、EGFPコード配列の上流側のヒトCMVエンハンサーおよびヒトGAPDHプロモーター、およびこれに続く、SV40ポリアデニル化シグナル、ヒトガストリン転写終結コドンおよびSV40エンハンサーで構成されていた。2つのヒトMAR由来の遺伝エレメントは、発現カセットおよびSV40プロモーターから発現するピューロマイシン耐性遺伝子に隣接しているが、SLXプラスミド_082は、発現カセットの上流側に位置するMARエレメントの種類によって異なる(hMAR 1−68およびhMAR X−29;Girod et al.,2007)。
懸濁チャイニーズハムスター卵巣細胞(CHO−K1)をL−グルタミン(PAA社、オーストリア)およびHT栄養補助剤(Gibco、Invitrogen life sciences社)を補充したSFM4CHOハイクローン無血清培地(ThermoScientific社)中で、加湿空気中37℃、5%CO2で維持した。CHO−K1細胞を、製造業者の推奨する方法に従って電気穿孔(Neon devices、Invitrogen社)により、ピューロマイシン耐性遺伝子を保有するトラスツズマブまたはインフリキシマブの重鎖および軽鎖発現ベクターでトランスフェクトした。2日後、これらの細胞をT75プレート中の10μg/mlのピューロマイシンを含む培地に移し、これらの細胞を2週間選択剤の存在下でさらに培養した。次いで、トラスツズマブまたはインフリキシマブのIgGを発現する個々の安定細胞クローンを限界希釈法により作製し、増殖させ、増殖能およびIgG産生レベルについて分析した。最高レベルのIgGを発現するトラスツズマブまたはインフリキシマブIgG産生細胞クローンを、さらなる生化学実験のために選択した。次いで、これらのクローンのいくつかを、電気穿孔法によりSRP14発現ベクターおよびネオマイシン耐性遺伝子を保有するプラスミドでコトランスフェクトした。次いで、上記のとおり、300μg/mlのG418を含む培地中で細胞を2週間培養した。安定クローンを限界希釈法により単離し、生化学分析のための培養増殖前にQ−PCRアッセイによってSRP14発現を確かめた。
50mlのミニバイオリアクター(TPP社、スイス)において、トラスツズマブまたはインフリキシマブを発現する個々のクローンの増殖能および産生能を、37℃、5%CO2加湿インキュベーター中7日間の流加培養で評価した。細胞培養の3日目、4日目、および7日目に、細胞の密度および生存率をGuava EasyCyteフローサイトメトリーシステム(Millipore社)を用いて評価した。細胞培養上清中のIgG抗体価をサンドイッチELISAによって測定した。示したプロセス時間のサンプリング日における細胞密度(Cv/ml)およびIgG抗体価(μg/ml)の値をプロットした。トラスツズマブまたはインフリキシマブ発現クローンのIgG比生産性を、3日目〜7日目(産生段階)に計算したIgG濃度対生存細胞の整数(IVCD)の勾配として決定し、細胞あたりおよび1日あたりのpg(pcd)として表した。
可溶性細胞質タンパク質を、プロテアーゼ阻害剤カクテル(Roche社)の存在下でPBS緩衝液中1%Triton X−100を用いて細胞を透過処理することによって抽出した。氷上で30分インキュベーションした後、細胞を14,000rpmで10分間遠心分離した。上清を「可溶性細胞質およびERタンパク質」画分と命名した。ペレットを尿素Laemmli緩衝液(62.5mMのTris、2% SDS、8M尿素、5%グリセロール、ブロモフェノールブルー色素)中で超音波処理により溶解し、凝集した小胞の不溶性タンパク質画分を得た。次いで、可溶性画分および不溶性画分を2−メルカプトエタノールを含むまたは含まないLaemmli緩衝液で調整し、8分間95℃で煮沸た。還元試料および非還元試料を、ドデシル硫酸ナトリウムポリアクリルアミド遺伝子電気泳動(polyacrylamide gene electrophoresis)(SDS−PAGE)によって、それぞれ10%または4〜10%勾配のアクリルアミドゲルで分離した。
高(HP)および低(LP)IgG生産CHO−K1クローン上でシクロヘキシミドベースの追跡実験を行った。同数の細胞を6ウェルプレート中の100μM シクロヘキシミド(Sigma社)を補充した完全培地に播種した。様々な時点で細胞を採取し、1%Triton X−100含有PBSに溶解した。次いで、Tx可溶性画分およびTx不溶性画分を4〜10%アクリルアミド非還元SDS−PAGE上で分離し、抗ヒトIgG抗体で免疫ブロットした。
膜タンパク質由来の細胞質の分画を、細胞ペレットのディファレンシャル界面活性剤抽出によって行った。最初に細胞を1mlのPBSで洗浄し、HPおよびLP細胞の細胞膜を、1% Triton X−100の存在下または非存在下で、0.01%ジギトニン(Sigma社)を含むKHM緩衝液(110mM KAc、20mM HEPES、2mM MgCl2、pH7.2)で10分間透過処理した。半透過細胞をKHM緩衝液で1回洗浄し、トリプシンを室温で10分間かけて50μg/mlまで添加し、可溶性タンパク質を消化した。トリプシン消化を1mM PMSFおよび4mMのAEBSFの添加によって停止した。セクション2.4に記載のとおり、細胞を遠心分離によって収集し、Triton X−100およびプロテアーゼインヒビターの存在下で可溶性タンパク質を抽出した。2−メルカプトエタノールを含むLaemmli還元緩衝液をペレットおよび上清画分に添加し、次いで、8%SDS−PAGEに供した。免疫ブロットを行い、IgGおよびBiPタンパク質を検出した。
インフリキシマブLP細胞をPBS中で1回洗浄し、氷上で30分間、1mMのジチオビス(プロピオン酸スクシンイミジル)(DSP)架橋剤(ThermoScientific社)の存在下および非存在下でインキュベートした。50mMのTris−HCl(pH7.4)の添加によって架橋結合を10分間停止し、その後、タンパク質の抽出を1% Triton X−100含有PBS緩衝液中で行った。微量遠心管で14,000rpmで10分間遠心分離した後、Triton X−100不溶性画分または全タンパク質抽出物を還元条件下のSDS−PAGEによって分析し、免疫ブロットし、抗BiP抗体および抗LC抗体を用いて調べた。等量のTx不溶性画分のタンパク質を並行して分析した。
Claims (60)
- 以下を含む組換え核酸分子:
(a)5’および3’トランスポゾン特異的逆位末端反復配列(ITR);
(b)前記5’ITRと3’ITRの間に位置し、プロモーターの制御下にある導入遺伝子発現プロセシング(TEP)タンパク質またはTEP機能性RNAをコードする少なくとも1つの核酸配列、および
(c)任意選択で、同様に前記5’ITRと3’ITRの間に位置し、導入遺伝子プロモーターの制御下にある少なくとも1つの導入遺伝子(ここで、前記核酸分子は、任意に、ベクターの一部である)。 - 少なくとも1つのエピジェネティック調節エレメント、特に少なくとも1つのMAR(マトリックス結合領域)エレメントをさらに含む、請求項1に記載の組換え核酸分子。

- 前記TEPタンパク質がタンパク質分泌経路のタンパク質、組換え経路のタンパク質、BiPなどのシャペロンを含むタンパク質プロセシングもしくは代謝に係るタンパク質、またはそれらの組合せである、先行するいずれかの請求項の一項に記載の組換え核酸分子。
- 前記TEPタンパク質が、タンパク質分泌経路の以下のタンパク質:hSRP14、hSec61α1、hSec61β、hSec61γ、hSRP54、hSRP9、hSRPRα、hSRPβ、およびhCANXのうちの1つまたは複数である、請求項5に記載の組換え核酸分子。

- 前記哺乳類細胞が、前記少なくとも1つの単離TEP機能性RNAおよびベクターでトランスフェクトされることを特徴とし、前記ベクターは前記少なくとも1つの導入遺伝子および任意選択で前記少なくとも1つの導入遺伝子に隣接する3’ITRおよび5’ITRを含み、任意選択で前記5’ITRおよび前記3’ITRを認識するトランスポゼースを発現する単離核酸またはmRNAと共に、前記トランスフェクトされることを特徴とする、請求項33に記載の方法。
- 前記導入遺伝子が、免疫グロブリン、ホルモン、サイトカインまたは成長因子などの治療タンパク質である、請求項33または請求項33以降の請求項のいずれかに記載の方法。
- 前記組換え核酸分子が、任意選択で選択マーカーを含み、前記少なくとも1つの導入遺伝子が、
(a)前記マーカーについての選択なしで、または
(b)前記マーカーについての、例えば、培地中に含まれる選択剤による選択を行うことによって、および
(c)トランスポゼースの非存在下で、または
(d)トランスポゼースの存在下で、
発現する、請求項33または請求項33以降の請求項のいずれかに記載の方法。 - 前記TEP機能性RNAが、shRNAをコードする組換え核酸配列によってコードされるか、または限定されないが、Rad51、Rad51B、Rad51C、Rad51D、Xrcc2、Xrcc3、Rad52、Rad54、Brca1、Brca2、サイクリンD1、Ercc1、MDC1、Bard1、リガーゼ1、Mre11および/または53BP1などの少なくとも1つのHR遺伝子の発現を妨げるsiRNAを含む/からなる、請求項33に記載の方法。
- 請求項1〜13に記載の組換え核酸分子のうちのいずれか1つを含む少なくとも1つのベクターを1つの容器内に含み、互換性のあるトランスポゼースをコードするベクターを第2の任意の容器内に含み、前記ベクター(複数可)の使用法についての説明書をさらに別の容器内に含むキット。
- 2種類以上のベクターが1つまたは複数の容器に入れられた状態で提供され、前記TEPタンパク質が、シャペロン、SRP14、SRP9、またはSRP54のうちの少なくとも2つである、請求項44に記載のキット。
- 前記ベクター(複数可)内のTEP機能性RNA(複数可)が、限定されないが、Rad51、Rad51B、Rad51C、Rad51D、Xrcc2、Xrcc3、Rad52、Rad54、Brca1、Brca2、サイクリンD1、Ercc1、MDC1、Bard1、リガーゼ1、Mre11および/または53BP1などの少なくとも1つのHR遺伝子の発現を妨げるmiRNA、siRNAまたはshRNAをコードする核酸配列を含み/からなり、任意選択で、限定されないが、Rad51、Rad51B、Rad51C、Rad51D、Xrcc2、Xrcc3、Rad52、Rad54、Brca1、Brca2、サイクリンD1、Ercc1、MDC1、Bard1、リガーゼ1、Mre11および/または53BP1などの少なくとも1つのHR遺伝子の発現を妨げるsiRNA(複数可)をさらに別の容器内に含む、請求項44に記載のキット。
- 好ましくは、導入遺伝子の組換えおよび/または発現を増加させるための、請求項1〜13に記載の組換え核酸および/または請求項22〜31に記載の組換え哺乳類細胞の使用。
- (a)上流側でプロモーターと、下流側でポリアデニル化シグナルと隣接する導入遺伝子、
(b)ポリアデニル化シグナルの下流側の単一MARエレメント、または
(c)目的の導入遺伝子の上流側の第1のMARエレメントおよび前記導入遺伝子の組込み部位の下流側の第2のMARエレメント、
を含む発現ベクター。 - 単一または第1および第2のMARエレメントが、MARエレメント1_68、1_6、1_6R2、1_68R、1_68R2、X_29R3もしくはX_29または配列番号1、2、3、6、7、8、9もしくは10と少なくとも80%、90%、95%、98%、99%または100%の配列同一性を有するエレメントから選択される、請求項48に記載の発現ベクター。
- 前記単一または第1および第2のMAR(複数可)が、再構成したMARエレメント1_6R2、1_68R、1_68R2もしくはX_29R3または配列番号6、7、8、もしくは10と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有するエレメントから選択され、任意選択で、MARエレメント(複数可)が、それらの再構成していない対応物と比較して、目的の導入遺伝子の発現を少なくとも10%、少なくとも20%、少なくとも30%、少なくとも40%、少なくとも50%、少なくとも60%または少なくとも70%増加させる、請求項48に記載の発現ベクター。
- 前記プロモーターが、EF1αプロモーターであり、ポリアデニル化シグナルがBGHポリAシグナルである、請求項48〜50に記載の発現ベクター。
- 前記ベクターが、GAPDH、CGAPDH、CSV40p、CMVp、CHO Ef1α、CHO ActbまたはCHO Hspa5などのプロモーター(複数可)および/またはエンハンサー(複数可)またはそれらの融合体を含む、請求項48〜51に記載の発現ベクター。
- 前記プロモーターが、GAPDHプロモーターであり、CMVエンハンサーを含む、請求項52に記載の発現ベクター。
- 前記1つのMARまたは第1および第2のMAR、エンハンサー、プロモーター、目的の導入遺伝子ならびにポリアデニル化シグナルが、5’ITRと3’ITRの間に位置する、請求項48〜53に記載の発現ベクター。
- (a)ポリアデニル化シグナルの下流側の単一MARエレメント(ここで、前記単一MARエレメントが、好ましくはMAR 1−68もしくはMARX−29エレメントおよび/または配列番号1もしくは3と少なくとも80%、90%、95%、98%、99%または100%の配列同一性を有するエレメント、具体的には、MAR 1−68もしくはMARX−29に基づいて再構成したMAR、具体的には、配列番号6、7もしくは10(MAR 1_68R、1_68R2もしくはX_29R3)または配列番号9と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有するエレメントであり、好ましくは、MARX−29エレメントであり、かつ/または配列番号3と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有する)、あるいは
(b)目的の導入遺伝子の上流側の第1のMARエレメントおよび目的の前記導入遺伝子の下流側の第2のMARエレメント(ここで、第1のMARエレメントは、好ましくは1_6エレメントを含み、および/または配列番号2、具体的には、MAR 1−6に基づいて再構成したMARと少なくとも80%、90%、95%、98%、99%または100%の配列同一性を有し、具体的には、配列番号8(MAR 1_6R2)と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有するエレメントであり、第2のMARエレメントは、好ましくは、MAR 1−68エレメントを含み、かつ/または配列番号1と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有する)
を含む、請求項48〜54に記載の発現ベクター。 - 前記単一MARエレメントを含み、前記単一MARエレメントが、ポリアデニル化部位の下流側に位置し、MAR 1−68またはMAR X−29であり、および/または、配列番号1または3と少なくとも80%、90%、95%、98%、99%または100%の配列同一性を有し、具体的には、MAR 1−68もしくはMAR X−29に基づいて再構成したMARであり、具体的には、配列番号6、7もしくは10(MAR 1_68R、1_68R2もしくはX_29R3)または配列番号9と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有するエレメントであり、および好ましくはMAR X−29由来のエレメントであり、および/または配列番号3と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有する、請求項55に記載の発現ベクター。
- 前記導入遺伝子の上流側に第1のMARエレメントと、前記目的の導入遺伝子の下流側に第2のMARエレメントを含み、前記第1のMARエレメントがMAR 1_6エレメントみ、および/または配列番号2と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有し、第2のMARエレメントがMAR 1_68エレメントを含み、および/または配列番号1と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有する、請求項55に記載の発現ベクター。
- 導入遺伝子を発現させるための発現方法であって、
前記導入遺伝子を含む、請求項48〜57に記載のベクターのうちの1つを含む組換え哺乳類細胞を提供すること、および
導入遺伝子を発現させること、を含み、
前記MARエレメント(複数可)が、前記導入遺伝子の発現を好ましくは少なくとも10%、少なくとも20%、少なくとも30%、少なくとも40%、少なくとも50%、少なくとも60%または少なくとも70%増加させる、方法。 - 前記ベクターが、単一MAR X_29エレメントおよび/または配列番号3と少なくとも80%、90%、95%、98%、99%もしくは100%の配列同一性を有する核酸を含み、4、5、6、7、8、9、10、11、12、13または14週を超える培養後に、前記MARエレメントが、目的の導入遺伝子の発現を少なくとも10%、少なくとも20%、少なくとも30%、少なくとも40%、少なくとも50%、少なくとも60%または少なくとも70%増加させる、請求項58に記載の方法。
- 前記単離核酸分子および/または前記少なくとも1つの単離TEP機能性RNAでトランスフェクトされていない細胞と比較して、そのような細胞において導入遺伝子の組込みおよび/または発現が増加することを特徴とする、請求項33または請求項34〜43のいずれかに記載の方法。
