培養細胞。ヒト胎児腎臓(HEK293T)、HeLaおよび3T3細胞をダルベッコ改変イーグル培地(DMEM、Life Technologies)中の10%ウシ胎児血清中で培養した。ヒト誘導多能性幹細胞(iPS株WTSIi008−A、EBiSC、UK)を、Geltrexコーティング(Life Technologies)上のE8培地(Life Technologies)中で培養し、EDTAで継代した。マウスES細胞(C57BL/6×129/Sv、株KH2)(Beardら,2006)を、初代胚性線維芽細胞フィーダ細胞上で培養した。
分析されたサンプルの数を、図の凡例に示す。統計分析を、RまたはGraphPad Prismソフトウェアを使用して実行した。有意性を、スチューデントのt、x2、クラスカル・ウォリス(一元配置分散分析)検定、およびノンパラメトリックなマンホイットニーU検定を使用して評価した。データは、平均+SEM、ns、p>0.05、*p<0.05;**p<0.01、***p<0.001を意味する。
piggyBac転位系に基づく転位依存性発現スイッチ(PB−IDスイッチ)
「カットアンドペースト」機構に従って機能するトランスポゾン系(例えば、piggyBac、Tol2、Sleeping Beauty;Wuら,2006)は、ランダムな遺伝子座でのゲノム組込みを実現するために広く使用されている。組込み駆動型(ID)遺伝子スイッチを構築するために、その既知の効率、大きな貨物容量、および正確なカットアンドペースト機構(Lacosteら,2009、Woodard and Wilson,2015、Wuら,2006)のために、piggyBac転位系を使用した。piggyBacトランスポサーゼ(以下、設計されたPBase)は、piggyBacトランスポゾンの2つの末端反復に隣接するエレメントを動員し(Lacosteら,2009、以下、設計された5’TRおよび3’TR)、これらのエレメントをTTAA部位での標的細胞のゲノムに挿入する。トランスポゾンを切除すると、これらのTTAA部位は正確に復元される。piggyBac系に基づく古典的なプラスミドベクター(図1A、左)では、プロモータ(つまり、転写開始を駆動する配列)と、通常は転写ターミネータが後に続く目的の遺伝子(GOI)で構成される導入遺伝子が、2つのpiggyBac TRの間に置かれ、トランスポサーゼが最初に骨格ベクターからそれを切除し、続いてそれを宿主細胞のゲノムに挿入する。したがって、導入遺伝子は、ドナープラスミドからのPBaseを介した切除の前に発現される(図1B、左)。対照的に、PB−IDベクターは、導入遺伝子をエピソーム形でサイレントに維持するプロモータ、GOI、およびTRの配置を備えている(図1A、右)。つまり、まず、2つのpiggyBac ITRの1つが、2つのITRが逆平行ではなく平行になる、古典的なトランスポゾン構成(5’ITRと3’ITRの両方がセンス方向)と比較して、ドナープラスミドにおいて逆方向(センス方向に5’ITR、アンチセンス方向に3’ITR)に配置される。第二に、導入遺伝子が、完全に機能する産物を生成することができない2つのエレメントに分割され、TRの1つによって分離され、互いに対して反対の方向に置かれる。したがって、GOIは、組込み前は発現されないか(図1B、右)、またはその5’部分のみが発現される。この構成は、piggyBacを介した挿入と切除の両方が一致して、ドナープラスミドを宿主ゲノムに組み込むと同時に、導入遺伝子の2つの部分を機能的な発現ユニットに再結合する再配置を引き起こし、GOI発現をトリガーする状況を作り出す。古典的なpiggyBacベクターとは異なり、ベクター全体が宿主細胞のゲノムに組み込まれる。この導入遺伝子構成を、以後PB−IDと名付け、プロモータ(Prom)から目的の遺伝子(GOI)を発現するこの原理に基づく導入遺伝子を、以下のように表す:PB−IDProm∞GOI、ここで無限大記号∞は、GOIが構成的にプロモータの制御下にある古典的なトランスポゾン構成(以下、PBProm::GOIと表記)とは対照的に、転位中に起こる再配置を表す。PB−IDの原理に基づいて、以下に示すいくつかのタイプのベクターを設計および検証した。
2.1)最初にブロックされたGOIの転写がpiggyBacトランスポサーゼによって媒介されるゲノム組込みによって活性化されるPB−IDベクター(PB−ID転写スイッチ、図1および図2)
PB−ID転写スイッチでは、プロモータおよびGOIがエピソームベクター内でヘッドトゥーヘッド(反転)方向に置かれ、piggyBac ITRの1つによって分離されているため、転写が防止される。上で説明したように、PBase作用によるゲノム組込みの際、PB−IDベクターの再配置により、GOIがプロモータの制御下になり、正しい方向になり、その転写がトリガーされる(図1A右)。この状況は、GOIがその組込み前に既にプロモータの制御下にある古典的なpiggyBacトランスポゾンベクターの設計とは対照的である(図1A、左)。
プロモータ、GOI、末端反復配列、およびベクター骨格の相対的な配置に応じて、PB−ID転写スイッチの別個の構成を使用して、piggyBac転位によるGOIの発現をトリガーすることができる(図2A)。特に、以下の2つの側面に応じて、様々なタイプのPB−ID構成を考案した。
これらの配置の組合せにより、PB−ID(1A)、PB−ID(1B)、PB−ID(2A)、およびPB−ID(2B)で表記されるPB−IDスイッチの4つの構成が定義される(図2A)。
上で定義した様々なタイプのPB−ID構成が転位依存性GOI発現を駆動できるかどうかを試験するために、PB−IDベクターを、最小限のpiggyBac 5’および3’TR(Lacosteら,2009)、広く活性な合成CAGプロモータ(CMVエンハンサー、ニワトリベータアクチンプロモータ、およびウサギベータグロビンスプライスアクセプターからなる、(Niwaら,1991)、およびGOIとしてのRFP(mRFPl、Campbellら,2002)、それに続くウシ成長ホルモン転写ターミネータ(bGH polyA、Kakokiら,2004)を使用して、pUC57−miniプラスミド骨格でアセンブルした。3つのPB−IDCAG∞RFPベクターを、PB−ID(1A)、PB−ID(1B)、およびPB−ID(2A)設計に従って構築し、以下、PB−ID(1A)CAG∞RFP、PB−ID(1B)CAG∞RFP、およびPB−ID(2A)CAG∞RFPと表記し、これにより、タイプA対タイプB、およびタイプ1対タイプ2配置を試験することができた。
PB−IDCAG∞RFPプラスミドを、PBaseを発現する2番目のプラスミドを有するまたは有さないニワトリ胚脊髄に卵内にエレクトロポレーションした(図1D)。トランスフェクションの4日後、RFPの発現をVZから神経管の外層に流れる細胞の放射状の流れでPBaseに依存して観察した。この発現パターンは、心室表面を裏打ちするエレクトロポレーションされた神経前駆細胞のゲノムへの導入遺伝子組込みを示しており、したがって、放射状のクローンパターンを形成することが知られている、これらの細胞およびそれらのニューロンの子孫の長期標識を達成している(Leber and Sanes,1995;Loulierら,2014)。対照的に、非組込みベクター(CAG::Cerulean)からの発現は、エレクトロポレーションの直後に生まれた単離されたニューロンにほとんど制限されていた(図1D)。
古典的なエピソームベクター(CAG::GFP)を、E12で子宮内、マウス大脳皮質にエレクトロポレーションし、GFP発現をエレクトロポレーションの直後に生まれたニューロンで観察し、次いで、分裂する前駆細胞で急速に希釈されるため、中間層で観察した。対照的に、同じ方法でエレクトロポレーションされたPB−IDCAG∞RFPでは、RFPの発現は、前駆細胞および後期に生まれた上層ニューロンと星状細胞を含めたそれらの放射状に移動するすべての誘導体で、厳密なPBase依存的に観察された。同様に、マウス網膜では、神経発生の開始時にPB−IDベクターがエレクトロポレーションされ、RFPがすべての網膜層で観察されたが、エピソームは、初期に生じた網膜神経節細胞のみをマークした。重要なことに、それは、おそらく複数の組込みされていないコピーの継承のために、古典的なトランスポゾンおよびエピソームで得られた孤立したニューロンの強く不規則な標識を回避した。
auto−PB−IDスイッチでは、IDスイッチのすべてのコンポーネントが単一の「オールインワン」ベクターによってコードされる(図3)。設計は、以下の修正を有する転写IDスイッチ(1.2.1)の設計に従う。piggyBacトランスポサーゼ遺伝子(Yusaら,2011)が、それがベクターのエピソーム形で発現されるようにプロモータの制御下に置かれる。PBase作用によってゲノム組込みおよび再配置が引き起こされると、GOIは、プロモータの下流に再び置かれ、その転写がトリガーされる一方、PBase遺伝子は、プロモータに対して反対の方向に終了するため、もはやその制御下にない。したがって、発現は、PBaseからGOIのものに切り替わる(図3A)。これにより、単一のベクターで転位依存性GOI発現を実現できる。以下、この構成をPB−IDProm::PBase∞GOIと表記する。
auto−PB−IDベクターを、1.2.1に記載したPB−IDベクターにおいて、プロモータに続く5’TRの直後にpiggyBacトランスポサーゼ遺伝子(hyPBase;Yusaら,2011)を配置し、続いて転写ターミネータを配置することによってアセンブルした。PB−IDCAG::PBase∞RFPと名付けられたこの構成では、トランスポサーゼを介したゲノム組込み時に、発現がPBaseからRFP発現に切り替わる(図3A)。トランスフェクトされたHEK−293細胞での試験は、予想通り、PB−IDCAG::PBase∞RFPベクターが他のヘルパーベクターの非存在下でGOI発現を駆動することを示している(図3B)。
PB−ID転写スイッチ(I.2.1)では、エピソームベクターからの漏れのある転写が発生した場合は、GOIの翻訳および対応するタンパク質の生成が続くことになっている。このような可能性を回避するために、GOIの転写と翻訳の両方がベクターのエピソーム形においてブロックされるPB−ID翻訳スイッチを設計した(図4)。このスイッチでは、GOIオープンリーディングフレーム(ORF)が2つの部分に分割される:少なくとも翻訳開始コドンおよびタンパク質の非機能的N末端部分をコードする可変数のアミノ酸を含む5’部分、およびSTOPコドンまでの残りのタンパク質をコードする3’部分(図4A)。GOIの5’部分に関連するプロモータは、GOIの3’部分に対してヘッドトゥーヘッド(反転)方向に置かれ、導入遺伝子のこれら2つのエレメントは、piggyBac TRの1つによって分離されている。PBaseの作用に駆動されたゲノム組込み時に、PB−IDベクターの再配置により、GOIの2つの部分がプロモータの制御下に置かれた機能的なORFに再結合し、GOI転写および翻訳の完了が可能になる。GOI配列のマイナーな変更は、リーディングフレームを変更することなく、結果として生じるタンパク質の配列の最小限の変更で、ORFにpiggyBacフットプリント(組込み時にGOIの2つの部分の再結合から生じるTTAA配列)を組み込むために行われる。このような構成をPB−zIDProm∞GOIと表記する。
PB−ID翻訳スイッチに基づいて以下のGOIを発現するために4つの異なるベクターを設計した(図4および5)。EGFP(緑色蛍光タンパク質)、mRFP1(赤色蛍光タンパク質、Campbellら,2002)、IRFP670(近赤外蛍光タンパク質、Shcherbakova and Verkhusha,2013)、および部位特異的リコンビナーゼCre。GOI ORFを、以下の方法で5’と3’の部分に分割した:最初の3つのベクターである、CAGプロモータからの蛍光タンパク質を発現しているPB−zIDCAG∞EGFP、PB−zIDCAGcoRFP、およびPB−zIDCAG∞IRFPでは、GOIの5’部分は、タンパク質の最初の3〜6個のアミノ酸をコードしている(図4A)。CMVプロモータから部位特異的Creリコンビナーゼをコードする4番目のベクター(PB−zIDCMV∞Cre)では、CreORFは、別々に不活性であることが知られている2つの5’および3’部分に分割される(Jullien,2003)(図5A)。さらに、エピソームベクターによる組込み前に発現されるCreのNt部分(NtCre)を不安定化するために、以下の変更が行われる:Creコード配列の対応する5’部分の後に、急速分解のためのPEST配列である下流piggyBac5’TR(Rogersら,1986)、および生じた融合産物(NtCre−5’TR−PEST)が転写かつ翻訳され得るように配置された転写ターミネータが続く。
古典的なpiggyBacトランスポゾンおよびPB−IDベクターなどのpiggyBac系に基づくベクターでは、トランスポサーゼの作用は、piggyBacベースのベクター自体のエピソーム形(Wangら,2014)、またはpiggyBacトランスポサーゼをコードするベクターへの望ましくない自動または相互組込みにつながり得る。このようなエピソーム間(「自殺」とも称される)組込みの可能性を可能な限り減らし、ゲノム組込みを支持するために、pUC57−miniプラスミドに基づき、CAGプロモータおよび単一塩基対の置換によってすべてのTTAAサイトを除去したRFPレポーター導入遺伝子を含有するベクター骨格を設計し、アセンブルした。置換を、細菌におけるベクターの複製にも、真核細胞におけるその機能にも影響を及ぼさない方法で行った。この設計に基づいて、2つのベクターをアセンブルした。
TTAAのないPB*−zIDCAG∞RFPおよび*CAG::PBaseベクターを、E.Coliコンピテント細胞において形質転換し、それらは、正常に複製した。次いで、それらを、HEK−293細胞においてインビトロで試験し、ニワトリ胚脊髄での卵内エレクトロポレーションによってインビボで試験した(図6)。どちらの場合も、PBaseに依存したGOI(RFP)の発現が観察された。
3.1)PB−IDベクターを用いた迅速な薬物を含まない安定な細胞株の確立(図9)
古典的なゲノム組込みベクターで安定な細胞株を確立する場合、宿主細胞のゲノムに組み込まれた導入遺伝子からのGOI(または選択マーカ)の発現は、エピソームベクターの除去前に確実に評価することはできない。これは、典型的に、複数回の細胞分裂を必要とし、トランスジェニック細胞の選択と分析に数週間の遅延をもたらす。さらに、異常に高いレベルでのGOIの一過性エピソーム発現は、代謝および遺伝子発現を混乱させ、標的細胞の挙動、同一性、生存率に潜在的に有害な影響を与える可能性がある。PB−IDスイッチは、GOIおよび/またはマーカの発現をゲノム組込み導入遺伝子に制限することにより、これらの問題を回避することができる。したがって、導入遺伝子が組み込まれている細胞は、トランスフェクションの直後(48時間)にGOIおよび/またはマーカの発現によって同定でき、一過性のエピソームに関連する発現のバーストが回避される。
PB−IDスイッチによる高効率の薬物を含まない安定な細胞株の確立を、PB−ID転写スイッチ(PB−IDCAGcoRFP)に基づくRFPを発現するベクターで検証した。この目標のために、HEK−293細胞を、PBaseとともにPB−IDCAG∞RFPベクターまたは古典的なpiggyBacトランスポゾンベクター(PBCAG::RFP)のいずれかでトランスフェクトした。培養2日後、RFP陽性細胞を選別し、FACSにより96ウェルディッシュに単一細胞として蒔いた。10日間の増殖後(薬剤選択の非存在下)、PB−IDベクターでトランスフェクトされた細胞に由来するクローンの95.78%±0.73SEMがRFPを発現したが、同じ条件では、古典的なpiggyBacベクターに由来するクローンの55.18%±5.07SEMのみがRFP陽性であった(図9A)。これらの結果は、エピソーム希釈を待たずに、GOIまたはマーカ発現に基づいて陽性細胞を選別することにより、高効率でトランスジェニック細胞株を産出するためのツールとしてPB−IDスイッチを検証している。
マウスからのES細胞にPB−zIDCAG∞GFP−KrasおよびCAG::hyPBaseベクターをトランスフェクトした。マウスES細胞では、PB−zIDCAG∞GFP−Krasベクターを用いた蛍光ベースのクローン選択により、古典的なトランスポゾンと比較して組込み事象が高度に強化されていることが実証された(図11A)。したがって、転位へのPB−IDスイッチの依存性は、効率的な導入遺伝子活性化および効率的なゲノム組込みを可能にする。重要なことに、PB−IDベクターを用いた短期および長期の生存率は、古典的なトランスポゾンの生存率と同等であり(図11Bおよび11C)、スイッチは、HeFaおよび3T3を含めたすべての試験した細胞で活性であった(図11D)。これらの結果は、PB−IDを、エピソーム発現を妨げることなく、トランスフェクションの直後にGOIのゲノム発現を評価できる非常に効率的な薬物を含まない遺伝子組換えのツールとして確立している。
複数の導入遺伝子を安定して共発現する細胞株を選択して確立することは、古典的なプロトコルでは困難である。例えば、薬剤耐性に基づくマルチプレクシング選択には、いくつかの別個の薬剤の同時または連続適用、トランスフェクトされた細胞に有害な効果を与える可能性のある繊細で複雑な手順が必要である。さらに、安定な遺伝子導入のための古典的なアプローチの限定された収量のために、複数の導入遺伝子を共組込みする細胞は、典型的に、トランスフェクトされた細胞の少数のみを表す。別個の蛍光タンパク質マーカを有するPB−IDベクターを用いて細胞をコトランスフェクトすると、複数の導入遺伝子を迅速かつ高収率で共発現する細胞株を選択できる可能性がある。
PB−IDでの高効率の薬物を含まないマルチプレックスで安定な細胞株の確立を、PB−ID翻訳スイッチ(PB−zIDCAG∞RFP、PB−zIDCAG∞EGFP、およびPB−zIDCAG∞IRFP)に基づいてmRFP1、EGFP、およびIRFP670を発現する3つのベクターを使用して検証した。この目標のために、HEK−293細胞を、PBaseの存在下で、3つのPB−IDベクターの混合物、または古典的なpiggyBacトランスポゾンベクター(PBCAG::RFP、PBCAG::EGFPおよびPBCAG::IRFP)のいずれかでコトランスフェクトした。培養2日後、3つの蛍光タンパク質を共発現する細胞を選別し、FACSにより96ウェルディッシュに単一細胞として蒔いた。10日間の増殖後(薬物選択の非存在下)、3つのマーカを共発現する細胞に由来するクローンの大部分(79.64%±3.76SEM)は、発現を維持したが、3つのpiggyBacベクターでのトランスフェクションでは、トリプルトランスジェニッククローンの22.43%±2.22SEMのみをもたらした。これらの結果は、エピソーム希釈を待たずに、マーカ共発現に基づいて陽性細胞を選別することにより、高効率で複数の導入遺伝子を安定して発現する細胞株を産出するためのツールとしてPB−IDスイッチを検証している。
iPS細胞を、3つのPB−IDベクター(PB−zIDCAGcoRFP、PB−zIDCAG∞GFP、およびPB−zIDCAG∞IRFP)の混合物でコトランスフェクトした。3つの蛍光タンパク質を共発現するクローンは、同様に、複数の継代にわたってほぼ均一なレベルでそれらの発現を維持した(図12)。3色のPB−zID導入遺伝子は、iPS細胞が神経系統に分化する間の長期的な発現も可能にした。したがって、PB−IDは、細胞培養モデルにおけるマルチプレックス遺伝子組換えのための効率的なツールを提供する。
(エピソーム導入遺伝子の効果とは対照的に)トランスフェクトされた細胞のゲノムに組み込まれた導入遺伝子の効果をモニタリングすることは、例えば、一定の生理学的レベルで、かつ/または後成的因子の調節下で長期間発現された場合に、機能獲得および機能喪失実験において、ある遺伝子産物の機能を実験的に評価することなど、様々な意図のために望ましい。エピソーム形で活性な古典的なベクターでは、エピソームからの一過性の導入遺伝子の発現が、組み込まれた導入遺伝子の発現を隠し、非生理学的レベルでの発現のバーストおよび発現の急速な変化を引き起こす。誘導性戦略は、ある場合にこの問題を回避することを可能にする場合があるが、それでも分析を著しく遅らせるエピソーム希釈を必要とする。PB−IDスイッチは、発現をゲノム組込み導入遺伝子に制限するため、これらの導入遺伝子によって生成されるGOIの効果を明確にモニタリングできる。次いで、蛍光タンパク質などのマーカの共発現を使用して、機能的実験においてGOIを生成する細胞を特定できる。
PB−IDベクターを用いて遺伝子モザイク分析を行う可能性を検証するために、神経前駆細胞の分化に及ぼす阻害効果で知られるNotch受容体細胞内ドメインをGOIとして使用した(Pierfeliceら,2011)。RFPおよびNICDのPBase依存性共発現を可能にする(1.2.6)に提示したPB−zIDCAG∞RFP−2A−NICDベクターを使用した(図7)。このベクターを、ニワトリ胚脊髄において、PBaseおよびコントロールベクター(PB−zIDCAG∞EGFP)でコトランスフェクトした。インキュベーションの3日または6日後、標識された細胞の分析は、NICDの長期発現に期待される効果:GFP+細胞への影響が少ない一方でRFP+細胞に影響を与える神経新生の大幅な減少、およびコントロール条件と比較した心室表面に位置するRFP+前駆細胞の拡大(zPB−IDCAG∞EGFPのエレクトロポレーションに対応)を示した。したがって、PB−IDスイッチを使用して、特定の遺伝的動揺を誘導し、影響を受けた細胞および影響を受けていない細胞を別個の色標識で追跡できる。
細胞系統追跡およびクローン分析アプローチは、細胞集団または個々の細胞のレベルで、インビトロまたはインビボで幹/前駆細胞の運命を分析するための組織発達およびホメオスタシスの研究で広く使用されている。これには、目的の細胞を、それらの分裂を通して伝達される永続的な標識でマークする必要がある。この意図で利用できるツールの中で、ゲノム組込み型トランスポゾンベースのベクターは、それらの単純さおよび幅広いモデルへの直接的な適用性のために魅力的である。さらに、トランスポゾンに基づく多色標識アプローチが開発され、子孫または異なる個々の前駆細胞を別個の色マーカでマークすることにより、クローンおよび/またはクローン境界の同定を促進した(Figueres−Onateら,2016;Garcia−Morenoら,2014;Foulierら,2014;Siddiqiら,2014)。しかしながら、トランスポゾンベースのクローン標識スキームの一般的な問題は、組み込まれていないトランスポゾンベクターから発現されたマーカが、細胞分裂によるエピソーム希釈の前に組み込まれた導入遺伝子と最初に重なるため、クローン同定が妨げられることである。マーカ発現をゲノム組込み標識に制限することにより、PB−IDスイッチは、この問題を回避することを可能にする。したがって、発達中の脊椎動物の神経系におけるインビボでの長期の細胞系統追跡および多色クローン分析のためのPB−IDベクターの有効性が実証された。
−長期的な細胞系統追跡(図10A):
モデルとしてマウス大脳皮質を使用して前駆細胞系統を追跡するためのPB−IDベクターの適用性を検証した。PB−IDCAG∞RFPベクターを、PBaseおよびコントロールCAG::GFPプラスミドとともに、大脳室を裏打ちする胚性皮質前駆細胞においてE12で子宮内にエレクトロポレーションした。E18(エレクトロポレーションの6日後)では、予想通り、GFP発現は、エレクトロポレーション時またはその直後に生じた大脳皮質の中間層を占めるニューロンに制限された(前駆細胞分裂を通したCAG::GFPプラスミドの希釈から予想されるように、その後に生じた細胞は標識されなかった)。対照的に、RFPの発現は、親前駆細胞の恒久的な標識の場合に予想されるように、心室表面から中間および上部皮質層に向かって放射状に移動する細胞の流れで見出された。P10(エレクトロポレーションの15日後)では、RFP標識は、中間層および上部皮質層の分化したニューロン、ならびに皮質壁全体に位置する星状細胞で観察されたが、GFP発現は、再び中間層ニューロンに制限されていた。この発現のパターンは、PB−IDベクターを用いた皮質前駆細胞の恒久的な標識とのみ適合性がある。したがって、PB−IDスイッチを使用して、長期間の発生期間にわたって、インビボで前駆細胞/幹細胞の系統を追跡することができる。
蛍光タンパク質メーカーの別個の色を発現するベクターの混合物を使用して、PB−IDスイッチに基づく多色細胞系統追跡スキームを開発した。この目標のために、PBaseとともにE2ニワトリ胚の脊髄に半疎な方法で、卵内で同時エレクトロポレーションした(1.2.3)で述べた3つのベクター(PB−zIDCAG∞RFP、PB−zIDCAG∞EGFP、およびPB−zIDCAG∞IRFPベクター)を用いた。E6での分析により、様々な異なる色で均一に標識されている心室表面から放射状に移動する細胞の平行な流れが明らかになった。このような分布は、他のアプローチにより脊髄で観察されたクローンパターンに対応していた(Leber and Sanes,1995;Loulierら,2014)。細胞は、3つの蛍光マーカの豊富な組合せを発現し、単色のクローン標識で必要な密度よりも高い全体的な標識密度でクローンを同定することを可能にした。PB−IDベクターを用いて観察された色パレットは、以前のトランスポゾンベースの多色系統追跡方法で得られたものと同じかそれ以上に拡張されていた。したがって、PB−IDスイッチは、クローンに関連する細胞の群および/または他のクローンに対するそれらの境界を標識し、マッピングするための効率的なツールである。

f)piggyBacトランスポゾンの5’ITRを含むか、またはそれからなる第1の配列およびpiggyBacトランスポゾンの3’ITRを含むか、またはそれからなる第2の配列にある2つの配列のセットであって、前記配列が互いに対して反対の方向にある、2つの配列のセットと
を含み、
−前記目的の遺伝子の配列が前記目的の遺伝子の5’末端部分および前記目的の遺伝子の3’末端部分にある2つのフラグメントの形で本発明の組換えベクターに存在し、
−前記目的の遺伝子の配列の3’末端部分が前記プロモータの配列に対してアンチセンス方向にあり、
−前記目的の遺伝子の配列の5’末端部分が前記プロモータ配列の下流に局在しており、前記プロモータの方向に対してセンス方向にあり、
−piggyBacトランスポゾンの5’ITRを含むか、またはそれからなる前記セットの配列が前記プロモータ配列の下流に局在しており、前記プロモータの方向に対してセンス方向にあり、前記プロモータを前記目的の遺伝子の配列の3’末端から分離する、請求項5に記載の組換えベクター。 - トランスポサーゼをコードする配列をさらに含む、請求項1〜6のいずれかに記載の組換えベクター。
- 好ましくは請求項1〜7のいずれかに記載の組換えベクターを作製するための、空の組換えベクターであって、
a)第1のマルチクローニング部位と、
b)第2のマルチクローニング部位と、
c)DNAトランスポゾンの5’ITRを含むか、またはそれからなる第1の配列およびDNAトランスポゾンの3’ITRを含むか、またはそれからなる第2の配列からなる2つの配列のセットであって、前記配列が互いに対して反対の向きにある、2つの配列のセットと
を含み、
前記セットの2つの配列のうちの1つが、前記第1のマルチクローニング部位と前記第2のマルチクローニング部位との間に位置している、空の組換えベクター。 - 請求項8に記載の空の組換えベクターまたは請求項1〜7のいずれかに記載の組換えベクターと、
好ましくはタンパク質として、またはトランスポサーゼ発現ベクターの形で提供されるトランスポサーゼ酵素と、
を含む、キット。
